To elucidate the origin of collagen hyperfibrils, we have ultrastructurally investigated the development of collagen fibrils after in situ treatment of liver tissue sections by different proteolytic enzymes. After treatment, collagen fibrils were largely disorganized within the same fibril bundle. A wide range in diameters was observed. Hyperfibrils, which correspond to those fibrils with the largest diameters and having a flower-like section, were present. On the basis of these results, we can suggest that hyperfibrils reflect breakdown states of collagen fibrils.

In the present study rat hepatocytes isolated either by trypsin or by collagenase were investigated concerning its cultivability and its morphological properties. The cultivation was carried out as monolayer for 24 h. The yields of cells prepared by trypsin or collagenase amounted to 8 x 10(7) cells per liver and 15-30 x 10(7) cells per liver, respectively, with viabilities measured by trypan blue exclusion test of 70-80 and 90-95%, respectively. Nonhepatocytes were not taken into consideration. Using the electron microscopy it could be established that both freshly isolated and 24 h cultured hepatocytes were intact. There were no morphological differences between cells isolated by trypsin and cells isolated by collagenase. After 24 h cultivation hepatocytes prepared by trypsin showed a little tendency in forming a slightly flattened appearance and in forming intercellular contacts.

1. The influences of enzyme treatments (trypsin and collagenase) on responses to perfused acetylcholine were examined on physically isolated single Aplysia neurons, using the voltage-clamp, internal perfusion, and rapid external perfusion technique. 2. During treatment with trypsin (0.025 to 0.1%) for 10 to 30 min at room temperature (22 to 25 degrees C), the peak amplitude of the Na current induced by acetylcholine increased in a time- and dose-dependent manner, and the decay in the continued presence of acetylcholine was slowed. This effect of trypsin treatment was irreversible after washing for 60 min without enzyme. 3. Edrophonium, a cholinesterase inhibitor, has previously been shown to augment the Na acetylcholine response in this preparation by inhibition of acetylcholinesterase. After treatment of the neuron with trypsin, the augmentation after edrophonium was abolished. Furthermore, in the presence of edrophonium, trypsin also failed to increase the response. The dose-response curve for acetylcholine after treatment of trypsin was similar to that in the presence of edrophonium. These results suggest that the modification of the current response by trypsin is a result of removal of cholinesterase activity from the membrane. 4. In contrast to the effects of trypsin, collagenase (0.03 to 0.1%) for 10 to 60 min did not change the current amplitude of the acetylcholine response. However, collagenase treatment did alter the kinetics of the acetylcholine response in a dose-dependent manner, in that the rate of decay was accelerated. A similar acceleration was seen in the acetylcholine responses on other neurons which were due to Cl or K currents, suggesting that the effect was independent on the type of channel. This effect of collagenase was reversible after 30 to 60 min of washing of the neuron. 5. In the presence of edrophonium or after the treatment with trypsin, collagenase still accelerated the current kinetics of the acetylcholine response, indicating that cholinesterase activity is not related to this effect. Furthermore, heated collagenase (presumably inactivated) had a similar action, suggesting that the enzymatic activity of collagenase is not related to the modification of the response. 6. These results suggest that Aplysia acetylcholinesterase is sensitive to trypsin but not to collagenase. However, the preparation of a collagenase used in these studies contains some factor which alters the response to acetylcholine, but this effect is reversible and unrelated to enzymatic activity.

Transmission electron microscopy was used to study the effects of proteolytic enzymes (collagenase, trypsin, clostripain), the calcium chelator ethyleneglycol-bis-(beta-aminoethyl ether) N,N,N',N'-tetraacetic acid (EGTA), and the calcium ionophore A 23187 on substrate adhesion and fine structure of chondrocytes and fibroblasts. Monolayer-cultured cells responded to treatment with the proteolytic enzymes followed by EGTA or A 23187 by rounding and detaching from the substrate. This was accompanied by the formation of a microvillous surface, deep nuclear folds, and numerous cytoplasmic vacuoles. Labeling experiments with colloidal thorium dioxide indicated that the vacuoles were formed by endocytosis and fusion of endocytic vesicles with preexisting lysosomes. To a variable extent, similar changes were produced by trypsin or EGTA alone. The cells regained their normal fine structure after withdrawal of the reagents and when seeded onto a substrate. In suspension culture, recovery was incomplete; the cells retained a rounded shape and an increased number of cytoplasmic vacuoles. The results suggest that changes in plasma membrane composition and its permeability to calcium represent the primary signal for cell rounding and detachment. The cellular mechanisms responsible for the associated folding of the nuclear envelope and the cell surface remain unidentified. Nevertheless, this is believed to represent a means of handling of excess membrane during sudden transition from a flattened to a rounded shape. Membrane stored in folds and vacuoles is reutilized when the cells reattach and spread out on a substrate.

The electrophysiological and pharmacological properties of aggregates prepared from cells of 7-day-old chick embryo heart ventricles depend on the enzyme used for cell dissociation. The mean beat rate of aggregates formed from trypsin-dissociated cells was about 53 beats/min whereas aggregates formed from collagenase-dissociated cells had a mean beat rate of more than twice this value. Spontaneous activity of most aggregates formed from trypsin-dissociated cells was inhibited by elevating external potassium or by adding tetrodotoxin to the medium. A similar response to potassium was seen in all aggregates formed from collagenase-dissociated cells. However, approximately half of the aggregates formed from collagenase-dissociated cells were tetrodotoxin insensitive. Intracellular microelectrode recordings demonstrated that aggregates formed from collagenase-dissociated cells typically had reduced action potential maximal upstroke velocities and depolarized threshold potentials in comparison to those recorded from aggregates formed from trypsin-dissociated cells. In the presence of tetrodotoxin the maximal upstroke velocity of aggregates formed from either collagenase- or trypsin-dissociated cells decreased markedly. In the case of the collagenase-treated cells, the spontaneous activity which persisted in the presence of tetrodotoxin was abolished by the slow channel blocker D-600. Computer simulation of membrane depolarization supports the view that aggregates formed from collagenase-treated cells have a reduced fast inward sodium current and a significant leakage current. Aggregates prepared from trypsin-dissociated cells display properties which more closely resemble those of intact 7-day embryonic ventricular tissue. We therefore conclude that, contrary to previous reports, collagenase is not the enzyme necessarily best suited for cell dissociation in all tissue culture studies.

The authors showed previously that the effusions of the experimental polyseritis after intraperitoneal injections of trypsin and elastase, go from peritoneum to the pleural cavities and never from thorax to the peritoneum. The intraperitoneal injections of collagenase or of collagenase and trypsin can also cause polyseritis in the rat; but more often they provoke heavy hemorrhagic lesions of the wall of the abdomen and the diaphragm. Perforations of the diaphragm were observed in 8 rats of 32, with in 6, a intrathoracic hernia of the liver or the stomach. After the intrapleural injection of collagenase or of collagenase and trypsin, hemorrhagic lesions were seen in the thorax, but not in the abdomen. These facts are a new proof for the transdiaphragmatic propagation of the proteasic solutions injected in the peritoneum.

Experiments are presented in which membrane lipids of endothelial cells in monolayer culture were labelled with [14C]linoleic acid. Approx. 90% of the radioactive label were incorporated into phospholipids. A comparison of various harvesting methods showed that during the disruption of the labelled endothelial cell monolayer, 0.25% trypsin and 0.125% trypsin (+0.01% EDTA) released 650 and 470% more radioactivity, respectively, than did 0.01% collagenase (+0.01% EDTA). Parallel studies were performed on a green monkey kidney cell line. In this case, 0.25% trypsin released 520% more radioactivity than did 0.1% collagenase (+0.01% EDTA), although 0.125% trypsin in the presence of EDTA (0.01%) was much less traumatic than trypsin alone, the released radioactivity being of the same order of magnitude as that for collagenase. Morphological studies on endothelial cell cultures failed to reveal any distinctive differences in surface morphology following the various enzyme treatments. The results suggest that collagenase treatment of endothelial cell monolayers is the least traumatic harvesting or subculturing method as far as the integrity of the lipids in the cell membrane is concerned.

The present study deals on the modifications produced by three proteolytic enzymes in the interiorization of cultured metacyclic forms of Trypanosoma cruzi in peritoneal macrophages of non-stimulated mice. The proteolytic enzymes used were trypsin, papain and collagenase. Pretreatment of metacyclic forms or macrophages with the enzymes resulted in lower penetration ratios with respect to controls. On the other hand, percentages of parasitization were maintained throughout the periods of time assayed.

In the rat peritoneal injections of collagenase or trypsin give rise to severe lesions. In our experience 20% of the animals remain intact. The frequency of lesions increases with older and heavier subjects. Moreover 25% of the rats who remained free of lesions after a first injection of collagenase resist to a second one. This shows that they are strongly protected against the enzyme. The exact nature and location of this protective mechanism are not known.

A transplantable rodent tumor producing multiple layers of basement membrane was used to study the effects of trypsin, hyaluronidase and collagenase on basement membranes. Treatment with trypsin resulted in an increase in the distance between adjacent lamellae and a loss of granular structures. Treatment with hyaluronidase separated basement membrane layers only in the outer lamellae, whereas collagenase resulted in extensively folded sheets which consisted predominantly of granules. From these findings it may be concluded that the granular structures represent the morphological equivalent of glycoproteins which are interlinked by a collagenous filamentous network. Hence, the BM represents a functional unit of proteoglycans, glycoproteins and collagen.

Recent studies with long-term mouse marrow cultures have indicated the importance of the adherent layer as a primary reservoir of the most primitive stem cells, from which derivative stem cells and more differentiated progenitors are continuously generated. We have now examined the role of the adherent cell layer in long-term human marrow cultures from this point of view. Prerequisite to such an undertaking was the development of a nontoxic and reproducible method for detaching the adherent layer and making it into a single-cell suspension suitable for characterization by colony assays. Both trypsin and collagenase could be used to obtain suspensions that met these criteria. Lack of toxicity was demonstrated by the preservation of CFU-E, BFU-E, and CFU-C plating efficiency in fresh human marrow cell suspensions exposed to the same enzymatic treatments. Collagenase treatment of long-term marrow culture adherent layers was considered superior because it freed all hemopoietic colony-forming cells but left some of the other cells still adherent. Using this method, we found that CFU-C, BFU-E, and CFU-G/E were consistently detectable in the adherent layer for at least 8 wk, with the majority of the BFU-E and CFU-G/E being located in the adherent layer (70%-75% after 2-3 wk and more than 90% by 7-8 wk). Although corresponding numerical differences in adherent and nonadherent CFU-C populations were not observed, the colonies derived from them showed marked differences in the size they achieved; the adherent layer being the exclusive site of CFU-C, with a very high proliferative capacity. These findings emphasize the importance of assessing the progenitor content of the adherent layer of long-term human marrow cultures and provide an appropriate methodology.

It was demonstrated in this study that the 3-dimensional, "pseudo-random" architecture of collagen in the general matrix of normal cartilage can be transformed enzymatically into a radial array of fibril aggregates or fibers. By first degrading the proteoglycans and then attacking the collagen, it is possible to produce a collagenous structure almost identical to that observed in matrices exhibiting both nonprogressive softening and osteoarthritic changes, and in matrices subjected to dynamic overloading. This structural transformation is explained as a breakdown in the fibril interlinking system.

A new technique for pancreatic islet isolation, based on trypsin administered into the pancreatic duct system and a reduced amount of collagenase for digestion of the removed and chopped pancreatic tissue, yielded viable islets as judged by the metabolic response of 27 inbred, streptozotocin-diabetic rats after intraportal transplantation of the islets: all recipients of greater than 240 islets normalized their blood glucose, plasma insulin, urine volume and urinary glucose. The number of islets isolated was the same as with the conventional collagenase technique.

Frog dorsal root ganglion (DRG) cells treated with collagenase and trypsin were dissociated mechanically. The characteristics of dose-response curves of GABA-induced Cl- currents (ICl) were mainly studied using the "concentration-clamp" technique, by which the external solution around isolated single neurons was rapidly changed within a few milliseconds. The saturating maximum currents induced by various concentrations of GABA normalized to the peak ICl induced by 10(-5) M GABA on the same neuron varied from 1.5 to 5.5 among the neurons, though the absolute maximum currents varied little. The Hill coefficients (n = 1.92 +/- 0.09, mean +/- S.D.) also varied little on the neurons. The time constants of the desensitization phase of fully activated GABA responses were not so markedly different among the neurons. The present results suggested a possibility that the enzyme treatment produced a selective inactivation of alpha 1 unit, a high affinity subtype of the GABA-receptor.
