The roots of Ligusticum porteri J.M. Coult. & Rose, Apiaceae, are used in Mexican traditional medicine for the treatment of gastrointestinal disorders. The dimeric phthalide (Z´)-diligustilide found in their organic extracts is associated with the gastroprotective properties. In this study, the gastroprotective effect of nanoemulsion of the volatile oil from the roots of L. porteri (at doses of 1, 3, 10, and 30 mg/kg) in the indomethacin-induced gastric damage model was tested, and nitric oxide (NO) and hydrogen sulfide (H2S) gastric levels were determinate. The volatile oil obtained by steam distillation (0.63% yield) contained 37 constituents, as assessed by GC-MS analysis, where the significant compounds were 4-phenyl-3-buten-2-ol (20.54%) and (Z)-3-butylidenephthalide (20.54%) that correspond to phthalide monomers. These results demonstrate that nanoemulsion of the volatile oil from L. porteri (at 10 and 30 mg/kg) shows a significant gastroprotective effect (72 and 92% respectively) against indomethacin-induced gastric damage, which is not different from the gastroprotective effect of omeprazole (69%) (p

The inhabitants of the floodplain of the Mazagão River in the State of Amapá in the Brazilian Amazon have inherited from indigenous African and Cabocla cultures indications for the use and forms of preparation of medicinal plants to cure diseases of the body and spirit. This study aimed to perform an ethnopharmacological survey of medicinal plants used by the riparian community of the floodplains of the Mazagão River, in the State of Amapá. In this study, we chose semistructured interviews with socioeconomic, ethnopharmacological, and ethnobotanical aims. The collection of medicinal plants occurred during guided tours. The Use Value (UV), Informant Consensus Factor (ICF), Correction Factor (CF), and Fidelity level (FL) were calculated. There were 130 species of medicinal plants, distributed in 116 genera and 57 families; Fabaceae (16), Lamiaceae (14), Euphorbiaceae (7), and Arecaceae (6) include 33.33% of the total species sampled. All 95 native species of floodplain forests were previously described, and 35 are exotic species. The species with the highest UV (≥ 0.5) at the mouth of the Mazagão River were Carapa guianensis (0.91), Pentachlethra macroloba (0.83), Dalbergia subcymosa (0.77), Uncaria tomentosa (0.75), Otacanthus azureus (0.62), Virola surinamensis (0.62), Hura crepitans (0.58), Euterpe oleracea (0.56), and Arrabidaea chica (0.51). These species were also the ones that presented the highest ICF among the informants and 100% in FL for a specific therapeutic use. The study is comprised of 16 categories of therapeutic use, of which the majority of the plants used are related to diseases such as microbial infections (20.67%, 73 species), gastrointestinal disorders (13.31%), and inflammation (11.61%). The results showed that knowledge about the use of medicinal plants along the rivers and streams that form the mouth of the Mazagão River is evenly distributed. Most of the interviewees present diversified knowledge about the medicinal resources because they have a close relationship with the floodplain forest. Native species of this forest predominate among the most commonly used medicinal plants as subsidies for future pharmacological studies.

Ethnopharmacological relevance Plant species Rosmarinus officinalis L. (Lamiaceae; Synonyms: Salvia rosmarinus Schleid. and Rosmarinus angustifolius Mill.) is a herb widely used worldwide. In local and traditional medicine, its used for inflammation-related diseases. Currently, studies report anti-inflammatory activity in its essential oil (EORO). However, to better understand EORO’s anti-inflammatory activity its necessary to understand its phytochemistry and the signaling pathways affected by it. Hence, this review aimed to describe EORO phytochemical profile, ethnopharmacological uses, some biological activities of EORO will be described but emphasizing its anti-inflammatory potential and possible mechanisms of action involved. Materials and methods The research was performed using the databases Medline, Embase, BVS Regional Portal, Science Direct, CAPES Journals, and Scopus; using the keywords “Rosmarinus officinalis”, “anti-inflammatory” and “essential oil”. Additional information was gathered from related textbooks, reviews, and documents. Results and discussion Until now about 150 chemical compounds were identified in EORO samples, more frequently reported molecules were 1,8-cineole, α-pinene, and camphor. Studies suggest that the anti-inflammatory activity of EORO occur mainly through inhibition of NF-κB transcription and suppression of arachidonic acid cascade. Its antioxidant activity also aids by preventing injury caused by the reactive species of inflammation; its smooth muscle relaxant activity contributes to ameliorating airway inflammatory diseases. Lastly, toxicity assessments indicate low toxicity to EORO. Conclusions Current evidence indicates anti-inflammatory activity in EORO, supporting its ethnopharmacological uses in inflammatory-related diseases, and potential future applications. However, although considerable acute inflammatory models were tested, more chronic inflammatory models are needed; clinical studies are still absent, this may be due to the high doses needed for essential oils to exert pharmacological effects, but recent studies show this issue can be bypassed using the oil formulated as nanoemulsions to improve its bioavailability.

Ethnobotanical relevance Spondias mombin L. is a plant dispersed throughout the tropical regions of South America, Africa, and Asia, being found mainly in the North and Northeast of Brazil, where the leaves are used in preparations for neuropsychiatric disorders. Therefore, it is of great importance to carry out studies in different pharmacological models that can prove the traditional use of this plant species. Materials and methods the hydroethanolic extract from S. mombin leaves (HELSm) was evaluated by oral administration (25 mg/kg) and by immersion (25 mg/l) in scototaxis test in zebrafish (Danio rerio). For this study, caffeine (100 mg/kg) and buspirone (25 mg/kg) were used as standard drugs. The antidepressant action of the HELSm was evaluated assessed in the novel tank diving test (NTDT). In this study, a group with 1% ethanol, one with unpredictable chronic mild stress (UCMS), and another with developmental, social isolation (DSI) were used as induction groups for depression-like behavior and fluoxetine (20 mg/kg) as a drug pattern. Results by the HPLC-UV fingerprint analysis, the HELSm presented several derivatives of polyphenolic compounds and flavonoids and identified ellagic acid and isoquercitrin, and by the gas-chromatographic, the majority of the identified compounds were fatty acids, esters, and alcohols. By immersion, the LC50 was 49.86 mg/l and by oral via the LD50 in 48 h, was 4.515 g/kg in zebrafish. For all spatiotemporal and behavioral variables (time spent, white compartment, latency, toggle, erratic swimming, freezing duration, thigmotaxis, and risk assessment), the treatment with HELSm produced a similar effect to buspirone and was significant when compared to the caffeine and control group (p Conclusions Our results suggest that the acute administration of the HELSm in the scototaxis and NTDT tests in a zebrafish model (Danio rerio) produced anxiolytic and antidepressant effects, devoid of hypnotic and sedative actions by immersion, and this action was improved when administered by oral via. Possibly, the presence of isoquercitrin in the leaves of Spondias mombin participates in the anxiolytic and antidepressant effects.

The plant species Libidibia ferrea (Mart. ex Tul.) LP Queiroz var. ferrea basionym of Caesalpinia ferrea (Mart. ex Tul.) is used in various regions of Brazil in folk medicine in the treatment of several health problems, especially in acute and chronic inflammatory processes. Most of the preparations employed are alcoholic. Therefore, this study aimed to evaluate the acute toxicity of the hydroethanolic extract of fruits of Libidibia ferrea (EHEFLf) in zebrafish, emphasizing the possible changes in the organic-cellular level of the gills, liver, kidneys, and intestine and on embryos. The result obtained by LC-M/MS from EHEFLf indicated a high concentration of possible polyhydroxylated substances. EHEFLf, at a dose of 2 g/kg orally, produced non-significant alterations of the analyzed organs. However, for embryos, the treatment with different concentrations demonstrated heart toxicity that was concentration-dependent. There is no evidence of a correlation of the observed effects with the phytochemical composition, and considering the species of animal used, it can be suggested that the oral use of L. ferrea hydroethanolic extract has an acceptable degree of safety for use as an oral medicinal product. and embryo results have shown significant affinity to the heart, however, it is perceived to be related to the concentrations used.

Mosquitoes, such as Aedes aegypti and Culex quinquefasciatus, are important vectors of diseases, such as dengue fever, chikungunya fever, Zika virus, and filariasis, and these diseases are public health problems. The present study was carried out to evaluate the larvicidal activity of the hydroethanolic extract from leaves of Acmella oleracea leaves against 3rd instar larvae of the Ae. aegypti dengue vector and the Cx. quinquefasciatus filariasis vector. The hydroethanolic extract caused significant mortality in Ae. aegypti and Cx. quinquefasciatus. After 24 h of exposure to the extract, it was possible to establish the LC50 values for the extract: 11.41 ppm for Ae. aegypti and LC50 32.40 ppm for Cx. quinquefasciatus. The hydroethanolic extract from leaves of A. oleracea showed very low ecotoxicity suggesting that it can be used without causing environmental damage. This is the first study that shows the use of hydroethanolic extract from leaves of A. oleracea as an alternative to synthetic larvicides to eliminate larvae of Ae. aegypti and Cx. quinquefasciatus in an easy, cheap and safe way.

Ethnopharmacological relevance Endopleura uchi (Huber) Cuatrec is a plant species from the Brazilian Amazon. The barks of this tree are used in folk medicine – mainly as a decoction – for dyslipidemia, uterine infection, fibroids, polycystic ovary, menstrual disorders, as a contraceptive and abortive agent, among others. However, the data available about its developmental toxicity are still insufficient. Aim of the study This study aimed to evaluate the reproductive toxicity and teratogenic effects in embryos from zebrafish treated with the hydroethanolic extract from the barks of Endopleura uchi (EEu). Materials and methods Both sexes of zebrafish (Danio rerio) were treated with EEu either through immersion (1.2, 2.5, and 5 mg/L) or orally (75, 200, and 500 mg/kg) over 21 consecutive days. Next, we assessed their fertility and gonads’ histopathology; in their embryos were assessed teratogenesis, lethalities, and heart rate during daily observations (24, 48, 72, and 96 hpf). Results The phytochemical analysis of EEu through HPLC/MS shows bergenin as the major compounds. After 21 days of treatment were detected minor histopathological changes in parental fishes, such as atretic oocytes, interstitial fibrosis, and decreased the percentage of early vitellogenic oocytes, but without impairing the reproduction of treated animals. However, in the embryos was observed significantly increased frequency of malformation in all the groups treated through immersion, and in the group treated orally with the highest concentration (500 mg/kg). Conclusion Based on the results, EEu caused no adverse effects in the progenitors on both treatments (immersion and oral). However, it was observed that the concentrations 1.2, 2.5, and 5 mg/L (immersion), and the dose 500 mg/kg (oral) caused malformations in the offspring (F1 generation). These results emphasize the need for attention when using preparations from E. uchi, mainly for pregnant women. Further studies are needed to compare its effects with the extract's primary compound (bergenin).

The gastroprotective effect of a turmeric acetone extract (TAE) (Curcuma longa L. [Zingiberaceae]) was evaluated and compared against its major curcuminoids; curcumin (CUR), demethoxycurcumin (DMC) and bisdemethoxycurcumin (BDMC). Additionally, to demonstrate the importance of the metabolites’ ratio in the extract on the synergistic effect, different mixtures were evaluated. An ethanol-induced gastric injury model was used to evaluate the gastroprotection activity in Wistar rats. The pharmacologic interaction analysis was performed using the Combination Index (CI)-Isobologram Equation method. The CI calculated at 0.5 of affected fraction (fa) for the TAE indicated a synergistic interaction between its components. However, when the proportion of curcuminoids changed from 3.7:1:10 in TAE to a 1:1:1 ratio, the CI implied an antagonistic effect. The binary combinations of curcuminoids (1:1) also showed an antagonistic interaction. The results of this work suggest that the proportion of curcuminoids in the TAE is crucial for the gastroprotective effect against ethanol-induced damage.

The treatment of Diabetes mellitus (DM) involves drugs for controlling the blood glucose level, and for treating concomitant disorders such as obesity. Based on the potent antioxidant activity, the α-glycosidase and lipase inhibitory effect, and the potent hypoglycemic effect of the fruit extract of Cassia grandis Lf (CgE), in this work it was developed a novel tablet formulation (CgET) using CgE as the active ingredient. The excipients proportion was optimized by using a D-optimal mixture design. The effect of the compaction force (10, 13, 16, 19, and 22 kN) on the technological properties of tablets was evaluated. The optimized granules formulation was compressed at 16 kN, for producing tablets with excellent technological properties. Tablets showed a potent antioxidant effect (IC50 = 1.45 μg/mL), and α-glycosidase inhibitory activity (IC50 = 34.96 μg/mL) more potent than Acarbose (IC50 = 63.25 μg/mL). Tablets exhibited a strong antiglycant effect by the oxidative (IC50 = 22.45 ± 1.80 μg/mL) and non-oxidative (IC50 = 25.49 ± 1.80 μg/mL) pathways, and produced (at 100 mg/kg) a hypoglycemic effect statistically equal to glibenclamide (25 mg/kg). CgET did not show oral acute toxicity (LC50 > 2000 mg/kg), framed as a non-toxic products. Fruits of the Cassia grandis L f, an underutilized species from the Cuban’ eastern region; allow developing a novel tablet formulation that could be used for the treatment of diabetic patients.

Baccharis reticularia DC. is a plant species from the Asteraceae family that is endemic to Brazil. Despite the great importance of Baccharis genus, no study has been carried out regarding either the phytochemical composition of B. reticularia or the evaluation of its larvicidal potential. Considering the intrinsic immiscibility of essential oils, this study shows larvicidal nanoemulsions containing the B. reticularia phytochemically characterized essential oil and its main constituent against Aedes aegypti. The major compound found was d-limonene (25.7%). The essential oil inhibited the acetylcholinesterase, one of the main targets of insecticides. The required hydrophile-lipophile balance of both nanoemulsions was 15.0. The mean droplet sizes were around 90.0 nm, and no major alterations were observed after 24 h of preparation for both formulations. After 48 h of treatment, the estimated LC50 values were 118.94 μg mL−1 and 81.19 μg mL−1 for B. reticularia essential oil and d-limonene nanoemulsions, respectively. Morphological alterations evidenced by scanning electron micrography were observed on the larvae treated with the d-limonene nanoemulsion. This paper demonstrated a simple and ecofriendly method for obtaining B. reticularia essential oil and d-limonene aqueous nanoemulsions by a non-heating and solvent-free method, as promising alternatives for Aedes aegypti control.

The species Acmella oleracea (L.) R.K. Jansen (Asteraceae), popularly known as jambú, is marketed in fairs as a female aphrodisiac and has several pharmacological activities already confirmed, among them the sexual stimulant action. The objective of this study was to evaluate the effects of the oral administration of the hydroethanolic extract of A. oleracea flowers (EHAo) on wistar rats during the pre-mating, mating, and pre-implantation period.During the treatment period, measurements of feed intake, water intake, weight, estrous cycle, behavior, reproductive parameters, biochemical parameters, hematological parameters, and histopathology of ovaries were performed daily.In the gas chromatography analysis - mass spectrometry characterization, the compound (2E, 6Z, 8E) -N-isobutyldeca-2,6,8-trienamide (spilanthol) was detected as the majority compound at the 84% concentration. In the conditions of this study, EHAo did not cause maternal toxicity. However, in the estrous cycle, the frequency of the Proestrous (P) and Estrous (E) phase was significantly increased with the doses of 88.91 and 444.57mg/kg of the EHAo in relation to the control. On the other hand, the metaestrous (M) and diestrous (D) phases showed a significant reduction in their frequency in the groups treated with EHAo. Water intake increased significantly (p0.01), as well as the triglyceride levels, the total cholesterol and fractions (p0.05), and the percentage of neutrophils (p0.05).It is concluded, therefore, that the treatment with EHAo, which is one of the forms popularly used, is safe in the concentrations and time of treatment studied as it is able to influence the estrous cycle without altering folliculogenesis and fertility.

Euterpe oleracea Mart., popularly known as “acai”, is a tropical fruit from the Amazon region where it has considerable economic importance. Acai has been used as food and for several medicinal purposes. Despite the widespread use of this fruit, there is a lack of data regarding the safety of using this fruit oil exclusively. Therefore, we evaluated the in vitro cytotoxic, genotoxic, and antigenotoxic effects of E. oleracea fruit oil (EOO) in cultured human lymphocytes (non-metabolizing cells) and HepG2 cell line (human hepatoma) (metabolizing cells) by using MTT, comet, and micronucleus assays. A wide range of EOO concentrations was tested with a preliminary MTT assay, which allowed selecting five concentrations for comet and micronucleus assays: 2.5, 10, 100, 500, and 1000 µg/mL. The results showed that none of the EOO tested concentrations presented cytotoxic effects. The genotoxic assessment revealed an absence of significant DNA and chromosome damage in human lymphocytes and HepG2 cells but did not show chemoprotection against the DNA damage induced by methyl methanesulfonate and benzo[a]pyrene, used as DNA-damaging agents.

Snakebites envenomations are a problem public healt h in worldwide due to the high rates of morbidity a nd mortality. The Bothrops venom causes local tissue damage and inflammation is one of the most important events that occur. At present, effective treatment for snakebites is serum therapy with antivenom, whi ch neutralizes systemic alterations but does not preve nt local damage that can cause disabilities. Many p lants are used in popular medicine to treat these acciden ts but few attempts have been made to investigate t he scientific validity of these assertions. In Amazon region, indigenous and local people use the macerat ed bark of Brosimum guinanensis applied in the form of cataplasm, on the site of s nakebite. This study aimed to analyze the ability of the Brosimum guianensis aqueous extract in the neutralization several effe cts induced by Bothrops atrox snake venom to investigate the scientific validity of folk medicine informations by means of controlled experiments. Our results sho wed that Brosimum guianensis aqueous extract was not effective to inhibit oedema, peritonitis, coagulant , myotoxic, phospholipase A2 activity (indirect hem olytic method) induced by B. atrox venom , but was able to inhibited significantly hemorrhag ic and nociceptive activities. These results support a potential effec t of this extract as a compounds source for biotechonological application and synthesis of new drugs with therapeutic purpose.

Manilkara subsericea(Mart.) Dubard (Sapotaceae) is popularly known in Brazil as “guracica.” Studies withManilkaraspp indicated the presence of triterpenes, saponins, and flavonoids. Several activities have been attributed toManilkaraspp such as antimicrobial, antiparasitic and antitumoral, which indicates the great biological potential of this genus. In all, 87.19% of the hexanic extract from fruits relative composition were evaluated, in which 72.81% were beta- and alpha-amyrin esters, suggesting that they may be chemical markers forM. subsericea. Hexadecanoic acid, hexadecanoic acid ethyl ester, (E)-9-octadecenoic acid ethyl ester, and octadecanoic acid ethyl ester were also identified. Ethanolic crude extracts from leaves, stems, and hexanic extract from fruits exhibited antimicrobial activity againstStaphylococcus aureusATCC25923. These extracts had highIC50values against Vero cells, demonstrating weak cytotoxicity. This is the first time, to our knowledge, that beta- and alpha-amyrin caproates and caprylates are described forManilkara subsericea.

No. of bitstreams: 0 Previous issue date: 2016-07-01 E. oleracea is a tropical plant from the Amazon region, with its fruit used for food, and traditionally, as an antioxidant, anti-inflammatory, hypocholesterolemic, for atherosclerotic disease, and has anticancer properties. The oil of the fruit has antidiarrheic, anti-inflammatory and antinociceptive activities, but without genotoxicity evaluation. Therefore, the aim of this study was to evaluate the genotoxic potential of E. oleracea fruit oil (EOO), in rat cells. Male Wistar rats were treated with EOO by gavage at doses of 30, 100 and 300 mg/kg, for 14 days, within a 24 h interval. The DNA damage in the leukocytes, liver, bone marrow and testicular cells, was assessed by the comet assay, and the clastogenic/aneugenic effects in the bone marrow cells, by the micronucleus test. Our phytochemicals characterization of the EOO showed the presence of vanillic, palmitic, γ-linolenic, linoleic, oleic, cinnamic, caffeic, protocatechuic, ferulic, syringic acids, and flavonoids quercetin and kaempferol rutinoside as the main constituents. Both cytogenetic tests performed showed that EOO presented no significant genotoxic effects in the analyzed cells, at the three tested doses. These results indicate that, under our experimental conditions, E. oleracea fruit oil did not reveal genetic toxicity in rat cells. Medicinal plants encompass a rich source of active compounds that can neutralize snake venoms or toxins. Costus spicatus (Jacq.) Sw. (Costaceae) is used by the Amazonian population to treat inflammation, pain and other pathological manifestations.To evaluate the influence of C. spicatus aqueous extract on edema, peritonitis, nociception, coagulation, haemorrhage and indirect haemolytic activity induced by Bothrops atrox venom (BAV).Dried and pulverized leaves were extracted with distilled water. Envenoming was induced by administration of B. atrox snake venom in Swiss Webster mice. The experimental groups consisted of BAV (at the minimum dose to induce measurable biological responses) and C. spicatus extract (CSE, 1.25, 2.5, 5.0, 7.5 and 10 mg/kg/25 μl phosphate-buffered saline) administered individually and in combination (BAVCSE). PBS was used as a control. In vitro assays were also conducted in order to evaluate phospholipase A2 coagulant activities (indirect haemolytic method).CSE significantly reduced the venom-induced edema and nociception at all concentrations tested and inhibited migration of inflammatory cells at the three least concentrations (5.0, 7.5 and 10 mg/kg/25 μl PBS). CSE was not effective in inhibiting coagulant, haemorrhagic and indirect haemolytic activities of the venom.The data suggest that CSE could exhibit a central mechanism for pain inhibition, and may also inhibit prostaglandin synthesis. These findings corroborate the traditional administration of C. spicatus decoction to treat inflammatory disorders, including those caused by B. atrox envenomation.

The aim of this study was to evaluate the effect of the Sanguinaria canadensis tincture associated to a chewing gum regarding to dental plaque score (O'Leary) and Streptococcus CUFs. Forty eight students of the University Jose do Rosario Vellano - Brazil took part in this double blind, placebo control study, with ages ranging from 18 to 25 years old, patterned into three groups: placebo group; Sanguinaria canadensis 2.1 mg/day, and Sanguinaria canadensis 4.2 mg/day. Chewing gums were used three times a day for ten days. During the first phase of the study, the chewing gum action was evaluated on dental plaque already installed and in the second phase its action was analyzed on the forming plaque. The results showed statistically significant differences among group I in the first and second phases of the study (p < 0.05 Anova One Way - Tukey Test, t student). The results related to the number of Streptococcus sp. showed statistically sig- nificant differences among groups I, II and III, with 0, 5, 15 and 30 minutes after using the chewing gums (p < 0.01 t Stu- dent test), These values suggest that Sanguinaria canadensis associated to a chewing gum decreased significantly dental plaque scores and number of Streptococcus sp. when compared to placebo chewing gums.

Un echantillon de la boisson ayahuasca contenant les alcaloides harmalol, harmol, harmane et N,N-dimethyltryptamine a ete evalue dans les tests de nage forcee et d’open-field. Dans le test de nage forcee, les doses ont diminue le temps d’immobilite d’une maniere inversement proportionnelle a la dose. Des doses de 2,5 mg/kg et de 10 mg/kg d’ayahuasca ont diminue aussi l’activite locomotrice et l’exploration verticale dans le test d’open-field. La dose de 2,5 mg/kg a amene a une diminution de l’activite locomotrice de 44,95% et de 62,12% pour l’exploration verticale. On peut avancer l’hypothese d’une relation inversement proportionnelle entre dose et effet; la dose de 2,5 mg/kg a montre une activite antidepressive.

We have evaluated the anti-inflammatory and analgesic properties of the leaves (LCE) and stem bark (BCE) crude extracts of Zanthoxylum riedelianum (Rutaceae). Different fractions of the stem bark extract (hexane, BCEH; dichloromethane, BCED; ethyl acetate, BCEE; and lyophilized aqueous residual, BCEW) were also investigated. We studied the effects of the extracts and fractions using the rat paw oedema test induced by carrageenan, dextran, histamine or nystatin; the mouse abdominal constriction test; the mouse hot-plate test (only for LCE and BCE); and the mouse formalin test. Both extracts and all BCE fractions displayed anti-inflammatory activity in the carrageenan-induced oedema model, but not for dextran, histamine or nystatin. Considering the analgesic models, both extracts showed antinociceptive activity, but BCE was more active than LCE in models of central pain. All BCE fractions showed significant inhibition in the abdominal constriction test and in both phases of the formalin test. When BCED was submitted to phytochemical procedures it led to the isolation of six lignans (sesamin, methylpluviatolide, dimethylmatairesinol, piperitol-4′-O-γ,γ-dimethylallyl ether, kaerophyllin and hinokinin), and a triterpene (lupeol). Inhibition of cyclooxygenase and its metabolites may have been involved in the mechanism of action of this plant, considering previous studies reporting the anti-inflammatory and analgesic activity for the identified lignans, as well as anti-inflammatory activity for lupeol.

Austroplenckia populnea (Reiss) Lund is a Brazilian plant from "cerrado", which belongs to Celastraceae family, popularity know as "marmelinho-do campo, mangabeira-brava, mangabarana, vime and maria-mole". This plant is used in folk medicine to treat dysenteries and inflammatory disorders, such as rheumatism. Austroplenckia populnea bark hydroalcoholic crude extract, and its hexane, chloroform and ethyl acetate fractions, obtained by partition, as well as the isolated populnoic acid were investigated for their anti-inflammatory (carrageenan, dextran and histamine-induced rat paw oedema, histamine-induced increase in vascular permeability, and granulomatous tissue induction) and analgesic activities (writhing and hot plate tests). The ED(50) (oral) of the crude extract for the inhibition of carragenan-induced rat paw oedema assay was determined to be 200 mg/kg, which was also used in the assays with the extract and its fractions in all other experiments. The aim of this work was to use in vivo models to evaluate the analgesic and anti-inflammatory activities of ursolic acid (UA) and oleanoic acid (OA), the major compounds isolated as an isomeric mixture from the crude methylene chloride extract of Miconia albicans aerial parts, in an attempt to clarify if these compounds are responsible for the analgesic properties displayed by this plant. Ursolic acid inhibited abdominal constriction in a dose-dependent manner, and the result obtained at a content of 40 mg kg-1 was similar to that produced by administration of acetylsalicylic acid at a content of 100 mg kg-1. Both acids reduced the number of paw licks in the second phase of the formalin test, and both of them displayed a significant anti-inflammatory effect at a content of 40 mg kg-1. It is noteworthy that the administration of the isolated mixture, containing 65% ursolic acid/35% oleanolic acid, did not display significant analgesic and anti-inflammatory activities. On the basis of the obtained results, considering that the mixture of UA and OA was poorly active, it is suggested that other compounds, rather than UA and OA, should be responsible for the evaluated activities in the crude extract, since the crude extract samples displayed good activities.

Extracts of the fruits of Caesalpinia ferrea Mart. (Leguminosae) are widely consumed in folkloric medicine in Brazil and several other countries without any genetic toxicity evaluation. In this study we investigated, the clastogenic and cytotoxic potential of the crude aqueous extract of the fruits of C. ferrea in Wistar rat bone marrow cells using the micronucleus and chromosomal aberration test systems. The animals were treated by gavage with 3 concentrations of the extract, 500, 1000 and 1500 mg/kg, and cyclophosphamide 30 mg/kg. Bone marrow cells were collected 24 h after the treatment. There was no statistically significant difference in the mean of micronucleated polychromatic erythrocytes (MNPCE), mean number of chromosomal aberrations or mitotic index (MI) for the 3 concentrations compared with negative control suggesting that the crude aqueous extract from the fruits of the C. ferrea has no clastogenic and cytotoxic effect in Wistar rat bone marrow cells.

Anti-inflammatory and analgesic activities as well as the median lethal dose (LD50) of water-ethanolic extract (PHE) of the aerial parts of Pothomorphe umbellata were evaluated in animal models. The ED(50) (oral) for the inhibition of carrageenan-induced rat paw edema assay was determined to be 550 mg/kg, while the LD(50) was higher than 2.0 g/kg. At a dose of 550 mg/kg, PHE inhibited the inflammatory process by 48.7% (P0.05) on the third hour of the assay (edema peak) when compared to the untreated control. Indomethacin, the positive control used in this test, inhibited the edema by 58.6% at a dose of 10 mg/kg, when compared to the untreated control (P0.05). All three fractions--hexane, methylene chloride and ethyl acetate--obtained by partition of PHE with respective solvents also showed inhibition of the edema induced by carrageenan over a period of 4h but the methylene chloride fraction showed the best activity. The activity shown by the methylene chloride fraction at 200 mg/kg was comparable to that exhibited by indomethacin at a dose of 10mg/kg. The number of writhings induced by a 0.6% acetic acid solution intraperitoneal injection was decreased by 22% (P0.05) in the group treated orally with Pothomorphe umbellata crude extract. PHE also inhibited the granulomatous tissue formation in rats by 6.2% (P0.05). In the same assay, topically applied dexamethasone decreased the granuloma formation by 14.2%. The above results suggest that Pothomorphe umbellata crude extract has analgesic and anti-inflammatory properties supporting its folkloric use for the treatment of these conditions.

The toxic effects of copaíba oil-resin (ORC) and the copaiba oil-resin vaginal cream (CVC) were evaluated in subacute treatment phase, in wistar rats, which were treated orally (p.o) and intravaginal (ivag). Treated groups received the dose of, 0.04 mg/kg, 28 mg/kg, and 32 mg/kg, respectively, with ORC (p.o), Copaifera duckei oil-resin (ORCV-ivag) and vaginal cream with oil-resin C. duckei (CVC – ivag). The treatment for 22 days with ORC and CVC did not cause clinical signs of toxicity, no deaths have been reported, and did not change the development weight of the animals. The females treated with ORC (p.o), exhibited greater intake of water, but the feed intake was not different in males and females. The use of CVC did not change the water intake of females, but altered feed intake. The ORCV was capable of causing hypoglycemic effect and elevated serum creatinine levels. The hematology parameters of the animals were not changed by ORC (p.o, 32 mg/kg). The use of CVC changed hematocrit, lymphocytes, and the concentration of hemoglobin. The use of CVC and ORCV (ivag), did not altered the biochemicals parameters. Females treated with ORC (p.o, 32 mg/kg) showed some kind of susceptibility to specific use (elevation of total cholesterol, HDL and alkaline phosphatase). The elevation of serum ALT and AST enzymes was not attributed to the use of ORC orally and intravaginal and the use of CVC, as well as the product of its formulation (BVC). The subacute treatment with C. duckei oil-resin and cream (CVC) did not cause clinical signs of toxicity, no deaths have been reported and did not change significantly the parameters evaluated in this study.

Background Plants have been recognized as a good source of insecticidal agents, since they are able to produce their own defensives to insect attack. Moreover, there is a growing concern worldwide to develop pesticides with low impact to environment and non-target organisms. Hexane-soluble fraction from ethanolic crude extract from fruits of Manilkara subsericea and its triterpenes were considered active against a cotton pest (Dysdercus peruvianus). Several natural products with insecticidal activity have poor water solubility, including triterpenes, and nanotechnology has emerged as a good alternative to solve this main problem. On this context, the aim of the present study was to develop an insecticidal nanoemulsion containing apolar fraction from fruits of Manilkara subsericea. Results It was obtained a formulation constituted by 5% of oil (octyldodecyl myristate), 5% of surfactants (sorbitan monooleate/polysorbate 80), 5% of apolar fraction from M. subsericea and 85% of water. Analysis of mean droplet diameter (155.2 ± 3.8 nm) confirmed this formulation as a nanoemulsion. It was able to induce mortality in D. peruvianus. It was observed no effect against acetylcholinesterase or mortality in mice induced by the formulation, suggesting the safety of this nanoemulsion for non-target organisms. Conclusions The present study suggests that the obtained O/A nanoemulsion may be useful to enhance water solubility of poor water soluble natural products with insecticidal activity, including the hexane-soluble fraction from ethanolic crude extract from fruits of Manilkara subsericea.

The vegetal species Pterodon emarginatus Vog. (Leguminosae/Papilonaceae), popularly known in Brazil as ‘sucupira branca’, is widely used by domestic medicine as an anti-inflammatory. From these observations, the hexanic crude extract (HCE) of the fruits was obtained and submitted for assessment of its anti-inflammatory activity. For this purpose, the following tests were used: (1) Determination of ED50 and LD50; (2) Paw edema induced by carrageenin, dextran, histamine and nystatin; (3) Peritonitis caused by carrageenin and (4) Granuloma test. Methylene chloride, ethyl acetate and methanolic extracts from the stem bark of Bauhinia guianensis (Leguminosae, Caesalpinoideae) were obtained. These extracts were evaluated for antiinflammatory activity which was conducted using carrageenin, dextran and histamine-induced paw edema in rats. The extracts of B. guianensis were also assessed for analgesic activity which was conducted using the writhing test in mouse. The different animal groups were treated with these extracts (100 mg/kg i.p. and p.o, IC 50) 30 min prior to the application of stimuli. The methanolic extract demonstrated significant inhibition in the carrageenin-induced edema model. In the dextran-induced edema model, all three extracts inhibited the inflammatory process significantly, with the methanolic extract being the most active. The ethyl acetate extract was the only one shown to be effective in the histamine-induced edema model. Este trabalho descreve os resultados da atividade antiinflamatória de um granulado composto pelos extratos secos de Calendula officinalis L. e Matricaria recutita L. (GECOMR® - granulado encapsulado de Calendula officinalis e Matricaria recutita). Estas espécies são utilizadas na medicina popular devido às suas atividades antiinflamatória, antiúlcera gástrica e antiespasmódica. A redução do processo edematogênico nos grupos de animais tratados com o GECOMR foi observada para todos agentes inflamatórios testados (Carragenina, dextrana e histamina). Esta associação mostrou-se eficaz sobre os modelos de edema de pata induzidos pelas diferentes substâncias inflamatórias, com as doses de 100 e 250 mg.kg-1. A associação dos extratos hidroalcoólicos destas espécies medicinais não modificou a indicação isolada de cada uma. This study shows the results of the anti-inflammatory activity of a phytocomplex composed by dry extracts of Calendula officinalis L. and Matricaria recutita L. (GECOMR®). These plants have been used by folk medicine for its anti-inflammatory, antiulcerogenic and antispasmodic activities. The reduction of the edematogenic process in the treated animals with GECOMR was observed for all inflammatory agents tested, (Carrageenan, Dextran and Hystamine). Its association has been efficient in rat paw edema induced by inflammatory substances in 100 and 250 mg.kg-1 doses. This association of the ethanolic extracts did not modify the indication of each plant isolated.

In rats, administration of Casearia sylvestris extract derived from fresh and dried leaves protects stomach mucosa without changing gastric pH. In stress-induced acute lesions, the preventive effect of extract from dried leaves is more effective than extract from fresh leaves. In acetic acid-induced chronic ulcer, both extracts reduce the size of ulceration and increase the number of collagen fibers after 5 days of treatment, and these effects are similar with both extracts. These antiulcer activities of Casearia sylvestris may be due to the presence of volatile oils, tannins and triterpenerelated compounds.

Many plant crude extracts and their isolated compounds are the most attractive sources of new drugs and show promising results for the treatment of gastric ulcers. Austroplenckia populnea is commonly known as “marmelinho-do campo, mangabeira-brava, mangabarana and vime” and it has been used in folk medicine as anti-dysenteric and anti-rheumatic. Powdered bark wood (3.25 kg) was macerated with aqueous ethanol (96%) and the extract was concentrated under reduced pressure to yield 406 g of crude hydralcoholic extract. The hydralcoholic extract was suspended in aqueous methanol and partitioned with hexane, chloroform and ethyl acetate (EtOAc) in sequence, yielding 8.0 g, 9.5 g and 98.17 g of crude extracts, respectively. Chromatography of the hexane extract over a silica gel column led to the isolation of the triterpene populnoic acid. The oral administration of hydralcoholic, hexane, chloroform and EtOAc extracts (200 mg/kg) decreased the ulcer lesion index (ULI) by 83.15%, 46.87%, 32.2%, 68.12%, respectively. Oral administration of populnoic acid (100 mg/ kg) diminished the ULI by 55.29%. All the obtained results were significant in comparison with the negative control, with exception of the chloroform extract.

To investigate the antiulcer and antiinflammatory activities of the essential oil from Casearia sylvestris leaves (EOCS) the following tests were used: rat paw edema, granulomatous tissue test, vascular permeability, writhing test, gastric ulcer stress-induced and evaluation of gastric secretion (pylorus ligation test). The total yield of EOCS was 2.5% with LD50 of 1100 mg/kg in mouse. The major compounds identified using gas chromatography were caryophyllene, thujopsene, alfa-humulene, beta-acoradiene, germacrene-d, bicyclogermacrene, calamenene, germacrene B, spathulenol and globulol. The EOCS orally administered to the rats at 125 mg/kg resulted 36% of inhibition in carrageenan-induced edema in the rat paw assay (p0.05, Student's t-test). However, both rat paw edema dextran-induced and vascular permeability assay using histamine showed no significant inhibition. Mice submitted to the writhing test using acetic acid presented 58% and 56% of inhibition in writhes with EOCS and indomethacin, respectively. Furthermore, EOCS inhibited 90% of stress-induced gastric ulcer, while cimetidine inhibited 70% (p0.05, Student's t-test). The volume of gastric secretion in the group treated with EOCS was greater than the group treated with cimetidine. The EOCS also showed no changes related to H+ levels in the gastric secretion. With the overall results obtained in this study the authors suggest an anti-inflammatory activity for the EOCS of Casearia sylvestris beyond its anti-ulcer activity.

The toxic effect of the hydroalcoholic crude extract (HACE) from the fruits of the Solanum grandiflorum was evaluated following subchronic treatment (p.o.) in Wistar rats for 65 days using the dose of 2 mg/kg. The LD(50) in mice was 50 mg/kg (p.o.). At the end of treatment, the consumption of water remained within the normal patterns. On the other hand, the consumption of food had 40% reduction in the period between the 18th and the 40th day after the beginning of the study. The diuresis showed a wide variation in volume which started at the 8th day of treatment until the end of the study. This finding was correlated with macroscopic observations of the kidneys which presented nephromegaly (70%) when compared to the control group. Moreover, for the histopathological study, it was observed degeneration and glomerular necrosis (100%), tubular lesions (80%), and hypercellular lesions (100%) in the treated group. The ponderal development of the treated group was increased compared to the control group. The anti-inflammatory and antinociceptive effects of trans-dehydrocrotonin, isolated from the bark of Croton cajucara (Euphorbiaceae), were investigated using several animal models. The trans-dehydrocrotonin produced a significant inhibition of carrageenin-induced paw edema and cotton pellet granuloma in rats. It also inhibited the writhings in mice induced by acetic acid, but did not show a significant effect in the hot-plate test in mice. The LD50 of t-DCTN was 555.0 mg/kg (p.o.) for mice.

The 19-nor-clerodane trans-crotonin (CTN) and the triterpene acetyl aleuritolic acid (AAA), isolated from the stem bark of Croton cajucara Benth (Euphorbiaceae), a traditional medicinal plant from Amazon region of Brazil, as well as the aqueous extract (AE) from its stem bark, were submitted to pharmacological screening for anti-inflammatory and antinociceptive activities in animal models. The oral administration of AAA (50 mg/kg), CTN (50 mg/kg) or AE (300 mg/kg) inhibited the acetic acid-induced writhing in mice. The AE, CTN and AAA had shown significant inhibition of carrageenin-induced edema in rats, in all time intervals measured after the injection of the stimulus, with the greatest inhibition at the first hour for AAA (47.7%) and the second hour for CTN (54.4%). They have also exhibited significant inhibition in the dextran-induced edema 90 minutes after the stimulus: 31.9% for CTN and 28.5% for AAA. In the histamine-induced edema, the inhibition showed by CTN and AAA were 43.2% and 40.5%, respectively, 90 minutes after the injection of stimulus. This study extends and supports the popular medicine and folkloric uses of Croton cajucara in the Amazon region of Brazil. O 19-nor-clerodano trans-crotonina (CTN) e o triterpeno ácido acetil aleuritólico (AAA), isolados das cascas do caule de Croton cajucara Benth (Euphorbiaceae), uma planta tradicional da Região Amazônica do Brasil, bem como o extrato aquoso (EA) das cascas do caule deste Croton, foram submetidos a experimentos farmacológicos utilizando animais, para avaliação das atividades anti-inflamatória e antinociceptiva. A administração oral de AAA (50 mg/kg), CTN (50 mg/kg) ou AE (300 mg/kg) inibiram as contorções em ratos, induzidas por ácido acético. Os terpenóides AAA e CTN, bem como o extrato polar EA, inibiram significativamente o edema de pata em ratos, induzido por carragenina. As inibições foram observadas em todos os intervalos de medições, tendo sido evidenciado melhores inibições para os terpenóides AAA (47,7%, após a primeira hora) e CTN (54,4%, após a segunda hora). Evidenciou-se ainda, após 90 minutos do estímulo, significante inibição no edema induzido por dextrana (31,9% para CTN e 28,5% para AAA) e por histamina (43,2% para CTN e 40,5% para AAA). Estes resultados confirmam o uso popular de Croton cajucara na região Amazônica do Brasil, no combate a inflamações.

Diabetes mellitus is a syndrome that reaches more than 382 million people worldwide. It interferes with the metabolism of carbohydrates, causing chronic hyperglycemia. The objective of this study was to evaluate the effect of the Copaifera duckei, Dwyer, Fabaceae, oleoresin on streptozotocin-induced (STZ) diabetic rats. This study was based on the induction of diabetes mellitus by streptozotocin (55 mg/kg, i.p.) in Wistar rats and treated with doses of C. duckei oleoresin (250 and 500 mg/kg, p.o.). Subsequently, the clinical, biochemical and histopathological of the pancreas parameters were evaluated. Copaiba (Copaifera duckei Dwyer, Fabaceae) oleoresin is an important Amazonian raw material. Despite its insecticidal potential, poor water solubility remains a challenge for the development of effective and viable products. Nanotechnology has emerged as a promising area to solve this problem, especially oil-in-water nanoemulsions. On this context, the aim of the present study was to develop oil-in-water nanoemulsions using copaiba oleoresin dispersed through a high internal phase; and evaluate its potential insecticidal action against Aedes aegypti larvae. Overall, 31 formulations were prepared, ranging from 11.5 ± 0.2 to 257.3 ± 4.1 nm after one day of manipulation. Some of them reached small mean droplet sizes (< 200 nm) and allowed achievement of a nanoemulsion region. The formulation consisted of 5% (w/w) of copaiba oil, 5% (w/w) of surfactant and 90% (w/w) of water, which presented mean droplet size of 145.2 ±0.9 nm and polidispersity of 0.378 ± 0.009 after one day of manipulation, and these were evaluated for larvicidal potential. According to mortality level (250 ppm - 93.3 after 48 h), this nanoemulsion was classified as a promising insecticidal agent against Aedes aegypti larvae. The present study allowed the development of low-cost ecofriendly green natural-based nanoformulations with potential larvicidal activity, using a nanobiotechnology approach. Background The increasing efforts to reduce the environmental impact on the Amazon’s natural resources are focusing on watercourses that pass through effluents with high concentrations of heavy metals. The adsorption by absorbent is one of the methods used to remove metallic ions. In this assignment, the preparation of activated carbon from Brazil nut bark (Bertholletia excelsa l.), which is a waste material produced from the use of seeds in foodstuffs and cosmetics, is shown. Results The absorbent was carbonized at 400 °C in 3 h and activated at 800 °C in 2 h, having received the name of AC2, and, the specific area, pore size, real and apparent densities, porosity, scanning electron microscopy (SEM) coupled to energy-dispersive X-ray spectroscopy (EDS), X-ray diffraction (XRD), pH, moisture, fixed carbon and surface functional groups by Boehm method and Fourier transformed infrared spectroscopy (FTIR) were characterized. According to the results, the carbon presented alkaline characteristic, mesoporosity, average pore diameters of 2.203 nm and specific surface area by BET of 464.835 m2 g-1. The efficiency of removal was performed in synthetic solutions of copper sulphate (II) pentahydrate (CuSO4.5H2O), evaluating the influence of pH, initial concentration of copper solution (II), particle diameter and time contact of the adsorbent in solution. As atividades antiinflamatória e antinociceptiva do extrato padronizado de Hypericum brasiliense (HBSE) (Guttiferae) foi avaliada em modelos animais. Ratos Wistar machos foram tratados com extrato de H. brasiliense (50, 250 e 500 mg/kg, v.o.) em solução 3% Tween 80 0,9% NaCl. O tratamento com HBSE (500 mg/kg) mostrou inibição significativa sobre o edema induzido por carragenina comparado ao grupo controle. Nessa dose, o edema foi reduzido em 31,25% na terceira hora (pico do edema) após o tratamento, mas na dose de 50 mg/kg, o edema apresentou redução de 53,13% (p < 0,05). Ainda com a dose de 50 mg/kg, a diminuição do edema induzido por dextrana foi similar ao controle positivo, ciproeptadina. Houve diminuição na formação do tecido granulomatoso (6,6%) comparável ao grupo controle. O HBSE também inibiu o número de contorções abdominais em 46,4%, estatisticamente igual ao controle positivo, tratado com indometacina (42,9%). Na dose de 250 mg/kg, houve inibição do número de contorções em 70,7% quando comparado ao grupo controle (p < 0,001). No teste da placa-quente, foi verificado aumento no tempo de latência com a dose de 50 mg/kg. Os resultados demonstram que o HBSE possui atividade antiinflamatória sobre processos agudos, principalmente quando sua gênese está relacionada à síntese dos derivados do ácido araquidônico, e seu efeito analgésico provavelmente envolve ação sobre o Sistema Nervoso Central. The anti-inflammatory and antinociceptive activities of the standardized leaves extract (HBSE) of Hypericum brasiliense (Guttiferae) were evaluated in animal models. Male Wistar rats were treated with H. brasiliense extract (50, 250 and 500 mg/kg, p.o.) in 3% Tween 80 0.9% saline solution. The treatment of the edema induced by carrageenin with HBSE (500 mg/kg) showed significant inhibition when compared to the control group. At this dose, the edema decreased by 31.25% in the third hour after treatment (edema peak), but the dose of 50 mg/kg has inhibited the edema by 53.13% (p < 0.05). At the dose of 50 mg/kg, the decrease of the edema induced by dextran was similar to that caused by cyproheptadine. The decrease of the formation of granulomatous tissue (6.6%) was comparable to the control group. The HBSE inhibited the abdominal constrictions induced by acetic acid. At a dose of 50 mg/kg, the inhibition of the abdominal constrictions (46.4%) was comparable to that produced by indomethacin (42.9%). A dose of 250 mg/kg inhibited these constrictions by 70.66% when compared to control (p < 0.001). In the hot-plate test, an increase in the latency time was observed at a 50 mg/kg dose. These data suggest that HBSE has anti-inflammatory activity on acute process, developed principally by arachdonic acid derivates and analgesic effect due to its probable involvement in the Central Nervous System.

Due to its therapeutic action, the phytochemical study of the essential oil of Aniba canelilla H.B.K Mez. (Lauraceae) and its toxicological preclinical trial were evaluated. The essential oil extracted from the bark (OEAC) yielded 1.0% and the major constituents were 1-nitro-2-phenylethane (52.9%) and methyl eugenol (38.7%), both identified by GC/MS and GC-FID. The observed LD50 (oral) was 720 ± 66.4 μg/kg.
