Porcine circovirus 3 (PCV-3) has emerged as a significant pathogen affecting global swine populations, yet its epidemiology and clinical implications remain incompletely understood. This retrospective study aimed to investigate the prevalence and histopathological features of PCV-3 infection in pigs from Switzerland, focusing on archival cases of suckling and weaner piglets presenting with suggestive lesions. An in-house qPCR assay was developed for detecting PCV-3 in frozen and formalin-fixed paraffin-embedded tissues, enhancing the national diagnostic capabilities. Histopathological reassessment identified PCV-3 systemic disease (PCV-3-SD) compatible lesions in 19 (6%) of archival cases, with 47% testing positive by qPCR across various organs. Notably, vascular lesions predominated, particularly in mesenteric arteries, heart, and kidneys. The study confirms the presence of PCV-3 in Switzerland since at least 2020, marking the first documented cases within the Swiss swine population. Despite challenges in in situ hybridization validation due to prolonged formalin fixation, the findings indicate viral systemic dissemination. These results contribute to the understanding of PCV-3 epidemiology in Swiss pigs, emphasizing the need for continued surveillance and further research on its clinical implications and interaction with host factors. Our study underscores the utility and limitations of molecular techniques in confirming PCV-3 infections.

Objective: Milk samples from clinically healthy sows often contain a similar bacterial content as samples from mastitic sows. To verify whether contamination during sample collection is a possible reason for this or bacterial content in the mammary gland postpartum is a regular finding, the aim of the present study was to compare the suitability of milk samples and mammary gland biopsies for assessing the bacteriological status of healthy sows., Material and Methods: Twenty-five clinically healthy sows of different parities were selected. The mammary skin and teats were cleaned and disinfected before biopsies and milk samples were taken from the second mammary gland on the left udder side one day postpartum. Needle biopsies were performed after local anaesthesia. Samples were investigated bacteriologically for aerobic bacteria and a semi-quantitative classification of bacterial growth was carried out. Additionally, formalin-fixed paraffin-embedded biopsies from 18 of the healthy sows were stained and scored for inflammatory cells., Results: A low bacterial content could be found in 96 % of milk samples and in 92 % of biopsies from the healthy sows. Both Gram-positive (mostly streptococci and staphylococci) and Gram-negative bacteria ( Escherichia coli) were detected. Histopathological examination revealed mild inflammatory cell infiltration with mainly plasma cells and lymphocytes, in rare cases neutrophilic granulocytes., Conclusion: Biopsies of the mammary gland provided similar results with regard to bacteriological investigation compared to milk sampling although these were collected under aseptic conditions. Therefore, it is assumed that ubiquitous bacteria are regularly present in the colostrum and in the mammary gland tissue of clinically healthy sows after parturition. This report describes the detection of "Brachyspira (B.) hampsonii" clade I in Belgian pigs imported to Germany. Two of seventeen pigs from one herd were reported positive for Brachyspira hyodysenteriae by culture in a Belgian diagnostic laboratory, but negative for this Brachyspira species by specific PCR. In this study, from 22 fecal samples and 2 colon contents of these animals various Brachyspira species were cultured and identified by nox-RFLP as Brachyspira murdochii, Brachyspira innocens and Brachyspira intermedia. Albeit the six B. intermedia isolates proved to be negative in a species specific PCR. Sequencing of the nox-gene of three of these isolates revealed that the sequences were 99% identical to published sequences of "B. hampsonii" clade I. From one pig which was positive for "B. hampsonii" clade I histopathology was done and showed moderate lesions consistent with brachyspiral disease., (Copyright © 2013 Elsevier B.V. Skin tumors are rarely observed in pigs, and, in particular, only a few cases of squamous cell carcinoma have been reported. The present case report describes the occurrence of a squamous cell carcinoma located in the skin at the center of the udder of a 14-year-old German Landrace sow. The tumor was spherical and 17.0 cm in diameter. The tissue of the distal pole had a cauliflowerlike appearance, was black and ulcerated with a malodorous exudation. The sow did not show any signs of reduced general condition caused by the tumor. No evidence of metastasis was noted in regional lymph nodes and in the lung. The tumor, which weighed 2.4 kg, was surgically removed. The histopathology revealed infiltratively growing pleomorphic tumor cells and cell aggregations with prominent squamous differentiation. Cells were strongly positive for several cytokeratins by immunhistochemistry. The present findings confirmed the diagnosis of squamous cell carcinoma. After surgery, the sow recovered without complications and remained without recurrence at the follow-up three months later. This is the first report ofa squamous cell carcinoma in the skin in the udder in an old sow.

In industrial countries, Hepatitis E virus (HEV) transmission to humans is predominantly assumed to be a zoonotic infection. Recently, it has been demonstrated that about 50% of domestic pigs in Germany carry HEV-specific antibodies. However, further investigations concerning the distribution of HEV in different age groups of German domestic pigs, phylogenetic analyses and the viral load in the porcine liver are still pending. Liver samples of all age groups from herds in a pig-dense region in north-western Germany were investigated for the presence and quantity of HEV RNA and subsequently genotyped. Out of 251 liver samples, 34 contained ORF2-specific RNA, whereas 19 samples were positive using ORF1-specific primers, resulting in an overall detection rate of 13.5% and 7.6%, respectively. Especially nursery pigs and growers were tested positive for viral RNA. Furthermore, determination of the HEV copy numbers revealed high replication levels. Up to 10(9) genome copies per g of liver tissue could be detected suggesting a likely high degree of viral spread to the environment. In the HEV-positive liver samples we found no hints for pathohistological changes reflecting the HEV status.The HEV sequences showed marked diversity but could be assigned to HEV genotype 3 without exception. However, by comparing two different genomic fragments, we found indications for infections with two different HEV variants in domestic pigs. Apart from this, the current study confirms the outcome of our recent serological HEV survey and for the first time gives direct proof of HEV infections in the German domestic pig population.
