Invasive predators often impose devastating selection pressures on native prey species. However, their effects can be regionally dependent and influenced by the local ecological conditions of their invaded habitats. Evolved behavioral phenotypes are important mechanisms by which prey adapt to the presence of novel predators. Here, we asked how behavior and behavioral plasticity of threespine stickleback (Gasterosteus aculeatus) populations have evolved following the introduction of the invasive predator, northern pike (Esox lucius). We examined the behavior of F1 offspring generated from three pike-free and three pike-invaded populations and measured how stickleback activity and plant use behaviors, and their plasticity, have evolved following pike introduction. To evaluate plasticity, we exposed juvenile stickleback to predator cues during their first year of development and then evaluated how this repeated exposure influenced behavioral responses to an artificial predation event. We found no overarching effect of pike in either evolved behaviors or behavioral plasticity, and no evidence for the presence of developmental plasticity. Furthermore, we found that depending on the phenotype, pike-invaded stickleback populations have either more or less among-population variation than pike-free populations. Predation often has consistent effects on prey behavior and morphology, but whether the physiological mechanisms underlying these effects show similarly consistent patterns across different populations remains an open question. In vertebrates, predation risk activates the hypothalamic-pituitary-adrenal (HPA) axis, and there is growing evidence that activation of the maternal HPA axis can have intergenerational consequences via, for example, maternally-derived steroids in eggs. Here, we investigated how predation risk affects a suite of maternally-derived steroids in threespine stickleback eggs across nine Alaskan lakes that vary in whether predatory trout are absent, native, or have been stocked within the last 25 years. Using liquid chromatography coupled with mass spectroscopy (LC-MS/MS), we detected 20 steroids within unfertilized eggs. Factor analysis suggests that steroids covary within and across steroid classes (i.e. glucocorticoids, progestogens, sex steroids), emphasizing the modularity and interconnectedness of the endocrine response. Surprisingly, egg steroid profiles were not significantly associated with predator regime, although they were more variable when predators were absent compared to when predators were present, with either native or stocked trout. Despite being the most abundant steroid, cortisol was not consistently associated with predation regime. Thus, while predators can affect steroids in adults, including mothers, the link between maternal stress and embryonic development is more complex than a simple one-to-one relationship between the population-level predation risk experienced by mothers and the steroids mothers transfer to their eggs. The Fourspine Stickleback (Apeltes quadracus) is a diminutive member of the Gasterosteidae inhabiting a rather narrow zone comprising coastal and near-coastal brackish and freshwater habitats from the Gulf of St. Lawrence to North Carolina. Behavior, morphology, and several other aspects of the biology of A. quadracus have been well studied, yet there are surprisingly few investigations of its life history. Here, we provide data on four reproductive traits for female A. quadracus from seven sites in Nova Scotia, Canada. The sites occur within three distinct regions of Nova Scotia, and encompass a variety of brackish-water and freshwater habitats. Female size, clutch mass adjusted to a common body size, and clutch size adjusted to a common body size differed significantly among the three regions. These traits also showed significant variation across sites within regions. Egg dry mass did not differ across regions, but there was significant variation across sites within regions. Our study, and earlier ones from more southern populations, indicate that there may be variation of evolutionary interest in several traits on multiple spatial scales in this species. Isolation from predators can lead to the reduction or loss of ancestral behavioral defenses in prey, but does not always do so. Predators introduced to populations that have experienced relaxed selection from some ancestral predators can favor the evolution of antipredator behavior that has been lost. We examined these possibilities by eliciting antipredator behavior in three populations of threespine stickleback fish, Gasterosteus aculeatus: an oceanic population thought to resemble the ancestral form, and two populations historically (up to 20 000 yr) devoid of piscine predators (relaxed selection), one of which has been stocked with salmonids for nearly 25 yr (reversed selection). We used three kinds of predator models: a sculpin (ambush predator), a rainbow trout (chasing predator), and an overhead silhouette of an arctic tern. Stickleback reacted differently to the three models, indicating that they distinguished among them. Individuals from all populations responded similarly to the tern model. The ancestral population showed the weakest response to the sculpin model despite being the only population that encounters these predators naturally. Stickleback from the trout-free population displayed slightly reduced responses to the trout model, and recovery times like those in the ancestral population providing only weak evidence for loss of the ancestral antipredator repertoire. Fish from the reverse-selected population exhibited fascinating, elevated responses to both the trout and sculpin models relative to the other two populations. These findings offer initial evidence of (1) a partial alteration of the ancestral behavioral repertoire during a long period of relaxed selection from piscine predators, and (2) rapid acquisition of extreme responses to piscine predators under reverse selection. Most research on ontogenetic niche shifts has focused on changes in habitat or resource use related to food resource distribution and heterospecific size-limited predation. Cannibalism, an intraspecific interaction, can also affect habitat selection or resource use by vulnerable size classes. Morphological defenses, such as spines, increase the effective size of an individual, making it more difficult to consume. The importance of such defense structures in affecting niche shifts in early life history stages is unclear. Using a combination of field observations and experiments in aquaria and wading pools, we examined the relative roles of cannibalism and morphology in determining juvenile habitat use in two populations of threespine stickleback that differ in pelvic spine morphology. Juveniles were categorized into three size classes: small (5-10 mm), medium (11-15 mm), and large (15-25 mm). In experiments assessing the relative vulnerability of juveniles to cannibalism by adults, we documented a significant difference among size classes in the number of juveniles eaten such that more large juveniles were eaten from the population lacking pelvic spines. The natural distribution of small and large juveniles in two distinct littoral microhabitats, open water and vegetation, was determined in each lake. In both populations, small juveniles were more abundant in vegetation. In the population with pelvic spines, a greater proportion of large juveniles was observed in open water than in vegetation. In the population without pelvic spines, the proportion of large juveniles did not differ between the two habitats. Experiments comparing juvenile habitat use in the presence or absence of adult conspecifics suggest that differences in habitat use may not only depend on the size of the individual, or the size of the individual relative to the size of the adult predator, but also on the degree of development or expression of defensive structures. Abstract Hormones play a prominent role in animal development, mediating the expression of traits and coordinating phenotypic responses to the environment. Their role as physiological integrators has implications for how populations respond to natural selection and can impact the speed and direction of evolutionary change. However, many emerging and established fish models with the potential to be ecologically or evolutionarily informative are small-bodied, making hormone sampling through traditional methods (whole-body or plasma) lethal or highly disruptive. Sampling methodology has thus restricted study design, often limiting sample sizes, and has prevented the study of at-risk/endangered populations. We utilize water-borne hormone sampling, a minimally invasive method of measuring the rate of steroid hormone release across the gills and further validate this method in a novel, evolutionary context. First, we compare water-borne hormone measures of cortisol with those quantified from plasma and whole-body samples collected from the same individuals to establish the relationship between concentrations quantified via the three methods. We then compare the release of steroid hormones in three populations of threespine stickleback to establish the sensitivity of this tool in measuring within-individual and between-individual variation in biologically relevant contexts (reproductive stages), and in assessing differences among populations with distinct evolutionary histories. We demonstrate a strong positive relationship between cortisol concentrations measured with water-borne, plasma, and whole-body collection techniques. Tracking estradiol and testosterone throughout clutch production in females produced anticipated patterns associated with growing and maturing eggs, with divergence in estradiol production in one population. Additionally, differences among populations in cortisol levels at ovulation paralleled the relative presence of a social stressor, and thus expected energetic needs within each population. We confirm that water-borne hormone sampling is sufficiently sensitive to capture biologically relevant fluctuations in steroid hormones between environmental contexts and demonstrate that among-population differences are detectable. This technique can be applied broadly to small fish to answer important ecological and evolutionary questions. By linking population variation in hormones and the multivariate phenotype, this technique will help elucidate both proximate mechanisms underlying phenotypic development and variation, and the way hormone networks alter evolutionary responses to selection. Abstract: Recent adaptive radiations offer special insight into the processes that generate biodiversity. The loss of unique members of such radiations undermines their collective evolutionary potential, as well as the ability of scientists to compare divergent populations, thereby devaluing the entire radiation as a system of study. To illustrate our case, we consider the adaptive radiation of the threespine stickleback fish (Gasterosteus aculeatus) in light of anthropogenic threats. Specifically, we consider the effects of stocking with rainbow trout, Onchorynchus mykiss, on populations of stickleback in lakes of two types in the Cook Inlet Region of Alaska, USA: (1) those in which salmonids are native and (2) those historically devoid of predatory fish. Many populations in this second class exhibit reduced pelvic armor that is, in part, a consequence of the historical absence of piscine predators that prey upon stickleback. Long-term trapping data from 77 lakes show that stocking in lakes where trout are not native is associated with large fluctuations in stickleback abundance, relative to lakes in which trout are native, or to lakes in which trout are neither native nor stocked. Pelvic-reduced populations appear to be at most risk from stocking. We discuss how these populations have responded to, and may yet respond to such threats, and how the information we have on these populations can be used to identify evolutionarily significant units (ESUs: ), and certainly, distinct populations segments (DPSs: ) deemed worthy of protection under Section 3(15) of the 1973 US Endangered Species Act as amended in 1978.
