Abstract There is need for advancing minimally invasive diagnostic techniques for sex differentiation at early life-stage classes in sea turtles. The objectives of this study were to determine whether there are sex-specific effects on the metabolome of the loggerhead sea turtle (Caretta caretta) and to identify potential biomarkers for sex classification at early life-stage classes (50 post-hatchling and 50 juvenile). Comprehensive analyses including plasma chemistry (n = 100) and untargeted (n = 48) metabolomic analyses were performed. Unlike with plasma chemistry analytes, there were significant differences in the metabolomic profiles between sexes. Nine metabolites differed significantly between sexes, with several metabolites deserving greater attention as potential biomarkers: C22H32O6 and pantothenic acid was higher in females than in males, and carboxylic acid ester (C10H16O5), 1-methylhistidine and C7H14N2O7 was higher in males than females. These differences remained constant both across and within the same life-stage class. This work identified a discernible effect on the metabolic profile based on sex of loggerhead sea turtles and it suggests that these metabolites and/or their ratios could serve as potential biomarker candidates for sex classification in this species.

Aerogel scaffolds are nanostructured materials with beneficial properties for tissue engineering applications. The tracing of the state of the aerogels after their implantation is challenging due to their variable biodegradation rate and the lack of suitable strategies capable of in vivo monitoring the scaffolds. Upconversion nanoparticles (UCNPs) have emerged as advanced tools for in vitro bioimaging because of their fluorescence properties. In this work, highly fluorescent UCNPs were loaded into aerogels to obtain theranostic implants for tissue engineering and bioimaging applications. 3D-printed alginate-hydroxyapatite aerogels labeled with UCNPs were manufactured by 3D-printing and supercritical CO2 drying to generate personalize-to-patient aerogels. The physicochemical performance of the resulting structures was evaluated by printing fidelity measurements, nitrogen adsorption-desorption analysis, and different microscopies (confocal, transmission and scanning electron microscopies). Stability of the aerogels in terms of physicochemical properties was also tested after 3 years of storage. Biocompatibility was evaluated in vitro by different cell and hemocompatibility assays, in ovo and in vivo by safety and bioimaging studies using different murine models. Cytokines profile, tissue index and histological evaluations of the main organs unveiled an in vivo downregulation of the inflammation after implantation of the scaffolds. UCNPs-decorated aerogels were first-time manufactured and long-term traceable by fluorescence-based bioimaging until 3 weeks post-implantation, thereby endorsing their suitability as tissue engineering and theranostic nanodevices (i.e. bifunctional implants).

Abstract Background There are many gaps in our understanding of the mechanisms involved in ovarian follicular development in cattle, particularly regarding follicular deviation, acquisition of ovulatory capacity, and preovulatory changes. Molecular evaluations of ovarian follicular cells during follicular development in cattle, especially serial transcriptomic analyses across key growth phases, have not been reported. This study aims to address this gap by analyzing gene expression using RNA-seq in granulosa and antral cells recovered from ovarian follicular fluid during critical phases of ovarian follicular development in Holstein cows. Results Integrated analysis of gene ontology (GO), gene set enrichment (GSEA), protein–protein interaction (PPI), and gene topology identified that differentially expressed genes (DEGs) in the largest ovarian follicles at deviation (Dev) were primarily involved in FSH-negative feedback, steroidogenesis, cell proliferation, apoptosis, and the prevention of early follicle rupture. In contrast, DEGs in the second largest follicles (DevF2) were mainly related to loss of cell viability, apoptosis, and immune cell invasion. In the dominant (PostDev) and preovulatory (PreOv) follicles, DEGs were associated with vascular changes and inflammatory responses. Conclusions The transcriptome of ovarian follicular fluid cells had a predominance of granulosa cells in the dominant follicle at deviation, with upregulation of genes involved in cell viability, steroidogenesis, and apoptosis prevention, whereas in the non-selected follicle there was upregulation of cell death-related transcripts. Immune cell transcripts increased significantly after deviation, particularly in preovulatory follicles, indicating strong intrafollicular chemotactic activity. We inferred that immune cell invasion occurred despite an intact basal lamina, contributing to follicular maturation. Graphical Abstract

Invasive alien species are an important component of global change, threatening biodiversity, ecosystem functioning, economy, and human health. The number of alien species that attain the invasive status has experienced an exponential increase in recent years, leading some government agencies and stakeholders to allocate substantial resources to early detection, control, mitigation, and eradication programs. To develop effective nature conservation strategies, it is crucial to understand the invasive status of alien species and to identify priority species for management at spatial scales with a biogeographical basis. Despite significant progress in producing lists of alien species at the country level, a standard methodology for species assessment within ecological regions (i.e., regions with similar environmental or biogeographical characteristics) is still lacking. Here, we develop a systematic approach to determine invasion status and to prioritize invasive alien plant species within an ecoregion. We apply this approach in the Cantabrian Mixed Forests ecoregion, which encompasses biogeographically related areas from N Portugal, NW Spain, and SW France, and is strongly affected by plant invasions. By combining scientific evidence with expert opinion on the ecological characteristics of alien plants, we identified 175 invasive plant species in the study ecoregion, of which 37 cause massive environmental and/or socio-economic impacts. For each species, we provide comprehensive information and recommendations for scientists, land managers, policy makers, and other stakeholders under a biogeographical basis. This information includes species characteristics, invasion status/level, population trends, geographic locations and range size, local abundance, environmental and socio-economic impacts, and invaded habitats. We also accounted for administrative divisions within the ecoregion to facilitate the use of such evaluations in local-scale management and conservation plans. Our framework may be applied to any ecoregion worldwide, enhancing the assessment and management of invasive species pools within biogeographically meaningful regions.

Abstract Background Determining the postmortem interval (PMI) accurately remains a significant challenge in forensic sciences, especially for intervals greater than 5 years (late PMI). Traditional methods often fail due to the extensive degradation of soft tissues, necessitating reliance on bone material examinations. The precision in estimating PMIs diminishes with time, particularly for intervals between 1 and 5 years, dropping to about 50% accuracy. This study aims to address this issue by identifying key protein biomarkers through proteomics and machine learning, ultimately enhancing the accuracy of PMI estimation for intervals exceeding 15 years. Methods Proteomic analysis was conducted using LC–MS/MS on skeletal remains, specifically focusing on the tibia and ribs. Protein identification was performed using two strategies: a tryptic-specific search and a semitryptic search, the latter being particularly beneficial in cases of natural protein degradation. The Random Forest algorithm was used to model protein abundance data, enabling the prediction of PMI. A thorough screening process, combining importance scores and SHAP values, was employed to identify the most informative proteins for model’s training and accuracy. Results A minimal set of three biomarkers—K1C13, PGS1, and CO3A1—was identified, significantly improving the prediction accuracy between PMIs of 15 and 20 years. The model, based on protein abundance data from semitryptic peptides in tibia samples, achieved sustained 100% accuracy across 100 iterations. In contrast, non-supervised methods like PCA and MCA did not yield comparable results. Additionally, the use of semitryptic peptides outperformed tryptic peptides, particularly in tibia proteomes, suggesting their potential reliability in late PMI prediction. Conclusions Despite limitations such as sample size and PMI range, this study demonstrates the feasibility of combining proteomics and machine learning for accurate late PMI predictions. Future research should focus on broader PMI ranges and various bone types to further refine and standardize forensic proteomic methodologies for PMI estimation.

During a parasitological survey of tapeworms from Myliobatis goodei Garman, 1885 (Myliobatiformes: Myliobatidae) in coastal waters off Argentina in the Southwestern Atlantic, a new rhinebothriidean cestode species, Anindobothrium danielae sp. nov., is described using morphological and molecular techniques. This species differs from its congeners by a particular combination of features, including the configuration of the bothridia, the number of marginal loculi, and the number and distribution of testes. Additionally, Anindobothrium myliobatidis comb. nov. is proposed based on several morphological traits, including the presence of stalked bothridia with marginal loculi and an apical sucker, euapolytic strobila, and postvaginal testes. The diagnosis of the genus Anindobothrium Marques, Brooks & Lasso, 2001 is amended to include the features exhibited by these two species; two species subsets are suggested based on the configuration of the bothridia. The presence of A. danielae sp. nov. and A. myliobatidis comb. nov. in the studied area not only increases the number of cestodes in M. goodei here from eight to ten but also represents the first report of a rhinebothriidean cestode parasitizing stingrays of the family Myliobatidae in the Southwestern Atlantic.

Abstract CRISPR-based genome editing of pseudogene-associated disorders, such as p47 phox -deficient chronic granulomatous disease (p47 CGD), is challenged by chromosomal rearrangements due to presence of multiple targets. We report that interactions between highly homologous sequences that are localized on the same chromosome contribute substantially to post-editing chromosomal rearrangements. We successfully employed editing approaches at the NCF1 gene and its pseudogenes, NCF1B and NCF1C, in a human cell line model of p47 CGD and in patient-derived human hematopoietic stem and progenitor cells. Upon genetic engineering, a droplet digital PCR-based method identified cells with altered copy numbers, spanning megabases from the edited loci. We attributed the high aberration frequency to the interaction between repetitive sequences and their predisposition to recombination events. Our findings emphasize the need for careful evaluation of the target-specific genomic context, such as the presence of homologous regions, whose instability can constitute a risk factor for chromosomal rearrangements upon genome editing.

Abstract Peptides are promising drug development frameworks that have been hindered by intrinsic undesired properties including hemolytic activity. We aim to get a better insight into the chemical space of hemolytic peptides using a novel approach based on network science and data mining. Metadata networks (METNs) were useful to characterize and find general patterns associated with hemolytic peptides, whereas Half-Space Proximal Networks (HSPNs), represented the hemolytic peptide space. The best candidate HSPNs were used to extract various subsets of hemolytic peptides (scaffolds) considering network centrality and peptide similarity. These scaffolds have been proved to be useful in developing robust similarity-based model classifiers. Finally, using an alignment-free approach, we reported 47 putative hemolytic motifs, which can be used as toxic signatures when developing novel peptide-based drugs. We provided evidence that the number of hemolytic motifs in a sequence might be related to the likelihood of being hemolytic.

Abstract We describe the geographical variation in tree species composition across Amazonian forests and show how environmental conditions are associated with species turnover. Our analyses are based on 2023 forest inventory plots (1 ha) that provide abundance data for a total of 5188 tree species. Within-plot species composition reflected both local environmental conditions (especially soil nutrients and hydrology) and geographical regions. A broader-scale view of species turnover was obtained by interpolating the relative tree species abundances over Amazonia into 47,441 0.1-degree grid cells. Two main dimensions of spatial change in tree species composition were identified. The first was a gradient between western Amazonia at the Andean forelands (with young geology and relatively nutrient-rich soils) and central–eastern Amazonia associated with the Guiana and Brazilian Shields (with more ancient geology and poor soils). The second gradient was between the wet forests of the northwest and the drier forests in southern Amazonia. Isolines linking cells of similar composition crossed major Amazonian rivers, suggesting that tree species distributions are not limited by rivers. Even though some areas of relatively sharp species turnover were identified, mostly the tree species composition changed gradually over large extents, which does not support delimiting clear discrete biogeographic regions within Amazonia.

Abstract Bioactive metabolites play a crucial role in shaping interactions among diverse organisms. In this study, we identified cyclo(Pro-Tyr), a metabolite produced by Bacillus velezensis, as a potent inhibitor of Botrytis cinerea and Caenorhabditis elegans, two potential cohabitant eukaryotic organisms. Based on our investigation, cyclo(Pro-Tyr) disrupts plasma membrane polarization, induces oxidative stress and increases membrane fluidity, which compromises fungal membrane integrity. These cytological and physiological changes induced by cyclo(Pro-Tyr) may be triggered by the destabilization of membrane microdomains containing the [H+]ATPase Pma1. In response to cyclo(Pro-Tyr) stress, fungal cells activate a transcriptomic and metabolomic response, which primarily involves lipid metabolism and Reactive Oxygen Species (ROS) detoxification, to mitigate membrane damage. This similar response occurs in the nematode C. elegans, indicating that cyclo(Pro-Tyr) targets eukaryotic cellular membranes.

Dirofilaria immitis is a mosquito-borne nematode of dogs, other carnivores and, occasionally, humans. Globally, D. immitis infection (which causes heartworm) is typically more prevalent in tropical than temperate regions. In this study, the seroprevalence of D. immitis was determined from a sample of 335 non-stray dogs from four municipalities, two each from the states of Puebla and Guerrero in Mexico, using polyclonal antibodies to detect serum antigens using an enzyme-linked immunosorbent assay (ELISA). The accuracy of the assay was compared with the modified Knott’s test. The polyclonal antibody used in the direct ELISA had a high sensitivity (100%) with variable specificity (98.2–98.8%) in the municipalities of Puebla and Guerrero. The area under the curve for the four municipalities was 1.0, indicating a high accuracy test, with a cut-off value ranging from 0.45 to 0.50. The overall prevalence of D. immitis infection was 17.56% (59 out of 335). The highest prevalence was in Acapulco (24.78%), followed by Chilpancingo (20.93%), Tecamachalco (10.81%) and Quecholac (8.06%). The highest percentage of positive samples was detected in tropical regions (23.12%) and the lowest in temperate regions (9.56%). This study demonstrates that polyclonal anti-D. immitis antibodies can successfully diagnose heartworm-infected dogs and be used to monitor prevalence effectively and develop prevention strategies against Dirofilaria infection.

The growing interest in microalgae is driven by their potential in various bioindustries, such as biofuel production, bioremediation, and the generation of high-value biomolecules. This paper aims to systematically review the state of research on bioprospecting microalgae, their applications, and recognize trends. This study employs an exploratory and descriptive research approach, using bibliometric methods to analyze scientific production and identify emerging trends in bioprospecting microalgae research. The analysis reveals exponential publication growth, with multidisciplinary sources indicating a strong applied focus. Leading countries in this research field benefit from clear technology transfer policies, and the prevalent terms “production” and “biomass” underscore the industrial relevance. Key research areas include biofuels and bioremediation, with a combined emphasis that is often studied in cultivation and biomass production. Bioactive compounds derived from microalgae are a current trend for industrial, medical, and food applications. Although the potential for CO2 capture is acknowledged, direct studies are limited. This systematic review provides a comprehensive overview of current trends and identifies opportunities and challenges in microalgae research, highlighting its significance for sustainable development and industrial applications.

This study investigated the potential of chicken byproduct hydrolysates (CBH) characterized by a mixture of low-molecular-weight peptides (B. pinguin, BK: B. karatas, BRO: bromelain) on the histological and histopathological analysis of the liver and kidney in an MS-induced murine model. Methods: Thirty adult male Wistar rats were randomly assigned to six groups (n = 5): (1) standard diet (STD); (2) MS with a hypercaloric diet (MS + HC); (3) CBH-BP (200 mg/kg of body weight); (4) CBH-BK (200 mg/kg of body weight); (5) CBH-BRO (200 mg/kg of body weight); (6) carnosine (CAR) 50 mg/kg of body weight. Liver and kidney samples were processed by conventional hematoxylin and eosin (H&E) histological techniques, Masson’s trichrome stain (MTS), and the periodic acid–Schiff (PAS) histochemical method. A scoring scale was used for the histopathological evaluation with scores ranging from 0 (normal tissue) to 4 (severe damage). Results: CBHs demonstrated a significant therapeutic effect (p < 0.05) on hepatic and renal morphological alterations induced by MS. Hepatic scores for lipid inclusions, vascular congestion, and cellular alteration were all reduced to below two. Similarly, renal scores for tubular degeneration, vascular congestion, and dilation of Bowman’s space were also decreased to less than two. The therapeutic efficacy of CBHs was comparable to that of the positive control, CAR (β-alanyl-L-histidine). Conclusions: CBH-BP, CBH-BK, and CBH-BRO treatments reduced morphological alterations observed in liver and kidney tissues, which is relevant since from a histological and histopathological point of view, it allows us to understand at the cellular and tissue level the effects that these treatments can have on a living organism, indicating a potential to improve organ health in people with MS.

Prostate cancer (PCa) is a common and deadly disease in men. It is often diagnosed at advanced stages, at which point patients are treated mainly with docetaxel (DTX), which is effective but limited by resistance and side effects. Overactivation of the transcription factors NF-κB and STAT-3 plays a critical role in the development, progression, and chemoresistance of PCa. In this regard, the blockade of NF-κB with pentoxifylline (PTX) or STAT-3 with Stattic (STT) is known to increase the sensitivity of tumor cells to chemotherapy in both in vitro and in vivo models. We investigated whether simultaneous blockade with PTX and STT increases the efficacy of the DTX treatment in inducing apoptosis in metastatic castration-resistant PCa DU-145 cells. Our results showed that the combination of PTX + STT led to higher levels of apoptosis, regardless of whether or not DTX was present in the treatment. Determining caspases and ΔΨm indicates that the intrinsic caspase pathway of apoptosis is principally favored. In addition, this combination inhibited proliferation and colony formation and arrested the cell cycle in the G1 phase. These results indicate that the combination of the PTX + STAT-3 inhibitor could potentiate DTX effectively, opening the possibility of effective treatments in PCa.

High-density olive groves, despite their interesting production potential, have several limitations, including their high fruit load and irrigation requirements. This study aimed to evaluate the effects of fruit load and deficit irrigation on oil yield, fruit quality, and olive chemical composition in a high-density olive grove (cv Sikitita). Our main hypothesis was that primary metabolism, as influenced by crop load and stress, could modify the accumulation of different phenolic classes. Different fruit loads were generated through flower thinning (66%, 50%, 33%, 0%), and two deficit irrigation treatments (−60%, −75%) were compared to the well-watered control (920 m3/ha). Thinning treatments had a limited effect on oil yield; on the other hand, deficit irrigation caused considerably less oil accumulation in the fruit on all sampling dates. Thinning 66% and deficit irrigation 75% were considered with the control for untargeted metabolomic analysis, including three sampling dates. A total of 233 distinct phenolic compounds were annotated. Multivariate HCA results indicated that harvest time had an impact on the phenolic profile of olive fruits, obtaining two separated clusters that grouped t1 and t2 together and apart from t3, which clustered independently. Regarding agronomic techniques, they played a differential role in the phenolic profile (supervised OPLS-DA). Fruit load mostly affected flavonoid glycosides. In contrast, the phenolic response to deficit irrigation was more heterogeneous, with phenolic acids (35%), flavonoids (25%), LMW, and other phenols (25%).

Abstract In several carcinomas, including hepatocellular carcinoma, it has been demonstrated that cancer stem cells (CSCs) have enhanced invasiveness and therapy resistance compared to differentiated cancer cells. Mathematical-computational tools could be valuable for integrating experimental results and understanding the phenotypic plasticity mechanisms for CSCs emergence. Based on the literature review, we constructed a Boolean model that recovers eight stable states (attractors) corresponding to the gene expression profile of hepatocytes and mesenchymal cells in senescent, quiescent, proliferative, and stem-like states. The epigenetic landscape associated with the regulatory network was analyzed. We observed that the loss of p53, p16, RB, or the constitutive activation of β-catenin and YAP1 increases the robustness of the proliferative stem-like phenotypes. Additionally, we found that p53 inactivation facilitates the transition of proliferative hepatocytes into stem-like mesenchymal phenotype. Thus, phenotypic plasticity may be altered, and stem-like phenotypes related to CSCs may be easier to attain following the mutation acquisition.

Cacalol (C), a sesquiterpene isolated from Psacalium decompositum, has demonstrated anti-inflammatory and antioxidant activities. Its cytotoxic, antiproliferative, and pro-apoptotic effects have been previously shown in an in vitro breast cancer model. A derivative, cacalol acetate (CA), shows potential in regulating these processes, which has not been previously reported. This study focused on an in vitro cervical cancer model, assessing CA’s antiproliferative, pro-apoptotic, cytostatic, and anti-migratory activities using the HeLa cell line. The natural anticancer agent indole-3-carbinol (I3C) was used as a control for comparison. CA demonstrated significant antitumor activities, including inhibiting cell growth, inducing apoptosis, arresting cells in the G2 phase of the cell cycle, and inhibiting cell migration. These effects were notably greater compared to I3C. I3C, while following a similar trend, did not induce Cas-3 expression, suggesting a different apoptotic pathway. Neither CA nor I3C increased p62 and LC3B levels, indicating they do not stimulate autophagy marker expression. Both compounds inhibited HeLa cell migration and induced cell cycle arrest. Despite both holding promise as anticancer agents for cervical cancer, CA’s lower cytotoxicity and stronger regulation of tumor phenotypes make it a more promising agent compared to I3C.

Nematode surveys in natural environments in the Iberian Peninsula detected three unidentified Xenocriconemella populations that closely resembled the X. macrodora-species complex, but utilization of integrative taxonomy confirmed that they comprised a new taxon described in this paper as X. andreae sp. nov. Only females were detected in the new species (considered parthenogenetic) and delineated with a bare body (274–353 µm); lip region with two annuli, continuous with body delineation; second lip annulus enclosed by the first one. Flexible and thin stylet (88.0–99.0 µm), representing 30.4–47.8% of total body length. The excretory pore is positioned 2–3 annuli posterior to the level of stylet knobs, at 101.5 (87–107) µm from the lip region. Female genital tract: monodelphic, prodelphic, large, and representing 34.4–52.4% of the body length; vagina slightly ventrally curved. The anus is located at (6–9) annuli from the rear end. Tail short, conoid, and blunt round terminus. Ribosomal and mitochondrial markers (D2-D3 expansion domains of 28S, ITS, partial 18S rRNA, and COI), as well as molecular phylogenetic analyses of sequences, confirmed this new taxon, and it was clearly delineated from X. macrodora and species within the species complex (X. costaricense, X. iberica, X. paraiberica, and X. pradense).

The intestinal microbiota and the human body are in a permanent interaction. There is a symbiotic relationship in which the microbiota plays a vitally important role in the performance of numerous functions, including digestion, metabolism, the development of lymphoid tissue, defensive functions, and other processes. It is a true metabolic organ essential for life and has potential involvement in various pathological states, including cancer and pathologies other than those of a digestive nature. A growing topic of great interest for its implications is the relationship between the microbiota and cancer. Dysbiosis plays a role in oncogenesis, tumor progression, and even the response to cancer treatment. The effect of the microbiota on tumor development goes beyond a local effect having a systemic effect. Another aspect of great interest regarding the intestinal microbiota is its relationship with drugs, modifying their activity. There is increasing evidence that the microbiota influences the therapeutic activity and side effects of antineoplastic drugs and also modulates the response of several tumors to antineoplastic therapy through immunological circuits. These data suggest the manipulation of the microbiota as a possible adjuvant to improve oncological treatment. Is it possible to manipulate the microbiota for therapeutic purposes?

The known distribution of Bothriechis aurifer (Salvin, 1860) is outdated, which impedes an accurate evaluation of its conservation status. Here, we clarify the locality-level distribution of B. aurifer and announce a 101-km range extension. Based on this and other data, we recommend its recategorization as Endangered under the International Union for Conservation of Nature’s Red List of Threatened Species, and its addition to Appendix I of the Convention on International Trade in Endangered Species of Wild Fauna and Flora. ABSTRACTHeterogeneous nuclear ribonucleoproteins (hnRNPs) are multifunctional proteins with integral roles in RNA metabolism and the regulation of alternative splicing. These proteins typically contain prion-like domains of low complexity (PrLDs or LCDs) that govern their assembly into either functional or pathological amyloid fibrils. To date, over 60 mutations targeting the LCDs of hnRNPs have been identified and associated with a spectrum of neurodegenerative diseases including amyotrophic lateral sclerosis (ALS), frontotemporal dementia (FTD), and Alzheimer’s disease (AD). The cryo-EM structures of pathological and functional fibrils formed by different hnRNPs have been recently elucidated, including those of hnRNPA1, hnRNPA2, hnRNPDL-2, TDP-43, and FUS. In this review, we discuss the structural features of these amyloid assemblies, placing particular emphasis on scrutinizing the impact of prevalent disease-associated mutations mapping within their LCDs. By performing systematic energy calculations, we reveal a prevailing trend of destabilizing effects induced by these mutations in the amyloid structure, challenging the traditionally assumed correlation between pathogenicity and amyloidogenic propensity. Understanding the molecular basis of this discrepancy might provide insights for developing targeted therapeutic strategies to combat hnRNP-associated diseases.

CULLIN4-RING (CRL4) E3 ubiquitin ligases use the substrate receptor module CONSTITUTIVELY PHOTOMORPHOGENIC1 (COP10)-DDB1-DET1-DDA1 (CDDD) to target Arabidopsis ABA receptor PYL8, acting as negative regulators of ABA responses. Conversely, ABA treatment attenuates PYL8 receptor degradation, although the molecular mechanism remained elusive. Here, we show that ABA promotes the disruption of CRL4-CDDD complexes, leading to PYL8 stabilization. ABA-mediated CRL4-CDDD dissociation likely involves an altered association between DDA1-containing complexes and the COP9 signalosome (CSN), a master regulator of the assembly of cullin-based E3 ligases, including CRL4-CDDD. Indeed, treatment with CSN inhibitor CSN5i-3 suppresses the ABA effect on CRL4-CDDD assembly. Our findings indicate that ABA stabilizes PYL8 by altering the dynamics of the CRL4-CDDD-CSN complex association, showing a regulatory mechanism by which a plant hormone inhibits an E3 ubiquitin ligase to protect its own receptors from degradation.

Abstract The placenta, a pivotal organ in mammalian reproduction, allows nutrient exchange and hormonal signaling between the mother and the developing fetus. Understanding its molecular intricacies is essential for deciphering normal embryonic development and pathological conditions such as tumorigenesis. Here, we explore the multifaceted role of the tumor suppressor BRCA1-associated protein 1 (BAP1) in cancer and placentation. Initially recognized for its tumor-suppressive properties, BAP1 has emerged as a key regulator at the intersection of tumorigenesis and placental development. BAP1 influences crucial cellular processes such as cell death, proliferation, metabolism, and response to hypoxic conditions. By integrating insights from tumor and developmental biology, we illuminate the complex molecular pathways orchestrated by BAP1. This perspective highlights BAP1's significant impact on both cancer and placental development, and suggests novel therapeutic strategies that could improve outcomes for pregnancy disorders and cancer.

Abstract The structure of microbial communities arises from a multitude of factors, including the interactions of microorganisms with each other and with the environment. In this work, we sought to disentangle those drivers by performing a cross-study, cross-biome meta-analysis of microbial occurrence data in more than 5000 samples, applying a novel network clustering algorithm aimed to capture conditional taxa co-occurrences. We then examined the phylogenetic and functional composition of the resulting clusters, and searched for global patterns of assembly both at the community level and in the presence/absence of individual metabolic pathways. Our analysis highlighted the prevalence of functional redundancy in microbial communities, particularly between taxa that co-occur in more than one environment, pointing to a relationship between functional redundancy and environmental adaptation. In spite of this, certain pathways were observed in fewer taxa than expected by chance, suggesting the presence of auxotrophy, and presumably cooperation among community members. This hypothetical cooperation may play a role in genome reduction, since we observed a negative relationship between the size of bacterial genomes and the size of the community they belong to. Overall, our results suggest the microbial community assembly is driven by universal principles that operate consistently across different biomes and taxonomic groups.

Abstract Brain damage triggers diverse cellular and molecular events, with astrocytes playing a crucial role in activating local neuroprotective and reparative signaling within damaged neuronal circuits. Here, we investigated reactive astrocytes using a multidimensional approach to categorize their responses into different subtypes based on morphology. This approach utilized the StarTrack lineage tracer, single-cell imaging reconstruction and multivariate data analysis. Our findings identified three profiles of reactive astrocyte responses, categorized by their effects on cell size- and shape- related morphological parameters: “moderate”, “strong,” and “very strong”. We also examined the heterogeneity of astrocyte reactivity, focusing on spatial and clonal distribution. Our research revealed a notable enrichment of protoplasmic and fibrous astrocytes within the “strong” and “very strong” response subtypes. Overall, our study contributes to a better understanding of astrocyte heterogeneity in response to an injury. By characterizing the diverse reactive responses among astrocyte subpopulations, we provide insights that could guide future research aimed at identifying novel therapeutic targets to mitigate brain damage and promote neural repair. Mattirolomyces mexicanus Kovács, Trappe & Alsheikh is recorded for the first time from the Yucatan Peninsula, Mexico, based on morphological and molecular data. This species is characterized by the whitish membranous peridium with inflate hyphae over 100 µm in diameter, pale gleba, subreticulat-ed ascospores of 15–21 µm, and the putative association with Fabaceae species. This is first record of a sequestrate Ascomycota from the Yucatan Peninsula and the second record of the species worldwide. A description, photographs, and a molecular analysis of two DNA markers (ITS and LSU) are presented.

Abstract Mesoscale eddies influence the distribution of diazotrophic (nitrogen-fixing) cyanobacteria, impacting marine productivity and carbon export. Non-cyanobacterial diazotrophs (NCDs) are emerging as potential contributors to marine nitrogen fixation, relying on organic matter particles for resources, impacting nitrogen and carbon cycling. However, their diversity and biogeochemical importance remain poorly understood. In the subtropical North Atlantic along a single transect, this study explored the horizontal and vertical spatial variability of NCDs associated with suspended, slow-sinking, and fast-sinking particles collected with a marine snow catcher. The investigation combined amplicon sequencing with hydrographic and biogeochemical data. Cyanobacterial diazotrophs and NCDs were equally abundant, and their diversity was explained by the structure of the eddy. The unicellular symbiotic cyanobacterium UCYN-A was widespread across the eddy, whereas Trichodesmium and Crocosphaera accumulated at outer fronts. The diversity of particle-associated NCDs varied more horizontally than vertically. NCDs constituted most reads in the fast-sinking fractions, mainly comprising Alphaproteobacteria, whose abundance significantly differed from the suspended and slow-sinking fractions. Horizontally, Gammaproteobacteria and Betaproteobacteria exhibited inverse distributions, influenced by physicochemical characteristics of water intrusions at the eddy periphery. Niche differentiations across the anticyclonic eddy underscored NCD-particle associations and mesoscale dynamics, deepening our understanding of their ecological role and impact on ocean biogeochemistry.

The Tropical Indo-Pacific (TIP) includes about two thirds of the world’s tropical oceans and harbors an enormous number of marine species. The distributions of those species within the region is affected by habitat discontinuities and oceanographic features. As well as many smaller ones, the TIP contains seven large recognized biogeographic barriers that separate the Red Sea and Indian Ocean, the Indian from the Pacific Ocean, the central and eastern Pacific, the Hawaiian archipelago, the Marquesas and Easter Islands. We examined the genetic structuring of populations of Cirrhitichthys oxycephalus, a small cryptic species of reef fish, across its geographic range, which spans the longitudinal limits of the TIP. We assessed geographic variation in the mitochondrial cytb gene and the nuclear RAG1 gene, using 166 samples collected in 46 localities from the western to eastern edges of the TIP. Sequences from cytb show three well-structured groups that are separated by large genetic distances (1.58–2.96%): two in the Tropical Eastern Pacific (TEP), one at Clipperton Atoll another occupying the rest of that region and the third that ranges across the remainder of the TIP, from the central Pacific to the Red Sea and South Africa. These results indicate that the ~4,000 km wide Eastern Pacific Barrier between the central and eastern Pacific is an efficient barrier separating the two main groups. Further, the ~950 km of open ocean that isolates Clipperton Atoll from the rest of the TEP is also an effective barrier. Contrary to many other cases, various major and minor barriers from the Central Indo-Pacific to the Red Sea are not effective against dispersal by C. oxycephalus, although this species has not colonized the Hawiian islands and Easter Island. The nuclear gene partially supports the genetic structure evident in cytb, although all haplotypes are geographically mixed.

CXCR4 is a ubiquitously expressed chemokine receptor that regulates leukocyte trafficking and arrest in both homeostatic and pathological states. It also participates in organogenesis, HIV-1 infection, and tumor development. Despite the potential therapeutic benefit of CXCR4 antagonists, only one, plerixafor (AMD3100), which blocks the ligand-binding site, has reached the clinic. Recent advances in imaging and biophysical techniques have provided a richer understanding of the membrane organization and dynamics of this receptor. Activation of CXCR4 by CXCL12 reduces the number of CXCR4 monomers/dimers at the cell membrane and increases the formation of large nanoclusters, which are largely immobile and are required for correct cell orientation to chemoattractant gradients. Mechanistically, CXCR4 activation involves a structural motif defined by residues in TMV and TMVI. Using this structural motif as a template, we performed in silico molecular modeling followed by in vitro screening of a small compound library to identify negative allosteric modulators of CXCR4 that do not affect CXCL12 binding. We identified AGR1.137, a small molecule that abolishes CXCL12-mediated receptor nanoclustering and dynamics and blocks the ability of cells to sense CXCL12 gradients both in vitro and in vivo while preserving ligand binding and receptor internalization.

The taxonomic status of butterfly rays within the genus Gymnura remains a subject of ongoing debate among researchers. Some authors recognize up to five valid species for the Americas, while others considered several to be synonyms, which has posed a persistent challenge. We aimed to shed light on this complexity by employing molecular operational taxonomic units (MOTUs) based on the mitochondrial gene cytochrome oxidase I (COI). Genetic sequences were obtained from fresh muscle tissue collected in the marine ecoregions corresponding to the type locality from all the nominal butterfly ray species distributed along the Eastern Tropical Pacific (ETP). Our results unveiled compelling findings; all the species delimitation models used consistently identified seven MOTUs for the American continent and an extra G. altavela MOTU restricted to Africa. In addition, our results and models exceeded the worldwide accepted interspecific threshold of 2.0%. Remarkably, our results support the taxonomic reinstatement of Gymnura afuerae (Hildebrand, 1946) as a valid species, with a range expanding into the ETP in the Southern Hemisphere. Similarly, our data support the recent suggestion of resurrecting Gymnura valenciennii (Duméril, 1865) as a valid species in the western Atlantic. These findings urge a reassessment of the conservation status and a comprehensive taxonomic revision of American butterfly rays.

Lipoatrophia semicircularis is a benign pathology characterized by subcutaneous tissue atrophy that affects the skin and related structures. Its etiology remains unclear; however, in the recent few years, it has been proposed that electrostatic charges could be a potential factor. Based on this hypothesis, the aim of this work is to study the cause–effect relation between electrostatic energy and LS, providing insights into the molecular mechanisms. For this purpose, an experimental murine model was created using obese mice. One group served as a control and the other groups involved charging clothes with varying connections to the ground: through the skin, through the clothes or not connected to the ground). Skin biopsies showed that the most significant lesions, including lipophagic granulomas with inflammatory infiltrate, were found in the first group (connected to the ground through the skin). Lipophagic reactions without an inflammatory infiltrate were observed in the other groups subjected to electrical discharges. In the control mice, no histological changes were observed. Oxidative processes were also measured in lower limbs tissue. Malondialdehyde levels significantly increased in the lower limbs after electrostatic discharges. However, the presence of ground through a wire attached to highly conductive clothes around the thigh significantly reduced the effect of electrostatic charges on lipid peroxidation. To our knowledge, this is the first study in which an experimental model has been used to reproduce LS induced by electrostatic energy, suggesting a cause–effect relationship between electrostatic charge and discharge with fat tissue lesion.

Abstract Actin dynamics control early T-cell receptor (TCR) signalling during T-cell activation. However, the precise regulation of initial actin rearrangements is not completely understood. Here, we have investigated the regulatory role of the phosphatase Slingshot-1 (SSH1) in this process. Our data show that SSH1 rapidly polarises to nascent cognate synaptic contacts and later relocalises to peripheral F-actin networks organised at the mature immunological synapse. Knockdown of SSH1 expression by CRISPR/Cas9-mediated genome editing or small interfering RNA reveal a regulatory role for SSH1 in CD3ε conformational change, allowing Nck binding and proper downstream signalling and immunological synapse organisation. TCR triggering induces SSH1-mediated activation of actin dynamics through a mechanism mediated by Limk-1 inactivation. These data suggest that during early TCR activation, SSH1 is required for rapid F-actin rearrangements that mediate initial conformational changes of the TCR, integrin organisation and proximal signalling events for proper synapse organisation. Therefore, the SSH1 and Limk-1 axis is a key regulatory element for full T cell activation.

Pereskia sacharosa Griseb. is a plant used in traditional herbal medicine to treat inflammation. We analyzed the phenolic content of P. sacharosa leaves (EEPs) by liquid chromatography–tandem mass spectrometry (LC-MS/MS) and investigated the anti-inflammatory properties of EEPs and its flavonoid fraction (F10) in animal models subjected to acute neuroinflammation induced by bacterial lipopolysaccharide (LPS). Coronal brain sections of C57BL/6JN male mice or Wistar male rats administered with EEPs or F10 before LPS were subjected to in situ hybridization to determine c-fos and CD14 mRNA levels in the hypothalamus or GABAA γ2 mRNA levels in the hippocampus. Theta oscillations were recorded every 6 h in the hippocampus of Wistar rats. In total, five flavonoids and eight phenolic acids were identified and quantified in P. sacharosa leaves. Either EEPs or F10 crossed the blood–brain barrier (BBB) into the brain and reduced the mRNA expression of c-fos, CD14, and GABAA γ2. A decrease in theta oscillation was observed in the hippocampus of the LPS group, while the F10 + LPS group overrode the LPS effect on theta activity. We conclude that the bioactive compounds of P. sacharosa reduce the central response to inflammation, allowing the early return of ambulatory activity and well-being of the animal.

A new belondirid, dorylaimid taxon, Oleaxonchium olearum gen. et sp. nov., collected from an olive grove in the Andalusia region of Spain, is characterised, including its morphological description, morphometrics, SEM study, and molecular (18S-, 28S-rDNA) analyses. The new genus displays a unique combination of traits that distinguishes it from its closest genera: a rectangular lip region with sclerotized margins when observed in lateral view and visibly hexagonal in face view under SEM; a comparatively long cheilostom with thickened walls at its anterior part; a short isthmus-like section separating both pharyngeal regions; a mono-opistho-ovarian didelphic female genital system without pars refringens vaginae; and a short and rounded tail. The new species is characterised by its 2.44–2.87 mm long body, lip region 7–7.5 µm wide, odontostyle 10–10.5 µm long, neck 723–973 µm long, pharyngeal expansion occupying 63–72% of the total neck length, female anterior genital branch 4–6% of body length, tripartite posterior uterus 1.9–2.6 body diameters long, with a short intermediate section bearing sclerotized elements, vulva (V = 58–61) a transverse slit, caudal region 29–35 µm long (c = 74–89, c’ = 0.9–1.1), and male unknown. As derived from an integrative approach combining morphological and molecular data, the new genus is close to Metaxonchium, the polyphyly of Belondiridae is confirmed, and support is provided in favour of the monophyly of Axonchiinae.

Effective long-term management of invasive non-native species (INNS) in South America is a pressing yet complex task. Critically, the environmental, historical, cultural, and economic idiosyncrasies of the region call for the inclusion of a plurality of views from those sectors of society receiving the negative and positive impacts of INNS. This is a multifaceted, and often daunting, task that can be aided by an early identification of stakeholders – those affected by or with an interest in INNS and their management – accompanied by targeted stakeholder engagement. Here, we report the procedures and results of a stakeholder mapping activity aimed at identifying stakeholders and designing engagement strategies. Using expert knowledge procedures, we compiled comprehensive lists of stakeholders for six case-studies in South America: (i) invasive grasses (Urochloa spp.) in Brazil; (ii) glossy privet (Ligustrum lucidum) in Argentina; (iii) lodgepole and Monterey pines (Pinus contorta and P. radiata) in Argentina; (iv) American mink (Neogale vison) in Argentina and Chile; (v) lodgepole and Monterey pines in Chile; and (vi) German yellow-jacket (Vespula germanica) in Chile. Overall, we identified 250 stakeholders, which, based on their interest and influence, were classified into “context settlers” (2%), “key players” (47%), “crowd” (5%), and “subjects” (49%). We outlined strategies to engage with each of these four groups and for each of our six case-studies. Across case studies, communication with stakeholders was the most common engagement strategy proposed (27%; 19 of 70 strategies), followed by active involvement of stakeholders in INNS research and management (23%). Our results highlight the importance of considering power imbalances, as those stakeholders more likely to benefit from INNS were assessed to have more influence over INNS management relative to local and indigenous communities. Our work illustrates how to identify stakeholders in a rigorous and rapid manner, which should be complemented with the involvement of the stakeholders themselves.

We describe a new species of Centrolene from the Subandean Cordillera of Kutukú in southeastern Ecuador. The new species differs from all other glassfrogs by the combination of the following characters: presence of processes of vomers but without vomerine teeth; humeral spines in males; dorsum green with light green dots and without dark marks; dorsal skin with abundant tubercles; all visceral peritonea translucent (except for pericardium); and small body size (snout-vent length 21.5–21.9 mm in adult males). The new species is sister to Centrolene camposi from the Western Cordillera of the Andes of southwestern Ecuador, and together they form a clade with C. condor from the Subandean Cóndor Cordillera in southeastern Ecuador. Our time tree suggests that the new species originated at the end of the Pliocene. In addition, we present new information for C. zarza, expanding its geographic range across the southeastern Andes and the Kutukú and Cóndor cordilleras, amending its definition and diagnosis, and offering new information on its natural history and extinction risk. We also discuss the taxonomic status of Ecuadorian populations reported as C. huilensis and conclude that they are C. muelleri based on their close phylogenetic relationships and morphological similarity to samples of C. muelleri from Peru. Centrolene huilensis is a valid species and not closely related to C. muelleri.

Abstract Background The gut microbiota controls broad aspects of human metabolism and feeding behavior, but the basis for this control remains largely unclear. Given the key role of human dipeptidyl peptidase 4 (DPP4) in host metabolism, we investigate whether microbiota DPP4-like counterparts perform the same function. Results We identify novel functional homologs of human DPP4 in several bacterial species inhabiting the human gut, and specific associations between Parabacteroides and Porphyromonas DPP4-like genes and type 2 diabetes (T2D). We also find that the DPP4-like enzyme from the gut symbiont Parabacteroides merdae mimics the proteolytic activity of the human enzyme on peptide YY, neuropeptide Y, gastric inhibitory polypeptide (GIP), and glucagon-like peptide 1 (GLP-1) hormones in vitro. Importantly, administration of E. coli overexpressing the P. merdae DPP4-like enzyme to lipopolysaccharide-treated mice with impaired gut barrier function reduces active GIP and GLP-1 levels, which is attributed to increased DPP4 activity in the portal circulation and the cecal content. Finally, we observe that linagliptin, saxagliptin, sitagliptin, and vildagliptin, antidiabetic drugs with DPP4 inhibitory activity, differentially inhibit the activity of the DPP4-like enzyme from P. merdae. Conclusions Our findings confirm that proteolytic enzymes produced by the gut microbiota are likely to contribute to the glucose metabolic dysfunction that underlies T2D by inactivating incretins, which might inspire the development of improved antidiabetic therapies.

Introducción: La celiaquía es una patología multisistémica con base autoinmune que provoca una reacción a la proteína gluten encontrada en alimentos como o trigo, centeno, triticale, cebada, espelta y avena. Los síntomas más comunes de la celiaquía son la pérdida de peso, pérdida de apetito y fatiga general entre otros. A consecuencia de estos síntomas el rendimiento deportivo puede verse afectado. Objetivo: Para comprender como la celiaquía impacta en el rendimiento de los deportistas, el objetivo de este estudio es realizar una revisión sistemática relacionada con el rendimiento deportivo en personas celíacas. Materiales y métodos: Para ello, se utilizó la siguiente frase final de búsqueda: “Celiac” AND “Sport” AND “Performance”, que fue introducida por igual en las bases de datos: Web of Science, Scopus y PubMed. Resultados: Los resultados muestran la existencia de escasos documentos relacionados con el objeto de estudio (n=11), dónde se muestra una variabilidad de tipos de diseño en los trabajos seleccionados. Por otro lado, se extrae que la única solución para mantener un adecuado rendimiento deportivo en una persona celiaca es llevar una dieta libre de gluten de por vida. Conclusiones: Los documentos seleccionados subrayan la importancia de que los deportistas con enfermedad celiaca mantengan una dieta sin gluten de manera constante a lo largo de su vida. Debido a la escasez de estudios relacionados con la temática, se recomienda aumentar el número de investigaciones relacionadas con el objeto de estudio, con la finalidad de mejorar los estilos y hábitos de vida de los deportistas celíacos.

Biogeography has as a central theme, which is the study of geographic ranges of species that are determined by evolutionary history, abiotic factors, and biotic interactions. Understanding the influence of biotic interactions on geographic ranges is a topic that has been little explored, especially in a way that compares species that vary in intensity of interaction. Here, we assessed interaction intensity as a determinant of geographic range overlap between ant-following birds and army ants in Mexico and Central America. We hypothesized that the intensity of the interaction between army ants and ant-following birds, obligate or facultative, predicts the overlap of geographic ranges of interacting species, as well as the extension of geographic ranges. We generated species distribution models with MAXENT and estimated the percentage of overlap between two species of army ants and 10 species of ant-following birds. Contrary to our predictions, Bayesian regression models found no support for an estimated higher range overlap for obligate ant-following birds and army ants, or wider geographic ranges for facultative ant-following bird species. However, our results suggested trends for higher percentages of range overlap between obligate ant-following birds and army ants, and for geographic ranges of facultative ant-following birds extending to areas without the presence of army ants. Our research encourages further exploration of the biogeography of biotic interactions as part of a quantitative gradient of intensities and not as qualitative categories, integrating spatial and temporal variation in the intensity of interaction.

Aquatic shredder insect diversity declines latitudinally toward the equator, contrary to the general latitudinal biodiversity gradient. Shredder diversity along tropical elevational gradients mimics this shredder latitudinal pattern. One of the hypotheses proposed to explain this pattern of diversity is that dispersal capacity drives variation in shredder assemblages given their low dispersal capacity in tropical streams. Additionally, tropical shredders probably have lower dispersal capacity than the rest of tropical aquatic insects, which have lower dispersal capacities than their counterparts in temperate areas. We tested this hypothesis in an elevational gradient of more than 2000 m in 16 reaches of streams distributed in the Usumacinta, Cahabon, and Polochic river watersheds. We quantitatively sampled aquatic insects and measured 12 environmental variables. We found a regional pool of 118 taxa, with 13 taxa classified as shredders, and 2 taxa of predator-shredders. Contrary to expectations, shredder rarefied richness decreased with increasing elevation, which suggests that dispersal capacity did not change with elevation. Assemblage similarity decreased with increasing distance between reaches due to low capacity to fly long distances. This relationship had a smaller slope when using the shortest spatial distances between pairs of reaches due to potential lateral scatter by flying adults. In sum, the results support the hypothesis that dispersal capacity drove aquatic shredder assemblage structure in these 16 tropical streams.

In the SARS-CoV-2 lineage, RNA elements essential for its viral life cycle, including genome replication and gene expression, have been identified. Still, the precise structures and functions of these RNA regions in coronaviruses remain poorly understood. This lack of knowledge points out the need for further research to better understand these crucial aspects of viral biology and, in time, prepare for future outbreaks. In this research, the in silico analysis of the cis RNA structures that act in the alpha-, beta-, gamma-, and deltacoronavirus genera has provided a detailed view of the presence and adaptation of the structures of these elements in coronaviruses. The results emphasize the importance of these cis elements in viral biology and their variability between different viral variants. Some coronavirus variants in some groups, depending on the cis element (stem-loop1 and -2; pseudoknot stem-loop1 and -2, and s2m), exhibited functional adaptation. Additionally, the conformation flexibility of the s2m element in the SARS variants was determined, suggesting a coevolution of this element in this viral group. The variability in secondary structures suggests genomic adaptations that may be related to replication processes, genetic regulation, as well as the specific pathogenicity of each variant. The results suggest that RNA structures in coronaviruses can adapt and evolve toward different viral variants, which has important implications for viral adaptation, pathogenicity, and future therapeutic strategies.

Abstract Background Prenatal alcohol exposure (PAE) has serious physical consequences for children such as behavioral disabilities, growth disorders, neuromuscular problems, impaired motor coordination, and decreased muscle tone. However, it is not known whether loss of muscle strength occurs, and which interventions will effectively mitigate physical PAE impairments. We aimed to investigate whether physical alteration persists during adolescence and whether exercise is an effective intervention. Results Using paradigms to evaluate different physical qualities, we described that early adolescent PAE animals have significant alterations in agility and strength, without alterations in balance and coordination compared to CTRL animals. We evaluated the effectiveness of 3 different exercise protocols for 4 weeks: Enrichment environment (EE), Endurance exercise (EEX), and Resistance exercise (REX). The enriched environment significantly improved the strength in the PAE group but not in the CTRL group whose strength parameters were maintained even during exercise. Resistance exercise showed the greatest benefits in gaining strength, and endurance exercise did not. Conclusion PAE induced a significant decrease in strength compared to CTRL in PND21. Resistance exercise is the most effective to reverse the effects of PAE on muscular strength. Our data suggests that individualized, scheduled, and supervised training of resistance is more beneficial than endurance or enriched environment exercise for adolescents FASD.

The port fouling fauna from Mexico has recieved greater study in the last 15 years. By improving our knowledge of fouling invertebrates on artificial substrates on the Southern Pacific coast of Mexico, we can better understand their distribution and prevent bioinvasions. The specimens listed in this present study come from PVC plates that were placed between June 2019 to June 2020 submerged at 1 m depth, on four docks of the Chahué Marina, Oaxaca, Mexico. Forty-four taxa, representing nine phyla were identi-fied: Sipuncula (1 species), Entoprocta (1), Bryozoa (3), Arthropoda (3), Porifera (4), Cnidaria (5), Chordata (6), Mollusca (10) and Annelida (11). Thirty taxa were identified to species, with two still requiring confirmation. Five were identified to genus, two to family, and five to class level. From the 30 identified species, 18 were non-native species: one entoproct, one mollusk, two bryozoans, three hydrozoans, three barnacles, four polychaetes, and four ascidians. These results include seven new records from the state of Oaxaca: one entoproct, one barnacle, two bryozoans, and three hydrozoans.

Abstract With the main aim of identifying biomarkers that contribute to defining the concept of ideal protein in growing rabbits under the most diverse conditions possible this work describes two different experiments. Experiment 1: 24 growing rabbits are included at 56 days of age. The rabbits are fed ad libitum one of the two experimental diets only differing in lysine levels. Experiment 2: 53 growing rabbits are included at 46 days of age, under a fasting and eating one of the five experimental diets, with identical chemical composition except for the three typically limiting amino acids (being fed commercial diets ad libitum in both experiments). Blood samples are taken for targeted and untargeted metabolomics analysis. Here we show that the metabolic phenotype undergoes alterations when animals experience a rapid dietary shift in the amino acid levels. While some of the differential metabolites can be attributed directly to changes in specific amino acids, creatinine, urea, hydroxypropionic acid and hydroxyoctadecadienoic acid are suggested as a biomarker of amino acid imbalances in growing rabbits’ diets, since its changes are not attributable to a single amino acid. The fluctuations in their levels suggest intricate amino acid interactions. Consequently, we propose these metabolites as promising biomarkers for further research into the concept of the ideal protein using rabbit as a model.

Based on molecular and morphological evidence, we report the first record of Eleutherodactylus nitidus (Peters, 1870) from Mexico City, in the urban area of Panteón Xilotepec, Xochimilco. We discuss the remarkable urban adaptability of Eleutherodactylus and the need for additional sampling to determine if E. nitidus is native or introduced to Mexico City.

Abstract The selection of highly productive genotypes with stable performance across environments is a major challenge of plant breeding programs due to genotype-by-environment (GE) interactions. Over the years, different metrics have been proposed that aim at characterizing the superiority and/or stability of genotype performance across environments. However, these metrics are traditionally estimated using phenotypic values only and are not well suited to an unbalanced design in which genotypes are not observed in all environments. The objective of this research was to propose and evaluate new estimators of the following GE metrics: Ecovalence, Environmental Variance, Finlay–Wilkinson regression coefficient, and Lin–Binns superiority measure. Drawing from a multi-environment genomic prediction model, we derived the best linear unbiased prediction for each GE metric. These derivations included both a squared expectation and a variance term. To assess the effectiveness of our new estimators, we conducted simulations that varied in traits and environment parameters. In our results, new estimators consistently outperformed traditional phenotype-based estimators in terms of accuracy. By incorporating a variance term into our new estimators, in addition to the squared expectation term, we were able to improve the precision of our estimates, particularly for Ecovalence in situations where heritability was low and/or sparseness was high. All methods are implemented in a new R-package: GEmetrics. These genomic-based estimators enable estimating GE metrics in unbalanced designs and predicting GE metrics for new genotypes, which should help improve the selection efficiency of high-performance and stable genotypes across environments.

The exhibition of paternal behaviour has been associated with high peripheral concentrations of oxytocin (OT) and levels of oxytocin receptors (OTRs) in the paraventricular and supraoptic nuclei of the hypothalamus. The presence of OTRs in the medial preoptic area (mPOA), bed nucleus of the stria terminalis (BNST), medial amygdala (MeA), and olfactory bulb (OB) has not been studied extensively in the paternal context, although these nuclei are essential in the regulation of paternal behaviour. The aim of this study was to determine whether there is an association between paternal response of sexually inexperienced males of the Mongolian gerbils (Meriones unguiculatus) and OT concentrations in plasma, as well as the presence of OTRs in the mPOA, BNST, MeA, and OB. We used sexually inexperienced gerbils: ten paternal (PAT) males and 10 males aggressive with pups (AGG). PAT males and AGG males were subjected to paternal behaviour tests. Then, blood samples were taken to quantify OT concentrations. The brains were dissected for OTR immunohistochemistry analyses. PAT males had significantly higher plasma levels of OT and numbers of cells with OTR immunoreactivity (ir) in the mPOA/BNST. These results indicated an association between paternal behaviour and peripheral concentrations of OT, as well as the presence of OTRs in the mPOA and BNST. This suggests that OT and OTRs could participate in the regulation of paternal behaviour in the Mongolian gerbil.

Sex-specific differences in lifespan and ageing are observed in various species. In humans, women generally live longer but are frailer and suffer from different age-related diseases compared to men. The hallmarks of ageing, such as genomic instability, telomere attrition or loss of proteostasis, exhibit sex-specific patterns. Sex chromosomes and sex hormones, as well as the epigenetic regulation of the inactive X chromosome, have been shown to affect lifespan and age-related diseases. Here we review the current knowledge on the biological basis of sex-biased ageing. While our review is focused on humans, we also discuss examples of model organisms such as the mouse, fruit fly or the killifish. Understanding these molecular differences is crucial as the elderly population is expected to double worldwide by 2050, making sex-specific approaches in the diagnosis, treatment, therapeutic development and prevention of age-related diseases a pressing need.

Amyotrophic lateral sclerosis (ALS) is a devastating neuromuscular disease characterized by progressive motor function and muscle mass loss. Despite extensive research in the field, the underlying causes of ALS remain incompletely understood, contributing to the absence of specific diagnostic and prognostic biomarkers and effective therapies. This study investigates the expression of long-non-coding RNAs (lncRNAs) in skeletal muscle as a potential source of biomarkers and therapeutic targets for the disease. The expression profiles of 12 lncRNAs, selected from the literature, were evaluated across different disease stages in tissue and muscle biopsies from the SOD1G93A transgenic mouse model of ALS. Nine out of the 12 lncRNAs were differentially expressed, with Pvt1, H19 and Neat1 showing notable increases in the symptomatic stages of the disease, and suggesting their potential as candidate biomarkers to support diagnosis and key players in muscle pathophysiology in ALS. Furthermore, the progression of Myhas and H19 RNA levels across disease stages correlated with longevity in the SOD1G93A animal model, effectively discriminating between long- and short-term survival individuals, thereby highlighting their potential as prognostic indicators. These findings underscore the involvement of lncRNAs, especially H19 and Myhas, in ALS pathophysiology, offering novel insights for diagnostic, prognostic and therapeutic targets.

Los consumidores suelen asociar sus preferencias con sabores específicos según la procedencia de los alimentos y bebidas; sin embargo, no siempre se consideran estas diferencias cuando se exportan dichos alimentos. Esta investigación presenta los resultados de un estudio diseñado para evaluar diferencias interculturales en la percepción de los consumidores mexicanos y colombianos de características organolépticas (gusto general, frescura, intensidad de color, intensidad de aroma, acidez) entre el arándano cultivar biloxi cultivado en México y Colombia. Se diseñó un estudio experimental basado en un cuestionario en el que 397 consumidores de ambos países (192 en México, 205 en Colombia) evaluaron distintos atributos sensoriales de dos muestras: un arándano mexicano y uno colombiano, ambos de la variedad biloxi. Adicionalmente, se realizó un análisis de microexpresiones faciales para complementar los resultados del experimento con indicadores de valencia emocional. Se demostró que ciertas correspondencias entre las propiedades organolépticas son consistentes en ambas culturas. Específicamente, en ambas nacionalidades, se evidencia un efecto significativo en los atributos acidez, amargura y dulzura. El arándano colombiano fue percibido más dulce, mientras que el arándano mexicano fue evaluado como más amargo y ácido. Cuando el arándano evaluado fue clasificado como más ácido, la calificación de gusto general fue más baja. Estos resultados son relevantes para los productores, comercializadores y exportadores de arándanos, sabiendo que aquellas variedades más dulces serán las que tendrán mayor aceptación por parte de los consumidores colombianos y mexicanos.
