Mesoporous silica nanoparticles (MSNs) are physically and chemically stable inorganic nanomaterials that have been attracting much attention as carriers for drug delivery systems in the field of nanomedicine. In the present study, we investigated the potential of MSN vaccines that incorporate antigen peptides for use in cancer immunotherapy. In vitro experiments demonstrated that fluorescently labeled MSNs accumulated in a line of mouse dendritic cells (DC2.4 cells), where the particles localized to the cytosol. These observations could suggest that MSNs have potential for use in delivering the loaded molecules into antigen-presenting cells, thereby stimulating the host acquired immune system. In vivo experiments demonstrated prolonged survival in mice implanted with ovalbumin (OVA)-expressing lymphoma cells (E.G7-OVA cells) following subcutaneous inoculation with MSNs incorporating OVA antigen peptides. Furthermore, OVA-specific immunoglobulin G antibodies and cytotoxic T lymphocytes were detected in the serum and the spleen cells, respectively, of mice inoculated with an MSN-OVA vaccine, indicating the induction of antigen-specific responses in both the humoral and cellular immune systems. Advanced breast cancer remains a significant oncological challenge, requiring new approaches to improve clinical outcomes. This study investigated an innovative theranostic agent using the MCM-41-NH2-DTPA-Gd3⁺-MIH nanomaterial, which combined MRI imaging for detection and a novel chemotherapy agent (MIH 2.4Bl) for treatment. The nanomaterial was based on the mesoporous silica type, MCM-41, and was optimized for drug delivery via functionalization with amine groups and conjugation with DTPA and complexation with Gd3+. MRI sensitivity was enhanced by using gadolinium-based contrast agents, which are crucial in identifying early neoplastic lesions. MIH 2.4Bl, with its unique mesoionic structure, allows effective interactions with biomolecules that facilitate its intracellular antitumoral activity. Physicochemical characterization confirmed the nanomaterial synthesis and effective drug incorporation, with 15% of MIH 2.4Bl being adsorbed. Drug release assays indicated that approximately 50% was released within 8 h. MRI phantom studies demonstrated the superior imaging capability of the nanomaterial, with a relaxivity significantly higher than that of the commercial agent Magnevist. In vitro cellular cytotoxicity assays, the effectiveness of the nanomaterial in killing MDA-MB-231 breast cancer cells was demonstrated at an EC50 concentration of 12.6 mg/mL compared to an EC50 concentration of 68.9 mg/mL in normal human mammary epithelial cells (HMECs). In vivo, MRI evaluation in a 4T1 syngeneic mouse model confirmed its efficacy as a contrast agent. Coal fly ash (CFA) is an industrial byproduct produced during the production of electricity in thermal power plants from the burning of pulverized coal. It is considered hazardous due to the presence of toxic heavy metals while it is also considered valuable due to the presence of value-added minerals like silicates, alumina, and iron oxides. Silica nanoparticles' demands and application have increased drastically in the last decade due to their mesoporous nature, high surface area to volume ratio, etc. Here in the present research work, short rod-shaped, mesoporous silica nanoparticles (MSN) have been synthesized from coal fly ash by using Bacillus circulans MTCC 6811 in two steps. Firstly, CFA was kept with the bacterial culture for bioleaching for 25 days in an incubator shaker at 120 rpm. Secondly, the dissolved silica in the medium was precipitated with the 4 M sodium hydroxide to obtain a short rod-shaped MSN. The purification of the synthesized silica particle was done by treating them with 1 M HCl at 120 °C, for 90 min. The synthesized short rod-shaped MSN were characterized by UV–vis spectroscopy (UV–Vis), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), Particle size analyzer (PSA), Field emission scanning electron microscopy (FESEM), and transmission electron microscope. The microscopic techniques revealed the short rod-shaped mesoporous silica nanoparticles (MSN) for the final nano-silica, whose size varies from 40 to 80 nm, with an average size of 36 ± 5 nm. The XRD shows the crystalline nature of the synthesized MSN having a crystallite size of 36 nm. The FTIR showed the three characteristic bands in the range of 400–1100 cm−1, indicating the purity of the sample. The energy dispersive X-ray (EDX) showed 53.04 wt% oxygen and 43.42% Si along with 3.54% carbon in the final MSN. The particle size analyzer revealed that the average particle size is 368.7 nm in radius and the polydispersity index (PDI) is 0.667. Nisin was loaded in mesoporous silica nanoparticles (MSN) (0, 1, 2 and 3 wt%) and incorporated into poly (lactic acid) (PLA) matrix by casting method. The mechanical property, water vapor permeability, thermal property, and antimicrobial property of PLA nanocomposite films were measured. And the internal structure of the composite film was observed through the Scanning electron microscope. Meanwhile, the kinetic of nisin releasing from food simulated solution was investigated and the migration model was established. The result showed that mesoporous silica had an effect on the mechanical and thermal property of the composite films. Both water vapor transmission rate (WVP) and oxygen permeation rate (OTR) decreased, especially for 3MSN/NI/PLA, where WVP decreased by about 9.64% and OTR decreased by 35.19%, which was beneficial to improve the preservation effect. With the addition of MSN, the amount of loaded nisin increased, and at 3MSN/NI, the nisin loading reached 88.75%, and the mesoporous silica could effectively control the release of nisin, thus prolonging the preservation effect. The release kinetic of nisin was described by Fick's law and Weibull's model. In short, the PLA nanocomposite film has the potential to be used as an antimicrobial active film by adding nisin as an antimicrobial agent. As a potential therapeutic solution for cancer, the combination of naturally occurring bioactive compounds with nanocarriers could be of great importance. In this study, a mesoporous silica nanoparticle (MSN) surface decorated with polydopamine (PDA) has been prepared using myricetin (MYR)-loaded mesoporous silica nanoparticles (MYR-MSN-PDA) as a means of drug delivery against cancer. Using dynamic light scattering (DLS), scanning electron microscopy (SEM) techniques, and Fourier-transform infrared spectroscopy (FTIR) the MYR-MSN-PDA was characterized. A cytotoxic study was then carried out by testing the MYR-MSN-PDA against normal and cancer cell lines to assess their effects on cytotoxicity. MYR-MSN-PDA were studied using flow cytometry as well as real-time quantitative PCR to determine their apoptotic properties. Furthermore, MYR-MSN-PDA was evaluated for its anti-oxidant properties as well. Compared to normal cells, MYR-MSN-PDA showed higher cytotoxicity against cancer cells, indicating that the synthesized nanoparticles do not pose any potential health risks. It is also important to note that MYR-MSN-PDA also has statistically significant level of effectiveness as an antioxidant agent in combating free radicals in both DPPH and ABTS assays (***p < 0.001). In addition, MYR-MSN-PDA at concentrations of 7, 14, and 28 µg/mL was shown to induce 6.74%, 40.1%, and 55.1% subG1 arrest in cells treated with nanoparticles, respectively, indicating that the nanoparticles can induce apoptosis. According to the results of this study, MYR-MSN-PDA synthesized in this study demonstrated potency against cancer cells in vitro, and has anti-oxidant properties which it is recommended that more research is carried out in vitro and in vivo. The goal of tissue-engineered heart valves (TEHV) is to replace normal heart valves and overcome the shortcomings of heart valve replacement commonly used in clinical practice. However, calcification of TEHV is the major bottleneck to break for both clinical workers and researchers. Endothelialization of TEHV plays a crucial role in delaying valve calcification by reducing platelet adhesion and covering the calcified spots. In the present study, we loaded RunX2-siRNA and VEGF into mesoporous silica nanoparticles and investigated the properties of anti-calcification and endothelialization in vitro. Then, the mesoporous silica nanoparticle was immobilized on the decellularized porcine aortic valve (DPAV) by layer self-assembly and investigated the anti-calcification and endothelialization. Our results demonstrated that the mesoporous silica nanoparticles delivery vehicle demonstrated good biocompatibility, and a stable release of RunX2-siRNA and VEGF. The hybrid decellularized valve exhibited a low hemolysis rate and promoted endothelial cell proliferation and adhesion while silencing RunX2 gene expression in valve interstitial cells, and the hybrid decellularized valve showed good mechanical properties. Finally, the in vivo experiment showed that the mesoporous silica nanoparticles delivery vehicle could enhance the endothelialization of the hybrid valve. [Display omitted] In recent years, injectable stimuli-sensitive hydrogels are employed as suitable drug delivery carriers for the release of various anti-cancer drugs. However, large pore size of the microporous hydrogel trigger release of small molecular anticancer drug that limits hydrogel application in cancer therapy. Therefore, introducing reinforcing fillers such as mesoporous silica nanoparticles (MSNs) can not only load different type of anticancer drugs but also prevent the premature release of drugs due to the strengthening of the networks. Furthermore, high specific surface area, suitable size, large pore volume, and stable physicochemical properties of MSNs can improve the therapeutic efficacy. In this study, to sustain the release of hydrophobic anticancer drug, camptothecin (CPT) was loaded into MSNs, and then imbibed into the physiological stimuli-sensitive poly(ethylene glycol)-poly(β-aminoester urethane) (PAEU) hydrogels. MSN-imbibed PAEU hydrogels exhibited prolonged release of CPT than MSNs and PAEU hydrogel alone. Furthermore, MSN-imbibed PAEU copolymers form stable viscoelastic gel depot into the subcutaneous layers of Sprague-Dawley rats and found to be safe and not induced toxicity to healthy organs, implying biodegradability and safety of the hydrogels. Interestingly, CPT-loaded hydrogels shown dose-dependent toxicity to A549 and B16F10 cells. These results demonstrated that MSN-imbibed PAEU hydrogel with biocompatible, biodegradable, and in situ gel forming property could be a useful drug delivery depot for sustained release of anticancer drugs. Interfacial nanobubbles on a superhydrophobic surface can serve as ultrasound cavitation nuclei for continuously promoting sonodynamic therapy, but their poor dispersibility in blood has limited their biomedical application. In this study, we proposed ultrasound-responsive biomimetic superhydrophobic mesoporous silica nanoparticles, modified with red blood cell membrane and loaded with doxorubicin (DOX) (F-MSN-DOX@RBC), for RM-1 tumor sonodynamic therapy. Their mean size and zeta potentials were 232 ± 78.8 nm and −35.57 ± 0.74 mV, respectively. The F-MSN-DOX@RBC accumulation in a tumor was significantly higher than in the control group, and the spleen uptake of F-MSN-DOX@RBC was significantly reduced in comparison to that of the F-MSN-DOX group. Moreover, the cavitation caused by a single dose of F-MSN-DOX@RBC combined with multiple ultrasounds provided continuous sonodynamic therapy. The tumor inhibition rates in the experimental group were 71.5 8 ± 9.54%, which is significantly better than the control group. DHE and CD31 fluorescence staining was used to assess the reactive oxygen species (ROS) generated and the broken tumor vascular system induced by ultrasound. Finally, we can conclude that the combination of anti-vascular therapy, sonodynamic therapy by ROS, and chemotherapy promoted tumor treatment efficacy. The characteristics of global prevalence and high recurrence of bladder cancer has led numerous efforts to develop new treatments. The spontaneous voiding and degradation of the chemodrug hamper the efficacy and effectiveness of intravesical chemotherapy following tumor resection. Herein, the externally thiolated hollow mesoporous silica nanoparticles (MSN-SH(E)) is fabricated to serve as a platform for improved bladder intravesical therapy. Enhanced mucoadhesive effect of the thiolated nanovector is confirmed with porcine bladder. The permeation-enhancing effect is also verified, and a fragmented distribution pattern of a tight junction protein, claudin-4, indicates the opening of tight junction. Moreover, MSN-SH(E)-associated reprogramming of M2 macrophages to M1-like phenotype is observed in vitro. The antitumor activity of the mitomycin C (MMC)-loaded nanovector (MMC@MSN-SH(E)) is more effective than that of MMC alone in both in vitro and in vivo. In addition, IHC staining is used to analyze IFN-y, TGF-/1, and TNF-a. These observations substantiated the significance of MMC@MSN-SH(E) in promoting anticancer activity, holding the great potential for being used in intravesical therapy for non-muscle invasive bladder cancer (NMIBC) dueto its mucoadhesivity, enhanced permeation, immunomodulation, and prolonged and very efficient drug exposure. Liver cancer, especially hepatocellular carcinoma, is an important cause of cancer-related death, and its incidence is increasing worldwide. Nano drug delivery systems have shown great promise in the treatment of cancers. In order to improve their therapeutic efficacy, it is very important to realize the high accumulation and effective release of drugs at the tumor site. In this manuscript, using doxorubicin (DOX) as a model drug, CD13-targeted mesoporous silica nanoparticles coated with NGR-peptide-modified pegylated carboxymethyl chitosan were constructed (DOX/MSN-CPN). DOX/MSN-CPN comprises a spherical shape with an obvious capping structure and a particle size of 125.01 ± 1.52 nm. With a decrease in pH, DOX/MSN-CPN showed responsive desorption from DOX/MSN-CPN and pH-responsive release of DOX was observed. Meanwhile, DOX/MSN-CPN could be efficiently absorbed through NGR-mediated internalization in vitro and could efficiently deliver DOX to tumor tissues with long accumulation times in vivo, suggesting good active targeting properties. Moreover, significant tumor inhibition has been observed in antitumor studies in vivo. This study provides a strategy of utilizing DOX/MSN-CPN as a nano-platform for drug delivery, which has superb therapeutic efficacy and safety for the treatment of hepatocellular carcinoma both in vivo and in vitro. Micro-ribonucleic acid (miRNA)-based therapies show advantages for bone regeneration but need efficient intracellular delivery methods. Inorganic nanoparticles such as mesoporous bioactive glass nanoparticles (MBGN) and mesoporous silica nanoparticles (MSN) have received growing interest in the intracellular delivery of nucleic acids. This study explores the capacity of MBGN and MSN for delivering miRNA to bone marrow mesenchymal stem cells (BMSC) for bone regenerative purposes, with a focus on comparing the two in terms of cell viability, transfection efficiency, and osteogenic actions. Spherical MBGN and MSN with a particle size of ~200 nm and small-sized mesopores were prepared using the sol-gel method, and then the surface was modified with polyethyleneimine for miRNA loading and delivery. The results showed miRNA can be loaded into both nanoparticles within 2 h and was released sustainedly for up to 3 days. Confocal laser scanning microscopy and flow cytometry analysis indicated a high transfection efficiency (>64%) of both nanoparticles without statistical difference. Compared with MSN, MBGN showed stronger activation of alkaline phosphatase and activation of osteocalcin genes. This translated to a greater osteogenic effect of MBGN on BMSC, with Alizarin red staining showing greater mineralization compared with the MSN group. This review focuses on the biomedical application of mesoporous silica nanoparticles (MSNs), mainly focusing on the therapeutic application of MSNs for cancer treatment and specifically on overcoming the challenges of currently available anthelmintics (e.g., low water solubility) as repurposed drugs for cancer treatment. MSNs, due to their promising features, such as tunable pore size and volume, ability to control the drug release, and ability to convert the crystalline state of drugs to an amorphous state, are appropriate carriers for drug delivery with the improved solubility of hydrophobic drugs. The biomedical applications of MSNs can be further improved by the development of MSN-based multimodal anticancer therapeutics (e.g., photosensitizer-, photothermal-, and chemotherapeutics-modified MSNs) and chemical modifications, such as poly ethyleneglycol (PEG)ylation. In this review, various applications of MSNs (photodynamic and sonodynamic therapies, chemotherapy, radiation therapy, gene therapy, immunotherapy) and, in particular, as the carrier of anthelmintics for cancer therapy have been discussed. Additionally, the issues related to the safety of these nanoparticles have been deeply discussed. According to the findings of this literature review, the applications of MSN nanosystems for cancer therapy are a promising approach to improving the efficacy of the diagnostic and chemotherapeutic agents. The clinical utilization of fenbendazole (FBZ) as a potential anticancer drug has been limited due to its low water solubility, which causes its low bioavailability. The development of a drug nanoformulation that includes the solubilizing agent as a drug carrier can improve solubility and bioavailability. In this study, Mobil Composition of Matter Number 48 (MCM-48) nanoparticles were synthesized and functionalized with succinylated β-lactoglobulin (BLG) to prevent early-burst drug release. The BLG-modified amine-functionalized MCM-48 (MCM-BLG) nanoparticles were loaded with FBZ to produce the drug nanoformulation (FBZ-MCM-BLG) and improved the water solubility and, consequently, its anticancer effects against human prostate cancer PC-3 cells. The prepared FBZ-MCM-BLG was characterized in terms of size, zeta potential, drug loading capacity, morphology, thermal and chemical analyses, drug release, cellular uptake, cell viability, cell proliferation, production of reactive oxygen species (ROS), and cell migration. The results demonstrated that the FBZ-MCM-BLG nanoparticles have a spherical morphology with a size and zeta potential of 369 ± 28 nm and 28 ± 0.4 mV, respectively. The drug loading efficiency of the new nanoformulation was 19%. The release of FBZ was pH-dependent; a maximum cumulative release of about 76 and 62% in 12 h and a burst release of 53 and 38% in the first 0.5 h was observed at pH 1.2 and 6.8, respectively. The prepared FBZ-MCM-BLG formulation demonstrated higher cytotoxicity effects against PC-3 cells by 5.6- and 1.8-fold, respectively, when compared to FBZ and FBZ-MCM nanoparticles. The new formulation also increased the production of ROS by 1.6- and 1.2-fold and inhibited the migration of PC-3 cells when compared to the FBZ and FBZ-MCM nanoparticles, respectively. Overall, FBZ-MCM-BLG nanoparticles improved FBZ delivery to PC-3 cells and have the potential to be evaluated for the treatment of prostate cancer following a comprehensive in vivo study. Tacrolimus (FK506) is an effective therapeutic for transplant rejection in clinical practice, primarily inhibiting rejection by suppressing the activation and proliferation of allogeneic T cells in the lymph nodes (LNs). However, conventional administration methods face challenges in directly delivering free FK506 to the LNs. In this study, we introduce a novel LN-targeted delivery system based on mesoporous silica nanoparticles (MSNs-FK506-MECA79). These particles were designed to selectively target high endothelial venules in LNs; this was achieved through surface modification with MECA79 antibodies. Their mean size and zeta potential were 201.18 ± 5.98 nm and − 16.12 ± 0.36 mV, respectively. Our findings showed that MSNs-FK506-MECA79 could accumulate in LNs and increase the local concentration of FK506 from 28.02 ± 7.71 ng/g to 123.81 ± 76.76 ng/g compared with the free FK506 treatment group. Subsequently, the therapeutic efficacy of MSNs-FK506-MECA79 was evaluated in a skin transplantation model. The treatment with MSNs-FK506-MECA79 could lead to a decrease in the infiltration of T cells in the grafts, a reduction in the grade of rejection, and a significant prolongation of survival. In this study, mesoporous silica nanoparticles (MSN) as the core, loaded with paclitaxel (PTX), were coated with a non-porous silica shell functionalized with disulfide bonds. The nanoparticles were further coated with polyethylene glycol (PEG) via disulfide linkages. We analyzed the physicochemical properties of nanoparticles, including hydrodynamic size via Dynamic Light Scattering (DLS), zeta potential, X-ray Diffraction (XRD) patterns, Fourier-Transform Infrared (FTIR) spectra, and imaging through Transmission Electron Microscopy (TEM) and Scanning Electron Microscopy (SEM). The drug release profile in two distinct glutathione (GSH) concentrations of 2 µM and 10 µM was measured. The cellular uptake of nanoparticles by MCF-7 cell line was determined using Confocal Laser Scanning Microscopy (CLSM) images and flow cytometry. Furthermore, the cell viability and the capability of nanoparticles to induce apoptosis in MCF-7 cell line were studied using the MTT assay and flow cytometry, respectively. Our investigations revealed that the release of PTX from the drug delivery system was redox-responsive. Also, results indicated an elevated level of cellular uptake and efficient induction of apoptosis, underscoring the promising potential of this redox-responsive drug delivery system for breast cancer therapy. To improve the anticancer properties of elesclomol (ELC), targeted theranostic nanoparticles (NPs; APT-PEG-Au-MMNPs@ELC) were designed to increase the selectivity of the drug delivery system (DDS). ELC was synthesized and entrapped in the open porous structure of magnetic mesoporous silica nanoparticles (MMNPs). The pore entrance of MMNPs was then blocked using gold gatekeepers. Finally, the external surfaces of the particles were grafted with functional polyethylene glycol (PEG) and EpCAM aptamer to generate biocompatible and targeted NPs. In the next step, the physicochemical properties of prepared NPs were fully evaluated and their anticancer potential was evaluated both in vitro and in vivo. The targeted NPs were successfully synthesized with a final size diameter of 81.13 ± 7.41 nm. The results indicated a pH-dependent release pattern, which sustained for 72 h despite an initial rapid release. Upon exposure to APT-PEG-Au-MMNPs@ELC, higher cytotoxicity was observed in human prostate cancer cells (PC-3) as compared with control Chinese hamster ovary (CHO) cells, indicating higher specificity of targeted NPs against EpCAM-positive cancerous cells. Moreover, APT-PEG-Au-MMNPs@ELC could induce apoptosis in PC-3 cells. In vivo results on a PC-3 xenograft tumor model demonstrated that targeted NPs could significantly inhibit tumor growth and diminish severe side effects of ELC, compared to the free drug. Premature leakage of drugs into the bloodstream remains a problem in the process of drug delivery, resulting in higher toxic and side effects on normal tissue cells. Novel drug carriers enabling effective drug accumulation in the tumor site have got much attention of researchers. Poly (methyl acrylic acid diisopropyl amino ethyl ester) (PDPA) is a polymer with pH-sensitive response, the diisopropylamine in the side chains are protonated in an acidic environment, resulting in its changing from hydrophobic to hydrophilic. PDPA was grafted onto the surface of mesoporous silica nanoparticles (MSNs) by atom transfer radical polymerization (ATRP) reaction. Then folic acid (FA) was introduced onto PDPA as a targeting ligand for enabling the drug carrier to enter tumor cells effectively. Simultaneously poly [di (ethylene glycol) methyl ether methacrylate-co-oligo (ethylene glycol) methacrylate]-b-poly[methyl acrylic acid diisopropyl amino ethyl ester] (P(MEO2MA90-co-OEGMA10)-b-PDPA10) with dual sensitivity of temperature and pH was synthesized and the lower critical solution temperatures (LCST) of the polymer was adjusted to 44 ℃ by adjusting the proportion of three monomers. A drug carrier with a core-shell structure was self-assembled through hydrophobic force by covering the later polymer on the surface of MSNs-PDPA-FA. Finally, the drug release kinetics of the carrier was studied by using doxorubicin(DOX) as a model drug, and the result showed that the total leakage of the model drug DOX was less than 10% after 48 h in normal physiological environment (pH=7.4, 37 ℃). However, when the pH value was changed to 5.0 at 44 ℃, the shell was dissociated from the core and the drug was released. Then, the drug was released rapidly within 5 h and the cumulative drug release could reach to 65% in 48 h. Therefore, this research was expected to construct a shielding system based on the pH/temperature dualresponsive polymer, which could be used to avoid premature exposure of certain functional groups or molecules to the normal physiological environment, and it could also prevent the premature release of drugs in the normal tissues. Background: Sonodynamic therapy (SDT) may be a new hopeful non-invasive method for cancer treatment, which incorporates a combination of low-intensity ultrasound and a sonosensitive chemical. The goal of this study was to evaluate the effect of dual-frequency sonication (1 and 3 MHz) and injected Hematoporphyrin encapsulated in mesoporous silica nanoparticles (HPMSNs) as a sensitizer in the treatment of mice grafted with breast adenocarcinoma. Methods: In this research, one hundred and thirty-two female mice with grafted breast adenocarcinoma were separated into 22 groups including control, sham, 4 groups of sonication 1 or 3 MHz (1 and 2 W/cm² ), and 16 groups of SDT with Hematoporphyrin (HP) and HP-MSNs (2.5 and 5 mg/kg). The tumor growth factors and tumor grading were used to assess the treatment management. Results: The results indicate that dual-frequency sonication has a delayed effect on tumor growth. The required time of T5 to the initial volume in all groups of SDT with HP (5 mg/kg) was greater than that in the control group (P<0.05). It was observed that SDT with an injection of HPMSNs was effective in delaying tumor growth and the time of T2 and T5 was higher than that of other groups (P<0.05). This group had Grade II (intermediate), while others had Grade III (high) malignancy in the histological study of mice breast adenocarcinoma. Conclusion: Our results reveal that dual-frequency SDT therapy with HP-MSN has a delaying tumor growth effect on mice breast adenocarcinoma. As pharmaceutical excipients, mesoporous silica nanoparticles (MSNs) have attracted considerable concern based on potential risks to the public. The impact of MSNs on biochemical metabolism is poorly understood, and few studies have compared the effects of MSNs administered via different routes. To evaluate the hepatotoxicity of MSNs, metabolomics, proteomics and transcriptomic analyses were performed in mice after intravenous (20 mg/kg/d) or oral ad-ministration (200 mg/kg/d) of MSNs for 10 days. Intravenous injection induced significant hepatic injury based on pathological inspection and increased the levels of AST/ALT and the inflammatory factors IL-6, IL-1β and TNF-a. Omics data suggested intravenous administration of MSNs perturbed the following metabolites: succinate, hypoxanthine, GSSG, NADP+, NADPH and 6-phosphogluconic acid. In addition, increases in GPX, SOD3, G6PD, HK, and PFK at proteomic and transcriptomic levels suggested elevation of glycolysis and pentose phosphate pathway, synthesis of glutathione and nucleotides, and antioxidative pathway activity, whereas oxidative phosphorylation, TCA and mitochondrial energy metabolism were reduced. On the other hand, oral administration of MSNs disturbed inflammatory factors and metabolites of ribose-5-phosphate, 6-phosphogluconate, GSSG, and NADP+ associated with the pentose phosphate pathway, glutathione synthesis and oxidative stress albeit to a lesser extent than intravenous injection despite the administration of a ten-fold greater dose. Overall, systematic biological data suggested that intravenous injection of nanoparticles of pharmaceutical excipients substantially affected hepatic metabolism function and induced oxidative stress and inflammation, whereas oral administration exhibited milder effects compared with intravenous injection. Mesoporous silica nanoparticles (MSNs) have been well-researched in the design and fabrication of advanced drug delivery systems (DDSs) due to their advantages such as good biocompatibility, large specific surface area and pore volume for drug loading, easily surface modification, adjusted size and good thermal/chemical stability. For MSN-based DDSs, gate materials are also necessary. And natural polysaccharides, one kind of the most abundant natural resource, have been widely applied as the "gatekeepers" in MSN-based DDSs. Polysaccharides are cheap and rich in sources with good biocompatibility, and some of them have important biological functions. In this review article, polysaccharides including chitosan, hyaluronic acid, sodium alginate and dextran, et al. are briefly introduced. Imbalance of oxidative and inflammatory regulation is the main contributor to neurofunctional deterioration and failure of rebuilding spared neural networks after spinal cord injury (SCI). As an emerging biosafe strategy for protecting against oxidative and inflammatory damage, hydrogen (H2) therapy is a promising approach for improving the microenvironment to allow neural regeneration. However, achieving release of H2 at sufficient concentrations specifically into the injured area is critical for the therapeutic effect of H2. Thus, we assembled SiO2@mSiO2 mesoporous silica nanoparticles and loaded them with ammonia borane (AB), which has abundant capacity and allows controllable release of H2 in an acid-dependent manner. The release of H2 from AB/SiO2@mSiO2 was satisfactory at pH 6.6, which is approximately equal to the microenvironmental acidity after SCI. After AB/SiO2@mSiO2 were intrathecally administered to rat models of SCI, continuous release of H2 from these nanoparticles synergistically enhanced neurofunctional recovery, reduced fibrotic scar formation and promoted neural regeneration by suppressing oxidative stress reaction. Furthermore, in the subacute phase of SCI, microglia were markedly polarized toward the M2 phenotype by H2 via inhibition of TLR9 expression in astrocytes. In conclusion, H2 delivery through AB/SiO2@mSiO2 has the potential to efficiently treat SCI through comprehensive modulation of the oxidative and inflammatory imbalance in the microenvironment. In the present work, a mesoporous silica nanoparticle (MSN@RB-NA)-based fluorescent nanomaterial was developed for fast and selective detection of a nerve agent simulant, diethylchlorophosphate (DCP), where two chromophores, naphthalimide and rhodamine B, were co-functionalized on the surface of MSN through their silane derivatives. The successful immobilization of the two fluorescent derivatives on MSN was demonstrated by a variety of characterization methods including FT-IR, BET-BJH, XPS, TEM, TGA, and solid-state 13C NMR, etc. A filter-paper sensor based on the dual-chromophore-functionalized MSN was tested for DCP vapor detection. Fluorescence measurements revealed that DCP can induce fluorescence reduction of naphthalimide unit at 476 nm and enhanced emission of rhodamine B at 575 nm of the paper sensor. Control experiments reveal that the piperazine group connected with the naphthalimide unit is responsible for DCP recognition, and a FRET process from naphthalimide to rhodamine B is believed the reason for the observed enhanced rhodamine emission. The paper sensor exhibits high sensitivity to DCP vapor with detection limit at 4.58 ppb, excellent selectivity over other 17 interferents, and fast response speed (< 1 s). Tacrolimus (TAC), a potent immunosuppressive macrolide, has been investigated for ocular diseases due to promising results in the treatment of anterior and posterior segments eye diseases. Mesoporous and functionalized silica nanoparticles show potential as TAC delivery platforms owing to their interesting characteristic as large surface area, uniform pore size distribution, high pore volume, and excellent biocompatibility. The purpose of this study was to incorporate TAC in functionalized silica nanoparticles with 3-aminopropyltriethoxysilane (MSNAPTES) and investigate the safety and biocompatibility of the systems. The MSNAPTES and MSNAPTES TAC nanoparticles were characterized. The in vitro cytotoxicity of MSNAPTES and MSNAPTES load with TAC (MSNAPTES-TAC) in retinal pigment epithelial cells (ARPE-19) was determined, chorioallantoic membrane (CAM) assay model was used to investigate the in vivo biocompatibility, and safety of intravitreal injection was evaluated using clinical examination (assessment of intraocular pressure and indirect fundus ophthalmoscopy), electroretinographic (ERG) and histologic studies in rats' eyes. The elemental analysis (CHN), thermogravimetric (TGA), photon correlation spectroscopy and Fourier transform infrared (FTIR) analysis confirmed the presence of functionalized agent and TAC in the MSNAPTES nanoparticles. TAC loading was estimated at 7% for the MSNAPTES TAC nanoparticles. MSNAPTES and MSNAPTES TAC did not present in vitro cytotoxicity. The drug delivery systems showed good biocompatibility on CAM. No retinal abnormalities, vitreous hemorrhage, neovascularization, retinal detachment, and optic nerve atrophy were observed during the in vivo study. Follow-up ERGs showed no changes in the function of the retina cells after 15 days of intravitreal injection, and histopathologic observations support these findings. In conclusion, MSNAPTES TAC was successfully synthesized, and physicochemical analyses confirmed the presence of TAC in the nanoparticles. In vitro and in vivo studies indicated that MSNAPTES TAC was safe to intravitreal administration. Background: A low pH environment is created due to the production of acids by oral biofilms that further leads to the dissolution of hydroxyapatite crystal in the tooth structure significantly altering the equilibrium. Although the overall bacterial counts may not be eradicated from the oral cavity, however, synthesis of engineered anti-bacterial materials are warranted to reduce the pathogenic impact of the oral biofilms. The purpose of this study was to synthesize and characterize chlorhexidine (CHX)-loaded mesoporous silica nanoparticles (MSN) grafted with poly-L-glycolic acid (PGA) and to test the in vitro drug release in various pH environments, cytotoxicity, and antimicrobial capacity. In addition, this study aimed to investigate the delivery of CHX-loaded/MSN-PGA nanoparticles through demineralized dentin tubules and how these nanoparticles interact with tooth dentin after mixing with commercial dentin adhesive for potential clinical application. Results: Characterization using SEM/TEM and EDX confirmed the synthesis of CHX-loaded/MSN-PGA. An increase in the percentage of drug encapsulation efficiency from 81 to 85% in CHX loaded/MSN and 92-95% in CHX loaded/MSN-PGA proportionately increased with increasing the amount of CHX during the fabrication of nanoparticles. For both time-periods (24 h or 30 days), the relative microbial viability significantly decreased by increasing the CHX content (P < 0.001). Generally, the cell viability percentage of DPSCs exposed to MSN-PGA/Blank, CHX-loaded/MSN, and CHX-loaded/MSN-PGA, respectively was > 80% indicating low cytotoxicity profiles of experimental nanoparticles. After 9 months in artificial saliva (pH 7.4), the significantly highest micro-tensile bond strength value was recorded for 25:50 CHX/MSN and 25:50:50 CHX/MSN-PGA. A homogenous and widely distributed 50:50:50 CHX-loaded/MSN-PGA nanoparticles exhibited excellent bonding with the application of commercially available dentin adhesive. Conclusions: A pH-sensitive CHX release response was noted when loaded in MSN grafted PGA nanoparticles. The formulated drug-loaded nanocarrier demonstrated excellent physicochemical, spectral, and biological characteristics. Mesoporous silica nanoparticles (MSNs) functionalized with redox‐sensitive or pH‐sensitive nanovalves for doxorubicin delivery and release by using recombinant human H chain ferritin (HFn) as a cap have been designed and fabricated. In both cases, transmission electron microscope observatory, dynamic light scattering change, Fourier transform infrared spectra examination, thermogravimetric analysis show that HFn can be chemically bonded to MSNs while retaining its ability to target transferrin receptor 1 (TfR1). Cargo loading and release studies demonstrate that HFn is an efficient capping agent, blocking the pores of MSN preventing cargo molecules from diffusing out, and is responsive to redox stimuli or pH changes. More importantly, HFn can not only cap the MSNs, but also enables targeted cargo delivery to malignant cells by binding to the TfR1 that has been overexpressed in various tumors, which can be reflected by the cell viability and fluorescence microscope analysis results comparing with cyclodextrin as the capping agent and TfR1 blocking assay. The in vivo study reveals the excellent efficacy of doxorubicin loaded and HFn capped MSNs on suppression of tumor growth. Mesoporous silica nanoparticles (MSNs) are known as carriers with high loading capacity and large functionalizable surface area for target-directed delivery. In this work, large pore size MSNs were prepared by the co-condensation method. Tioguanine (TG) as a hydrophobic drug was loaded into the synthesized MSNs to prepare TG@MSNs. The results show MSNs were successfully synthesized and they have good loading efficiency and capacity for TG. Silk fibroin (SF) as an active targeting agent and pH-sensitive targeting release, is a natural protein from silkworm cocoons. It was used to coating MSNs to prepare SF/TG@MSNs. SF was Degummed and it was characterized by nuclear magnetic resonance (NMR), fourier transform infrared (FTIR) and field emission scanning electron microscopy (FESEM) methods. The MSNs were fully characterized by FESEM, transmission electron microscopy (TEM), zeta potential analysis, dynamic light scattering (DLS), nitrogen adsorption/desorption analysis, thermogravimetric analysis (TGA), X-ray diffraction (XRD) analysis, FTIR and UV-visible spectroscopies. The mechanism of TG release from SF/TG@MSNs were pH-sensitive. The in vitro release modeling revealed that TG released via the Higuchi model with fickian diffusion mechanism. Mesoporous silica nanoparticle (MSN), one of the most widely used nanomaterials, is of poor biodegradability in vivo due to its highly stable Si–O–Si structure. Here, the structural stability of Si–O–Si was regulated by doping strontium ion. Sr-doped MSNs (Sr-MSNs) were synthesized by a cetyltrimethylammonium-bromide-mediated template method and their phase, morphology, structure, physicochemical properties, in vitro degradability and cytocompatibility were investigated. Results indicated that Sr2+ was successfully incorporated into MSN with the collapse of the ordered mesoporous structure. Sr-doping significantly improved the specific surface area, in vitro degradability and cytocompatibility of MSN in a Sr-content-dependent manner. Particularly, excessive Sr-doping gave rise to generating impure strontium silicate which converted into disordered amorphous silica during degradation and hindered the biodegradable behaviors of MSN. Hence, the synthetic Sr-MSNs with excellent surface nature, biodegradability and biocompatibility were supposed to be applied as potential carriers for the controllable release of drugs and ions in numerous clinical applications. Mesoporous silica nanoparticles (MSNs) have ideal characteristics as next generation of controlled drug delivery systems. In this study, a MSN-based nanocarrier was fabricated and gold nanoparticle (GNP)-biotin conjugates were successfully grafted onto the pore outlets of the prepared MSN. This bioconjugate served as a capping agent with a peptide-cleavable linker sensitive to matrix metalloproteinases (MMPs), which are overexpressed extracellular proteolytic enzymes in cancerous tissue. The prepared nanocarriers were fully characterised by scanning electron microscopy (SEM), transmission electron microscopy (TEM), nitrogen adsorption/desorption, Fourier transform infra-red spectroscopy (FTIR), dynamic light scattering (DLS) and thermo gravimetric analysis (TGA). In vitro release studies showed efficient capping of MSNs with gold gate and controlled release of Doxorubicin (DOX) in the presence of matrix metalloproteinase-2 (MMP-2) and acidic pH values. High DOX-loading capacity (21%) and encapsulation efficiency (95.5%) were achieved using fluorescence technique. DOX-loaded nanocarriers showed high cytocompatibility and could efficiently induce cell death and apoptosis in the MMP-2 overexpressed cell lines. Ultrasmall silver nanoclusters (Ag NCs) are one of the emerging and highly efficient antibacterial agents, owing to the unique features of sub-2 nm particle size and the high abundance of the active Ag+ species. However, practical applications of Ag NCs in biological environment are often hampered by silver oxidization, which results in particle aggregation and loss of antibacterial activity. In this study, for the first time, we develop a facile method to synthesize highly dispersed Ag NCs decorated mesoporous silica nanoparticles (Ag NC-MSNs) capable of long-term and efficient release of Ag+ ions. This novel Ag NC-MSNs nanocomposite was demonstrated as an effective antibacterial agent against both Gram-positive and Gram-negative pathogenic bacteria. Compared with the counterparts Ag NCs and silver nanoparticles decorated mesoporous silica nanoparticles (Ag NP-MSNs), Ag NC-MSNs exhibit 17-fold and 27-fold enhancement in antibacterial potency, respectively. The homogeneous distribution of ultrasmall Ag NCs in the mesoporous architecture of supporting MSNs matrix is crucial for the controlled release of Ag+ ions, leading to the superior broad-spectrum antimicrobial activity. Moreover, the cytotoxicity assay indicated that the effective antibacterial concentration of Ag NC-MSNs shows minimum toxicity on mammalian cells. Cinnamon essential oil (CEO) was successfully loaded into mesoporous silica nanoparticles (MSNPs). To protect and preserve post-harvest white mushrooms (Agaricus bisporus) from CNRMA 03.0371 strain (Mucor circinelloides) and FJ09 species (Mucor sp.), MSNP-CEO/potato starch films were prepared by a casting method to develop a functional packaging design. The cooperation of MSNPs-CEO markedly enhanced the tensile strength and physical properties of films. MSNPs-CEO appeared to be more evenly distributed in the films by scanning electron microscopy analysis. Fourier-transform infrared spectroscopy (FTIR) confirmed that inter-molecular interactions were formed between MSNPs-CEO and potato starch. In addition, the in vitro release rate of CEO in the MSNPs from the films was investigated to improve its antimicrobial activity against the CNRMA 03.0371 strain and the FJ09 species commonly found in post-harvest white mushrooms stored at 4 °C. The MSNP-CEO/potato starch films showed better antimicrobial activity against FJ09 species than against CNRMA 03.0371 strain. Proteins and peptides (PPs), as therapeutics are widely explored in the past few decades, by virtue of their inherent advantages like high specificity and biocompatibility with minimal side effects. However, owing to their macromolecular size, poor membrane permeability, and high enzymatic susceptibility, the effective delivery of PPs is often challenging. Moreover, their subjection to varying environmental conditions, when administered orally, results in PPs denaturation and structural conformation, thereby lowering their bioavailability. Hence, for effective delivery with enhanced bioavailability, protection of PPs using nanoparticle-based delivery system has gained a growing interest. Mesoporous silica nanoparticles (MSNs), with their tailored morphology and pore size, high surface area, easy surface modification, versatile loading capacity, excellent thermal stability, and good biocompatibility, are eligible candidates for the effective delivery of macromolecules to the target site. This review highlights the different barriers hindering the oral absorption of PPs and the various strategies available to overcome them. In addition, the potential benefits of MSNs, along with their diversifying role in controlling the loading of PPs and their release under the influence of specific stimuli, are also discussed in length. Further, the tuning of MSNs for enhanced gene transfection efficacy is also highlighted. Since extensive research is ongoing in this area, this review is concluded with an emphasis on the potential risks of MSNs that need to be addressed prior to their clinical translation. Background. Rise in the number of healthcare associated or hospital acquired infections is a major problem affecting the global healthcare sector. We evaluated superior antibacterial and antibiofilm photodynamic therapy (aPDT) using malachite green encapsulated mesoporous silica nanoparticles (MG-MSN) against Staphylococcus aureus and Escherichia coli, which are known to be major causative agents of nosocomial infections. Methods. Malachite green (MG) was encapsulated on mesoporous silica nanoparticles (MSN). Fourier-transform infrared spectroscopy, Transmission electron microscopy, and spectroscopic analysis were performed to characterize the MG-MSN. The antimi-crobial efficacies of MSN, MG, and MG-MSN were investigated and the results were recorded. Results. MG-MSN was effective against both the tested bacteria. S. aureus was more phototoxic to MG-MSN compared to E. coli. The antibiofilm efficacy of MG-MSN on E. coli and S. aureus was also studied. Biofilm inhibition was 65.68 ± 2.62% in E. coli and 79.66 ± 3.82% in S. aureus. Cell viability assay, exopolysaccharides quantification, and confocal laser scanning microscopy studies also revealed the enhanced antibiofilm activity of MG-MSN when used as a potential photosensitizer for aPDT. Abstract Curcumin loaded mesoporous silica nanoparticle (SBA-15) was incorporated into chitosan (CS) film to improve the functional properties of pure CS film. Curcumin was loaded into SBA-15 (SBA-15-Cur) through a rotavapor method. The structural properties of SBA-15-Cur were characterised in detail by small-angle X-ray scattering, fourier transform-infrared (FT-IR) spectroscopy, transmission electron microscopy and N 2 adsorption-desorption analyses. The CS/SBA-15-Cur bionanocomposite film was prepared by solvent casting. The mechanical properties of the bionanocomposite film were improved by the addition of the SBA-15-Cur nanofiller, as revealed by the FT-IR analysis of the biocomposite film. However, the water vapour permeability of the films was not significantly influenced by the filler. Release studies suggested that the CS/SBA-15-Cur bionanocomposite film exhibited pH-responsive and sustained release behaviour of curcumin. The CS/SBA-15-Cur film demonstrated efficient antimicrobial activity against Staphylococcus aureus and Escherichia coli. In order to avoid cumulative toxicity of the remained mesoporous silica nanoparticles (MSNs) in biological systems, three amino acids (aâs), i.e., glycine (Gly), aspartic acid (Asp) and cysteine (Cys), were incorporated into MSNs for accelerating their biodegradation. Aâ was conjugated with 3-isocyanatopropyl triethoxysilane (ICPTES) before reacting with tetraethyl orthosilicate to form aâ-MSNs via the sol-gel process based co-condensation. FTIR spectra confirmed the urea bond formation in aâ-ICPTES, whereas silicon resonances of T2 and T3 in 29Si NMR spectrum indicated the incorporation of aâ in MSNs. Spherical bare-MSNs (112 nm) were obtained while the rod-like particles were formed in the case of Gly-MSNs (73 nm in length), Asp-MSNs (90 nm in length), and Cys-MSNs (163 nm in length). The trend of %Si dissolution rate analyzed from microwave plasma-atomic emission spectrometer (MP-AES) of aâ-MSNs in phosphate buffer saline (PBS)/trypsin enzyme (pH 7.4) was 3–5 times higher than in PBS (pH 7.4) and 7–8 times higher than in acetate buffer (pH 5.2), respectively. The Asp-MSNs having two carboxylic groups showed the highest degradability, followed by Cys-MSNs, Gly-MSNs, and bare-MSNs in all three media. The objectives were to identify the functional domains of a potential oncoprotein, cell migration inducing hyaluronidase 2 (CEMIP2), evaluate its expression levels and roles in colorectal cancer (CRC), and develop an aptamer-based nanoparticle for targeted therapy. Data mining on TCGA identified that CEMIP2 might play oncogenic roles in CRC. In a local cohort, CEMIP2 mRNA levels significantly stepwise increase in CRC patients with higher stages, and high CEMIP2 confers worse disease-free survival. In addition, CEMIP2 mRNA levels significantly correlated to hyaluronan levels in sera from CRC patients. Deletion mapping identified that CEMIP2 containing G8 and PANDER-like domains preserved hyaluronidase activity and oncogenic roles, including cell proliferation, anchorage-independent cell growth, cell migration and invasion, and human umbilical vein endothelial cell (HUVEC) tube formation in CRC-derived cells. A customized monoclonal mouse anti-human CEMIP2 antibody probing the PANDER-like domain (anti-289307) counteracted CEMIP2-mediated carcinogenesis in vitro. Cell-SELEX pinpointed an aptamer, aptCEMIP2(101), specifically interacted with the full-length CEMIP2, potentially involving its 3D structure. Treatments with aptCEMIP2(101) significantly reduced CEMIP2-mediated tumorigenesis in vitro. Mesoporous silica nanoparticles (MSN) carrying atpCEMIP2(101) and Dox were fabricated. Bone related infections elicit excessive inflammatory responses, progressive osteolysis, and persistent bacterial load, which impede bone regeneration. Conventional antibiotics can mitigate bacterial load; however, they fail to inhibit bone resorption and the inflammatory response. To enhance bone regeneration during bacterial infection, an optimal therapeutic strategy should not only eradicate bacteria but also inhibit inflammation and osteolysis. In this study, we fabricated a multi-biofunctional coating, designated MSNs-BMP4-EN, by covalently immobilizing mesoporous silica nanoparticles (MSNs) with human bone morphogenetic protein 4 (BMP4) and loading with enoxacin (EN) for antibacterial effect with inhibiting inflammation and osteolysis. Antimicrobial assessments revealed that MSNs-BMP4-EN markedly obstructed bacterial adhesion and colonization. Subsequent in vitro assessments demonstrated that it facilitated osteogenic differentiation of bone mesenchymal stem cell and impeded early osteoclastogenesis. Moreover, MSNs-BMP4-EN effectively restrained the secretion of pro-inflammatory mediators, thereby counteracting the infection-induced inflammatory milieu and promoting bone regeneration. Collectively, these findings indicate that MSNs-BMP4-EN exhibits excellent antibacterial, anti-inflammatory, and bone resorption inhibitory attributes during initial stages of bone-related infection, thereby further improving bone regeneration. In order to optimize the chemotherapeutic efficacy of doxorubicin (DOX) and improve the photodynamic therapeutic effectiveness of rose bengal (RB), a mesoporous silica nanoparticle system was designed as the carrier of RB and DOX for chemo-photodynamic combination therapy. A pH-sensitive strategy has been exploited to enhance the delivery efficiency. Our results suggested that the production of singlet oxygen was independent of the release of RB while strongly influenced by the external DOX layer. This method showed several benefits, including accelerating cellular uptake of the payloads and enabling chemo-photodynamic combination therapy for synergistic cancer treatment. Mesoporous silica nanoparticles (MSN) have several advantages as carriers for drug delivery, including high drug loading capacity, good biocompatibility, excellent stability, and easily tailorable surface properties. This study describes the design of an MSN-based carrier for use in a heat shock responsive drug delivery system. MSN were functionalized with the temperature-sensitive PEG/PCL multiblock copolymer, as gatekeepers, allowing the release of entrapped drugs in response to heat shock stimuli (MBC-MSN). In the absence of heat shock, doxorubicin (Dox)-loaded MBC-MSN showed very low cytotoxicity, as PEG/PCL inhibited the premature release of Dox from MBC-MSN. In response to heat-shock stimuli, however, these Dox@MBC-MSN showed significant cytotoxicity, similar to that of free Dox. Dox release was due to the loosening of the structure of the gatekeeper, PEG/PCL, in response to the heat shock stimuli. Amino-functionalized mesoporous silica nanoparticles (MSN-NH2) were synthesized by a post-grafting method and further studied as carriers for doxorubicin hydrochloride (DOX) delivery. The morphology, structure, and property of MSN-NH2 and DOX-loaded MSN-NH2 (DOX@MSN-NH2) were studied using various techniques, such as transmission electron microscopy, Fourier transformed infrared spectroscopy, N2 adsorption-desorption isotherms, and zeta potentials. The drug loading and release profile as well as the in vitro cell cytotoxicity were detaily investigated. The results indicated that the loading content of DOX increased with the decrease of MSN-NH2/DOX mass ratio and/or the increase of amino density. DOX@MSN-NH2 exhibited a pH-dependent drug release, drug release increased as the pH value decreased. Compared with MSN-NH2, which were neglectable cytotoxicity against non-small-cell lung cancer (A549) cells, DOX@MSN-NH2 displayed remarkable cytotoxicity toward A549 cells in dose- and time-dependent manners. Colorectal cancer (CRC) is one of the leading causes of cancer-deaths worldwide. Methods for the early in situ detection of colorectal adenomatous polyps and their precursors – prior to their malignancy transformation into CRC – are urgently needed. Unfortunately at present, the primary diagnostic method, colonoscopy, can only detect polyps and carcinomas by shape/morphology; with sessile polyps more likely to go unnoticed than polypoid lesions. Here we describe our development of polyp-targeting, fluorescently-labeled mesoporous silica nanoparticles (MSNs) that serve as targeted endoscopic contrast agents for the early detection of colorectal polyps and cancer. In vitro cell studies, ex vivo histopathological analysis, and in vivo colonoscopy and endoscopy of murine colorectal cancer models, demonstrate significant binding specificity of our nanoconstructs to pathological lesions via targeting aberrant α-L-fucose expression. Our findings strongly suggest that lectin-functionalized fluorescent MSNs could serve as a promising endoscopic contrast agent for in situ diagnostic imaging of premalignant colonic lesions. Composite levofloxacin (LVF)/nanofibers have been fabricated through electrospinning. Slow release was achieved by covalently binding LVF to mesoporous silica nanoparticles (MSN) through a cleavable thioester bond and then blending the MSN into poly(ϵ-caprolactone) (PCL) nanofibers. Conjugated LVF–MSN was characterized by FTIR, DSC, TGA, and solid-state C13NMR. The structure of composite nanofibers was studied by scanning electron microscopy (SEM). Drug release profiles showed that burst release was decreased from 59% in the uniform PCL/LVF electrospun mats to 20% in the PCL/conjugated LVF–MSN mats after 1 day in phosphate buffer at 37°C, and gradual release in the latter was observed over the next 13 days. This slow release is due to the cleavable bond between LVF and MSN that can be hydrolyzed over a time and results in slow release of LVF. Over the past decade, mesoporous silica nanoparticles (MSNs) smaller than 200 nm with a high colloidal stability have been extensively studied for systemic drug delivery. Although small molecule delivery via MSNs has been successful, the encapsulation of large therapeutic biomolecules, such as proteins or DNA, is limited due to small pore size of the conventional MSNs obtained by soft-templating. Here, we report the synthesis of mesoporous silica nanoparticles with extra-large pores (XL-MSNs) and their application to in vivo cytokine delivery for macrophage polarization. Uniform, size-controllable XL-MSNs with 30 nm extra-large pores were synthesized using organic additives and inorganic seed nanoparticles. XL-MSNs showed significantly higher loadings for the model proteins with different molecular weights compared to conventional small pore MSNs. XL-MSNs were used to deliver IL-4, which is an M2-polarizing cytokine and very quickly degraded in vivo, to macrophages and polarize them to anti-inflammatory M2 macrophages in vivo. XL-MSNs induced a low level of reactive oxygen species (ROS) production and no pro-inflammatory cytokines in bone marrow-derived macrophages (BMDMs) and in mice injected intravenously with XL-MSNs. We found that the injected XL-MSNs were targeted to phagocytic myeloid cells, such as neutrophils, monocytes, macrophages, and dendritic cells. Finally, we demonstrated that the injection of IL-4-loaded XL-MSNs successfully triggered M2 macrophage polarization in vivo, suggesting the clinical potential of XL-MSNs for modulating immune systems via targeted delivery of various cytokines. Herein, we have formulated folic acid conjugated polyacrylic acid capped mesoporous silica nanoparticles (∼47.6 nm in diameter) for pH-dependent targeted delivery of chrysin to breast cancer (MCF-7) cells. Chrysin loaded mesoporous silica nanoparticles (Chr- mSiO 2 @PAA/FA) have been noted to induce apoptosis in MCF-7 cells through oxidative insult and mitochondrial dysfunction with subsequent G 1 arrest. Further, in tumor bearing mice, intravenous incorporation of Chr-mSiO 2 @PAA/FA has been noticed to enhance the anti-neoplastic effects of chrysin via tumor site-specific accumulation. Enhanced cytotoxicity of chrysin contributed towards in vivo tumor regression, restoration of normalized tissue architecture and maintenance of healthy body weight. Besides, no serious systemic toxicity was manifested in response to Chr-mSiO 2 @PAA/FA administration in vivo. Mesoporous silica nanoparticles (MSNs) are known as carriers with high loading capacity and large functionalizable surface area for target-directed delivery. In this study, a series of docetaxel-loaded folic acid- or methionine-functionalized mesoporous silica nanoparticles (DTX/MSN-FA or DTX/MSN-Met) with large pores and amine groups at inner pore surface properties were prepared. The results showed that the MSNs were successfully synthesized, having good pay load and pH-sensitive drug release kinetics. The cellular investigation on MCF-7 cells showed better performance of cytotoxicity and cell apoptosis and an increase in cellular uptake of targeted nanoparticles. In vivo fluorescent imaging on healthy BALB/c mice proved that bare MSN-NH2 are mostly accumulated in the liver but MSN-FA or MSN-Met are more concentrated in the kidney. Importantly, ex vivo fluorescent images of tumor-induced BALB/c mice organs revealed the ability of MSN-FA to reach the tumor tissues. Central nervous system (CNS) drug delivery is often hampered due to the insidious nature of the blood-brain barrier (BBB). Nose-to-brain delivery via olfactory pathways have become a target of attention for drug delivery due to bypassing of the BBB. The antioxidant properties of phytochemicals make them promising as CNS active agents but possess poor water solubility and limited BBB penetration. The primary aim of this study was the development of mesoporous silica nanoparticles (MSNs) loaded with the poorly water-soluble phytochemicals curcumin and chrysin which could be utilised for nose-to-brain delivery. We formulated spherical MSNP using a templating approach resulting in ∼220 nm particles with a high surface porosity. Curcumin and chrysin were successfully loaded into MSNP and confirmed through Fourier transformation infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA) and HPLC approaches with a loading of 11–14% for curcumin and chrysin. Release was pH dependant with curcumin demonstrating increased chemical stability at a lower pH (5.5) with a release of 53.2% ± 2.2% over 24 h and 9.4 ± 0.6% for chrysin. MSNP were demonstrated to be non-toxic to olfactory neuroblastoma cells OBGF400, with chrysin (100 μM) demonstrating a decrease in cell viability to 58.2 ± 8.5% and curcumin an IC 50 of 33 ± 0.18 μM. Furthermore confocal microscopy demonstrated nanoparticles of <500 nm were able to accumulate within cells with FITC-loaded MSNP showing membrane localised and cytoplasmic accumulation following a 2 h incubation. MSNP are useful carriers for poorly soluble phytochemicals and provide a novel vehicle to target and deliver drugs into the CNS and bypass the BBB through olfactory drug delivery. Peroxidase mimetic catalytic atom transfer radical polymerization (ATRP) was first used to install tertiary amine-functionalized polymer brushes on the surface of mesoporous silica nanoparticles (MSNs) in a facile and highly efficient manner. Poly(N,N-dimethylaminoethyl methacrylate) (PDMAEMA) brushes-grafted MSNs were fabricated by biocompatible deuterohemin-β-Ala-His-Thr-Val-Glu-Lys (DhHP-6)-catalyzed surface-initiated ATRP (SI-ATRP). The resulting organic-inorganic hybrid nanocarriers were fully characterized by Fourier transform-infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA), X-ray photoelectron spectroscopy (XPS), powder X-ray diffraction (XRD), SEM, TEM, Elemental analysis, Zeta-potential, and N2 adsorption-desorption isotherms, which demonstrated the successful coating of pH-responsive polymers on the MSN surface. Rhodamine 6G (Rh6G) dyes were further loaded within the mesopores of this nanocarrier, and the release of Rh6G out of MSNs in a controlled fashion was achieved upon lowing the solution pH. The electrostatic repulsion of positively-charged tertiary ammonium of PDMAEMAs in acidic environments induced the stretching out of polymer brushes on MSN surfaces, thus opening the gates to allow cargo diffusion out of the mesopores of MSNs. Small-molecule drug combination therapies are an attractive approach to enhancing cancer chemotherapeutic responses. Therefore, this study aimed to investigate the potential of axitinib (AXT) and celastrol (CST) in targeting angiogenesis and mitochondrial-based apoptosis in cancer. Therefore, we prepared AXT/CST-loaded combination nanoparticles (ACML) with CST loaded in the mesoporous silica nanoparticles (MSN) and AXT in PEGylated lipidic bilayers. We showed that ACML effectively inhibited angiogenesis and mitochondrial function and was efficiently internalized in SCC-7, BT-474, and SH-SY5Y cells. Furthermore, hypoxia-inducible factor (HIF)-1α expression, which increased under hypoxic conditions in all cell lines exposed to ACML, markedly decreased, which may be critical for tumor inhibition. Western blotting showed the superior anticancer effect of combination nanoparticles in different cancer cells. Compared to the cocktail (AXT/CST), ACML induced synergistic cancer cell apoptosis. The AXT/CST-based combination nanoparticle synergism might be mediated by AXT, which controls vascular endothelial growth factor receptors while CST acts on target cell mitochondria. Importantly, ACML-treated mice showed remarkably higher tumor inhibition (64%) than other groups did in tumor xenograft models. Tumor xenograft immunohistochemistry revealed elevated caspase-3 and poly (ADP-ribose) polymerase and reduced CD31 and Ki-67 expression, clearly suggesting tumor apoptosis through mitochondrial and antiangiogenic effects. Overall, our results indicate that ACML potentially inhibited cell proliferation and induced apoptosis by blocking mitochondrial function, leading to enhanced antitumor efficacy. The enzymatic production of biodiesel by methanolysis of canola oil was studied using self-made biocatalysts. Mesoporous SBA-15 nanoparticles were prepared, characterized and functionalized by 3-glycidyloxypropyl trimethoxysilane. Lipases from Candida antarctica (CALB), Thermomyces lanuginosus (TLL) and Rhizomucor miehei (RML) were covalently immobilized onto SBA-epoxy. Thermal stability and the influence of methanol concentration on the catalytic activity were also evaluated. Higher thermal stability and methanol tolerance for immobilized derivatives were achieved compared to the free enzyme. In an optimization study, the effect of water, t -butanol and blue silica gel as water adsorbent on the yield of FAME was considered. For the SBA-RML catalysed reaction, water had little effect in increasing FAME yield, but when 20 wt% water by substrate weight was added to the SBA-TLL catalysed reaction, the methyl ester content reached nearly complete conversion (98% FAME). t -Butanol had a great effect on yield, with almost complete conversion for SBA-RML and SBA-TLL. The effect of blue silica gel also was investigated. The immobilized TLL was quite stable and can be reused for 20 cycles without significant loss in activity (6%). Novel bilayer coated mesoporous silica nanoparticle (MCM-41) based on pH sensitive poly(acrylic acid-co-itaconic acid) and human serum albumin (HSA) was designed for controlled delivery of gemcitabine (anticancer drug) to cancer cells. The shell around the mesoporous silica has bilayer structure. Poly(acrylic acid-co-itaconic acid) was used as pH-sensitive inner shell and human serum albumin, HSA, was used as outer shell. The core–shell structure was formed due to electrostatic interaction between ammonium groups of modified MCM-41 and carboxylate groups of copolymer. Also, the albumin layer was wrapped around the copolymer coated nanoparticle by electrostatic interaction between ammonium groups from protein and carboxylate ions of copolymer shell. Moreover, the maximum release occurred at pH 5.5 (pH of endosomes) because the bilayer shell collapsed at this pH. In this paper, a multifunctional theranostic magnetic mesoporous silica nanoparticle (MMSN) with magnetic core was developed for magnetic-enhanced tumor-targeted MR imaging and precise therapy. The gatekeeper β-cyclodextrin (β-CD) was immobilized on the surface of mesoporous silica shell via platinum(IV) prodrug linking for reduction-triggered intracellular drug release. Then Arg-Gly-Asp (RGD) peptide ligand was further introduced onto the gatekeeper β-CD via host–guest interaction for cancer targeting purpose. After active-targeting endocytosis by cancer cells, platinum(IV) prodrug in MMSNs would be restored to active platinum(II) drug in response to the innative reducing microenvironment in cancer cells, resulting in the detachment of β-CD gatekeeper and thus simultaneously triggering the in situ release of anticancer drug doxorubicin (DOX) entrapped in the MMSNs to kill cancer cells. It was found that with the aid of an external magnetic field, drug loaded MMSNs showed high contrast in MR imaging in vivo and exhibited magnetically enhanced accumulation in the cancer site, leading to significant inhibition of cancer growth with minimal side effects.
