Steady-state and time-resolved fluorescence techniques were employed to study the photophysics and photochemistry of trans-resveratrol. trans-Resveratrol is found in large quantities in fungi-infected grapevine-leaf tissue and plays a direct role in the resistance to plant disease. We found that trans-resveratrol in liquid solution undergoes a trans-cis isomerization process in the excited state at a rate that depends partially on the solvent viscosity, as was found in previous studies on trans-stilbene. The hydroxyl groups of the phenol moieties in resveratrol are weak photoacids. In water and methanol solutions containing weak bases such as acetate, a proton is transferred to the base within the lifetime of the excited state. When resveratrol is adsorbed on cellulose (also a component of the plant's cell wall), the cis-trans process is slow and the lifetime of the excited state increases from several tens of picoseconds in ethanol to about 1.5 ns. Excited-state proton transfer occurs when resveratrol is adsorbed on cellulose and acetate ions are in close proximity to the phenol moieties. We propose that proton transfer from excited resveratrol to the fungus acid-sensing chemoreceptor is one of the plant's resistance mechanisms to fungal infection.

Steady-state and time-resolved fluorescence techniques were employed to study the excited-state proton transfer (ESPT) from a photoacid adsorbed on starch to a nearby water molecule. Starch is composed of ∼30% amylose and ∼70% amylopectin. We found that the ESPT rate of adsorbed 8-hydroxy-1,3,6-pyrenetrisulfonate (HPTS) on starch arises from two time constants of 300 ps and ∼3 ns. We explain these results by assigning the two different ESPT rates to HPTS adsorbed on amylose and on amylopectin. When adsorbed on amylose, the ESPT rate is ∼3 × 10(9 )s(-1), whereas on amylopectin, it is only ∼3 × 10(8) s(-1).

Time-resolved and steady-state florescence measurements were used to study the photoprotolytic process of an adsorbed photoacid on cellulose and chitin. For that purpose we used the 8-hydroxy-1,3,6-pyrenetrisulfonate (HPTS) photoacid which transfers a proton to water with a time constant of 100 ps, but is incapable of doing so in methanol or ethanol. We found that both biopolymers accept a proton from the electronically excited acidic ROH form of HPTS. The excited-state proton-transfer (ESPT) rate of HPTS adsorbed on chitin is greater than that on cellulose by a factor of 5. The ESPT on chitin also occurs in the presence of methanol or ethanol, but at a slower rate. The transferred protons can recombine efficiently with the conjugate excited base, the RO(-) form of HPTS.

The main features of the photoinduced kinetics of both ultrafast excited-state proton and electron transfer reactions that occur in the picosecond (ps) and femtosecond (fs) time domains are compared. Proton transfer (PT) reaction kinetics can be described in terms of several discrete values of rate coefficients in the form of polyexponential functions where each value of the rate coefficient can be attributed to a definite physical behavior of the reaction mechanism. In contrast, electron transfer (ET) reaction kinetics requires a consideration of a continuous distribution of rate coefficients. This difference can be related to structure of the ground-state reactant pairs for each reaction. Excited-state ET can occur at various configurations of reactant molecules and its rate reflects the fluctuations of the distances and orientations of these molecules. In contrast, excited-state PT requires preliminary formation of a ground-state H-bonded complex with definite structure where the reaction occurs after photoexcitation.

UV-vis steady-state and time-resolved techniques were employed to study the excited-state proton-transfer process from two weak photoacids positioned next to the surface of chitosan and cellulose. Both chitosan and cellulose are linear polysaccharides; chitosan is composed mainly of d-glucosamine units. In order to overcome the problem of the high basicity of the glucosamine, we chose 2-naphthol (pKa* ≈ 2.7) and 2-naphthol-6-sulfonate (pKa* ≈ 1.7) as the proton emitters because of their ground state pKa (≈9). Next to the 1:1 cellulose:water weight ratio, the ESPT rate of these photoacids is comparable to that of bulk water. We found that the ESPT rate of 2-naphthol (2NP) and 2-naphthol-6-sulfonate (2N6S) next to chitosan in water (1:1) weight ratio samples is higher than in bulk water by a factor of about 5 and 2, respectively. We also found an efficient ESPT process that takes place from these photoacids in the methanol environment next to the chitosan scaffold, whereas ESPT is not observed in methanolic bulk solutions of these photoacids. We therefore conclude that ESPT occurs from these photoacids to the d-glucosamine units that make up chitosan.

Mutations near the fluorescing chromophore of the green fluorescent protein (GFP) have direct effects on the absorption and emission spectra. Some mutants have significant band shifts and most of the mutants exhibit a loss of fluorescence intensity. In this study we continue our investigation of the factors controlling the excited state proton transfer (PT) process of GFP, in particular to study the effects of modifications to the key side chain Ser205 in wt-GFP, proposed to participate in the proton wire. To this aim we combined mutagenesis, X-ray crystallography, steady-state spectroscopy, time-resolved emission spectroscopy and all-atom explicit molecular dynamics (MD) simulations to study the double mutant T203V/S205A. Our results show that while in the previously described GFP double mutant T203V/S205V the PT process does not occur, in the T203V/S205A mutant the PT process does occur, but with a 350 times slower rate than in wild-type GFP (wt-GFP). Furthermore, the kinetic isotope effect in the GFP double mutant T203V/S205A is twice smaller than in the wt-GFP and in the GFP single mutant S205V, which forms a novel PT pathway. On the other hand, the crystal structure of GFP T203V/S205A does not reveal a viable proton transfer pathway. To explain PT in GFP T203V/S205A, we argue on the basis of the MD simulations for an alternative, novel proton-wire pathway which involves the phenol group of the chromophore and water molecules infrequently entering from the bulk. This alternative pathway may explain the dramatically slow PT in the GFP double mutant T203V/S205A compared to wt-GFP.

Steady-state and time-resolved techniques were employed to study the excited-state proton-transfer (ESPT) rate of two newly synthesized 8-hydroxy-1,3,6-pyrenetrisulfonate (pyranine, HPTS) derived photoacids in three protic solvents, water, methanol and ethanol. The ESPT rate constant k(PT) of tris(1,1,1,3,3,3-hexafluoropropan-2-yl)-8-hydroxypyrene-1,3,6-trisulfonate, 1a, whose pK(a)* ~ -4, in water, methanol and ethanol is 3 × 10(11) s(-1), 8 × 10(9) s(-1) and 5 × 10(9) s(-1) respectively. (8-Hydroxy-N1,N3,N6-tris(2-hydroxyethyl)-N1,N3,N6-trimethylpyrene-1,3,6 trisulfonamide, 1b) is a weaker acid than 1a but still a strong photoacid with pK(a)* ~ -1 and the ESPT rate in water, methanol and ethanol is 7 × 10(10) s(-1), 4 × 10(8) s(-1) and 2 × 10(8) s(-1). We qualitatively explain our kinetic results by a Marcus-like free-energy correlation which was found to have a general form suitable for describing proton transfer reactions in both the proton-adiabatic and the proton-non-adiabatic limits.

Steady-state and time-resolved emission techniques were used to study the protolytic processes in the excited state of dehydroluciferin, a nonbioluminescent product of the firefly enzyme luciferase. We found that the ESPT rate coefficient is only 1.1 × 10(10) s(-1), whereas those of d-luciferin and oxyluciferin are 3.7 × 10(10) and 2.1 × 10(10) s(-1), respectively. We measured the ESPT rate in water-methanol mixtures, and we found that the rate decreases nonlinearly as the methanol content in the mixture increases. The deprotonated form of dehydroluciferin has a bimodal decay with short- and long-time decay components, as was previously found for both D-luciferin and oxyluciferin. In weakly acidic aqueous solutions, the deprotonated form's emission is efficiently quenched. We attribute this observation to the ground-state protonation of the thiazole nitrogen, whose pK(a) value is ~3.

The crystal structure of 2-butylamino-4-nitro-5-methyl pyridine N-oxide (2B5M) and solution studies of both 2B5M and 2-methylamino-4-nitro-5-methyl pyridine (2M5M) N-oxide are presented. Steady-state absorption and emission measurements were employed for both molecules while a picosecond fluorescence up-conversion technique was used to follow the dynamic behavior of the 2M5M system. The experimental methods were complemented by DFT and TD DFT B3LYP/6-31G(d,p) calculations involving ground and excited-state optimization which in the case of the smaller 2M5M molecule were extended to the CAM-B3LYP/6-31G(d,p) method. The solvent effect is incorporated by applying the polarizable continuum (PCM) model. The data reveal that the 2B5M molecule crystallizes in the monoclinic space group P2(1)/c and its crystal lattice is composed of monomers with intramolecular N-H···O [2.572(3) Å] hydrogen bonds, connected into a polymer network by weak intermolecular C-H…O [3.2-3.4 Å]-type interactions. Quantum-chemical calculations show that the aminoalkyl substitutent in aminoalkyl-pyridine N-oxides is a specific determinant of the CT nature of the lowest-lying excited electronic ππ* state, distinguishing them from other nitroaromatic compounds. The results of both picosecond fluorescence up-conversion experiments in different solvents and quantum-chemical calculations suggest that in nonpolar media the ESIPT process in 2M5M is favored, while in polar acetonitrile, the N* → PT* transition demands barrier-crossing and thus unfavorable thermodynamic conditions do not allow the ESIPT to occur. The signals of picosecond fluorescence up-conversion of 2M5M are solvent- and emission-wavelength dependent. The three time components found in a weakly polar isooctane-dioxane mixture have been attributed to solvation dynamics (∼500 fs), and to relaxation of N* and PT* forms while in acetonitrile, a very rapid fluorescence decay with a time constant (2.3-4.0 ps) indicative of the presence of the normal (N*) form was observed. Much shorter fluorescence lifetimes in alcohols (a few picoseconds) and in D(2)O (less than 200 fs) than in aprotic solvents suggest that in protic media, the solvent molecules participate in the ESIPT, bridging between the methylamine group and the N-oxide group of 2M5M.

To further explore excited state proton transfer (ESPT) pathways within green fluorescent protein (GFP), mutagenesis, X-ray crystallography, and time-resolved and steady-state optical spectroscopy were employed to create and study the GFP mutant S205A. In wild type GFP (wt-GFP), the proton transfer pathway includes the hydroxyl group of the chromophore, a water molecule, Ser205, and Glu222. We found that the ESPT rate constant of S205A is smaller by a factor of 20 than that of wt-GFP and larger by a factor of 2 in comparison to the ESPT rate of S205V mutant which we previously characterized. (1) High resolution crystal structures reveal that in both S205A and S205V mutants, an alternative proton transfer pathway is formed that involves the chromophore hydroxyl, a bridging water molecule, Thr203 and Glu222. The slow PT rate is explained by the long (∼3.2 Å and presumably weak) hydrogen bond between Thr203 and the water molecule, compared to the 2.7 Å normal hydrogen bond between the water molecule and Ser205 in wt-GFP. For data analysis of the experimental data from both GFP mutants, we used a two-rotamer kinetic model, assuming only one rotamer is capable of ESPT. Data analysis supports an agreement with the underlying assumption of this model.

Time-resolved emission as well as steady-state UV-vis techniques were employed to study the photoprotolytic processes that d-luciferin, the natural substrate of the firefly luciferase, undergoes in both acidic aqueous solutions and ice. The emission spectrum of D-luciferin in a 20 mM HCl aqueous solution or higher has an additional emission band at 590 nm red-shifted with respect to the strongest emission band positioned at 530 nm of the deprotonated NRO(-*) form in a pH-neutral aqueous solution. We attribute this emission band to the zwitterion form designated as (+)HNRO(-). The time-resolved emission signals show that the NRO(-*) emission band at 530 nm and the zwitterion emission band at 590 are strongly quenched by a recombination process with a proton in an acidic solution and in ice. In ice, the quenching rate is 10 times faster than in the liquid state. We attribute the fast quenching rate to the high value of the proton diffusion constant in ice.

Steady-state and time-resolved techniques were employed to study the excited-state proton transfer (ESPT) from d-luciferin, the natural substrate of the firefly luciferase, to the mild acetate base in aqueous solutions. We found that in 1 M aqueous solutions of acetate or higher, a proton transfer (PT) process to the acetate takes place within 30 ps in both H(2)O and D(2)O solutions. The time-resolved emission signal is composed of three components. We found that the short-time component decay time is 300 and 600 fs in H(2)O and D(2)O, respectively. This component is attributed either to a PT process via the shortest water bridged complex available, ROH··H(2)O··Ac(-), or to PT taking place within a contact ion pair. The second time component of 2000 and 3000 fs for H(2)O and D(2)O, respectively, is attributed to ROH* acetate complex, whose proton wire is longer by one water molecule. The decay rate of the third, long-time component is proportional to the acetate concentration. We attribute it to the diffusion-assisted reaction as well as to PT process to the solvent.

Steady-state emission and time-resolved techniques were employed to study the photoprotolytic processes d-luciferin undergoes in water-methanol mixtures over a wide range of molar fractions (chi(MeOH)) of methanol. We found that in the concentration range of 0 < chi(MeOH) < 0.8 the rate constant of the excited-state proton transfer (ESPT) to the solvent decreases nearly exponentially with increasing chi(MeOH). At chi(MeOH) > 0.8 the proton transfer rate constant decreases with an even steeper slope. The kinetic isotope effect (KIE) maintains a constant value of 2.4 +/- 0.2 at all the mixture's compositions.

Steady-state absorption and emission as well as time-resolved emission spectroscopies were employed to study the photophysics and photochemistry of D-luciferin, the firefly active bioluminescent compound. In aqueous solution the electronically excited-state protonated D-luciferin compound undergoes an efficient process of proton transfer to the solvent, with a rate constant k(PT) = 3.0 x 10(10) s(-1). We found a kinetic isotope effect of about 2.5 for this process. The deprotonated form of D-luciferin in the excited state recombines irreversibly with the geminate proton. Hence, the fluorescence decay of the deprotonated form is nonexponential, and the fluorescence quantum yield is low.

mKeima is an unusual monomeric red fluorescent protein (lambda(em)(max) approximately 620 nm) that is maximally excited in the blue (lambda(ex)(max) approximately 440 nm). The large Stokes shift suggests that the chromophore is normally protonated. A 1.63 A resolution structure of mKeima reveals the chromophore to be imbedded in a novel hydrogen bond network, different than in GFP, which could support proton transfer from the chromophore hydroxyl, via Ser142, to Asp157. At low temperatures the emission contains a green component (lambda(em)(max) approximately 535 nm), enhanced by deuterium substitution, presumably resulting from reduced proton transfer efficiency. Ultrafast pump/probe studies reveal a rising component in the 610 nm emission with a lifetime of approximately 4 ps, characterizing the rate of proton transfer. Mutation of Asp157 to neutral Asn changes the chromophore resting charge state to anionic (lambda(ex)(max) approximately 565 nm, lambda(em)(max) approximately 620 nm). Thus, excited state proton transfer (ESPT) explains the large Stokes shift. This work unambiguously characterizes green emission from the protonated acylimine chromophore of red fluorescent proteins.

The effects of excess protons on the fluorescence quenching process of 1-naphthol-4-sulfonate (1N4S) and 1-naphthol-3-sulfonate (1N3S) in methanol-doped ice samples were studied by employing a time-resolved emission technique. We found that the fluorescence quenching of the deprotonated form RO(-)* of both photoacids by protonation is very efficient in ice, whereas in liquid water the proton fluorescence quenching is rather small. Using the Smoluchowski diffusion-assisted binary collision model under certain assumptions and approximations, we found that the calculated proton diffusion constant in ice in the temperature range of 240-260 K was 10 times greater than that of water at 295 K.

We used an inhomogeneous excited-state proton-transfer kinetics model to explain the origin of the non-exponential time-resolved emission of the A-band of wt-green fluorescence protein. The calculated fit is rather good for both H 2O and D 2O samples in a wide temperature range of 80-229 K. We attribute the inhomogeneous kinetics to the distance dependence of the excited-state proton-transfer rate between the proton donor (the hydroxyl group of the chromophore) and the oxygen of a nearby water molecule.

Steady-state emission and time-correlated single-photon counting (TCSPC) are used to measure the temperature dependence of the proton-transfer rate of wt-GFP in H2O and D2O. As the temperature decreases, the proton-transfer rate from the protonated form slows down. At about 80 K, the rate is about 10-fold slower than the rate at room temperature. At lower temperatures of 70 K down to 13 K (the lowest temperature studied), the rate of proton transfer is almost temperature independent. We explain the temperature dependence of the proton-transfer rate by an intermolecular vibration assisted tunneling mechanism. We attribute the specific intermolecular vibration to the oscillation of two oxygen atoms: the chromophore's phenol ring and the nearby water molecule. The kinetic isotope effect is about 5 and is almost temperature independent.

Wild-type green fluorescent protein (wt-GFP) has a prominent absorbance band centered at approximately 395 nm, attributed to the neutral chromophore form. The green emission arising upon excitation of this band results from excited-state proton transfer (ESPT) from the chromophore hydroxyl, through a hydrogen-bond network proposed to consist of a water molecule and Ser205, to Glu222. Although evidence for Glu222 as a terminal proton acceptor has already been obtained, no evidence for the participation of Ser205 in the proton transfer process exists. To examine the role of Ser205 in the proton transfer, we mutated Ser205 to valine. However, the derived GFP variant S205V, upon excitation at 400 nm, still produces green fluorescence. Time-resolved emission spectroscopy suggests that ESPT contributes to the green fluorescence, and that the proton transfer takes place approximately 30 times more slowly than in wt-GFP. The crystal structure of S205V reveals rearrangement of Glu222 and Thr203, forming a new hydrogen-bonding network. We propose this network to be an alternative ESPT pathway with distinctive features that explain the significantly slowed rate of proton transfer. In support of this proposal, the double mutant S205V/T203V is shown to be a novel blue fluorescent protein containing a tyrosine-based chromophore, yet is incapable of ESPT. The results have implications for the detailed mechanism of ESPT and the photocycle of wt-GFP, in particular for the structures of spectroscopically identified intermediates in the cycle.

The reversible proton dissociation and geminate recombination of a photoacid is studied as a function of temperature in water electrolyte solutions and binary water-methanol mixtures, containing 0.1 and 0.2 mole fractions of methanol. 8-Hydroxypyrene-1,3,6-trisulfonate trisodium salt (HPTS) is used as the photoacid. The experimental data are analyzed by the reversible geminate recombination model. We found that the slope of the logarithm of the proton-transfer rate constant as a function of the inverse of temperature (Arrhenius plot) in the liquid phase of these samples are temperature-dependent, while in the solid phase, the slope is nearly constant. The slope of the Arrhenius plot in frozen electrolyte solution is larger than that of the water-methanol mixtures, which is about the same as in pure water. Careful examination of the time-resolved emission in ice samples shows that the fit quality using the geminate recombination model is rather poor at relatively short times. We were able to get a better fit using an inhomogeneous kinetics model assuming the proton-transfer rate consists of a distribution of rates. The model is consistent with an inhomogeneous frozen water distribution next to the photoacid.

In a previous work, we proposed an extended model for intermolecular excited-state proton transfer to the solvent. The model invoked an intermediate species, the contact ion-pair RO(-)...H(3)O(+), where a proton is strongly hydrogen bonded to the conjugated photabase RO(-). In this study we tested the extended model by measuring the transient absorption and emission of 8-hydroxypyrene-1,3,6-trisulfonate (HPTS) in an aqueous solution in the presence of a large concentration of mineral acids. In a neutral pH solution, the pump-probe signal consists of three time components, <1, 4, and 100 ps. The 4 ps time component, with a relative amplitude of about 0.3, was attributed to the formation of the contact ion-pair and the long 100 ps component to the dissociation of the ion-pair to a free proton and RO(-). In the presence of acid, the recombination of an excess proton competes with the geminate recombination. At a high acid concentration, the recombination process alters the time-dependent concentrations of the reactant, product and intermediate contact ion-pair. We observed that when the acid concentration increases, the amplitude of both the long and intermediate time components decreases. At about 3 M of acid, both components almost disappear. Model calculations of the acid effect on the transient HPTS signal indeed showed that the amplitude of the intermediate time component decreases as the excess proton concentration increases.

A femtosecond pump-probe, with approximately 150 fs resolution, as well as time-correlated single photon counting with approximately 10 ps resolution techniques are used to probe the excited-state intermolecular proton transfer from HPTS to water. The pump-probe signal consists of two ultrafast components (approximately 0.8 and 3 ps) that precede the relatively slow (approximately 100 ps) component. From a comparative study of the excited acid properties in water and methanol and of its conjugate base in basic solution of water, we propose a modified mechanism for the ESPT consisting of two reactive steps followed by a diffusive step. In the first, fast, step the photoacid dissociates at about 10 ps to form a contact ion pair RO-*...H3O+. The contact ion pair recombines efficiently to re-form the photoacid with a recombination rate constant twice as large as the dissociation rate constant. The first-step equilibrium constant value is about 0.5 and thus, at short times, <10 ps, only approximately 30% of the excited photoacid molecules are in the form of the conjugated base-proton contact ion pair. In the second, slower, step, of about 100 ps, the proton is separated by at least one water molecule from the conjugate base RO-. The separated proton and the conjugated base can recombine geminately as described by our previous diffusion-assisted model. The new two-step reactive model predicts that the population of the ROH form of HPTS will decrease with two time constants and the RO- population will increase by the same time constants. The proposed model fits the experimental data of this study as well as previous published experimental data.

The pressure dependence of the excited-state proton dissociation rate constant of four photoacids, 2-naphthol-6,8-disulfonate (2N68DS), 10-hydroxycamptothecin (10-CPT), 5-cyano-2-naphthol (5CN2), and 5,8-dicyano-2-naphthol (DCN2), are studied in methanol. The results are compared with the results of the pressure dependence study we recently conducted for several photoacids in water, ethanol, and propanol. The pressure dependence is explained using an approximate stepwise two-coordinate proton transfer model. The increase in rate, as a function of pressure, manifests a strong dependence of proton tunneling on the distance which decreases with an increase of pressure between the two oxygen atoms involved in the process. The decrease in the proton transfer rate with increasing pressure reflects the dependence of the reaction on the solvent relaxation rate. We found that, for the relatively weak photoacids 2N68DS, 10-CPT, and 5CN2, the proton transfer rate constant increases by a factor of about 5-8 at a pressure of about 1.5 GPa. For a strong photoacid like DCN2, the rate increase was only by a factor of 2.

Steady-state and time-resolved fluorescence techniques were employed to study the excited-state proton transfer (ESPT) from a reversibly dissociating photoacid, 2-naphthol-6,8-disulfonate (2N68DS). The reaction was carried out in water and in acetonitrile–water solutions. We find by carefully analyzing the geminate recombination dynamics of the photobase–proton pair that follows the ESPT reaction that there are two targets for the proton back-recombination reaction: the original O− dissociation site and the SO3− side group at the 8 position which is closest to the proton OH dissociation site. This observation is corroborated in acetonitrile-water mixtures of χwater < 0.14, where a slow intramolecular ESPT occurs on a time scale of about 1 ns between the OH group and the SO3− group via H-bonding water. The proton-transferred R*O− fluorescence band in mixtures of χwater < 0.14 where only intramolecular ESPT occurs is red shifted by about 2000 cm−1 from the free R*O− band in neat water. As the water content in the mixture increases above χwater = 0.14, the R*O− fluorescence band shifts noticeably to the blue region. For χwater > 0.23 the band resembles the free anion band observed in pure water. Concomitantly, the ESPT rate increases when χwater increases because the intermolecular ESPT to the solvent (bulk water) gradually prevails over the much slower intramolecular via the water-bridges ESPT process. The isotope effect of the proton diffusion constant in ice Ih was studied by using a time-resolved emission technique. For that purpose we used three photoactive molecules that are sensitive to the excess mobile proton in ice. We found that the diffusion constant isotope effect was smaller by less than a factor of two in the temperature range of 240-270 K. Previously, we reported that the proton diffusion constant in ice is very large, in fact, 10 times greater than that of water at 295 K. [ABSTRACT FROM AUTHOR]

The water layers interspacing between the phospholipid membranes of a multilamellar vesicle are 3-10 water layers across and their width is adjusted by osmotic pressure (Parsegian, V.A., et al., 1986. Methods Enzymol. 127:400-416). In these thin water layers we dissolved pyranine (8 hydroxypyrene 1,3,6 trisulfonate), a compound which, upon photo excitation, ejects it hydroxy proton with time constant of 100 psec. (Gutman, M. 1986. Methods Enzymol. 127:522-538). In the present study we investigated how the width of the aqueous layer, the density of phosphomoieties on the membrane's surface and the activity of water in the layer affect the capacity of protons to diffuse out from the electrostatic cage of the excited anion before it decays to the ground state. Using a combination of steady-state and subnanosecond time-resolved fluorescence measurements we determined the average number of proton excited-anion recombinations before the proton escapes from the Coulomb cage. The probability of recombination in thin water layer is significantly higher than in bulk. The factor contributing most to enhancement of recombination is the diminished water activity of the thin aqueous layer. The time frame for proton escape from an electrostatic trap as big as a membrane-bound protein is 3 orders of magnitude shorter than turnover time of membrane-bound enzymes. Thus the effects of local forces on proton diffusion, at the time scale of physiological processes, is negligible. [ABSTRACT FROM AUTHOR]
