Sanguinarine (SAN), as the main active component of a traditional Chinese veterinary medicine, has been widely used in the animal husbandry and breeding industry. However, the metabolites of SA are still uncertain. Therefore, this research aimed to investigate the metabolites of SA based on rats in vivo. The blood, feces, and urine of rats were collected after the oral administration of 40 mg/kg SAN. Ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS) was employed to identify the metabolites of SAN. The elemental composition of sanguinarine metabolites was inferred by analyzing their exact molecular weight, and the structures of the metabolites were predicted based on their fragment ions and cleavage pathways. A total of 12 metabolites were identified, including three metabolites in the plasma, four in the urine, and nine in the feces. According to the possible metabolic pathways deduced in this study, SAN was mainly metabolized through reduction, oxidation, demethylation, hydroxylation, and glucuronidation. Background: The dry root of Salvia miltiorrhiza Bunge (Danshen in Chinese) is an used-widely traditional Chinese herbal medicine with and promising efficacy. This herbal plant has been extensively cultivated in China. Currently, people usually rely on its morphological features to evalaute its pharmaceutical quality. In this study, laser micro-dissection system (LMD) was applied to isolate single fresh tissue of root of S. miltiorrhiza. Under fluorescent microscopic model, five tissues namely cork, cortex, phloem, xylem ray and vessel were well recognized and isolated accurately by LMD, respectively and then the distribution pattern of the major bioactive compounds in various tissues was investigated by ultra-performance liquid chromatography-quadrupole/time of flight-mass spectrometry, which aims to validate the traditional experience on evaluating pharmaceutical quality of Danshen by morphological features. Results: Total 62 chemical peak signals were captured and 58 compounds including 33 tanshinones, 23 salvianolic acids and 2 others were identified or tentatively characterized in micro-dissection tissues. Further semi-quantitative analysis indicated that the bioactive components such as tanshinones and salvianolic acids were mainly enriched in cork tissue. Conclusion: In the present study, analysis of metabolic profile in different tissues of roots of S. miltiorrhiza is reported for the first time. Bopu powder® and Sangrovit® were developed from Macleayacordata and are widely used in agriculture and animal husbandry, but their impurities have been rarely reported in the literature. Impurity analysis is of great importance to the quality and safety of veterinary drugs. In this study, high-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF-MS) combined with a screening method was used to screen and characterize the impurities in Bopu powder® and Sangrovit®. A total of 58 impurities were screened from Bopu powder® and Sangrovit® using the screening strategies, of which 39 were identified by their accurate m/z value, characteristic MS/MS data, and fragmentation pathways of references. This established method was used for impurity analysis for the first time and proved to be a useful and rapid tool to screen and identify the impurities of Bopu powder® and Sangrovit®, especially for those at trace levels in a complex sample. In addition, this study marks the first comprehensive research into impurities in these two products and has great significance for the systematic detection of impurities in other plant-derived drugs. Although Lilium brownii (L. brownii) bulbs are popular fresh vegetables, a series of quality problems still remain after harvest. In this study, fresh L. brownii bulbs were placed in the dark at 25, 4, and −20 °C and under light at 25 °C from 0 to 30 days; the chemical compositions were analyzed by ultraviolet spectrophotometry (UV) and high-performance liquid chromatography quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF-MS). During the 30-day storage period, the browning degree increased over the storage time and with increasing temperature, but the contents of proteins and free amino acids decreased and were aggravated by light. The total polyphenol content increased until the 6th day at 25 °C (dark or light), but it did not significantly accumulate at −20 or 4 °C. The reducing sugar content showed a dynamic balance, but the total polysaccharide content decreased constantly in the four storage conditions. The polyphenol oxidase (PPO) activity increased with storage time and increasing temperature, while it was inhibited by light. The increase rates of malondialdehyde (MDA) content at −20 °C and light (25 °C) were higher than those at 4 and 25 °C. In addition, 12 secondary metabolites were identified, most of which accumulated during the storage period, for example, 1-O-feruloyl-3-O-β-D-glucopyranosylglycerol; 1,3-O-di-p-coumaroylglycerol; 1-O-feruloyl-3-O-p-coumaroylglycerol; and 1,2-O-diferuloylglycerol. The variations in nutrient levels had a low correlation with browning, but the variations in MDA, PPO, and secondary metabolite (phenolic acids) levels had a high correlation with browning. In conclusion, fresh L. brownii bulbs should be stored at a low temperature (4 °C) and in dark condition, and browning bulbs are excellent materials for secondary metabolite utilization. Bitter orange, Citrus aurantium L. var. amara (CAVA), is an important crop and its flowers and fruits are widely used in China as a food spice, as well as in traditional Chinese medicine, due to its health-promoting properties. The secondary metabolites that are present in plant-derived foods or medicines are, in part, responsible for the health benefits and desirable flavor profiles. Nevertheless, detailed information about the bioactive ingredients in CAVA is scarce. Therefore, this study was aimed at exploring the phytochemicals of CAVA by high performance liquid chromatography/quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF-MS). Here, a systematic screening method combined with HPLC-Q-TOF-MS was presented. This technique was used to systematically screen metabolites, primarily from the complex matrix of CAVA, and to identify these compounds by their exact masses, characteristic fragment ions, and fragmentation behaviors. A total of 295 metabolites were screened by the screening method and 89 phytochemicals were identified in the flowers, fruits, roots, leaves, and branches of CAVA. For the first time, 69 phytochemicals (flavonoids, alkaloids, terpenoids, etc.) were reported from CAVA. Aurantii fructus (zhiqiao, ZQ) is a traditional Chinese medicine (TCM) and raw material of TCM healthcare food (TCM-HF), mainly focused on the regulation of gastrointestinal disorders and the abundant application of antioxidants. Pharmacological investigations of ZQ flavonoids have identified them as the main bioactive components in recent years, but little has been reported on the extraction processes of antioxidant flavonoids (AFs). The aim of this study was to establish an efficient ultrasonic-assisted extraction (UAE) method for the extraction of AFs from ZQ using a response surface methodology (RSM), analyze the composition of AFs, and develop a qualitative evaluation method for ZQ. Flavonoid yield and antioxidant ability were selected as the responses to optimize the extraction of AFs, and the multiple effects of independent variables were investigated. The optimized conditions for the extraction of AFs based on the Box-Behnken design (BBD) were as follows: ethanol concentration, 58%; extraction temperature, 70 °C; and extraction time, 17 min. The flavonoid yield and antioxidant activity reached 241.70 mg/g and 59.42%, respectively, which matched the predicted values. Furthermore, optimized UAE processes were first established for the efficient and fast extraction of AFs. Flavanones and polymethoxyflavonoids (PMFs) were identified as potential AFs using time-of-flight mass spectrometry. Meanwhile, the quality of ZQ was evaluated using the criteria importance through intercriteria correlation (CRITIC) method for the first time, and Yuanjiang ZQ was considered as an excellent raw material of TCM-HF. Hemerocallis citrina Borani is an important crop and its flower buds are widely consumed in East Asian areas as a vegetable, as well as in traditional Chinese medicine, due to its health-promoting properties. Metabolites present in plant-derived foods or medicines are in part responsible for their desirable flavor profiles and health benefits. Nevertheless, detailed information about these compounds in H. citrina is scarce. Therefore, this study aimed to investigate the metabolites by high-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF-MS). In this study, a total of 144 compounds, including 14 amides, 25 polyphenols, 44 flavonols, 35 anthraquinones, 15 naphthols, and 11 other components, were detected by the established screening method and were identified by their precise m/z values, characteristic tandem mass spectrometry (MS/MS) data and fragmentation pathways of references, 111 of which were reported in this plant for the first time. The distribution of identified ingredients in different parts of H. citrina was determined. Interestingly, colchicine, which had been reported as a toxic compound in the fresh flower buds in previous studies and various news reports, was not found.
