The genes for the production of elloramycin (ELM) from Streptomyces olivaceus (So) Tü2353 were cloned using a polyketide synthase gene probe from the tetracenomycin pathway. A cosmid clone (16F4) isolated from a gene library of So Tü2353 conferred tetracenomycin C and ELM resistance to S. lividans TK64 and complemented a mutation in So Tü2353R. Introduction of cosmid 16F4 into S. lividans TK64 resulted in the production of 8-demethyl-tetracenomycin C, an intermediate of ELM biosynthesis.

In a screening programme for substances with morphogenic effects on the nikkomycin producer strain Streptomyces tendae Tü 901 we identified a metabolite, which induced aerial mycelium formation in the bld mutant Tü 901/S 2566-EM 1. By using a HPLC UV/Vis absorbance spectral library we could confirm that this compound was identical with the macrolide antibiotic borrelidin. 100 ng borrelidin/paperdisc were sufficient to show an evident morphological effect.

During germination spores of Streptomyces viridochromogenes NRRL B-1551 excrete a compound, germicidin, which has an inhibitory effect on the germination of its own arthrospores at a concentration as low as 200 pM (40 pg/ml). At higher concentrations germicidin inhibits porcine Na+/K(+)-activated ATPase and retards the germination of the cress Lepidium sativum. Germicidin is the first known autoregulative inhibitor of spore germination in the genus Streptomyces and was isolated from the supernatant of germinated spores, but also from the supernatant of the submerged culture. Spectroscopic analysis and derivatization reactions revealed germicidin to be 6-(2-butyl)-3-ethyl-4-hydroxy-2-pyrone (C11H16O3). Crude isolates of germicidin from the supernatant of submerged culture, but not from the spores, contained a second, structurally very similar compound (C10H14O3), in which in contrast to germicidin a 2-propyl instead of the 2-butyl chain was bound to C-6 and which did not show any activity in the germination and ATPase assay. The germination assay was evaluated as a new screening model for specifically active compounds.

New intermediates associated with nikkomycin biosynthesis, called nikkomycins SZ, SX, SoZ and SoX, were isolated and characterized from the culture broth of Streptomyces tendae Tü 901/S 2566. They are analogues to octosyl acids, shunt metabolites of polyoxin biosynthesis. The decreasing amounts of nikkomycins SZ and SX, produced in the culture medium, shows a significant correlation to the increasing amounts of the biologically active nikkomycins Z and X, dependent on the increasing concentration of iron.

Streptomyces olivaceus TU 2718 produces the siderophore desferrioxamine E. Production depends on L-lysine and iron concentrations in the medium. With optimized conditions the yield of desferrioxamine E could be increased to 12 g/l in feeding fermentations. Supplementation of the basic production medium with natural and synthetic precursors of desferrioxamine E led to the production of twelve new analogues of desferrioxamine E.

Two new dipeptidyl nikkomycins of the Z and X type were isolated from the culture broth of Streptomyces tendae TU 901/395-11/32 and characterized. They show a variation in the amino acid moiety of the molecule. Nikkomycin Wz is composed of L-tyrosine and 5-amino-5-deoxy-D-allo-furanuronic acid N-glycosidally bound to uracil, whereas nikkomycin Wx is composed of L-tyrosine and 5-amino-5-deoxy-D-allo-furanuronic acid N-glycosidally bound to 4-formyl-4-imidazolin-2-one. The new nikkomycins are good inhibitors of chitin synthetase from Coprinus cinereus but they did not inhibit growth of fungi and yeasts.

From the cultures of Streptomyces violaceoniger, strain Tü 96, two new lipophilic antibiotics, Lysolipin I and Lysolipin X were isolated. The latter one is chemically unstable and is easily transformed to Lysolipin I. The deeply yellow Lysolipin I has a molecular formula C29H24CINO11. It was characterized by the ir, uv, H-nmr and 13C-nmr spectra, which make a quinone structure very probable. Lysolipin I is active against Gram-positive and Gram-negative bacteria. However, enterobacteriae are only inhibited in high dilution, when the membrane permeation is damaged. Lysolipin I acts lytically against bacterial cells. Its activity is decreased by several lipids. The site of action is the biosynthesis of bacterial cell walls, an interaction with the carrier lipid for mureine intermediates being probable.

Four new dipeptidyl nikkomycins of the Z- and the X-series with a variation in the amino acid moiety of the molecule were isolated from the mutant Streptomyces tendae 901/395 and characterized. Nikkomycins Kz and Kx contain 2-amino-4-hydroxy-4-(2-pyridyl)butyric acid, and nikkomycins Oz and Ox 2-amino-4-hydroxy-4-(5-hydroxy-2-pyridyl)butyric acid. In contrast to nikkomycins Z and X, nikkomycins Kx and Ox are quite stable at alkaline pH and exhibit a lower biological activity against various test organisms. From the mutant S. tendae 901/C37, which is auxotrophic for methionine and threonine, enhanced amounts of two tripeptidyl nikkomycins, Qz and Qx, were produced which are analogues of nikkomycins J and I and contain a homoserine residue instead of glutamic acid. These nikkomycins exhibit a high pH instability.

A new antibiotic rinamycin was isolated from the culture mycelium of a streptomycete designated as Streptomyces venezuelae TU 1102. The antibiotic was obtained as a honey-coloured powder having the tentative molecular formula C20H55N08. It exhibits a significant inhibition of fungi, yeasts, gram-negative bacteria. The primary site of action of the antibiotic was suggested to be the RNA metabolism of rinamycin-sensitive cells.

From a strain of Streptomyces antibioticus seven yellow phenazines were isolated. The antibacterially most active antibiotic was identified as (-)-saphenamycin, a second one with compound DC-86-Y (saphenic acid). Three compounds were new: Saphenic acid methyl ether, 6-acetylphenazine-1-carboxylic acid and an inseparable mixture of fatty acid esters of saphenic acid. Two simple phenazines were phenazine-1-carboxylic acid (tubermycin B) and unsubstituted phenazine, which was isolated for the first time from a microorganism.

Streptomyces glaucescens, strain Tü 49 = ETH 22794, produces hydroxystreptomycin as well as the tetracenomycins, a mixture of several lipophilic antibiotics. The main component and the most active one is tetracenomycin C. Tetracenomycin C has a molecular formula C23H20O11 and is chemically related to tetracyclines and anthracyclinones. The pale yellow antibiotic is active against some gram-positive bacteria, especially against streptomycetes. Gram-negative bacteria and fungi are not inhibited. In considering the differences of biological activity and the functional groups of the molecule, tetracenomycin C is not a member of the tetracycline or anthracyclinone group of antibiotics.

Two new nikkomycins were isolated from the culture broth of Streptomyces tendae Tü 901/PF 53+-3. The new compounds are the dipeptide nikkomycin pseudo-Z (psi Z) and tripeptide nikkomycin pseudo-J (psi J), which are analogues to nikkomycins Z and J. Nikkomycins pseudo-Z and pseudo-J have a C-glycosidic linkage between uracil and 5-amino-5-deoxy-D-allo-furanuronic acid, which is comparable to the C-glycosidic bond in pseudouridine. The new Cc-nucleoside nikkomycins exhibit a lower biological activity than the CN-nucleoside nikkomycins.

From a new strain of Streptomyces, Streptomyces parvulus, strain Tü 2480 three glucosides of the alkaloid pyridindolol were isolated. The structure elucidation is based on spectroscopic investigations and degradation to pyridindolol and alpha,D-methyl glucoside.

The colored urdamycins A to F, six new angucycline antibiotics produced by Streptomyces fradiae strain Tü 2717, were detected by chemical screening. They are biologically active against Gram-positive bacteria and stem cells of murine L1210 leukemia. The urdamycins are glycosides and differ in their aglycones, which can be liberated by acidic hydrolysis besides the sugars D-olivose and L-rhodinose. The structure of the main compound, urdamycin A, follows from the spectroscopic and chemical data in connection with an X-ray analysis. The aglycone urdamycinone A is identical with aquayamycin. The structures of urdamycin B, E, F and partial structures of urdamycin C and D, will be presented in a subsequent paper. The new term "angucycline/angucyclinone" is used for an increasing group of related antibiotics.

Tetracenomycins B3 and D3, besides tetracenomycin D (D1), were produced by a blocked mutant of the elloramycin producer Streptomyces olivaceus TU 2353. The compounds were isolated as red powders, and their structures were elucidated by comparing their physicochemical data with those of the known tetracenomycins A2, B1, B2, D and E. Tetracenomycin B3 (2), the main compound, and tetracenomycin D (3) were antibiotically inactive against Gram-positive and Gram-negative bacteria, whereas tetracenomycin D3 (1) showed a moderate activity against Bacillus subtilis and Arthrobacter aurescens. Tetracenomycin B3 (2) is the key intermediate where the biosynthesis of the elloramycins branches off from the line leading to tetracenomycin C (5) as the final product of the tetracenomycin biosynthesis branch.

Bacimethrin (1), known as a thiamine antagonist produced by Bacillus megatherium, was isolated from Streptomyces albus and has been further characterized by NMR spectra and acetylation. A new easy three step synthesis for 1 is described. The biological activity of 1, and its mode of action were discussed. There are indications that bacimethrin inhibits the phosphorylation of 4-amino-5-hydroxymethyl-2-methylpyrimidine (Pyr-OH) during thiamine biosynthesis.

Streptomyces olivaceus, strain Tü 1379 produces beside the red dyestuff prodigiosin several closely related hitherto unknown cyclodepsipeptide antibiotics, mainly imacidine B and C and imacidinic acids. The antibiotics affect murein biosynthesis. Only actinomycetes are inhibited.

To isolate Streptomyces tendae mutants blocked in the biosynthesis of the nikkomycin nucleoside base 4-formyl-4-imidazoline-2-one, an assay was developed to detect the formation of nikkomycins containing this base during growth on solid medium. The assay is based on the reaction of the 4-formylimidazolone structure of nikkomycins with the aldehyde reagent barbituric acid leading to red-colored products. Among 18,000 N-methyl-N'-nitro-N-nitrosoguanidine treated clones tested in the barbituric acid assay, we isolated one mutant which was incapable of forming any nikkomycins containing the 4-formylimidazolone base (nikkomycins Cx, X and I) but instead produced nikkomycins containing uracil (nikkomycins C, Z and J). In addition, we isolated strains with mutations affecting the biosynthesis of 2-amino-4-hydroxy-4-(5-hydroxy-2-pyridyl)-3-methylbutyric acid, the unusual amino acid of nikkomycins Z, X, J and I. By analyzing colonies derived from single spores or protoplasts of S. tendae Tü901/395, a mutant producing besides nikkomycins Z, X, J and I, also nikkomycins Kz/Kx and Oz/Ox, we obtained strains which only formed nikkomycins Kz/Kx and Oz/Ox with 2-amino-4-hydroxy-4-(2-pyridyl)butyric acid and 2-amino-4-hydroxy-4-(5-hydroxy-2-pyridyl)-butyric acid as amino acids. Mutation of such a strain (Tü901/395-11) by UV365nm in the presence of 8-methoxypsoralen and selection of S-2-aminoethyl-L-cysteine-resistant clones led to the isolation of Tü901/AEC1 and AEC2 which produced exclusively nikkomycins Kz and Kx. According to their nikkomycin spectrum, these strains were blocked at the hydroxylation step occurring at the pyridyl residue during biosynthesis of the nikkomycin amino acid.

(S)-3-p-methoxyphenyl-3-acetamidopropan-1-ol was isolated from cultures of an actinomycete (Streptomyces michiganensis). Its structural determination by spectroscopic means and its synthesis are described.

Elloramycin (1), a new antibiotic produced by Streptomyces olivaceus strain Tü 2353, was detected by chemical screening. The dark yellow compound, molecular formula C32H36O15, is weakly active against a variety of Gram-positive bacteria, especially streptomycetes and against stem cells of L-1210 leukemia. Acidic hydrolysis of the antibiotic liberated elloramycinone (3) as aglycone and 2,3,4-tri-O-methyl-L-rhamnose, which was identified as methyl glycoside 5b. The structure of elloramycin was established by comparison of the spectra (UV, 1H NMR, 13C NMR) with those of the known tetracenomycin C (2), 3 and the fact that 2 and 3 gave the same tetramethyl ether after permethylation. Elloramycin is an anthracycline-like antibiotic, the aglycone resembles tetracenomycin C, the sugar is connected in a phenolic alpha-glycosidic linkage.

Four new and two known peptide antibiotics containing amiclenomycin (Acm) have been isolated from a culture of Streptomyces venezuelae Tü 2460: L-MeIle-L-Acm (1), L-Ile-L-Acm (2), L-MeVal-L-Acm (3), L-MeIle-L-Acm-L-Gln (4), L-Ile-L-Acm-L-Gln (5) and L-Val-L-Acm-L-Gln (6). These di- and tripeptides exhibited antimicrobial activity on a minimal medium against Gram-negative bacteria, which could be reversed by biotin. It was shown that the antibiotics were decomposed by peptidases to provide amiclenomycin (7) after their uptake into cells of Escherichia coli via peptide-permeases. The antimicrobial activity of the amiclenomycin-peptides was the inhibition of DAPA-aminotransferase by the amiclenomycin-warhead, however, amiclenomycin itself was hardly transported into the cells. Since the amiclenomycin peptides misuse general transport systems, they are presented here as examples for the illicit transport concept.

Fermentations of Streptomyces violaceoniger TU 905 produce the antifungal antibiotics niphithricins A, B, elaiophylin and nigericin. The niphithricins have been characterized as new macrolide antibiotics, and the previously unknown structure of elaiophylin was determined to be a macrodiolide. The niphithricins were biologically active against Gram-positive bacteria and fungi. The mode of action is attributed to an alteration of the membrane permeability.

From the fermentation broth of Streptomyces tendae Tü 901 a substance was isolated which inhibits the growth of several fungi. The new antibiotic affects the chitchin biosynthesis. Its structure was identified by mass spectrometry of the products obtained after chemical degradation. Nikkomycin is a nucleoside-peptide antibiotic consisting of uracil, an amino hexuronic acid and a new amino acid containing a pyridin ring.
