Abstract During bacterial cytokinesis, polymers of the bacterial tubulin FtsZ coalesce into the Z ring to orchestrate divisome assembly and septal cell wall synthesis. Previous studies have found that Z ring condensation and stability is critical for successful cell division. However, how FtsZ filaments condense into a Z ring remains enigmatic and whether septal cell wall synthesis can feedback to the Z ring has not been investigated. Here, we show that FtsZ-associated proteins (Zaps) play important roles in Z ring condensation and stability, and discover septal cell wall synthesis as a novel player for Z ring condensation and stabilization in Escherichia coli and Caulobacter crescentus. Moreover, we find that the interaction between the Z ring membrane anchor, FtsA, and components of the septal cell wall synthetic complex are critical for septal cell wall synthesis-mediated Z ring condensation. Altogether, these findings suggest that the divisome is a self-enhancing machine in these two gram-negative bacteria, where the Z ring and the septal cell wall synthetic complex communicate with and reinforce each other to ensure robustness of cell division.

Summary: Intestinal dysbiosis is prominent in systemic sclerosis (SSc), but it remains unknown how it contributes to microvascular injury and fibrosis that are hallmarks of this disease. Trimethylamine (TMA) is generated by the gut microbiome and in the host converted by flavin-containing monooxygenase (FMO3) into trimethylamine N-oxide (TMAO), which has been implicated in chronic cardiovascular and metabolic diseases. Using cell culture systems and patient biopsies, we now show that TMAO reprograms skin fibroblasts, vascular endothelial cells, and adipocytic progenitor cells into myofibroblasts via the putative TMAO receptor protein R-like endoplasmic reticulum kinase (PERK). Remarkably, FMO3 was detected in skin fibroblasts and its expression stimulated by TGF-β1. Moreover, FMO3 was elevated in SSc skin biopsies and in SSc fibroblasts. A meta-organismal pathway thus might in SSc link gut microbiome to vascular remodeling and fibrosis via stromal cell reprogramming, implicating the FMO3-TMAO-PERK axis in pathogenesis, and as a promising target for therapy.

Abstract Antioxidant-rich diets serve as protective factors in preventing obesity. The composite dietary antioxidant index (CDAI) represents a novel, comprehensive metric for assessing the antioxidant capacity of diets. Our objective is to investigate the relationship between the CDAI and obesity prevalence among adults in the United States. Dietary and anthropometric information about adults aged 20 years and older were obtained from the National Health and Nutrition Examination Survey (NHANES) 2007–2018. The CDAI was derived from six dietary antioxidants. Obesity is defined as a body mass index (BMI, kg/m2) ≥ 30 kg/m2, and abdominal obesity as a waist circumference (WC, cm) ≥ 102 cm for men and ≥ 88 cm for women. The relationship between CDAI and obesity, including abdominal obesity, was analyzed using logistic regression and subgroup analyses. A total of 25,553 participants were analyzed. Abstract To achieve high power conversion efficiency in perovskite/silicon tandem solar cells, it is necessary to develop a promising wide-bandgap perovskite absorber and processing techniques in relevance. To date, the performance of devices based on wide-bandgap perovskite is still limited mainly by carrier recombination at their electron extraction interface. Here, we demonstrate assembling a binary two-dimensional perovskite by both alternating-cation-interlayer phase and Ruddlesden−Popper phase to passivate perovskite/C60 interface. The binary two-dimensional strategy takes effects not only at the interface but also in the bulk, which enables efficient charge transport in a wide-bandgap perovskite solar cell with a stabilized efficiency of 20.79% (1 cm2). Based on this absorber, a monolithic perovskite/silicon tandem solar cell is fabricated with a steady-state efficiency of 30.65% assessed by a third party. Moreover, the tandem devices retain 96% of their initial efficiency after 527 h of operation under full spectral continuous illumination, and 98% after 1000 h of damp-heat testing (85 °C with 85% relative humidity).

Abstract The adsorption behavior of gold thiosulfate ions on the surface of kaolinite was studied using a combination of experimental research and quantum chemical calculations. Under the condition of a stirring time of 30 min, a stirring speed of 500 r·min−1, and a mass ratio of 30% kaolinite in the slurry, when the initial gold concentration of 56.50 mg·L−1,the adsorption rate of gold-thiosulfate ions from a kaolinite-containing solution was 7.44%. The results of Fourier transform infrared spectroscopy (FTIR) and energy dispersive spectroscopy (EDS) showed that the physical and chemical adsorption of kaolinite and gold thiosulfate occurred in solution. Quantum chemical calculations were performed using the CASTEP module in Materials Studio. The adsorption energy of gold thiosulfate on the surface of kaolinite (001) was calculated as − 438.01 kJ·mol−1.The calculated H76–O289 distance was 1.615 Å. Mulliken Charge population analysis and bond population analysis showed that gold thiosulfate ions form relatively stable bonds on the kaolinite surface (001). In the process of thiosulfate immersion, part of gold is adsorbed by kaolinite, which affects the extraction of gold. These results indicate that during the leaching process of gold thiosulfate, kaolinite has the ability to "catch" gold, which affects the leaching efficiency.

Abstract To evaluate the effect of diabetic retinopathy (DR) status or severity on all-cause and cause-specific mortality among diabetic older adults in the United States using the most recent National Health and Nutrition Examination Survey (NHANES) follow-up mortality data. The severity of DR was graded according to the Early Treatment Diabetic Retinopathy Study (ETDRS) grading scale. Multiple covariate-adjusted Cox proportional hazards regression models, Fine and Gray competing risk regression models, and propensity score matching (PSM) methods were used to assess the risk of all-cause and cause-specific mortality in individuals with diabetes. All analyses adopted the weighted data and complex stratified design approach proposed by the NHANES guidelines. Time to death was calculated based on the time between baseline and date of death or December 31, 2019, whichever came first. Ultimately 1077 participants, representing 3,025,316 US non-hospitalized individuals with diabetes, were included in the final analysis. After a median follow-up of 12.24 years (IQR, 11.16–13.49), 379 participants were considered deceased from all-causes, with 43.90% suffering from DR, including mild DR (41.50%), moderate to severe DR (46.77%), and proliferative DR (PDR) (67.21%). DR was associated with increased all-cause, cardiovascular disease (CVD) and diabetes mellitus (DM)-specific mortality, which remained consistent after propensity score matching (PSM). Results of DR grading assessment suggested that the presence of mild, moderate to severe NPDR was significantly associated with increased risk of all-cause and CVD-specific mortality, while the presence and severity of any DR was associated with increased DM-specific mortality, with a positive trend. The presence of DR in elderly individuals with diabetes is significantly associated with the elevated all-cause and CVD mortality. The grading or severity of DR may reflect the severity of cardiovascular disease status and overall mortality risk in patients with diabetes.

Abstract Cancer models play critical roles in basic cancer research and precision medicine. However, current in vitro cancer models are limited by their inability to mimic the three-dimensional architecture and heterogeneous tumor microenvironments (TME) of in vivo tumors. Here, we develop an innovative patient-specific lung cancer assembloid (LCA) model by using droplet microfluidic technology based on a microinjection strategy. This method enables precise manipulation of clinical microsamples and rapid generation of LCAs with good intra-batch consistency in size and cell composition by evenly encapsulating patient tumor-derived TME cells and lung cancer organoids inside microgels. LCAs recapitulate the inter- and intratumoral heterogeneity, TME cellular diversity, and genomic and transcriptomic landscape of their parental tumors. LCA model could reconstruct the functional heterogeneity of cancer-associated fibroblasts and reflect the influence of TME on drug responses compared to cancer organoids. Notably, LCAs accurately replicate the clinical outcomes of patients, suggesting the potential of the LCA model to predict personalized treatments. Collectively, our studies provide a valuable method for precisely fabricating cancer assembloids and a promising LCA model for cancer research and personalized medicine.

Labile C inputs into soils will be partially transformed into soil organic carbon (SOC) through microbial assimilation or physicochemical protection as such mineral-organic interactions and soil aggregation. The C inputs may stimulate the decomposition of native SOC, inducing a phenomenon known as the priming effect. Increasing C inputs may increase SOC content, yet the relative role of these mechanisms in controlling the magnitudes of SOC increase among soils remains unclear. Four soils differing in microbiology and N content were incubated with 13C-labeled glucose at the amounts of 0.5 (G0.5), 1.0 (G1.0), and 2.0 (G2.0) g C kg−1 soil for 48 days. The objectives of this study were 1) to quantify the fates of added glucose-C, the priming effect, and the changes in SOC and mineral N contents after the incubation, and 2) to identify the main mechanisms for SOC increase with the increased amount of C input and the effects of initial soil N availability and microbial composition. 13C isotope probing showed that 19 to 26 % and 37 to 96 % of added glucose were utilized for microbial respiration and growth, respectively, after the incubation in all soils, with the glucose-C use efficiency ranging from 30 to 60 %. The remaining accounted for 4 to 37 % in all cases except for UltisolHN at G1.0 and G2.0 (–23 % and −2%), which was confirmed as the fraction protected in soil or re-utilized by microbes via acid extraction. The soil-protected glucose-C was higher, while glucose-C use efficiency was lower with the increased amount of added glucose only in the soils with low N. Glucose addition induced a positive priming effect, a SOC increase, and a mineral N loss irrespective of soil type and addition amount. Glucose addition increased the cumulative fungal biomass in all cases and the cumulative bacterial biomass only at G2.0 in all soils and at G1.0 in the high-N soils. It increased the final fungal necromass only at G0.5 in MollisolLN and UltisolHN and at G2.0 in MollisolHN and the final bacterial necromass in all cases except for the low-N soils and UltisolHN at G2.0. With the increased amount of added glucose, the SOC content increased linearly and the priming effect and fungal biomass increased non-linearly, with larger magnitudes in the soils with lower N. These results indicated that glucose was quickly protected after addition and remained in the soil until soil N became a limiting factor for microbial utilization. The availability of soil N relative to input C mediated microbial growth and its controls over SOC change via the soil protection and priming effects. The carbon sequestration potential with increased input of dissolved substrates as such glucose e. g. through root exudation was likely larger in soils with lower fertility due to reduced substrate-C use efficiency and increased effect of soil protection despite an increased priming effect. Further studies are needed on more substrate types as they affect substrate-C use efficiency and soil protection mechanisms.

Abstract Radiation triage and biological dosimetry are critical for the medical management of massive potentially exposed individuals following radiological accidents. Here, we performed a genome-wide screening of radiation-responding mRNAs, whose N6-methyladenosine (m6A) levels showed significant alteration after acute irradiation. The m6A levels of three genes, Ncoa4, Ate1 and Fgf22, in peripheral blood mononuclear cells (PBMCs) of mice showed excellent dose-response relationships and could serve as biomarkers of radiation exposure. Especially, the RNA m6A of Ncoa4 maintained a high level as long as 28 days after irradiation. We demonstrated its responsive specificity to radiation, conservation across the mice, monkeys and humans, and the dose-response relationship in PBMCs from cancer patients receiving radiation therapy. Finally, NOCA4 m6A-based biodosimetric models were constructed for estimating absorbed radiation doses in mice or humans. Collectively, this study demonstrated the potential feasibility of RNA m6A in radiation accidents management and clinical applications.

Abstract Inflammatory bowel disease (IBD) and periodontitis are reported to be closely associated; however, whether there is a causal association between them remains unclear. To explore the existence of this causality, this study applied a bidirectional two-sample Mendelian randomization (MR). The genetic variants were obtained from the summary statistics of genome-wide association studies of IBD, including its subtypes CD and UC, and periodontitis. 175, 148, 113, and six single-nucleotide polymorphisms were selected as instrumental variables for IBD, CD, UC, and periodontitis, respectively. In MR analysis, random-effects inverse-variance weighted was used as the primary method, and weighted median and MR Egger regression were applied as the complementary method. A series of sensitivity analyses were also conducted to ensure the reliability of the results. None of these analyses found a significant effect of genetically proxied IBD and its subtypes on periodontitis, and vice versa. Subsequent sensitivity analyses did not detect any horizontal pleiotropy and heterogeneity. Caution should be exerted when it comes to clinical relevance and further studies are needed to clarify the relationship between IBD and periodontitis.

Dioscorea cirrhosa is a natural plant with advantageous properties in both health care and dyeing. In this work, cotton fabric was dyed with Dioscorea cirrhosa dry extract by the traditional, repeated, dip-spray dyeing process and single-side sunlight fixation method. The color characteristics, surface morphology and structure, chemical composition, and mechanical properties of the dyed cotton fabric were characterized. The flame-retardant and waterproof functions of the dyed cotton fabric were also evaluated. The results showed that the dyed cotton fabric presented the limiting oxygen index (LOI) with one side displaying a reddish-brown coating and the other side being red. The limiting oxygen index of the cotton fabric increased from 18% to 28% after dyeing treatment.

Abstract Post-traumatic epilepsy (PTE) is an important cause of poor prognosis in patients with cerebral contusions. The primary purpose of this study is to evaluate the high-risk factors of PTE by summarizing and analyzing the baseline data, laboratory examination, and imaging features of patients with a cerebral contusion, and then developing a Nomogram prediction model and validating it. This study included 457 patients diagnosed with cerebral contusion who met the inclusion criteria from November 2016 to November 2019 at the Qinghai Provincial People's Hospital. All patients were assessed for seizure activity seven days after injury. Univariate analysis was used to determine the risk factors for PTE. Summary: Pancreatic stellate cells (PSCs) are suggested to play an important role in the development of pancreas and islet fibrosis. However, the precise contributions and solid in vivo evidence of PSCs to the fibrogenesis remain to be elucidated. Here, we developed a novel fate-tracing strategy for PSCs by vitamin A administration in Lrat-cre; Rosa26-tdTomato transgenic mouse. The results showed that stellate cells give rise to 65.7% of myofibroblasts in cerulein-induced pancreatic exocrine fibrosis. In addition, stellate cells in islets increase and contribute partly to myofibroblasts pool in streptozocin-induced acute or chronic islet injury and fibrosis. Furthermore, we substantiated the functional contribution of PSCs to fibrogenesis of pancreatic exocrine and islet in PSCs ablated mice. We also found stellate cells’ genetic ablation can improve pancreatic exocrine but not islet fibrosis. Together, our data indicates that stellate cells are vital/partial contributors to myofibroblasts in pancreatic exocrine/islet fibrosis.

Introduction: The stereo matching technology of satellite imagery is an important way to reconstruct real world. Most stereo matching technologies for satellite imagery are based on depth learning. However, the existing depth learning based methods have the problems of holes and matching errors in stereo matching tasks.Methods: In order to improve the effect of satellite image stereo matching results, we propose a satellite image stereo matching network based on attention mechanism (A-SATMVSNet). To solve the problem of insufficient extraction of surface features, a new feature extraction module based on triple dilated convolution with attention module is proposed, which solves the problem of matching holes caused by insufficient extraction of surface features. At the same time, compared with the traditional weighted average method, we design a novel cost-volume method that integrates attention mechanism to reduce the impact of matching errors to improve the accuracy of matching.Results and discussion: Experiments on public multi-view stereo matching dataset based on satellite imagery demonstrate that the proposed method significantly improves the accuracy and outperforms various previous methods. The validation of satellite remote sensing surface reflectance products is aimed at comparing the reflectance pixel values of products with ground measurement values at the pixel scale. Due to the existence of surface heterogeneity, we cannot obtain the satellite pixel scale truth value through ground sampling, and only the satellite relative pixel scale truth value that closely approximates it can be acquired. The process of converting the point-scale spectrum of ground sampling into a pixel-scale spectrum will produce certain errors, known as point-to-pixel-scale conversion uncertainty, which is closely related to the type of sample area and the ground sampling method. In this study, we conducted research on the uncertainty of point-to-pixel-scale conversion generated via different ground sampling methods in the upscaling process. We utilized unmanned aerial vehicle (UAV) hyperspectral images to invert the surface reflectance spectral curves of wheat, corn, bare soil, and soybeans at the pixel scale, and simulate the ground measurement spectra and satellite pixel scale ground truth of different sampling methods, so as to realize the quantitative calculation of the uncertainty of the ground truth at the satellite pixel scale. On this basis, we analyzed in depth the effects of the sampling method, measurement height, and number of spectra on the scale conversion uncertainty. The research results show that airborne hyperspectral images can accurately simulate the spectra of ground measurements, and can be used as an effective means of ground spectral sampling and uncertainty analysis. When using the systematic sampling method, the more the sampling points, the smaller the uncertainty. However, the uncertainty of scale conversion tends to stabilize when the number of sampling points is increased to a certain quantity. As the height of ground measurement increases, the number of spectra within the elementary sampling unit (ESU) increases, leading to smaller scale conversion uncertainties. The research results of this study will provide support for the subsequent optimization of ground sampling methods and the improvement of measurement efficiency and measurement accuracy.

Abstract Click-through rate prediction, which aims to predict the probability of the user clicking on an item, is critical to online advertising. How to capture the user evolving interests from the user behavior sequence is an important issue in CTR prediction. However, most existing models ignore the factor that the sequence is composed of sessions, and user behavior can be divided into different sessions according to the occurring time. The user behaviors are highly correlated in each session and are not relevant across sessions. We propose an effective model for CTR prediction, named Session Interest Model via Self-Attention (SISA). First, we divide the user sequential behavior into session layer. A self-attention mechanism with bias coding is used to model each session. Since different session interest may be related to each other or follow a sequential pattern, next, we utilize gated recurrent unit (GRU) to capture the interaction and evolution of user different historical session interests in session interest extractor module. Then, we use the local activation and GRU to aggregate their target ad to form the final representation of the behavior sequence in session interest interacting module. Experimental results show that the SISA model performs better than other models.

Lithium dendrite and parasitic reactions are two major challenges for lithium metal anode. Here, the authors show suppression of lithium-dendrite and elimination of continuous parasitic reactions by tuning the reduction kinetics of lithium-ion through a uniform nano-porous separator.

K+ channels of the Shaker family have been shown to play crucial roles in K+ uptake and transport. Cotton (Gossypium hirsutum) is an important cash crop. In this study, the 24 Shaker family genes were identified in cotton. Phylogenetic analysis suggests that they were assigned to five clusters. Additionally, their chromosomal location, conserved motifs and gene structure were analyzed. The promoter of cotton Shaker K+ channel genes comprises drought-, low-temperature-, phytohormone-response elements, etc. As indicated by qRT-PCR (quantitative real-time PCR), cotton Shaker K+ channel genes responded to low K+ and NaCl, and especially dehydration stress, at the transcript level. Moreover, one of the Shaker K+ channel genes, GhKAT1aD, was characterized. This gene is localized in the plasma membrane and is predicted to contain six transmembrane segments. It restored the growth of the yeast mutant strain defective in K+ uptake, and silencing GhKAT1a via VIGS (virus-induced gene silencing) resulted in more severe symptoms of K+ deficiency in cotton leaves as well as a lower net K+ uptake rate. The results of this study showed the overall picture of the cotton Shaker K+ channel family regarding bioinformatics as well as the function of one of its members, which provide clues for future investigations of cotton K+ transport and molecular insights for breeding K+-efficient cotton varieties.

As a critical component of many remote sensing satellites and model validation, pixel-scale surface quantitative parameters are often affected by scale effects in the acquisition process, resulting in deviations in the accuracy of image scale parameters. Consequently, various successive scale conversion methods have been proposed to correct the errors caused by scale effects. In this study, we propose ResTransformer, a deep learning model for scale conversion of surface reflectance using UAV images, which fully extracts and fuses the features of UAV images in the sample area and sample points and establishes a high-dimensional nonlinear spatial correlation between sample points and sample area in the target sample area, so that the scale conversion of surface reflectance at the pixel-scale can be completed quickly and accurately. We collected and created a dataset of 500k samples to verify the accuracy and robustness of the model with other traditional scale conversion methods. The results show that the ResTransformer deep learning model works best, providing average MRE, average MRSE, and correlation coefficient R values of 0.6440%, 0.7460, and 0.99911, respectively, and the baseline improvements compared with the Simple Average method are 92.48%, 92.45%, and 16.59%, respectively. The ResTransformer model also shows the highest robustness and universality and can adapt to surface pixel-scale conversion scenarios with different sizes, heterogeneous sample areas, and arbitrary sampling methods. This method provides a promising, highly accurate, and robust method for converting pixel-scale surface reflectance scale.

Abstract Our aims were to determine the accuracy of an improved formula for determining the minimum occlusive force (MOF) of a vascular clamp on rats’ abdominal aortas, compare our findings with the calculated theoretical MOF, and provide reference data for clinical research and development of medical instruments that cause minimal damage. We created a vessel closure model and developed a formula for calculating the theoretical MOF of arterial vessels when they are occluded. This formula utilises the blood pressure in the blood vessel, its diameter, and the width of the vascular clamp. We then measured the actual MOF in 24 rat abdominal aortic segments with different diameters and different blood pressures and compared the theoretical and actual MOFs. Analysis of the experimental data showed a probability of 0.315, which means that, under the condition of normal distribution, the difference between the theoretical and actual MOF is not significant at the α = 0.05 level. Thus, the actual measured MOF tended to be consistent with the theoretical MOF calculated by the formula we developed. The improved formula will provide a reference for clinical research and development of medical instruments that cause minimal injury, thus contributing to the development of microsurgery.

In modern society, dairy products have become increasingly important in our diet because of changes in consumption patterns due to urbanization. However, Chinese residents' dairy consumption remains at a relatively low level, with great potential for growth. Exploring the main determinants of dairy consumption and their effect mechanisms not only helps to improve the health status of residents, but also has important policy implications for the development of China's dairy industry. Based on the data of China Health and Nutrition Survey (CHNS) from 1989 to 2011, this study empirically analyzes the impact of urbanization on residents' dairy consumption. The results indicate that urbanization could significantly promote residents' consumption of dairy products and the effect is higher in areas with low urbanization levels and in midwestern regions than in areas with high urbanization levels and in midwestern regions. From the perspective of effect mechanism, income growth, employment structure transition and the rise of modern markets are three important mediating paths. Additionally, the results imply that in areas with low urbanization levels, income growth and the rise of modern markets are the main significant mediators; while in areas with high urbanization levels, employment structure transition is a significant mediator. Moreover, in midwestern regions, income growth is a significant mediator, and employment structure transition is a significant mediator in all regions. These findings have practical implications for understanding the relationship between urbanization and residents' food consumption and for further promoting residents' dairy consumption and the development of China's dairy industry.

Climate is a dominant factor affecting the potential geographical distribution of species. Understanding the impact of climate change on the potential geographic distribution of species, which is of great significance to the exploitation, utilization, and protection of resources, as well as ecologically sustainable development. Betula platyphylla Suk. is one of the most widely distributed temperate deciduous tree species in East Asia and has important economic and ecological value. Based on 231 species distribution data points of Betula platyphylla Suk. in China and 37 bioclimatic, soil, and topography variables (with correlation coefficients < 0.75), the potential geographical distribution pattern of Betula platyphylla Suk. under Representative Concentration Pathway (RCP) climate change scenarios at present and in the 2050s and 2070s was predicted using the MaxEnt model. We analyzed the main environmental variables affecting the distribution and change of suitable areas and compared the scope and change of suitable areas under different climate scenarios. This study found: (1) At present, the main suitable area for Betula platyphylla Suk. extends from northeastern to southwestern China, with the periphery area showing fragmented distribution. (2) Annual precipitation, precipitation of the warmest quarter, mean temperature of the warmest quarter, annual mean temperature, and precipitation of the driest month are the dominant environmental variables that affect the potential geographical distribution of Betula platyphylla Suk. (3) The suitable area for Betula platyphylla Suk. is expected to expand under global warming scenarios. In recent years, due to the impact of diseases and insect infestation, and environmental damage, the natural Betula platyphylla Suk. forest in China has gradually narrowed. This study accurately predicted the potential geographical distribution of Betula platyphylla Suk. under current and future climate change scenarios, which can provide the scientific basis for the cultivation, management, and sustainable utilization of Betula platyphylla Suk. resources.

Click-through rate prediction has become a hot research direction in the field of advertising. It is important to build an effective CTR prediction model. However, most existing models ignore the factor that the sequence is composed of sessions, and the user behaviors are highly correlated in each session and are not relevant across sessions. In this paper, we focus on user multiple session interest and propose a hierarchical model based on session interest (SIHM) for CTR prediction. First, we divide the user sequential behavior into session layer. Then, we employ a self-attention network obtain an accurate expression of interest for each session. Since different session interest may be related to each other or follow a sequential pattern, next, we utilize bidirectional long short-term memory network (BLSTM) to capture the interaction of different session interests. Finally, the attention mechanism based LSTM (A-LSTM) is used to aggregate their target ad to find the influences of different session interests. Experimental results show that the model performs better than other models.

Summary: Nowadays, wearable devices mainly exist in the form of portable accessories with various functions, connecting various kinds of terminals like mobile phones to form various wearable systems. In a wearable system, the wearable power supply device is the key component as energy dispenser for all devices. Nanosheets, a kind of two-dimensional material, which always displays a high surface-to-volume ratio and thus is lightweight and has remarkable conductive as well as electrochemical properties, have become the optimal choice for wearable power supply devices. The development and status of nanosheet-based wearable power supply devices including nanosheet-based wearable batteries, nanosheet-based wearable supercapacitors, nanosheet-based wearable self-powered energy suppliers are introduced in this article. Besides, the future opportunities and challenges of wearable devices are discussed.

Although microrobotic swarms are attractive for biomedical applications, controlling their collective movement in specific bio-fluids is difficult. Here, the authors report navigated locomotion and collective behaviors of magnetic field- and medium-induced micro-robotic swarms in various bio-fluids.

Summary: Combining catalysts with active colloidal matter could keep catalysts from aggregating, a major problem in chemical reactions. We report a kind of ensemble of bubble-cross-linked magnetic colloidal swarming nanomotors (B-MCS) with enhanced catalytic activity because of the local increase of the nanocatalyst concentration and three-dimensional (3D) fluid convection. Compared with the two-dimensional swarming collective without bubbles, the integral rotation was boosted because of the dynamic dewetting and increased slip length caused by the continuously ejected tiny bubbles. The bubbles cross-link the nanocatalysts and form stack along the vertical axis, generating the 3D network-like B-MCS ensemble with high dynamic stability and low drag resistance. The generated B-MCS ensemble exhibits controllable locomotion performance when applying a rotating magnetic field. Summary: The processes underlying synchronous multiple organ fibrosis in systemic sclerosis (SSc) remain poorly understood. Age-related pathologies are associated with organismal decline in nicotinamide adenine dinucleotide (NAD+) that is due to dysregulation of NAD+ homeostasis and involves the NADase CD38. We now show that CD38 is upregulated in patients with diffuse cutaneous SSc, and CD38 levels in the skin associate with molecular fibrosis signatures, as well as clinical fibrosis scores, while expression of key NAD+-synthesizing enzymes is unaltered. Boosting NAD+ via genetic or pharmacological CD38 targeting or NAD+ precursor supplementation protected mice from skin, lung, and peritoneal fibrosis. In mechanistic experiments, CD38 was found to reduce NAD+ levels and sirtuin activity to augment cellular fibrotic responses, while inhibiting CD38 had the opposite effect. Thus, we identify CD38 upregulation and resulting disrupted NAD+ homeostasis as a fundamental mechanism driving fibrosis in SSc, suggesting that CD38 might represent a novel therapeutic target.

Abstract The ability to successfully regulate negative emotions such as fear and anxiety is vital for mental health. Intranasal administration of the neuropeptide oxytocin (OXT) has been shown to reduce amygdala activity but to increase amygdala–prefrontal cortex connectivity during exposure to threatening stimuli suggesting that it may act as an important modulator of emotion regulation. The present randomized, between‐subject, placebo‐controlled pharmacological study combines the intranasal administration of OXT with functional magnetic resonance imaging (fMRI) during an explicit emotion regulation paradigm in 65 healthy male participants to investigate the modulatory effects of OXT on both bottom‐up and top‐down emotion regulation. OXT attenuates the activation in the posterior insular cortex and amygdala during anticipation of top‐down regulation of predictable threat stimuli in participants with high trait anxiety. In contrast, OXT enhances amygdala activity during the bottom‐up anticipation of unpredictable threat stimuli in participants with low trait anxiety. OXT may facilitate top‐down goal‐directed attention by attenuating amygdala activity in high anxiety individuals, while promoting bottom‐up attention/vigilance to unexpected threats by enhancing amygdala activity in low anxiety individuals. OXT may thus have the potential to promote an adaptive balance between bottom‐up and top‐down attention systems depending on an individual's trait anxiety level.

Abstract RVX-208 is a recently reported inhibitor of bromo and extraterminal (BET) family proteins (including BRD2-4 and BRDT) with selectivity for the second bromodomain (BD2), currently in phase III clinical trials. Despite of its promising antitumor activity, due to the conserved folds of the first and second bromodomains (BD1 and BD2), the detailed selectivity mechanism of RVX-208 towards BD2 over BD1 is still unknown. To elucidate selective inhibition mechanism of RVX-208 to BD2, microsecond molecular dynamics simulations were performed in this study for BRD2-BD1, BRD2-BD2 and BRD4-BD1 with and without RVX-208, respectively. Binding free energy calculations show that there exists strongest interaction between RVX-208 and BRD2-BD2. Leu383 and Asn429 are two most important residues of BRD2-BD2 for binding to RVX-208. Structural network analysis reveals that RVX-208 can shorten the communication path of ZA and BC loops in BRD2-BD2 pocket, making pocket more suitable to accommodate RVX-208. Additionally, different behaviors of His433 (Asp160 in BRD2-BD1) and Val435 (Ile162 in BRD2-BD1) in BRD2-BD2 are key factors responsible for selective binding of RVX-208 to BRD2-BD2. The proposed selective inhibition mechanism of RVX-208 to BRD2-BD2 can be helpful for rational design of novel selective inhibitors of the second bromodomain of BET family proteins.

Abstract The chloride/phosphate ratio (Cl/PO4) has been suggested to have a role in primary hyperparathyroidism (PHPT), but the associations between Cl/PO4 combined with ALP level and PHPT has not been well-studied. Our aim was to investigate the predictive value of combination Cl/PO4 with ALP for PHPT. A cross-sectional retrospective analysis was conducted to examine 172 patients diagnosed with PHPT categorized into two groups: normocalcaemic primary hyperparathyroidism (NPHPT) group and hypercalcaemia PHPT group. We found that Cl/PO4 levels and ALP levels in the NPHPT and hypercalcaemia PHPT group were both significantly higher than normal controls. Cl/PO4 and ALP levels were an independent risk factor for PHPT. Cl/PO4 combined with ALP increased the receiver-operating characteristic curves (ROC-AUC) and the diagnostic value in NPHPT and hypercalcaemia PHPT group (0.913; 95% CI, 0.744–1.000 and 0.932; 95% CI, 0.897–0.966, respectively), specificity of 92.8% and sensitivity of 98%. In conclusion, combination Cl/PO4 with ALP might be a low-cost, simple, available predictive marker of PHPT in Chinese individuals, particularly Chinese remote region where the method used to measure PTH cannot be done. Moreover, due to serum calcium level in NPHPT, Cl/PO4 combined with ALP level measurement have great potential to predict significant occurrence of NPHPT.

Abstract Asynchronous calibration quantitative computed tomography (QCT) is a new tool that allows the quantification of bone mineral density (BMD) without the use of a calibration phantom during scanning; however, this tool is not fully validated for clinical use. We used the European spine phantom (ESP) with repositioning during scanning and assessed the accuracy and short-term reproducibility of asynchronous QCT. Intra-scanner and intra-observer precision were each calculated as the root mean square of the standard deviation (RMSSD) and the coefficient of variation (CV-RMSSD). We also compared asynchronous and conventional QCT results in 50 clinical subjects. The accuracy of asynchronous QCT for three ESP vertebrae ranged from 1.4–6.7%, whereas intra-scanner precision for these vertebrae ranged from 0.53–0.91 mg/cc. Asynchronous QCT was most precise for a trabecular BMD of 100 mg/cc (CV-RMSSD = 0.2%). For intra-observer variability, overall precision error was smaller than 3%. In clinical subjects there was excellent agreement between the two calibration methods with correlation coefficients ranging from 0.96–0.99. A Bland–Altman analysis demonstrated that methodological differences depended on the magnitude of the BMD variable. Our findings indicate that the asynchronous QCT has good accuracy and precision for assessing trabecular BMD in the spine.

Abstract Coalescence and splitting of liquid marbles (LMs) are critical for the mixture of precise amount precursors and removal of the wastes in the microliter range. Here, the coalescence and splitting of LMs are realized by a simple gravity‐driven impact method and the two processes are systematically investigated to obtain the optimal parameters. The formation, coalescence, and splitting of LMs can be realized on‐demand with a designed channel box. By selecting the functional channels on the device, gravity‐based fusion and splitting of LMs are performed to mix medium/drugs and remove spent culture medium in a precise manner, thus ensuring that the microenvironment of the cells is maintained under optimal conditions. The LM‐based 3D stem cell spheroids are demonstrated to possess an approximately threefold of cell viability compared with the conventional spheroid obtained from nonadhesive plates. Delivery of the cell spheroid to a hydrophilic surface results in the in situ respreading of cells and gradual formation of typical 2D cell morphology, which offers the possibility for such spheroid‐based stem cell delivery in regenerative medicine.

Identification of the most influential spreaders that maximize information propagation in social networks is a classic optimization problem, called the influence maximization (IM) problem. A reasonable diffusion model that can accurately simulate information propagation in social networks is the key step to efficiently solving the IM problem. Synergism of neighbor nodes plays an important role in information propagation dynamics. Some known diffusion models have considered the reinforcement mechanism in defining the activation threshold. Most of these models focus on the synergetic effects of nodes on their common neighbors, but the accumulation of synergism has been neglected in previous studies. Inspired by these facts, we first discuss the catalytic role of synergism in the spreading dynamics of social networks and then propose a novel diffusion model called the synergism-based three-step cascade model (TSSCM) based on the above analysis and the three-degree influence theory. Finally, we devise an algorithm for solving the IM problem based on the TSSCM. Experiments on five real large-scale social networks demonstrate the efficacy of our method, which achieves competitive results in terms of influence spreading compared to the four other algorithms tested.

Although critical for studies of gut motility and intestinal regeneration, the in vitro culture of intestinal muscularis with peristaltic function remains a significant challenge. Periodic contractions of intestinal muscularis result from the coordinated activity of smooth muscle cells (SMC), the enteric nervous system (ENS), and interstitial cells of Cajal (ICC). Reproducing this activity requires the preservation of all these cells in one system. Here we report the first serum-free culture methodology that consistently maintains spontaneous and periodic contractions of murine and human intestinal muscularis cells for months. In this system, SMC expressed the mature marker myosin heavy chain, and multipolar/dipolar ICC, uniaxonal/multipolar neurons and glial cells were present. Furthermore, drugs affecting neural signals, ICC or SMC altered the contractions. Combining this method with scaffolds, contracting cell sheets were formed with organized architecture. With the addition of intestinal epithelial cells, this platform enabled up to 11 types of cells from mucosa, muscularis and serosa to coexist and epithelial cells were stretched by the contracting muscularis cells. The method constitutes a powerful tool for mechanistic studies of gut motility disorders and the functional regeneration of the engineered intestine.

Wound healing is a complex biological process, and current research finds that jellyfish have a great capacity for promoting growth and healing. However, the underlying mechanisms remain unclear. Thus, this study was conducted to investigate the molecular mechanisms and effects of a tentacle extract (TE) from the jellyfish Cyanea capillata (C. capillata) on cell proliferation and migration in human umbilical vein endothelial cells (HUVECs). First, our results showed that TE at the concentration of 1 μg/ml could promote cell proliferation over various durations, induce a transition of the cells from the G1-phase to the S/G2-phase of the cell cycle, and increase the expression of cell cycle proteins (CyclinB1 and CyclinD1). Second, we found that TE could activate the PI3K/Akt, ERK1/2 and JNK MAPK signaling pathways but not the NF-κB signaling pathway or the apoptosis signaling cascade. Finally, we demonstrated that the TE-induced expression of cell cycle proteins was decreased by ERK1/2 inhibitor PD98059 but not by PI3K inhibitor LY294002 or JNK inhibitor SP600125. Similarly, the TE-enhanced migration ability of HUVECs was also markedly attenuated by PD98059. Taken together, our findings indicate that TE-induced proliferation and migration in HUVECs mainly occurred through the ERK1/2 MAPK signaling pathway. These results are instructively important for further research on the isolation and purification of growth-promoting factors from C. capillata and are hopeful as a means to improve human wound repair in unfavorable conditions.

Bisphenol A (BPA) can interact with nuclear receptors and affect the normal function of nuclear receptors in very low doses, which causes BPA to be one of the most controversial endocrine disruptors. However, the detailed molecular mechanism about how BPA interferes the normal function of nuclear receptors is still undiscovered. Herein, molecular dynamics simulations were performed to explore the detailed interaction mechanism between BPA with three typical nuclear receptors, including hERα, hERRγ and hPPARγ. The simulation results and calculated binding free energies indicate that BPA can bind to these three nuclear receptors. The binding affinities of BPA were slightly lower than that of E2 to these three receptors. The simulation results proved that the binding process was mainly driven by direct hydrogen bond and hydrophobic interactions. In addition, structural analysis suggested that BPA could interact with these nuclear receptors by mimicking the action of natural hormone and keeping the nuclear receptors in active conformations. The present work provided the structural evidence to recognize BPA as an endocrine disruptor and would be important guidance for seeking safer substitutions of BPA.

BackgroundJellyfish contain diverse toxins and other bioactive components. However, large-scale identification of novel toxins and bioactive components from jellyfish has been hampered by the low efficiency of traditional isolation and purification methods.ResultsWe performed de novo transcriptome sequencing of the tentacle tissue of the jellyfish Cyanea capillata. A total of 51,304,108 reads were obtained and assembled into 50,536 unigenes. Of these, 21,357 unigenes had homologues in public databases, but the remaining unigenes had no significant matches due to the limited sequence information available and species-specific novel sequences. Functional annotation of the unigenes also revealed general gene expression profile characteristics in the tentacle of C. capillata. A primary goal of this study was to identify putative toxin transcripts. As expected, we screened many transcripts encoding proteins similar to several well-known toxin families including phospholipases, metalloproteases, serine proteases and serine protease inhibitors. In addition, some transcripts also resembled molecules with potential toxic activities, including cnidarian CfTX-like toxins with hemolytic activity, plancitoxin-1, venom toxin-like peptide-6, histamine-releasing factor, neprilysin, dipeptidyl peptidase 4, vascular endothelial growth factor A, angiotensin-converting enzyme-like and endothelin-converting enzyme 1-like proteins. Most of these molecules have not been previously reported in jellyfish. Interestingly, we also characterized a number of transcripts with similarities to proteins relevant to several degenerative diseases, including Huntington's, Alzheimer's and Parkinson's diseases. This is the first description of degenerative disease-associated genes in jellyfish.ConclusionWe obtained a well-categorized and annotated transcriptome of C. capillata tentacle that will be an important and valuable resource for further understanding of jellyfish at the molecular level and information on the underlying molecular mechanisms of jellyfish stinging. The findings of this study may also be used in comparative studies of gene expression profiling among different jellyfish species.

Thioredoxins (Trx proteins) are a family of small, highly-conserved and ubiquitous proteins that play significant roles in the resistance of oxidative damage. In this study, a homologue of Trx was identified from the cDNA library of tentacle of the jellyfish Cyanea capillata and named CcTrx1. The full-length cDNA of CcTrx1 was 479 bp with a 312 bp open reading frame encoding 104 amino acids. Bioinformatics analysis revealed that the putative CcTrx1 protein harbored the evolutionarily-conserved Trx active site 31CGPC34 and shared a high similarity with Trx1 proteins from other organisms analyzed, indicating that CcTrx1 is a new member of Trx1 sub-family. CcTrx1 mRNA was found to be constitutively expressed in tentacle, umbrella, oral arm and gonad, indicating a general role of CcTrx1 protein in various physiological processes. The recombinant CcTrx1 (rCcTrx1) protein was expressed in Escherichia coli BL21 (DE3), and then purified by affinity chromatography. The rCcTrx1 protein was demonstrated to possess the expected redox activity in enzymatic analysis and protection against oxidative damage of supercoiled DNA. These results indicate that CcTrx1 may function as an important antioxidant in C. capillata. To our knowledge, this is the first Trx protein characterized from jellyfish species.

BACKGROUND AND AIMS: MET, the hepatocyte growth factor receptor, is a receptor tyrosine kinase overexpressed and activated in a subset of gastric cancer. Several studies investigated the relationship between MET amplification and expression with the clinical outcome in patients with gastric cancer, but yielded conflicting results. We performed a systematic review and meta-analysis to determine the influence of MET amplification and expression on prognosis in gastric cancer. METHODS: MEDLINE and EMBASE were searched for studies that explored the association between MET amplification and expression with survival in patients with gastric cancer up to 1 April, 2013. Data of individual hazard ratios (HRs) and 95% confidence intervals (CIs) for meta-analyses were extracted from the publications and combined in pooled HRs. RESULTS: Fourteen studies involving 2,258 patients with gastric cancer were included. It was suggested that MET overexpression had an unfavorable impact on survival of patients with gastric cancer, with HRs (95% CIs) of 2.57 (95% CI: 1.97-3.35) overall, 2.82 (95% CI: 1.86-4.27) among studies using amplification for measure scale of MET and 2.42 (95% CI: 1.66-3.54) for expression. The magnitude of association was reduced whereas remained statistically significant in high quality studies or in larger sample size studies and corresponding HRs were 2.18(1.76, 2.70) and 2.35(1.93, 2.87), respectively, without significant heterogeneity. CONCLUSION: The findings from present study indicated that higher MET gene amplification and expression in gastric cancer was an indicator of poor prognosis.

EGCG possesses the ability of disaggregating the existing amyloid fibrils which were associated with many age-related degenerative diseases. However, the molecular mechanism of EGCG to disaggregate these fibrils is poorly known. In this work, to study the influence of EGCG on the full-length human islet amyloid polypeptide 1-37 (hIAPP1-37) oligomers, molecular dynamics simulations of hIAPP1-37 pentamer and decamer with EGCG were performed, respectively. The obtained results indicate that EGCG indeed destabilized the hIAPP1-37 oligomers. The nematic order parameter and secondary structure calculations coupled with the free-energy landscape indicate that EGCG broke the initial ordered pattern of two polymers, greatly reduced their β-sheet content and enlarged their conformational space. On this basis, three possible target sites were identified with the binding capacity order of S1>S2>S3. After a deeper analysis of each site, we found that S1 was the most possible site on which residues B-Ile26/Ala25, A-Phe23, B/C-Leu27 and E-Tyr37 played an important role for their binding. The proposal of this molecular mechanism can not only provide a prospective interaction figure between EGCG and β-sheet-rich fibrils of hIAPP1-37, but also is useful for further discovering other potential inhibitors.

BackgroundCell-free microRNAs stably and abundantly exist in body fluids and emerging evidence suggests cell-free microRNAs as novel and non-invasive disease biomarker. Deregulation of miR-29 is involved in the pathogenesis of diabetic nephropathy and insulin resistance thus may be implicated in diabetic vascular complication. Therefore, we investigated the possibility of urinary miR-29 as biomarker for diabetic nephropathy and atherosclerosis in patients with type 2 diabetes.Methods83 patients with type 2 diabetes were enrolled in this study, miR-29a, miR-29b and miR-29c levels in urine supernatant was determined by TaqMan qRT-PCR, and a synthetic cel-miR-39 was added to the urine as a spike-in control before miRNAs extraction. Urinary albumin excretion rate and urine albumin/creatinine ratio, funduscopy and carotid ultrasound were used for evaluation of diabetic vascular complication. The laboratory parameters indicating blood glucose level, renal function and serum lipids were also collected.ResultsPatients with albuminuria (n = 42, age 60.62 ± 12.00 yrs) showed significantly higher comorbidity of diabetic retinopathy (p = 0.015) and higher levels of urinary miR-29a (p = 0.035) compared with those with normoalbuminuria (n = 41, age 58.54 ± 14.40 yrs). There was no significant difference in urinary miR-29b (p = 0.148) or miR-29c level (p = 0.321) between groups. Urinary albumin excretion rate significantly correlated with urinary miR-29a level (r = 0.286, p = 0.016), while urinary miR-29b significantly correlated with carotid intima-media thickness (cIMT) (r = 0.286, p = 0.046).ConclusionUrinary miR-29a correlated with albuminuria while urinary miR-29b correlated with carotid intima-media thickness (cIMT) in patients with type 2 diabetes. Therefore, they may have the potential to serve as alternative biomarker for diabetic nephropathy and atherosclerosis in type 2 diabetes.
