Despite the accumulating evidence showing the essential role of pentatricopeptide repeat (PPR) proteins in organellar biogenesis and plant development in Arabidopsis thaliana and maize (Zea mays), the functions of most PPR proteins in rice (Oryza sativa) are still unknown. A former study demonstrated that the mitochondria-localized Arabidopsis PPR19 is crucial for mitochondrial function and normal plant growth and development. In this study, we characterized the functional role of a rice ortholog (LOC_Os12g04110) of Arabidopsis PPR19 protein. The loss-of-function osppr19 mutant displayed delayed seed germination and stunted root and seedling growth compared with wild-type. The height of the osppr19 mutant was significantly shorter, and the grain mass of the mutant was lower than that of the wild-type. The osppr19 mutant carried few filled grains and a higher number of aborted seeds than the wild-type. The structures of mitochondria in the osppr19 mutant were abnormal, and more reactive oxygen species were accumulated in the mutant, suggesting defective mitochondrial biogenesis and function in the osppr19 mutant. Importantly, the amount of mature mitochondrial transcripts was significantly decreased in the mutant. Taken together, these results suggest that the mitochondrial OsPPR19 is essential for mitochondrial biogenesis and function, which is crucial for plant growth and development of rice grain.

Strigolactones (SL) are crucial plant hormones that regulate plant growth. We investigated genetic and metabolic changes in tobacco axillary flower buds following application of GR24 (SL synthetic analogue), administered 2 and 6 days later. The results indicated that GR24 effectively inhibited the growth of axillary buds. RNA sequencing revealed 1 781 differentially expressed genes in axillary buds after 6 days of GR24 treatment compared to untreated controls. Among them, 882 genes were up-regulated following GR24 treatment, suggesting substantial number of genes experienced significant changes in expression following GR24 treatment. Four carbohydrate metabolites exhibited altered abundance after 6 days of GR24 treatment; one increased and three decreased. In this study, GR24 induces substantial changes in the transcriptome and metabolome of tobacco axillary buds, with the starch and sucrose metabolic pathways and the phenylpropane biosynthesis pathway playing essential roles in the regulation of tobacco axillary bud development. Transcriptomic and metabolomic analyses highlighted that GR24 treatment significantly modulated the starch and sucrose metabolic pathways and the phenylpropane biosynthesis pathway. Our results suggest that the metabolic pathways of starch and sucrose and the biosynthesis pathway of phenylpropane play important roles in the regulation of growth and development of tobacco axillary buds by GR24.

Phosphorus is a key limiting factor for plant growth. Several approaches are developed to mitigate the impact of P shortage on plants and to the selection of crops with high P mobilizing capacity from P-deficient soils. In this work, four Medicago truncatula genotypes (A17, TN8.20, TN1.11, and TN6.18) were compared for their efficiency to cope with P limiting conditions using several criteria. Significant differences between genotypes, P deficiency treatments, and the interaction of genotypes with P deficiency treatments were found. P limitation resulted in an important decrease in shoot biomass, P content, P use efficiency, and photosynthetic parameters. A significant variability was found between the four genotypes, with A17 and TN8.20 being the most tolerant genotypes to P deficiency. This was consistent with the better ability of these genotypes to acidify rhizosphere and stimulate the activity of acid phosphatase and its relative gene (MtPAP1). The expression of P transporter genes (MtPT1, MtPT3, and MtPT5) was induced by P deficiency, however, the overexpression of those genes was more pronounced in tolerant genotypes. Overall, our data indicate that A17 and TN8.20 are more efficient in mobilizing P under limiting conditions and could be cultivated in P-deficient soils as forage crops.

The intensification of agriculture is closely linked to high emissions of greenhouse gases. To address the challenges, the European Commission published the European Green Deal in 2019. The aim of our study was to compare the yield of maize genotypes bred in Martonvásár in three different cropping environments (organic, irrigated conventional, and non-irrigated conventional). The silage and grain yields of different maize hybrids and parental lines were evaluated in a three-replicate small plot experiments. The green mass yield of the organic area was 19 and 15% lower compared to the irrigated conventional and non-irrigated conventional treatments. The dry matter yield of the maize hybrids was 12.9 t ha-1 in the organic area, 15.7 t ha-1 in the irrigated, and 15.8 t ha-1 in the non-irrigated environment. Hybrids had significantly better grain yield in the conventional systems (irrigated: 10.0 t ha-1 and non-irrigated: 9.8 t ha-1) than in the organic environment (7.6 t ha-1). The difference in yield results was not as considerable for the parental lines as for the hybrids. In addition, our results indicated high presence of heterosis for yields. The heterosis of the grain yields was two times higher than for silage yields. Heterosis was highest at the non-irrigated conventional area.

Perennial ryegrass (PRG; Lolium perenne) remains the backbone of grass swards in Northern Ireland due to its improved digestibility persistence, and ease of management compared with other grass species. However, innovative breeding approaches are needed that include positive environmental outcomes, as well as improved productivity in ruminants. The objective of this study was to evaluate the feasibility of root-trait screening and selection using the in situ coring method under commercial grass breeding field conditions. 108 root cores were sampled over a 2-year period from a field trial sown in autumn 2021. Root cores were washed, scanned, and analysed using the open-access root scanning platform Rhizovision. A seasonal effect was noted whereby significant differences were detected in October for root volume, network area, and surface area, but no significant differences for any root parameter were detected in April. No association was observed between root volume, network area, or surface area at the October sampling with either dry matter (DM) yield at the 4th cut (October) or annual DM yield. These results suggest that this method may be useful for identifying improved germplasm in PRG for root characteristics; however, being comparatively labour and time intensive this method may not be practicable for large-scale breeding programmes.

Pod shattering is an undesirable process leading to loss of harvestable yields. In the present study, we sought to undertake the first comprehensive phenotyping in 254 bean (Phaseolus vulgaris L.) genotypes for pod shattering including various mechanistic aspects as well as assess natural variation in the germplasm set for 16 seed physical traits including shattering score. There was substantial variability in 16 pod physical traits in the material. Significant diversity of the material in respect of pod traits was indicated by the broad range and coefficient of variation (CV) values. Using Random Impact Assessment (RIA), we found substantial variability in pod shattering score in common bean genotypes indicating significant diversity. Shattering score had a mean value of 6.098 with a range of 1.07 to 9.13. Highest shattering score was recorded in WB-6, WB-20-247, and N-7 while the lowest value for shattering score was recorded in WB-1129 and WB-216. Shattering score was negatively correlated with pod thickness (r = -0.698) followed by ventral/dorsal length ratio (r = -0.468) and positively correlated with breadth/thickness ratio (r = 0.599) and string % (r = 0.590). The principal component analysis (PCA) concentrated 86.91% variability in the first five principal components, and the first two PCs accounted for 55.62% of the total variation.

An untargeted metabolomic analysis of five Eucalyptus species was used to compare their chemical profiles. Gas chromatography-mass spectrometry (GC-MS) along with multivariate analyses including principal component analysis (PCA) and hierarchical cluster analysis (HCA) were applied to assess variations in the leaf chemical profiles of E. maidenii F. Muell, E. robusta Smith, E. citriodora Hook (syn Corymbia citriodora), E. tereticornis Smith and E. camaldulensis Dehnh. In total, 299 relevant metabolites were identified and subjected to statistical analyses in order to detect either the major common metabolites of the genus or the specific compounds contributing to the characteristic chemical composition of each species. Organic acids, nitrogenous compounds, and phenolic constituents were the main discriminatory groups between the investigated eucalypts. The cluster analysis revealed that fatty acids and fatty alcohols were more concentrated in the leaves of E. citriodora and distinguished it from other species, whereas sugars, polyols, and glycosides were characteristic to E. camaldulensis.

Excess water is an abiotic stress in plants, but the level at which excess water becomes varies widely between plant species. We conducted a two growing season replicated excess flooding experiment that was planted with 24 accessions of perennial ryegrass which had been vegetatively propagated to ensure equal representation of genotypes within an accession, both cultivars and ecotypes, from various geographical origins. The excess water treatment applied over the winter periods was achieved with irrigation. Yields increased in the winter-flooded treatment in contrast to the non-artificial watered control treatment significantly in 2017. In 2018 the same trend could be seen, but was not significant. Differences in composition of macro- and micronutrient profiles were observed. Sulphur was the only element with highly significantly increased concentration (0.25%) in flooded samples compared to control. Phosphorus, copper, iron, manganese, and molybdenum decreased statistically significantly under flooded conditions. In conclusion, perennial ryegrass is coping extremely well with excess water supplied over the winter period and can utilise it effectively in spring.

In plant breeding, polyploidization is an established technique to obtain superior phenotypic characteristics. In seed propagated agricultural crops, seed treatments with antimitotic agents are often used to obtain chromosome doubling. Here, we developed a method to induce polyploidization in clonally propagated fodder grasses Lolium, Festuca, and the intergeneric hybrid Festulolium. The aim was to obtain specific genotypes at both the diploid and tetraploid levels. We evaluated different types of plant explants, and the effects of the type, concentration, and application mode of three antimitotic agents (oryzalin, colchicine, and trifluralin) on the survival rate and the polyploidization efficiency. The treatment of greenhouse-grown tillers with antimitotics only resulted in mixoploids, while tissue culture propagated plants were successfully polyploidized. A shock pretreatment, using a high concentration of an antimitotic agent during a short period, successfully induced tetraploids in all three genera. Additionally, supplementing the tissue culture medium with a lower dosage of an antimitotic agent during minimal four weeks after the shock pretreatment further promoted polyploidization. By our methods, we were able to generate diploid and tetraploid plants with an identical genomic constitution but different ploidy allowing investigation of the effects of polyploidization on plant physiology and gene regulatory networks.

Opportunities exist to accelerate genetic gain in forage breeding using genome-wide selection approaches. In this study, we evaluated rapid cycle recurrent genomic selection (GS) as a means of improving genetic gain for value of annual forage yield. A small population of tetraploid half-sib families was evaluated for seasonal forage yield over two years, and the maternal parent plants were genotyped and genomic prediction models developed. The GS model for value of annual forage yield had a predictive ability of 0.23. An initial round of among-family selection based on field evaluations and within-family selection using genomic estimated breeding values was performed. This was followed by two further GS cycles. New synthetics were produced after each round of selection and were established in a field trial alongside the starting population. A positive response to selection was observed in new synthetics after two successive rounds of rapid cycle recurrent genomic selection before declining in the third round. The genetic gain for the value of annual forage yield was 2.4% from C0 SYN-1 to C1 SYN-1 and 6.4% from C1 SYN-1 to C2 SYN-1. In the case of C0 to C1, genetic gain was compounded by among-family selection based on field evaluations. The implementation of rapid cycle recurrent genomic selection offers an opportunity to increase genetic gain; however, the predictive ability is likely to decay rapidly as selection candidates become more distant from the training population.

Martaban camphor [Cinnamomum parthenoxylon (Jack) Meisn] is a woody tree in India, China, Indonesia, Thailand, and Vietnam and has been widely utilized for commercial purposes. It is threatened due to fragmented habitats, over-deforestation, and oil extraction. To conserve this species, the investigation of genetic diversity and population structure of this species is essential. Herein, we analyzed 192 adult trees from eight populations covering its natural distribution range in Vietnam using ten polymorphic EST-SSR markers. Medium levels of genetic diversity (R = 2.7, Ho = 0.399, He = 0.426) and genetic differences between populations (Fst = 0.223) were determined. Two populations, Cuc Phuong and Xuan Nha have undergone recent bottlenecks. These results indicated that anthropogenic activities may be the major factor for the low heterozygosity and influenced the number of alleles in all C. parthenoxylon populations. Clustering analyses revealed three genetic clusters that related to gene flow between different areas. We proposed in situ conservation for some populations with high levels of allelic richness, genetic diversity, or private alleles. The collecting of the seeds of the remaining populations for ex-situ conservation could be performed.

The coastal areas of western Taiwan feature acidic and saline soils with low fertility. Sodium bicarbonate (NaHCO3), produced readily by carbon capture and storage technologies, could be suitable for the neutralization of acidic soils, but its effects on plant growth and the ability of Bacillus subtilis var. natto to confer salinity tolerance remain unclear. In this study, we examined the potential of sodium bicarbonate and B. subtilis var. natto (NTU18) to improve the growth of tobacco (Nicotiana tabacum L.) under salt stress conditions. We found that salt stress was the main factor affecting tobacco growth, resulting in shorter roots and shoots, a reduced leaf area and leaf number, and clustered dark green leaves. The addition of sodium bicarbonate exacerbated the symptoms of salinity stress. Inoculating the soil with B. subtilis did not enhance salinity tolerance, but intriguingly it increased shoot and root growth under normal conditions and in the presence of sodium bicarbonate. The mechanism of growth promotion mediated by the bacteria is unknown and should be investigated in more detail.

Flavonoids are a class of phenolic compounds that are widely distributed in nature. They have a variety of physiological and pharmacological activities. They exist in free form or in the form of glycosides. The glycosylation occurs by glycosyltransferases, which is a common modification of plant secondary metabolites and the last step of their biosynthesis. Glycosylation can increase the diversity of the structure and function of flavonoids, and is currently a research hotspot. Based on the classification of flavonoids, this paper describes and summarizes the biotransformation and characteristics of glycosylation modification of flavonoids with different structural types, and describes the effects of enzymatic glycosylation using different types of glycosyltransferases on the biological activity and function of flavonoids depending on sugar connection position, sugar quantity, and type. This paper provides a reference for the development and application of flavonoids glycosylation and glycosyltransferases and also opens up a new direction for plant breeding.

Lettuce is one of our most important leaf vegetables that can be cultivated safely in organic farming, which is not only pesticide-free, but also aims to maintain and stimulate the presence of naturally occurring beneficial organisms, such as algae, mosses, bacteria, or arbuscular mycorrhiza (AM) fungi. These organisms are all beneficial for soil life and nutrient decomposition. The positive effects of beneficial microorganisms could be enhanced by mulching which is a widely used practice in organic farming. Mulching may also increase soil nutrient substance after decomposition and inhibit weed growth. In our experiment, we sought to determine the effect of different mulching techniques (alfalfa, rye, black foil) on AM root colonisation, leaf chlorophyll (Chl) content, and on peroxidase (POD) activity in Lactuca sativa plants and observe whether there are correlations between these parameters. Results show natural mulching has a positive effect on mycorrhiza fungi root colonisation and therefore lowers the stress in lettuce plant. On the other hand, there was no significant correlation between root colonisation and Chl content. As POD enzymes are directly linked to enzymatic browning, the high colonisation rate of AM may consequently lower post-harvest browning in lettuce.

This study aimed to evaluate the effect of putrescine priming on the initial growth, chlorophyll fluorescence, primary metabolites accumulation, and antioxidant enzyme activities in two maize hybrids with contrasting drought tolerances. Seeds of Zea mays L. hybrids DKB 390 (drought tolerant) and BRS 1030 (drought sensitive) were primed with putrescine (10 or 100 µM). Paper rolls moistened with distilled water or mannitol (-0.6 MPa) were maintened at 30°C for 7 d. The growth parameters were higher in the DKB hybrid than in the BRS hybrid. Putrescine priming (10 µM) promoted the root growth of BRS at levels similar to those of DKB and improved photochemical and non-photochemical quenching and maximum quantum efficiency of BRS seedlings. Higher levels of reducing sugars were found in DKB seedlings when compared to BRS in both roots and leaves, especially with 100 µM putrescine. Total soluble sugar and starch were lower in the maize roots under water deficit and with 10 µM putrescine for both hybrids. BRS seedlings showed higher starch content in the leaves in the control and 10 µM putrescine treatments. Superoxide dismutase was activated in BRS plants by the priming, especially in the roots, but this effect was not observed for catalase, ascorbate, or guaiacol peroxidase, although the DKB seedlings presented much higher guaiacol peroxidade activity than BRS seedlings in both the roots and shoots. In conclusion, putrescine priming (10 M) improved the morphological and biochemical responses of the drought sensitive maize hybrid BRS.

Phytoplankton can encounter dynamic changes in their environment including fluctuating nutrient supply, and therefore require survival mechanisms to compete for such growth-limiting resources. Diatoms, single-celled eukaryotic microalgae, are typically first responders when crucial macronutrients phosphorus (P) and nitrogen (N) enter the marine environment and therefore must have tightly regulated nutrient sensory systems. While nutrient starvation responses have been described, comparatively little is known about diatom nutrient sensing mechanisms. We previously identified that the model diatoms Phaeodactylum tricornutum and Thalassiosira pseudonana use calcium (Ca2+) ions as a rapid intracellular signaling response following phosphate resupply. This response is evident only in phosphate deplete conditions, suggesting that it is coordinated in P-starved cells. Rapid increases in N uptake and assimilation pathways observed following phosphate resupply, indicate tight interplay between P and N signaling. To regulate such downstream changes, Ca2+ ions must bind to Ca2+ sensors following phosphate induced Ca2+ signals, yet this molecular machinery is unknown. Here, we describe our findings in relation to known diatom P starvation signaling mechanisms and discuss their implications in the context of environmental macronutrient metadata and in light of recent developments in the field. We also consider the importance of studying phytoplankton nutrient signaling systems in the face of future ocean conditions.

Hydrogen sulfide (H2S) is a crucial signaling molecule in plants. Recent studies have shown that H2S plays an equally important role as nitric oxide (NO) and hydrogen peroxide (H2O2) in plant signaling. Previous studies have demonstrated the involvement of H2S in regulating drought and other stressful environmental conditions, but the exact downstream molecular mechanisms activated by the H2S signaling molecule remain unclear. In this study, we conducted a comprehensive genome-wide transcriptomic analysis of both wild type (WT) and double mutant (lcd/des1). Arabidopsis thaliana plants were exposed to 40% polyethylene glycol (PEG) to induce drought stress and 20 µM sodium hydrosulfide (NaHS). The resulting transcriptome data were analyzed for differentially significant genes and their statistical enrichments in the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. The results indicated significant upregulation of genes related to photosynthesis, carbon fixation, plant secondary metabolite biosynthesis, inositol and phosphatidylinositol signaling pathways, and stress-responsive pathways in mutant plants under drought stress. Mutant plants with impaired H2S signaling mechanisms displayed greater susceptibility to drought stress compared to wild-type plants. In summary, all findings highlight the pivotal role of H2S signaling in stimulating other drought-responsive signaling pathways.

Newly discovered plant behaviors, linked to historical observations, contemporary technologies, and emerging knowledge of signaling mechanisms, argue that plants utilize complex information processing systems. Plants are goal-oriented in an evolutionary and physiological sense; they demonstrate agency and learning. While most studies on plant plasticity, learning, and memory deal with the responsiveness of individual plants to resource availability and biotic stresses, adaptive information is often perceived from and coordinated with neighboring plants, while competition occurs for limited resources. Based on existing knowledge, technologies, and sustainability principles, climate-smart agricultural practices are now being adopted to enhance crop resilience and productivity. A deeper understanding of the dynamics of plant behavior offers a rich palette of potential amelioration strategies for improving the productivity and health of natural and agricultural ecosystems.

The rising prevalence of viral-induced diseases, particularly those caused by certain strains, poses a substantial risk to the genetic diversity of Solanaceae crops and the overall safety of horticultural produce. According to the “gene-for-gene” hypothesis, resistance proteins are capable of selectively identifying nontoxic effectors produced by pathogens, as they are under purview of the host’s immune defenses. The sensitivity and responsiveness of Solanaceae plants to viral attacks play a crucial role in shaping the outcomes of their interactions with viruses. Pathogenic organisms, devise an array of infection tactics aimed at circumventing or neutralizing the host’s immune defenses to facilitate effective invasion. The invasion often accomplishes by suppressing or disrupting the host’s defensive mechanisms or immune signals, which are integral to the infection strategies of such invading pathogens. This comprehensive review delves into the myriad approaches that pathogenic viruses employ to infiltrate and overcome the sophisticated immune system of tomatoes. Furthermore, the review explores the possibility of utilizing these viral strategies to bolster the resilience of horticultural crops, presenting a hopeful direction for forthcoming progress in plant health and agricultural stability.

Currently, applying lime to cadmium (Cd)-contaminated paddy fields to increase pH and reduce Cd availability is an effective method to control excessive Cd levels in rice grain. However, under hydroponic conditions, the impact of increased pH on Cd accumulation in different rice varieties remains unclear. This study employed three rice varieties (Yuzhenxiang, Shaoxiang 100, Xiangwanxian 12) with different Cd accumulation characteristics under different pH and long-term treatment with 1 μM CdCl2, to study the effect of pH on growth and Cd accumulation in different rice varieties. The result showed that as pH shifted from 5 to 8, the SPAD values, shoot dry weight, and plant height of the three rice varieties significantly decreased. The main root length, root volume, and root dry weight of Yuzhenxiang, and Shaoxiang100 significantly decreased. Conversely, the root architecture indicators of Xiangwanxian 12 did not change significantly. As for element accumulation, increasing the pH significantly increased the content of Mn in both the shoots and roots of all three varieties. Yuzhenxiang significantly reduced Cd content in both the shoots and roots of rice, while Shaoxiang100 significantly increased Cd content in both parts. Xiangwanxian 12 showed a significant increase in Cd content in the shoots but a decrease in the roots. In terms of subcellular distribution, Yuzhenxiang significantly reduced Cd concentrations in the cell wall and organelles of root cells, resulting in lower Cd concentrations in the root tissue. Conversely, Shaoxiang100 significantly increased Cd concentrations in the cell wall, organelles, and soluble fractions of root cells, leading to higher Cd concentrations in the root tissue. Xiangwanxian 12 also exhibited a decrease in Cd concentrations in the cell wall, organelles, and soluble fraction of root cells, resulting in lower Cd concentrations in the root tissue. Additionally, the expression of the OsNRAMP5 and OsHMA3 gene was significantly increased in Shaoxiang 100, while no significantly change in Yuzhenxiang and Xiangwanxian 12. These results provide important guidance on the impact of pH on Cd accumulation during the vegetative growth stage of different rice varieties.

Ralstonia solanacearum and R. pseudosolanacearum, the causative agents of bacterial wilt, ranks as the second most devastating phytopathogens, affecting over 310 plant species and causing substantial economic losses worldwide. R. solanacearum and R. pseudosolanacearum infect plants through the underground root system, where it interacts with both the host and the surrounding microbiota and multiply in the xylem where bacteria cell and its polysaccharide product block the water transportation from root to aboveground. Currently, effective control methods are limited, as resistance genes are unavailable and antibiotics prove ineffective. In current Commentary, we review recent advancements in combating bacterial wilt, categorizing the approaches (weapons) into three distinct strategies. The physical and chemical weapons focus on leveraging sound waves to trigger crop immunity and reducing bacterial virulence signaling, respectively. The biological weapon employs predatory protists to directly consume Ralstonia cells in the root zone, while also reshaping the protective rhizosphere microbiome to fortify the plant. We believe that these novel methods hold the potential to revolutionize crop protection from bacterial wilt and inspire new era in sustainable agriculture.

The plant root absorbs water and nutrients, anchors the plant in the soil, and promotes plant development. Root is developed from root apical meristem (RAM), which is formed during embryo stage and is maintained by dividing stem cells. Plant hormones have a predominant role in RAM maintenance. This review evaluates the functional crosstalk among three major hormones (auxin, cytokinin, and brassinolide) in RAM development in Arabidopsis, integrating a variety of experimental data into a regulatory network and revealing multiple layers of complexity in the crosstalk among these three hormones. We also discuss possible directions for future research on the roles of hormones in regulating RAM development and maintenance.

Tripartite interactions among plants, fungi, and bacteria are critical for maintaining plant growth and fitness, and volatile organic compounds (VOCs) play a significant role in these interactions. However, the functions of VOCs within the niche of mycoheterotrophic plants, which represent unique types of interactions, remain poorly understood. Gastrodia elata, a mycoheterotrophic orchid species, forms a symbiotic relationship with specific Armillaria species, serving as a model system to investigate this intriguing issue. Rahnella aceris HPDA25 is a plant growth-promoting bacteria isolated from G. elata, which has been found to facilitate the establishment of G. elata-Armillaria symbiosis. In this study, using the tripartite symbiotic system of G. elata-Armillaria gallica-R. aceris HPDA25, we investigate the role of VOCs in the interaction among mycoheterotrophic plants, fungi, and bacteria. Our results showed that 33 VOCs of HPDA25-inducible symbiotic G. elata elevated compared to non-symbiotic G. elata, indicating that VOCs indeed play a role in the symbiotic process. Among these, 21 VOCs were accessible, and six active VOCs showed complete growth inhibition activities against A. gallica, while R. aceris HPDA25 had no significant effect. In addition, three key genes of G. elata have been identified that may contribute to the increased concentration of six active VOCs. These results revealed for the first time the VOCs profile of G. elata and demonstrated its regulatory role in the tripartite symbiotic system involving G. elata, Armillaria, and bacteria.

Lead is one of the major environmental pollutants which is highly toxic to plants and living beings. The current investigation thoroughly evaluated the synergistic effects of oxalic acid (OA) and salicylic acid (SA) on Zea mays L. plants subjected to varying durations (15, 30, 30, and 45 days) of lead (Pb) stress. Besides, the effects of oxalic acid (OA) combined with salicylic acid (SA) for different amino acids at various periods of Pb stress were also investigated on Zea mays L. The soil was treated with lead nitrate Pb (NO3)2 (0.5 mM) to induce Pb stress while the stressed plants were further treated using oxalic acid (25 mg/L), salicylic acid (25 mg/L), and their combination OA + SA (25 mg/L each). Measurements of protein content, malondialdehyde (MDA) levels, guaiacol peroxidase (GPOX) activity, catalase (CAT) activity, GSH content, and Pb concentration in maize leaves were done during this study. MDA levels increased by 71% under Pb stress, while protein content decreased by 56%, GSH content by 35%, and CAT activity by 46%. After treatment with SA, OA, and OA+SA, there was a significant reversal of these damages, with the OA+SA combination showing the highest improvement. Plant Class III peroxidases have diverse roles in controlling root hair growth, anther development, and abiotic and biotic stress responses. However, their abiotic stress response mechanism in rice remains elusive. Here, we identified a peroxidase precursor gene, OsPRX83, and investigated its role in enhancing osmotic stress tolerance in rice. We used OsPRX83 overexpression and CRISPR-Cas9-generated mutant lines to elucidate OsPRX83‘s function and expression patterns under stress conditions. The expression of OsPRX83 was induced by H2O2, PEG, NaCl, and ABA treatments. Using qRT-PCR, RNA sequencing, and physiological assays, we demonstrated that overexpression of OsPRX83 enhanced the osmotic and oxidative stress tolerance as compared to the wild-type and mutant seedlings, as evident from the higher survival rates, enhanced peroxidase (POD) and ascorbate peroxidase (APX) activities, and increased ABA sensitivity compared with mutants and wild-type plants. Transcriptome analysis further supported the involvement of OsPRX83 in the ROS scavenging, by modulating the expression of OsDREB1B, OsDREB1E, OsDREB1F, OsDREB1G in response to osmotic treatment. In summary, our study suggests that OsPRX83 plays a pivotal role in enhancing stress tolerance in rice through ABA-dependent pathways and ROS scavenging. Therefore, this study elucidates the function of a novel abiotic stress response gene in rice, thereby may contribute to a new genetic engineering resource for engineering drought-resistant rice varieties.

SMALL AUXIN UP RNAs (SAURs), the largest family of early auxin response genes, plays crucial roles in multiple processes, including cell expansion, leaf growth and senescence, auxin transport, tropic growth and so on. Although the rice SAUR gene family was identified in 2006, it is necessary to identify the rice SAUR gene due to the imperfection of its analysis methods. In this study, a total of 60 OsSAURs (including two pseudogenes) distributed on 10 chromosomes were identified in rice (Oryza sativa). Bioinformatics tools were used to systematically analyze the physicochemical properties, subcellular localization, motif compositions, chromosomal location, gene duplication, evolutionary relationships, auxin-responsive cis-elements of the OsSAURs. In addition, the expression profiles obtained from microarray data analysis showed that OsSAUR genes had different expression patterns in different tissues and responded to auxin treatment, indicating functional differences among members of OsSAUR gene family. In a word, this study provides basic information for SAUR gene family of rice and lays a foundation for further study on the role of SAUR in rice growth and development.

Arabidopsis EARLY LIGH-INDUCIBLE PROTEIN 2 (ELIP2) is a chlorophyll- and carotenoid-binding protein and is involved in photoprotection under stress conditions. Because its expression is induced through high light, cold, or UV-B stressors, its mechanism of induction has been studied. It is known that a functional unit found in the promoter, which is composed of Element B and Element A, is required and sufficient for full activation by these stressors. In this study, the role of each element in the unit was analyzed by introducing weak mutations in each element as synthetic promoters in addition to intensive repeat constructs of each single element. The results suggest that a stressor like cold stress generates two parallel signals in plant cells, and they merge at the promoter region for the activation of ELIP2 expression, which constitutes an “AND” gate and has a potential to realize strong response with high specificity by an environmental trigger.

Most studies on anesthesia focus on the nervous system of mammals due to their interest in medicine. The fact that any life form can be anaesthetised is often overlooked although anesthesia targets ion channel activities that exist in all living beings. This study examines the impact of lidocaine on rice (Oryza sativa). It reveals that the cellular responses observed in rice are analogous to those documented in animals, encompassing direct effects, the inhibition of cellular responses, and the long-distance transmission of electrical signals. We show that in rice cells, lidocaine has a cytotoxic effect at a concentration of 1%, since it induces programmed reactive oxygen species (ROS) and caspase-like-dependent cell death, as already demonstrated in animal cells. Additionally, lidocaine causes changes in membrane ion conductance and induces a sharp reduction in electrical long-distance signaling following seedlings leaves burning. Finally, lidocaine was shown to inhibit osmotic stress-induced cell death and the regulation of Ca2+ homeostasis. Thus, lidocaine treatment in rice and tobacco (Nicotiana benthamiana) seedlings induces not only cellular but also systemic effects similar to those induced in mammals.

Salvia miltiorrhiza, known as Danshen, is a traditional Chinese medicinal plant with significant cardiovascular benefits, attributed to its secondary metabolites, particularly tanshinones. Despite their medicinal value, tanshinones occur in low natural abundance, necessitating research to increase their content. This study explores the role of the ARF transcription factor (SmARF1) in tanshinone accumulation in Danshen. Overexpressing SmARF1 in hairy roots significantly increased tanshinone levels. EMSA and Dual-LUC assays revealed that SmMYB36, a transcription factor interacting with SmMAPK3, binds to and regulates the SmARF1 promoter. SmMYB36 alone inhibited the expression of SmARF1 gene, while its interaction with SmMAPK3 enhanced SmARF1 promoter activity. This MAPK3-MYB36-ARF1 module elucidates a complex regulatory mechanism for tanshinone biosynthesis, offering insights for targeted enhancement of tanshinone content through advanced biotechnological approaches.

Sophora davidii is a cross-pollinated plant with important ecological protection and medicinal value in China, but its seed yield is low due to backward and nonstandard production technology. Therefore, we divide the flowering period of Sophora davidii into initial, full and final flowering period, measuring the floral morphology, pollen viability, stigma receptivity, nectar volume and nectar concentration, foraging behavior of pollinators, fertilization physiology, seed yield and quality through field observation and indoor testing to explore whether the flowering period affects the floral traits, pollinator behavior and seed production of plants. Our results revealed that the nectar volume, nectar concentration, pollen viability and stigma receptivity at full flowering period were the highest. The single visit time and visit time per flower of Chinese honey bees were the longest in the full flowering period, while the number of transfer, visit frequency and number of touching stigma were the least. The visiting number of the bees was the most and the most active in the full flowering period. The bees pollination not only improved the pollen amount, germination rate, pollen tube length and the ovule number of S. davidii, but also their effect was the most obvious in full flowering period. The principal component analysis showed that pollination by Chinese honey bees during the whole flowering period of S. davidii was the best way to produce seeds. We can conclude that flowering period affects flower traits, foraging behavior of pollinators, seed yield and quality of S. davidii.

To investigate the synergistic effect of IAA and melatonin (MT) on three plants to alleviate the effects of salt damage on plants, we aim to determine the optimal concentrations of exogenous hormone treatments that improve salinity resistance for each species. In this experiment, three desert plants, Sarcozygium xanthoxylon, Nitraria tangutorum, and Ammopiptanthus mongolicus, which are common in Wuhai City, were used as plant materials. Two time periods (12 h,24 h) of exogenous hormone IAA (100 μmol/L) and exogenous melatonin concentration (0, 100, 200, 300 μmol/L) were used to treat the three desert plants in saline soil under different conditions of exogenous IAA and exogenous melatonin. The results indicate that under different concentrations of exogenous IAA and melatonin, the germination rate and vigor of the three desert plant species in saline-alkaline soil improved. However, as the concentration of melatonin increased, the germination rate and vigor of these desert plants were inhibited. Whereas, plant height, root length, leaf length, fresh weight, dry weight, and root vigor of the three desert plants were alleviated under different conditions of exogenous IAA and exogenous melatonin. under the action of two exogenous hormones, the low concentration of melatonin decreased their malondialdehyde content and increased their proline content. As melatonin levels increased, the activity of antioxidant enzymes also rose initially, followed by a subsequent decline. This study highlights the synergistic effects of two exogenous hormones on the critical role of cell osmomodulators and antioxidant enzyme activity in combating salinity damage in three desert plants.

Parthenocarpy, characterized by seedless fruit development without pollination or fertilization, offers the advantage of consistent fruit formation, even under challenging conditions such as high temperatures. It can be induced by regulating auxin homeostasis; PAD1 (PARENTAL ADVICE-1) is an inducer of parthenocarpy in Solanaceae plants. However, precise editing of PAD1 is not well studied in peppers. Here, we report a highly efficient clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) ribonucleoprotein (RNP) for CaPAD1 editing in three valuable cultivars of pepper (Capsicum annuum L.): Dempsey, a gene-editable bell pepper; C15, a transformable commercial inbred line; and Younggo 4, a Korean landrace. To achieve the seedless pepper trait under high temperatures caused by unstable climate change, we designed five single guide RNAs (sgRNAs) targeting the CaPAD1 gene. We evaluated the in vitro on-target activity of the RNP complexes in three cultivars. Subsequently, we introduced five CRISPR/Cas9-RNP complexes into protoplasts isolated from three pepper leaves and compared indel frequencies and patterns through targeted deep sequencing analyses. We selected two sgRNAs, sgRNA2 and sgRNA5, which had high in vivo target efficiencies for the CaPAD1 gene across the three cultivars and were validated as potential off-targets in their genomes. These findings are expected to be valuable tools for developing new seedless pepper cultivars through precise molecular breeding of recalcitrant crops in response to climate change.

Plant memory plays an important role in the efficient and rapid acclimation to a swiftly changing environment. In addition, since plant memory can be inherited, it is also of adaptive and evolutionary importance. The ability of a plant to store, retain, retrieve and delete information on acquired experience is based on cellular, biochemical and molecular networks in the plants. This review offers an up-to-date overview on the formation, types, checkpoints of plant memory based on our current knowledge and focusing on its transcriptional aspects, the transcriptional memory. Roles of long and small non-coding RNAs are summarized in the regulation, formation and the cooperation between the different layers of the plant memory, i.e. in the establishment of epigenetic changes associated with memory formation in plants. The RNA interference mechanisms at the RNA and DNA level and the interplays between them are also presented. Furthermore, this review gives an insight of how exploitation of plant transcriptional memory may provide new opportunities for elaborating promising cost-efficient, and effective strategies to cope with the ever-changing environmental perturbations, caused by climate change. The potentials of plant memory-based methods, such as crop priming, cross acclimatization, memory modification by miRNAs and associative use of plant memory, in the future’s agriculture are also discussed.

The process of silique dehiscence is essential for the proper dispersal of seeds at the end of a dehiscent fruit plants lifecycle. Current research focuses on genetic manipulation to mitigate this process and enhance shatter tolerance in crop plants, which has significant economic implications. In this study, we have conducted a time-course analysis of cell patterning and development in valve tissues of Arabidopsis thaliana and closely related Triangle of U species (Brassica juncea, Brassica carinata, Brassica napus, Brassica rapa, and Brassica nigra) from Brassicaceae. The goal was to decipher the detailed temporal developmental patterns of the endocarp a and b cell layers of the valve, specifically their degradation and lignification respectively. Additionally, we propose a new classification system for the lignification of the endocarp a cell layer: L1 indicates the cell closest to the replum, with L2 and L3 representing the second and third cells, respectively, each numerical increment indicating lignified cells farther from the replum. Our findings provide a foundational framework absent in current literature, serving as an effective blueprint for future genomic work aimed at modifying valve structures to enhance agronomic traits, such as reducing fiber (lignin) or increasing shatter tolerance.

The CLAVATA pathway plays a key role in the regulation of multicellular shoot and root meristems in flowering plants. In Arabidopsis, CLAVATA 3-like signaling peptides (CLEs) act via receptor-like kinases CLAVATA 1 and CRINKLY 4 (CR4). In the moss Physcomitrium patens, PpCLAVATA and PpCR4 were previously studied independently and shown to play conserved roles in the regulation of cell proliferation and differentiation. The plant calpain DEFECTIVE KERNEL 1 (DEK1) has been identified as another key regulator of cell division and cell fate in vascular plants and bryophytes. The functional interaction between CLAVATA, CR4, and DEK1 remains unknown. Here, we show that P. patens crinkly4 and dek1 mutants respond differently to CLE peptide treatments suggesting their distinct roles in the CLAVATA pathway. Reduced CLAVATA-mediated suppression of leafy shoot growth in Δcr4 mutants indicates that PpCR4 is involved in CLV3p perception, most likely as a receptor. The CLV3p strongly suppressed leaf vein development in Δcr4 mutants, suggesting that other receptors are involved in these processes and indicating a potential role of PpCR4 in organ sensitization to CLEs.

Lilium pumilum DC (L. pumilum DC) plays an important role in the rational utilization of salinized soil. To explore the molecular mechanism of salt-tolerant L. pumilum, the LpMYB4 was cloned. LpMYB4 close relationship with Bambusa emeiensis and Zea mays MYB4 throughout the phylogenetic tree construction. LpMYB4 protein was found to be localized in the nucleus. Prokaryotic and eukaryotic bacterial solution resistance experiments proved that the exogenous introduction of LpMYB4 made the overexpression strains obtain better survival ability under saline-alkaline stress. Compared with wild-type plants, tobacco plants overexpressing LpMYB4 had better growth and lower leaf wilting and lodging, the content of chlorophyll was higher, the content of hydrogen peroxide and superoxide anion was lower, the activity of peroxidase and superoxide dismutase was higher and the relative conductivity was lower under saline-alkaline stress. The analysis of seed germination and seedling resistance of transgenic plants under salt stress showed that LpMYB4 transgenic seeds were more tolerant to salt stress during germination and growth. Yeast two-hybrid and two-luciferase complementation experiments showed that LpMYB4 interacted with yeast two-hybrid and LpGPX6. The analysis of the role of LpMYB4 in improving plant saline-alkali resistance is helpful to the transformation of plant germplasm resources and has great significance for agriculture and sustainable development.

Rapeseed (Brassica napus L.) is an important oilseed crop widely cultivated worldwide, and drought is the main environmental factor limiting its yield enhancement and the expansion of planted areas. SIMILAR TO RCD ONE (SRO) is a plant-specific small gene family that plays a crucial role in plant growth, development, and responses to abiotic stresses such as drought. However, the functional role of SROs in rapeseed remains poorly understood. In this study, 19 BnaSROs were identified from the rapeseed genome, with 9, 10, 10, 18, and 20 members identified from the genomes of Brassica rapa, Brassica nigra, Brassica oleracea, Brassica juncea, and Brassica carinata, respectively. We then analyzed their sequence characteristics, phylogenetic relationships, gene structures, and conserved domains, and explored the collinearity relationships of the SRO members in Brassica napus and Brassica juncea. Next, we focused on the analysis of tissue expression and stress-responsive expression patterns of rapeseed SRO members and examined their expression profiles under ABA, MeJA and water-deficit drought treatments using qPCR. Transcriptome data analysis and qPCR detection indicated that BnaSROs exhibit multiple stress-responsive expression patterns. BnaSRO1 and BnaSRO11, which are likely to function through interactions with NAC transcription factors, were screened as major drought-regulated members. Our results provide a solid foundation for functional analysis of the role of the SRO gene family in abiotic stress responses, especially drought stress responses, in rapeseed.

One of the main signal transduction pathways that modulate plant growth and stress responses, including drought, is the action of phytohormones. Recent advances in omics approaches have facilitated the exploration of plant genomes. However, the molecular mechanisms underlying the response in the crown of barley, which plays an essential role in plant performance under stress conditions and regeneration after stress treatment, remain largely unclear. The objective of the present study was the elucidation of drought-induced molecular reactions in the crowns of different barley phytohormone mutants. We verified the hypothesis that defects of gibberellins, brassinosteroids, and strigolactones action affect the transcriptomic, proteomic, and hormonal response of barley crown to the transitory drought influencing plant development under stress. Moreover, we assumed that due to the strong connection between strigolactones and branching the hvdwarf14.d mutant, with dysfunctional receptor of strigolactones, manifests the most abundant alternations in crowns and phenotype under drought. Finally, we expected to identify components underlying the core response to drought which are independent of the genetic background. Large-scale analyses were conducted using gibberellins-biosynthesis, brassinosteroids-signaling, and strigolactones-signaling mutants, as well as reference genotypes. Detailed phenotypic evaluation was also conducted. The obtained results clearly demonstrated that hormonal disorders caused by mutations in the HvGA20ox2, HvBRI1, and HvD14 genes affected the multifaceted reaction of crowns to drought, although the expression of these genes was not induced by stress. The study further detected not only genes and proteins that were involved in the drought response and reacted specifically in mutants compared to the reaction of reference genotypes and vice versa, but also the candidates that may underlie the genotype-universal stress response. Furthermore, candidate genes involved in phytohormonal interactions during the drought response were identified. We also found that the interplay between hormones, especially gibberellins and auxins, as well as strigolactones and cytokinins may be associated with the regulation of branching in crowns exposed to drought. Overall, the present study provides novel insights into the molecular drought-induced responses that occur in barley crowns.

G protein-coupled receptors (GPCRs) constitute the largest family of transmembrane proteins in metazoans that mediate the regulation of various physiological responses to discrete ligands through heterotrimeric G protein subunits. The existence of GPCRs in plant is contentious, but their comparable crucial role in various signaling pathways necessitates the identification of novel remote GPCR-like proteins that essentially interact with the plant G protein α subunit and facilitate the transduction of various stimuli. In this study, we identified three putative GPCR-like proteins (OsGPCRLPs) (LOC_Os06g09930.1, LOC_Os04g36630.1, and LOC_Os01g54784.1) in the rice proteome using a stringent bioinformatics workflow. The identified OsGPCRLPs exhibited a canonical GPCR ‘type I’ 7TM topology, patterns, and biologically significant sites for membrane anchorage and desensitization. Cluster-based interactome mapping revealed that the identified proteins interact with the G protein α subunit which is a characteristic feature of GPCRs. Computational results showing the interaction of identified GPCR-like proteins with G protein α subunit and its further validation by the membrane yeast-two-hybrid assay strongly suggest the presence of GPCR-like 7TM proteins in the rice proteome. The absence of a regulator of G protein signaling (RGS) box in the C- terminal domain, and the presence of signature motifs of canonical GPCR in the identified OsGPCRLPs strongly suggest that the rice proteome contains GPCR-like proteins that might be involved in signal transduction.

MYB transcription factor is one of the largest families in plants. There are more and more studies on plants responding to abiotic stress through MYB transcription factors, but the mechanism of some family members responding to salt stress is unclear. In this study, physiological and transcriptome techniques were used to analyze the effects of the R2R3-MYB transcription factor AtMYB72 on the growth and development, physiological function, and key gene response of Arabidopsis thaliana. Phenotypic observation showed that the damage of overexpression strain was more serious than that of Col-0 after salt treatment, while the mutant strain showed less salt injury symptoms. Under salt stress, the decrease of chlorophyll content, the degree of photoinhibition of photosystem II (PSII) and photosystem I (PSI) and the degree of oxidative damage of overexpressed lines were significantly higher than those of Col-0. Transcriptome data showed that the number of differentially expressed genes (DEGs) induced by salt stress in overexpressed lines was significantly higher than that in Col-0. GO enrichment analysis showed that the response of AtMYB72 to salt stress was mainly by affecting gene expression in cell wall ectoplast, photosystem I and photosystem II, and other biological processes related to photosynthesis. Compared with Col-0, the overexpression of AtMYB72 under salt stress further inhibited the synthesis of chlorophyll a (Chla) and down-regulated most of the genes related to photosynthesis, which made the photosynthetic system more sensitive to salt stress. AtMYB72 also caused the outbreak of reactive oxygen species and the accumulation of malondialdehyde under salt stress, which decreased the activity and gene expression of key enzymes in SOD, POD, and AsA-GSH cycle, thus destroying the ability of antioxidant system to maintain redox balance. AtMYB72 negatively regulates the accumulation of osmotic regulatory substances such as soluble sugar (SS) and soluble protein (SP) in A. thaliana leaves under salt stress, which enhances the sensitivity of Arabidopsis leaves to salt. To sum up, MYB72 negatively regulates the salt tolerance of A. thaliana by destroying the light energy capture, electron transport, and antioxidant capacity of Arabidopsis.

The potential of rhizobacteria with plant growth promoting (PGP) traits in alleviating abiotic stresses, especially drought, is significant. However, their exploitation in the semi-arid regions of Ethiopian soils remains largely unexplored. This research aimed to isolate and evaluate the PGP potential of bacterial isolates collected from groundnut cultivation areas in Ethiopia. Multiple traits were assessed, including phosphate solubilization, indole-3-acetic acid (IAA) production, ammonia production, salt and heavy metal tolerance, drought tolerance, enzyme activities, hydrogen cyanide production, antibiotic resistance, and antagonistic activity against fungal pathogens. The identification of potent isolates was carried out using MALDI-TOF MS. Out of the 82 isolates, 63 were gram-negative and 19 were gram-positive. Among them, 19 isolates exhibited phosphate solubilization, with AAURB 34 demonstrating the highest efficiency, followed by AURB 12. Fifty-six isolates produce IAA in varying amounts and all isolates produce ammonia with AAURB12, AAURB19, and AAURB34 displaying strong production. Most isolates demonstrated tolerance to temperatures up to 40°C and salt concentrations up to 3%. Notably, AAURB12 and AAURB34 exhibited remarkable drought tolerance at an osmotic potential of −2.70 Mpa. When subjected to levels above 40%, the tested isolates moderately produced lytic enzymes and hydrogen cyanide. The isolates displayed resistance to antibiotics, except gentamicin, and all isolates demonstrated resistance to zinc, with 81–91% showing resistance to other heavy metals. AAURB34 and AAURB12 exhibited suppression against fungal pathogens, with percent inhibition of 38% and 46%, respectively. Using MALDI-TOF MS, the promising PGP isolates were identified as Bacillus megaterium, Bacillus pumilus, and Enterobacter asburiae. This study provides valuable insights into the potential of rhizobacteria as PGP agents for mitigating abiotic stresses and contribute to the understanding of sustainable agricultural practices in Ethiopia and similar regions facing comparable challenges.

Cyanobacterium Nostoc commune is a filamentous terrestrial prokaryotic organism widely distributed, which suggest its high adaptive potential to environmental or abiotic stress. Physiological parameters and proteomic analysis were performed in two accession of N. commune with the aim to elucidate the differences of physiological trails between distant geotypes, namely Antarctic (AN) and central European (CE). The result obtained clearly showed that the AN geotype demonstrates elevated levels of total phenols, flavonoids, carotenoids, and phycobiliproteins, indicative of its adaptation to environmental stress as referred by comparison to CE sample. Additionally, we employed LC-MS analysis to investigate the proteomes of N. commune from AN and CE geotypes. In total, 1147 proteins were identified, among which 646 proteins expressed significant (up-regulation) changes in both accessions. In the AN geotype, 83 exclusive proteins were identified compared to 25 in the CE geotype. Functional classification of the significant proteins showed a large fraction involved in photosynthesis, amino acid metabolism, carbohydrate metabolism and protein biosynthesis. Further analysis revealed some defense-related proteins such as, superoxide dismutase (SOD) and glutathione reductase, which are rather explicitly expressed in the AN N. commune. The last two proteins suggest a more stressful condition in AN N. commune. In summary, our findings highlight biochemical processes that safeguard the AN geotype of N. commune from extreme environmental challenges, not recorded in CE accession, probably due to less stressful environment in Europe. This study brings the first ever proteomic analysis of N. commune, emphasizing the need for additional investigations into the climate adaptation of this species with rather plastic genome.

Extracellular ATP (eATP) orchestrates vital processes in plants, akin to its role in animals. P2K1 is a crucial receptor mediating eATP effects. Immunoprecipitation tandem mass spectrometry data highlighted FERONIA’s significant interaction with P2K1, driving us to explore its role in eATP signaling. Here, we investigated putative P2K1-interactor, FERONIA, which is a versatile receptor kinase pivotal in growth and stress responses. We employed a FERONIA loss-of-function mutant, fer-4, to dissect its effects on eATP signaling. Interestingly, fer-4 showed distinct calcium responses compared to wild type, while eATP-responsive genes were constitutively upregulated in fer-4. Additionally, fer-4 displayed insensitivity to eATP-regulated root growth and reduced cell wall accumulation. Together, these results uncover a role for FERONIA in regulating eATP signaling. Overall, our study deepens our understanding of eATP signaling, revealing the intricate interplay between P2K1 and FERONIA impacting the interface between growth and defense.

Soil toxicity is a major environmental issue that leads to numerous harmful effects on plants and human beings. Every year a huge amount of Pb is dumped into the environment either from natural sources or anthropogenically. Being a heavy metal it is highly toxic and non-biodegradable but remains in the environment for a long time. It is considered a neurotoxic and exerts harmful effects on living beings. In the present review article, investigators have emphasized the side effects of Pb on the plants. Further, the authors have focused on the various sources of Pb in the environment. Investigators have emphasized the various responses including molecular, biochemical, and morphological of plants to the toxic levels of Pb. Further emphasis was given to the effect of elevated levels of Pb on the microbial population in the rhizospheres. Further, emphasized the various remediation strategies for the Pb removal from the soil and water sources.

In the decades since their discovery in plants in the mid-to-late 1900s, melatonin (N-acetyl-5-methoxytryptamine) and serotonin (5-methoxytryptamine) have been established as their own class of phytohormone and have become popular targets for examination and study as stress ameliorating compounds. The indoleamines play roles across the plant life cycle from reproduction to morphogenesis and plant environmental perception. There is growing interest in harnessing the power of these plant neurotransmitters in applied and agricultural settings, particularly as we face increasingly volatile climates for food production; however, there is still a lot to learn about the mechanisms of indoleamine action in plants. A recent explosion of interest in these compounds has led to exponential growth in the field of melatonin research in particular. This concept paper aims to summarize the current status of indoleamine research and highlight some emerging trends.

Potassium (K+) plays a role in enzyme activation, membrane transport, and osmotic regulation processes. An increase in potassium content can significantly improve the elasticity and combustibility of tobacco and reduce the content of harmful substances. Here, we report that the expression analysis of Nt GF14e, a 14-3-3 gene, increased markedly after low-potassium treatment (LK). Then, chlorophyll content, POD activity and potassium content, were significantly increased in overexpression of Nt GF14e transgenic tobacco lines compared with those in the wild type plants. The net K+ efflux rates were severely lower in the transgenic plants than in the wild type under LK stress. Furthermore, transcriptome analysis identified 5708 upregulated genes and 2787 downregulated genes between Nt GF14e overexpressing transgenic tobacco plants. The expression levels of some potassium-related genes were increased, such as CBL-interacting protein kinase 2 (CIPK2), Nt CIPK23, Nt CIPK25, H+-ATPase isoform 2 a (AHA2a), Nt AHA4a, Stelar K+ outward rectifier 1(SKOR1), and high affinity K+ transporter 5 (HAK5). The result of yeast two-hybrid and luciferase complementation imaging experiments suggested Nt GF14e could interact with CIPK2. Overall, these findings indicate that NtGF14e plays a vital roles in improving tobacco LK tolerance and enhancing potassium nutrition signaling pathways in tobacco plants.

Earthworms’ coelomic fluid (CF) has been discovered to possess properties that promote plant development. In particular, the earthworm’s coelomic fluid-associated bacteria (CFB) are the primary factor influencing the plants’ response. To investigate this, we used bacteria isolated from the CF and selected based on different plant growth-promoting traits, in a mesocosm ecosystem that includes plants. This experiment aimed to assess their impact on the metabolism of plants growing under abiotic stress environments (alkaline soil and nitrogen (N), phosphate (P), and potassium (K) deficit) and compare the lipid profiles of plants under the various treatments. We used seven different bacterial species isolated from the CF of Aporrectodea molleri and as a plant model Zea mays L. For the metabolomic analysis method, we used gas chromatography-mass spectrometry lipidomic. After observing the metabolomic profiles, we found that a few molecular pathways are involved in how plants react to bacterial biostimulants. The bacterial isolates belonging to Pantoea vagans, Pseudomonas aeruginosa, Bacillus paramycoides, and Bacillus thuringiensis have led to a significant increase in synthesizing several metabolites belonging to various chemical categories. Contrary to predictions, abiotic stress did not cause a drop in the composition and concentration of lipids in plants treated with the CFB, demonstrating the rigidity of the protective mechanisms. The statistical analysis based on the Pearson method revealed a positive significant correlation between plant growth parameters (length of the aerial part, surface of the leaves, and biomass) and some metabolites belonging to fatty acids, carboxylic acids, benzene derivatives, and alkanes. Moreover, the standard metabolic components of all treatments in much higher concentrations during bacterial treatments than the control treatment suggests that the bacteria have stimulated the overexpression of these metabolic components. According to these results, we could assume that plants treated with CFB exhibit an adaptability of abiotic stress defense mechanisms, which may be attributed to the upregulation of genes involved in lipid biosynthesis pathways.

Foeniculum vulgare Mill. commonly known as fennel, is a globally recognized aromatic medicinal plant and culinary herb with widespread popularity due to its antimicrobial, antioxidant, carminative, and diuretic properties, among others. Although the phenotypic effects of salinity stress have been previously explored in fennel, the molecular mechanisms underlying responses to elevated salinity in this plant remain elusive. MicroRNAs (miRNAs) are tiny, endogenous, and extensively conserved non-coding RNAs (ncRNAs) typically ranging from 20 to 24 nucleotides (nt) in length that play a major role in a myriad of biological functions. In fact, a number of miRNAs have been extensively associated with responses to abiotic stress in plants. Consequently, employing computational methodologies and rigorous filtering criteria, 40 putative miRNAs belonging to 25 different families were characterized from fennel in this study. Subsequently, employing the psRNATarget tool, a total of 67 different candidate target transcripts for the characterized fennel miRNAs were predicted. Additionally, the expression patterns of six selected fennel miRNAs (i.e. fvu-miR156a, fvu-miR162a-3p, fvu-miR166a-3p, fvu-miR167a-5p, fvu-miR171a-3p, and fvu-miR408-3p) were analyzed under salinity stress conditions via qPCR. This article holds notable significance as it identifies not only 40 putative miRNAs in fennel, a non-model plant, but also pioneers the analysis of their expression under salinity stress conditions.

Tea plantations in Karst regions suffer from the serious effects of frequent temporary karst droughts, leading to a decline in tea production and quality in the region. The close relationship between growth and electrical parameters of plants, including physiological capacitance, resistance and impedance, can be used to accurately monitor their plant water status online, quickly, accurately, timely and nondestructively. In this study, three tea tree cultivars of Zhonghuang No.2 (ZH), Wuniuzao (WNZ), and Longjing 43 (LJ) with different levels of drought resistance were selected as experimental materials, and experiments were carried out under controlled conditions according to control (soil water content of 40–45%, D0), (keeping D0 no watering to 5 days, D5), (keeping D0 no watering to 10 days, D10), (the first day after D10 is rehydrated to D0 is regarded as R1) and (the fifth day after D10 rehydration to D0 is regarded as R5), to determine intracellular water metabolism and nutrient translocation characteristics based on intrinsic electrical parameters. The photosynthetic characteristics and chlorophyll fluorescence parameters were also determined to investigate the response of water metabolism to simulated karst drought in the three tea tree cultivars. The results indicated that the water metabolism patterns responded to environmental water changes with a medium water-holding capacity, medium water transport rate, and low water-use efficiency, and the nutrient patterns in those tea tree varieties demonstrated with a high nutrient flux per unit area, low nutrient transfer rate, and high nutrient transport capacity. After rehydration, only the electrical characteristics of WNZ returned to the D0 levels, but the net photosynthetic rate of all varieties returned to or even exceeded the D0 levels. The chlorophyll fluorescence parameters could not be used to characterize the recoverability of metabolism in tea trees. The electrical characteristics quickly reflected the response of the water metabolism in plants to environmental changes, and the fusion of electrical characteristics and photosynthetic characteristics was able to more quickly, accurately, and comprehensively reflect the response of water metabolism to temporary karst drought.
