PREMISE Plant domestication can be detected when transport, use, and manipulation of propagules impact reproductive functionality, especially in species with self-incompatible breeding systems. METHODS Evidence for human-caused founder effect in the Four Corners potato (Solanum jamesii Torr.) was examined by conducting 526 controlled matings between archaeological and non-archaeological populations from field-collected tubers grown in a greenhouse. Specimens from 24 major herbaria and collection records from >160 populations were examined to determine which produced fruits. RESULTS Archaeological populations did not produce any fruits when self-crossed or outcrossed between individuals from the same source. A weak ability to self- or outcross within populations was observed in non-archaeological populations. Outcrossing between archaeological and non-archaeological populations, however, produced fully formed, seed-containing fruits, especially with a non-archaeological pollen source. Fruit formation was observed in 51 of 162 occurrences, with minimal evidence of constraint by monsoonal drought, lack of pollinators, or spatial separation of suitable partners. Some archaeological populations (especially those along ancient trade routes) had records of fruit production (Chaco Canyon), while others (those in northern Arizona, western Colorado, and southern Utah) did not. CONCLUSIONS The present study suggests that archaeological populations could have different origins at different times-some descending directly from large gene pools to the south and others derived from gardens already established around occupations. The latter experienced a chain of founder events, which presumably would further reduce genetic diversity and mating capability. Consequently, some archaeological populations lack the genetic ability to sexually reproduce, likely as the result of human-caused founder effect.

Potato has about 100 related wild Solanum species growing naturally in the Americas. The US Potato Genebank aims to keep samples useful for research and breeding to improve the crop, often in the form of botanical seed families. A key component of genebank efficiency is assessing diversity within and among populations, and DNA marker sequence diversity is a powerful proxy for trait diversity. We previously reported on three factors which can cause under-estimation of heterozygosity: ascertainment, allele frequency, and ploidy bias. We here report, using GBS data for four diploid potato species, that average percent of apparent heterozygosity increases as data is more complete—the maximum difference was 2% heterozygotes when only a few individuals are called, to 36% when nearly all individuals were called. However, there was evidence that estimates of average heterozygosity based only on loci for which every individual has data can also be biased upward. Implausibly high levels of heterozygosity suggest non-segregating non-homologous SNPs, which occurred as 5–9% of all loci with complete data. We propose that best estimates of average heterozygosity in unselected seedlings should be based on loci with data for all samples after eliminating those loci that appear to be artificially fixed as heterozygous, which reduces observed heterozygote frequency by 16–26%. On that basis, the wild species examined have similar heterozygosity to the cultivated phureja.

Potato has about 100 related wild Solanum species growing naturally in the Americas. Solanum jamesii (jam), native to the southwest USA and Mexico, has many valuable traits for breeding, but making hybrids is extremely difficult. We investigated the approach of using the bridge species Solanum verrucosum (ver). A survey of all ver populations in the US Potato Genebank identified the best females. The standard bridge crossing technique using emasculation of ver and “mentor” double pollination after first pollinating with jam was very inefficient for most ver because: 1) emasculation depressed seedset in ver females, 2) despite very careful emasculation many accidental ver selfs resulted, and 3) mentor pollination produced many unwanted hybrids with the mentor pollen parent. We therefore produced populations of ver by backcrossing five generations into tuberosum cytoplasm. The BC5 lines are very vigorous in growth and flowering, but having very low male fertility and complete self-incompatibility they can be mass crossed with jam pollen without emasculation. Even with no mentor pollen, most pistils of verBC5 pollinated with jam produce a small fruit with an average of less than one normal seed. Resulting seedlings were confirmed ver-jam hybrids by DNA markers.

Potato cultivars are propagated by tubers which are sensitive to damage by freezing. Potato has about 100 related wild Solanum species growing naturally in the Americas. When tubers of a spectrum of these species were slowly cooled, most were killed at a few degrees below 0 C. Only tubers of Solanum jamesii, native to the southwest USA, remained firm, sprouted and grew after one week exposure to freezing temperatures as cold as -15C. Differential Thermal Analysis was performed to detect low temperature exotherms (LTEs) in jamesii and similar-sized tubers of cultivars Russet Burbank and Snowden. LTEs, indicating supercooling, were detected only in the jamesii tubers. Survival of potato tubers at freezing temperatures could be useful in several ways. Even non-lethal cold exposure of potato tubers at harvest reduces quality for eating and for seed, but storage at freezing temperatures without damage could help preserve tuber dormancy and limit diseases.

Crop genebanks keep a sample of the diversity in wild and cultivated germplasm. Knowing the structure of diversity within and among populations informs best decisions on efficient collecting, preservation and use. The US Potato Genebank has over 90 Solanum species, ranging in representation from only a few populations to several hundred. In a previous study we showed that populations of three species captured nearly all AFLP bands in polymorphic loci when represented by 100 populations that had been added over time to the genebank. In the current study we add analysis of two additional species and propose a simple metric, L100, to compare the expected marker diversity capture of 100 populations among different species. L100 detects when there is less population overlap and more rare markers, indicating that more populations are needed to capture the available diversity in a species.

AFLP markers were used to build a core subset in 144 accessions of the United States Potato Genebank (USPG) ex situ collection of the species formerly known as Solanum phureja (now reclassified as Solanum tuberosum L. subsp. andigenum). A core subset aims to sample the minimum number possible of non-redundant germplasm units capturing the maximum diversity of the entire collection. Characterization and trait evaluation of a core subset is expected to be easier and more effective when compared to the entire germplasm collection. In this study 1534 polymorphic AFLP markers were generated and demonstrated to be useful to determine genetic relationships in the materials used. All the accessions were clearly discriminated from each other with genetic similarity levels ranging from 62 to 89%, and no duplicate germplasm samples were detected. To create the core subset, the presence of the AFLP marker was considered as the genetic trait to retain and the selection of accessions was determined by their rankings on the number of most markers added to the core. The results of this selection process revealed that the 9 top-ranked accessions of phureja, when combined, achieved the benchmark of capturing 85% of the markers detected in the complete collection. Further selection to increase the percentage of marker capture added 17 accessions with unique marker contributions which resulted in a final core subset of 26 accessions capturing 96% of the marker diversity. A Principal Component analysis determined that spatial distribution of diversity in the core was a proportional sampling of the entire diversity of the collection validating the selecting approach used. A review of the public database for valuable traits for the USPG phureja germplasm revealed that the core subset also included accessions with desirable pest/disease resistances and stress tolerances (in 25 out of the 30 traits reported in the database). The USPG expects this core subset would create opportunities for facilitating evaluation and research by using a reduced set of germplasm units with nearly all the diversity of the entire set.

Ploidy manipulation and the resulting need for rapid ploidy screening can be important in potato research and breeding programs. We tested three predictors of ploidy, particularly seeking the quickest, simplest, and most reliable: Chloroplast number per guard cell (C#), guard cell length (GC), and pollen diameter (P), with a total of seven variations in methods of preparation. Time required for each preparation was assessed, and a panel of inexperienced volunteers compared these methods for accuracy using a standard set of coded samples of known ploidy. The common method of counting C# with iodine stain took longer and was no more accurate than observing C# or GC in tap water. GC from tape impressions of the underside of leaves was reliable and has the advantage of permanent slides for later reference. We recommend GC, whether in water, stained, or as tape impressions. GC is significantly different in diploids and tetraploids, but the distributions do overlap, so experience and care in selecting a representative sample of cells contributes to accuracy. The standard measurement of P after staining with aceto-carmine was faster to prep and just as reliable as epidermal methods for some technicians, even with no previous experience. Pursuit of ultra-simplified methods led us to measure P in tap water. Diameters of pollen in plain water are significantly larger, but only for living pollen, suggesting this method might also be developed into a rapid and reliable way to estimate pollen viability.

Potato has about 100 wild species relatives that are multiplied in the form of botanical seed populations by genebanks and distributed for use in research and breeding, so understanding factors that affect seed germination is valuable. This is especially true for problematic seedlots that have slow and low (“trickle”) germination even when subjected to germination techniques that give optimal results in most other seedlots in the genebank. Twelve seedlots with trickle germination even after at least 2 years of storage and 2 robustly-germinating positive control seedlots encompassing 9 species were germinated in petri plates, and subjected to day/night = 10 h/14 h = 20C/6C alternating temperature (AT) trials along with constant room temperature (RT) controls, in the spring of 2015 and 2016. When Relative Area Under the Germination Progress Curve (RAUGPC) was calculated, normal germinating seedlots’ germination in AT was not significantly different from RT control. Some species’ seedlots’ RAUGPC germination in AT was nearly zero, some were about 50% of RT control, and some in AT germination had RAUGPC double or even triple that of RT control. In a separate trial in 2017, four seedlots most responsive to AT were shown to germinate significantly better in constant cold than their RT controls, but not as well as seeds germinated under AT. Also in 2017, two seedlots known to be very dependent on gibberellin pretreatment were tested. RT germination without gibberellin had RAUGPC of only 13% of RT control, but AT without gibberellin pretreatment increased this to 65%, showing AT substantially substitutes for gibberellin pretreatment in these seedlots. AT could be a useful application to all seed germination assays in the genebank if it facilitated a better assessment of the potential viability of trickle-germinating seedlots. But one would need to identify and exclude those seedlots for which alternating temperature greatly depresses germination.

Potato has about 100 wild species relatives that are multiplied in the form of botanical seed populations by genebanks, and distributed for use in research and breeding, so factors that affect long term seed germination are of interest. In 1987 the US Potato Genebank conducted routine seed multiplication on 127 populations of 31 species as greenhouse-grown potted plants. Half of the parent plants in each population were given two doses of liquid fertilizer in addition to the standard slow-release granular fertilizer which had been mixed into the medium for all plants. Plants with this extra liquid fertilizer application grew larger, and produced significantly more fruit and seeds. However, seeds from parents fertilized extra did not have improved seed weight or germination after 26 years of storage. We conclude that optimizing the fertilizer regime used on parent plants, while doubling the average seed yield, does not result in higher quality seeds.

Potato has two wild relatives native to the USA, Solanum jamesii (jam) and S. fendleri (fen). Core collections are a useful tool for genebanks, identifying a ranked minimum number of samples that together encompass most of the total genetic diversity. With diversity measured as presence of AFLP bands, we made core collections for each species such that >90 % of diversity was captured in a minimum of populations. For fen, bulks containing about 25 % of populations accomplished that standard. For jam, a single “mega-population” at Mesa Verde, CO, consisting of many thousands of plants, was found to encompass 82 % of the AFLP bands detected in all samples across the entire USA range, and adding three more populations captured a total of >90 %. Core members for both jam and fen with the most diversity originated from the northern part of the range. That suggests that these areas merit more collecting. If resources for germplasm preservation and evaluation are limited, samples in the core collections should take priority.

The US Potato Genebank in Sturgeon Bay, Wisconsin, is the national germplasm collection for the world’s most important vegetable crop. It contains about 6,000 accessions of 100 species of tuber-bearing relatives of Solanum tuberosum. The potato of commerce is a clonal crop susceptible to many systemic pathogens, so the genebank routinely uses in vitro clonal maintenance and distribution for named cultivars. In vitro management is also the tool of choice for various breeding and genetic stocks of interest to breeders and researchers. Long-term backup cryopreservation of clones is done at the base collection in Fort Collins, Colorado. In vitro techniques also play an important role in virus elimination from clones. Recently, in vitro propagation has expanded to the temporary safekeeping of meristem propagules in antimicrobial medium during plant collecting expeditions in the southwest USA. The genebank’s mission includes promoting technology that supports expanded use of the germplasm, in particular, finding ways to overcome interspecific hybridization barriers. Thus, in vitro techniques such as pollen viability testing, ploidy manipulation, protoplast fusion, and embryo rescue have contributed technology for major advances in interspecific hybridization, utilization of noncommercial species, and introgression of genes from wild near-relatives. Finally, advancing in vitro technology holds promise as a tool for mass bioassay and selection of seeds, pollen, or somaclones for useful traits.

Knowing how genetic diversity is partitioned among and within wild potato species populations is important for efficient sampling for collection, preservation and evaluation. We sought to evaluate the effectiveness of SNPs for assessing germplasm by using the exact set of four model species previously evaluated by RAPDs. To avoid large numbers of SNP samples, population bulks of 25 plants were used, and original RAPD data was adjusted to match SNP genotype data. It was noted that especially for SNPs, it was necessary to examine only loci polymorphic within species to get a realistic view of genetic partitioning within species. This resulted in only a few hundred useful loci for some species. When considering among-population versus within-population partitioning of diversity, both SNPs and RAPDs distinguished the species as expected according to their known breeding system. Primitive wild species were confirmed as very heterogeneous within their populations. Both SNP and RAPD markers can be used to help breeders and genebank managers understand patterns and use in potato germplasm diversity.

Our previous study examining the effects of agrichemicals on the reproductive capacity of potato species revealed that the pesticide carbofuran negatively influenced flowering duration and pollen viability. These changes could limit reproductive ability non-randomly, modify allelic frequencies, and cause genetic drift. This study utilized Simple Sequence Repeat (SSR) markers to examine that possibility by assessing the genetic structure of progenies derived from potato populations exposed to two levels of carbofuran. A total of eight populations of five potato species (acl, buk, hcr, med and rap) were evaluated. An untreated, uncontaminated population was included as control for comparison. The results revealed that most of the 116 SSR allele frequencies assessed did not differ significantly between control and progenies of pesticide-contaminated parents. A few markers showed a significant frequency shift in some species, specifically buk and med. However, this study reveals that although the pesticide reduces reproduction in wild potato species populations, changes in population genetics are minor and do not significantly threaten diversity.

Gibberellin deficient mutants in potato have been published as ga1 (from S. tuberosum ssp. tuberosum and andigena), pito (from the tuberosum cultivar Pito), and ga2 (from the phureja haploid inducing clone “1.22”). We conducted crossing experiments to investigate genetic similarities. When the cultivar Pito was testcrossed with ga1 4x dwarfs, 1/6 of progeny were dwarf, exactly the expectation if Pito is duplex for ga1. Cultivars Pito and known ga1-carriers Superior and Eramosa all have ga1-carrier Early Rose as a common parent, supporting a hypothesis that the mutant described as pito is actually the same allele as ga1 by common descent. When reputed ga2 heterozygous carrier 1.22 was ga1 testcrossed, only one of 418 progeny was dwarf, suggesting that ga1 and ga2 are not allelic. We then attempted to create mutant ga2 in homozygous form. Since 1.22 does not self, it was outcrossed to individuals from S. bukasovii and S. microdontum populations devoid of dwarfs, then backcrossed to 1.22. No dwarfs were found in over 30,000 BC progeny of 44 different F1 hybrids. A possible explanation is that the ga2 mutant may be present only in a particular clonal sport of 1.22 in which it was reported, and not in the original 1.22 clone tested at the US Potato Genebank. Like pito, ga2 may be identical or allelic to ga1, since mutation from the normal to ga1 dwarf allele in 1.22 pollen was evident in the single dwarf recovered in the ga1 testcross progeny of 1.22. Some special genetic mechanism for relatively frequent mutation at this locus must be present, since normal shoots that occasionally arise on dwarf plants show that the recessive ga1 can also spontaneously revert to the dominant functional allele, as confirmed by testcrosses. However, this phenomenon does not appear to explain spontaneous clonal sports of cv Russet Norkotah selected for agronomic characters, which one might expect to result from the presence of more gibberellin-producing alleles.

A basic question in germplasm collecting is whether the in situ genetic diversity in a given geographic range has been adequately sampled. While one would ideally sample all diverse sites with appropriate habitat, there is usually a practical bias against visiting relatively inaccessible sites. For wild potato in the USA, mountain habitats often include easy access locations (near roads, usually at lower altitudes), and relatively remote locations (usually high altitude crests that can be accessed only by trail hiking and camping). This work used AFLP markers to compare three southeastern Arizona mountain ranges for which multiple "easy" and "remote" Solanum fendleri populations had been collected. Of the total markers detected, 24%, 6% and 3% were unique to the "remote" locations, and 3%, 21% and 34% were unique to "easy" locations. This case study demonstrates that populations at such locations are not identical, but the most unique alleles are sometimes captured at the remote location, sometimes at the easy. The practical conclusion is that both locations need to be sampled and compared empirically in the lab for unique allele richness to identify locations with highest priority for additional collecting.

Variation in tuber pH has been previously reported in cultivated potato and shown to be associated with economic traits. We here report a broad survey of potato germplasm for tuber pH, including wild Solanum species. Cultivar pH ranged from about 5.5 to 6.2, while six wild species were significantly lower. Solanum microdontum had the lowest pH average of the wild species examined at 5.17 with the lowest observed reading of 4.99. This species also exhibited a strong environmental effect, in which field tubers (5.28) were much lower than greenhouse tubers (5.63). Long storage generally did not have a significant effect on wild species tubers, except for S. jamesii, which reduced from 5.78 to 5.54. Various types of sampling were tested. Skins were generally lower than flesh, with a high correlation between the tissues. Samples that were frozen, cooked, or reconstituted from dried powder had different absolute pH values, but these were highly correlated, so germplasm would be ranked for pH similarly regardless of method. Although not tested here, physiological links between pH and economic traits like disease resistance, nutrition and tuber quality are plausible. If so, pH testing might serve as a rapid and inexpensive screening tool in breeding programs. The association of pH and useful traits might also provide clues for exploring underlying genetic and physiological mechanisms in potatoes.

Some tuber-bearing wild potato species are reportedly higher in potential health-promoting traits, such as antioxidant activity (AOA) and total phenolic content (TP), than commercial cultivars; therefore, they could be used as parental material in breeding for high AOA and TP. However, using wild species might result in progenies that are toxic for human consumption because of the presence of high total glycoalkaloids (TGAs) and other unknown compounds. Therefore, wild potato accessions should be screened for cytotoxicity before their introduction into breeding programs. The objective of this study was to investigate antiproliferative activity and cytotoxicity of tuber extracts from 15 Solanum jamesii accessions on human HT-29 colon and LNCaP prostate cancer cell lines in vitro. Also, correlations among AOA, TP, TGA, and antiproliferative activity were determined. The tuber extracts significantly inhibited proliferation of HT-29 and LNCaP cell lines and were not cytotoxic to the cells compared to the control (DMSO). The antiproliferative activity exhibited by tuber extracts was not due to necrosis, because the amount of lactate dehydrogenase (LDH) released from cells incubated with the extracts was not significantly different from that released from cells incubated without extracts (control). Colon cancer cells were more responsive to tuber extract treatment than prostate cancer cells. In both HT-29 and LNCaP cells, there were no observable significant correlations between antioxidant activity (DPPH and ABTS) and inhibition of cell proliferation or between TP and cell proliferation inhibition. Also, glycoalkaloids did not exhibit significant correlations with the inhibition of cancer cell proliferation. Findings of this study show that S. jamesii accessions probably pose no cytotoxic effects when used as parental material in improving the nutritional value of potato cultivars. Correlation results, along with cell proliferation data, suggest that not only the compounds measured in this study but also other bioactive compounds present in the matrix acting additively or synergistically may be more responsible for the antiproliferative effects of potato tuber extracts than higher concentrations of a single or group of compounds.

Although the potato of commerce, the autotetraploid Solanum tuberosum is a clonal crop, genebank maintenance of wild and cultivated relatives is commonly done as populations of true seeds. In order to help equalize gamete contribution and prevent genetic drift over seed regenerations, an equal number of seeds from each mother plant may be bulked as a source of parents for future regenerations. To determine if this extra sample is beneficial, we selected two S. andigena populations with great variation of seed production among mother plants. Seed production of each mother plant was recorded over 12 standard seed regenerations and genetic indicators were developed in the form of 105 polymorphic RAPD bands. For each polymorphic band, the variation of seed production among mother plants was used to estimate the potential impact on band phenotypic frequency due to an over-all seed bulk. Most bands were fixed or nearly fixed within at least one of the populations, such that the variation in maternal seedset observed would not have put them at signficant risk of being lost in an over-all bulk. For balanced bulks to provide a significant preservation advantage, a band must be rare in all populations and associated with mother plants that produce significantly fewer seeds. Of over 100 loci assessed here, none met these conditions. We therefore conclude that the duplication of documentation and storage space invested in making balanced seed bulks probably return little genetic benefit to the genebank.

Aim To assess evidence for geographical and environmental range expansion through polyploidy in wild potatoes ( Solanum sect. Petota ). There are diploids, triploids, tetraploids, pentaploids and hexaploids in this group. Location Wild potatoes occur from the south-western USA (Utah and Colorado), throughout the tropical highlands of Mexico, Central America and the Andes, to Argentina, Chile and Uruguay. Methods We compiled 5447 reports of ploidy determination, covering 185 of the 187 species, of which 702 determinations are presented here for the first time. We assessed the frequency of cytotypes within species, and analysed the geographical and climatic distribution of ploidy levels. Results Thirty-six per cent of the species are entirely or partly polyploid. Multiple cytotypes exist in 21 species, mostly as diploid and triploid, but many more may await discovery. We report the first chromosome count (2 n = 24) for Solanum hintonii . Diploids occupy a larger area than polyploids, but diploid and tetraploid species have similar range sizes, and the two species with by far the largest range sizes are tetraploids. The fraction of the plants that are polyploids is much higher from Mexico to Ecuador than farther south. Compared with diploids, triploids tend to occur in warmer and drier areas, whereas higher-level polyploids tend to occur in relatively cold areas. Diploids are absent from Costa Rica to southern Colombia, the wettest part of the group’s range. Main conclusions These results suggest that polyploidy played an important role in this group’s environmental differentiation and range expansion.

Malnutrition is one of the world’s largest health concerns. Folate (also known as vitamin B9) is essential in the human diet, and without adequate folate intake, several serious health concerns, such as congenital birth defects and an increased risk of stroke and heart disease, can occur. Most people’s folate intake remains sub-optimal, even in countries that have a folic acid food fortification program in place. Staple crops, such as potatoes, represent an appropriate organism for biofortification through traditional breeding based on their worldwide consumption and the fact that modern cultivars only contain about 6% of the daily recommended intake of folate. To start breeding potatoes with enhanced folate content, high folate potato material must be identified. In this study, 250 individual plants from 77 accessions and 10 Solanum species were screened for their folate content using a tri-enzyme extraction and microbial assay. There was a 10-fold range of folate concentrations among individuals. Certain individuals within the species Solanum tuberosum subsp. andigenum, Solanum vernei and Solanum boliviense have the potential to produce more than double the folate concentrations of commercial cultivars, such as Russet Burbank. Our results show that tapping into the genetic diversity of potato is a promising approach to increase the folate content of this important crop.

Because of their adaptation to a diverse set of habitats and stresses, wild species of cultivated crops offer new sources of genetic diversity for germplasm improvement. Using an Infinium array representing a genome-wide set of 8303 single nucleotide polymorphisms (SNPs), phylogenetic relationships and allelic diversity were evaluated within a diversity panel of germplasm from Solanum sect. Petota. This panel consists of 74 plant introductions (PIs) representing 25 species and provides a diverse representation of tuber-bearing Solanum germplasm. Unlike other molecular markers, genome-wide SNPs have not been widely implemented in potato. To determine relatedness between current species classifications and SNP-based genetic distances, a phylogeny was generated based on random individuals from each core collection PI. With few exceptions, SNP-based estimates of species relationships revealed general agreement with the existing taxonomic grouping of species in Solanum sect. Petota. Genotype comparisons between the Solanum sect. Petota diversity panel and a panel of 213 tetraploid cultivars and breeding lines indicated a greater extent of diversity between populations of native Andean landraces than among modern cultivated varieties. Comparison of SNP allele frequencies between the Solanum sect. Petota panel and tetraploid cultivars identified loci with extreme divergence between cultivated potato and its tuber-bearing relatives. Several of these loci are associated with genes related to carbohydrate metabolism and tuber development, suggesting potential roles in potato domestication. The Infinium SNP data offer a new taxonomic view of potato germplasm, while further identifying candidate alleles likely to differentiate wild germplasm and cultivated potato, possibly underlying key agronomic traits.

A basic goal of the U.S. Potato Genebank (USPG) and others is to test assumptions about the stability of genetic diversity in their collections. For example, when heterogeneous seed populations are regenerated, one assumes that using a careful regeneration protocol will result in very little diversity loss in the progeny. However, even the most careful mating scheme cannot prevent genetic selection if it happens earlier—when seedlings are transplanted to become the seedincrease parents. The objective of this work was to assess the prospect of losing diversity at the seedling transplant step. Seeds of a total of 245 original seedlots (from the wild) of 11 inbreeding species [Solanum acaule Bitter, albicans (Ochoa) Ochoa, demissum Lindl., etuberosum Lindl., fendleri A. Gray, polyadenium Greenm., palustre Poepp. ex Schltdl., polytrichon Rydb., papita Rydb., stoloniferum Schltdl. & Bouche´, and verrucosum Schltdl.] were sown. The most vigorous, uniform ‘‘normal’’ (N) seedlings were distinguished from any that were small (S) or otherwise would likely be avoided when transplanting the parent plants to be used for seed increase. Bulks of N and S types within seedlots were compared by random amplified polymorphic DNA (RAPD). Genetic similarity (GS) was calculated as average percentage matching band status at RAPD loci. About 25% of the seedlots exhibited S types, but in no case were these significantly different from their N sibs (none with lower than GS 5 98%). These results suggest that inbred species’ original seedlots are homogeneous and not subject to unwanted seedling selection at transplanting, despite random environmental effects that sometimes produce S seedlings.

Most wild potato germplasm in genebanks is collected, preserved, and evaluated as botanical seed populations that may be highly heterozygous and heterogeneous. However, some species are selfers so potentially very homozygous, perhaps also homogeneous. Intrapopulation heterogeneity increases sampling error that can undermine consistency in seed regeneration in the genebank, screening results, germplasm collecting, and estimates of taxonomic relationships. Thus, knowledge of genetic heterogeneity (GH) can predict the need to commit more resources for larger sample sizes or replication when populations of a given species are being regenerated, evaluated, collected, and classified. This study investigated withinpopulation GH in 32 potato populations comprising four different breeding systems observed inSolatium species:S. fendleri (2n=4x=48, disomic selfer),S. jamesii (2n=2x=24, outcrosser),S. sucrense (2n=4x=48, tetrasomic outcrosser), andS. verrucosum (2n=2x=24, selfer). RAPD markers were used to estimate heterogeneity among 24 individuals per population. Populations ofS. verrucosum were quite homogeneous with an average GH of 6.0%. Similarly low heterogeneity was detected among the eight populations ofS. fendleri (average GH=7.1%). In contrast,S. jamesii andS. sucrense had a much higher GH of 29.4% and 44.1%, respectively. These results demonstrate and quantify the great difference in intrapopulation heterogeneity among wild potato species. Calculations based on intrapopulation heterogeneity indicate that samples should be composed of 25 to 30 random plants for low sample variation that is uniform for all species.

In previous work at the US Potato Genebank, RAPDs were used to detect large differences between genebank-conserved samples and their correspondingin situ populations re-collected from the wild. This work investigates one possible explanation for these differences: a large genetic change in the sample when it undergoes “domestication” by a forced sexual seed increase and subsequent adaptation to cultivation in the genebank. However, when 11Solanum fendleri and 17S. jamesii populations were collected from the wild and compared to their sexual progeny generated at the genebank, no significant differences in RAPD finger-prints were detected. These results show that theory of “genebank domestication” is not supported. However, when plants of one pair of populations were tested individually, the seed increase population was significantly more heterogeneous than its clonally collectedin situ parents. Thus, while genebank populations have the same genes as their wild counterparts, they may contain genotypes not present in the wild.

The use of genetic resources could be more effective and efficient if we were able to predict the presence or absence of useful traits in different populations or accessions. We analyzed the extent to which taxonomic, geographic and ecological factors can predict the presence of frost tolerance in wild potatoes. We used screening data for 1646 samples from 87 species that had been collected in 12 countries in the Americas. There was a strong association of frost tolerance with species and to a lesser extent with taxonomic series. There was significant geographic clustering of areas with wild potatoes with similar levels of frost tolerance. Areas with a high level of frost tolerance are the central and southern Peruvian Andes, the lowlands of Argentina and adjacent areas, and a small area in the central Chilean Andes. There is a greater chance of finding wild potatoes with high levels of frost tolerance in areas with a yearly mean minimum temperature below 3 °C than there is in warmer areas. However, temperature is only a weak predictor of frost tolerance. Temperature data alone did not predict observed frost tolerance in eastern Argentina/Uruguay and falsely predicted it in the southwestern United States. Because many wild potato species occur over small areas, taxonomic, ecological, and geographical factors are difficult to disentangle.

Micronutrient deficiency, also known as the hidden hunger, affects over two billion people worldwide. Potato is the third most consumed food crops in the world, and is therefore a fundamental element of food security for millions of people. Increasing the amount of micronutrients in food crop could help alleviate worldwide micronutrient malnutrition. In the present study, we report on the identification of single nucleotide polymorphism (SNP) markers associated with folate, an essential micronutrient in the human diet. A high folate diploid clone Fol 1.6 from the wild potato relative Solanum boliviense (PI 597736) was crossed with a low/medium folate diploid S. tuberosum clone USW4self#3. The resulting F1 progeny was intermated to generate an F2 population, and tubers from 94 F2 individuals were harvested for folate analysis and SNP genotyping using a SolCap 12K Potato SNP array. Folate content in the progeny ranged from 304 to 2,952 ng g-1 dry weight. 6,759 high quality SNPs containing 4,174 (62%) polymorphic and 2,585 (38%) monomorphic SNPs were used to investigate marker-trait association. Association analysis was performed using two different approaches: survey SNP-trait association (SSTA) and SNP-trait association (STA). A total of 497 significant SNPs were identified, 489 by SSTA analysis and 43 by STA analysis. Markers identified by SSTA were located on all twelve chromosomes while those identified by STA were confined to chromosomes 2, 4, and 6. Eighteen of the significant SNPs were located within or in close proximity to folate metabolism-related genes. Forty two SNPs were identical between SSTA and STA analyses. These SNPs have potential to be used in marker-assisted selection for breeding high folate potato varieties.

Greenhouse assays were carried out to characterizeSolanum accessions previously reported to be late blight resistant and to identify individuals within the accessions with high levels of resistance to late blight. Foliage of wildSolanum species accessions or hybrid cultivated potato x wild species (diploid or tetraploid) from Mexico, Russia, or South America was inoculated withPhytophthora infestans (US-8) and was scored for severity of foliar symptoms at 7 and 14 days after inoculation (DAI). Mexican accessions (S. brachycarpum,S. pinnatisectum,S. guerreroense, andS.fendleri) were significantly more resistant (1.1% infection at 14 DAI) than either Russian (59.1% infection at 14 DAI) or South American (53.4% infection at 14 DAI) accessions. Moreover, the genotypes within the Mexican accessions tended to be more uniformly resistant whereas genotypes within the Russian and South American accessions tended to segregate for resistance. The more resistant genotypes of Russian and South American accessions (176/546) were retested, and 56 genotypes were identified as having potential for use in potato breeding programs for resistance to late blight. Fifty percent of the selected genotypes were fromS. microdontum accessions PI498124, PI595510, and PI595511. The potential of these species for incorporation into breeding programs for late blight resistance is discussed.

Populations of two wild potato species known to have high levels of green peach aphid, PLRV, and PVY resistance (Solanum polytrichon PI 184170 andS. stoloniferum PI 160226) were assessed for heterogeneity of RAPD markers among plants within these populations and among other populations of the same species. These species are reputed selfers, so were suspected of being relatively inbred. About 75% of the bands segregating within their species were fixed within these populations, and more than 90% of the total bands observed within these populations were fixed. These levels of homogeneity were not significantly different from those of an inbred population ofS. albicans known to be extremely homogeneous. It can be expected that populations such as these, that are relatively homogeneous, are also more likely to be homogeneous for specific traits of interest. Thus, there may be less need for rigorous fine screening of these materials at the genotype level before making breeding crosses.

Two major components of frost resistance are freezing tolerance in the nonacclimated state (growing in normal condition) and capacity to cold acclimate (increase in freezing tolerance upon exposure to chilling temperatures). In addition to these two major components, numerous factors contribute to frost survival. Although the rate of cold acclimation and deacclimation have been recognized as important factors contributing to frost survival, very little information about them is available. Our objective was to determine if there is variability in the rate of cold acclimation and deacclimation among tuber-bearing wild potato species: S. acaule Bitter, S. commersonii Dunal, S. megistacrolobum Bitter, S. multidissectum Hawkes, S. polytrichon Rydb., S. sanctae-rosae Hawkes, and S. megistacrolobum subsp. toralapanum (Cárdenas & Hawkes) Giannattasio&Spooner. Relative freezing tolerance of these species was measured after 0, 3, 6, 9 and 12 days of cold acclimation and after 12 and 24 hours deacclimation. Our results showed there were differences in the rates of cold acclimation and deacclimation among these species. With respect to the rate of acclimation we found these species can be divided into four groups: (i) early; (ii) late acclimators; (iii) progressive acclimators, and (iv) nonacclimators. Likewise, a wide range of cold deacclimation behavior was found. Some species showed as low a loss of 20% of their freezing tolerance, others showed as much as >60% loss after 12 hours of deacclimation. Significant deacclimation was observed in all cold acclimating species after 1 day. These results demonstrate that the rates of cold acclimation and deacclimation were not necessarily related to the cold acclimation capacity of a species. Rapid acclimation in response to low temperatures preceding a frost episode and slow deacclimation in response to unseasonably warm daytime temperatures could be advantageous for plants to survive frost events. Thus, in addition to nonacclimated freezing tolerance and acclimation capacity, it would be very desirable to be able to select for rapid acclimation and slow deacclimation abilities. Results demonstrate that variability for these two traits exists in Solanum L. (potato) species.

Genetic characterization of germplasm is important for setting objective guidelines for conservation. One common problem found in genebanks is determining the value of populations with insufficient or unreliable data regarding their geographic origin. In this study, a genetic analysis based on RAPD markers was conducted to characterize a `mystery' population of Solanum sucrense, a polysomic tetraploid potato (2n=4x=48), for which adequate documentation was lacking. The comparative analysis of genetic similarities between this mystery population and each one of 30 other S. sucrense populations in the genebank revealed that all populations within this species, including the mystery population, are significantly different from being duplicates, and are therefore worthy of separate conservation. RAPD markers also distinguished the mystery population from closely related tetraploid species S. oplocense, S. gourlayi and S. tuberosum ssp. andigena, suggesting that it is also not a duplicate of a population of these species. If RAPDs can clearly differentiate populations within highly heterogeneous tetraploids like S. sucrense, they should be generally useful for determining germplasm organization within potato species.

The expression of freezing tolerance was examined in interspecific F1 and somatic hybrids of potatoes using 20 species and 34 different combinations between hardy and sensitive species. In the field, the frost tolerance of hybrids resembled either that of the hardy parent, the sensitive parent, or the parental mean, depending on the species combination and the genomic ratio (ratio of the number of sets of chromosomes contributed from each parent). Similar phenomena were observed when the non-acclimated freezing tolerance (NA) and the acclimation capacity (ACC) (two independent genetic components of freezing tolerance) were evaluated separately under controlled environments. In general, the expression level of freezing tolerance was higher in hybrids with more genomes contributed from the hardy parent than from the sensitive parent. In addition, the effectiveness or combining ability of genes conferring freezing tolerance from the hardy species also showed some influence on the expression of freezing tolerance. All three parameters, namely NA, ACC and acclimated freezing tolerance (AA) (NA plus ACC), were significantly correlated to the frost tolerance exhibited in the field. This indicates that the controlled freezing test used in this study could provide a good estimate of field performance. The implications of these results in breeding for freezing tolerance in potatoes are discussed.

Modern potato cultivars (Solanum tuberosum L.) require high rates of fertilizer nitrogen (N). This practice is costly and can pose a serious threat to surface and groundwater. Previous evaluation of wild potato germplasm demonstrated the existence of species capable of producing high total biomass under low N conditions, with the ability to make maximum use of added N. Therefore, a two-year field experiment was conducted in 1994 and 1995 to investigate the response of selected wild potato accessions and their hybrids with the haploid USW551 (USW) to low and high N environments. The haploid USW and cultivars Russet Burbank, Red Norland, and Russet Norkotah were also included in the study. Uniform propagules and seedlings from the variousSolanum species were transplanted to a Hubbard loamy sand (Udic Haploboroll) at Becker, Minn. and were subjected to two N treatments: 0 and 225 kg N ha-1. At harvest, total dry biomass of wild and hybrid potato germplasm was equal to or higher than that of the cultivars. However, cultivar biomass partitioning was 1% to roots, 15% to shoots, 0% to fruits, and 84% to tubers, whereas wild potato species partitioned 18% to roots plus nontuberized stolons, 52% to shoots, 23% to fruits, and only 7% to tubers. Hybrids were intermediate, allocating 9% of their biomass to roots plus nontuberized stolons, 39% to shoots, 14% to fruits, and 38% to tubers. Nitrogen use efficiencies for many of the species and crosses were comparable to that for Russet Burbank and greater than those for Red Norland and Russet Norkotah. Of the wild species tested,S. chacoense accessions had the highest biomass accumulation and N uptake efficiencies and may be the best source of germplasm for improving NUE in a potato breeding program.

The expression of freezing tolerance was characterized in interspecific somatic hybrids between S. tuberosum (tbr) and two cold-hardy wild species, S. brevidens (brd) and S. commersonii (cmm). The nonacclimated freezing tolerance (NA) and acclimation capacity (ACC, increase in freezing tolerance in response to low nonfreezing temperature), two main genetic components of freezing tolerance, were evaluated separately. In contrast to cmm, which exhibited excellent NA and ACC, the freezing tolerance of brd was mainly due to ACC. However, the ACC of brd was only moderately expressed in the somatic hybrids. The NA of cmm was also suppressed in combination with tbr genomes. However, with acclimation, some of the tbr (+) cmm somatic hybrids achieved freezing tolerance comparable to pure hardy species such as brd used in this study. Analysis of chloroplast DNA type by RFLP markers revealed no significant difference in ACC between somatic hybrids carrying chloroplasts from either tbr or cmm. The reasons for the reduced expression of freezing tolerance from either the brd or cmm parent and the utilization of these somatic hybrids in breeding programs are discussed.

A representative sample of the US potato germplasm collection was screened in the greenhouse for ability to accumulate tuber calcium. Germplasm with very high and very low calcium accumulation capacity has been identified by progressively screening for extremes among species, among populations (accessions), and finally among genotypes (i.e., fine screening). Among accessions, the bestS. gourlayi andmicrodontum populations averaged more than six-fold greater tuber calcium than the poorestS. kurtzianum population. When screening was performed at the genotype level, the best individuals ofgourlayi andmicrodontum had over eight-fold greater calcium than the poorestkurtzianum individual. When grown in the field, calcium levels ofS. microdontum selections were confirmed to be significantly higher thantuberosum cultivars. When selected genotypes were intercrossed and their progeny were compared to their original populations, mean tuber calcium was significantly changed in the intended direction of selection forS. kurtzianum (made lower) andS. microdontum (made higher). These stocks are expected to be useful tools for study of the genetic and physiological bases of tuber calcium accumulation, refining screening methods, and breeding improved cultivars.

Exotic potato germplasm may contain useful traits for improving nitrogen (N) use efficiency in cultivated potatoes (Solanum tuberosum L.). The objective of this study was to evaluate a “mini” core collection of wild germplasm for biomass production, N uptake, and N use efficiency. A field study was conducted during the 1993 growing season on a loamy sand soil at Becker, Minnesota. Uniform transplants for 39 wild accessions and 3 varieties were grown under greenhouse conditions, and after 40 days, they were transplanted to the field. Plants were subjected to two N treatments, 0 and 225 kg N ha1, replicated three times in a split-plot design. Plant parts were collected separately 111 days after transplanting, dried, weighed, and analyzed for N concentration. Nitrogen rate and potato species had significant effects on total dry weight, N content, and tissue N concentration. The regression coefficient of N concentration on total dry weight was very low (r=0.22, NS), whereas the regression coefficient of total N content on total dry weight was high (r=0.94, P>0.001). At both low and high N environments, Russet Burbank had greater dry weight than all the accessions. Several wild accessions, especiallyS. chacoense, S. commersonii, S. kurtzianum, S. microdontum, andS.phureja, had equal to or greater dry weights than Russet Norkotah or Red Norland. For N uptake efficiency, 2,7, and 20 accessions were ranked better than Russet Burbank, Russet Norkotah, and Red Norland, respectively. Recovery of soil applied N by the varieties ranged from 16 to 36%; the top seven wild accessions recovered between 27 and 49%. Based on plant growth without added N fertilizer and relative response to N fertilizer, the species were categorized into four relative N use efficiency groups: good foragers with good response to N, poor foragers with good response to N, good foragers with poor response to N, and poor foragers with poor response to N.

The main objective of the present study was to investigate the influence of supplemental calcium fertilization on the levels of freezing tolerance in different wild potato species. For this purpose, the freezing tolerance of a broad spectrum of theSolatium taxa was evaluated with and without supplemental calcium fertilization. Previous studies have shown that there is a large variation in the calcium accumulation capabilities among species and among various accessions within species. While this study confirms such variation, no direct relationship between the leaf calcium content and the improvement in freezing tolerance was found. Nevertheless, overall 45% of the species accessions had significant higher leaf calcium content and 52% of the species accessions showed a significant drop in the frost score average at the p

This study was conducted to investigate the utility of systematic screening at the genotype level withinSolanum accessions highly resistant to the Colorado potato beetle. Evaluations of clonally replicated genotypes showed that most accessions reported to be uniform when screened as populations contained small but significant variation among genotypes for resistance to oviposition, larvae, and defoliation, differences for numbers of larvae being most common. Adult counts and percentage defoliation were not as useful in evaluating among-genotype variability in beetle resistance. Genotypes ofS. pinnatisectum WRF 343 andS. tarijense PI 473227 were the most uniformly and highly resistant to Colorado potato beetle. Genotypes of S.berthaultii PI 473331,S. chacoense PI 473405, andS. tarijense PI 473336 were moderately to highly resistant, and genotypes ofS. bukasovii PI 473494 andS. canasense PI 230511 were uniformly susceptible to Colorado potato beetle. Nonparametric correlation analyses indicated that number of egg masses, small larvae, large larvae, and defoliation scores were positively correlated, negatively correlated, or not correlated, depending on the species. One generation of selection attempting to segregate resistance and susceptibility in nearly uniform and highly resistantS. pinnatisectum WRF 343 resulted in “divergent” populations that could not be distinguished from each other or the base population. Thus, through genotype (“fine”) screening and selection, we showed that some existing populations are virtually pure for extreme resistance. Use of individuals from such families would make screening breeding populations more efficient, and reduce the risk of losing resistance genes that have non-dominant effects. Fine screening, recurrent selection, and maintenance of such elite populations is recommended as an extension of population-based evaluation usually done by genebanks.

The purpose of this study was to investigate differences in stomatal index among backcross progeny ofSolanum commersonii (freezing tolerant) andSolanum cardiophyllum (freezing sensitive) to assess the feasibility of using this trait as a marker for screeningSolanum populations for freezing tolerance. Measurements were taken on three terminal fully-expanded leaflets per genotype by completing microscopic examination of epidermal impressions made in partially dissolved cellophane tape. Freezing tolerance was estimated in parallel studies on the same plant material. Values of SI were significantly greater (Fisher T-test, 0.05) for theS. cmm. group (parent + backcross progeny) compared with theS. cph. group. Stomatal index of the F1 was significantly greater than SI ofS. cph. parents and similar toS. cmm. parents. Values of SI for both backcross progenies were greater than parental values. Non-acclimated relative freezing tolerance values were in the following order:S. cmm. parents >S. cmm. backcrosses > F1 >S. cph. back-crosses >S. cph. parents. Stomatal index values followed a similar pattern with the exceptionS. cmm. backcross >S. cmm. parents. These data suggest: a) increased SI is inherited as a dominant trait, b) SI may be a useful screening marker in breeding programs interested in improving freezing tolerance.

Potato is one of the most important world vegetables with respect to value of production and nutritional impact. Frost susceptibility reduces or precludes production on millions of hectares worldwide. In contrast to disease and pest problems, cultural or chemical treatments are not likely to be helpful in combating frost, so genetic improvement must be sought. Fortunately, wild relatives of the cultivated potato have been shown to exhibit frost tolerance far superior to that of cultivated species. In this research, over 2,600 accessions representing 101 species in the US potato collection at Sturgeon Bay, Wisconsin were visually screened after natural frosts in a uniform field in 1992. This provided a more comprehensive survey of germplasm accessions, further characterized species’ hardiness and intraspecific variation. The most hardy species wereSolanum acaule, S. albicans, S. commersonii, andS. demissum. An additional previously unreported extremely frost hardy species (Solanum paucissectum) was discovered. This information should expand the insights and germplasm options available to those intent on breeding desirable varieties with high levels of frost hardiness.

Heat stress limits botanical seed production by inhibiting flowering, pollen production and pollen viability. Three accessions (PIs) of each of 23 diverseSolanum species were screened for stability of fertility parameters under heat stress. Seedlings were grown to flowering in temperate conditions (16–25 C), then treated with three weeks of heat stress of up to 45 C for 4–6 hours per day at midday with nighttime lows of 18 C, and compared to temperate-grown controls. A highly significant effect of species, temperature, and their interaction was detected for flowering. This means species were inherently different and heat had a general depressing effect, but the degree of that effect varied among species. While most species had little flowering in the hot house,S. commersonii andmicrodontum flowered significantly more there than in the temperate house. Of species which flowered in hot conditions, some shed no pollen and some had good shed of mostly dead pollen, but onlyS. commersonii, jamesii, kurtzianum andmegistacrolobum had good flowering, pollen shed and viability. This work is expected to contribute to the study of the genetic and physiological bases of heat stress fertility, true potato seed (TPS) breeding, and possibly improvement of tomato production under heat stress.

We conducted a joint Colombia/United States/Venezuela wild potato (Solanum sect. Petota Dumort.) germplasm collecting expedition in Colombia from June 27–August 24, and in Venezuela from August 17–September 15, 1992. The goals of the expedition were to collect germplasm and study the species boundaries of all of the 23 Colombian and Venezuelan taxa accepted by current taxonomists. We made 128 collections of 16 of these taxa, 96 as true seed collections. We collected the first available germplasm collections of S. cacetanum, S. cuatrecasasii, S. estradae, S. lobbianum, S. orocense, S. paramoense, and S. sucubunense, and obtained germplasm collections of S. neovalenzuelae and S. pamplonense as a germplasm exchange from the Colombian national germplasm collection. We had problems identifying some of our collections, and currently are investigating them for species status and interrelationships. We summarize the state of germplasm collections for Colombia and Venezuela, provide our field data regarding the taxonomy of Colombian and Venezuelan wild potatoes, and provide recommendations for future collecting.

The inheritance of endosperm balance number (EBN), a genetic, dose-dependent crossability system functioning in tuber-bearing Solanum (potato) species, was investigated for certain wild potato species having an EBN equal to one half of their ploidy. The EBN of Solanum acaule, a disomic 4(2EBN) South American species, was investigated by producing F1 and F2 hybrids with artificial 4x(2EBN) S. commersonii. This allowed assessment of recombination among the two genomes of disomic S. acaule and that of S. commersonii. When crossability of the hybrids with 1EBN, 2EBN and 4EBN standards was tested, no variation for EBN was detected. The apparent lack of recombination and segregation for EBN in these hybrids indicates that the genomes of S. acaule and S. commersonii carry EBN in a genetically-similar way. Combined with previous reports, these data indicate that the inheritance of EBN is similar in widely-separated taxa from South America and Mexico.

The United States Potato Introduction Station Herbarium at Sturgeon Bay, Wisconsin, serves the National Research Support Program-6 (NRSP-6; formerly known as the Inter-Regional Potato Introduction Project IR-1). NRSP-6 is part of the US germplasm system and is the sole gene bank for wild and cultivated potatoes in the USA. The gene bank and herbarium are devoted entirely to wild and cultivated potatoes (Solanum sect. Petota). NRSP-6 is charged with the introduction, preservation, classification, evaluation and distribution of potato germplasm worldwide. Recent expeditions and grants for upgrading the herbarium and associated facilities have provided a valuable new international resource for researchers in sect. Petota.

Frost or winter survival is regarded as a complex trait with polygenic inheritance. Two major components of this survival in crop plants are freezing tolerance in the nonacclimated state and cold acclimation capacity. To date researchers have not distinguished the two components as separate heritable traits. The mode of inheritance of these two traits was investigated in F1 and backcross populations of two wild diploid potato species (Solanum commersonii and Solanum cardiophyllum) exhibiting extremes of freezing tolerance and acclimation capacity. Precise assessment of these two traits allowed distinction of small but significant differences among genotypes. The two traits were not correlated in segregating populations, suggesting independent genetic control. Analyses of generation means indicate that all of the variance for acclimation capacity and a major proportion of the variance for the nonacclimated freezing tolerance can be best explained by an additive-dominance model with both traits being partially recessive. Recovery of parental phenotypes in limited populations suggests that both traits are controlled by relatively few genes. To our knowledge this is the first study demonstrating independent genetic control of the two main traits associated with frost or winter survival. Our results show that it should be possible to incorporate these traits from wild germ plasm into cultivated crop plants by independent selection. These results help explain the lack of progress in improving winter survival through field selection. Furthermore, our study demonstrates relative simplicity of the inheritance of cold acclimation, thus providing avenues for understanding the link between biochemical and genetic aspects of low-temperature stress in crop plants.

La resistencia de especies silvestres de Solanum a Phytophthora infestans (Mont). de Bary es multigénica. Para cuantificar algunas respuestas enzimáticas al ataque del patógeno, tres especies silvestres resistentes y una cultivada, susceptible, de Solanum, se expusieron a la infección natural por P. infestans en el Valle de Toluca, México. No se presentó la enfermedad en las especies silvestres (Solanum microdontum Bitt, S. demissum Buk. y S. pinnatisectum Dun.). En la primera se obtuvieron correlaciones significativas (P= ¿0,05) entre fenoles totales (FEN) y fenilalanina-amonio-liasa (PAL) (r= 0,93); peroxidasa (POX) y superóxido dismutasa (SOD) (r= 0,97); y PAL/POX (r= 0,79). En S. demissum las correlaciones con significancia estadística fueron entre FEN/PAL (r= 0,94) y POX/SOD (r= 0,88). En S. pinnatisectum hubo significancia entre FEN/PAL (r= 0,81), POX/SOD (r= 0,89) y FEN/POX (r= 0,95). Para S. tuberosum L., susceptible, la enfermedad no permitió que la planta completara su ciclo. La actividad enzimática decreció en términos absolutos y fue menor que en las silvestres. No obstante, en S. tuberosum hubo correlaciones significativas entre FEN/PAL (r= 0,97), POX/SOD (r= 0,93) y FEN/POX (r= 0,93). En las especies resistentes, la actividad de las enzimas y de los fenoles totales se incrementó a medida que el patógeno se establecía, no así en la susceptible. Se corrobora la poligenia de defensa de las especies silvestres incluidas en el presente estudio contra el oomiceto.

A joint Chilean, United States potato (Solanum sect.Petota) germplasm collecting expedition was conducted in Chile between January 21–March 30, 1989. The trip resulted in collections ofSolanum brevidens (50 accessions),S. etuberosum (28 accessions),S. maglia (3 accessions) and indigenous primitive landraces ofS. tuberosum (24 accessions). Comments are presented on the habitats, distribution, and potential and realized breeding value of these species.

In vitro culture of pollen tubes is a convenient way to distinguish inviable pollen from stylar incompatibilities which prevent pollen tube growthin vivo. This usefulness ofin vitro pollen tube culture depends on how successful the technique is in promoting optimal pollen tube growth. Experiments were conducted to compare the effects of the type of sugar and method of aeration of cultures ofSolanum species (potato) pollen. Lactose was superior to sucrose in promoting pollen germination and pollen tube growth on hanging drop cultures. Germination and growth were greatly enhanced when pollen was cultured in actively aerated lactose medium. Since pollen growth comparable to that occurringin vivo was observed using lactose medium with active aeration, this technique may be useful in various types of research related to reproductive biology.

A joint Danish, Mexican, United States wild potato (Solanum sect.Petota) germplasm collecting expedition was conducted in Mexico between Aug. 21-Oct. 20, 1988. The trip resulted in 93 seed and 25 tuber collections of 18 species and one putative natural hybrid,S. xmichoacanum. Rare species collected include:Solanum darum,S. hintonii,S. lesteri, andS. xmichoacanum. First germplasm collections were made of the disjunct populations ofS. fendleri from Baja California. The potential and realized breeding value of these species is discussed.

Total antioxidant activity was evaluated in 40 tuber-bearing Solanum species. Accessions identified in a broad screen as having high antioxidant activity were fine screened via high performance liquid chromatography (HPLC) to determine specific phenolic compounds present. The objective of the study was to identify tuber-bearing Solanum species that are higher in antioxidants than in currently available cultivated varieties and breeding lines, and to investigate the variability for this trait among accessions within species and genotypes within accessions. The identified accessions will be used as a source of germplasm to develop potato cultivars containing increased levels of antioxidant compounds. In a broad screen for total antioxidant activity, the 40 tuber-bearing species showed a wider range of variability than cultivated varieties and breeding lines. Solanum pinnatisectum and S. jamesii accessions consistently ranked among the highest in antioxidant activity and phenolic content. Based on the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay, antioxidant activity in the wild species ranged from 48 to 824 μg trolox eq/gfw. HPLC analysis revealed that the phenolic content of these species was primarily composed of chlorogenic and caffeic acids. Other phenolics identified were p-coumaric acid, rutin hydrate, vanillic acid, epicatechin, t-cinnamic acid, gallic acid, and salicylic acid. The highest phenolic content found among the accessions was five-fold higher than the highest of the cultivated genotypes.

Endosperm Balance Number (EBN) is a genetic, dose-dependent crossability system functioning in tuber-bearing Solanum species. Each species has been assigned 1EBN, 2EBN, or 4EBN. Species thus designated cross only within their EBN group. Doubling of chromosome number also doubles the EBN. The ploidy: EBN ratio is not consistent among Solanum species. Some diploids are 2EBN while others are 1EBN. Some tetraploids are 4EBN while others are 2EBN. Species from Mexico typically have EBNs that are one-half of their ploidy [e.g. 2x(1EBN), 4x(2EBN)]. Hybrids of Mexican species and a South American species, 2x(1EBN) S. Commersonii, and its 4x(2EBN) colchicine derivative were made and crossed to 1, 2, and 4EBN standard testers to determine the relationship of the genetic organization of EBN among and within these species. Diploid hybrids crossed only to 1EBN standard testers. Hybrids of 4x(2EBN) S. commersonii and 4x(2EBN) Mexican species crossed almost exclusively to 2EBN standard testers. Complex tetraploid hybrids involving S. commersonii, S. stenophyllidium (a Mexican diploid), and Mexican tetraploids of series Longipedicellata also crossed only to 2EBN testers. The apparent lack of recombination and segregation for EBN in these hybrids indicates that the genomes of the Mexican diploid and tetraploid species carry EBN in a way genetically similar to that of the South American species S. Commersonii.
