The objective of this study was to investigate the effects of yucca extract (YE) supplementation on estrus, nutrient digestibility, antioxidant capacity and fecal microorganisms of gilts. Twenty gilts were randomly divided into two groups: basal diet (CONT) and basal diet + 0.25 g/kg YE (YETG). The results showed that supplementing 0.25 g/kg YE in the diet of gilts significantly increased the apparent digestibility of dietary energy, crude fat and crude protein (p < 0.05). In addition, YE could also improve the antioxidant capacity of gilts, significantly increase the serum total antioxidant capacity (T-AOC) activity and decrease the malondialdehyde (MDA) content of gilts (p < 0.05). In terms of fecal microorganisms, YE significantly increased the Shannon index and Simpson index of fecal microorganisms of gilts (p < 0.05), decreased the abundance of Proteobacteria, Actinobacteriota and Streptococcus sp., and increased the abundance of Muribaculaceae and Prevotalla sp. in the feces of gilts (p < 0.05). In conclusion, dietary YE increased the apparent digestibility of nutrients, improved the antioxidant status of gilts and increased the α diversity of fecal microorganisms. These results provide a reference for the application of YE in gilts production.

Pectin is a proven prebiotic and widely used in human health products. This study aims to assess the impact of dietary pectin supplementation during gestation on sow vaginal microbiota and the offspring's intestinal composition. Thirty sows were randomly allocated to two groups and fed a standard diet (CON) or a standard diet supplemented with 3 g/kg pectin (PEC). Blood, feces, and vaginal swab samples from the sows and blood, intestines issue, and colonic content samples from the offspring were collected and analyzed. The results indicate that the relative abundance of vaginal Lactobacillus was notably enhanced in the PEC group and fecal β-glucuronidase (β-G) activity and plasma 17β-estradiol (E2) concentration were also significantly increased in the PEC group. Newborn piglets were found to host different microbial communities as well. At the phylum level, Proteobacteria dominated in the CON group, and Firmicutes was predominant in the PEC group. Newborn piglets in the PEC group had a lower interleukin-6 (IL-6) concentration in their plasma. The expression of intestinal cytokines of offspring was improved as well. Villus height and villus height/crypt depth (V/C) in the PEC group were extremely higher than those in the CON group. In conclusion, dietary pectin supplementation can be of benefit to both sows and newborn piglets.

Abstract Background Two studies were designed to determine standard ileal crude protein (CP) and amino acid (AA) digestibility of soybean meal (SBM) from different origins fed to non-pregnant and pregnant sows. Seven solvent-extracted SBMs from soybeans produced in the USA, Brazil, and China were selected. In Exp. 1, eight different diets were created: a nitrogen (N)-free diet and 7 experimental diets containing SBM from different origins as the only N source. Eight non-pregnant, multiparous sows were arranged in an 8 × 8 Latin square design (8 periods and 8 diets). In Exp. 2, the diet formula was the same as in Exp. 1. Eight gestating sows (parity 3) were assigned to 4 different diets in a replicated 4 × 3 Youden square design (three periods and four diets) in mid-gestation and again in late-gestation stages. Results When fed to non-pregnant and late-gestating sows, the standardized ileal digestibility (SID) of CP and most AAs from different SBM were not significantly different (P > 0.05). Abstract Background The feed intake of sows during lactation is often lower than their needs. High-fiber feed is usually used during gestation to increase the voluntary feed intake of sows during lactation. However, the mechanism underlying the effect of bulky diets on the appetites of sows during lactation have not been fully clarified. The current study was conducted to determine whether a high-fiber diet during gestation improves lactational feed intake (LFI) of sows by modulating gut microbiota. Methods We selected an appropriate high-fiber diet during gestation and utilized the fecal microbial transplantation (FMT) method to conduct research on the role of the gut microbiota in feed intake regulation of sows during lactation, as follows: high-fiber (HF) diet during gestation (n = 23), low-fiber (LF) diet during gestation (n = 23), and low-fiber diet + HF-FMT (LFM) during gestation (n = 23). Results Compared with the LF, sows in the HF and LFM groups had a higher LFI, while the sows also had higher peptide tyrosine tyrosine and glucagon-like peptide 1 on d 110 of gestation (G110 d). The litter weight gain of piglets during lactation and weaning weight of piglets from LFM group were higher than LF group. Sows given a HF diet had lower Proteobacteria, especially Escherichia-Shigella, on G110 d and higher Lactobacillus, especially Lactobacillus_mucosae_LM1 and Lactobacillus_amylovorus, on d 7 of lactation (L7 d). The abundance of Escherichia-Shigella was reduced by HF-FMT in numerically compared with the LF. In addition, HF and HF-FMT both decreased the perinatal concentrations of proinflammatory factors, such as endotoxin (ET), lipocalin-2 (LCN-2), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β). The concentration of ET and LCN-2 and the abundance of Proteobacteria and Escherichia-Shigella were negatively correlated with the LFI of sows. Conclusion The high abundance of Proteobacteria, especially Escherichia-Shigella of LF sows in late gestation, led to increased endotoxin levels, which result in inflammatory responses and adverse effects on the LFI of sows. Adding HF during gestation reverses this process by increasing the abundance of Lactobacillus, especially Lactobacillus_mucosae_LM1 and Lactobacillus_amylovorus.

This study aimed to investigate the effects of the dietary fiber pectin on the gut microbiota and health of parturient sows. A total of 30 parity 5–7, multiparous gestation sows (Large White × Landrace) were randomly assigned to two treatment groups after mating: Con (control, basic diet) and Pec (pectin, 3%). The sows received the two diets during gestation, and all sows were fed the same standard basic diet during lactation. The results of β-diversity showed that the composition of the gut microbiota was different in the Con and Pec groups. Compared with the sows in the Con group, the Pec sows showed a higher abundance of the gut bacteria Clostridium and Romboutsia and a lower abundance of harmful bacteria (Micrococcaceae, Coriobacteriaceae, Dorea, Actinomyces). On the other hand, the SCFA plasma concentration was increased in the Pec group, while pro-inflammatory cytokine (IL-6, IL-1β, and TNF-α) concentrations were decreased. In conclusion, the soluble dietary fiber pectin could improve the reproductive performance and health of sows by increasing the abundance of some commensal bacteria enhancing the metabolite SCFA levels and reducing the pro-inflammatory cytokine plasma levels.

The purpose of this study is to investigate the effects of supplementing Yeast−derived postbiotics (Y−dP) to the diet of sows during late pregnancy and lactation on fecal microbiota and short−chain fatty acids (SCFA) in sows and their offspring weaned piglets, as well as the relationship between gut microbiota and SCFA, serum cytokines, and sow reproductive performance. A total of 150 sows were divided into three groups: control diet (CON), CON + Y−dP 1.25 g/kg, and CON + Y−dP 2 g/kg. The results showed that supplementing 0.125% Y−dP to the diet of sows can increase the content of isobutyric acid (IBA) in the feces of pregnant sows and reduce the content of butyric acid (BA) in the feces of weaned piglets (p < 0.05). The fecal microbiota of pregnant sows β diversity reduced and piglet fecal microbiota β diversity increased (p < 0.05). Y−dP significantly increased the abundance of Actinobacteria and Limosilactobacilli in the feces of pregnant sows (p < 0.05), as well as the abundance of Verrucomicrobiota, Bacteroidota, and Fusobacteriota in the feces of piglets (p < 0.05). The abundance of Bacteroidota in the feces of pregnant sows is positively correlated with propionic acid (PA) (r > 0.5, p < 0.05). The abundance of Prevotellaceae_NK3B31_group was positively correlated with Acetic acid (AA), PA, Valerate acid (VA), and total volatile fatty acid (TVFA) in the feces of pregnant sows (r > 0.5, p < 0.05), and Bacteroidota and Prevotellaceae_NK3B31_group were negatively correlated with the number of stillbirths (r < −0.5, p < 0.05). The abundance of Lactobacillus and Holdemanella in piglet feces was positively correlated with TVFA in feces and negatively correlated with IgA in serum (r > 0.5, p < 0.05). In conclusion, supplementing Y−dP to the diet of sows from late gestation to lactation can increase the chao1 index and α diversity of fecal microorganisms in sows during lactation, increase the abundance of Actinobacteria and Limosilactobacilli in the feces of sows during pregnancy, and increase the abundance of beneficial bacteria such as Bacteroidetes in piglet feces, thereby improving intestinal health. These findings provide a reference for the application of Y−dP in sow production and a theoretical basis for Y−dP to improve sow production performance.

Animal feeds contain a substantial number and diversity of microorganisms, and some of them have pathogenic potential. The objectives of this study were to investigate the effects of different doses of gamma (γ)-ray irradiation on the bacteria count in different types of feed and then to test the effect of γ-ray-irradiation-treated fishmeal on the gut health and growth performance of weaning piglets. In trial 1, three fishmeal samples, two feather meal samples, three meat meal samples, three soybean meal samples, and three vitamin complexes were treated with γ-ray irradiation doses of 0, 3, 6, or 9 kGy. The 6 and 9 kGy doses eliminated most of the bacteria in the feed but also resulted in a loss of vitamin C and B1. In trial 2, 96 weaning piglets were fed one of the following three diets with eight replicates (pens) per group over a 14-day period: (1) the control diet—the basal diet supplemented with 6% fishmeal with a low bacteria count (40 CFU/g) and no E. coli; (2) the fishmeal-contaminated diet (FM-contaminated) diet—the basal diet supplemented with 6% fishmeal with a high bacteria count (91,500 CFU/g) and E. coli contamination; and (3) the irradiated fishmeal (irradiated FM) diet—the basal diet supplemented with γ-ray-irradiation-treated E. coli-contaminated fishmeal. The piglets that received the FM-contaminated diet had significantly lower average daily gain and a greater diarrhea index compared to those fed the control diet, whereas γ-ray irradiation treatment abrogated the negative effect of the E. coli-contaminated fishmeal. Collectively, γ-ray irradiation at a dose of 6–9 kGy was sufficient to eliminate the microorganisms in the feed, thereby benefitting the growth performance and gut health of the weaning piglets.

Abstract Background Dietary fiber (DF) is often eschewed in swine diet due to its anti-nutritional effects, but DF is attracting growing attention for its reproductive benefits. The objective of this study was to investigate the effects of DF intake level on oocyte maturation and uterine development, to determine the optimal DF intake for gilts, and gain microbial and metabolomic insight into the underlying mechanisms involved. Methods Seventy-six Landrace × Yorkshire (LY) crossbred replacement gilts of similar age (92.6 ± 0.6 d; mean ± standard deviation [SD]) and body weight (BW, 33.8 ± 3.9 kg; mean ± SD) were randomly allocated to 4 dietary treatment groups (n = 19); a basal diet without extra DF intake (DF 1.0), and 3 dietary groups ingesting an extra 50% (DF 1.5), 75% (DF 1.75), and 100% (DF 2.0) dietary fiber mixture consisting of inulin and cellulose (1:4). Oocyte maturation and uterine development were assessed on 19 d of the 2nd oestrous cycle. Microbial diversity of faecal samples was analysed by high-throughput pyrosequencing (16S rRNA) and blood samples were subjected to untargeted metabolomics. Results The rates of oocytes showing first polar bodies after in vitro maturation for 44 h and uterine development increased linearly with increasing DF intake; DF 1.75 gilts had a 19.8% faster oocyte maturation rate and a 48.9 cm longer uterus than DF 1.0 gilts (P 0.1%) increased linearly or quadratically with increasing DF intake. Untargeted metabolic analysis revealed significant changes in serum metabolites that were closely associated with microbiota, including serotonin, a gut-derived signal that stimulates oocyte maturation. Conclusions The findings provide evidence of the benefits of increased DF intake by supplementing inulin and cellulose on oocyte maturation and uterine development in gilts, and new microbial and metabolomic insight into the mechanisms mediating the effects of DF on reproductive performance of replacement gilts.

This study was conducted to investigate the effects of dietary multi-enzyme (multi-carbohydrase and phytase complex, MCPC) supplementation on digestibility, growth performance, bone mineralization, and carcass yield and traits in growing–finishing pigs fed diets with adequate or deficient net energy (NE), amino acids (AA), calcium (Ca) and phosphorus (P) levels. A total of 576 crossbred [Duroc × (Landrace × Yorkshire)] barrows (~25 kg) were fed one of the six diets till live weight approached 130 kg. Basal diets included a positive control (PC), negative control 1 (NC1) and 2 (NC2), while another three diets were prepared by adding MCPC to the three basal diets. The final body weight was lower (p < 0.05) in NC2 than in NC1 and PC treatments, while overall feed intake and feed-gain ratio were higher (p < 0.05) in NC1 and NC2 than in PC treatment. The NC2 treatment showed lower (p < 0.05) carcass weight but higher (p < 0.05) lean meat percentage than the PC treatment. The apparent ileal digestibility (AID) of gross energy (GE), crude protein (CP) and AA was decreased (p < 0.05) or tended (p < 0.10) to decrease in NC1 and/or NC2 diets compared with a PC diet. MCPC supplementation improved (p < 0.05) AID of Ca, P and AA (Lys, Leu, Val, Phe, Gly, Tyr and Pro), apparent total-tract digestibility (ATTD) of GE, CP, bone strength, Ca, and P retention. In conclusion, MCPC supplementation improved nutrient digestibility, bone mineralization, and growth performance of fattening pigs, regardless of the nutritional level of the basal diet.

Abstract Background Replacement gilts are typically fed ad libitum, whereas emerging evidence from human and rodent studies has revealed that time-restricted access to food has health benefits. The objective of this study was to investigate the effect of meal frequency on the metabolic status and ovarian follicular development in gilts. Methods A total of 36 gilts (Landrace × Yorkshire) with an age of 150±3 d and a body weight of 77.6±3.8 kg were randomly allocated into one of three groups (n = 12 in each group), and based on the group allocation, the gilts were fed at a frequency of one meal (T1), two meals (T2), or six meals per day (T6) for 14 consecutive weeks. The effects of the meal frequency on growth preference, nutrient utilization, short-chain fatty acid production by gut microbial, the post-meal dynamics in the metabolic status, reproductive hormone secretions, and ovarian follicular development in the gilts were measured. To study the effects of energy and dietary fiber on breast development in gilts and its possible mechanisms, 32 gilts (Landrace × Yorkshire) were randomly allocated into a 2 × 2 factorial design to receive a diet with low or high energy [LE: 33.37 MJ/d digestible energy (DE); HE: 41.87 MJ/d DE] and low or high fiber (LF: 0.3 kg/d dietary fiber, HF: 0.6 kg/d dietary fiber). The weight of breast tissue was recorded. The mammary glands were collected for further analyses. The high energy intake increased the relative weight of breast tissue (p < 0.05) and the content of breast fat (p < 0.05). At the same time, the oil red staining of breast slices also showed an increase in breast fat content in high-energy treatment. High energy intake increased the DNA concentration in breast tissues (p < 0.05). In addition, high energy intake increased the concentration of triglycerides, free fatty acids, and total cholesterol in the blood of gilts (p < 0.05), and the supplementation of high fiber tended to reduce free fatty acids, total cholesterol, and estradiol (p < 0.1). Proteomic analysis suggested that there were notable differences in the cytoskeleton, intracellular non–membrane-bounded organelle, apoptosis, receptor activity, and endopeptidase inhibitor activity in molecular function between the energy and fiber effects (p < 0.05). High fiber intake also decreased the mRNA expression of 5-HT7, Bax, and caspase-3 in the breast tissue of gilts (p < 0.05), which further confirmed the importance of fiber in regulating breast development in gilt. Our results indicate that increasing gilt energy intake improved breast weight and fat deposition and increased breast cell apoptosis. Increased fiber intake reduced breast fat deposition and breast cell apoptosis at high energy intake in gilts. These results provide a potential strategy for dietary intervention against high energy intake in gilts and even in humans.

This study investigated the effects of dietary choline levels during gestation on reproductive performance of sows. In addition, the plasma metabolome and gut microbiota of sows was studied. A total of 260 multiparous sows were allocated to five dietary treatment groups with increasing choline concentrations (1,050, 1,450, 1,850, 2,250, and 2,650 mg/kg) in a randomized complete block design. The sows were fed experimental diets from breeding until farrowing and a common lactating diet during lactation. The results showed that the backfat (BF) gain of sows during gestation, individual birth weight for total piglets born, piglets born alive, average piglet weight at weaning increased linearly (P < 0.05), whereas the within-litter birth weight variation coefficient (CV) of piglets born alive and suckling piglet mortality decreased linearly (P < 0.05) as dietary choline level increased. A quadratic effect of dietary choline level was observed for the average daily feed intake (ADFI) of sows during lactation (P < 0.05). ADFI was maximized when the dietary choline concentration reached 1,910 mg/kg. Plasma H2O2 concentration at day 30 of gestation in the 1,050 mg/kg group was greater than that in the 1,850 and 2,650 mg/kg groups (P < 0.05). Plasma metabolomics identified 46 metabolites among the three groups. Specifically, plasma concentrations of trimethylamine-N-oxide (TMAO), dopamine, and L-proline increased while 1-methylhistidine concentration decreased as dietary choline levels increased. In addition, bacterial observed species and richness (Chao 1 and ACE) at day 110 of gestation decreased as dietary choline levels increased (P < 0.05). For the gut microbiota composition, the enhanced dietary choline level decreased the abundance of phylum Proteobacteria (P < 0.05) and increased the abundance of phylum Actinobacteria (P < 0.05) at day 30 of gestation. Compared with the 1,050 mg/kg group, the abundance of genus Terrisporobacter was less in the 1,850 mg/kg group, and genera Bacillus and Cellulomonas were greater in the 2,650 mg/kg group. In summary, increasing dietary choline levels improved the birth weight, uniformity of neonatal piglets and litter performance during lactation. This may be associated with better antioxidant capability, metabolic status, and gut microbiota of sows during gestation.

Abstract Background This study aimed to investigate the effects of oral administration of Enterococcus faecium NCIMB 10415 (E. faecium) on intestinal development, immunological parameters and gut microbiota of neonatal piglets challenged with enterotoxigenic Escherichia coli K88 (ETEC). A total of 96 1-day-old sow-reared piglets were randomly assigned to 2 groups, with 48 piglets in each group. The piglets were from 16 litters (6 piglets each litter), and 3 piglets each litter were allocated to the E. faecium-supplemented (PRO) group, while the other 3 piglets were allocated to the control (CON) group. After colostrum intake, piglets in the PRO group were orally administrated with 3 × 109 CFU E. faecium per day for a period of one week. On day 8, one piglet per litter from each group was challenged (CON+ETEC, PRO+ETEC) or not (CON-ETEC, PRO-ETEC) with ETEC in a 2 × 2 factorial arrangement of treatments. On day 10 (2 days after challenge), blood and tissue samples were obtained from piglets. Results Before ETEC challenge, there were no significant differences for the average daily gain (ADG) and fecal score between the two groups of piglets. After ETEC challenge, the challenged piglets had greater fecal score compared to the non-challenged piglets, whereas E. faecium administration was able to decrease the fecal score. Piglets challenged with ETEC had shorter villous height, deeper crypt depth, and reduced number of goblet cells in the jejunum and decreased mRNA abundance of claudin-1 in the ileum, whereas increased the percentage of lymphocytes, concentrations of IL-1β in the plasma and TNF-α in the ileal mucosa, as well as increased the mRNA abundances of innate immunity-related genes in the ileum tissue. These deleterious effects caused by ETEC were partly alleviated by feeding E. faecium. In addition, piglets in PRO-ETEC group had decreased the percentage of CD8+ T cells of the peripheral blood when compared to those in CON-ETEC group. Moreover, E. faecium administration increased Verrucomicrobia at phylum level and decreased Bilophila at genus level. Conclusions These results suggest that oral administration of E. faecium alleviated the intestinal injury and diarrhea severity of neonatal piglets challenged by ETEC, partly through improving the intestinal microbiota and immune response. This offers a potential strategy of dietary intervention against intestinal impairment by ETEC in neonatal piglets.

This study examined the effects of multi-strain probiotics (BL) and Perilla frutescens seed extract (PSE), alone or in combination, on weaning piglets. In total, 96 weaning piglets were allocated into four treatments: CON group (the basal diet), PSE group (basal diet + 1g/kg PSE), BL group (basal diet + 2 g/kg BL), and BL+PSE group (basal diet +1 g/kg PSE + 2 g/kg BL) according to a 2 × 2 factorial arrangement. The supplementation of BL or PSE improved the gain to feed ratio. Dietary BL reduced diarrhea occurrence and Escherichia coli, but increased Lactobacillus counts in the ileal digesta. Dietary PSE tended to increase Lactobacillus counts in the ileal digesta. Interactive effects were found in terms of ileal villus height, the gene expression of IL-1β, and malondialdehyde in the ileal mucosa. Dietary BL lowered malondialdehyde in the spleen, liver, and jejunal mucosa but increased the total antioxidant capacity (T-AOC) in the liver and ileum mucosa. The supplementation of PSE improved superoxide dismutase in serum and T-AOC in the liver, and reduced MDA in liver, spleen, and jejunum mucosa. Taken together, BL or PSE showed positive effects, improving growth and intestinal morphology and enhancing antioxidant capacity. However, their interaction showed no beneficial effects on the antioxidant indices and the intestinal morphology of weaned piglets.

Inflammatory responses reduce milk production in lactating sows. Silymarin may modulate inflammatory reactions. Here, we aimed to verify whether dietary silymarin supplementation could alleviate inflammatory responses in lactating sows through microbiota change in the gut. We also investigated how silymarin impacts inflammatory response in lactating sows. One hundred and ten sows were randomly assigned to a control diet (basal diet) or treatment diet (basal diet and 40 g/d silymarin) from the 108th day of gestation to weaning. Blood, milk, and feces from sows were collected for analysis. It was shown in the results that dietary silymarin supplementation decreased the level of pro-inflammatory cytokine IL-1β (p < 0.05) on the 18th day of lactation in the blood of the sows. Dietary silymarin supplementation tended to decrease (p = 0.06) somatic cell count in the colostrum of sows. Dietary silymarin supplementation reduced the gut bacterial community and the richness of the gut microbial community (p < 0.01) using 16S rRNA gene sequencing. The fecal microbes varied at different taxonomic levels in the lactating sows with silymarin supplementation. The most representative changes included an increase in the relative abundance of Fibrobacteres and Actinobacteria (p < 0.05) and tended to reduce the relative abundance of Spirochaetaes and Tenericutes (p = 0.09, 0.06) at the phylum level. It is suggested that dietary silymarin supplementation in late gestation until lactation has anti-inflammatory effects in lactation sow, which could be associated with the modulation of gut microbiota.

This study was aimed to explore how a high-quality diet or a flavor plus multi-enzyme diet affects the feed intake, nutrient digestibility and antioxidation capacity of lactating sows and the growth of their progeny. Thirty primiparous sows were randomly assigned to three treatments from d 2 of lactation until weaning (d 21): control (CON), with a basal diet; high quality (HQ), with 200 kcal/kg higher net energy than CON; or the CON diet supplemented with 500 mg/kg flavor and 100 mg/kg multi-enzymes (F + E). Sows fed with the HQ or F + E diets improved piglets’ live weight (p < 0.05) and average daily weight gain (p < 0.10), litter weight gain (p < 0.10) and piglet growth to milk yield ratio (p < 0.10). Compared with CON, the HQ and F + E groups increased the digestibility of ether extract, ash, neutral detergent fiber, crude fiber and phosphorus (p < 0.10), and the HQ group also increased dry matter, gross energy, crude protein, acid detergent fiber and energy intake (p < 0.05). Compared with CON, the F + E group decreased serum urea nitrogen and aspartate aminotransferase (p < 0.05) and enhanced superoxide dismutase, catalase and glutathione peroxidase, but it decreased malondialdehyde in milk supernatant (p < 0.05).

To investigate the effects of dietary fiber (DF), crude protein (CP) level, and gestation stage on nitrogen utilization, 28 Landrace-Yorkshire cross gestating sows at parity two were randomly divided into four dietary treatments with seven duplicates of one pig with a repeated-measures design. The diets comprised one with normal crude protein (CP) of 13.3%, one with a low CP diet of 10.1%, and two diets, one with dietary fiber (DF) supplementation of inulin and cellulose at the ratio of 1:1 and one without DF. The total litter size, litter size alive, and newborn birthweight of piglets did not differ between treatment groups. Sows that received high DF levels had greater nitrogen output in feces, lower urinary nitrogen, and increased nitrogen retention. Sows that received a low CP diet had reduced nitrogen excretion in feces and urine, lower nitrogen retention, and an unchanged nitrogen retention ratio. Sows at the late stage of gestation on days 95 to 98 had lower nitrogen excretion in urine and greater nitrogen retention than in the early stage of gestation on days 35 to 38, associated with a significant decrease in serum amino acids in late gestation. Maternal protein deposition was increased by high DF, decreased by low CP, and lower in late gestation compared with early gestation. Collectively, DF improved nitrogen utilization by decreasing urine nitrogen output, and nitrogen utilization increased as gestation advanced.

Abstract Background Weanling pigs, with immature immune system and physiological function, usually experience post-weaning diarrhea. This study determined the effects of dietary Clostridium butyricum supplementation on growth performance, diarrhea, and immunity of weaned pigs challenged with lipopolysaccharide (LPS). Methods In Experiment (Exp.) 1, 144 weaned piglets were weaned at 21 d and randomly assigned to six groups, with six replicates per group and four pigs per replicate, receiving a control diet (CON) or diet supplemented with antibiotics (AB) or C. butyricum (CB) (0.1%, 0.2%, 0.4%, or 0.8%), respectively. All diets in Exp. 1 were a highly digestible basal diet, with 3,000 mg/kg zinc oxide supplied in the first 2 wk only. In Exp. 2, 180 piglets were weaned at 21 d and randomly assigned to five groups, with six replicates per group and six pigs per replicate, receiving CON, AB, or CB (0.2%, 0.4%, or 0.6%) diets. The digestibility of diets was lower than those in Exp. 1, and did not include zinc oxide. At 36 d of Exp. 2, 12 piglets were selected from each of the CON and 0.4% CB groups, six piglets were intraperitoneally injected with LPS (50 μg/kg body weight) and the other six piglets with normal saline; animals were killed at 4 h after injection to collect blood, intestine, and digesta samples for biochemical analysis. To investigate the effects of the ratio of insoluble fiber to soluble fiber (ISF:SF) on sow performance and piglet intestinal development, we randomly assigned 64 gilts to four treatments comprising diets with the same level of dietary fiber, but different ISF:SF values of 3.89 (T1), 5.59 (T2), 9.12 (T3), and 12.81 (T4). At birth and weaning, six piglets per treatment at each phase were slaughtered for sampling. As ISF:SF increased, the mean piglet body weight (BW) at weaning and piglet BW gain, which were all significantly higher in T1 and T2 compared with T3 and T4 (p < 0.05), showed a linear decrease (p < 0.05); the crypt depth of the jejunum in weaned piglets linearly increased, whereas the duodenal weight, jejunal villus height, and villus height/crypt depth in newborn piglets and enzymatic activity of lactase, sucrase, and maltase linearly decreased (p < 0.05). No differences were observed in the yield and composition of milk (p > 0.05). Moreover, when the ISF:SF was 3.89 in gestation diets, higher piglet BW at weaning occurred, possibly because the ISF:SF affected development and enzymatic activity in the small intestine—effects related to digestion and absorption of nutrients—and consequently enhanced piglet BW gain.
