Behavioral embodied research shows that words evoking limb-specific meanings can affect responses performed with the corresponding body part. However, no study has explored this phenomenon's neural dynamics under implicit processing conditions, let alone by disentangling its conceptual and motoric stages. Here, we examined whether the blending of hand actions and manual action verbs, relative to nonmanual action verbs and nonaction verbs, modulates electrophysiological markers of semantic integration (N400) and motor-related cortical potentials during a lexical decision task. Relative to both other categories, manual action verbs involved reduced posterior N400 amplitude and greater modulations of frontal motor-related cortical potentials. Such effects overlapped in a window of ∼380-440 msec after word presentation and ∼180 msec before response execution, revealing the possible time span in which both semantic and action-related stages reach maximal convergence. In construing meaning, the brain recruits multimodal (conceptual) systems and embodied (modality-specific) mechanisms. Yet, no consensus exists on how crucial the latter are for the inception of semantic distinctions. To address this issue, we combined electroencephalographic (EEG) and intracranial EEG (iEEG) to examine when nouns denoting facial body parts (FBPs) and nonFBPs are discriminated in face-processing and multimodal networks. First, FBP words increased N170 amplitude (a hallmark of early facial processing). Second, they triggered fast (~100 ms) activity boosts within the face-processing network, alongside later (~275 ms) effects in multimodal circuits. Third, iEEG recordings from face-processing hubs allowed decoding ~80% of items before 200 ms, while classification based on multimodal-network activity only surpassed ~70% after 250 ms. Finally, EEG and iEEG connectivity between both networks proved greater in early (0-200 ms) than later (200-400 ms) windows. Comparisons between backward and forward translation (BT, FT) have long illuminated the organization of bilingual memory, with neuroscientific evidence indicating that FT would involve greater linguistic and attentional demands. However, no study has directly assessed the functional interaction between relevant mechanisms. Against this background, we conducted the first fMRI investigation of functional connectivity (FC) differences between BT and FT. In addition to yielding lower behavioral outcomes, FT was characterized by increased FC between a core semantic hub (the left anterior temporal lobe, ATL) and key nodes of attentional and vigilance networks (left inferior frontal, left orbitofrontal, and bilateral parietal clusters). Instead, distinct FC patterns for BT emerged only between the left ATL and the right thalamus, a region implicated in automatic relaying of sensory information to cortical regions. Research on how the brain construes meaning during language use has prompted two conflicting accounts. According to the 'grounded view', word understanding involves quick reactivations of sensorimotor (embodied) experiences evoked by the stimuli, with simultaneous or later engagement of multimodal (conceptual) systems integrating information from various sensory streams. Contrariwise, for the 'symbolic view', this capacity depends crucially on multimodal operations, with embodied systems playing epiphenomenal roles after comprehension. To test these contradictory hypotheses, the present magnetoencephalography study assessed implicit semantic access to grammatically constrained action and non-action verbs (n = 100 per category) while measuring spatiotemporally precise signals from the primary motor cortex (M1, a core region subserving bodily movements) and the anterior temporal lobe (ATL, a putative multimodal semantic hub). Convergent evidence from sensor- and source-level analyses revealed that increased modulations for action verbs occurred earlier in M1 (∼130-190 ms) than in specific ATL hubs (∼250-410 ms). Moreover, machine-learning decoding showed that trial-by-trial classification peaks emerged faster in M1 (∼100-175 ms) than in the ATL (∼345-500 ms), with over 71% accuracy in both cases. Considering their latencies, these results challenge the 'symbolic view' and its implication that sensorimotor mechanisms play only secondary roles in semantic processing. Instead, our findings support the 'grounded view', showing that early semantic effects are critically driven by embodied reactivations and that these cannot be reduced to post-comprehension epiphenomena, even when words are individually classified. Functional shifts (FSs) - morphosyntactically marked words evoking coherent but novel meanings - are ubiquitous in English and, specially, in Shakespearean literature. While their neural signatures have been explored in native speakers, no study has targeted foreign-language users, let alone comparing early and late bilinguals. Here, we administered a validated FS paradigm to subjects from both populations and evaluated time-frequency modulations evoked by FS and control sentences. Early bilinguals exhibited greater sensitivity towards FSs, indexed by reduced fronto-posterior theta-band oscillations across semantic- and structural-integration windows. Frontostriatal networks play critical roles in grounding action semantics and syntactic skills. Indeed, their atrophy distinctively disrupts both domains, as observed in patients with Huntington's disease (HD) and Parkinson's disease, even during early disease stages. However, frontostriatal degeneration in these conditions may begin up to 15 years before the onset of clinical symptoms, opening avenues for pre-clinical detection via sensitive tasks. Such a mission is particularly critical in HD, given that patients' children have 50% chances of inheriting the disease. Against this background, we assessed whether deficits in the above-mentioned domains emerge in subjects at risk to develop HD. We administered tasks tapping action semantics, object semantics, and two forms of syntactic processing to 18 patients with HD, 19 asymptomatic first-degree relatives, and sociodemographically matched controls for each group. The patients evinced significant deficits in all tasks, but only those in the two target domains were independent of overall cognitive state. More crucially, relative to controls, the asymptomatic relatives were selectively impaired in action semantics and in the more complex syntactic task, with both patterns emerging irrespective of the subjects' overall cognitive state. Our findings highlight the relevance of these dysfunctions as potential prodromal biomarkers of HD. Parkinson's disease (PD) patients show marked impairments in processing action verbs, and to a lesser extent, concrete (specially, manipulable) nouns. However, it is still unclear to what extent deficits in each of these categories are influenced by more general cognitive dysfunctions, and whether they are modulated by the words' implied motility. To examine these issues, we evaluated 49 non-demented PD patients and 49 healthy volunteers in an oral production task. The patients were divided into two groups depending on the presence or absence of mild cognitive impairment (PD-MCI and PD-nMCI, respectively). Participants named pictures of actions varying in motion content (low and high) and of objects varying in manipulability (low and high). The PD-MCI group showed deficits across all four categories. However, PD-nMCI patients exhibited a selective difficulty for high-motion action verbs. This finding corroborates and refines previous results suggesting that disturbances of action-related lexico-semantic information in PD constitute a sui generis alteration manifested early in the course of the disease's physiopathology. Processing action verbs, in general, and manual action verbs, in particular, involves activations in gross and hand-specific motor networks, respectively. While this is well established for receptive language processes, no study has explored action-semantic integration during written production. Moreover, little is known about how such crosstalk unfolds from motor planning to execution. Here we address both issues through our novel "action semantics in typing" paradigm, which allows to time keystroke operations during word typing. Specifically, we created a primed-verb-copying task involving manual action verbs, non-manual action verbs, and non-action verbs. Motor planning processes were indexed by first-letter lag (the lapse between target onset and first keystroke), whereas execution dynamics were assessed considering whole-word lag (the lapse between first and last keystroke). Each phase was differently delayed by action verbs. When these were processed for over one second, interference was strong and magnified by effector compatibility during programming, but weak and effector-blind during execution. Instead, when they were processed for less than 900ms, interference was reduced by effector compatibility during programming and it faded during execution. Finally, typing was facilitated by prime-target congruency, irrespective of the verbs' motor content. Thus, action-verb semantics seems to extend beyond its embodied foundations, involving conceptual dynamics not tapped by classical reaction-time measures. Background: Integration of compatible or incompatible emotional valence and semantic information is an essential aspect of complex social interactions. A modified version of the Implicit Association Test (IAT) called Dual Valence Association Task (DVAT) was designed in order to measure conflict resolution processing from compatibility/incompatibly of semantic and facial valence. The DVAT involves two emotional valence evaluative tasks which elicits two forms of emotional compatible/incompatible associations (facial and semantic)., Methods: Behavioural measures and Event Related Potentials were recorded while participants performed the DVAT., Results: Behavioural data showed a robust effect that distinguished compatible/incompatible tasks. The effects of valence and contextual association (between facial and semantic stimuli) showed early discrimination in N170 of faces. The LPP component was modulated by the compatibility of the DVAT., Conclusions: Results suggest that DVAT is a robust paradigm for studying the emotional interference effect in the processing of simultaneous information from semantic and facial stimuli.

Gestures should play a role in second language comprehension, given their importance in conveying contextual information. In this study, the N400 and the LPC were evaluated in a task involving the observation of videos showing utterances accompanied by gestures. Students studying advanced (G-High participants) and basic German (G-Low participants) as a second language were investigated. The utterance-gesture congruence and metaphoric meaning of content were manipulated during the task. As in previous ERP reports with native speakers, metaphorical expressions were sensitive to gestures. In G-Low participants, no modulation in the 300-500 ms window was observed, and only a modest effect was observed for the 500-700 ms window. More subtle differences of verbal expression were not processed in this group. Consistent with previous reports of the same paradigm with native speakers, the N400 from G-High group discriminated both congruent and incongruent gestures as well as literal and metaphorical sentences. Our results suggest that semantic processing is robust in the learning of a second language, although the amplitude modulation and latency of ERPs might depend on the speaker's proficiency level., (2010 Elsevier Ireland Ltd. Endophenotypes is one emerging strategy in schizophrenia research that is being used to identify the functional importance of genetically transmitted, brain-based deficits present in this disease. Currently, event-related potentials (ERPs) are timely used in this search. Several ERPs, including N400, present deficits in relation to schizophrenia. In order to assess the genetic liability of N400 as a possible endophenotype, a picture semantic matching task (congruent and incongruent pairs of pictures) was performed by 21 unaffected first-degree relatives of patients with schizophrenia, 21 DSM-IV diagnosed schizophrenia probands, and 21 control subjects, matched by age, gender and educational level. Probands and relatives were selected form Multiplex schizophrenia families. Significantly reduced N400 amplitude for congruent categories in N400 was found in probands and relatives in relation to controls. The latency onset and the maximum peak latency of N400 were delayed in both, relatives and probands groups compared to control. The voltage maps of incongruous-minus-congruous difference indicate a more reduced right restricted negativity in probands and relatives, when compared to a widely extended bilateral negativity in controls. No general differences were found between patients and relatives. These results demonstrate an electrophysiological deficit in semantic match processing in clinically unaffected first-degree relatives of patients with schizophrenia, suggesting a possible use of this marker as endophenotype.

We explore the hypothesis that induction of holistic or analytic strategies influences comprehension and processing of highly contextualized expressions of ordinary language, such as irony. Twenty undergraduate students were asked to categorize as coherent or incoherent a group of sentences. Each sentence completed a previous story, so that they could be ironical, literal or nonsensical endings. Participants were asked to evaluate whether each sentence was coherent or incoherent. Half of them were initially instructed to consider whether the sentences made sense (holistic condition); the other half were instructed to consider whether the sentences were congruent or incongruent (analytic condition). Behavioral responses and Event Related Potentials were registered during the experiment. Both behavioral and electrophysiological results allow clearly distinguishing between the holistic and the analytic strategies. The fact that the same set of stimuli elicits different ERP waveforms, depending on the strategy with which they are analyzed, suggests that different cognitive processes and different areas of the brain are operating in each case.

This study explores whether the brain can discriminate degrees of semantic congruency during wakefulness and sleep. Experiment 1 was conducted during wakefulness to test degrees of congruency by means of N400 amplitude. In Experiment 2, the same paradigm was applied to a different group of participants during natural night sleep. Stimuli were 108 sentences (definitions with two attributes) with four possible degrees of congruence as ending targets. In both studies, the amplitude of N400-like effect showed modulation according to the degree of congruency. The results indicate that the brain can accomplish sentential semantic discriminations not only in wakefulness but also in sleep.

Recent embodied cognition research shows that access to action verbs in shallow-processing tasks becomes selectively compromised upon atrophy of the cerebellum, a critical motor region. Here we assessed whether cerebellar damage also disturbs explicit semantic processing of action pictures and its integration with ongoing motor responses. We evaluated a cognitively preserved 33-year-old man with severe dysplastic cerebellar gangliocytoma (Lhermitte-Duclos disease), encompassing most of the right cerebellum and the posterior part of the left cerebellum. The patient and eight healthy controls completed two semantic association tasks (involving pictures of objects and actions, respectively) that required motor responses. Accuracy results via Crawford’s modified t-tests revealed that the patient was selectively impaired in action association. Moreover, reaction-time analysis through Crawford’s Revised Standardized Difference Test showed that, while processing of action concepts involved slower manual responses in controls, no such effect was observed in the patient, suggesting that motor-semantic integration dynamics may be compromised following cerebellar damage. Notably, a Bayesian Test for a Deficit allowing for Covariates revealed that these patterns remained after covarying for executive performance, indicating that they were not secondary to extra-linguistic impairments. Taken together, our results extend incipient findings on the embodied functions of the cerebellum, offering unprecedented evidence of its crucial role in processing non-verbal action meanings and integrating them with concomitant movements. Action verbs are critically embodied in motor brain networks. In Parkinson's disease (PD), damage to the latter compromises access to such words. However, patients are not fully incapable of processing them, as their performance is far from floor level. Here we tested the hypothesis that action-verb processing in PD may rely on alternative disembodied semantic circuits. Seventeen PD patients and 15 healthy controls listened to action verbs and nouns during functional MRI scanning. Using cluster-mass analysis with a permutation test, we assessed task-related functional connectivity considering seeds differentially engaged by action and non-action words (namely, putamen and M1 versus posterior superior temporal lobe, respectively). The putamen seed showed reduced connectivity within the basal ganglia in patients for both lexical categories. However, only action verbs recruited different cortical networks in each group. Specifically, the M1 seed exhibited more anterior connectivity for controls and more posterior connectivity for patients, with no differences in the temporal seed. Moreover, the patients' level of basal ganglia atrophy positively correlated with their reliance on M1-posterior connectivity during action-verb processing. PD patients seem to have processed action verbs via non-motor cortical networks subserving amodal semantics. Such circuits may afford alternative pathways to process words when default embodied mechanisms are disturbed. Moreover, the greater the level of basal ganglia atrophy, the greater the patients' reliance on this alternative route. Recent studies suggest that action-verb processing is particularly affected in early stage Parkinson's disease (PD), highlighting the potential role of subcortical areas in language processing and in the semantic integration of actions. However, this disorder-related language impairment is frequently unrecognized by clinicians and often remains untreated. Early detection of action-language processing deficits could be critical for diagnosing and developing treatment strategies for PD. In this article, we review how action-verb processing is affected in PD and propose a model in which multiple and parallel frontotemporal circuits between the cortex and the basal ganglia provide the anatomic substrate for supporting action-language processing. We hypothesize that contextual coupling of action-language networks are partially dependent on cortical-subcortical integration, and not only on somatotopic motor cortical organization or in a mirror neuron system. This hypothesis is supported by both experimental and clinical evidence. Then, we identify further research steps that would help to determine the reliability of action-language impairments as an early marker of PD. Finally, theoretical implications for clinical assessment and for models of action-language interaction (action-perception cycle theories, mirror system models of language, and embodied cognition approaches to language) are discussed. Background: Research suggests that individuals with different attachment patterns process social information differently, especially in terms of facial emotion recognition. However, few studies have explored social information processes in adolescents. This study examined the behavioral and ERP correlates of emotional processing in adolescents with different attachment orientations (insecure attachment group and secure attachment group; IAG and SAG, respectively). This study also explored the association of these correlates to individual neuropsychological profiles. Methodology/Principal Findings We used a modified version of the dual valence task (DVT), in which participants classify stimuli (faces and words) according to emotional valence (positive or negative). Results showed that the IAG performed significantly worse than SAG on tests of executive function (EF attention, processing speed, visuospatial abilities and cognitive flexibility). In the behavioral DVT, the IAG presented lower performance and accuracy. The IAG also exhibited slower RTs for stimuli with negative valence. Compared to the SAG, the IAG showed a negative bias for faces; a larger P1 and attenuated N170 component over the right hemisphere was observed. A negative bias was also observed in the IAG for word stimuli, which was demonstrated by comparing the N170 amplitude of the IAG with the valence of the SAG. Finally, the amplitude of the N170 elicited by the facial stimuli correlated with EF in both groups (and negative valence with EF in the IAG). Conclusions/Significance: Our results suggest that individuals with different attachment patterns process key emotional information and corresponding EF differently. This is evidenced by an early modulation of ERP components’ amplitudes, which are correlated with behavioral and neuropsychological effects. In brief, attachments patterns appear to impact multiple domains, such as emotional processing and EFs.

Several studies have illuminated how processing manual action verbs (MaVs) affects the programming or execution of concurrent hand movements. Here, to circumvent key confounds in extant designs, we conducted the first assessment of motor–language integration during handwriting—a task in which linguistic and motoric processes are co‐substantiated. Participants copied MaVs, non‐manual action verbs, and non‐action verbs as we collected measures of motor programming and motor execution. Programming latencies were similar across conditions, but execution was faster for MaVs than for the other categories, regardless of whether word meanings were accessed implicitly or explicitly. In line with the Hand‐Action‐Network Dynamic Language Embodiment (HANDLE) model, such findings suggest that effector‐congruent verbs can prime manual movements even during highly automatized tasks in which motoric and verbal processes are naturally intertwined. To assess the impact of Parkinson's disease (PD) on spontaneous discourse, we conducted computerized analyses of brief monologues produced by 51 patients and 50 controls. We explored differences in semantic fields (via latent semantic analysis), grammatical choices (using part-of-speech tagging), and word-level repetitions (with graph embedding tools). Although overall output was quantitatively similar between groups, patients relied less heavily on action-related concepts and used more subordinate structures. Also, a classification tool operating on grammatical patterns identified monologues as pertaining to patients or controls with 75% accuracy. Finally, while the incidence of dysfluent word repetitions was similar between groups, it allowed inferring the patients' level of motor impairment with 77% accuracy. Our results highlight the relevance of studying naturalistic discourse features to tap the integrity of neural (and, particularly, motor) networks, beyond the possibilities of standard token-level instruments.
