Abstract Drug screening based on in-vitro primary tumor cell culture has demonstrated potential in personalized cancer diagnosis. However, the limited number of tumor cells, especially from patients with early stage cancer, has hindered the widespread application of this technique. Hence, we developed a digital microfluidic system for drug screening using primary tumor cells and established a working protocol for precision medicine. Smart control logic was developed to increase the throughput of the system and decrease its footprint to parallelly screen three drugs on a 4 × 4 cm2 chip in a device measuring 23 × 16 × 3.5 cm3. We validated this method in an MDA-MB-231 breast cancer xenograft mouse model and liver cancer specimens from patients, demonstrating tumor suppression in mice/patients treated with drugs that were screened to be effective on individual primary tumor cells. Mice treated with drugs screened on-chip as ineffective exhibited similar results to those in the control groups. The effective drug identified through on-chip screening demonstrated consistency with the absence of mutations in their related genes determined via exome sequencing of individual tumors, further validating this protocol. Therefore, this technique and system may promote advances in precision medicine for cancer treatment and, eventually, for any disease.

Abstract The microbiome in a specific human organ has been well-studied, but few reports have investigated the multi-organ microbiome as a whole. Here, we aim to analyse the intra-individual inter-organ and intra-organ microbiome in deceased humans. We collected 1608 samples from 53 sites of 7 surface organs (oral cavity, esophagus, stomach, small intestine, appendix, large intestine and skin; n = 33 subjects) and performed microbiome profiling, including 16S full-length sequencing. Microbial diversity varied dramatically among organs, and core microbial species co-existed in different intra-individual organs. We deciphered microbial changes across distinct intra-organ sites, and identified signature microbes, their functional traits, and interactions specific to each site. We revealed significant microbial heterogeneity between paired mucosa-lumen samples of stomach, small intestine, and large intestine. Finally, we established the landscape of inter-organ relationships of microbes along the digestive tract. Therefore, we generate a catalogue of bacterial composition, diversity, interaction, functional traits, and bacterial translocation in human at inter-organ and intra-organ levels.

Abstract Preeclampsia (PE) is considered as a disease of placental origin. However, the specific mechanism of placental abnormalities remains elusive. This study identified thrombospondin‐1 (THBS1) is downregulated in preeclamptic placentae and negatively correlated with blood pressure. Functional studies show that THBS1 knockdown inhibits proliferation, migration, and invasion and increases the cycle arrest and apoptosis rate of HTR8/SVneo cells. Importantly, THBS1 silencing induces necroptosis in HTR8/SVneo cells, accompanied by the release of damage‐associated molecular patterns (DAMPs). Necroptosis inhibitors necrostatin‐1 and GSK′872 restore the trophoblast survival while pan‐caspase inhibitor Z‐VAD‐FMK has no effect. Mechanistically, the results show that THBS1 interacts with transforming growth factor B‐activated kinase 1 (TAK1), which is a central modulator of necroptosis quiescence and affects its stability. Moreover, THBS1 silencing up‐regulates the expression of neuronal precursor cell‐expressed developmentally down‐regulated 4 (NEDD4), which acts as an E3 ligase of TAK1 and catalyzes K48‐linked ubiquitination of TAK1 in HTR8/SVneo cells. Besides, THBS1 attenuates PE phenotypes and improves the placental necroptosis in vivo. Taken together, the down‐regulation of THBS1 destabilizes TAK1 by activating NEDD4‐mediated, K48‐linked TAK1 ubiquitination and promotes necroptosis and DAMPs release in trophoblast cells, thus participating in the pathogenesis of PE.

Abstract KRAS is an important tumor intrinsic factor driving immune suppression in colorectal cancer (CRC). In this study, we demonstrate that SLC25A22 underlies mutant KRAS-induced immune suppression in CRC. In immunocompetent male mice and humanized male mice models, SLC25A22 knockout inhibits KRAS-mutant CRC tumor growth with reduced myeloid derived suppressor cells (MDSC) but increased CD8+ T-cells, implying the reversion of mutant KRAS-driven immunosuppression. Mechanistically, we find that SLC25A22 plays a central role in promoting asparagine, which binds and activates SRC phosphorylation. Asparagine-mediated SRC promotes ERK/ETS2 signaling, which drives CXCL1 transcription. Secreted CXCL1 functions as a chemoattractant for MDSC via CXCR2, leading to an immunosuppressive microenvironment. Targeting SLC25A22 or asparagine impairs KRAS-induced MDSC infiltration in CRC. Finally, we demonstrate that the targeting of SLC25A22 in combination with anti-PD1 therapy synergizes to inhibit MDSC and activate CD8+ T cells to suppress KRAS-mutant CRC growth in vivo. We thus identify a metabolic pathway that drives immunosuppression in KRAS-mutant CRC.

Abstract Excessive CO2 and food shortage are two grand challenges of human society. Directly converting CO2 into food materials can simultaneously alleviate both, like what green crops do in nature. Nevertheless, natural photosynthesis has a limited energy efficiency due to low activity and specificity of key enzyme D‐ribulose‐1,5‐bisphosphate carboxylase/oxygenase (RuBisCO). To enhance the efficiency, many prior studies focused on engineering the enzymes, but this study chooses to learn from the nature to design more efficient reactors. This work is original in mimicking the stacked structure of thylakoids in chloroplasts to immobilize RuBisCO in a microreactor using the layer‐by‐layer strategy, obtaining the continuous conversion of CO2 into glucose precursor at 1.9 nmol min−1 with enhanced activity (1.5 times), stability (≈8 times), and reusability (96% after 10 reuses) relative to the free RuBisCO. The microreactors are further scaled out from one to six in parallel and achieve the production at 15.8 nmol min−1 with an energy conversion efficiency of 3.3 times of rice, showing better performance of this artificial synthesis than NPS in terms of energy conversion efficiency. The exploration of the potential of mass production would benefit both food supply and carbon neutralization.

This study aims to compare rumen microbiome and metabolites between second lactation dairy cows in the 75th percentile (n = 12; 57.2 ± 5.08 kg/d) of production according to genomic predicted transmitting ability for milk (GPTAM) and their counterparts in the 25th percentile (n = 12; 47.2 ± 8.61 kg/d). It was hypothesized that the metagenome and metabolome would differ between production levels. Cows were matched by days in milk (DIM), sire, occurrence of disease, and days open in previous lactation. For an additional comparison, the cows were also divided by phenotype into high (n = 6; 61.3 ± 2.8 kg/d), medium (n = 10; 55 ± 1.2 kg/d), and low (n = 8; 41.9 ± 5.6 kg/d) based on their milk production. Samples were collected 65 ± 14 DIM. Rumen content was collected using an oro-gastric tube and serum samples were collected from the coccygeal vessels. High-resolution liquid chromatography-mass spectrometry (LC-MS) was used for rumen and serum metabolite profiling. Shotgun metagenomics was used for rumen microbiome profiling. Microbiome sample richness and diversity were used to determine alpha and Bray-Curtis dissimilarity index was used to estimate beta diversity. Differences in metabolites were determined using t-tests or ANOVA. Pearson correlations were used to consider associations between serum metabolites and milk production. There was no evidence of a difference in rumen metabolites or microbial communities by GPTAM or phenotype. Cows in the phenotypic low group had greater serum acetate to propionate ratio and acetate proportion compared to the cows in the phenotypic medium group. Likewise, serum propionate proportion was greater in the medium compared to the low phenotypic group. Serum acetate, butyrate, and propionate concentrations had a weak positive correlation with milk production. When investigating associations between rumen environment and milk production, future studies must consider the impact of the ruminal epithelium absorption and post-absorption processes in relation to milk production.

Land Use/Land Cover (LULC) is one of the most significant human variables influencing the efficiency of Ecosystem Services (ESs) in terrestrial ecosystems. Theoretical and technical assistance for regional sustainable land use planning and management, as well as ecosystem conservation and restoration, is provided by investigating the influence of changes in the LULC pattern on the efficiency of ESs. This research focuses on the interactions between socioeconomic activities and natural ecological processes in the Three Gorges Reservoir Area (TGRA). We use LULC data from the TGRA for the years 1990, 2000, 2010, and 2020. The study includes the analysis and calculation of the spatiotemporal evolution features of the current LULC pattern and the efficiency of ESs, including their spatiotemporal distribution. Considering the TGRA's national development orientation and guidance, three potential LULC patterns are constructed under various develop-ment scenarios. To calculate the efficiency of ESs, the GeoSOS-FLUS future LULC simulation model is linked, and several methodologies such as INVEST, RUSLE, and CASA are used. The goal is to investigate the influence of future changes in LULC patterns on ESs efficiency. The findings show the following: (1) From 1990 to 2020, the values of water conservation services in the TGRA decreased and subsequently increased. High-value areas are primarily located in the reservoir's centre and eastern sections, whereas low-value areas are mostly found in the western section. Soil conservation service values initially declined and later climbed. The TGRA's carbon storage services have in-creased yearly, from 552.64 g/m2 in 2000 to 615.92 g/m2 in 2020. (2) In the ecological protection scenario, carbon storage and soil erosion increased compared to the ecosystem services in 2020. The ecological system service benefits are greater when compared to the natural development scenario. (3) The four ESs show positive spatial correlations across all three scenarios, and local spatial au-tocorrelation analysis findings demonstrate that carbon storage, water yield, and habitat quality have comparable spatial distributions across all three scenarios. To some extent, high-value areas for water conservation, soil retention, carbon storage, and habitat quality overlap.

BackgroundStudies in general population reported a positive association between tobacco smoking and airflow obstruction (AFO), a hallmark of chronic obstructive pulmonary disease (COPD). However, this attempt was less addressed in silica dust-exposed workers.MethodsThis retrospective cohort study consisted of 4481 silicotic workers attending the Pneumoconiosis Clinic during 1981-2019. The lifelong work history and smoking habits of these workers were extracted from medical records. Spirometry was carried out at the diagnosis of silicosis (n = 4177) and reperformed after an average of 9.4 years of follow-up (n = 2648). AFO was defined as forced expiratory volume in one second (FEV1)/force vital capacity (FVC) less than lower limit of normal (LLN). The association of AFO with smoking status was determined using multivariate logistics regression, and the effect of smoking cessation on the development of AFO was evaluated Cox regression.ResultsSmoking was significantly associated with AFO (current smokers: OR = 1.92, 95% CI 1.51-2.44; former smokers: OR = 2.09, 95% CI 1.65-2.66). The risk of AFO significantly increased in the first 3 years of quitting smoking (OR = 1.23, 95% CI 1.02-1.47) but decreased afterwards with increasing years of cessation. Smoking cessation reduced the risk of developing AFO no matter before or after the confirmation of silicosis (pre-silicosis cessation: HR = 0.58, 95% CI 0.46-0.74; post-silicosis cessation: HR = 0.62, 95% CI 0.48-0.79).ConclusionsSmoking cessation significantly reduced the risk of AFO in the workers with silicosis, although the health benefit was not observed until 3 years of abstinence. These findings highlight the importance of early and long-term smoking cessation among silicotic or silica dust-exposed workers.

This study aimed to evaluate the seroprevalence and spatial and temporal clustering of SARS-CoV-2 antibodies in household cats within 63 counties in Illinois from October 2021 to May 2023. The analysis followed a stepwise approach. First, in a choropleth point map, we illustrated the distribution of county-level seroprevalence of SARS-CoV-2 antibodies. Next, spatial interpolation was used to predict the seroprevalence in counties without recorded data. Global and local clustering methods were used to identify the extent of clustering and the counties with high or low seroprevalence, respectively. Next, temporal, spatial, and space-time scan statistic was used to identify periods and counties with higher-than-expected seroprevalence. In the last step, to identify more distinct areas in counties with high seroprevalence, city-level analysis was conducted to identify temporal and space-time clusters. Among 1,715 samples tested by serological assays, 244 samples (14%) tested positive. Young cats had higher seropositivity than older cats, and the third quarter of the year had the highest odds of seropositivity. Three county-level space-time clusters with higher-than-expected seroprevalence were identified in the northeastern, central-east, and southwest regions of Illinois, occurring between June and October 2022. In the city-level analysis, 2 space-time clusters were identified in Chicago's downtown and the southwestern suburbs of Chicago between June and September 2022. Our results suggest that the high density of humans and cats in large cities such as Chicago, might play a role in the transmission and clustering of SARS-CoV-2. Our study provides an in-depth analysis of SARS-CoV-2 epidemiology in Illinois household cats, which will aid in COVID-19 control and prevention.

Appropriate environmental sensing methods and visualization representations are crucial foundations for the in situ exploration of planets. In this paper, we developed specialized visualization methods to facilitate the rover’s interaction and decision-making processes, as well as to address the path-planning and obstacle-avoidance requirements for lunar polar region exploration and Mars exploration. To achieve this goal, we utilize simulated lunar polar regions and Martian environments. Among them, the lunar rover operating in the permanently shadowed region (PSR) of the simulated crater primarily utilizes light detection and ranging (LiDAR) for environmental sensing; then, we reconstruct a mesh using the Poisson surface reconstruction method. After that, the lunar rover’s traveling environment is represented as a red-green-blue (RGB) image, a slope coloration image, and a theoretical water content coloration image, based on different interaction needs and scientific objectives. For the rocky environment where the Mars rover is traveling, this paper enhances the display of the rocks on the Martian surface. It does so by utilizing depth information of the rock instances to highlight their significance for the rover’s path-planning and obstacle-avoidance decisions. Such an environmental sensing and enhanced visualization approach facilitates rover path-planning and remote–interactive operations, thereby enabling further exploration activities in the lunar PSR and Mars, in addition to facilitating the study and communication of specific planetary science objectives, and the production and display of basemaps and thematic maps.

In this paper, a theoretical study was introduced to discussion the Influence of donor senstizer on efficiency of solar cell with clear focusing on dye senstized solar cell DSSCs applications was presented. Use of donor as -sensitizer dye in solar cells was a viable contender in photovoltaics due to their spectrum of excited state to transfer more elkectrons to conduction band of semiconductor .In this study, two systems Alq3/ZnO and D149/ZnO devices taken with same two solvents .Transtion energy ,coupling strength and transtion parameters are used to calculate the electron current density , it uses to calculate the photovoltic characteristic I-V ,fill factor and the efficiency of the solar cell. Especially, the largest performance was for the both D149/ ZnO and Alq3/ZnO solar cells based on Acetonitrile as a solvent with electron current density of (430 0.329-13.8) mA/cm^2 between (1-6) V and produced the highest efficiency calculated 2.593 and 0.938for the both solar cell was corresponds to the lowest efficiency about 0.536 for Alq3/ZnO with Ethanol solvent

Abstract Weakened circadian activity rhythms (CARs) were associated with mild cognitive impairment (MCI) in the general population. However, it remains unclear among pneumoconiosis patients. We aimed to address this knowledge gap. This cross-sectional study comprised 186 male pneumoconiosis patients (71.3 ± 7.8 years) and 208 healthy community men. Actigraphy was used to determine CARs parameters (percent rhythm, amplitude, MESOR, and acrophase). Values below the corresponding medians of the CARs parameters represented weakened CARs. The Cantonese version of Mini-Mental State Examination (CMMSE) was used to assess cognitive function, MCI, and the composite outcome of MCI plus cognitive impairment. Compared with the community referents, pneumoconiosis patients had worse cognition and dampened CARs. Compared with the community referents or pneumoconiosis patients with robust circadian rhythm, pneumoconiosis patients with weakened circadian rhythm were consistently associated with increased risk of MCI and the composite outcome. However, significant association was only observed between MESOR and the composite outcome (adjusted OR = 1.99, 95%: 1.04–3.81). A delayed phase of CARs was insignificantly associated with MCI and the composite outcome. Our findings showed that weakened CARs were associated with worse cognitive function among male pneumoconiosis workers. Intervention in improving CARs may mitigate cognitive deterioration in male pneumoconiosis workers.

Abstract Negaprion acutidens (sicklefin lemon shark) is distributed in the Indo-Pacific and in close association with coral reefs. Under the protection of the Dongsha Atoll National Park, a small but well-established juvenile population of N. acutidens inhabiting coastal areas of Dongsha Island was recently observed to display site fidelity by using acoustic telemetry. This study was designed to reveal the fine scale genetic structure and relatedness within and among 5 juvenile shark cohorts inhabiting 3 sampling sites at Dongsha Island. A total 188 juveniles were caught and sampled between 2016 and 2017, and genotyped with twelve loci. They were assigned to 5 year cohorts (2013–2017) based on the body length and date they were caught, also assigned to 3 sites based on where they were caught. Among five cohorts, the percentage of unrelated pairs within a cohort is more than 62% in average, suggesting a potential high mortality during their early life stage. The results of Fst and assignment testing showed that there was no significant genetic structure between sites and cohorts indicating that there was no fine scale genetic structure, even though the juveniles possessed strong site fidelity. A small effective population size (Ne) was detected (Ne = 86.7) which indicates the presence of a potentially isolated and vulnerable population at Dongsha. These results provide the genetic diversity as a baseline for future management and conservation of N. acutidens in the South China Sea.

Ship detection aims to automatically identify whether there are ships in the images, precisely classifies and localizes them. Regardless of whether utilizing early manually designed methods or deep learning technology, ship detection is dedicated to exploring the inherent characteristics of ships to enhance recall. Nowadays, high-precision ship detection plays a crucial role in civilian and military applications. In order to provide a comprehensive review of ship detection in optical remote-sensing images (SDORSIs), this paper summarizes the challenges as a guide. These challenges include complex marine environments, insufficient discriminative features, large scale variations, dense and rotated distributions, large aspect ratios, and imbalances between positive and negative samples. We meticulously review the improvement methods and conduct a detailed analysis of the strengths and weaknesses of these methods. We compile ship information from common optical remote sensing image datasets and compare algorithm performance. Simultaneously, we compare and analyze the feature extraction capabilities of backbones based on CNNs and Transformer, seeking new directions for the development in SDORSIs. Promising prospects are provided to facilitate further research in the future.

Summary: As we interact with our surroundings, we encounter the same or similar objects from different perspectives and are compelled to generalize. For example, despite their variety we recognize dog barks as a distinct sound class. While we have some understanding of generalization along a single stimulus dimension (frequency, color), natural stimuli are identifiable by a combination of dimensions. Measuring their interaction is essential to understand perception. Using a 2-dimension discrimination task for mice and frequency or amplitude modulated sounds, we tested untrained generalization across pairs of auditory dimensions in an automatized behavioral paradigm. We uncovered a perceptual hierarchy over the tested dimensions that was dominated by the sound’s spectral composition. Stimuli are thus not perceived as a whole, but as a combination of their features, each of which weights differently on the identification of the stimulus according to an established hierarchy, possibly paralleling their differential shaping of neuronal tuning.

Rapid industrialisation and urbanisation have moved contemporary civilization ahead but also deepened clashes with nature. Human society’s long-term evolution faces a number of serious problems, including the climate issue and frequent natural disasters. This research analyses the spatiotemporal evolution features of land use remote sensing data from 2005, 2010, 2015, and 2020. Under the Nature-based Solutions (NbS) idea, four scenarios are established: Business as Usual (BAU), Woodland Conservation (WLC), Arable Land Conservation (ALC), and Urban Transformation and Development (UTD). The RF–Markov–CA model is used to simulate the spatiotemporal patterns of land use for the years 2025 and 2030. Furthermore, the InVEST model is utilised to assess and forecast the spatiotemporal evolution features of carbon storage. The findings show that (1) the primary land use categories in the Three Gorges Reservoir Area (TGRA) from 2005 to 2020 are arable land and woodland. Arable land has a declining tendency, whereas woodland has an increasing–decreasing trend. (2) The WLC scenario exhibits the greatest growth in woodland and the lowest drop in grassland from 2020 to 2030, indicating a more stable ecosystem. (3) The TGRA demonstrates substantial geographic variation in carbon storage from 2005 to 2030, with a broad distribution pattern of “higher in the north, lower in the south, higher in the east, lower in the west, with the reservoir head > reservoir centre > reservoir tail”. (4) In comparison to the other three scenarios, the WLC scenario sees a slower development of construction and arable land from 2020 to 2030, whereas the ecological land area rises the highest and carbon storage increases. As a result, the WLC scenario is the TGRA’s recommended development choice. The study’s findings have substantial implications for the TGRA’s ecological preservation and management, as well as for the optimisation of ecosystem carbon cycling and the promotion of regional sustainable development.

The decline in neurogenesis following birth is accompanied with a quiescent state characteristic of neural progenitors of the adult brain. Here, the authors identify the Wnt pathway antagonist SFRP1 as a potential signal that promotes quiescence and show that its inhibition stimulates stem cell activation.

Summary: Different from traditional procedures of using solid stabilizers like polymers and surfactants, here we demonstrate that water, as a very “soft” matter, could function as a “spacer” to prevent the aggregation of metal-organic frameworks (MOFs) in aqueous dispersions. Our theoretical calculations reveal in case of an excess of positively charged metal nodes of MOFs, where water molecules are ligated to metal nodes that greatly enhance MOFs’ solution dispersibility through electrostatic stabilization. This discovery has motivated us to develop a facile experimental approach for producing a category of “clean” MOF dispersions without foreign additives. Potential application has been demonstrated for the size fractionation of MOFs, which results in small-size MOFs (50–80 nm) characteristic of superior electrocatalytic oxygen evolution activities (256 mV at 10 mA cm−2, Tafel slope of 49 mV dec−1 and durability >30 h). This work would provide new clues for aqueous processing of MOFs for many emerging applications.

Abstract Summer-spring predominance of tuberculosis (TB) has been widely reported. The relative contributions of exogenous recent infection versus endogenous reactivation to such seasonality remains poorly understood. Monthly TB notifications data between 2005 and 2017 in Hong Kong involving 64,386 cases (41% aged ≥ 65; male-to-female ratio 1.74:1) were examined for the timing, amplitude, and predictability of variation of seasonality. The observed seasonal variabilities were correlated with demographics and clinical presentations, using wavelet analysis coupled with dynamic generalised linear regression models. Overall, TB notifications peaked annually in June and July. No significant annual seasonality was demonstrated for children aged ≤ 14 irrespective of gender. The strongest seasonality was detected in the elderly (≥ 65) among males, while seasonal pattern was more prominent in the middle-aged (45–64) and adults (30–44) among females. The stronger TB seasonality among older adults in Hong Kong suggested that the pattern has been contributed largely by reactivation diseases precipitated by defective immunity whereas seasonal variation of recent infection was uncommon.

The continental lake basin is a favorable accumulation area of shale oil and gas; however, the organic matter enrichment regularity in continental lake facies is still controversial, which hinders the exploration of continental shale oil and gas in the Sichuan Basin. In this study, the relationship between a sedimentary environment and organic matter enrichment of continental shale was analyzed by petrology and organic and inorganic geochemistry tests of 153 samples. The results show that different lithologic associations in the Da’anzhai member have different contents of organic matter. Among them, the TOC content of unit II (shale-limestone interbed) is the highest, mainly, type II kerogen, followed by unit III (shale clip shell limestone belts), mainly, type III kerogen. Geochemical indicators show that different paleoenvironmental factors play different leading roles in organic matter enrichment in different periods of the Da’anzhai member. Unit Ⅱ is deposited in an arid environment with less fresh water supply; therefore, the water cycle is hampered. Due to the effect of salinity stratification, anoxic water was formed, which was beneficial to the preservation of algae and plankton in the lake basin, resulting in the formation of type Ⅱ2 kerogen. Unit III is deposited in a semi-humid and semi-arid climate, and its water is in an oxidizing environment. Precipitation and river runoff increase the input of terrigenous debris and higher plants to form type III kerogen. This study has guiding significance for the cause of organic matter enrichment in the Da’anzhai member and the prediction of favorable zones.

Hyperspectral image (HSI) super-resolution (SR) is a classical computer vision task that aims to accomplish the conversion of images from lower to higher resolutions. With the booming development of deep learning (DL) technology, more and more researchers are dedicated to the research of image SR techniques based on DL and have made remarkable progress. However, no scholar has provided a comprehensive review of the field. As a response, in this paper we aim to supply a comprehensive summary of the DL-based SR techniques for HSI, including upsampling frameworks, upsampling methods, network design, loss functions, representative works with different strategies, and future directions, in which we design several sets of comparative experiments for the advantages and limitations of two-dimensional convolution and three-dimensional convolution in the field of HSI SR and analyze the experimental results in depth. In addition, the paper also briefly discusses the secondary foci such as common datasets, evaluation metrics, and traditional SR algorithms. To the best of our knowledge, this paper is the first review on DL-based HSI SR.

Some cephalopods carry microorganisms in two specialized organs, the light organ and the accessory nidamental gland (ANG). For the light organ, comprehensive mechanisms have been described for winnowing (bacterial selection) and maintenance of the symbiotic luminescent bacterium Vibrio fischeri (V. fischeri). However, the mechanisms controlling bacterial selection and maintenance during bacterial colonization of the ANG are open biological issues with physiological significance. Our recent study on bigfin reef squid (Sepioteuthis lessoniana) already showed that the ANG bacterial community shifts gradually and exhibits decreased diversity throughout maturation. This study further describes a potential role of an innate immunity-involved molecule, peptidoglycan recognition proteins (PGRPs), in the ANG of bigfin reef squid during bacterial transmission and colonization. First, we found that four homologs of the PGRP family are expressed in the ANG of bigfin reef squid (slPGRP2-5), but only slPgrp2 transcript levels are highly correlated with ANG development and bacterial colonization. Besides, slPgrp2 transcripts are mainly expressed in the epithelial cells of certain secondary tubules of ANG, and the expression levels are varied in the epithelial cells of other secondary tubules. This data reveals that slPgrp2 transcripts may associate with the composition of bacterial consortium and its secretary factors. Moreover, recombinant slPGRP2 had a negative effect of Escherichia coli (E.coli) which inhibited bacterial growth in culture. Therefore, our data suggest slPgrp2 expression in the epithelial cells of secondary tubules in the ANG may have an essential role in the winnowing and maintenance of holobiont homeostasis in bigfin reef squid.

Thorium (Th) isotopes were applied to quantify the contributions of lithogenic inputs to the Qinghai Lake (QHH). Concentrations of dissolved 232Th and 230Th were measured in 59 water samples collected from Qinghai Lake and its exogenous recharge rivers. There are significant differences in the concentration of 232Th of the sampled water in QHH that confirm the input of variable lithogenic material sources. The 230Th concentrations were used to calculate a scavenging residence time for Th, which was then applied to calculate the flux of dissolved 232Th by matching the measured concentrations of dissolved 232Th. Then the 232Th content of lithogenic material was used with the solubility of Th from the preliminary particle data from the Qinghai–Qaidam district. When using a Th solubility from particles of 1%, the fluxes of lithogenic material range from 0.03 to 25.25 g/m2/yr in the surface water, consistent with the flux results of settled particles from the previous study. When a large number of exogenous recharge rivers are mixed into the northwest basin of Qinghai Lake, the 232Th content and lithogenic flux of the lake water are mainly influenced by the type and content of the particles in the Buha and Shaliu rivers. Conversely, in south basin with limited recharging rivers, the 232Th content of the lake water away from the estuary is mainly influenced by atmospheric dust. Furthermore, based on the 230Th normalization method (combining with 232Th and τTh), the Buha and Shaliu rivers located in the northwest basin contribute about 90% of the detrital flux to the lake. The lithogenic flux in the southeast lake is dominated by dust flux with a value of ∼0.109 g/m2/yr, while the higher lithogenic flux at the bottom of the lake was likely generated by accumulated sinking particulate matter and resuspension of bottom sediments in September. This study confirms the utility of long-lived Th isotopes to quantify lithogenic inputs based on the Th content of the dissolved lake water and also supply deposition resolution information for QHH sediment records with some certainty.

Abstract Hydraulic fracture is widely performed to enhance oil and gas production in the development of unconventional reservoirs. This paper proposes a numerical model for the calculation of the unpropped fracture conductivity of shale. This numerical model consists of three basic models, namely, the numerical reconstruction model of fracture morphology, fracture deformation model under closed stress, and fluid flow model. The numerical model can be used to analyze the influencing factors of the unpropped fracture conductivity of shale quantitatively. The results demonstrate that the fracture misalignment of shale is a prerequisite for unpropped fractures with conductivity at high closed stress. Furthermore, the unpropped fracture conductivity of shale decreases exponentially with the increase in closed stress. Moreover, closed stress is the factor having the most significant influence on the unpropped fracture conductivity of shale, followed by roughness and fracture morphology. The negative effect of closed stress can be reduced by increasing the fracture morphology during the fracturing operation.

Abstract Digital nucleic acid amplification tests enable absolute quantification of nucleic acids, but the generation of uniform compartments and reading of the fluorescence requires specialized instruments that are costly, limiting their widespread applications. Here, the authors report deep learning‐enabled polydisperse emulsion‐based digital loop‐mediated isothermal amplification (deep‐dLAMP) for label‐free, low‐cost nucleic acid quantification. deep‐dLAMP performs LAMP reaction in polydisperse emulsions and uses a deep learning algorithm to segment and determine the occupancy status of each emulsion in images based on precipitated byproducts. The volume and occupancy data of the emulsions are then used to infer the nucleic acid concentration based on the Poisson distribution. deep‐dLAMP can accurately predict the sizes and occupancy status of each emulsion and provide accurate measurements of nucleic acid concentrations with a limit of detection of 5.6 copies µl‐1 and a dynamic range of 37.2 to 11000 copies µl‐1. In addition, deep‐dLAMP shows robust performance under various parameters, such as the vortexing time and image qualities. Leveraging the state‐of‐the‐art deep learning models, deep‐dLAMP represents a significant advancement in digital nucleic acid tests by significantly reducing the instrument cost. We envision deep‐dLAMP would be readily adopted by biomedical laboratories and be developed into a point‐of‐care digital nucleic acid test system.

The dissolved uranium (U) content in the water column of saline lakes varies little between ice-free seasons throughout the whole water column. Such uniformity allows for the potential absolute dating and/or paleohydrologic interpretations of lake sediments and biogenic shell materials using U isotopes. Before using these methods in cold regions, however, it is necessary to evaluate the effects that ice freeze-thaw processes have on the distribution of U isotopes in saline lake waters, and to determine the amount of variation in U isotopic values when such processes occur. In this paper, we collected ice and dissolved water samples from six lakes with variable salinity in February 2021. Five groundwater and three water samples from rivers into Qinghai Lake were sampled in November 2020. The sampled water was analyzed for dissolved concentrations of 238U and the activity ratio of 234U/238U ([234U/238U]AR). The results show that the 238U concentration of ice samples was less than that of the underlying water. The [234U/238U]AR of ice in the five saline lakes was similar to that of the underlying water with less than a 10‰ variation, suggesting no observable fractionation between ice and dissolved water. Thus, the ice freeze-thaw processes have almost no effect on the uranium content and [234U/238U]AR of the sampled saline lakes, which were characterized by a limited recharge volume from surface runoff, groundwater, and ice volume, namely the close saline lake in arid alpine background. The results from the indoor freeze-thaw experiments also showed that the U isotopic composition of Qinghai Lake waters and ice were similar with the 238U concentration of the ice was about 40% of that of the dissolved lake water, supporting the data obtained from natural saline lakes. The above results provide important insights into whether it is feasible to use U isotopes for absolute dating and/or paleohydrologic analysis of lake sediments or biogenic shell materials. In addition, the results are important for evaluating the [234U/238U]AR and uranium concentrations in seawater when there exists a process of melting polar ice, and for determining the initial delta 234U variations needed for dating of coral and other fossil materials.

Experience-dependent plasticity is a key feature of brain synapses for which neuronal N-Methyl-D-Aspartate receptors (NMDARs) play a major role, from developmental circuit refinement to learning and memory. Astrocytes also express NMDARs, although their exact function has remained controversial. Here, we identify in mouse hippocampus, a circuit function for GluN2C NMDAR, a subtype highly expressed in astrocytes, in layer-specific tuning of synaptic strengths in CA1 pyramidal neurons. Interfering with astrocyte NMDAR or GluN2C NMDAR activity reduces the range of presynaptic strength distribution specifically in the stratum radiatum inputs without an appreciable change in the mean presynaptic strength. Mathematical modeling shows that narrowing of the width of presynaptic release probability distribution compromises the expression of long-term synaptic plasticity. Our findings suggest a novel feedback signaling system that uses astrocyte GluN2C NMDARs to adjust basal synaptic weight distribution of Schaffer collateral inputs, which in turn impacts computations performed by the CA1 pyramidal neuron.

BackgroundAs advocated by WHO in "Closing the Health Gap in a Generation", dramatic differences in child health are closely linked to degrees of social disadvantage, both within and between communities. Nevertheless, research has not examined whether child health inequalities include, but are not confined to, worse acute respiratory infection (ARI) symptoms among the socioeconomic disadvantaged in Pakistan. In addition to such disadvantages as the child's gender, maternal education, and household poverty, the present study also examined the linkages between the community environment and ARI symptoms among Pakistan children under five. Furthermore, we have assessed gender contingencies related to the aforementioned associations.MethodsUsing data from the nationally representative 2017-2018 Pakistan Demographic and Health Survey, a total of 11,908 surviving preschool age children (0-59 months old) living in 561 communities were analyzed. We employed two-level multilevel logistic regressions to model the relationship between ARI symptoms and individual-level and community-level social factors.ResultsThe social factors at individual and community levels were found to be significantly associated with an increased risk of the child suffering from ARI symptoms. A particularly higher risk was observed among girls who resided in urban areas (AOR = 1.42; pDiscussionsOur results underscore the need for socioeconomic interventions in Pakistan that are targeted at densely populated households and communities within urban areas, with a particular emphasis on out-migration, in order to improve unequal economic underdevelopment. This could be done by targeting improvements in socio-economic structures, including maternal education.

The aim of this study was to investigate whether measures of regional adiposity-including subcutaneous adipose tissue index (SATI) and visceral adipose tissue index (VATI)-can be associated with overall survival (OS) in Taiwanese patients with bone metastases. METHODS:This is a retrospective analysis of prospectively collected data. We examined 1280 patients with bone metastases who had undergone radiotherapy (RT) between March 2005 and August 2013. ProblemTo investigate the relation of inflammation-related parameters and pregnancy outcome in women with the early threatened abortion.Method of study630 women with early threatened abortion were divided into two groups based on the pregnancy outcome. All of them had the blood routine examination before treating. The differences between two groups were analyzed by the Chi-squared test, Student T test, Mann-Whitney U test, Binary Logistic Regression, Marginal Structural Model and Threshold effect analysis.ResultsWe found that there is no significant difference in the pregnancy outcome for NLR (OR:0.92, CI95%:0.72, 1.17) and PLR (OR:1.00, CI%:0.99, 1.01). However, a difference had a statistical significance in the pregnancy outcome when LMR less than 2.19 (OR:0.39, CI95%:0.19,0.82).ConclusionsThis study suggested that higher LMR was related to the lower risk of miscarriage in the women with early threatened abortion in a way.

Abstract The membrane potentials of cortical neurons in vivo exhibit spontaneous fluctuations between a depolarized UP state and a resting DOWN state during the slow-wave sleeps or in the resting states. This oscillatory activity is believed to engage in memory consolidation although the underlying mechanisms remain unknown. Recently, it has been shown that UP-DOWN state transitions exhibit significantly different temporal profiles in different cortical regions, presumably reflecting differences in the underlying network structure. Here, we studied in computational models whether and how the connection configurations of cortical circuits determine the macroscopic network behavior during the slow-wave oscillation. Inspired by cortical neurobiology, we modeled three types of synaptic weight distributions, namely, log-normal, sparse log-normal and sparse Gaussian. Both analytic and numerical results suggest that a larger variance of weight distribution results in a larger chance of having significantly prolonged UP states. However, the different weight distributions only produce similar macroscopic behavior. We further confirmed that prolonged UP states enrich the variety of cell assemblies activated during these states. Our results suggest the role of persistent UP states for the prolonged repetition of a selected set of cell assemblies during memory consolidation.

The inflammatory process is proposed to be one of the factors to benign prostatic enlargement (BPH), and this is the first study examining the anti-inflammatory ability of phloretin in treating rats with testosterone-induced BPH. BPH would be induced by testosterone (10 mg/kg/day testosterone subcutaneously for 28 days), and the other groups of rats were treated with phloretin 50 mg/kg/day or 100 mg/kg/day orally (phr50 or phr100 group) after induction. Prostate weight and prostate weight to body weight ratio were significantly reduced in the Phr100 group. Reduced dihydrotestosterone without interfering with 5α-reductase was observed in the phr100 group. In inflammatory proteins, reduced IL-6, IL-8, IL-17, NF-κB, and COX-2 were seen in the phr100 group. In reactive oxygen species, malondialdehyde was reduced, and superoxide dismutase and glutathione peroxidase were elevated in the phr100 group. In apoptotic assessment, elevated cleaved caspase-3 was observed in rats of the phr100 group. Enhanced pro-apoptotic Bax and reduced anti-apoptotic Bc1-2 could be seen in the phr100 group. In histological stains, markedly decreased glandular hyperplasia and proliferative cell nuclear antigen were observed with reduced expression in the phr100 group. Meanwhile, positive cells of terminal deoxynucleotidyl transferase dUTP nick end labeling were increased in the phr100 group. In conclusion, the treatment of phloretin 100 mg/kg/day could ameliorate testosterone-induced BPH.

Abstract The role that serum uric acid (UA) plays in the pathophysiological development of erectile dysfunction (ED) is controversial. We aimed to screen the factors related with ED, and to examine the association between serum UA and ED. Our data were derived from a cross-sectional Survey on Prevalence in East China for Metabolic Diseases and Risk Factors study in 2014–2015. Questionnaire of International Index of Erectile Dysfunction-5 was used for assessment of ED. Data were collected in three general communities respectively. A total of 1365 men were enrolled with an overall mean age 55.5 ± 10.8 years (range: 20–83 years). The prevalence of ED was 62.4% (51.4% standardized) in the population. Males with ED were older, and more prone to have a higher follicle-stimulating hormone, luteinizing hormone, sex hormone-binding globulin, glycated hemoglobin, fasting plasma glucose levels and lower free androgen index (FAI), UA levels, and more likely to have diabetes and elevated blood pressure compared with those without ED. Age and UA were independent influencing factors for ED. Besides, UA was positively correlated with FAI after adjustment for age. In conclusion, our study demonstrated the protective role that UA might play in development of ED.
