Objective: The semen quality of stallions including sperm motility is an important target of selection as it has a high level of individual variability. However, effects of the molecular architecture of the genome on the mechanisms of sperm formation and their preservation after thawing have been poorly investigated. Here, we conducted a genome-wide association study (GWAS) for the sperm motility of cryopreserved semen in stallions of various breeds. Methods: Semen samples were collected from the stallions of 23 horse breeds. The following semen characteristics were examined: progressive motility (PM), progressive motility after freezing (FPM), and the difference between PM and FPM. The respective DNA samples from these stallions were genotyped using Axiom Equine Genotyping Array. Results: We performed a GWAS search for single nucleotide polymorphism (SNP) markers and potential genes related to motility properties of frozen-thawed semen in the stallions of various breeds. As a result of the GWAS analysis, two SNP markers, rs1141327473 and rs1149048772, were identified that were associated with preservation of the frozen-thawed stallion sperm motility, the relevant putative candidate genes being NME/NM23 family member 8 (NME8), olfactory receptor family 2 subfamily AP member 1 (OR2AP1), and olfactory receptor family 6 subfamily C member 4 (OR6C4). Potential implications of effects of these genes on sperm motility are herein discussed. Conclusion: The GWAS results enabled us to localize novel SNPs and candidate genes for sperm motility in stallions. Implications of the study for horse breeding and genetics are a better understanding of genomic regions and candidate genes underlying stallion sperm quality, and improvement in horse reproduction and breeding techniques. The identified markers and genes for sperm cryotolerance and the respective genomic regions are promising candidates for further studying the biological processes in the formation and function of the stallion reproductive system. Simple Summary: Artificial insemination is actively used in animal husbandry. It is important to know the quality of the sperm for artificial insemination. One of the indicators of sperm quality can be an assessment of energy metabolism, since energy is needed for sperm to move and fertilize the egg. We studied the respiration rate in spermatozoa of different animal species: bulls, stallions, boars, reindeer and roosters. To determine the production of energy (ATP), the substance 2.4-dinetrophenol (2.4-DNP) was used, which stopped the production of ATP. Semen was assessed before and after freezing. The evaluation showed the same response to the addition of 2.4-DNP to the semen of different species, as well as a sufficient relationship between the reaction of semen respiration to the addition of 2.4-DNP and the fertilizing ability of sperm. At the same time, no relationship was found between the respiratory rate and fertility. The 2.4-DNP test can be a suitable additional measure of sperm quality. To assess sperm quality, it is important to evaluate energy metabolism. The test substance 2.4-dinitrophenol (2.4-DNP) is an agent for destroying oxidative phosphorylation. 2.4-DNP shuts off the production of adenosine triphosphate (ATP) from oxidation and then, the respiration rate increases. If the respiratory chain is damaged, there is little or no response to adding 2.4-DNP. The aim of this study was to analyze the respiratory activity and oxidative phosphorylation in semen before and after freezing and compare the obtained data with the fertilizing ability of sperm. There was a reduction in sperm respiration rates in all species after thawing. The respiration of spermatozoa of boars, bulls, stallions, reindeers and chicken showed responses to 2.4-dinitrophenol. The only difference is in the strength of the response to the test substance. After freezing and thawing, respiratory stimulation by 2.4-DNP decreased. The results of our study show that respiration rate is not correlated with pregnancy rates and egg fertility. However, there was a high correlation between the stimulation of respiration by 2.4-dinitrophenol and pregnancy rates. The test for an increase in respiration rate after adding 2.4-dinitrophenol could be a suitable test of the fertilizing ability of sperm. [ABSTRACT FROM AUTHOR]

The formation and functioning of the animal reproductive system occurs as a result of the coordinated interaction of a wide range of genes. To search for causal mutations, work was carried out to search for polymorphic variants in the marker regions detected using GWAS. The four single-nucleotide substitutions in the exon of the GRM8 gene identified during the studies and the association of these SNPs with sperm quality was carried out. Semen from 22 stallions was collected. Sperm volume, concentration and progressive motility were assessed. Sequencing of the sections of the candidate GRM8 gene was carried out using an Applied Biosystems 3500 genetic analyzer. For the rs1138419111 genotype, no significant differences were found in the studied parameters. According to the identified single nucleotide substitution rs1147388106, the highest ejaculate volume was in stallions with the GG genotype (55.9±26.5 ml) compared to stallions with the GA genotypes (32.5±13.9 ml) and AA (18.0±33,6) (p < 0.05). When analyzing data on SNP rs395286150, stallions with a heterozygous CT genotype had the best sperm quality. Thus, the cell concentration was 317.0±66.5 million/ml in stallions with the CT genotype, 209.6±58.2 and 189.5±74.9 % with the CC and TT genotypes, respectively (P < 0.05). The progressive sperm motility of stallions with the CT genotype was 65.5±20.5% versus 48.7±22.0% in stallions with the TT genotype and 48.4±18.6% with CC. Analysis of data on SNP rs394524550 revealed a significant effect of the genotype on progressive motility. Stallions with the AG genotype had a progressive motility of 64.6±16.3%, and those with GG and AA 32.7±15.7 and 49.6±18.1%, respectively (P < 0.05). Thus, as a result four single nucleotide polymorphisms were identified in the exon of the GRM gene. Simple Summary: The cryopreservation and long-term storage of semen is one of the methods for accelerated improvement of the genetic qualities of animals. However, horse breeders prefer to use fresh or chilled semen, as the fertilizing capacity of frozen equine semen is much lower. It is important to find extenders, or a combination of extenders, that will improve semen survival after freezing. It is also important that the extender can be easily and simply prepared for use. Steridyl is a concentrate to which you just need to add sterilized water. This extender was developed for ruminants. In this study we tested Steridyl for freezing stallion semen. The motility, morphology, energy metabolism, DNA damage, and fertility of sperm frozen in Steridyl were evaluated. As a result, Steridyl was shown to be a good extender for equine semen freezing. The fertilizing ability of stallion sperm after freezing is lower than in other species. The search for the optimal extender, combination of extenders, and the freezing protocol is relevant. The aim of this study was to compare lactose-chelate-citrate-yolk (LCCY) extender, usually used in Russia, and Steridyl® (Minitube) for freezing sperm of stallions. Steridyl is a concentrated extender medium for freezing ruminant semen. It already contains sterilized egg yolk. Semen was collected from nine stallions, aged from 7 to 12 years old. The total and progressive motility of sperm frozen in Steridyl was significantly higher than in semen frozen in LCCY. The number of spermatozoa with normal morphology in samples frozen in LCCY was 60.4 ± 1.72%, and with Steridyl, 72.4 ± 2.10% (p < 0.01). Semen frozen in Steridyl showed good stimulation of respiration by 2.4-DNP, which indicates that oxidative phosphorylation was retained after freezing–thawing. No differences among the extenders were seen with the DNA integrity of spermatozoa. Six out of ten (60%) mares were pregnant after artificial insemination (AI) by LCCY frozen semen, and 9/12 (75%) by Steridyl frozen semen. No differences among extenders were seen in pregnancy rate. In conclusion, Steridyl was proven to be a good diluent for freezing stallion semen, even though it was developed for ruminants. [ABSTRACT FROM AUTHOR]
