Background: The association between age at fist calving (AFC) and production, fertility and survivability in Jersey heifers requires evaluation and comparison with published literature on Holstein heifers., Method: Lactation records from 7256 pedigree Jersey heifers were grouped by AFC. Analysis comprised mixed effects multivariable regression modelling and binary logistic regression modelling., Results: Increased lifetime daily yield (LDY) was significantly associated with a reduced AFC; calving UK Jersey heifers at 24 months or below is optimal. Heifers calving at 25-26 months had a significantly lower mean calving interval (391 days; 95 per cent CI 386 to 397) than those calving over 30 months (399; 95 per cent CI 394 to 405) (P=0.03). Heifers with an AFC over 30 months were less likely to survive to second lactation than those with an AFC 18-24 months. Spring calving heifers produced significantly less milk per day (9.87 kg; 95 per cent CI 9.64 to 10.1) than autumn (10.18; 95 per cent CI 9.94 to 10.41) or winter calvers (10.07; 95 per cent CI 9.84 to 10.3) (P≤0.01). Lactating cows and nulliparous heifers are in distinctive and unique physiological conditions when they are approaching the planned time of breeding, at approximately 60 days in milk and 13-15 months of age, respectively. This study aimed to profile the metabolic milieu in heifers (N = 14) and lactating cows (N = 15) in the weeks leading up to planned time of breeding. All cows were followed for a period of 15 weeks, from 3 weeks pre-calving to 12 weeks post-calving, while heifers were monitored for a period of 4 weeks leading up to the tentative week of breeding (pre-breeding period). For data analysis, we further divided cows into primiparous (N = 8) and multiparous (N = 7) cows owing to the significant difference in their milk yield. Assessment of reproductive performance showed that primiparous and multiparous cows tended to have lower pregnancy rates compared to heifers (P < 0.1). Plasma concentrations of β-hydroxybutyric acid were about 2-fold higher in multiparous cows than those of heifers in the week leading up to planned time of breeding (P < 0.05). Total bile acid levels during the pre-breeding period were higher in all lactating cows compared to heifers (P < 0.05) and glucose levels were lower in lactating cows (P < 0.05). Triglyceride concentrations were lowest in multiparous cows compared to both primiparous cows and nulliparous heifers (P < 0.05). In addition, lactating cows had higher concentrations of total-cholesterol and the high-density lipoprotein and low-density lipoprotein compared to heifers (P < 0.05). Conversely, concentrations of very low-density lipoprotein were lower in multiparous cows than primiparous cows and nulliparous heifers (P < 0.05). There were no differences in plasma glutathione levels, as measured by liquid chromatography-tandem mass spectrometry, between the groups, but the ferric reducing ability of plasma was higher in lactating cows compared to heifers (P < 0.05). These data establish the differences in the profile of metabolic and oxidative markers during the period approaching planned time of breeding in lactating cows compared to nulliparous heifers. As certain metabolites in the plasma have been shown to be represented in the ovarian follicular microenvironment, the unique profiles may influence reproductive performance in dairy cattle in different physiological stages., (Copyright © 2019. Age at puberty is a critical trait, because pregnancy success during the breeding season is correlated with the percentage of heifers that reach puberty before or early in the breeding season. A negative genetic correlation between age at puberty and heifer pregnancy rate indicate that selection to decrease age at puberty would increase heifer pregnancy rates. Calving late has been reported to increase the chance of calving late or not calving the following year, and heifers need to wean 3 to 5 calves to pay for development costs. In tropical regions, cattle raising faces a critical issue with performance deficits associated to nutrition, this parameter directly affects fertility. The present research had a quantitative-qualitative approach, studying the cause and effect reaction of injectable casein and calcium on productive and reproductive parameters in a batch of 82 crossbred zebu heifers. The batch was divided into treatment and control groups, both of them had identical sanitary plans, environmental conditions, and feeding management systems. Initially, an anthropometric technique was used, measuring weight (kg) before and after the study, as well as uterine diameter (mm). The results showed that the weights of the treatment group went from 277.46 ± 30.92 to 299.07 ± 29.51, and the control group from 274.98 ± 30.32 to 298.43 ± 28. In terms of uterine diameter, the treatment group went from 16.38 ± 3.3 to 20.5 ± 3.67, and the control group from 15.17 ± 3.04 to 20.54 ± 3.42. The presence of the corpus luteum was 12.20% in the treatment group and 4.88% in the control group. Background: Anti‐Müllerian hormone (AMH) holds potential as a biomarker for assessing the superovulation (SO) response in cattle. Nonetheless, there exists scant information regarding this aspect in the literature concerning dairy heifers. Given this gap, our objective is to explore the viability of AMH as an indicator for gauging the SO response specifically in Holstein heifers. Furthermore, our aim encompasses examining the variations in AMH levels within the same individuals before and after undergoing SO. Methods: The study included 41 Holstein heifers. All heifers were superovulated and blood samples were taken both before and after the SO protocol. Results: The findings revealed that the mean values of serum AMH concentrations before and after SO were 0.122 ng/mL (0.093–0.248 ng/mL) and 0.119 ng/mL (0.084–0.170 ng/mL), respectively. AMH concentrations in heifers were stratified into low (<0.106 ng/mL), medium (0.107–0.126 ng/mL) and high (>0.127 ng/mL) categories both before and after SO. Conclusions: There was no significant correlation between AMH levels in the heifers both before and after SO treatment with the number of follicles, corpora lutea, total embryos collected or embryos transferred (p > 0.05). Furthermore, this study showed that serum AMH concentrations in Holstein heifers did not change after SO treatment. In this study, as AMH levels in Holstein heifers were in a narrow range, a relationship between AMH and SO response could not be determined. In future studies, we believe that it would be more useful to plan more studies in Holstein donor heifers, taking into account the number of animals and AMH levels. [ABSTRACT FROM AUTHOR]

During Taurus' virtual anniversary event, the loss of pregnancy after IATF in breeding herds was analyzed. 300 attendees from different locations participated, including specialists in the field. Compiled information on early embryonic and fetal losses was presented, and the maximum acceptable value for these losses was discussed. Percentages of pregnancy loss in primiparous and multiparous cows, as well as in heifers, were mentioned based on the reviewed literature. Context. Fodder beet (FB) is a widely used winter feed in New Zealand's southern regions due to its high yield, consistent quality, and environmental benefits. However, FB is low in crude protein, phosphorus and fibre, and a survey showed that many dairy farmers were concerned that feeding FB may have negative impacts on lifetime performance. Aim. To determine whether winter feeding FB or kale (Ka) to dairy cows, and then their heifer offspring at 1 and 2 years old, affects the heifer performance from birth until the end of their first lactation. Methods. In June-August 2017, pregnant dairy cows were fed FB or kale over winter and then 140 of the resultant heifer offspring were divided into four treatment groups at 9-11 months old according to their dam's winter diet and offered either FB or kale, with pasture baleage. The groups were as follows (dam crop-heifer crop): FB-FB, FB-Ka, Ka-FB, Ka-Ka. The heifers' subsequent performance (growth, stature, milk production, and reproduction) was measured to the end of their first lactation (May 2020), and blood mineral status was monitored from May 2018 to May 2019. This observational study has its limitations, but is useful for generating hypotheses to test in more controlled research. Key results. Heifers grazing FB in winter (June-August) 2018 had lower blood plasma urea and phosphate concentrations than did heifers grazing kale, despite similar dietary crude protein contents. Treatments had no effect on milk production. However, the heifer liveweights were affected and there was an indication of poorer reproductive performance. The FB-FB heifer treatment group had a lower average liveweight than the FB-Ka heifers from 1 to 2.8 years old. Conclusions. Although no impact on milk production for the first lactation was observed, the key results suggest possible negative impacts of feeding heifers FB during winter, particularly if their dam also wintered on FB. Implications. The combined effects of FB dam and heifer winter diets on lifetime productivity warrant further investigation to help develop sustainable FB feeding systems. When feeding FB, we recommend that farmers test their crops and supplements to ensure that sufficient nutrition, in particular protein and phosphorus are provided. [ABSTRACT FROM AUTHOR]

Context. Broader applications of milk mid-infrared spectral data could add value to milk-recording data. One such application is to rank cows on the probability of conception to first service (MFERT) which could help prioritise cows for insemination with dairy sexed semen (SS). Aims. This study compared the use of MFERT estimates against two other approaches, to (1) identify most and least fertile dairy cows and (2) prioritise cows predicted to be most fertile for first service insemination with SS. Methods. Mid-infrared spectral data from first herd test after calving was used to generate 13 379 MFERT predictions for 76 cohorts. Reproduction records were used to calculate reproductive parameters, calf numbers and net benefit, i.e. calf values minus mating costs, for two breeding programs. Breeding program 1 used SS and conventional dairy semen, while Breeding program 2 used SS, conventional dairy and beef semen. Three semen-allocation approaches were compared, namely, allocation via MFERT, calving date (CDATE) or assignment via random number generator (RANDOM). Key results. MFERT significantly outperformed (1) RANDOM in identifying cows most and least likely to calf after first insemination (P < 0.05), and (2) both CDATE and RANDOM in identifying cows most and least likely to calf overall (P < 0.05). This resulted in up to 1.5 and 4.5 more dairy heifer calves, in Breeding programs 1 and 2 respectively, and up to six fewer dairy-beef calves in Breeding program 2. Differences in net benefit among semen-allocation approaches were modest, although generally favoured MFERT. Few significant differences between MFERT and CDATE were found. However, significant net benefit differences among all three semen-allocation approaches were seen in Breeding program 2. Conclusions. MFERT outperformed CDATE and RANDOM in identifying most and least fertile cows. Realised net benefits of semen allocation by MFERT over other approaches were modest. Given the impact of semen type and dairy-beef calf prices value proposition will vary. Implications. Our study confirmed that MFERT can add value to milk recording data by identifying the most and least fertile cows. Successful reproduction of lactating dairy cattle, or in other words its reproductive performance is a limiting factor in the profit ratio of cattle breeding regarding market production. The aim of this research was to evaluate the success rate of synchronization protocols Ovsynch, Presynch/Ovsynch, and Double Ovsynch within the period of 24 months. The success rate of the 1st insemination, 2nd insemination, and after more than 2 inseminations was evaluated. The cattle were of Holstein Friesian breed with a milk yield of 10 200 kg. The animals were sorted into two categories – heifers (protocol Ovsynch) and primiparous, multiparous cows (protocols Presynch/Double Ovsynch). The research proved a 54 % success rate in the category of heifers after the 1st insemination. In the category of primiparous and multiparous cows was the success rate of 41 % after the 1st insemination, 39 % after the 2nd insemination, and 52 % after more than 2 inseminations for the Presynch/Ovsynch protocol. The success rate for Double Ovsynch protocol was 45 % after the 1st insemination, 42 % after the 2nd insemination and 51 % after more than 2 inseminations. The results for the given geographic region of Slovakia exceeded the average.

The objectives of this study were to investigate the effect of presynchronization or dose of GnRH on ovulatory response to initial GnRH, estrus expression and reproductive outcomes in Holstein heifers subjected to a progesterone based 5-d CO-Synch protocol. Heifers were fitted with a collar-mounted automated activity monitoring system on Day −5 and were assigned randomly to one of three groups: 1) G100 (n = 111), a 5-d CO-Synch plus PRID protocol with administration of 100 μg of GnRH at the time of PRID insertion (Day 0), 500 μg of cloprostenol (PGF) at the time of PRID removal (Day 5) and again 24 h later (Day 6); 2) G200 (n = 112), same as G100 except 200 μg GnRH was administered on Day 0; 3) P10 (n = 111), PRID + PGF on Day −5 followed by a 5 d CO-Synch with administration of 100 μg of GnRH on Day 0, PGF at the time of PRID removal (Day 5) and again 24 h later (Day 6). Approximately 72 h after PRID removal (Day 8), all heifers were timed-inseminated (TAI) and concurrently 100 μg of GnRH was administered to those not exhibiting estrus. All inseminations were done by one technician using either sex-sorted (n = 265) or conventional (n = 69) frozen-thawed semen. Transrectal ultrasonography was done to determine ovarian cyclicity and normalcy of the reproductive tract, ovarian dynamics and pregnancy. All heifers were cyclic and ovulatory response to initial GnRH was greater (P < 0.01) in G200 (51.8%) and P10 (47.7%) compared to G100 (27.9%). Estrus expression rate tended to be greater (P = 0.08) in G100 (93.7%) compared to G200 (85.7%) and intermediate in P10 (89.2%). Expression of estrus was associated positively with P/AI at 45 d post-TAI (P < 0.01; 70.2 vs. 31.4% for heifers expressing or not expressing estrus). Heifers in the G200 group had greater P/AI at 30 (P < 0.01) and 45 (P = 0.01) d post-TAI (79.5 and 75.9%, respectively) compared to those in G100 (63.1 and 60.4%) or P10 (64.0 and 62.2%) groups. Pregnancy loss did not differ among treatment groups (overall 3.9%). Results indicate that increasing the dose of the initial GnRH from 100 to 200 μg resulted in increased ovulatory response and improved P/AI in Holstein heifers subjected to a progesterone based 5-d CO-Synch protocol. Although heifers presynchronized with a combination of PRID and PGF had an increased ovulatory response to the initial GnRH, P/AI did not differ from the standard progesterone based 5-d CO-Synch protocol. • Colazo et al. Context: The performance of commercial beef-breeding herds in northern Australia is highly variable. Identifying and understanding the major factors that influence this is critical in determining which management interventions should be implemented to best manage these factors. Aims: This study aimed to describe the occurrence and magnitude of the risk factors identified as being strongly associated with one or more measures of cow performance in northern Australia. Methods: A prospective epidemiological study was conducted in a cross-section of commercial beef-breeding herds in northern Australia, to determine and quantify the major associations of up to 83 candidate herd management, nutritional and environmental risk factors with cow performance. Descriptive analyses of significant risk factors were conducted. Key results: Unfavourable levels of risk factors were observed for all country types and across three cow-age groups. However, generally, adverse property-level nutritional, environmental and management risk factors had a higher incidence in the Northern Forest, which was associated with significantly lower performance of heifers and cows. This was reflected in generally lower body condition of heifers and cows in this country type. Although the performance of heifers and cows was generally higher in the Southern Forest, the irregular incidence of adverse risk factors contributed to the observed quite variable performance. Conclusions: The factors significantly affecting the performance of cows in the major beef-breeding regions of northern Australia are described. These factors were additively more adverse in the Northern Forest. Implications: In this study, the necessary understanding of the factors most likely to be affecting the performance beef cows in this environment has been described. This is required to make appropriate decisions about management interventions to control these factors. This experiment investigated the effects of diet composition on rumen, vaginal, and uterine microbiota of beef heifers. Fifteen rumen-cannulated, pubertal Angus-influenced heifers were used in a replicated 3 × 3 Latin square design (28-d periods and 21-d washout intervals). Dietary treatments included diets based on (as-fed) 100% grass hay (HF), 60% grass hay + 40% corn-based concentrate (INT), or 25% grass hay + 75% corn-based concentrate (HG). Treatments were offered individually to heifers once daily at 2% body weight. Rumen, vaginal, and uterine samples were collected on days 0 and 28 of each period. Data were analyzed using orthogonal contrasts (linear and quadratic), using results from day 0 as independent covariates and heifer as the experimental unit. Ruminal pH on day 28 decreased linearly (P < 0.01) as concentrate inclusion increased. Uterine and vaginal pH on day 28 were not affected by treatments (P ≥ 0.35). Within the rumen samples, Bacteriodetes was the most abundant phylum and its relative abundance linearly decreased (P ≤ 0.01) with the inclusion of concentrate. Prevotella was the most abundant genus within the rumen but was not affected by treatments (P ≥ 0.44). Genera with relative abundance ≥1% (average across treatments) in the rumen that were impacted by treatments (P ≤ 0.01) included Bacteroides , Pedobacter , Dysgonomonas , Caloramator, and Ruminococcus. Firmicutes was the most abundant phylum in the vagina and uterus, but it was unaffected by treatments (P ≥ 0.16). Prevotella was the most abundant genus in the vagina, and its relative abundance increased (P < 0.01) with the inclusion of concentrate. Other genera with relative abundance ≥1% that were significantly affected (P ≤ 0.05) by treatments were Clostridium, Pedobacter , Roseburia, Oscillospira, Faecalibacterium, Caloramator, Paludibacter, Rhodothermus, and Porphyromonas. In uterine samples , Prevotella was the most abundant genus but was unaffected by treatments (P ≥ 0.29). Genera with relative abundance ≥1% in the uterus that were significantly affected (P < 0.01) by treatments were Caloramator , Paludibacter , and Thalassospira. Collectively, inclusion of concentrate in the diet altered the bacterial composition within the rumen as well as shifting bacterial populations within the vagina and uterus. Objective: Pregnant Nelore heifers (n = 417) were used to evaluate the effects of supplementation with monensin-molasses multinutrient block (B) during pre and/or postpartum on reproductive and progeny performance. Methods: Heifers were allocated in four treatments: i) CC: heifers received control supplement (C) in loose meal form (0.06% of body weight [BW] offered daily before and after parturition; n = 108); ii) CB: received C before parturition and B (0.07% of BW offered weekly after parturition; n = 117); iii) BC: received B before and C after parturition (n = 103) and iv) BB: received B before and after parturition (n = 89). During pre and postpartum periods, concentration of metabolites/hormones and cow/calf performance was evaluated over time. Cows were synchronized twice for fixed timed artificial insemination (FTAI) using an estradiol/progesterone-based protocol. Data was analyzed by orthogonal contrasts (C). Results: B increased pregnancy at first FTAI (p = 0.04) and overall pregnancy rate (C1: CC vs BB+BC+CB; p = 0.05). Supplemented cows had greater body condition score (BCS) only at parturition (D0; p = 0.04) and at D40 (p = 0.02). B increased BW (p = 0.03), glucose concentrations (p = 0.01) and subcutaneous fat thickness (p = 0.03) only at D40. Concentrations of insulin were higher in supplemented cows (p = 0.008). Calves born by cows supplemented before and after parturition (C2: BB vs BC+CB) were heavier at 80 (p<0.001), 120 (p<0.001), 170 (p = 0.002) and 210 (p = 0.02) days old. Conclusion: Regardless of period of treatment, block supplementation increased pregnancy at first FTAI and overall pregnancy rate. Additionality, block supplementation during both pre and postpartum periods improved progeny weight until weaning. A retrospective analysis of 5 studies (2011 to 2022) evaluated the effects of body weight and synchronization protocols before the start of breeding season on reproduction of Bos indicus-influenced beef heifers. Data were collected from 594 Brangus crossbred heifers weaned on average at 280 ± 26 d of age, allocated to bahiagrass pastures (4 to 5 heifers and 0.8 to 1.0 ha/pasture), and supplemented with soybean hulls-based concentrate (0.5 to 1.8% of body weight, (BW); dry matter basis] from 2 weeks post-weaning until the end of the breeding season (189 ± 24 days of supplementation). Heifers were initially sorted into those that were supplement (S) and were not supplemented (NS) assigned to an estrous synchronization protocol before the start of the breeding season. Then, within each synchronization group, heifers were sorted into those that were below (BE) or above (AB) the average BW of all heifers (300 kg) at the start of the breeding season (n = 130, 126, 146, and 192 for BE+NS, BE+S, AB+NS, and AB+S heifers, respectively). Data were analyzed using GLIMMIX procedure of SAS using synchronization, BW at breeding, and resulting interaction as fixed effects, and heifer (synchronization x BW), study, and pasture(study) as random effects. Effects of synchronization x BW at breeding and synchronization were not detected for heifer BW and overall average daily gain (ADG). However, AB heifers were always (P < 0.0001) heavier from post-weaning until the end of the breeding season and had (P < 0.0001) greater overall ADG than BE heifers. Effects of synchronization x BW at breeding tended (P = 0.07) to detected for puberty attainment at the start of the breeding season, which was least (P < 0.01) for BE+NS, did not differ between BE+S vs. AB+NS (P = 0.37), and was greatest for AB+S heifers (31, 66, 60, and 81 ± 7.4%, respectively). However, final pregnancy and calving percentages differed (P < 0.01) only for BE vs. AB heifers (68 vs. 81 ± 4.4% and 63 vs. 75 ± 5.2%, respectively). Effects of synchronization x BW at breeding tended (P = 0.10) to detected for percent of heifers calving within the first 30 days of the calving season, which was least (P < 0.03) for BE+NS and did not differ (P = 0.20) among BE+S, AB+NS, and AB+S heifers (48, 69, 61, and 64 ± 5.9%, respectively). Hence, in Bos indicus-influenced beef heifers, BW above 300 kg at the start of the breeding season led to improved overall reproductive performance compared to BW < 300 kg, whereas estrous synchronization protocols had positive effects to puberty attainment (regardless of heifer BW) and improved early calving distribution only when heifers were below 300 kg at the start of the breeding season. Summary: Lipopolysaccharide (LPS) endotoxemia has been negatively associated with fertility. This study aimed to investigate the effect of LPS-induced inflammation on gene expression associated with bovine fertility in the uterus and oviduct. Sixteen healthy heifers were divided into two groups. The LPS group (n = 8) received two intravenous (i.v.) injections of 0.5 µg/kg of body weight of LPS with a 24-h interval, and the control group (n = 8) received two i.v. injections of saline solution with the same interval of time. All the animals had the follicular wave synchronized. Three days after the second injection of LPS, all animals were slaughtered and uterine and oviduct samples were collected. Gene expression associated with inflammatory response, thermal and oxidative stresses, oviduct environment quality, and uterine environment quality was evaluated. Body temperature and leucogram demonstrated that LPS induced an acute systemic inflammatory response. In the uterus, the expression of PTGS2 and NANOG genes was downregulated by the LPS challenge. However, no change in expression was observed in the other evaluated genes in the uterus, nor those evaluated in the oviduct. In conclusion, the inflammatory process triggered by LPS did not persist in the uterus and oviduct 3 days after challenge with LPS. Nonetheless, reduction in PTGS2 and NANOG expression in the uterus suggested that, indirectly, LPS may have a prolonged effect, which may affect corpus luteum and endometrial functions. This observational study aimed to determine the effect of genetic merit for fertility traits on estrous expression and estrous cycle duration in grazing dairy cows, as measured by an activity monitoring device. A secondary aim was to describe changes in expression of estrus that occur during successive estrous cycles postpartum. Neck-mounted, activity-monitoring devices (Heatime, SCR Engineers Ltd.) were fitted to nulliparous Holstein-Friesian heifers with positive (POS FertBV) or negative genetic merit for fertility traits (NEG FertBV) to capture activity data during their first and second lactations (POS FertBV: n = 242, n = 188; NEG FertBV: n = 159, n = 87 in lactation 1 and 2, respectively). An estrous event was identified when the activity change index exceeded 26 activity units (AU) for 4 h. A total of 1,254 and 892 estrous events were identified in lactation 1 and 2, respectively. Estrous duration was defined as the interval between when the threshold was first exceeded and when activity dropped below the threshold, with no new event starting within 24 h of the end of the previous event. This definition of estrus included cows in which activity crossed the threshold multiple times in a day and were classified as a single estrous event. A second measure, high activity duration, was defined as the total hours that activity exceeded the threshold. To characterize estrous activity, peak activity (above baseline) and total activity (area under the curve of activity above baseline) were measured. Compared with NEG FertBV cows, POS FertBV cows had more active, longer estrous events. In lactation 1, the POS FertBV group had a mean estrous duration and a high activity duration of 12.5 and 12.4 h compared with 11.4 and 11.3 h for the NEG FertBV group [standard error of the difference (SED) = 0.5 and 0.4 h, respectively]. This significant difference also occurred in lactation 2, with a mean estrous duration of 13.1 versus 11.8 h (SED = 0.5 h) and a high activity duration of 13.0 versus 11.8 h (SED = 0.4 h) in the POS and NEG FertBV groups, respectively. Total activity and peak activity were greater in the POS compared with the NEG FertBV group in lactation 1 (peak activity: 65.5 vs. 55.8 AU, SED = 2.4 AU; total activity: 588 vs. 494 AU, SED = 25 AU) and lactation 2 (peak activity: 72.5 vs. 61.2 AU, SED = 2.9 AU; total activity: 648 vs. 541 AU, SED = 30 AU). Estrous cycle duration did not differ between the POS and NEG FertBV groups (lactation 1: 20.4 vs. 20.6 d, SED = 0.25; lactation 2: 20.8 vs. 21.0 d, SED = 0.28). Less estrous activity of the cow was associated with the first postpartum estrus. In contrast, the number of previous estrous events did not consistently affect the duration of the subsequent estrous cycle. The outcomes of this study provide evidence that positive genetic merit for fertility traits is associated with more overt estrous expression. Post-operative pain and inflammation are normal physiological reactions to caesarean section. Their management in cattle have rarely been investigated. This surgical procedure negatively affects reproductive function with, for example, a reduction in fertility resulting in an increase in calving interval. In this multicenter clinical trial, the objective was to evaluate the impact on reproductive performance of meloxicam injected before caesarean section to manage post-operative pain and inflammation. Meloxicam is a non-steroidal anti-inflammatory drug. One hundred and twenty-seven Charolais heifers (n = 127) were recruited from 47 farms in six French veterinary practices in the Burgundy region. The heifers underwent a non-elective standardized caesarean section operation. Heifers were randomly assigned to one of two groups: meloxicam (n = 66), intravenous meloxicam injection before surgery, or control (n = 61). Reproductive performance and health information were recorded from the time of the caesarean section to the next calving or to culling. In our study, meloxicam administration before caesarean section had no effect on the incidence of retained placenta (18.2% of treated vs 25.0% of control cows, p = 0.35). The pregnancy rate was higher in treated than in control cows (83.1% vs 67.8%, p = 0.04 after multivariate analysis) and a survival analysis showed that the median calving interval was 35 days shorter in the meloxicam (t 50% = 417 days) compared to the control group (t 50% = 452 days, p = 0.05). A trend was also observed for culling rate to be lower in treated (4.7%) compared to control cows (13.3%, p = 0.09). Polymorphisms in μ-calpain (CAPN1) that beneficially associate with beef tenderness are reported to antagonistically associate with calving day in beef heifers and post-partum interval to estrus in beef cows. We, therefore, hypothesized that a molecular breeding value for slice shear force, calculated based on CAPN1 and calpastatin (CAST) genotypes, would demonstrate an antagonistic relationship between genomically predicted slice shear force and ordinal calving date in replacement beef heifers. A secondary objective of this study was to evaluate the association of a polymorphism in diacylglycerol O-acyltransferase (DGAT1) with reproductive traits in beef heifers. One hundred eighty-seven MARC III heifers (¼ Angus, ¼ Hereford, ¼ Red Poll, and ¼ Pinzgauer) that had been selectively bred to increase the frequency of these polymorphisms were submitted for monthly ultrasound exams beginning at 333 d of age and continuing until the start of breeding to determine pubertal status. At the last exam before breeding, all antral follicles were counted, and the length and height of each ovary was measured to determine if genomic selection for slice shear force associated with ovarian follicle number. Calving date, calf gender, and calf birth weight were recorded at parturition. Regression analysis of the molecular breeding value for slice shear force of the heifers on ordinal calving date indicated no association between genomic prediction of tenderness and calving date (P = 0.16); however, there was a tendency for age at puberty to be delayed in heifers as genetic merit for tenderness improved (P = 0.09). The results of the present study indicate that within experimental precision, selecting for tenderness using genomic predictions had minimal or no antagonistic association with reproductive performance in heifers. Context: Heat stress has led to a serious reduction in dairy cows production performance, thus increasing the stress of feeding and reproduction management. Aims: Heat stress arises when cows are unable to dissipate excess body heat, we aimed to investigate the effects of ambient temperature (AT) and humidity on diurnal body temperature and activity. Methods: For improving the technology for rearing dairy cows, the vaginal temperature (VT) and activity of 60 Holstein heifers in summer (n = 20), autumn (n = 20), and winter (n = 20) were measured using the oestrus monitoring system. Key results: We found that VT fluctuated slightly (~38.22–38.32°C) when AT and temperature-humidity index (THI) were lower than 20°C and 68, respectively. However, when this threshold is reached, VT increased significantly with increasing AT and THI, whereas activity decreased significantly. Conclusions: Heat stress may be caused when THI is above 68 and cow's VT reaches 38.32°C. Evidently, when the THI exceeds 68 and VT is more than 38.32°C, suitable measures for reducing the effect of heat stress on the productivity of dairy cows should be taken. Implications: The combined monitoring of VT and THI might provide accurate guidance for preventing and controlling heat stress. The effect of incorporation of two different mineral mixtures and/or oak leaves was studied on nutrient utilization and reproductive performance in anestrous heifers. Twenty-one anestrous heifers (18.2 ± 1.45 months; 229 ± 14.2 kg body weight) were randomly distributed into three similar groups. Heifers in control (T1) and first treatment group (T2) were fed concentrate mixture incorporated with Bureau of Indian Standards (BIS) specific mineral mixture and a customized mineral mixture developed specially for Kumaon hills (MMKH), respectively, along with local green grass (Pennisetum orientale). In the second treatment group (T3), concentrate mixture was the same as that of T2, while the source of roughage was local oak (Quercus leucotricophora) leaves containing 3.35% condensed tannin. A digestibility trial was conducted after 120 days of study. The feed intake was similar among the groups. Digestibility coefficient of crude protein (CP) was lower in T3 than T2 and comparable to T1. Feeding oak leaves improved absorption of calcium as compared to grass-fed animals. Bioavailability of copper and zinc was higher (P < 0.05) in oak leaves and MMKH fed group (T3) as compared to T1, but similar to T2. Conversely, absorption of iron had the reverse trend and was reflected in serum Fe concentration. Hematological, biochemical, enzyme and hormonal profiles were not influenced by any of the treatments. The relative occurrence of estrus cyclicity and conception rate was more in groups T2 and T3, respectively, than other groups. It was concluded that feeding oak foliage-based diet containing 1.87% tannin along with customized mineral mixture developed for Kumaon hills improved certain nutrient utilization and reproductive performance as compared to local green grass supplemented with BIS-specific mineral mixture or MMKH. Sire selection is important to maintain the desired genetic capability of any cattle herd; therefore, selection of economic production traits affect profitability by increasing pre- and post-weaning gain and carcass quality potential. Using heifers developed from sires selected for growth and milk traits may have long-term impact on sustainability of the cowherd by influencing mature cow size and milk production. Our objective was to determine the impacts of sires selected for high and moderate growth and high and low milk traits on growth and reproductive performance of replacement heifers. Therefore, we selected replacement heifers sired by bulls within the upper 5th and 50th per-centiles of the Angus breed for yearling weight (YW) EPD as high growth (HG) moderate growth (MG), the upper 15th percentile of the Angus breed for milk EPD as high milk (HM), and the lower 15th percentile for low milk (LM) in a 2x2 factorial design resulting in 4 treatments: HG/HM, HG/LM, MG/HM, and MG/LM. Heifers (n = 46) from appropriate AI sires that were born and developed at Oklahoma State University -Range Cow Research Center - South Range Unit were selected as replacements. They were raised on a base diet of dry, dormant tall grass prairie and moderate quality grass hay following weaning and 2 kg/d of 16% crude protein concentrate supplement. Body weights and biometrics were recorded at weaning and approximately at 1-yr of age, and blood was collected at 11 and 0 d before synchronization for progesterone analysis to determine pubertal status pre-breeding. Estrus was synchronized using the 14-d CIDR-PG with timed AI method followed by a 45-d natural service breeding period. Pregnancy checks were conducted by serum collection 35-d following AI and via rectal palpation by an experienced veterinarian 112 d after removal of the bulls. Statistical analyses were performed using the mixed procedure of SAS (SAS 9.4, SAS INC., Carey, NC). Heifers from high growth sires were 28 kg heavier at weaning (P < 0.01) and 26 kg heavier as yearlings (P < 0.01) than those sired by moderate growth sires, while sire milk EPD affected neither (P ≥ 0.55). High growth and high milk sired heifers had greater hip heights, frame scores and thus greater (P ≤ 0.05) estimated mature body weight (EMBW). Despite this, sire selection had no impact on percentage of heifers pubertal before breeding, AI pregnancy, or the total pregnancy rates (P > 0.24) Based on these data, selection of sires for HG and HM leads to increased productivity of the offspring but gives rise to larger mature cows, which may pressure grazing resources in limited resource environments impacting the long-term sustainability of the cow-calf enterprise. The aim of the present study was to evaluate the effects of zearalenone (ZEA) on the reproductive system morphometry, oestrogen (E2) levels and oocyte quality of beef heifers. Twenty non‐pregnant purebred Nellore (Bos indicus) heifers [age, ≥18 months; initial body weight, 348 ± 30 kg (mean ± standard deviation)] were used. The animals were randomly divided into experimental group and a control group of 10 animals each. Group experimental was administered 300 ppb ZEA per os for 98 days, and the control group was administered placebo per os for 98 days. The administration of ZEA was carried out daily by adding mycotoxin to the diet. All heifers were evaluated weekly via rectal ultrasound examinations (12 weeks). Diameters of the right and left uterine horns, right and left ovaries, largest antral follicle and corpus luteum were measured. Vulva size was also measured. Blood samples were collected to estimate E2 levels. At the end of 12 weeks, the heifers were slaughtered, and the ovaries were sent to the laboratory for in vitro embryo production. A completely randomized design was adopted, and repeated measures analysis of variance (p <.05) was performed (except for oocyte quality). Vulva size (p =.0985); diameters of uterine horns (p =.0522), ovaries (p =.6955), antral follicles (p =.6355) and corpus luteum (p =.3808); and E2 levels (p =.3379) were not affected by the treatments. ZEA‐contaminated diet significantly reduced (p =.05) the proportion of viable oocytes (49.4%, n = 207) compared with the control diet (59.9%, n = 222); however, the blastocyst rate did not differ between the groups (p =.9418). The results indicate that contamination of beef heifer's diet with 300 ppb ZEA affected neither morphometric parameters nor plasma oestrogen levels; however, ZEA contamination was detrimental to oocyte quality. The aim of this study was to compare conformation, age at puberty and subsequent milk production of Holstein–Friesian–Jersey crossbred heifers grown in a linear trajectory (Target; n = 55) between 6- and 15-month target liveweights (LWT), with heifers grown in a seasonal manner (Seasonal; n = 55) of slow over first winter and fast over spring. Heifers that grew to Target attained puberty 38 days earlier than heifers that grew in a seasonal manner; however, no difference between treatments in the date of first calving occurred. Heifers in the seasonal treatment were 1 cm (P =.032) taller than those in the target treatment, but similar in girth and length. There was no effect of treatment on first lactation milk production. These results suggest that provided heifers reached their pre-mating target LWT, the growth trajectory between target LWT at 6 and 15 months of age did not negatively impact frame size or first lactation milk production. Interest is growing in methods to apply precision nutritional supplements strategically during the peri-conceptual period to enhance progeny performance of beef females. Studies where supplemental methionine (MET) was provided to bovine females in the peri-conceptual period demonstrated epigenetic modifications in muscle of the progeny. Guanidinoacetic acid (GAA) has been fed directly to livestock to increase creatine synthesis, but has not been evaluated for use in fetal programming. We fed MET and GAA to crossbred heifers during the peri-conceptual period as a 2 x 2 factorial design to influence carcass characteristics of their steer calves. It was, therefore, important to determine if the interactions of dietary MET and GAA in the peri-conceptual period would alter reproductive efficiency. The null hypothesis was that these supplements would not alter reproductive efficiency of beef heifers when fed in the peri-conceptual period. Heifers were divided into four breeding groups for management purposes. On day -63, heifers (n = 20/diet, age = 384.1 ± 1.1 d) were assigned HeatTime collars and individually fed MET (10 g/d), GAA (40 g/d), the combination (MET + GAA), or ground corn carrier (CON) for 126 days. On day -9, heifers received 100 mg of gonadotropin releasing hormone and a CIDR. On day -2, the CIDR was removed and luteolysis was initiated with 25 mg of prostaglandin F2a. All heifers were inseminated with male sexed semen from a single sire and administered a second 100 mg injection of gonadotropin releasing hormone on day 0. On day 63, a blood sample was collected from all pregnant heifers (CON n = 10; MET n = 8; GAA n = 7; MET + GAA n =10) to evaluate circulating progesterone concentrations. Reproductive tracts were collected to determine CL weight, gravid uterine weight, and placentome number. Data were analyzed using the MIXED procedure of SAS with group, MET, GAA, and the interaction of MET and GAA as fixed effects. Days to first estrus were greater and number of estrus events prior to synchronization were fewer in CON heifers compared with supplemented heifers (P < 0.05). Luteal weight tended to be greater in heifers supplemented with MET compared with CON and GAA heifers (P = 0.09), but peak intensity of estrus, concentration of progesterone at day 63 of pregnancy, gravid uterine weight, and placentome number did not differ due to diet (P > 0.10). Supplementation with MET or GAA may improve onset of puberty but does not negatively alter reproductive performance in crossbred beef heifers. The objectives were to investigate the effects of differing planes of pre- and postweaning nutrition on prepubertal serum leptin concentrations, LH profiles, and age at puberty in Holstein heifers. Thirty-six Holstein calves were randomly assigned to either a low (5 L) or high (10 L of whole milk/d) preweaning plane of nutrition from 1 to 7 wk of age, a transition diet (a dry total mixed ration with 85% of concentrate) from wk 8 to 10, and to either a low (70%) or high (85% of concentrate dry total mixed ration) postweaning plane from 11 to 25 wk of age. Serum leptin concentrations were measured every 2 wk from 1 to 25 wk of age, and LH profiles were determined both at wk 15 and 25 based on sequential blood samples taken every 12 min over 10 h. Starting at 26 wk of age, ovaries were examined weekly by transrectal ultrasonography until first ovulation (puberty) was confirmed. Heifers that received the high preweaning plane diet had greater mean (± standard error; SE) leptin concentrations during the preweaning phase than those fed the low plane (2.9 ± 0.1 vs. 2.6 ± 0.1 ng/mL). During the postweaning phase, mean circulating leptin was greater in heifers fed the high compared with the low postweaning diet [3.2 ng/mL (95% confidence interval; CI = 2.7 to 3.4) vs. 2.5 ng/mL (95% CI = 2.3 to 2.8)]. Mean (± SE) amplitude (2.1 ± 0.1 vs. 1.7 ± 0.1 ng/mL), peak (2.2 ± 0.1 vs. 1.8 ± 0.1 ng/mL), and duration (35.6 ± 2.1 vs. 28.7 ± 2.0 min) of LH pulses assessed at wk 15 were greater in heifers offered the high than those offered the low preweaning plane, but no nutritional effects were observed on LH pulses at wk 25. Mean (± SE) age at puberty was 250 ± 9 d and was not influenced by planes of nutrition. However, the likelihood of attaining puberty by 30 wk of age was greater (hazard ratio = 3.8; 95% CI = 1.0 to 14.4) in heifers fed the high postweaning plane compared with the low plane. Heifers achieving puberty by 30 wk also had greater leptin concentrations from wk 1 to 25, whereas for every 1 ng/mL increase in circulating leptin at wk 15 and 25, days to puberty were estimated to decrease by 22 d (95% CI = 1 to 44) and 13 d (95% CI = 1 to 24), respectively. Increasing the preweaning plane from 5 to 10 L/d of whole milk increased serum leptin concentrations at wk 1, 3, and 5 and LH pulse amplitude, peak, and duration at wk 15. Induction protocols based on progesterone (P4) are used prior to a synchronization program for timed-AI (TAI) to increase number of pubertal heifers and pregnancy per AI (P/AI). Injectable, long-lasting P4 (iP4) is a novel, practical method to supplement P4 in cattle. Here, we aimed to test the effect of an induction protocol based on a single injection of iP4 on P/AI of heifers. Bos indicus (Nellore) heifers were classified as pubertal (PUB; n = 224) or prepubertal (PRE; n = 414) based on two ovarian ultrasonographyc exams conducted 10 d apart. Heifers with a corpus luteum (CL) in any of the exams were considered PUB. Within each puberty status, heifers were assigned to receive nothing (NoiP4) or an induction protocol (iP4). Induction consisted of a single injection of 150 mg of iP4 on D-31, followed by injections of 1 mg of estradiol benzoate (EB) and 150 μg of prostaglandin analogue (PGF) on D-21. On D-9, all heifers received 2 mg of EB + 75 μg of PGF associated to intravaginal P4-device insertion. On D-3, P4-releasing devices were removed and 150 μg of PGF injected. Heifers were inseminated based on estrus on D-1 or were TAI on D0. On D0, all heifers received a dose of GnRH analogue. On D-21, iP4 treatment stimulated a 50% increase in the uterine score (UTS) and a 19% increase in the diameter of the largest follicle of PRE heifers (P < 0.01). On D-9, PRE|iP4 group had a greater proportion (P < 0.01) of CL (63.3%) than PRE|NoiP4 group (11.6%). On D-3, exposure to 6 d P4-releasing device stimulated UTS of PRE|NoiP4 group in a similar fashion than the induction protocol, but it did not have any additional positive effect for PRE|iP4 heifers. P/AI of PRE|iP4 group was similar to that of the PUB groups (44.7 vs 46.9%), but was more than that of PRE|NoiP4 (34.2%). There was an overall 7.7% increment (P = 0.07) on P/AI of iP4 treated heifers (iP4: 46.0% vs. NoiP4: 38.3%). In conclusion, implementation of an induction protocol based on iP4 was efficacious to hasten puberty. Background: In this study, the effects of different protein and energy levels in mixed feed on growth performance and reproduction characteristics of heifers were investigated. Method: The material of the study consisted of 36 Holstein Friesian (black pied) breed cattle at the age of 6 months and average live body weight of 156.65 kg and the experiment was continued for 8 months. During the experiment, four different feeds containing 16% CP - 2500 kcal / kg ME for the 1st group, 16% CP - 2700 kcal / kg ME for the 2nd group, 18% CP - 2500 kcal / kg ME for the 3rd group, 18% HP - 2700 kcal / kg ME for the 4th group were given at 1.5% of the live weight of heifers. (Where CP and ME stand for Crude Protein and Metabolizable Energy, respectively). As roughage, an equal amount of straw and alfalfa hay mixture was given. Result: As a result of this study, the effects of the different protein levels on live body weight and body measurements (body length- BL, withers height-WH, hip height-HH, hip width-HW, body depth-BD, chest girth-CG) were not found significant (P> 0.05). Similarly, the effects of different energy levels on body weight and body measurements (P> 0.05) and protein X energy interaction in terms of live body weight and size were also not found significant (P> 0.05). Regarding the reproductive properties, the differences between the groups in terms of the effect of different protein and energy levels on the first estrus age were found statistically significant (P <0.05). As a result of the analysis made in terms of heifers age during the first insemination, the difference was found to be statistically significant (P <0.05). The effect of different protein and energy levels on the conception rate in the first insemination was found significant (P <0.05). As a result, it was concluded that affordable mixed feed combinations can be used within the limits of mixed feed combinations containing 2500-2700 kcal / kg ME and 16-18% CP, which were given in the investigation; however, low energy-high protein combinations would give more positive results considering their effects on the reproductive performances and conception rates at first insemination. The current review is designed with aims to highlight the impact of heat stress (HS) on calves and heifers and to suggest methods for HS alleviation. HS occurs in animals when heat gain from environment and metabolism surpasses heat loss by radiation, convection, evaporation and conduction. Although calves and heifers are comparatively heat resistant due to less production of metabolic heat and more heat dissipation efficiency, they still suffer from HS to some degree. Dry matter intake and growth performance of calves and heifers are reduced during HS because of redistributing energy to heat regulation through a series of physiological and metabolic responses, such as elevated blood insulin and protein catabolism. Enhanced respiration rate and panting during HS accelerate the loss of CO2, resulting in altered blood acid-base chemistry and respiratory alkalosis. HS-induced alteration in rumen motility and microbiota affects the feed digestibility and rumen fermentation. Decreased luteinizing hormone, estradiol and gonadotrophins due to HS disturb the normal estrus cyclicity, depress follicular development, hence the drop in conception rate. Prenatal HS not only suppresses the embryonic development by the impaired placenta, which results in hypoxia and malnutrition, but also retards the growth, immunity and future milk production of newborn calves. Based on the above challenges, we attempted to describe the possible impacts of HS on growth, health, digestibility and reproduction of calves and heifers. Likewise, we also proposed three primary strategies for ameliorating HS consequences. Genetic development and reproductive measures, such as gene selection and embryo transfers, are more likely long-term approaches to enhance heat tolerance. While physical modification of the environment, such as shades and sprinkle systems, is the most common and easily implemented measure to alleviate HS. Two experiments were conducted to evaluate the effects of the timing of artificial insemination (AI) and incorporation of the Slo3 K+ channel blocker 4-(4-chlorophenyl)butyl-diethyl-heptylammonium to semen extender (CSE) on pregnancy per AI (P/AI) and pregnancy loss in dairy heifers. In experiment 1, Holstein heifers were subjected to the 5-d CIDR-Synch protocol: d −8 GnRH and controlled internal drug-release device (CIDR); d −3 PGF 2α and CIDR removal; d −2 PGF 2α ; d 0 GnRH) and assigned randomly to receive timed AI with control semen on d 0 (72-CON; n = 104), control semen on d −1 (48-CON; n = 100), or CSE-treated semen on d −1 (48-CSE; n = 98). Heifers were fitted with collar-mounted automated estrus detection devices to monitor physical activity and rumination. In experiment 2, Holstein heifers were subjected to the 5-d CIDR-Synch protocol and received a mount detection patch at the first PGF 2α injection. Heifers detected in estrus before d 0 were inseminated on the same day, whereas those not detected in estrus received the second GnRH injection and timed AI on d 0. Heifers were assigned randomly to receive AI with control (AI-CON; n = 148) or CSE-treated semen (AI-CSE; n = 110). Four bulls with proven fertility were used in both experiments, and ejaculates from each sire were divided and processed as CON or CSE. Pregnancy was diagnosed by transrectal ultrasonography at 29 and 54 d after AI. Data were analyzed by logistic regression, and statistical models included the fixed effects of treatment and enrollment week. In experiment 1, orthogonal contrasts were built to assess the effects of day of AI (72-CON vs. 48-CON + 48-CSE) and treatment of semen with CSE (48-CON vs. 48-CSE). Pregnancy per AI on d 29 (72-CON = 60.8, 48-CON = 35.2, 48-CSE = 39.8%) and d 54 (72-CON = 58.2, 48-CON = 31.6, 48-CSE = 36.2%) was greater for heifers inseminated on d 0 compared with d −1. However, no effect of semen extender on P/AI was observed in heifers inseminated on d −1. In experiment 2, P/AI tended to be greater for AI-CSE than for AI-CON on d 29 (58.6 vs. 47.3%) and d 54 after AI (55.6 vs. 43.7%). Advancing AI by 24 h decreased the likelihood of pregnancy, and use of CSE was unable to overcome the expected asynchrony between insemination and ovulation. Nevertheless, incorporation of CSE in semen processing tended to improve P/AI when heifers received AI upon detected estrus or timed AI concurrently with the final GnRH of the 5-d CIDR-Synch protocol. This study investigated the relationships between body weight (BW) and stayability, and BW and calving pattern, of 189,936 New Zealand dairy heifers. Heifers were classified into 5 breed groups: Holstein-Friesian (F), Holstein-Friesian crossbred (FX), Jersey (J), Jersey crossbred (JX), and Holstein-Friesian × Jersey crossbred (FJ). Body weight was predicted using Legendre polynomials at 6, 12, and 15 mo of age, and we analyzed their relationships with stayability, calving rate, and re-calving rate over the first 3 calvings. Approximately 92% of heifers calved for the first time at age 2 yr, 76% a second time at 3 yr, and 61% a third time at 4 yr. Heifers that were heavier were more likely to remain in the herd for first, second, and third calving compared with heifers that were lighter. Furthermore, we found positive curvilinear relationships between pre-breeding BW and reproductive performance of dairy heifers. Heifers that were heavier at 6, 12, and 15 mo were more likely to calve early for first calving compared with heifers that were lighter, regardless of breed group. In addition, we found a large range in BW where the probability of calving or re-calving early was high. For example, FJ heifers that were between 255 and 396 kg at 15 mo of age had 21-d calving and re-calving rates above 75 and 70%, respectively. For second and third lactations, however, heifer pre-breeding BW showed only small effects on the 21-d calving and re-calving rates. For heifers that were at the heaviest end of the BW range in the current study, slight declines in stayability and reproductive performance occurred, compared with heifers in the mid-range of BW. Consequently, for heifers that were above average in BW, the benefit of increasing BW before first breeding would be small and might even result in slight declines in stayability and reproductive performance. For heifers that were below average in BW, considerable beneficial effects on stayability and reproductive performance are predicted as a result of improving rearing practices to produce heavier heifers throughout the pre-breeding rearing phase. Background: The greatest impact on profitability of a commercial beef operation is reproduction. However, in beef heifers, little is known about the vaginal and fecal microbiota with respect to their relationship with fertility. To this end, we followed heifers through gestation to examine the dynamics of vaginal and fecal microbial composition throughout pregnancy. Results: Heifers were exposed to an estrus synchronization protocol, observed over a 12-day period, artificially inseminated 12 h to 18 h after observed estrus, and subsequently exposed to bulls for a 50-day breeding season. Vaginal samples were taken at pre-breeding (n = 72), during the first (n = 72), and second trimester (n = 72) for all individuals, and third trimester for individuals with confirmed pregnancies (n = 56). Fecal samples were taken at pre-breeding (n = 32) and during the first trimester (n = 32), including bred and open individuals. Next generation sequencing of the V4 region of the16S rRNA gene via the Illumina MiSeq platform was applied to all samples. Shannon indices and the number of observed bacterial features were the same in fecal samples. However, significant differences in vaginal microbiome diversity between gestation stages were observed. No differences in beta-diversity were detected in vaginal or fecal samples regarding pregnancy status, but such differences were seen with fecal microbiome over time. Random Forest was developed to identify predictors of pregnancy status in vaginal (e.g., Histophilus, Clostridiaceae, Campylobacter) and fecal (e.g., Bacteroidales, Dorea) samples. Conclusions: Our study shows that bovine vaginal and fecal microbiome could be used as biomarkers of bovine reproduction. The objectives were to evaluate pregnancy per artificial insemination (AI), days to first AI, and proportion pregnant within 7 d of AI eligibility in dairy heifers subjected to presynchronization compared with dairy heifers not presynchronized. Thirty days before AI eligibility, Holstein heifers were assigned randomly to 1 of 3 groups: 14-d controlled internal drug release (CIDR; containing progesterone) presynchronization, PGF 2α presynchronization, or control (no presynchronization). Heifers in the 14-d CIDR presynchronization treatment (n = 119) received a CIDR on d −30, which was removed on d −16, followed by an injection of PGF 2α upon entry to the breeding program (d 0). Heifers in the PGF 2α presynchronization treatment (n = 118) received an injection of PGF 2α on d −11 and d 0. Control heifers (n = 121) were not presynchronized and received an injection of PGF 2α on d 0. All heifers received tail paint on d 0 to facilitate once-daily detection of estrus (based on paint removal). Heifers detected in estrus received AI with conventional semen on the same morning as detected estrus. Generalized linear mixed models were used to assess mean treatment differences. Following PGF 2α treatment on d 0, more heifers were detected in estrus in the first 7 d after eligibility in the 14-d CIDR group (95.8%) compared with the PGF 2α (74.6%) and control (66.9%) groups. Days to first AI differed between treatments (14-d CIDR = 3.6 d vs. PGF 2α = 5.0 d vs. control = 6.8 d). Pregnancy per AI for first AI within 7 d of eligibility was 71.9% (14-d CIDR), 58.0% (PGF 2α), and 61.7% (control), and differed between 14-d CIDR and PGF 2α heifers. Presynchronization with a 14-d CIDR increased the proportion of heifers pregnant in the first 7 d of eligibility (14-d CIDR = 68.9% vs. PGF 2α = 43.2% vs. control = 41.3%). Projected days on feed (d 0 to projected calving date) were 295 (14-d CIDR), 302 (PGF 2α), and 305 (control), and were different between the 14-d CIDR and control heifers. The potential economic benefit to the producer was $15.85 per heifer presynchronized with a 14-d CIDR protocol compared with the control group. Treatment of dairy heifers with a 14-d CIDR effectively presynchronized estrus, resulting in a greater proportion detected in estrus, reduced days to first AI, and an increased proportion of heifers pregnant within the first 7 d after breeding eligibility compared with heifers presynchronized with a single PGF 2α injection and control heifers. Impaired fertility during periods of heat stress is the culmination of numerous physiological responses to heat stress, ranging from reduced estrus expression and altered follicular function to early embryonic death. Furthermore, heat-stressed dairy cattle exhibit a unique metabolic status that likely contributes to the observed reduction in fertility. An understanding of this unique physiological response can be used as a basis for improving cow management strategies, thereby reducing the negative effects of heat stress on reproduction. Potential opportunities for improving the management of dairy cattle during heat stress vary greatly and include feed additives, targeted cooling, genetic selection, embryo transfer and, potentially, crossbreeding. Previous studies indicate that dietary interventions such as melatonin and chromium supplementation could alleviate some of the detrimental effects of heat stress on fertility, and that factors involved in the methionine cycle would likely do the same. These supplements, particularly chromium, may improve reproductive performance during heat stress by alleviating insulin-mediated damage to the follicle and its enclosed cumulus–oocyte complex. Beyond feed additives, some of the simplest, yet most effective strategies involve altering the timing of feeding and cooling to take advantage of comparatively low nighttime temperatures. Likewise, expansion of cooling systems to include breeding-age heifers and dry cows has significant benefits for dams and their offspring. More complicated but promising strategies involve the calculation of breeding values for thermotolerance, the identification of genomic markers for heat tolerance, and the development of bedding-based conductive cooling systems. Unfortunately, no single approach can completely rescue the fertility of lactating dairy cows during heat stress. Feed and energy efficiency have been topics of extensive research in recent years, with residual feed intake (RFI) being the preferred index to determine feed efficiency in beef cattle. Nevertheless, the biggest downside of RFI is that its measurement requires accurately determining individual feed intake, which leads to the need to carry out RFI trials in confinement conditions. In this context, residual heat production (RHP), defined as the difference between observed and predicted heat production, presents an alternative to determine efficiency in grazing conditions as it is independent of feed intake. This work aimed to evaluate the repeatability of RHP and study its relationship with metabolic, endocrine, and reproductive variables. Seventy-four Hereford heifers were used, and at the beginning of the experimental period, heifers were 18 ± 0.7-month-old and weighed on average 278 ± 26 kg. Heat production (HP) was determined at the beginning (March to May; fall) and at the end (September to November; spring) of the experiment using the heart rate-O 2 pulse technique to classify heifers according with the residual heat production (RHP). Efficient heifers showed lower HP (P < 0.01), ME (P < 0.01), and DM intake (P < 0.01) without differences in retained energy (RE; only in spring). During fall, average daily gain (ADG) was not included in the estimated HP model; therefore, although, as in RFI, RHP should be independent of BW and ADG, a trend for greater ADG and RE was found for efficient when compared with inefficient heifers. Decreased maintenance requirements (NEm and MEm) as well as higher partial efficiency of use of consumed ME (k) were estimated for more efficient heifers (low RHP; HH), while efficient heifers showed an earlier luteal phase and a shorter service-conception interval and calved earlier in the season, probably associated to an earlier development of fat tissue. Excluding glucose, no major differences were found in the plasma metabolites between groups. The objective was to assess heifer weight and reproduction response to a peanut skins plus corn supplement during spring wheat grazing. In spring of 2021 and 2022, 10 and 8 pastures (1.62 ha/pasture) of fall overseeded wheat were randomly assigned to supplement (SUP) or control (CONT) constrained to the same number of replications per treatment. Pastures were stocked with 6 heifers/pasture each year. The initial goal was to use peanut skins as a single ingredient; however, testing palatability in year 1, before spring grazing, demonstrated a need to create a mixed supplement for improved acceptance. Over 5 weeks from initiation of grazing to breeding, heifers were transitioned from a 33:67 to a 50:50 peanut skins:corn mix. Supplement was offered at 0.5% BW, as-fed, at the start of spring wheat grazing in early March. An additional 2.4% and 8.3% inclusion of dried molasses was incorporated to further enhance acceptance in year 1 and 2, respectively. Estrus synchronization (ES) began March 31 and April 5 for each year. Data were analyzed across year with year considered as a random effect and pasture the experimental unit. Initial weight was 12 ± 5.8kg greater (P = 0.05) for SUP. Weight at ES tended (P = 0.11) to be 9.4 ± 5.55kg greater for SUP. Reproduction tract score was similar (P = 0.55), averaging 4.2 ± 0.13, and heifers were of similar (P = 0.33) age (405 ± 4.1 days) at time of insemination. Average daily gain from initial wheat grazing until removal for natural service breeding did not differ between treatments (P = 0.92) and averaged 1.0 ± 0.21 kg. Body condition score was similar (P = 0.43) at ES and averaged 5.7 ± 0.7. The heavier initial BW of SUP heifers was sustained through wheat grazing (P = 0.05). Ultrasound determined pregnancies averaged 66.7 and 50.0 ± 6.6% for SUP and CONT, respectively, for an odds ratio of 2.1 ± 0.79 (P = 0.08) at 30 days after insemination. Overall pregnancy rates following natural service averaged 88.9 and 79.6 ± 4.88% for SUP and CONT, respectively with an odds ratio of 2.1±1.12 (P=0.19). High dietary protein has been associated with reduced reproductive performance. Using the peanut skins plus corn supplement was hypothesized to improve reproduction on a high protein diet due to the possible effect of peanut skins tannins on dietary protein utilization and corn altering the rumen degradable protein to organic matter ratio. While the fixed time artificial insemination pregnancy rates for CONT would not be considered poor, there was a tendency for SUP to secure more pregnancies among heifers of similar age, reproductive tract score, and body condition. The effect of average daily gain (ADG) on reproductive outcomes in replacement dairy heifers was investigated. All heifers were managed in the typical Irish spring calving, pasture-based system, where the herd calves in 1 block between January and April and the majority of the diet comprises grazed grass. Heifer calves (n = 399) from 7 herds were weighed at birth and at the beginning of the breeding season, and ADG was calculated. Service dates and pregnancy diagnosis results were recorded, and conception dates were calculated. Days open (DO) was defined as the number of days between the beginning of the breeding season and conception. Genetic data were retrieved from the Irish Cattle Breeding Federation database. A Cox proportional hazard model was constructed to identify variables with a significant effect on DO. An accelerated failure time model was used to predict survival curves and median survival times for different combinations of the significant variables. The ADG ranged from 0.41 to 0.91 kg/d, with a median of 0.70 kg/d. Frailty effect of farm within year, maintenance subindex of the economic breeding index, and ADG had a significant effect on DO. Derived from the final accelerated failure time model, the predicted median DO for a heifer with an ADG of 0.40, 0.70, or 0.90 kg/d aged 443 d at the beginning of the breeding season and with a maintenance subindex in the second tercile were 27, 16, and 11 d, respectively. The effects of 2 deleterious recessive haplotypes on reproduction performance of Ayrshire cattle, Ayrshire Haplotype 1 (AH1) and Ayrshire Haplotype 2 (AH2), were investigated in Canadian Ayrshire cattle. We calculated their phenotypic effects on stillbirth (SB) rate and 56-d nonreturn rate (NRR) by estimating the interaction of service sire carrier status with maternal grandsire carrier status using the official Canadian evaluation models for those 2 traits. The interaction term included 9 subclasses for the 3 possible statuses of each bull: haplotype carrier, noncarrier, or not genotyped. For AH1, 394 carriers and 1,433 noncarriers were available, whereas 313 carriers and 1,543 noncarriers were available for the AH2 haplotype. The number of matings considered for SB was 34,312 for heifers (first parity) and 115,935 for cows (later parities). For NRR, 49,479 matings for heifers and 160,528 for cows were used to estimate the haplotype effects. We observed a negative effect of AH1 on SB rates, which was 2.0% higher for matings of AH1-carrier sires to dams that had an AH1-carrier sire; this effect was found for both heifers and cows. However, AH1 had small, generally nonsignificant effects on NRR. The AH2 haplotype had a substantial negative effect on NRR, with 5.1% more heifers and 4.0% more cows returning to service, but the effects on SB rates were inconsistent and mostly small effects. Our results validate the harmful effects of AH1 and AH2 on reproduction traits in the Canadian Ayrshire population. Highlights • SNPs located on X chromosome can affect the expression of reproductive traits. • X chromosome showed higher linkage disequilibrium when compared to the largest autosomes. • A genetic perspective was proposed to explain the physiology behind the results. Abstract Sex chromosomes are generally excluded from genomic analyses, although X chromosome is the second largest chromosome and harbors many genes. The objective of this study was to evaluate the effect of X chromosome by identifying its regions associated with reproductive traits in Nelore cows through a genome-wide association and by estimating its linkage disequilibrium. Phenotypic data of 76,845 and 76,700 animals were used for pregnancy at 16 months and age at first calving, respectively, as well as genotypic data of 1,999 females. Linkage disequilibrium was estimated for X chromosome and for autosomes 1 and 2, due to similar size of chromosomes. Single-step method was used for association analysis and some genes with known role in reproduction were reported for both traits, such as KLHL4 which is associated with development of the female reproductive system, and PCDH11X which is associated with the onset of primary ovarian insufficiency. Moreover, some genes were associated with both traits, indicating a pleiotropic effect. However, contribution of the windows in X-chromosome is very small when compared to autosomes: an unexpected fact. The results revealed higher linkage disequilibrium on the X chromosome (0.2248 ± 0.17) compared to the other chromosomes (0.1965 ± 0.14 and 0.1967 ± 0.14 for autosomes 1 and 2, respectively), indicating low recombination rates. The objective of this trial was to determine the effect of the utilization of 30 % shrimp (Litopenaeus vannamei) waste meal on some productive and reproductive indicators of grazing heifers, in Culiacán -Sinaloa, Mexico-. Between February and August, 2016, a trial was conducted with 12 Swiss Brown heifers, of 10-14 months of age, distributed in a completely randomized design with two treatments of six animals each: control treatment: 3 kg of supplement, which contained 100 % concentrate feed; experimental treatment: 3 kg of supplement which contained 70 % concentrate feed and 30 % shrimp waste meal. The remainder of the diet consisted in grazing and grass hay. The daily weight gain was favorable and significant with the presence of the shrimp waste meal (0,520 vs. 0,410 kg/animal/day); while the reproductive response was shown in the percentages of pregnant heifers at the end of the test (83 % in the experimental treatment vs. 50 % in the control). It was proven that the inclusion of 30 % shrimp waste meal allowed higher daily weight gain, as well as a decrease of some indicators of caloric stress, such as the pH of urine and feces, in addition to the rectum temperature; this permitted pregnancies in a shorter time period, essential aspect for cattle husbandry in Sinaloa. Commercial Angus heifers (n = 190; body weight (BW) = 315 ± 49.3 kg) were used to determine the effects of trace mineral injections during gestation on heifer and subsequent calf performance. Heifers received three previous subcutaneous trace mineral (Multimin 90 [ MM ]; n = 93) or sterilized physiological saline (CON ; n = 97) injections approximately 90 d apart. These treatments were maintained and subsequent injections were given 205, 114, and 44 ± 26 d prepartum. Heifers were provided free-choice inorganic minerals. Heifer BW and body condition scores (BCS) were collected at trial initiation (296 ± 26 d prepartum) and 5- to 10-week intervals thereafter. Liver samples were collected at trial initiation, 5 and 176 ± 3 d postpartum from a subset of cows to determine trace mineral status. Milk production was assessed on 80 cow–calf pairs (40/treatment) at 71 ± 15 d postpartum. Cows were artificially inseminated (AI) 82 d postpartum and then exposed to bulls for 38 d. Data were reported from 174 calves (n = 87 calves/treatment). Calf liver samples were collected 5 and 147 ± 3 d postpartum to determine trace mineral status. Calf weaning BW was collected at 159 ± 26 d postpartum. Calf performance including calving date, birth BW, weaning BW, average daily gain (ADG), and health data were collected. Heifer BW and BCS did not differ (P ≥ 0.72) throughout the experiment. Multimin heifers tended (P = 0.08) to have greater initial liver Se and tended to have decreased (P = 0.08) initial liver Zn compared with CON. At calving, MM cows had increased (P ≤ 0.01) liver Cu and Se. There was no difference (P ≥ 0.47) in Julian calving date, calving percent, or unassisted births. Calf birth BW was lesser (P = 0.02) for MM than CON calves, and MM calves had greater (P = 0.03) liver Cu concentrations at birth than CON calves. Despite MM cows having increased (P < 0.01) milk production, calf weaning BW and ADG were not different (P ≥ 0.87). In addition, calf morbidity and mortality were not different (P ≥ 0.43) between treatments. Calf mineral status was not different (P ≥ 0.57) at the time of weaning regardless of treatment; however, MM cows had decreased (P = 0.03) liver Zn. Multimin cows had decreased (P = 0.05) AI pregnancy rates, yet there was no difference (P = 0.34) in overall pregnancy rate. Supplementing an injectable trace mineral during heifer development and gestation increased cow milk production and resulted in decreased AI pregnancy rates; however, there was no effect on overall pregnancy rates or preweaning calf health or performance. The objective of this experiment was to assess the effects of eprinomectin, an extended-release injectable parasiticide, on fescue toxicosis and its impacts on beef heifer performance and reproduction. Fall-born Angus × Simmental heifers (age = 246.3 ± 22.4 d; 264.8 ± 21.1 kg body weight [BW]) were randomly assigned to one of two treatments: extended-release eprinomectin injection (ERE; n = 100) or control (CON; saline; n = 99). Treatments were administered at a rate of 1 mL/50 kg BW. Prior to experiment, heifers were dosed with oral fenbendazole to minimize parasite load. All heifers grazed endophyte-infected tall fescue as a single group and were offered a 50:50 supplement mix of corn gluten feed and soybean hulls (2.7 kg as fed per heifer per day). Body condition scores (BCS), BW, hair coat score (HCS), blood, and fecal samples were collected throughout the experiment. A subset of 40 heifers were randomly selected (20 per treatment) to assess respiration rate (RR). On d 138, heifers began a 14-d controlled internal drug release + prostaglandin synchronization protocol. Following artificial insemination (AI), heifers were exposed to five bulls for 71 d. On d 214 and 291, AI and overall pregnancy rates, respectively, were determined. There was a treatment × time interaction (P < 0.01) for BW, BCS, and average daily gain (ADG). The ERE heifers had greater (P < 0.04) BW and BCS compared to CON heifers from d 55 and 112, respectively. In addition, ERE heifers had greater (P ≤ 0.04) ADG from d 0 to 56, 56 to 112, 112 to 171, and 171 to 214; however CON heifers had greater (P < 0.01) ADG from d 214 to 291. There was no treatment × time interaction or treatment difference (P ≥ 0.27) for HCS, RR, and serum prolactin concentrations. However, serum prolactin decreased (P < 0.01) in all heifers over time. There was a treatment × time interaction (P< 0.01) for fecal egg counts (FEC). The FEC did not differ (P ≥ 0.32) on d −1 or 55; however, ERE heifers had decreased (P < 0.01) FEC compared with CON heifers on d 111 (1.52 vs. 13.56 eggs per gram). The ERE heifers tended (P = 0.10) to have greater AI pregnancy rates (69% vs. 58%) and had greater (P = 0.01) overall pregnancy rates (84% vs. 68%) than CON heifers. Spring administration of extended-release eprinomectin improved BW, ADG, BCS, and AI and overall pregnancy rates in fall-born beef heifers. A retrospective analysis was conducted to evaluate the effect of number of estrous cycles exhibited before breeding on growth and reproductive performance of replacement beef heifers fed ad-libitum or restricted by 20% less than ad-libitum during postweaning development over a 9-yr period. Progesterone concentration in blood samples collected at 9- to 11-d intervals were used to assign heifers into groups by number of estrous cycles exhibited before the start of breeding: 0 (nonpubertal; n = 395), 1 (n = 205), 2 (n = 211), 3 (n = 116), or >3 (n = 249). Heifers (P < 0.01) in the 0 cycle group were born 6 d later than the 1, 2, or 3 cycle groups, which were born 4 d later (P < 0.01) than the >3 cycle group. Weight of heifers at birth decreased (P < 0.05) as the number of cycles increased. Weaning weight and ultrasound measures of loin area and fat thickness over the loin at 1 yr age increased as the number of cycles increased (P < 0.01). Postwean weight gain, hip height at 1 yr age, and weights from the start of breeding through precalving increased with cycle numbers in a quadratic fashion (P < 0.02) and were greater (P < 0.05) in ad-libitum than restricted-fed heifers. Pregnancy rate in the 0 cycle group was lower (84%; P < 0.05) than the 1 (90%) or >3 (94%) estrous cycles groups and tended to differ (P < 0.1) from the 2 (88%) and 3 (89%) estrous cycle groups. Interval from the start of breeding to calving was 3 to 5 d longer (P < 0.05) for the 0 cycle group (300 ± 1 d) than other groups. Proportion of heifers calving in the first 21 d was less (P < 0.05) in the 0 or 1 cycle groups than other groups. Pregnancy rates of 2-yr-old cows (n = 898) were lowest (P < 0.05) for the 0 (73%) and 2 (79%) estrous cycle groups than the 1 (85%), 3 (90%), or >3 (92%) estrous cycle groups. Restricted level of feeding during postweaning development resulted in greater (P < 0.05) proportion of heifers in 0 cycle group and lower (P < 0.05) proportion in >3 cycle group, but reproductive performance was not influenced (P > 0.1) by level of feeding or interaction of feeding and estrous cycle grouping. In summary, date of birth and rate of physical maturation (weight, height, and fat deposition) were associated with timing of puberty. Pregnancy rate was greater in heifers that exhibited estrus before the start of breeding, but did not improve from having more than one estrous cycle. Abstract The objective of the present study was to identify genetic variation in heifer pregnancy due to heat stress in Nelore cattle during the breeding season. The Nelore heifers were born between 2003 and 2013 on six farms located in three Brazilian States. Heifer pregnancy (HP) was analyzed as a binary trait, with a value of 1 (success) assigned to females that were diagnosed pregnant and a value of 0 (failure) assigned to those that were not pregnant after breeding season. A threshold-reaction norm model was adopted to estimate (co)variance components and genetic parameters over temperature-humidity index (THI) values for HP. On the phenotypic scale, HP of Nelore cattle was slightly reduced by the increase in THI values (-0.85%/THI). The posterior means of the heritability estimates ranged from 0.17 at THI 82.5 to 0.01 at THI 88. The mean genetic correlations for HP between different THI values were below the unity. The genetic correlation between extreme THI values (82.5 and 88) was -0.47 (0.10). Genetic variation in the response to heat stress and heterogeneity in the (co)variance components and genetic parameters were observed across the THI scale. Conception rates in nulliparous beef heifers can be greatly influenced by their ability to reach puberty at or prior to the initiation of a breeding season. Previous research has analyzed the efficacy of utilizing both long and short-term exposure to a source of progesterone via a controlled internal drug release insert (CIDR) prior to estrous synchronization with timed artificial insemination (TAI) and found improved conception rates. The purpose of this study was to evaluate the effects of feeding melengesterol acetate (MGA) immediately and 10 days before estrous synchronization. Ninety-three crossbred heifers (395.67 ± 5.37 kg) were blocked by BW, genetics, and reproductive tract scores and allotted to 3 treatments. The 3 treatment groups were: 7-day Co-synch + CIDR without presynchronization (CON, n = 31); 2cc PGF2a (Lutalyse) followed by melengesterol acetate feeding for 7 d before the start of the 7-day Co-synch + CIDR program (PRE, n = 31); and 2 mL Lutalyse followed by MGA feeding for 7 d followed 10 d later by the start of the 7-day Co-synch + CIDR program (PRE+10, n = 31). The 7-day Co-synch + CIDR protocol was initiated on d 0 by administering a 2 mL IM injection of GnRH and placing a CIDR into the vagina. The CIDRs were removed 7 days later and accompanied by 2 ml IM injection of PGF2a. An injection of GnRH occurred 60 to 66 hours following PGF2a at TAI with frozen semen from a single bull. Ten days after TAI, heifers were exposed to a bull. Estrotect patches were applied throughout the study to assess estrous behavior. Ovaries were visualized 24 hours post TAI to determine whether ovulation occurred. Pregnancy was determined on days 40, 64, and 109 post TAI via transrectal ultrasonography. Blood was collected via jugular venipuncture (d -19, MGA initiation and termination, d 0, d 7, and d 9) and serum progesterone determined. Performance data were analyzed using the MIXED procedure and conception data were analyzed using the GLIMMIX procedure of SAS. Heifers in the PRE+10 treatment group had higher levels of progesterone (P = 0.04) at d 0 than PRE heifers. At d7 there was a tendency (P = 0.07) for PRE+10 heifers to have greater concentrations of progesterone than PRE, but did not differ by d 9 (TAI, P = 0.36). TAI conception rates in heifers in the PRE treatment group (63%) tended (P = 0.09) to be greater compared with the controls (35%) and not different from PRE+10 (43%), with no differences in season-long pregnancy rates (P > 0.15). The main objective was to compare pregnancy per AI (P/AI) between sex-selected and conventional semen in cyclic beef heifers subjected to a 5-day Co-synch plus CIDR protocol and evaluated the usefulness of an estrus detection (ED) aid to identify heifers that were most likely to conceive. This study also determined if the expression of estrus before timed-AI (TAI) would be associated with increased P/AI in acyclic heifers inseminated with conventional semen. Heifers (n = 1690; 320–523 kg of body weight, and 13–15 mo of age) at three locations over 2 years were scanned by ultrasonography to determine cyclicity (presence of luteal tissue) and reproductive tract normalcy. Cyclic heifers (n = 1331) received a progesterone releasing device (CIDR) on Day 0, CIDR removal and 500 μg of cloprostenol (PGF) on Day 5, and 100 μg of GnRH along with TAI on Day 8. Acyclic heifers (n = 275) received the same treatment with the addition of GnRH on Day 0. On Day 5, all heifers received ED patches (Estrotect™) that were scored from 0 to 3, based on color change between initial application and Day 8; 0 = unchanged, 1 = ≤ 50% color change, 2 = > 50% color change, 3 = missing. Estrus was defined to have occurred when an ED patch was scored 2 or 3. Cyclic heifers were inseminated with either frozen-thawed sex-selected or conventional semen from either of three sires available commercially (two per year). Acyclic heifers were inseminated with conventional semen. Pregnancy diagnosis was performed by transrectal ultrasonography 28 or 48 d after TAI, depending on management. The percentage of cyclic heifers was 83.9% and the average estrus response was 63.8%. P/AI was greater ( P  < 0.01) in cyclic compared to acyclic heifers (53.3 vs. 36.0%) and tended to be greater ( P  = 0.07) for conventional semen (52.3 vs. 47.6%), despite all acyclic heifers being inseminated with conventional semen. Heifers with an ED patch scored 2 (61.1%) or 3 (58.6%) had greater (P < 0.01) P/AI than those scored 0 (31.8%) or 1 (33.1%), regardless of semen type. Pregnancy per AI was greater ( P  < 0.01) for heifers detected in estrus (60.6 vs . 32.3%). In cyclic heifers that did not exhibit estrus, P/AI was lower (P < 0.01) in those inseminated with sex-selected semen (27.8 vs . 45.9%), while in heifers that exhibited estrus, P/AI only tended to be lower (P = 0.08; 56.7 vs . 65.5%). In summary, P/AI was greater in cyclic heifers, in those inseminated with conventional semen and in those exhibiting estrus before TAI. To determine the effects of repeated trace mineral injections on heifer development and reproductive performance, commercial Angus heifers (n = 290; 199 ± 34.3 kg; 221 ± 22 d of age) were utilized in a completely randomized design. Heifers were stratified by body weight (BW) and were administered an injectable trace mineral (MM; Multimin 90) or saline (CON) given subcutaneously, post-weaning at 221, 319, 401, and 521 ± 22 d of age. Throughout development, heifers grazed endophyte-infected fescue, red clover pastures and were supplemented with corn distillers grains (2.7 kg per heifer per day) and given access to free choice inorganic minerals. Heifer BW and body condition scores (BCS) were collected at trial initiation and 4- to 7-wk intervals thereafter. Hair coat scores (HCS) and respiration rates (n = 30 heifers per treatment) were collected at 269, 310, and 361 ± 22 d of age. Blood and liver samples were collected at trial initiation and estrous synchronization from 30 heifers per treatment to determine trace mineral status. At 319, 372, and 421 ± 22 d of age, antral follicle count and ovarian size were determined via ultrasonography. Two blood samples from all heifers were collected 10 d apart, concurrent with ultrasound dates, for cyclicity determination. Estrous synchronization was initiated, and reproductive tract scores (RTS) were collected at 421 ± 22 d of age, and heifers were bred via artificial insemination (AI) at 430 ± 22 d of age. Heifer BW, BCS, and HCS did not differ (P = 0.12) throughout development, except at 268 ± 22 d of age when BCS was greater (P = 0.03) for MM than CON heifers. Respiratory rates were greater (P = 0.05) for MM than CON heifers at 269 ± 22 d of age but did not differ (P = 0.66) at 310 and 361 ± 22 d of age. Plasma Mn and Zn concentrations did not differ (P = 0.54). However, MM heifers had greater (P = 0.01) plasma and liver concentrations of Cu and Se compared to CON. Interestingly, MM decreased (P = 0.02) liver Zn concentrations compared to CON, and there was no difference (P = 0.60) in liver Mn. Antral follicle count and ovarian size did not differ (P = 0.51) due to treatment. Throughout development, number of heifers cycling was lesser (P < 0.01) for MM than CON heifers. However, there was no difference (P = 0.19) in RTS, AI pregnancy rates, or overall pregnancy rates. Objective: In a nucleus breeding scheme, the sire of dam’s pathway plays an important role in producing genetic improvement. Selection proportion is the key parameter for predicting selection intensity, through truncating the normal distribution. Semen sexing using flow cytometry reduces the number of vials of sperm that can be obtained from a proved bull. In addition, a lower fertility of this kind of sperm is expected because of the lower sperm dosage in sex sorted semen. Both of these factors could affect the selection proportion in the sire of dam’s pathway (pSD). Methods: In the current study, through a deterministic simulation, effect of utilizing sex sorted semen on selection (pSD) was investigated in three different strategies including 1: continuous use of sex sorted semen in heifers (CS), 2: the use of sex sorted semen for the first two (S2) and 3: the first (S1) inseminations followed by conventional semen. Results: Results indicated that the use of sex sorted semen has a negative impact on the sire of dams (SD) pathway due to increase in selection proportion. Consequently selection intensity was decreased by 10.24 to 20.57, 6.38 to 8.87 and 3.76 to 6.25 percent in the CS, S2, and S1 strategies, respectively. Conclusion: Considering the low effect of sexed semen on genetic improvement in dam pathways, it is necessary to consider the joint effect of using sex sorted semen on the sire and dams pathway to estimate about the real effect of sexed semen on genetic improvement in a nucleus breeding scheme. The current study evaluated exosomes isolated from plasma of heifers bred to have high or low fertility through developing extreme diversity in fertility breeding values, however, key animal traits (e.g., body weight, milk production, and percentage of North American genetics) remained similar between the 2 groups. The exosomes were isolated by a combined ultracentrifugation and size exclusion chromatography approach and characterized by their size distribution (nanoparticle tracking analysis), morphology (transmission electron microscopy), and presence of exosomal markers (immunoblotting). In addition, a targeted mass spectrometry approach was used to confirm the presence of 2 exosomal markers, tumor susceptibility gene 101 and flotillin 1. The number of exosomes from plasma of high fertility heifers was greater compared with low fertility heifers. Interestingly, the exosomal proteomic profile, evaluated using mass spectrometry, identified 89 and 116 proteins in the high and low fertility heifers respectively, of which 4 and 31 were unique, respectively. These include proteins associated with specific biological processes and molecular functions of fertility. Most notably, the tetratricopeptide repeat protein 41-related, glycodelin, and kelch-like protein 8 were identified in plasma exosomes unique to the low fertility heifers. These proteins are suggested to play a role in reproduction; however, the role of these proteins in dairy cow reproduction remains to be elucidated. Their identification underscores the potential for proteins within exosomes to provide information on the fertility status and physiological condition of the cow. The selection of the best dairy heifers is mainly driven by the genetic value of their parents. The phenotype analysis of cows and of the daughters of bulls has been used to identify the best genetic value for decades before being replaced by genomic selection of individuals that are not yet parents. Because it is possible to predict the future value of an individual by its genetic makeup, it becomes feasible to do it as early as the blastocyst stage and to decide which should be transferred or not. Because we know the genotype of an animal at birth, or even before, it is becoming desirable to reproduce this animal as soon as possible to reduce generation interval and improve selection speed. Nature provides constraints that can be overcome: a single oocyte per cycle and age at puberty. Indeed, it is now possible to super-stimulate the ovary at any age and to start collecting oocytes at 6 mo by trans-vaginal ultrasonography. The challenge becomes the production of good eggs and embryos capable of implanting and developing into healthy calves. Our understanding of ovarian follicular physiology has been instrumental in designing stimulation protocols that may be adjusted to any physiological context including age, and even the individual animal, to obtain a good response. Therefore, the combination of procedures developed in cows to optimize oocyte quality, for example, FSH coasting, in association with in vitro fertilization and optimal culture conditions can now result in the production of several female embryos twice a month from animals 6 to12 mo of age. The transcriptomic and epigenetic analyses of embryos produced from the same females at different ages were compared and few differences were noted in particular in relation to embryo metabolism. These embryos are as good as the ones obtained from adult animals and can be produced with sexed sperm of bulls 12 mo of age.
