Placental insufficiency results in fetal loss, low birth weight, stillbirth, preweaning mortality and poor growth. Placental development begins at conceptus elongation, which is a primary factor controlling the size of the placenta. After elongation, the allantois develops outward from the embryo to establish the allantochorion, which defines the size of the functional placenta. During implantation, chorionic trophoblasts adhere to endometrial epithelial cells. Placental structures known as areolae develop at the openings of the endometrial glands and take up endometrial gland secreted products (histotrophe). Between day 30 and 35 of gestation, the adhered trophoblast-endometrial epithelial bilayer undergoes microscopic folding. Fetal and maternal capillaries develop adjacent to the bilayer and blood flows are arranged in a cross-countercurrent manner. Except for nutrients secreted by the glands, nutrient exchange takes place between these capillaries within these folds. By day 85, the folds deepen and become more complex, increasing surface area. The epithelial bilayer thins and capillaries indent the plane of each layer (but do not penetrate), reducing distance between capillaries. The folded bilayer is surrounded by endometrial stroma on the maternal side and placental stroma on the fetal side. The fetal-placental stroma is partially composed of glycosaminoglycans, the most abundant being hyaluronan and heparan sulfate. Changes in both hyaluronoglucosaminidase and heparanase during placental development suggest that these enzymes play a role in placental development. In addition to structural modifications, various nutrient specific transport mechanisms exist. These mechanisms are likely to be as important to transport of specific nutrients as placental size or structure.

The efficiency of nutrient transport from the pregnant female pig to the developing fetus depends on the size and function of the placenta. It has been reported that maternal and fetal blood vessels are arranged in a cross-countercurrent arrangement within placental microscopic folds. Thus, the blood supplies are in close apposition to each other within these microscopic folds, and maternal and fetal blood flows in approximately opposite directions perpendicular to the plane of the placenta. This arrangement indicates that the width of the microscopic folds influences placental efficiency. The objective of this study was to determine whether differences in pig placental microscopic fold development are associated with differences in fetal size or are influenced by selection for ovulation rate or uterine capacity. Gilts from a randomly selected control line, a line selected for ovulation rate, and a line selected for uterine capacity were slaughtered, and uterine wall samples were collected within the placentas associated with the largest and smallest fetuses in each litter on d 45, 65, 85, and 105 of gestation. The uterine wall samples were processed for histology and analyzed using computer-assisted morphometry. Average width of the placental folds and average width of the placental stroma above the folds were measured. To measure fold complexity, the length of the epithelial bilayer for a given length of placenta was also measured. The width of the folded bilayer increased significantly from d 65 to 105 and was greater in placentas associated with small fetuses compared with large fetuses on d 105 of gestation. In contrast, the width of the placental stroma above the folded bilayer decreased with gestation and decreased more rapidly in placenta associated with the smallest compared with the largest fetus. These results indicate that the width of the microscopic folds of the placental trophoblast/endometrial epithelial bilayer is increased in placenta associated with small fetuses, which we hypothesize will increase the surface area for interaction between maternal and fetal blood supplies, thus improving placental efficiency in response to reduced placental size.

Previous experiments indicated that secreted (s) and membrane (m) forms of folate binding protein (FBP) are present in the intrauterine environment of the pig. Our previous studies indicated that the two forms were produced sequentially; the secreted form was present in the intrauterine glands until Day 20 of gestation, whereas binding analysis indicated that folate binding increased dramatically in placental membranes until Day 50 of gestation. However, the cell types expressing mFBP have not been investigated. In this experiment, uterine wall sections from Day 20, 35, 50, 70, 90, and 105 of gestation were collected at slaughter and fixed, and subjected to in situ hybridization analysis for mFBP expression. The mFBP mRNA was expressed by both columnar and cuboidal epithelia of the placental folds and expression appeared to be similar throughout gestation. Therefore, the placenta expressed mFBP from Day 35 of gestation onward, consistent with the concept that sFBP and mFBP occur sequentially during gestation in swine, and that placental mFBP expression plays a role in folate transport after a functional chorioallantoic placenta is established (between Day 20 and 35).

A study was conducted to evaluate the influence of maternal cortisol on early conceptus development in pigs (Sus scrofa). The corticosteroid synthesis inhibitor metyrapone was injected daily during days 14-19 of pregnancy, without (n = 6) and with commensurate administration of cortisol (n = 6). Blood samples were taken via an indwelling jugular catheter on days 14 and 18, and conceptuses were harvested during surgery on day 20. Compared with vehicle-injected control dams (n = 7) plasma cortisol and aldosterone concentrations were decreased (P < 0.01) by 52 and 29%, respectively, by metyrapone treatment. Cortisol administration reversed decreases in plasma cortisol by day 18. There were no treatment-associated effects on conceptus survival or size. Nor were there treatment-associated effects on allantoic fluid volume or content. Trophodermal glucocorticoid receptor (GR) mRNA expression decreased by 34% (P < 0.05) in metyrapone-treated pigs, and was not further influenced by concomitant administration of cortisol, thereby suggesting an influence of aldosterone on GR mRNA expression. Also, when all pigs were considered, there were treatment-independent second-order polynomial regressions (P < 0.05) between maternal plasma cortisol concentrations and embryonic weight, allantoic size and allantoic glucose concentrations, and between plasma aldosterone concentrations and trophodermal GR mRNA expression. Such biphasic corticosteroid concentration versus tissue parameter curves are noteworthy, but difficult to interpret validly. They may suggest that an appropriate corticosteroid environment is necessary for optimal porcine embryonic development during this stage of gestation, but cannot overshadow the absence of treatment effects on the porcine embryonic measures evaluated.

The objective was to understand how two forms of folate-binding protein interact to accomplish folate transport during pregnancy in swine. Specific folate binding was measured in uterine flushings during the estrous cycle and early pregnancy and in allantoic fluid (secreted form) and placental membranes (membrane form) throughout later pregnancy. In addition, the localization of the secreted form of folate-binding protein (sFBP) in uterine wall sections was assessed. Uterine flushings were collected on Days 10, 13, and 15 of the estrous cycle and pregnancy. Allantoic fluid and placentas were collected on Days 20, 35, 50, 70, 90, and 105 of pregnancy. Uterine-wall sections were collected on all days of the experiment. Folate binding was measured by incubation of aliquots of uterine flushings, allantoic fluid, or placental microsomal membranes with 0.5-4 nM [(3)H]folate. Uterine-wall sections were incubated with purified anti-FBP IgG or normal rabbit serum IgG to localize sFBP. Folate binding did not differ between early pregnancy and the estrous cycle in uterine flushings, was greatest from Day 50 to 70 of pregnancy in allantoic fluid, and was greatest from Day 50 of pregnancy onward in placental microsomal membranes. Staining for sFBP was present in the endometrial glands from Day 10 to 15 in cyclic gilts and from Day 10 to 20 in pregnant gilts. The pattern of folate binding and sFBP staining supports the concept that sFBP transports folate to the developing conceptus until placentation and then the placental form takes over folate transport.

During rapid development of the fetus, levels of high density lipoprotein (HDL) are elevated in pregnant women. The receptor for HDL, scavenger receptor class B type I (SR-BI), mediates selective cholesteryl ester uptake and is highly expressed in the human placenta. Because of the rapid growth of uterus during early pregnancy and differences in placentation between swine and humans, we hypothesized that SR-BI may be expressed in porcine endometrium to take up HDL cholesterol. The objectives of this study were to obtain the full coding region for porcine SR-BI, determine endometrial expression of SR-BI mRNA during the estrous cycle and early pregnancy, and map the gene. By iterative screening of a porcine expressed sequence tag library, we obtained the full coding region of SR-BI. Endometrial expression of SR-BI in White composite gilts (n = 3-4 each) was determined by Northern blotting on Days 10, 13, and 15 cyclic gilts and Days 10, 13, 15, 20, 30, and 40 pregnant gilts. In cyclic gilts, endometrial expression of SR-BI did not change between Days 10 and 13, but increased (P < 0.01) between Days 13 and 15. In pregnant gilts, endometrial expression of SR-BI increased (P < 0.01) between Days 10 and 13, remained elevated until Day 30, and decreased (P = 0.015) on Day 40. The SR-BI gene was mapped to 46.3 cM on chromosome 14. These results show that endometrial expression of SR-BI changes during the estrous cycle and early pregnancy, and suggest that SR-BI takes up HDL for endometrial development during early pregnancy.

The interrelationships among d-11 conceptus size, d-105 placental weight, placental efficiency (the ability of the placenta to support fetal growth and development), fetal erythropoiesis, and uterine capacity were examined in 1/2 Meishan, 1/2 White crossbred gilts that were unilaterally ovariohysterectomized at 90 to 100 d of age. In Exp. 1, gilts were mated after at least one normal estrous cycle and then slaughtered at 105 d of gestation and number of fetuses and CL, placental weights, fetal weights, hematocrits, fetal plasma iron, and fetal plasma folate were measured. In Exp. 2, gilts were mated and plasma progesterone was measured on d 2 and 3 of gestation. On d 11, the length of the remaining uterine horn was recorded and the uterine horn was flushed with minimal essential medium. Number of CL, conceptus number, conceptus diameters, and total uterine flush retinol-binding protein (tRBP), acid phosphatase (tAP), and folate-binding protein (tFBP) were measured. Gilts were mated again and slaughtered at 105 d of pregnancy and the same traits measured in Group 1 were recorded. Plasma progesterone concentrations on d 2 and 3 were correlated with average conceptus diameter on d 11 (r = 0.60, P < 0.01, for each day). In contrast, tRBP (r = 0.49, P < 0.01), tAP (r = 0.53, P < 0.01), and tFBP (r = 0.51, P < 0.01) in uterine flushings on d 11 were only correlated with d-3 plasma progesterone concentrations. No correlations between d-11 average conceptus diameter or d-11 uterine length with d-105 uterine capacity were observed. Uterine capacity was negatively correlated with placental weight, fetal weight and fetal hematocrit (r = -0.36, P < 0.01; r = -0.44, P < 0.01; r = -0.32, P < 0.01; respectively). Hematocrits were correlated with fetal plasma iron (r = 0.50, P < 0.01) and folates (r = 0.44, P < 0.01). Hematocrit, plasma iron, and plasma folate were each correlated with residual fetal weights after adjusting for placental weight (a measure of placental efficiency), and accounted for 11% of the variation in this trait. These data suggest that conceptus diameter and uterine protein secretion on d 11 may be influenced by the onset of progesterone secretion by the CL, but do not support an influence of conceptus growth during early pregnancy on uterine capacity. These results also suggest that reducing placental and fetal weights will likely result in increased uterine capacity.

Two distinct forms of endometrial folate binding protein (FBP) cDNAs were isolated using reverse transcription-polymerase chain reaction and 3' and 5' rapid amplification of cDNA ends (RACE) procedures. On the basis of the absence or presence of an intact glycophosphatidylinositol linkage site in the C terminus of the predicted amino acid sequences, the two forms appear to encode secreted and membrane-bound forms of FBP. The cDNAs for the putative secreted and membrane forms encoded 252- and 249-amino acid proteins, respectively, that were 73% identical with each other and were 66-82% identical with other known FBPs. However, the nucleotide sequences within the 5' untranslated region and from codons 224 and 223 of the secreted and membrane forms, respectively, to the 3' ends of each RNA, were divergent. The divergence in the 3' ends of the two cDNAs was exploited to determine changes in concentrations of each mRNA in the endometrium during the estrous cycle and early pregnancy. Northern blots of endometrial total RNA probed with a putative secreted FBP specific probe indicated that mRNA concentrations do not change during early pregnancy. In contrast, blots probed with a putative membrane FBP specific probe indicated that mRNA concentrations increase dramatically from Day 15 to Day 24 of pregnancy. Finally, N-terminal amino acid sequencing of FBP purified from Day 15 pregnant uterine flushings matched the secreted form of FBP mRNA. These data are consistent with a role for putative secreted and membrane-bound forms of FBPs in the transport of folate to the developing swine conceptus during early pregnancy.

This experiment consisted of the following treatment-breed groups: 1) White crossbred gilts, 2) White crossbred gilts treated with progesterone (200 mg/d in corn oil given on d 2 and 3 after estrus), and 3) Chinese Meishan gilts. Pregnant and nonpregnant gilts (n=3 to 6) from each treatment-breed combination were assigned to be slaughtered on d 10, 11, 12, 13, and 15. At slaughter each uterine horn was flushed with 20 mL of minimal essential medium. Uterine flushings were assayed for total protein, acid phosphatase, uteroferrin, retinol-binding protein, and oxytocin. Uterine flush total protein was increased by progesterone treatment, was unaffected by pregnancy status, and was less in Meishans. Similar patterns were found for retinol binding protein and uteroferrin, except that uteroferrin was greater in pregnant than in nonpregnant gilts. Oxytocin was greater in pregnant than in nonpregnant gilts, was not influenced by progesterone treatment, and was similar in Meishan and in White crossbred gilts. These results indicate that the conceptus does not influence secretion of either total protein or retinol binding protein during pregnancy and that the onset of secretion of these uterine proteins may be controlled by progesterone. The presence of the conceptus is associated with increased uteroferrin and oxytocin production. The decreased secretion of uterine proteins in Meishan gilts may partially explain the slower embryonic development that has been reported for this breed.

Studies were conducted to determine whether the corticosteroids cortisol and aldosterone, and corticosteroid-binding globulin (CBG) were present in the porcine early-embryonic environment. Cortisol was measured in uterine flushings from white crossbred gilts at Days 7, 10, 13, and 16 of the estrous cycle and pregnancy. Total content of cortisol increased (p < 0.01) between Days 13 and 16, and immunoreactive CBG (ir-CBG) increased (p < 0.01) between Days 10 and 13, in both cyclic and pregnant gilts. In a separate study with Chinese Meishan gilts, total cortisol and aldosterone content of uterine flushings increased (p < 0.02) between Days 10 and 15 of the estrous cycle and pregnancy. In another study with white crossbred gilts, CBG-like binding activity in uterine flushings was low at Day 10, then increased over 100-fold at Day 15 (p < 0.01). However, levels of CBG-like binding activity on Day 15 were 100-fold lower than those of ir-CBG measured in the previous study and could bind less than 4% of the uterine luminal cortisol. Differences between ir-CBG and CBG binding might be due to the ability of the CBG antibody to recognize either biologically inactive CBG or structurally similar molecules. CBG-like binding activity, which appeared unrelated to glucocorticoid receptors, was also present in the endometrial cytosol of white crossbred gilts. Concentrations (fmol/mg protein) of endometrial CBG-like activity decreased (p = 0.03) between Days 10 and 15 of the estrous cycle and pregnancy, did not differ with reproductive status, and on Day 15 were comparable to concentrations in uterine flushings but threefold lower (p < 0.01) than those in the serum. Equilibrium dissociation constants for CBG-like binding activities were comparable among the three locations. These studies indicate that corticosteroids are present-primarily in the free form-within the porcine uterine lumen and could influence early porcine conceptus development. Endometrial CBG-like binding activity could mediate actions of cortisol or progesterone on uterine function.

There are periods during pregnancy when sows may have a temporally high requirement for certain vitamins and minerals. Proteins transferring retinol and Fe to the developing pig fetus have been discovered, whereas transport mechanisms for other vitamins and minerals are probably present but have not yet been identified. Sow body tissues can serve as a reservoir for many micronutrients, but it is not known whether these reserves can supply an adequate quantity during critical fetal developmental periods. There is a low placental transfer of vitamin E to the fetus even if the dietary concentration fed to a gestating animal is high, but colostrum and milk concentrations can be increased when the nutrient is fed to sows. If the dam's diet contains inadequate Ca or P, the concentration of these elements in the developing fetus and milk will not be affected. Consequently, sow bone demineralization will occur under conditions of dietary inadequacy of Ca and P. Other nutrients can be depleted from sow tissue reservoirs over several parities (e.g., Se), resulting in low quantities being provided in the milk for nursing pigs. Scientific information involving adequate vitamin and mineral nutrition for female pigs to improve conception rate and embryonal survival that will result in optimum fetal and postnatal pig development can be considered to be in its infancy.

It has been hypothesized that conceptus estrogens influence endometrial protein secretion during pregnancy in swine. To test this hypothesis, the effect of estrone and estradiol treatment from d 30 to 60 of pregnancy or pseudopregnancy on endometrial protein secretion was investigated. Pregnant (P; n = 16) and pseudopregnant (PP) gilts (n = 18) received either sham treatment or estrone or estradiol implants (5 mg/d release rate; 60 d release) on d 30 of P or PP. Blood samples were collected on d 30, 40, 50, and 60 to measure estrone and estradiol. On d 60, gilts were hysterectomized. For P gilts, endometrium in apposition to one placenta from each uterine horn was collected. For PP gilts, each uterine horn was flushed with 40 mL of leucine-deficient minimal essential medium (MEM), and endometrial tissue was collected from each horn. Endometrial tissues were incubated in MEM in the presence of 50 microCi of [3H]leucine to examine protein secretion. Estrone and estradiol treatments increased both plasma and endometrial concentrations of estrone (P < .01 except endometrium for P gilts) and estradiol (P < .01, respectively). Endometrium from P gilts secreted more nondialyzable macromolecules (NDM), acid phosphatase activity (AP, a measure of uteroferrin), and retinol-binding protein (RBP) in culture than did endometrium from PP gilts. Estrone treatment increased (P < .01) endometrial NDM from P gilts but not that from PP gilts; estradiol had no effect. Both estrone and estradiol increased (P = .069) endometrial secretion of AP of PP but not of P gilts. Endometrial secretion of RBP was not affected by either estrone or estradiol treatment. Neither estrone nor estradiol affected total protein or AP and estrone treatment decreased (P < .05) RBP in uterine flushings from PP gilts. These data indicate that endometrium from P pigs secretes more protein than endometrium from PP pigs but neither estrone nor estradiol completely mimics the effect of pregnancy.

Secretion of proteins by the endometrium of pigs during pregnancy is reported to be influenced by progesterone and estrogen. The effect of estrone treatment from d 30 to 45 of pregnancy on endometrial protein secretion and uterine capacity was tested in unilaterally hysterectomized-ovariectomized (UHO) gilts. Ten intact and 16 UHO crossbred gilts were mated and assigned to the following treatments: 1) intact controls, n = 10; 2) UHO, n = 8; and 3) UHO + estrone (E; two 150-mg implants, 60-d release, implanted on d 30 of gestation), n = 8. Gilts were killed on d 45 of pregnancy. A sample of allantoic fluid was collected from each conceptus. Blood and endometrial tissue samples were collected to evaluate estrone concentrations, and placental and endometrial tissues were incubated to evaluate protein secretion. Estrone treatment increased (P < .01) plasma and endometrial tissue estrone concentrations. Unilateral hysterectomy-ovariectomy decreased (P < .01) fetal survival. No effect of estrone was detected on the number of live fetuses, protein secretion by endometrial tissue in culture, or allantoic fluid protein, acid phosphatase, retinol binding protein, or retinol. Estrone treatment decreased uterine weight (P = .05) and allantoic fluid fructose concentrations (P < .05). After correction for allantoic fluid volume, allantoic fluid protein, retinol, and acid phosphatase activity were decreased (P < .05) in UHO-treated pigs. The UHO treatment did not affect allantoic fluid fructose concentrations. These results indicate that 1) estrone treatment from d 30 to 45 of pregnancy and at the dosage used did not alter endometrial protein secretion or fetal survival, and 2) UHO decreased the amount of endometrial proteins available to conceptuses.

The effect of altering uterine space available to developing conceptuses on placental and endometrial protein secretion and endometrial 3H-thymidine incorporation on Days 25 and 35 of gestation was tested. Gilts bred at estrus (Day 0) were laparotomized on Day 4, and the uterine horn ipsilateral to the ovary with the most ovulations was ligated midway between the uterine tip and uterine bifurcation to create a crowded and a roomy uterine environment. On Day 25 (n = 7) or 35 (n = 6), gilts were killed and the reproductive tract was dissected from each. Each placenta and fetus was weighed, and a sample of endometrium adjacent to each placenta was collected. Within each uterine environment, tissues (500 mg) from the heaviest and lightest (by weight) placentae and adjacent endometrial tissue were cultured for 24 h at 37 degrees C in 15 ml Minimal Essential Medium having 0.1 times the normal amount of leucine plus 50 microCi 3H-leucine. Aliquots (2 ml) of dialyzed medium were subjected to two-dimensional PAGE and fluorography to examine proteins with molecular weight ranges of approximately M(r) 20,000-130,000, and a further aliquot was subjected to SDS-PAGE to examine low molecular weight proteins (M(r) 2000-20,000). In addition, endometrium (200 mg in duplicate) adjacent to the heaviest and lightest placentae within each uterine environment was incubated for 4 h in 5 ml MEM plus 1 microCi 3H-thymidine to measure DNA synthesis. After culture, 3H-thymidine incorporation and total DNA were determined. Fetal survival in the crowded uterine environment decreased (p < 0.05) on Day 35 compared to Day 25. On both days, the heaviest and lightest placentae from the crowded uterine environment weighed less (p < 0.01) than the heaviest and lightest placentae from the roomy uterine environment. The fetuses associated with the heaviest and lightest placentae from the crowded uterine environment also weighed less (p < 0.05) than fetuses associated with the heaviest and lightest placentae from the roomy uterine environment. Nondialyzable radioactivity in medium per gram of tissue cultured from placental cultures decreased (p < 0.05) and from endometrial culture increased (p < 0.05) between Days 25 and 35. Placental secretion of a protein with M(r) 46,000, pI 5, decreased between Days 25 and 35. Endometrial secretion of three proteins (M(r) 35,000, pI 6.2; M(r) 25,000, pI 4.9; and M(r) 14,000, pI unknown) increased, and secretion of three other proteins (M(r) 24,000, pI 6.5 doublet; M(r) 22,000, pI 7.5, and M(r) 7000, pI unknown) decreased from Day 25 to Day 35 of pregnancy.(ABSTRACT TRUNCATED AT 400 WORDS)

The Meishan pig provides a biological model with the genetic capacity to express a high prolificacy. This prolificacy can be partially attributed to a higher ovulation rate and a higher rate of prenatal survival at a given ovulation rate throughout gestation than in European breeds. Both early embryonic survival (factors inherent to the ovum and uterus, which occur before day 25 of gestation) and uterine capacity (factors inherent to uterine limitation, which occur from 30 days of gestation to parturition) may contribute to prenatal survival. Crossbreeding studies show that the prolificacy of Meishan pigs is primarily of maternal origin, but not whether the effects occur via the ovum, uterus or systemic factors. Although there have been few comparative studies on uterine function for prolific pigs, experiments relating to three general areas have been reported. During neonatal uterine development, endometrial gland development occurs shortly after birth in Meishan and European breeds. Tissue culture for 24 h showed that secretion of at least three uterine proteins (one identified as retinol-binding protein) increased in clear temporal association with development of the endometrial gland. The secretion of two additional proteins appears to change in a breed-specific manner. Although regulatory roles for uterine proteins remain to be defined, biochemical events associated with neonatal endometrial gland development may affect subsequent adult uterine function. During the time of maternal recognition of pregnancy or early gestation, reports suggest that uterine secretions are enhanced in Meishan pigs, which may partially explain their increased prolificacy. However, other studies show smaller embryo and placental weights at day 30 of gestation, suggesting that prolificacy of Meishan females is related to a maternal ability to control embryo/fetal and placental growth. During the later fetal period (uterine capacity) in European pigs, endometrial response to a crowded uterine environment is minimal, but the conceptus responds to a crowded uterine environment by altering placental function; similar experiments using Meishan pigs are in progress. Once the regulatory determinants of uterine function are understood, improvements can possibly be made in the reproductive efficiency of all swine.

A type I interferon (IFN) secreted by the trophoblast of early sheep and cow embryos is thought to be responsible for the maternal recognition of pregnancy. The expression of trophoblast IFN is tissue specific and temporally controlled. However, the isolated bovine trophoblast IFN promoter did not confer tissue specificity on the expression of a bacterial chloramphenicol acetyl transferase (CAT) reporter gene, and could not be induced by virus, unlike other type I IFNs. Trophoblast IFN acts locally within the uterus to prevent luteolysis and prolong progesterone secretion. Endometrial IFN receptors are present, and trophoblast IFN decreases expression of endometrial oxytocin receptor and increases expression of endometrial beta 2-microglobulin, MHC class I antigens and Mx (a mediator of IFN antiviral activity) only in the pregnant horn of pregnant ewes with a transected uterus. The primary effect of trophoblast IFN during early pregnancy appears to be an inhibition of oxytocin receptor expression, although studies in ovariectomized ewes suggest that luteal oxytocin may be required to facilitate the inhibition of prostaglandin F secretion by trophoblast IFN. An investigation of the isolated oxytocin receptor promoter should confirm its critical role in the maternal recognition of pregnancy.

In Expt 1, activity of 2',5'-oligoadenylate (2',5'-A) synthetase in endometrium collected on Day 16 (oestrus is Day 0) from the uterine horn ipsilateral to the corpus luteum was greater (P less than 0.001) for pregnant (135.5 +/- 1.72 nmol/mg protein/h) than for cyclic ewes (58.5 +/- 0.99 nmol/mg protein/h). In pregnant ewes, activity of 2',5'-A synthetase in endometrium collected from the contralateral uterine horn (119.5 +/- 1.72 nmol/mg protein/h) did not differ from that of the ipsilateral horn. In Expt 2, three ovariectomized ewes were treated with progesterone for 10 days and then with oestrogen for 2 days. Activity of 2',5'-A synthetase on Day 13 was 18% greater (P less than 0.10) in endometrium collected from the uterine horn receiving infusions of 30 micrograms ovine trophoblast protein-1 (oTP-1) twice a day on Days 10, 11 and 12(57.7 +/- 0.22 nmol/mg protein/h) than from the uterine horn receiving control infusions of serum protein (SP; 48.8 +/- 0.22 nmol/mg protein/h). In Expt 3, activity of 2',5'-A synthetase on Day 15 was not significantly greater in endometrium collected from the uterine horn of cyclic ewes receiving infusions of 30 micrograms oTP-1 twice a day on Days 12, 13 and 14 (46.5 +/- 0.37 nmol/mg protein/h) than in endometrium from the uterine horn receiving infusions of SP (38.2 +/- 0.37 nmol/mg protein/h). When results of Expt 2 and Expt 3 were combined, intrauterine infusion of oTP-1 increased (P less than 0.05) activity of 2',5'-A synthetase in endometrium by 20%.(ABSTRACT TRUNCATED AT 250 WORDS)

Ovine trophoblast protein-1 (oTP-1), stimulates the secretion of several proteins in explant culture of day-12 cyclic ovine endometrium. We partially purified and identified one of these proteins, an 11,000 Mr, pI approx. 6 protein by N-terminal amino acid sequencing and immunoprecipitation using antibody to human beta 2-microglobulin. The protein was purified from cultures of endometrium collected from day-16 pregnant ewes. The N-terminal amino acid sequence was 40-55% homologous to beta 2-microglobulin from a variety of species. Antibody to human beta 2-microglobulin immunoprecipitated the protein and another protein of similar molecular weight but more acidic pI. Using immunoprecipitation of radiolabelled proteins from culture, we demonstrated that oTP-1 increased production of this protein by 40% (P less than 0.05). We conclude that oTP-1 increases the secretion of a beta 2-microglobulin-like protein from day-12 non-pregnant endometrium in culture.

Southern blotting of bovine genomic DNA indicated the presence of at least 3 bovine tIFN genes. The full DNA sequence of one of these genes, thought to be expressed in trophoblast, has been determined, including 193 bp of 5' non-coding region. The inferred amino acid sequence of bovine tIFN is more similar to ovine tIFN (80%) than to bovine IFN-alpha II (70%). The 5' flanking sequence has some similarity with bovine IFN-alpha II, and may contain a viral response element. A recombinant bovine alpha I interferon (Ciba Geigy; brIFN), resembling tIFN, extended oestrous cycle length in sheep when administered by intrauterine infusion over the period, Days 12-15 after oestrus, when maternal recognition of pregnancy occurs. Intramuscular injection was only effective at the doses used if given over a longer period (i.e. Days 9-15). Our experiments indicate that both tIFN and brIFN inhibit luteolysis by preventing a rise in endometrial oxytocin receptor concentrations, and suggest that tIFN achieves this by extending the time for which progesterone suppresses oxytocin receptor development. Further studies are required to confirm this hypothesis and to elucidate the interaction of the effects of progesterone and tIFN in endometrial cells.

Three experiments (Exp) assessed the influence of stage of the estrous cycle, pregnancy, and intrauterine infusion of ovine conceptus secretory proteins (oCSP) on turnover of inositol trisphosphate (the putative second-messenger for oxytocin-stimulated secretion of prostaglandin F2 alpha) in ovine endometrium during luteolysis and maternal recognition of pregnancy. In Exp 1, endometrium was collected from 5 cyclic (Cy) and 6 pregnant (P) ewes on Day 16 after onset of estrus. In Exp 2, endometrium was collected from Day 12 Cy (n = 5), Day 12 P (n = 3), Day 16 Cy (n = 4), and Day 16 P (n = 3) ewes. In Exp 3, 12 Cy ewes were allotted randomly, in a 2 x 2 factorial arrangement, to receive serum protein (SP), or oCSP and estradiol-17 beta (E2), or vehicle treatments. Ewes were injected i.v. with 0.5 mg E2 or vehicle on Day 12 and received twice-daily infusions of 1.5 mg SP or oCSP (containing 25 micrograms ovine trophoblast protein-1 by radioimmunoassay [RIA]) + SP (1.5 mg total protein) into each uterine horn on Days 12, 13, and 14. Blood samples for RIA of plasma progesterone were collected on Days 10-15 (before treatment on each day) and endometrium was collected on Day 15. For each Exp, 100 mg endometrium was incubated, in duplicate, for 2 h with 10 microCi [3H] inositol and treated with 0 or 100 nM oxytocin (OT) for 20 min, then [3H]inositol mono-, bis-, and trisphosphates (IP1, IP2, and IP3, respectively) were quantified.(ABSTRACT TRUNCATED AT 250 WORDS)

Sheep conceptuses secrete a protein, oTP-1, between Days 10 and 21 of gestation which is responsible for establishment of pregnancy. oTP-1 inhibits uterine production of luteolytic amounts of PGF-2 alpha (PGF) produced in response to oestradiol and oxytocin. oTP-1 does not compete with oxytocin for binding to oxytocin receptors, but may interfere with oxytocin stimulation of the inositol phospholipid system. Pig conceptuses secrete oestrogens between Days 10 and 15 of pregnancy which are essential for establishment of pregnancy. Oestrogens, directly or indirectly, alter secretion of PGF from an endocrine direction (towards uterine vasculature) to an exocrine direction (towards the uterine lumen). PGF sequestered in the uterine lumen is unavailable to exert a luteolytic effect on the CL. Pig conceptus secretory proteins stimulate uterine production of PGF and PGE. Conceptus secretory proteins of sheep and pigs include proteins which have antiviral activity and may be considered interferons. In sheep, oTP-1 has both antiluteolytic and antiviral properties. The specific pig conceptus secretory proteins(s) possessing antiviral activity has not been established. Unlike oTP-1, however, it does not appear to possess antiluteolytic activity.

Pregnancy and intrauterine infusion of ovine trophoblast protein one (oTP-1) decrease oxytocin-induced secretion of prostaglandin F2 alpha (PGF) from the uterus. In the present study, effects of oTP-1 and pregnancy on endometrial secretion of PGF were examined in an in vitro perifusion system. In Experiment 1, endometrium from day 14 pregnant and cyclic ewes was perifused sequentially on both the lumenal and myometrial sides with Krebs Ringers Bicorbonate solution (KRB), KRB plus oxytocin (1 IU/ml) and KRB alone. Endometrium from pregnant ewes secreted more PGF from both lumenal and myometrial sides than endometrium from cyclic ewes (P less than 0.05). Oxytocin stimulated secretion of PGF from both sides of endometrium regardless of status. Secretion of PGF was greater from the lumenal surface of endometrium compared to myometrium (P less than 0.05) for pregnant and cyclic ewes. For Experiment 2, endometrium was collected from day 15 cyclic ewes and perifused sequentially with KRB, KRB plus 300 ng/ml of either Bovine Serum Albumin (BSA) or oTP-1, KRB with or without BSA or oTP-1 plus oxytocin (1 IU/ml) and then KRB alone. Oxytocin stimulated greater release of PGF from oTP-1-treated than BSA-treated endometrium. Pretreatment of endometrium with oTP-1 had the same effect on oxytocin-induced PGF secretion as cotreatment with oTP-1 and oxytocin. In Experiment 3, uterine horns of cyclic ewes were catheterized on day 10 of the estrous cycle, and infused with either oTP-1 or day 16 pregnant sheep serum proteins on days 12, 13 and 14. Endometrium was collected on day 15 and perifused sequentially with KRB, KRB plus oxytocin (1 IU/ml) and then KRB alone. Treatment of ewes with oTP-1 attenuated endometrial secretion of PGF in response to oxytocin. Results of this study indicate that: (1) pregnancy stimulates basal secretion of PGF from endometrium and has no effect on oxytocin-induced secretion of PGF in vitro; (2) short-term oTP-1 treatment enhances oxytocin-induced PGF secretion from day 15 cyclic endometrium and (3) long-term oTP-1 treatment in vivo inhibits oxytocin-induced PGF secretion in ewes.

The objective of the present study was to identify and characterize in a limited manner the major de novo oviductal secretory proteins (OSP) synthesized and released by the porcine oviduct. Oviductal tissue was collected on various days of the estrous cycle (EC) and early pregnancy (EP) and cultured in a modified minimal essential medium supplemented with 100 muCi L-[3H]-leucine. Oviductal secretory activity, as measured by the rate of incorporation of 3H-leucine (dpm/mg wet tissue weight) into nondialyzable macromolecules, was greatest (P less than .01) between days 0 and 2 and reached its lowest levels on days 10 to 15. There was no difference between left and right side or pregnancy status. This increased rate of incorporation at proestrus and estrus is temporally associated with elevated levels of estrogen. Incorporation rate for ampulla was greater than for the isthmus. Analysis of oviductal culture medium by one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis and fluorography revealed three protein bands of relative molecular weight (Mr) 335,000, 115,000, and 85,000, which were associated with proestrus, estrus, and metestrus and were not detectable on other days. All three proteins also incorporated 3H-glucosamine. The 115,000 Mr band was the major 3H-glucosamine-labeled protein. Two protein bands (Mr 60,000 and 20,000) were expressed with increasing progesterone during diestrus. Other de novo synthesized protein bands appear to be present throughout the EC and EP with little modulation by estrogen or progesterone. Thus, this study demonstrates that for the porcine oviduct, the increase in the incorporation rate of 3H-leucine into OSP by both whole oviduct and ampulla and de novo synthesis and secretion of three glycoproteins, Mr 335,000, 115,000, and 85,000, were associated with proestrus and estrus when events such as fertilization and early cleavage stages of embryo development occurred.

A radioimmunoassay has been developed for quantitation of ovine trophoblast protein-1 (oTP-1), a sheep conceptus secretory protein which allows for maintenance of the corpus luteum during early pregnancy. The assay was validated for dialysed and undialysed culture medium and pregnant uterine flushings ranging from no dilution (neat) to dilutions of 1:2500 for dialysed media, 1:100-1:1000 for undialysed media and 1:50-1:1000 for pregnant uterine flushings. The assay accurately measured oTP-1 added to undiluted and diluted dialysed and undialysed culture media and pregnant uterine flushings. No cross-reaction was detectable for bovine alpha or gamma interferon, bovine calmodulin, feline conceptus secretory proteins, equine conceptus secretory proteins, porcine conceptus secretory proteins, bovine conceptus secretory proteins and proteins in a uterine flushing collected from a non-pregnant ewe. Immunoreactivity in the assay matched that for oTP-1 throughout oTP-1 purification. This assay is the first validated assay which may be used to quantitate production of oTP-1 in culture or content of oTP-1 in uterine flushings.

Conceptuses produce steroids, prostaglandins, proteins and possibly other unidentified agents which may play a role in the establishment and maintenance of pregnancy. A key event in this process is protection of the corpus luteum (CL) from the luteolytic activity of prostaglandin (PG) F-2 alpha of uterine origin. Oestrogens produced by the pig conceptuses between Days 11 and 16 appear to exert an antiluteolytic effect resulting in the sequestering of PGF-2 alpha within the uterine lumen. Failure of the pregnant uterus to release PGF-2 alpha in an endocrine fashion, therefore, allows for maintenance of CL function. Conceptuses of sheep and cattle produce proteins which, when introduced into the uterine lumen of nonpregnant ewes and cows, suppress the ability of oestradiol and oxytocin to stimulate uterine production of PGF-2 alpha. These conceptus secretory proteins appear to exert an antiluteolytic effect by inhibiting uterine production of luteolytic amounts of PGF-2 alpha. The horse conceptus produces both oestrogens and proteins during early pregnancy when uterine production of PGF-2 alpha is suppressed. Co-culture of horse endometrium and conceptus inhibits endometrial production of PGF-2 alpha. Conceptuses of pigs, sheep and cattle undergo elongation to achieve apposition between trophectoderm and endometrium but the horse embryo migrates rapidly and consistently throughout the uterus to achieve endometrial contact.
