Although studies have suggested organochlorine pesticides (OCPs) exposure increased the risk of epithelial ovarian cancer, the mechanisms underlying its potential tumorigenic effects in the human ovary are not well understood. In this study, we investigated the impact of dichlorodiphenyldichloroethylene (DDE), endosulfan, and heptachlor exposure on epithelial cadherin (E-cadherin) and proinflammatory mediators in human ovary surface epithelial (HOSE) cells. We found that DDE, endosulfan, and heptachlor exposure resulted in epithelial differentiation accompanied by upregulation of E-cadherin expression and overexpression of proinflammatory cytokines (TNFα, IL-1β, and IL-6) in HOSE cells. The epithelial differentiation may accelerate HOSE cells to inclusion body formation, a common site for ovarian cancer initiation and persistent exposure to OCPs creates a chronic inflammatory microenvironment that may promote the neoplastic transformation of HOSE cells within the inclusion cyst.

The diamondback moth (DBM) Plutella xylostella (L.) (Lepidoptera: Plutellidae) is an insect pest found around the world that feeds on cruciferous crops. The DBM has become resistant to most insecticides in current use in the field. Broflanilide is a novel meta-diamide insecticide that binds to a new site on the γ-aminobutyric acid receptor and very efficiently protects against most pests in the order Lepidoptera, including DBM. In this study, the resistance of a laboratory-bred susceptible strain of DBM to broflanilide and the fitness costs posed by broflanilide to the DBM were evaluated. The DBM had no obvious resistance to broflanilide after 10 generations of selection. The realized heritability h2 was 0.033, suggesting a low risk of resistance developing in this strain. The F10 generation had no cross-resistance to the insecticides abamectin and endosulfan (which target the γ-aminobutyric acid receptor) and chlorantraniliprole (which targets a non-γ-aminobutyric acid receptor). The specific activities of important detoxification enzymes (cytochrome P450 monooxygenase, esterase, and glutathione S-transferase) were not obviously altered. However, the larval stage was prolonged and the adult stage was shortened significantly in F11 generation than the F0 generation. The total preoviposition period TPOP significantly prolonged 1.90 d in F11 generation. The fitness value Rf (0.93) was lower for the F11 generation than the F0 generation. The results indicated that long-term exposure to broflanilide exerts clear fitness costs in the DBM. This information will be useful in identifying reasonable broflanilide application guidelines for managing broflanilide resistance in the DBM.

Introduction. Farmers use a large number of pesticides to increase the productivity of Khat cultivation. Since Khat is consumed by many Ethiopians as a habit, there may be a significant concentration of pesticide residues in Khat leaves which result in adverse effects among the exposed population. Excess use of pesticides in Khat causes cancer, genotoxicity, and endocrine disturbances. The current study aimed to determine the concentration of selected pesticide residues on Khat leaves samples and to estimate the possible health risk on consumers. Methods. Thirty Khat samples were collected from Khat cultivators in the Amhara region, Ethiopia. Pesticide residue extraction and cleanup of Khat samples were performed using the QuEChERS method. Data were also collected about personal variables and Khat consumption habits. The pesticide concentrations were determined and the health risks were estimated. Results. Profenofos, dimethoate, and chlorpyrifos were detected in 6 (20%), 24 (80%), and 28 (93.3%) Khat samples, respectively. However, diazinon was detected in two samples and endosulfan I was not detected. The average concentration of chlorpyrifos, dimethoate, profenofos, and diazinon in Khat samples was 75.39 ± 20.26, 130 ± 48.94, 242.45 ± 119.79, and 81.5 ± 21.0 μg/kg, respectively. The HQ for diazinon was greater than 1 which shows the chronic health risks associated with intake of diazinon among Khat chewers. The acute health risks associated with intake of profenofos and diazinon were minimal. Discussion. Pesticide residue exposure is a risk factor for cardiac complications, reduction in antioxidant defense system, and developing neurological symptoms. All the pesticides were higher than the European Union Maximum Residue Levels (EU-MRL). Diazinon has the potential to cause higher chronic health risks. Chronic Khat chewers in the region may face potential health risks from pesticide residues. Awareness creation among people in the study area is necessary to avoid the adverse health effects of pesticide residues.

Endocrine disruptors are a major concern due to their possible association with hormone-dependent carcinogenesis. Some examples of compounds with such properties are organochlorine pesticides (OCPs). OCPs are persistent pollutants with high lipophilicity, long half-life, and bioaccumulation potential. In the past, some of the most commonly used OCPs were dichlorodiphenyltrichloroethane (DDT) and endosulfan. Here, we investigated the effects of o,p'-DDT, p,p'-DDT, and endosulfan and of hormones estradiol, testosterone, and progesterone on the expression of estrogen, progesterone, and androgen receptors (ER, PR, and AR) and of their target genes (KLF4, VEGFA, CCND1, PRLR, CDKN1A, and BCL6) in MCF-7 and MDA-MB-231 cells. The results confirmed that under the action of the insecticides, there are dose- and time-dependent changes in the expression of these receptors and target genes. As corroborated by an experiment with ER, PR, and AR negative MDA-MB-231 cells, the change in the expression of KLF4, VEGFA, CCND1, and PRLR in MCF-7 cells treated with o,p'-DDT and the change in CDKN1A and PRLR expression in MCF-7 cells treated with p,p'-DDT are likely mediated by ER, PR, and AR pathways. In conclusion, we have identified some targets of DDT and endosulfan and confirmed that the effects of insecticides on the expression of these target genes differ for breast cancer cell lines with different receptor statuses.

Organochlorine pesticides (OCPs) have been used globally to boost food production. Although banned, due to their prolonged toxic effects but their residue still impacts the quality of primary and processed agricultural products. This study assesses the levels of residual OCPs (α,β,δ-HCH, heptachlor, heptachlor epoxide, chlordane, methoxychlor, aldrin, dieldrin, endrin, endrin aldehyde, endrin ketone, endosulfan, endosulfan sulfate, 1,1-dichloro-2,2-bis(p-chlorophenyl)ethane (DDD), 1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene (DDE), and 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (DDT)) in food commonly consumed in Lagos and Ibadan, Southwest, Nigeria. Health risk associated with human exposure via food intake was evaluated with a statistical predictive model. About 248 composite food samples from 8 categories were analyzed in Lagos and Ibadan. Sample extraction and clean-up were by QueChERS method and extracts injected into GC-μECD. ƩDDT concentration was highest in meat products, aquatic foods, dairy products, edible oils, fruits, and cereals, while ƩHCHs were highest in chicken eggs and vegetables. ƩOCP concentrations (ng/g) in food categories were 6.09±1.6–6.85±0.9 (meat), 5.29±2.0–12.3±14 (aquatic foods), 4.86±1.7–5.89±0.8 (dairy products), 4.53±0.8–6.32±1.1 (edible oils), 3.32±1.3 (eggs), 3.54±1.0–4.80±1.5 (fruits), 4.16±2.7–4.40±0.8 (vegetables), and 6.12±2.0–6.62±0.9 (cereals). The estimated average daily intake of OCPs was 5.91, 12.5, 4.41, 6.40, 1.53, 5.14, 3.95, and 16.7 ng/kg bw/day through the consumption of meat products, aquatic foods, dairy products, edible oils, chicken eggs, fruits, vegetables, and cereals, respectively. BackgroundVegetables and rice produced in Cuba may contain residue of pesticides that exceed the maximum residue limits (MRLs). Pesticide residues on crop samples from Sancti Spíritus province were analyzed. Based on these residue data, a risk assessment of consumer exposure was conducted.MethodsSamples collected during the 2017, 2018, and 2019 periods were analyzed using UPLC-MS/MS and GC/ECD chromatography techniques. A deterministic risk assessment was performed, using consumption data collected by the authors. Also, a probabilistic risk assessment was worked out with @RISK for the Monte Carlo simulation.ResultsA broad range of pesticides were detected in the samples. Most of the detected active ingredients were insecticides and fungicides. A considerable part of the detected pesticide residues exceeded the EU and Cuban MRLs. However, the deterministic risk assessment revealed that the consumer risk of pesticide exposure is low. Only in the maximum exposure scenario children and adults were exposed to methamidophos residues in tomatoes higher than the acceptable daily intake.ConclusionsWhen a more realistic consumption (50th percentile) and higher (95th percentile) consumption pattern for tomatoes was assumed, the deterministic risk assessment found no chronic risk for children and adults being exposed to methamidophos present in the tomato samples. Furthermore, the probabilistic risk assessment concerning methamidophos exposure in tomato for children and adult strata showed no risk. To perform in the future a more accurate risk assessment for Cuban consumers it is important to have more detailed consumption data and an expanded dataset of the residues in different crops.

Background: Pesticides are applied on cowpeas to reduce the losses incurred to the attack by Callosobruchus maculatus which adversely affect its production and storage. Objectives: This study aims to evaluate the mean concentration of heptachlor epoxide, aldrin and endosulfan in randomly collected cowpeas obtained from local markets in South-West states of Nigeria and compare the mean concentration with the maximum residual limit (MRL) set by the regulatory body. Method: Seventy-six samples of cowpeas were collected from from local markets within the study area, pulverised, extracted with dichloromethane, cleaned-up and analysed by Gas Chromatography-Mass Spectroscopy (GC-MS) for aldrin, endosulfan and heptachlor epoxide. Results: The experiment showed that aldrin, endosulfan and heptachlor epoxide are widely used in South-West for preservation of cowpea. Mean concentration of aldrin (406.8±0.12 μg/kg) in Bodija market, Oyo state, was observed to be the highest. The market in Ile Epo market, Lagos state, was observed to have the highest mean concentration for endosulfan (315±0.164 μg/kg) while Okitipupa market, Ogun state, recorded a mean concentration for heptachlor epoxide (156±6.23 μg/kg), which is the highest. Conclusion: This shows that aldrin, endosulfan and heptachlor epoxide are used in the storage of cowpeas long after they were labelled probable carcinogens and their mean concentrations were above the MRL set by USEPA. Thus there is need for regular monitoring to be conducted on cowpeas for organochlorines.

The surface soil samples (0-15 cm) were found contaminated with different pesticides in vegetable growing areas in Bihar. Total twenty samples were analysed in which HCH and endosulfan were found as the major contaminants of soil. Nine samples were found contaminated with HCH (ND-0.055mg kg-1) and ten with endosulfan (ND-0.078mg kg-1). DDT residues were detected only in three soil samples. Among the organophosphate pesticide, residues of quinalphos (ND-0.062mg kg-1) and chlorpyriphos (ND-0.037mg kg-1) were detected in five samples each while dimethoate present in only one sample. Residues of cypermethrin, fenvalerate and deltamethrin were also detected in four, three and one soil sample, respectively.

Organochlorine pesticides (OCPs) residues were determined in a total of 160 samples comprising of cocoa beans and soil collected from four major cocoa farms in Ondo, Nigeria. The residues were extracted with dichloromethane, cleaned up by column chromatography and analysed with Gas Chromatography-Electron Capture Detector (GC-ECD). In the analysed cocoa samples, endrin and endosulfan I had the highest incidence of occurrence of 93% and 89%, respectively while lindane and heptachlor had the least occurrence of 19%. High incidences of occurrence in the soil samples were observed in endrin (93%) and endosulfan I (94%) while the least occurrence was α-HCH (13%). The total residual concentrations of the OCPs were higher in the cocoa samples compared to the soil samples. The mean residual levels of HCHs (α, β, δ) aldrin, dieldrin, endrin and endosulfan I and cis-permethrin in the cocoa samples were above Maximum Residual Limits (MRL) established by WHO/FAO while aldrin, dieldrin, endrin, endosulfans in the soil sample analysed were above MRL established by Netherlands. Bioaccumulation factor > 1 was recorded in most of the cocoa samples from the different farms especially for endrin, dieldrin, endosulfan I and trans-permethrin. This gives cause for concern considering the adverse health hazards pose by these pesticides.

In this study, the cytotoxic potential of fluoride and endosulfan in combination was investigated in Swiss albino mice bone marrow cells using the chromosomal aberration (CA) and micronucleus (MN) test systems. Fluoride (25.1 mg kg−1 body weight [bw] in water) and endosulfan (1.8 mg kg−1 bw by oral intubation) were administered orally alone and in combination (fluoride 25.1 mg kg−1 bw + endosulfan 1.8 mg kg−1 bw) to male Swiss albino mice daily for 30 days. A significant ( p < 0.01) increase in micronuclei (MNs) induction and decreased ratio ( p < 0.01) of polychromatic to normonochromatic erythrocytes (indicators of cytotoxicity) were observed compared with saline controls when animals were given the combination of fluoride and endosulfan. A significant ( p < 0.01) increase in MNs induction and no change in the polychromatic erythrocytes to erythrocyte ratio were also observed when endosulfan was given alone. CAs such as gaps, breaks, fragments, rings, exchanges, and polyploidy were recorded in the bone marrow cells. The mean percent frequency of CAs was increased ( p < 0.01) in all the treated groups compared with the control saline group. In the combination group (F + E), the percent frequencies of CAs were significantly higher (13.875%) compared with those in the individual treatment groups of fluoride (4.375%) and endosulfan (6.25%). The mitotic index was calculated as percentage of dividing cells. A significant ( p < 0.01) decrease in mitotic index was observed in all treated groups compared with controls. In the combination group (F + E), mitotic index was significantly less than ( p < 0.01; 4.1 ± 0.49) the saline control (10.8 ± 0.98). These results indicated that repeated intake of endosulfan through various sources in fluoride affected areas resulted in increased cytotoxic effects. The greater effect in the combination group indicated additive interaction of fluoride and endosulfan in inducing cytotoxicity in Swiss albino mice.

A survey was undertaken in five Village Development Committees (VDCs) of Lamjung District, Nepal from June to August 2018 to investigate major insect pests and their management practices in cucurbits. A total of ninety-five cucurbit growers were selected and interviewed using semi-structured questionnaires. The study revealed that the major insect pests attacking cucurbits were fruit fly, red pumpkin beetle, aphid, whitefly, epilachna beetle, cucurbit sting bug, cutworm, and blister beetle. The majority of farmers responded that fruit fly was the most prevalent insect pest, followed by aphid and red pumpkin beetle. Most of the farmers used chemical methods, that includes biological, mechanical, and cultural techniques to control the insects. For the mechanical method, they used sex-pheromone traps i.e. cue-lure. Among botanical pesticides, Neem (Azadirachta indica), Malabar Nut (Justicia adhatoda), Chinaberry (Melia azedarch), Mugwort (Artemisia spp.) were used. Commonly used insecticides by farmers were Cypermethrin, Dimethoate, Malathion, and Endosulfan. The indiscriminate use of chemical pesticides resulted in pest resistance, resurgence, and sometimes outbreak of insect pests. Majority of farmers were using chemical methods to control pests. Apart from this, Integrated Pest Management (IPM) was also adopted for good agricultural practices (GAP) to prevent chemical hazards on human health and the environment. To control insect pests, trained farmers should be encouraged to follow the sanitation of fields and protection of natural enemies by avoiding the use of pesticides a long as possible.

Endosulfan, a typical organochlorine pesticide, is widely used in agricultural countries and was detected in blood samples from the general population. Studies have shown a positive correlation between chronic kidney disease of unknown aetiology (CKDu) and endosulfan. CKDu has become endemic in agricultural countries, with clinical manifestations of tubulointerstitial fibrosis.The goal of this study was to investigate the effects of endosulfan in kidney cell injury in human renal tubular epithelial cells (HK-2), focusing on apoptosis, inflammatory response, and epithelial-mesenchymal transition (EMT). We found that endosulfan induced apoptosis in HK-2 cells by up-regulating the expression of BAX, APAF-1, Caspase-3 and mitochondrial Cytochrome c was released into the cytosol. Endosulfan caused an inflammatory response, showing the increase in the secretion and mRNA expression levels of IL-6/IL-8. Endosulfan triggered EMT, characterized by downregulation of E-cadherin and upregulation of Vimentin. Western blot results showed that p-Smad3 and Smad3 protein expression were elevated while the expression of Smad7 were decreased in endosulfan-exposed groups. Dual luciferase reporter assay confirmed the potential binding capacity of miR-429 to 3'-UTR of ACE2. Endosulfan causes upregulation of miR-429 and downregulation of ACE2 in HK-2 cells. Overexpression of miR-429 or silencing of ACE2 in HK-2 cells caused apoptosis, inflammation and EMT through TGF signaling pathway. These findings suggest that endosulfan can lead to kidney cell injury by modulating ACE2 through up-regulating miR-429, providing new evidence for the pathogenesis of CKDu.

Pesticides such as endosulfan, heptachlor and dieldrin persist in aquatic environments as a result of their resistance to biodegradation. However, there is no adequate information about the toxicity of endosulfan, heptachlor and dieldrin to the aquatic organism, African catfish (Clarias gariepinus)—a high valued widely distributed commercially interesting species. The current experiment was performed with the aim to determine the median lethal concentration (LC50) of endosulfan, heptachlor and dieldrin to African catfish (Clarias gariepinus); their behavioral abnormalities and histopathological alterations in several vital organs. A total of 324 juvenile fish were exposed for 96 h to six concentrations of endosulfan and dieldrin at 0, 0.001, 0.002, 0.004, 0.008 and 0.016 ppm, and to heptachlor at concentrations of 0, 0.02, 0.04, 0.08, 0.16 and 0.32 ppm for dose-response tests. The study demonstrated that the species is highly susceptible to those contaminants showing a number of behavioral abnormalities and histopathological changes in gill, liver and muscle. The 96-h LC50 value of endosulfan, dieldrin and heptachlor for the African catfish was found as 0.004 (0.001−0.01) mg/L, 0.006 mg/L and 0.056 (0.006−0.144) mg/L, respectively. Abnormal behaviors such as erratic jerky swimming, frequent surfacing movement with gulping of air, secretion of mucus on the body and gills were observed in response to the increasing exposure concentrations. Histopathological alterations of liver, gill and muscle tissues were demonstrated as vacuolization in hepatocytes, congestion of red blood cells (RBCs) in hepatic portal vein; deformed secondary lamellae and disintegrated myotomes with disintegrated epidermis, respectively. These findings are important to monitor and responsibly manage pesticide use in and around C. gariepinus aquacultural areas.

Endosulfan was widely used as an insecticide in the agricultural sector before its environmental persistence was fully understood. Although its fate and transport in the environment have been studied, the effects of historic endosulfan residues in soil and its bioaccumulation in crops are not well understood. This knowledge gap was addressed by investigating the dissipation and bioaccumulation of endosulfan in ginseng as a perennial crop in fresh and aged endosulfan-contaminated fields. In addition, the effect of granular biochar (GBC) treatment on the bioaccumulation factor (BAF) of endosulfan residue in ginseng was assessed. The 50% dissipation time (DT50) of the total endosulfan was over 770 days in both the fresh and aged soils under mulching conditions. This was at least twofold greater than the reported (6– > 200 days) in arable soil. Among the endosulfan congeners, the main contributor to the soil residue was endosulfan sulfate, as observed from 150 days after treatment. The BAF for the 2-year-old ginseng was similar in the fresh (1.682–2.055) and aged (1.372–2.570) soils, whereas the BAF for the 3-year-old ginseng in the aged soil (1.087–1.137) was lower than that in the fresh soil (1.771–2.387). The treatment with 0.3 wt% GBC extended the DT50 of endosulfan in soil; however, this could successfully suppress endosulfan uptake, and reduced the BAFs by 66.5–67.7% in the freshly contaminated soil and 32.3–41.4% in the aged soil. Thus, this adsorbent treatment could be an effective, financially viable, and sustainable option to protect human health by reducing plant uptake of endosulfan from contaminated soils.

In Northern Benin, insecticides are used for cotton production. These insecticides can be easily transferred to water ponds close to cotton fields. To monitor insecticides levels in water, sediments and fish samples from water ponds, a GC-MS analytical method was developed to detect residues of endosulfan, DDT and its parent compounds, isomers of HCH, pyrethroids and chlorpyrifos. In addition, the influence of storage conditions of water sample on pesticides determination performance has been studied. The limits of quantification were between 0.16 and 0.32 µg/L in water, 0.5 and 1 μg/kg in sediment and 1 and 2 μg/kg in fish. Twenty samples of water, twenty of sediments and forty of fish were taken in four different water reservoirs at five different times. Alpha-endosulfan, lambda-cyhalothrin and permethrin were the identified in sediment while p,p’-DDE, α- and β-HCH, chlorpyrifos, lambda-cyhalothrin and permethrin were detected in fish. Only organochlorines were determined in water because of the lack of recovery of pyrethroids from water stored in glass. Concentrations of insecticide residues in sediment for all water ponds ranged from non-detected to 101 µg/kg and from non-detected to 36 µg/kg in fish. Preliminary risk assessment for consumers of the North of Benin showed that the Estimated Daily Intakes were lower than the Acceptable Daily Intake sand Acute Reference Doses for all consumers. However, as one fish can be contaminated by 5 pesticide residues at the same time, it is not possible to exclude a risk for the consumer due to his exposure to mixtures of pesticides.

The nationwide occurrence of endosulfan residues in cotton fields has not yet been investigated. Therefore, in this study, 202 surface soil samples from 27 cities were collected from cotton fields in 8 major cotton-planting provinces of China, covering more than 97% of the national cotton sown area. The results showed that endosulfan residues were detected in cotton fields throughout the country. The main type of residue found was endosulfan sulfate (ES-sulfate), followed by β-endosulfan and α-endosulfan, with average concentrations of 0.475, 0.129, and 0.048 μg/kg, respectively. Significant spatial variations in the endosulfan residues was noted, and the highest concentration of endosulfan residues was observed in the northwest inland cotton-growing area, followed by that in the Yellow River basin and Yangtze River basin cotton-growing areas. The endosulfan residues showed significant positive correlations with soil organic matter and soil clay contents. The α/β endosulfan ratio was determined to be in the range of 0.02-1.20, indicating that endosulfan residues originated from the endosulfan application performed in historical cotton cultivation efforts. Together with the literature data, the concentrations of α-endosulfan and β-endosulfan residues peaked in 2015 and 2017, respectively, and showed an overall decreasing trend from 2002 to 2021. The results of the ecological risk assessment suggested that Folsomia candida was most sensitive to endosulfan residues, with 20.8% of the sites presenting a high risk. However, in general, the soil ecological risk of cotton fields across the country was low. Our study demonstrated that China has achieved promising results in controlling the use and pollution of endosulfan, especially after 2014.

Tea is the most frequently consumed drink in the sphere; next to the water. However, tea can be contaminated by different pesticides particularly those outdated pesticides which have been familiar as one of the main difficulties in Ethiopia and contaminate the food commodities. From a study done in south west of Ethiopia, Dichlorodiphenyltrichloroethane (DDT) and endosulfan were detected in commonly consumed food items. Therefore, the main aim of this study was to analyze the residue of organochlorine pesticides (OCPs) in tea collected from the market and assessing their risks to consumers. 19 different tea samples were randomly collected from supermarkets in Jimma town and Addis Ababa city. In the analytical procedure, QuEChERS (Quick, Easy, Cheap, Rugged, and Safe) method was applied for the extraction of tea samples. The finding of the study revealed that, five OCPs (aldrin, γ-chlordane, DDT, endrin, and dibutylchlorepoxide) were detected at a concentration of 0.1465, 0.167, 0.2044, 0.3067 and 0.4089 mg kg−1 in domestic tea, respectively, while endosulfan sulfate, methoxychlor, and heptachlor epoxide were detected at a concentration of 0.258, 0.458 and 1.427 mg kg-1 in imported tea samples respectively. From the pesticides detected, the mean concentration of DDT (0.292–0.825 mg kg−1) is above the Maximum Residue Limits (MRLs) established by China (0.2 mg kg−1). The estimated daily intake (EDI) of γ-chlordane and endrin were above the acceptable daily intakes (ADIs), this indicates that there is a health risk for tea consumers. The detection of pesticide residues in tea reveals that there is the widespread use of OCPs in the study area for agricultural purposes or as vector control. Seasonal levels of organochlorine pesticides (OCPs) in yam, cassava, sweet potato and cocoyam samples collected from Esa-Oke Farm Settlement, Osun State, Nigeria were evaluated. Sampling regiment for three sites lasted four months each during the dry and wet seasons. The dried samples were extracted using Soxhlet extractor with dichloromethane (DCM) as the extraction solvent, while the identification and estimation of OCPs in the crop filtrates, after clean-up, were carried out with Gas Chromatography equipped with Time-of-Flight Mass Spectroscopy Detector (GC-TOFMS). Ten OCPs determined in the crops had the overall seasonal mean levels that ranged from 158 ng/g (dieldrin) to 544 ng/g (heptachlor) and the seasonal mean burden per OCP occurred in the order: heptachlor epoxide (518) > heptachlor (447) > p,p’-DDE (431) > dieldrin (349) > chlordane (327) > aldrin (321) > p,p’-DDD (313) > methoxychlor (303) > endosulfan I (287) > p,p’-DDT (284) for wet season while this order was not the same for dry season of the same crop with heptachlor epoxide (415) > p,p’-DDE (373) > aldrin (305) > heptachlor (307) > methoxychlor (288) > chlordane (274) > p,p’-DDT (263) > p,p’-DDD (263) > endosulfan I (260) > dieldrin (246). For all of the OCPs, the estimated daily intake (EDI) and health risk indices (HRI) were significantly higher than the recommended values. Thus, a regular large-scale consumption of crop products from the farm settlement could result in grave public health concern in the course of time., Food Crops, Organochlorine Pesticides, Health Risks, Farm settlement, Esa-Oke.

Amaç: Bu çalışma; organoklorlu (Endosulfan) ve pyrethroid (Cypermethrin) pestisitler ile herbisitlerin (2,4-D ve Trifluralin) Hershberger metodu ile peri-pubertal erkek sıçan modelinde serum LH ve FSH düzeyleri ile androjen duyarlı dokular üzerine androjenik ve anti-androjenik etkilerini araştırmak amacıyla planlandı.Gereç ve Yöntem: Çalışmada 56 adet peri-pubertal 42 günlük erkek Sprague Dawley sıçan kullanıldı.İlk grup sham kastrasyon olarak ayrıldı. Geriye kalan 49 adet sıçan kastre edildi. On günlük iyileşme süresinin sonunda sıçanlar yedi gruba daha ayrıldı: Kastrasyon grubu; Testosteron propiyonat (TP) grubu; TP+Flutamide grubu; TP+Endosulfan grubu; TP+2,4-D grubu; TP+Cypermethrin grubu; TP+Trifluralin grubu. Son uygulamadan 24 saat sonra sıçanlar dekapite edilerek kan örnekleri toplandı. Tüm hayvanların prostat bezi, bulbouretral bezleri, seminal vezikülleri ve musculus levator ani (mLA) diseke edilerek tartıldı. Serum LH, FSH ve testosteron düzeyleri ELISA yöntemi ile belirlendi.Bulgular: Kastrasyon, tüm androjen-duyarlı doku ağırlıklarında azalmaya neden olurken (p0.05). 2,4-D, Cypermethrin ve Trifluralin serum FSH düzeylerinde anlamlı artışa yol açarken; serum LH düzeylerindeki artış sadece Trifluralin grubunda istatistiksel olarak anlamlı idi. Sonuç: Çalışmadan elde edilen bulgular, Hershberger metodunda Endosulfan, Cypermethrin ve Trifluralin’in bulbouretral bez üzerinde anti-androjenik; 2,4-D’nin ise vesicula seminalis üzerinde androjenik etkilere sahip olduğunu göstermiştir. Bu çevresel kirleticilerin erkek üreme fonksiyonlarını olumsuz yönde etkileyebileceği öngörülebilir., Aim: This study was aimed to investigate the androgenic and anti-androgenic effects of organochlorid (Endosulfan) and pyrethroid (Cypermethrin) pesticides and herbicides (2,4-D and Trifluralin), on serum LH and FSH levels and androgen-sensitive tissues in peri-pubertal rat model with Hershberger assay. Material and Method: 56 peri-pubertal 42 days old Sprague Dawley rats were used in the study. The first group was separated as sham castration. The remaining 49 rats were castrated. After 10 days recovery period, rats were divided into further seven groups: Castration group; Testosterone Propionate (TP) group; TP+Flutamide group; TP+Endosulfan group; TP+2,4-D group; TP+Cypermethrin group; TP+Trifluralin group. 24 hours after the last application, rats were decapitated and blood samples were obtained. Prostate, bulbourethral gland, vesicula seminalis and musculus levator ani (mLA) were dissected and weighed. Serum LH, FSH and testosteron levels were determined with ELISA. Result: Castration caused significant reduction in all androgen-sensitive tissue weights n(p0.05). 2,4-D, Cypermethrin and Trifluralin significantly increased serum FSH levels; but the increase in serum LH levels was only significant in Trifluralin group. Conclusion: Data obtained from this study showed that 2,4-D has androjenic effects on vesicula seminalis whereas Endosulfan, Cypermethrin and Trifluralin have anti-androgenic effects on bulbourethral gland. It can be suggested that, these environmental pollutants could affect male reproductive functions negatively.

A study to quantify the toxicity and ecotoxicological pressure of pesticides in the Sancti Spíritus province, Cuba, was carried out between 2011 and 2014. A longitudinal descriptive work was designed for the study period to identify potential risks to the environment and also to human health associated with the use of pesticides in the country. The Spread Equivalents (ƩSeq) and Pesticide Occupational and Environmental Risk (POCER) indicators, as well as the Toxic Load (TL) methodology of Instituto Cubano de Sanidad Vegetal, were used to determine the toxicity and ecotoxicity of pesticide use. One hundred and twenty- four active ingredients corresponding to 62 chemical families were applied in the province during the study period. Organophosphates, triazoles, sulfonylurea, pyrethroids, inorganic compounds (such as copper), carbamates, dithiocarbamates, neonicotinoids, aryloxyphenoxypropionates, and organochlorines predominated due to their use frequency. The use of toxic pesticides, and the lack of personal protection equipment, among others, made workers, residents, and applicators the toxicological modules with the highest risk of exposure. On the other hand, aquatic organisms, and the persistence of the pesticides in the soil and in groundwater, are the modules with the highest ecotoxicological pressure. By using the POCER and ƩSeq indicators, a more accurate toxicity and ecotoxicity assessment for certain pesticides can be performed in Cuba, in comparison to the one obtained when using only the TL equation currently employed in the country. In addition, substituting the most toxic pesticides (e.g., parathion, endosulfan, bifenthrin, copper oxychloride, mancozeb, paraquat, diquat, and ametryn) with less toxic ones (e.g., cypermethrin, tebuconazole, triadimenol, and bispyribac-sodium) could help reduce synthetic pesticide pressure on humans and the environment. Resumen Se realizó un estudio para cuantificar la toxicidad y la presión ecotoxicológica de los plaguicidas sintéticos en la provincia de Sancti Spíritus (Cuba) entre 2011 y 2014. Este trabajo puede ayudar a desarrollar políticas y prácticas de gestión para reducir los peligros del uso de plaguicidas sintéticos en el país. A través de un estudio longitudinal descriptivo, se identificaron los riesgos potenciales para el medioambiente y la salud humana asociados con el uso de plaguicidas. Para determinar la toxicidad y ecotoxicidad del uso de plaguicidas, se utilizaron los indicadores de aplicaciones equivalentes (ƩSeq) y de riesgos laborales y medioambientales (POCER, por su sigla en inglés), además de la metodología de carga tóxica (TL, por su sigla en inglés) del Instituto Cubano de Sanidad Vegetal. Durante el periodo de estudio, 124 ingredientes activos correspondientes a 62 familias químicas fueron aplicados. Por su frecuencia de uso, predominaron los organofosforados, triazoles, piretroides, compuestos inorgánicos (como el cobre), carbamatos, ditiocarbamatos, neonicotinoides, ariloxifenoxipropionato y organoclorados. El uso de plaguicidas tóxicos y la falta de equipos de protección personal, entre otros aspectos, hicieron que los trabajadores, los residentes y los aplicadores fueran los módulos humanos con el mayor riesgo de exposición. Por otro lado, los módulos de mayor presión ecotoxicológica son los organismos acuáticos, la persistencia en el suelo y el agua subterránea. Con el uso de los indicadores POCER y ƩSeq, se puede realizar una evaluación más precisa de la toxicidad y la ecotoxicidad en Cuba, en comparación con la obtenida al utilizar solo la ecuación TL, como se hace actualmente en el país. La sustitución de los plaguicidas más tóxicos (paratión, endosulfán, bifentrina, oxicloruro de cobre, mancozeb, paraquat, diquat y ametrina) por otros menos tóxicos (cipermetrina, tebuconazol, triadimenol y bispiribac-sodio) podría ayudar a reducir la presión de los plaguicidas sintéticos sobre los seres humanos y el medioambiente.

The object of this study was determination of organochlorine pesticide residues in freshwater samples collected along the Nile River at the Rosetta branch at Shubra Khait region El-Behera Governorate and Kufr EL-Zayat region, EL- Gharbia Governorate from two sites in different seasons during 2016-2017. The determination of organochlorine pesticide residues (PP-DDT, endosulfan, endrin, PP-DDD, OP-DDE, OP-DDD, Aldrin, Heptachlor- epoxide, heptachlor, d-HCH, a-HCH, g-HCH, dieldrin, PP-DDE, and methoxychlor) carried out by GC-µECD. The results indicated that the concentrations of pesticide residues monitored was below the permissible limits according to the World Health Organization (2011). Therefore, it is recommended to preserve the water of the Nile River by not dumpling sewage, agricultural and industrial waste without treatment in the Nile water and using pesticides in good way.

Metaflumizone is a novel semicarbazone insecticide. It functions as a sodium channel blocker insecticide (SCBI) with excellent insecticidal activity on most economically important lepidopterous pests. This study assessed the resistance risk of Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae) to metaflumizone in the laboratory and the effects of metaflumizone selection on toxicities to other insecticides. Spodoptera exigua collected from a field population at Huizhou in 2012 were successively challenged by metaflumizone to evaluate the risk of resistance evolution. Twelve generations of selection increased resistance to metaflumizone by 3.4-fold and threshold trait analysis revealed that the realized heritability (h2) of this resistance was 0.086. When h2 was equal to 0.086 and 90% of individuals were killed at each generation, LC50 to metaflumizone increased by 10-fold after 15 generations. The selection by metaflumizone did not increase the resistance to indoxacarb, chlorantraniliprole, spinosad, methomyl, or endosulfan, suggesting a lack of cross-resistance. However, metaflumizone challenge upheld the recession of resistance to emamectin benzoate, chlorfluazuron, and tebufenozide. The block of resistance drops by metaflumizone exposure implied a possible cross-resistance between metaflumizone and these three insecticides. These results contribute to integrated resistance management of S. exigua.

This work evaluates the factors affecting the presence of organochlorine pesticides in Pampeano aquifer in the Quequén Grande River watershed, Buenos Aires Province, Argentina. Eighteen sampling sites were selected in order to have representatives of different type of wells and types of soil. Among the analyzed compounds, endosulfan showed the highest concentrations(4.75 ng l−1 mean), which could be related to use in the recent past. Others important pesticides groups detected were HCHs (1.1 ng l−1 mean) and heptachlors (2.17 ng l−1mean). The analysis of the results show that the thickness of theunsaturated zone is the main factor related to the concentrations of pesticides and there were no differences according to wells or soil types. Although agricultural use of most OCPs has been banned in Argentina from more than 30 years ago, their residues are still detected in groundwater of the region, indicating their high persistence. Chronic exposure to pesticides is implicated in the etiopathogenesis of Parkinson’s disease (PD). Previously, we showed that dieldrin induces dopaminergic neurotoxicity by activating a cascade of apoptotic signaling pathways in experimental models of PD. Here, we systematically investigated endosulfan’s effect on the interplay between apoptosis and autophagy in dopaminergic neuronal cell models of PD. Exposing N27 dopaminergic neuronal cells to endosulfan rapidly induced autophagy, indicated by an increased number of autophagosomes and LC3-II accumulation. Prolonged endosulfan exposure (>9 h) triggered apoptotic signaling, including caspase-2 and -3 activation and protein kinase C delta (PKCδ) proteolytic activation, ultimately leading to cell death, thus demonstrating that autophagy precedes apoptosis during endosulfan neurotoxicity. Furthermore, inhibiting autophagy with wortmannin, a phosphoinositide 3-kinase inhibitor, potentiated endosulfan-induced apoptosis, suggesting that autophagy is an early protective response against endosulfan. Additionally, Beclin-1, a major regulator of autophagy, was cleaved during the initiation of apoptotic cell death, and the cleavage was predominantly mediated by caspase-2. Also, caspase-2 and caspase-3 inhibitors effectively blocked endosulfan-induced apoptotic cell death. CRISPR/Cas9-based stable knockdown of PKCδ significantly attenuated endosulfan-induced caspase-3 activation, indicating that the kinase serves as a regulatory switch for apoptosis. Additional studies in primary mesencephalic neuronal cultures confirmed endosulfan’s effect on autophagy and neuronal degeneration. Collectively, our results demonstrate that a functional interplay between autophagy and apoptosis dictate pesticide-induced neurodegenerative processes in dopaminergic neuronal cells. Our study provides insight into cell death mechanisms in environmentally linked neurodegenerative diseases.

Endosulfan is a DDT-era organochlorine pesticide. Due to past and current environmental contamination, investigation of endosulfan exposure is of current importance. Acute high dose exposure precipitates neural/endocrine system damage, but the effects on the immune system and of lower doses are not well-characterized. Two relatively low concentrations of endosulfan (i.e. 0.1 and 17 µM ENDO) were investigated in an in vitro study using human peripheral blood mononuclear cells (PBMC) to understand effects of relatively low doses (0.1–25.0 µM [≈0.04–10 ppm/40–10,000 ppb]) of ENDO upon normal human T- and B-lymphocytes and NK cells. The study here found that 17 µM ENDO inhibited phytohemagglutinin-M (PHA)-induced human PBMC proliferation. It was also seen that senescence and apoptosis among non-stimulated cells was increased, specifically within CD8 and NK populations, and that CD4:CD8 ratios also were increased. Treatment of non-stimulated PBMC with ENDO led to overall increases in production of tumor necrosis factor (TNF)-α, interferon (IFN)-γ, interleukin (IL)-2, -4, and -6, and decreased production of anti-inflammatory IL-10, suggesting an immunosenescence secretory phenotype. Interestingly, when the cells were pre-stimulated with mitogen (PHA), ENDO became inhibitory against the mitogen-induced proliferation and cytokine formation – with the exception of that of TNFα and IL-6, suggesting differential effects of ENDO on activated cells. Thus, at the organismal level, ENDO might also display differential effects during states of autoimmune disease or chronic viral infection in the exposed host.

Background: The study determined the presence and concentration of persistent organochlorine pesticides (OCPs) residues in cow milk from Ekiti State University Agricultural farm in Ado-Ekiti, Nigeria. The study was investigated in order to monitor consumer’s exposure to these chemicals pesticides. Methods: Qualitative identification and quantification evaluation of the extracted pesticides after clean-up on silica gel were done with a Gas Chromatography coupled with an Electron Capture Detector (GC-ECD). Results: The results revealed the presence of 11 OCPs residues in the milk samples, with concentration range of 0.001 - 0.189 mg/l, while α-BHC, endrin, endrin aldehyde, endosulfan II, endosulfan sulphate and methoxychlor were not detected. The analysis of variance revealed no significant variation in the levels of all the analysed pesticides except dieldrin. Conclusion: The hazard indices (HIs) were significantly lower than 1 with the range of 0.00063 - 0.107, indicating no potential health risk.

The extensive use of pesticides in agricultural practices has been associated with human health problems and environmental contamination worldwide. Brazil is the largest consumer of pesticides in the world and Espirito Santo State stands out as the second Brazilian producer of coffee. However, there is no information about environmental impact of coffee producing at Itapemirim River Basin (IRB) region, Espirito Santo State, Brazil; hence a simple and quick method using open access softwares (AGROSCRE and ARAqua Web) to estimate surface entrainment and pesticide leaching potential was carried out. AGROSCRE evaluates the contamination risk of superficial and groundwater by Active Ingredients (AIs), using GOSS Method, GUS index and US EPA criteria, while ARAqua Web estimates AI concentrations in water resources, indicating their suitability based on water quality requirements for different uses. Regarding pesticides used in coffee plantations, there is 44.7% chance of surface water and 23.7% chance of groundwater contamination. Results showed that ametryne, cyproconazole, diuron, epoxiconazole, flutriafol, triadimenol and triazophos pose contamination risk to both surface water and groundwater in the IRB region. Of these, 10.5% of the total AIs are triazoles and fall under environmental classes II and III (Product Very/Dangerous to the Environment). The AIs ametryne, thiamethoxam, iprodione, flutriafol, triazophos, endosulfan, triadimenol, cyproconazole, diuron, pendimethalin, chlorpyrifos, copper II hydroxide, etion, epoxiconazole and paraquat dichloride, were found to be potentially toxic, presenting environmental concentrations ranging from 123.40 μg L-1 to 0.14 μg L-1, which are higher than the safety standard for potable water (0.1 μg L-1). With respect to these AIs, chlorpyrifos, ethion and triazophos showed concentrations higher than EC50 values for aquatic invertebrates and ametryne and diuron for algae. Thus, local residents and environment may be at high risk of pesticide exposure, when these AIs are used in coffee plantations next to surface water or groundwater.

In the last decades, research regarding the dynamics of pesticides has grown, even in remote regions. Due to long-range atmospheric transport, environmental persistence and toxicological potential of organochlorine pesticides (OCPs), similar characteristics of current-use pesticides (CUPs) and their massive use in Brazil, these contaminants have become a major concern for environmental and human life. Thus, this study aimed to evaluate subgroups or individual chemicals of OCPs and CUPs, which could have travelled over two Conservation Unit sites in the Rio de Janeiro state. The study was carried out for 24 months, (2013–2015), in Itatiaia National Park (INP) and in the Serra dos Orgaos National Park (SONP), at ∼2400 and ∼2200 meters above sea level, respectively. The study was based on atmospheric passive sampling (polyurethane foam disks). Target pesticides were detected by means of gas chromatography device coupled with mass spectrometry (GC-MS). Significantly higher concentrations were measured in SONP when compared to INP. However, in broad terms, the contamination profile was quite similar for both national parks: The highest concentrations of endosulfan (INP - 1275 pg m−3 and SONP - 3202 pg m−3) were followed by cypermethrin (INP - 148 pg m−3 and SONP - 881 pg m−3) and chlorpyrifos (INP - 67 pg m−3 and SONP - 270 pg m−3). In agreement with previous studies, the atmospheric concentrations of legacy OCPs showed background air levels. The decrease of endosulfan over the years was highlighted with a parallel increase of chlorpyrifos, suggesting a collateral effect of the national bias of permissive and massive use of agrochemicals. CUPs seemed to behave like pseudo-persistent pollutants (pseudo-POPs). This is the first report of atmospheric concentrations of pyrethroids in Brazilian mountain regions, and possibly the first to investigate them in the air in South America or in any mountain region in the world.

Epimedium, is used traditionally in Chinese medicine to treat infertility problems. In this study, we establish the cell model to elucidate the protective effect of epimedium against ES by analyzing the molecular relationship between mitochondrial dynamics and steroidogenesis and to explore the molecular mechanism focusing on mitochondria function relating to fertility. ES induced ROS accumulation in mitochondria and the epimedium treatment significantly reduced the ROS accumulation. Furthermore, mitochondria morphology was restored to elongated shape following epimedium treatment. Epimedium treatment promoted dynamin-associated protein 1 (Drp1)-mediated steroidogenesis pathway by upregulating PKA, CREB, Drp1, and StAR protein expression in response to ES exposure in Leydig cells. Moreover, it was also identified that, CREB plays an important role in epimedium activation in Drp1-mediated steroidogenesis signaling pathway by increasing, phospho-CREB expression in nucleus. Testosterone level is decreased in ES-exposed cells; however, the testosterone level was increased after epimedium treatment. In conclusion, epimedium treatment improved mitochondria function in ES-exposed Leydig cells and activated downstream Drp1-dependent steroidogenesis by CREB mediated signaling pathway.

This study explored effects of dietary OCP intake from plant-origin foods (cereals, fruits, and vegetables) consumption on lipid metabolism and inflammation of women using a multiple follow-up study. The results showed that dietary intake of p,p′-dichlorodiphenyltrichloroethane (DDT) [β = − 10.11, 95% confidence interval (95%CI): − 17.32, − 2.905] and o,p′-dichlorodiphenyldichloroethylene (DDE) (β = − 6.077, 95%CI: − 9.954, − 2.200) were overall negatively associated with serum high-density lipoprotein cholesterol (HDL), whereas other OCPs were not. Serum interleukin (IL)-8 was positively associated with intake of dieldrin (β = 0.390, 95%CI: 0.105, 0.674), endosulfan-β (β = 0.361, 95%CI: 0.198, 0.523), total endosulfan (β = 0.136, 95%CI: 0.037, 0.234), and total OCPs (β = 0.084, 95%CI: 0.016, 0.153), and negatively correlated with intake of p,p′-DDE (β = − 2.692, 95%CI: − 5.185, − 0.198). We concluded that dietary intake of some individual DDT-, DDE- dieldrin-, and endosulfan-class chemicals from plant-origin foods may interfere with lipid metabolism and inflammation responses.

Background: Organochlorine pesticides used illegally in the storage of cowpeas by some farmers to prevent attack by weevils were known to affect the health of human adversely. Objectives: This study determined the mean concentration of aldrin, o,p΄-DDT, endosulfan, heptachlor epoxide and γ-HCH collected from some markets in South-South of Nigeria, compared these concentrations with set standards and ascertained the incidence of occurrence in cowpeas. Methods: Forty-eight samples of cowpeas were randomly collected from local markets, identified, authenticated, extracted with dichloromethane using Soxhlet apparatus, cleaned-up with silica gel and analysed in a GC-MS using Restek column. Results: The mean concentration of aldrin (445.08±6.43) µg/kg, heptachlor epoxide (188.84±3.42) µg/kg, o,p΄-DDT (182.24±5.13) µg/kg, endosulfan (142.94±2.05) µg/kg and γ-HCH (243.90±4.01) µg/kg in some markets were high and above the maximum residual limit. Conclusion: Organochlorine pesticides are used in most of the markets in south-south Nigeria for the preservation of cowpeas and in some cases, two or more were detected in a given sample, which could be due to the addition of a combination of pesticides. The need for regular monitoring by regulatory agencies and for sensitisation of both the farmers and merchants on the danger of using these pesticides in cowpeas preservation is therefore germane.

A monitoring network was established in streams within a catchment near the Costa Rican Pacific coast (2008–2011) to estimate the impact of pesticides in surface water (84 samples) and sediments (84 samples) in areas under the influence of melon and watermelon production. A total of 66 (water) and 47 (sediment) pesticides were analyzed, and an environmental risk assessment (ERA) was performed for four taxa (algae, Daphnia magna, fish and Chironomus riparius). One fungicide and seven insecticides were detected in water and/or sediment; the fungicide azoxystrobin (water) and the insecticide cypermethrin (sediments) were the most frequently detected pesticides. The insecticides endosulfan (5.76 μg/L) and cypermethrin (301 μg/kg) presented the highest concentrations in water and sediment, respectively. The ERA revealed acute risk in half of the sampling points of the melon-influenced area and in every sampling point from the watermelon-influenced area. Safety levels were exceeded within and around the crop fields, suggesting that agrochemical contamination was distributed along the catchment, with potential influence of nearby crops. Acute risk was caused by the insecticides chlorpyrifos, cypermethrin and endosulfan to D. magna, fish and C. riparius; the latter was the organism with the overall highest/continuous risk. High chronic risk was determined in all but one sampling point, and revealed a higher number of pesticides of concern. Cypermethrin was the only pesticide to pose chronic risk for all benchmark organisms. The results provide new information on the risk that tropical crops pose to aquatic ecosystems, and highlight the importance of including the analysis of sediment concentrations and chronic exposure in ERA. This study aims to determine the distribution of organochlorine pesticide pollution in water, sediments, mollusks, and fish at Saguling Dam as baseline data of organochlorine pollution. Samples were obtained from 12 locations, with 9 and 3 sampling points inside and outside the dam, respectively. Measurement of organochlorine residues was carried out using methods of extraction, purification, evaporation, and gas chromatography. Results showed the presence of several types of organochlorine compounds, namely, lindane, aldrin, dieldrin, heptachlor, dichlorodiphenyltrichloroethane (DDT), and endosulfan. Aldrin was dominant in water (2–37 μg/L) and sediments (2–1438 μg/L), while DDT and heptachlor were dominant organochlorine compounds in mollusks (13–2758 µg/L) and fish (11–104 μg/L), respectively. Sediments demonstrated higher organochlorine concentrations than water, mollusk, and fish. The distribution of organochlorine was affected by land use around the Citarum watershed and pollutant input from tributaries.

Pesticides are not only used on food but also on non-food crops, such as tobacco, to control a range of unwanted animal, plant, and microbial, fungal pests. The residue levels in tobacco leaves are expected to decline up to harvest, during drying, and when the leaves are further processed. Additional pesticides may also be applied to the finished product and residue levels may remain present even when the tobacco is burned. Human exposure to pesticide residues on tobacco occurs when residues remaining in cigarette smoke are inhaled. Based on this assumption, the objectives of this research were (i) to determine the level of pesticides residues in harvested tobacco leaves and (ii) to assess the risk of human exposure to these residues in tobacco smoke. Pesticide residues were detected in all analysed tobacco samples. These detected residues represent ten different active ingredients (AI), three of these AIs (thiodicarb, alachlor, and endosulfan) are no longer allowed in Europe. A 54.7% of these residues were quantifiable. Furthermore, it was found that with the use of solid-phase extraction sorbent (SPE) as adsorbent and n-hexane as solvent, higher recoveries of the pesticide residues in the tobacco smoke from the amount spiked can be obtained. It was also found that cigarette filters help to reduce the intake of residues of pesticides that may be present in cigarettes. Finally, the study concluded that both active smoking and passive smoking populations are exposed to pesticide residues in the tobacco smoke.

Chronic kidney disease of unknown etiology (CKDu), manifested clinically as tubulo interstitial fibrosis, has emerged as the second major cause of chronic kidney disease (CKD) in the Indian subcontinent and various agrochemicals have been implicated in its occurance. Among the agrochemicals organochlorine pesticides particularly endosulfan is well known for its toxicity and recent residue analysis have shown its presence in the blood samples of general population. In this present study, we have investigated the consequences of endosulfan exposure at a concentration (0.01 μM) equivalent to their highest reported presence in human blood sample of some CKDu patients, to human renal proximal tubular epithelial (HK-2) cell line with regard to ROS generation and expression of profibrotic and epithelial to mesenchymal (EMT) markers in order to find out endosulfan's ability to induce profibrotic changes in renal cell. We demonstrated a significant increase in intracellular ROS generation and increased expression of TGF-β1 when cells were incubated with β-endosulfan (0.01 μM) indicating occurrence of oxidative stress and fibrotic process. Again, decreased expression of epithelial marker E-cadherin and increase in the expression of mesenchymal marker α-smooth muscle actin (α-SMA) suggest possible onset of EMT process. Pre-treatment with 5 mM concentration of anti-oxidant N-acetyl cysteine partially attenuated the above process. In conclusion, these findings suggest possible involvement of β-endosulfan in the development of CKDu through oxidative stress and profibrotic signaling.

The presence of organochlorine pesticides (OCPs) and polybrominated diphenyl ethers (PBDEs) was analysed in air particulate matter ≤ 2.5 μm (PM2.5) and ≤10 μm (PM10) collected in the Metropolitan Zone of Mexico Valley (MZMV), during 2013 and 2014, respectively. Spatial and seasonal distributions of PM and their organic content named solvent extracted organic matter (SEOM) were determined. PM mass concentration and SEOM/PM ratios were compared with previous studies in 2006 in Mexico City. PM2.5 concentration was like found in 2006, however, PM10 decreased ∼43%. The SEOM/PM10 ratio was kept constant, suggesting a decrease in SEOM as well as PM10 emitted from natural sources, probably as a result of changes in the land use due to urban growth. A decrease ∼50% SEOM/PM2.5 ratio was observed in the same period, linked to adequate strategies and public policies applied by the local and federal governments to control the organic matter emitted from anthropogenic sources. Seven out of sixteen OCPs and five out of six PBDEs were found. The most common POPs were endosulfan I, endosulfan II, endosulfan sulfate, BDE-47 and BDE-99, present on >90% of the sampling days. OCPs in PM2.5 and PBDEs in PM10 showed seasonal variability. Higher PBDEs concentration in both particle sizes were observed at east and southeast of the MZMV, where one of the biggest landfills and wastewater treatment plants are located. OCPs in PM10 were mainly emitted from agricultural areas located to the southwest, southeast and east of the MZMV. OCPs in PM2.5 showed a regional contribution from the north and introduced into the valley. OCP degradation products were dominant over native OCPs, indicating no fresh OCP use. POPs comparison with other cities was made. Agreements and commissions created by the Mexican government reduced OCPs emissions, however, more effort must be made to control PBDE emission sources.

Rapid industrialization and urbanization have led to severe pollution into water resources which limits to reach safe drinking and irrigation water globally. One of the most important pollutants of environment that brought along with industrialization and technology are pesticides. In this study, it was aimed to investigate the pesticide residues in Dim Stream, due to intense touristic and agricultural activities in the region. Thus, four locations alongside the river were selected for sampling to evaluate the pesticide residue in the stream. The water samples were collected representing the rainy and dry seasons and extracted according to the Quechers method which is validated in terms of accuracy, specificity, limit of detection (LOD), and quantification (LOQ). Pesticide residues were analyzed by injecting LC-MSMS and GC-MS. The most recurrent pesticides were cypermethrin, endosulfan, deltamethrin, dicofol, metribuzin, parathion-methyl, permethrin, malathion, and tetradifon in the samples. Some of the levels of pesticides detected in water were significantly high compared with guideline values set by the Surface Water Quality Regulation of Turkey, EU, and World Health Organization, and this may be hazardous to aquatic life and human health. The obtained 18 recoveries of pesticides in the samples varied between 70 and 120%. LOD was ranged 19 from 0.23 to 9.67 μg/L. LOQ of 11 of the pesticides were higher than 1 μg/L.

The group of persistent organic pollutants (POPs) are particularly dangerous for the environment and by consequence for human health because of the risk to be transmitted in the food chain. Among them, the urgent problem of obsolete and forbidden organochlorinated pesticides (OCPs) needs a rigorous management in many countries, including Kazakhstan. The aim of our study was to evaluate the effect of pesticides content in food products on the genetic status and health of the population living on the contaminated areas near destroyed warehouses for OCPs (4 villages of Talgar district and 1 control site, Almaty region). The food products sampled in Taukaraturyk (control site), and in 4 villages where non-utilized obsolete pesticides were discovered: Beskainar, Kyzylkairat, Amangeldy, and Belbulak. The contents of 24 pesticides in food products from plant (apples, pears, tomatoes, cucumbers, sweet peppers) and animal (beef meat, cow milk, honey) origin, that grown in places of localization of non-utilized OCPs, were determined, sometimes in high and unacceptably high concentrations (before 2500 times over MRL). In pears, the pesticides content (especially DDT, γ-HCH, β-HCH, endosulfan, and aldrin pesticide group), was higher than in other fruits. Among vegetables, the highest levels of all groups of pesticide were found in cucumbers. Beef meat samples demonstrated increased contents of β-HCH, γ-HCH, endrin and dieldrin. In cow milk samples only the high concentration of dieldrin was found. The content of pesticides in meat was 4–5 times higher than in milk. The medical examinations, carried out among the cohorts living around the polluted by pesticides territories and control cohort from ecologically favorable village, showed that there were more individuals with high and middle levels of somatic health in the control group than in groups exposed to OCPs. The long-term effect of the pesticide contamination of the environment on genetic status of the population was assessed by chromosomal aberration (CA) frequencies. The highest level of chromosomal aberrations was identified for the examined residents of Kyzylkairat (41%) and Belbulak (38%), a high level in Amangeldy (12%), and middle level in Beskainar (6.5%). The association between the CA frequency, health status and the pesticides contents in food were assessed by a Spearman rank correlation. The low indicators of somatic health status were strictly associated with high levels of CA, and good health status indicates that the CA rates did not exceed the spontaneous level of mutagenesis. The strongest correlation was shown between high levels of chromosomal aberrations and the content of different pesticides in pears (Cr = 0.979–0.467), tomatoes (Cr = 0.877–0.476), cucumbers (Cr = 0.975–0.553) and meat (Cr = 0.839–0.368). The obtained results highlight the need to improve health protection by increasing the public awareness to the security of the storage of obsolete OCPs in order to strengthen food safety by efficient control services.

We investigated the levels and distribution patterns of α- and β-endosulfan and endosulfan sulfate in air, soil, water, and sediment samples collected from the South Korean persistent organic pollutants (POPs) monitoring networks. In the air samples, the highest concentrations of the total (Σ3) endosulfan (50.3–611 pg/m3, mean: 274 pg/m3) were observed during summer. Spearman analysis revealed a good correlation between agricultural land area and atmospheric concentrations of Σ3 endosulfan except during winter. Regardless of the season, the ratio of the two isomers (α/β) was 3.6–4.9 in the air samples, higher than that observed in technical mixtures (2.0–2.3), possibly due to the higher volatility of α-endosulfan, compared to β-endosulfan. Concentrations of Σ3 endosulfan in the soil samples (n.d.−13.4 ng/g, mean: 0.8 ng/g) were not significantly different except at some stations adjacent to large areas of farmland. The average levels of Σ3 endosulfan in the water and sediment samples were 2.1 ng/L and 0.1 ng/g dw, respectively. In analyzing the four largest rivers, it was observed that a few water stations during spring and fall and sediment stations in fall had high concentrations of the two isomers and endosulfan sulfate, particularly around the Yeoungsan and Nakdong Rivers near large areas of agricultural land. Endosulfan sulfate was dominant at most water and sediment sampling stations. This study demonstrates that the endosulfan found in most environmental compartments most probably derives from agricultural areas despite its ban as a pesticide. On the other hand, given that it was also detected in industrial and urban areas, in which pesticide application does not occur, it can be conjectured that endosulfan is aerially transported at higher temperatures and continuously circulates within the environment.

This study was carried out to investigate the risks of simultaneous exposure to pesticide residues and bacteria contaminants in locally produced fresh vegetables and vegetables in Tanzania. A total of 613 samples were analyzed for pesticide residues, out of which 250 were also analyzed for bacterial contamination. Overall, 47.5% had pesticide residues, 74.2% exceeded Maximum Residue Levels (MRLs). Organophosphorus (95.2%), organochlorines (24.0%), pyrethroids (17.3%), and carbamates (9.2%) residues dominated. MRL values were mostly exceeded in tomatoes, onions, watermelons, cucumbers, Chinese cabbage, and sweet paper. Tetramethrin (0.0329-1.3733 mg/kg), pirimiphos-methyl (0.0003-1.4093 mg/kg), permethrin (0.0009-2.4537 mg/kg), endosulfan (beta) (0.0008-2.3416 mg/kg), carbaryl (0.0215-1.5068 mg/kg), profenofos (0.0176-2.1377 mg/kg), chlorpyrifos (0.0004-1.2549 mg/kg) and dieldrin (0.0011-0.5271 mg/kg) exceeded MRLs. The prevalence of bacteria contamination was high (63.2%). Enterobacter (55.6%) Pseudomonas aeruginosa (32.4%), E. coli (28.2%), Citrobacter (26.8%), Klebsiella oxytoca (14.8%), and Salmonella (7.7%) were isolated. Furthermore, 46.4% tested positive for both pesticide residues and bacterial contaminants. Vegetables from farms (60.7%) contained more dual contaminants than market-based vegetables (41.8%). This may have resulted from excessive pesticide use and unhygienic handling of fresh fruits and vegetables at production level. Binary logistic regression showed that fresh fruits and vegetables with pesticide residues were 2.231 times more likely to have bacteria contaminants (OR: 2.231; 95% CI: 0.501, 8.802). The contamination levels of pesticide residues and bacterial contaminants could be perceived as a serious problem as most fresh fruits and vegetables recorded values of pesticide residues far above the MRLs with pathogenic bacteria isolated in higher proportions. MRLs was higher in most vegetables consumed raw or semi-cooked such as watermelons, carrots, cucumber, tomatoes, onion and sweet paper. There is an urgent need to develop pesticide monitoring and surveillance systems at farmer level, educating farmers and promoting the use of greener pesticides to mitigate the health effects of pesticides and bacterial contaminants.

Pesticides residue poses serious concerns to human health. The present study was carried out to determine the pesticide residues of peri-urban bovine milk (n = 1183) from five different sites (Bangalore, Bhubaneswar, Guwahati, Ludhiana and Udaipur) in India and dietary exposure risk assessment to adults and children. Pesticide residues were estimated using gas chromatography with flame thermionic and electron capture detectors followed by confirmation on gas chromatography-mass spectrometer. The results noticed the contamination of milk with hexachlorocyclohexane (HCH), dichloro-diphenyl trichloroethane (DDT), endosulfan, cypermethrin, cyhalothrin, permethrin, chlorpyrifos, ethion and profenophos pesticides. The residue levels in some of the milk samples were observed to be higher than the respective maximum residue limits (MRLs) for pesticide. Milk samples contamination was found highest in Bhubaneswar (11.2%) followed by Bangalore (9.3%), Ludhiana (6.9%), Udaipur (6.4%) and Guwahati (6.3%). The dietary risk assessment of pesticides under two scenarios i.e. lower-bound scenario (LB) and upper-bound (UB) revealed that daily intake of pesticides was substantially below the prescribed acceptable daily intake except for fipronil in children at UB. The non-cancer risk by estimation of hazard index (HI) was found to be below the target value of one in adults at all five sites in India. However, for children at the UB level, the HI for lindane, DDT and ethion exceeded the value of one in Ludhiana and Udaipur. Cancer risk for adults was found to be in the recommended range of United States environment protection agency (USEPA), while it exceeded the USEPA values for children.

Endosulfan, a mixture of α- and β-isomers, is used by farmers in the wet and dry season for khat and onion production. Khat leaf samples were collected in farmer fields at intervals of 1 h; 1, 5, 9 and 14 d after application. The dissipation rate of α- and β-isomers and residue level in khat were compared with residue levels in onion. The extraction was done by using Quick Easy Cheap Effective Rugged and Safe (QuEChERS) method and analyzed by Gas Chromatography – Electron Capture Detector (GC-ECD). Greater residue α- and β-isomer endosulfan levels were found in khat compared to onion as khat leaves are sprayed repeatedly in two week. Residue levels of khat exceeded the tolerable EU limit of 0.05 mg.kg−1 for leafy vegetables and herbs. For both raw and processed onion sample α- and β-endosulfan residues level were below the tolerable of limit EU regulation for bulb vegetables (i.e. 0. 1 mg.kg−1). The mean half-life for the α-isomer of endosulfan was 3.4 d in the wet season and 3.6 d in the dry season whilst that for the β-isomer was 5.0 d and 5.4 d respectively. Both isomers dissipated fastest in the wet season under conditions of high humidity and precipitation. The β-isomer persisted longer and had a lower dissipation rate from plants surface compared to the α-isomer.
