The analysis of critical states during fracture of wood materials is crucial for wood building safety monitoring, wood processing, etc. In this paper, beech and camphor pine are selected as the research objects, and the acoustic emission signals during the fracture process of the specimens are analyzed by three-point bending load experiments. On the one hand, the critical state interval of a complex acoustic emission signal system is determined by selecting characteristic parameters in the natural time domain. On the other hand, an improved method of b_value analysis in the natural time domain is proposed based on the characteristics of the acoustic emission signal. The K-value, which represents the beginning of the critical state of a complex acoustic emission signal system, is further defined by the improved method of b_value in the natural time domain. For beech, the analysis of critical state time based on characteristic parameters can predict the "collapse" time 8.01 s in advance, while for camphor pines, 3.74 s in advance. K-value can be analyzed at least 3 s in advance of the system "crash" time for beech and 4 s in advance of the system "crash" time for camphor pine. The results show that compared with traditional time-domain acoustic emission signal analysis, natural time-domain acoustic emission signal analysis can discover more available feature information to characterize the state of the signal. Both the characteristic parameters and Natural_Time_b_value analysis in the natural time domain can effectively characterize the time when the complex acoustic emission signal system enters the critical state. Critical state analysis can provide new ideas for wood health monitoring and complex signal processing, etc.

Abstract High quality is the main goal of today’s maize breeding and the investigation of grain quality traits would help to breed high-quality varieties in maize. In this study, genome-wide association studies in a set of 248 diverse inbred lines were performed with 83,057 single nucleotide polymorphisms (SNPs), and five grain quality traits were investigated in diverse environments for two years. The results showed that maize inbred lines showed substantial natural variations of grain quality and these traits showed high broad-sense heritability. A total of 49 SNPs were found to be significantly associated with grain quality traits. Among these SNPs, four co-localized sites were commonly detected by multiple traits. The candidate genes which were searched for can be classified into 11 biological processes, 13 cellular components, and 6 molecular functions. Finally, we found 29 grain quality-related genes. These genes and the SNPs identified in the study would offer essential information for high-quality varieties breeding programs in maize.

The ocean chlorophyll-a (Chl-a) concentration is an important variable in the marine environment, the abnormal distribution of which is closely related to the hazards of red tides. Thus, the accurate prediction of its concentration in the East China Sea (ECS) is greatly important for preventing water eutrophication and protecting the coastal ecological environment. Processed by two different pre-processing methods, 10-year (2011–2020) satellite-observed chlorophyll-a data and logarithmic data were used as the long short-term memory (LSTM) neural network training datasets in this study. The 2021 data were used for comparison to prediction results. The past 15 days’ data were used to predict the concentration of chlorophyll-a for the five following days. Results showed that the predictions obtained by both pre-processing methods could simulate the seasonal distribution of the Chl-a concentration in the ECS effectively. Moreover, the prediction performance of the model driven by the original values was better in the medium- and low-concentration regions. However, in the high-concentration region, the prediction of extreme concentrations by the two data-driven LSTM models showed underestimation, considering that the prediction performance of the model driven by the original values was better. Results of sensitivity experiments showed that the prediction accuracy of the model decreased considerably when the backward prediction time step increased. In this study, the neural network was driven only by chlorophyll-a, whose concentration in the ECS was forecasted, and the effect of other relevant marine elements on Chl-a was not considered, which is the current weakness of this study.

Sea surface temperature (SST) is an important physical factor in the interaction between the ocean and the atmosphere. Accurate monitoring and prediction of the temporal and spatial distribution of SST are of great significance in dealing with climate change, disaster prevention, disaster reduction, and marine ecological protection. This study establishes a prediction model of sea surface temperature for the next five days in the East China Sea using long-term and short-term memory neural networks (LSTM). It investigates the influence of different parameters on prediction accuracy. The sensitivity experiment results show that, based on the same training data, the length of the input data of the LSTM model can improve the model’s prediction performance to a certain extent. However, no obvious positive correlation is observed between the increase in the input data length and the improvement of the model’s prediction accuracy. On the contrary, the LSTM model’s performance decreases with the prediction length increase. Furthermore, the single-point prediction results of the LSTM model for the estuary of the Yangtze River, Kuroshio, and the Pacific Ocean are accurate. In particular, the prediction results of the point in the Pacific Ocean are the most accurate at the selected four points, with an RMSE of 0.0698 °C and an R2 of 99.95%. At the same time, the model in the Pacific region is migrated to the East China Sea. The model was found to have good mobility and can well represent the long-term and seasonal trends of SST in the East China Sea.

General combining ability (GCA) is an important index for inbred lines breeding of maize. To identify the genetic loci of GCA and associated agronomic traits, an association analysis with 195 SSRs was made in phenotypic traits of 240 F1 derived from 120 elite inbred lines containing current breeding resources of maize crossed with 2 testers (Zheng58 and Chang7-2) in two places in 2018. All of the 20 association loci detected for grain yield (GY), plant height (PH), ear height (EH) and GCA for the three traits in two places could explain a phenotypic variation range of 7.31%-9.29%. Among the 20 association loci, 9 (7.31%-9.04%) were associated with GY, 4 (7.22%-8.91%) were related to GCA of GY, 1 (7.56%) was associated with PH, and 3 (7.53%-8.96%) were related to EH. In addition, 3 loci (9.14%-9.29%) were associated with GCA of PH whereas no locus was identified for GCA of EH. In the comparison of the association loci detected in Baoding and Handan, interestingly, one locus (7.69% and 8.11%) was identified in both environments and one locus (7.52% and 7.82%) was identified for yield and GCA of yield. Therefore, the identification of GY-, PH-, EH- and GCA-related association loci could not only provide references for high yield breeding of maize, but also help us comprehend the relationships among GY, agricultural traits and GCA.

To identify electrical vehicle (EV) distribution paths with high robustness, insensitivity to uncertainty factors, and detailed road-by-road schemes, optimization of the distribution path problem of EV with multiple distribution centers and considering the charging facilities is necessary. With the minimum transport time as the goal, a robust optimization model of EV distribution path with adjustable robustness is established based on Bertsimas' theory of robust discrete optimization. An enhanced three-segment genetic algorithm is also developed to solve the model, such that the optimal distribution scheme initially contains all road-by-road path data using the three-segment mixed coding and decoding method. During genetic manipulation, different interlacing and mutation operations are carried out on different chromosomes, while, during population evolution, the infeasible solution is naturally avoided. A part of the road network of Xifeng District in Qingyang City is taken as an example to test the model and the algorithm in this study, and the concrete transportation paths are utilized in the final distribution scheme. Therefore, more robust EV distribution paths with multiple distribution centers can be obtained using the robust optimization model.

Abstract The retina plays a crucial role in processing and decoding visual information, both in normal development and during myopia progression. Recent advancements have introduced a library-independent approach for data-independent acquisition (DIA) analyses. This study demonstrates deep proteome identification and quantification in individual mice retinas during myopia development, with an average of 6,263 ± 86 unique protein groups. We anticipate that the use of a predicted retinal-specific spectral library combined with the robust quantification achieved within this dataset will contribute to a better understanding of the proteome complexity. Furthermore, a comprehensive mice retinal-specific spectral library was generated, encompassing a total identification of 9,401 protein groups, 70,041 peptides, 95,339 precursors, and 761,868 transitions acquired using SWATH-MS acquisition on a ZenoTOF 7600 mass spectrometer. This dataset surpasses the spectral library generated through high-pH reversed-phase fractionation by data-dependent acquisition (DDA). The data is available via ProteomeXchange with the identifier PXD046983. It will also serve as an indispensable reference for investigations in myopia research and other retinal or neurological diseases.

Abstract Photosynthesis, essential for life on earth, sustains diverse processes by providing nutrition in plants and microorganisms. Especially, photosynthesis is increasingly applied in disease treatments, but its efficacy is substantially limited by the well‐known low penetration depth of external light. Here, ultrasound‐mediated photosynthesis is reported for enhanced sonodynamic tumor therapy using organic sonoafterglow (ultrasound‐induced afterglow) nanoparticles combined with cyanobacteria, demonstrating the proof‐of‐concept sonosynthesis (sonoafterglow‐induced photosynthesis) in cancer therapy. Chlorin e6, a typical small‐molecule chlorine, is formulated into nanoparticles to stimulate cyanobacteria for sonosynthesis, which serves three roles, i.e., overcoming the tissue‐penetration limitations of external light sources, reducing hypoxia, and acting as a sonosensitizer for in vivo tumor suppression. Furthermore, sonosynthetic oxygenation suppresses the expression of hypoxia‐inducible factor 1α, leading to reduced stability of downstream SLC7A11 mRNA, which results in glutathione depletion and inactivation of glutathione peroxidase 4, thereby inducing ferroptosis of cancer cells. This study not only broadens the scope of microbial nanomedicine but also offers a distinct direction for sonosynthesis.

Abstract Myopia is an abnormal vision condition characterized by difficulties in seeing distant objects. Myopia has become a public health issue not only in Asian countries but also in Western countries. Previously, we found that violet light (VL, 360–400 nm wavelength) exposure effectively suppressed myopia progression in experimental chick and mice models of myopia. The inhibitory effects of VL on myopia progression are reduced in retina-specific opsin 5 (Opn5) knockout (KO) mice. Furthermore, VL exposure upregulated early growth response-1 (Egr-1) expression in the chorioretinal tissues of chicks. However, the expression of EGR-1 and role of OPN5 in mice following VL exposure remain unclear. In this study, we examined whether VL exposure-induced EGR-1 upregulation depends on Opn5 expression in the mouse retina. EGR-1 mRNA and protein expressions increased in the mouse retina and mouse retinal 661W cells following VL exposure. These increases were consistently reduced in retina specific Opn5 conditional KO mice and Opn5 KO 661W cells. Our results suggest that OPN5 mediates VL-induced EGR-1 upregulation in mice. These molecular targets could be considered for the prevention and treatment of myopia.

Organic fertilization and deep ploughing are expected to contribute to carbon (C) sequestration in arable soils. This hypothesis was evaluated over 11 years of chemical and organic fertilization under conventional tillage (20 cm) and deep ploughing (40 cm) in a rainfed sorghum-maize rotation system on the Loess Plateau. Soil was collected and analysed to determine the soil organic C (SOC) and total nitrogen (TN) contents in the soil profiles (0–60 cm) or in the macroaggregates, microaggregates and silt and clay fractions (0–20 cm only). The SOC and TN stocks at the 0–60 cm depth displayed a net decrease in the unfertilized control, a marginal increase with chemical fertilizer, a significant increase of 23–45 % and 19–42 %, respectively, with organic amendments and a significant increase of 54 % and 48 %, respectively, when combined with deep ploughing. A critical input amount of 2064 kg C ha−1 yr−1 or 162 kg N ha−1 yr−1 was found to be required to maintain the initial SOC or TN stock. Heatwaves have become increasingly frequent and intense, posing a significant threat to the survival and health of marine bivalves. The temperature fluctuations associated with heatwaves can cause significant alterations in the composition and quantity of microbial communities in bivalves, resulting in changes to their immunological responses, gut microbiome, oxidative stress levels, and other physiological processes and eventually making them more susceptible to diseases and mass mortalities. This is particularly concerning because some of these bivalves are consumed raw, which could represent a risk to human health. This paper provides an overview of the current state of knowledge regarding the impact of marine heatwaves on bivalves and their microbial communities, demonstrating the intricate relationship between heatwaves, microbial ecosystems, and bivalve health. Our analysis highlights the need for additional research to establish the underlying mechanisms of these reactions and to develop appropriate conservation and management strategies to limit the impact of heatwaves on bivalves and their microbial ecosystems.

Abstract As the most effective way to manage the surge in municipal solid waste (MSW) and global energy issues, waste‐to‐energy (WTE) incineration plants have been actively promoted by the Chinese government, and the public‐private partnership (PPP) procurement model is widely used to improve management efficiency. However, risk events have occurred frequently in this industry due to a lack of practice experience and to poor risk management. To investigate the potential risk factors in PPP WTE incineration projects in China, this paper aims to identify critical risk factors by assessing their severity and occurrence probability. Opinions of industry experts were collected via an empirical questionnaire survey. The respondents were invited to rate a total of 18 risk factors that impact the success of PPP WTE projects as gleaned from previous research. The overall results ranked risk factors based on risk impact and showed that public opposition, government decision making, defective legal and regulatory systems, environmental pollution, lack of supporting infrastructure, and government credit are the top six risk factors affecting the sustainable development of PPP WTE incineration projects. In addition, the causes and consequences of each critical risk factor are comprehensively analyzed. Lastly, practical and managerial implications are analyzed in terms of their management of these critical risks. The originality of this research lies in thoroughly identifying potential implicit risk factors according to the current social background using an empirical questionnaire survey based on case study findings. The results of this study are expected to improve risk management and facilitate the sustainable development of the PPP WTE incineration industry in China.

Overexpression and persistent activation of STAT5 play an important role in the development and progression of acute lymphoblastic leukemia (ALL), the most common pediatric cancer. Small interfering RNA (siRNA)-mediated downregulation of STAT5 represents a promising therapeutic approach for ALL to overcome the limitations of current treatment modalities such as high relapse rates and poor prognosis. However, to effectively transport siRNA molecules to target cells, development of potent carriers is of utmost importance to surpass hurdles of delivery. In this study, we investigated the use of lipopolymers as non-viral delivery systems derived from low molecular weight polyethylenimines (PEI) substituted with lauric acid (Lau), linoleic acid (LA) and stearic acid (StA) to deliver siRNA molecules to ALL cell lines and primary samples. Among the lipid-substituted polymers explored, Lau- and LA-substituted PEI displayed excellent siRNA delivery to SUP-B15 and RS4;11 cells. STAT5A gene expression was downregulated (36-92%) in SUP-B15 and (32%) in RS4;11 cells using the polymeric delivery systems, which consequently reduced cell growth and inhibited the formation of colonies in ALL cells. With regard to ALL primary cells, siRNA-mediated STAT5A gene silencing was observed in four of eight patient cells using our leading polymeric delivery system, 1.2PEI-Lau8, accompanied by the significant reduction in colony formation in three of eight patients. In both BCR-ABL positive and negative groups, three of five patients demonstrated marked cell growth inhibition in both MTT and trypan blue exclusion assays using 1.2PEI-Lau8/siRNA complexes in comparison with their control siRNA groups. Three patient samples did not show any positive results with our delivery systems. Differential therapeutic responses to siRNA therapy observed in different patients could result from variable genetic profiles and patient-to-patient variability in delivery. This study supports the potential of siRNA therapy and the designed lipopolymers as a delivery system in ALL therapy.

Use of small interfering RNA (siRNA) is a promising approach for AML treatment as the siRNA molecule can be designed to specifically target proteins that contribute to aberrant cell proliferation in this disease. However, a clinical-relevant means of delivering siRNA molecules must be developed, as the cellular delivery of siRNA is problematic. Here, we report amphiphilic carriers combining a cationic polymer (2 kDa polyethyleneimine, PEI2) with lipophilic moieties to facilitate intracellular delivery of siRNA to AML cell lines. Complete binding of siRNA by the designed carriers was achieved at a polymer:siRNA ratio of ≈ 0.5 and led to siRNA/polymer complexes of ≈ 100 nm size. While the native PEI2 did not display cytotoxicity on AML cell lines THP-1, KG-1 and HL-60, lipid-modification on PEI2 slightly increased the cytotoxicity, which was consistent with increased interaction of polymers with cell membranes. Cellular delivery of siRNA was dependent on the nature of lipid substituent and the extent of lipid substitution, and varied among the three AML cell lines used. Linoleic acid-substituted polymers performed best among the prepared polymers and gave a siRNA delivery equivalent to better performing commercial reagents. Using THP-1 cells and a reporter (GFP) and an endogenous (CXCR4) target, effective silencing of the chosen targets was achieved with 25 to 50 nM of siRNA concentrations, and without adversely affecting subsequent cell growth. We conclude that lipid-substituted PEI2 can serve as an effective delivery of siRNA to leukemic cells and could be employed in molecular therapy of leukemia.

Despite the initial response to the reversible, ATP-competitive quinazoline inhibitors that target ErbB-family, such a subset of cancer patients almost invariably develop resistance. Recent studies have provided compelling evidence that irreversible ErbB inhibitors have the potential to override this resistance. Here, we found that AST1306, a novel anilino-quinazoline compound, inhibited the enzymatic activities of wild-type epidermal growth factor receptor (EGFR) and ErbB2 as well as EGFR resistant mutant in both cell-free and cell-based systems. Importantly, AST1306 functions as an irreversible inhibitor, most likely through covalent interaction with Cys797 and Cys805 in the catalytic domains of EGFR and ErbB2, respectively. Further studies showed that AST1306 inactivated pathways downstream of these receptors and thereby inhibited the proliferation of a panel of cancer cell lines. Although the activities of EGFR and ErbB2 were similarly sensitive to AST1306, ErbB2-overexpressing cell lines consistently exhibited more sensitivity to AST1306 antiproliferative effects. Consistent with this, knockdown of ErbB2, but not EGFR, decreased the sensitivity of SK-OV-3 cells to AST1306. In vivo, AST1306 potently suppressed tumor growth in ErbB2-overexpressing adenocarcinoma xenograft and FVB-2/N(neu) transgenic breast cancer mouse models, but weakly inhibited the growth of EGFR-overexpressing tumor xenografts. Tumor growth inhibition induced by a single dose of AST1306 in the SK-OV-3 xenograft model was accompanied by a rapid (within 2 h) and sustained (≥24 h) inhibition of both EGFR and ErbB2, consistent with an irreversible inhibition mechanism. Taken together, these results establish AST1306 as a selective, irreversible ErbB2 and EGFR inhibitor whose growth-inhibitory effects are more potent in ErbB2-overexpressing cells.
