The vascular network of pregnant rabbit ovaries was studied by means of scanning electron microscopy (SEM) of corrosion casts, in order to evaluate the morphofunctional changes of the microcirculation of corpus luteum (CL). Pregnant rabbit ovary showed an overwhelming vascularization. Ovarian hilus displayed an increase in the arterial spirallisation. The arterial spiral pattern was present along the entire vessel course, up to CL tissues. The CL of pregnancy was supplied by wide vascular plexuses (2-5 plexuses were found in each pregnant ovary) whose major axis was about 2 mm. Luteal capillaries showed a tortuous course and were arranged in a three-dimensional, wide and rounded-meshed network. Postcapillary venoconstrictions were present. The venous drainage appeared more developed then the arterial supply. Tight artero-venous contacts in hilar, juxtamedullar and medullar regions of the ovary were observed. These results clearly show that the morphofunctional expression of CL of pregnancy is greatly dependent on its hemodynamic control. In particular, the increase of spirallisation exhibited by the arteries during pregnancy is likely to be considered a significant functional change. The spirallisation likely is a device for reducing the blood pressure through the CL. The artero-venous contacts, also previously described in hCG stimulated (pseudopregnant) ovaries, may support a counter-current like system that may allow a veno-arterial exchange of small molecules through the wall of the facing vessels. In addition, in 10-day pregnant rabbit CL the consolidation of a well-developed capillary network was revealed, which is a sign that the CL of pregnancy reached the full morphofunctional maturation. Furthermore, the CL of 10-day pregnant rabbit did not present significant capillary permeabilization and dilation or angiogenic processes, aspects that were previously found in stimulated periovulatory ovaries. Indeed, changes of the arterial supply and venous drainage of the CL of pregnancy were demonstrated. This suggests that the control of the blood flow through the CL of pregnancy may be transferred from the local capillary microcirculation to the regional artero/venous circulation. This may be probably related to the significant increase of the ovarian blood flow necessary for the maintenance of CL endocrine functions during pregnancy.

Electron-microscope immunocytochemistry was used to determine the subcellular distribution and presence of immunoreactive relaxin throughout pregnancy and early lactation in the corpus luteum of a marsupial, the tammar wallaby. Membrane-bound, electron-dense granules were a prominent feature of the luteal cell cytoplasm. The highest numbers of granules were observed between days 20 and 24 of the 26-day gestation, with a rapid clearance immediately after birth. Relaxin immunogold particles were present only in small, electron-dense granules (200-350 nm in diameter), with no particles observed in larger granules (>400 nm diameter), nuclei or mitochondria. Relaxin immunoreactivity was low throughout early and mid pregnancy but increased markedly between days 21 and 22 and remained high over the last 4 days of pregnancy. The number of granules containing relaxin immunogold particles and the density of immunostaining were both reduced on the day of expected births (day 26). Our data demonstrate that electron-dense granules in the luteal cell cytoplasm of a pregnant marsupial contain relaxin. The peptide is produced in greatest amounts at the end of pregnancy, consistent with a role in parturition.

The bovine pregnant corpus luteum was examined morphologically, to clarify the appearance and properties of the intramitochondrial bodies (IMB) in mitochondria of the large luteum cell (LLC). The incidence and diameter of the IMB (200-900 nm) showed a tendency to increase with the advance of pregnancy. Histochemically, the IMB reacted positively with protein, and immunohistochemically, they reacted IMB with 11 alpha-hydroxyprogesterone-1-11 succinyl-bovine serum albumin (P-BSA). The IMB seemed to combine with protein, and these granules did not undergo exocytosis. The cytoplasm of LLC contained another type of small electron-dense granules that were 150-200 nm in diameter, had an orifice for secretion, and reacted with relaxin. Small luteum cells (SLC) revealed crystalline-like inclusions. It is suggested that these inclusion cells participate in the synthesis of steroids.

In an attempt to characterize proteins secreted by the corpus luteum, explant cultures of luteal slices from cows taken on Days 3, 7, 11, 14, 17, and 19 of the estrous cycle, and Days 17, 88, 180, and > 240 of pregnancy were incubated with H-leucine for 24 h. Proteins in luteal-conditioned medium were separated by two-dimensional PAGE, transferred to polyvinylidene fluoride membrane, and subjected to N-terminal amino acid microsequencing. Microsequence analysis revealed that the bovine corpus luteum synthesized and released de novo synthesized apolipoproteins (Apo) E and A-I in culture during the estrous cycle and pregnancy. Release of Apo E was observed only on Day 3 of the estrous cycle. Release of Apo A-I was observed on Days 11, 14, 17, and 19 of the estrous cycle, and on all days of pregnancy examined. To demonstrate the presence of the appropriate mRNA and characterize the temporal relationship for these identified proteins, total RNA was isolated from corpora lutea on Days 2, 3, 7, 16, 17, and 20 of the estrous cycle, and on Days 17, 90, 170, 180, and 272 of pregnancy, and submitted to Northern and dot blot analysis. Apo E mRNA was expressed only on Days 2-3 of the estrous cycle and was not expressed on the other days of the cycle or during pregnancy. A single Apo E mRNA transcript about 1.0 kilobase (kb) in size was observed. Expression of Apo A-I mRNA was detected on all days of the estrous cycle and pregnancy examined. Apo A-I cDNA hybridized with a single mRNA transcript about 1.0 kb in size. Apo A-I mRNA levels did not differ among days of the estrous cycle, although higher levels of Apo A-I mRNA were observed during later stages of pregnancy. Serum concentrations of Apo A-I and progesterone were correlated across the estrous cycle but not during the prepartum period or after parturition. This study demonstrates for the first time that the corpus luteum synthesizes Apo E and Apo A-I and expresses their respective mRNAs. The pattern of expression of Apo E and Apo A-I mRNAs paralleled that of de novo synthesis of their respective proteins after incubation of luteal tissue with [H]leucine. The role of luteal apolipoproteins may involve an autocrine/paracrine function influencing luteal development, tissue remodeling, and steroidogenesis.

The microvasculature of bovine corpora lutea during pregnancy was studied by scanning electron microscopy using corrosion casts. Only one spiral arteriole branched directly from the ovarian artery entered the blood vascular bed of the pregnant corpora lutea. The bed consisted of numerous lobular sinusoidal capillary plexuses during pregnancy, and were characteristic to the microvasculature of a large corpus luteum. At the late stage of pregnancy, arteriovenous anastomoses were situated within the lobular sinusoidal capillary plexus composed of thin sinusoidal capillaries. These anastomoses may induce a decrease in blood flow to the lobular sinusoidal capillary plexus, suggesting that they may have an important role to induce the initial degeneration of the bovine pregnant corpora lutea.

In domestic ruminants the parenchyma of the corpus luteum consists of two subpopulations of steroidogenic cells commonly referred to as small and large luteal cells. These cells differ not only in size and structural characteristics, but also in functional properties. During the mid-luteal phase of the oestrous cycle approximately 60% of the corpus luteum is occupied by steroidogenic cells. Although the steroidogenic capacity of these cells declines as pregnancy advances, the cells persist as distinct populations throughout pregnancy and for several days following parturition. In general, structural changes typically observed at the end of the oestrous cycle also occur after parturition, but over a more extended period. These include deletion of endothelial cells and occlusion of capillary lumina with cellular debris and apoptotic bodies, an infiltration of eosinophils and macrophages, and fragmentation and lysis of parenchymal cells. However, not all parenchymal cells undergo lysis, nor are they rapidly phagocytosed by macrophages. Instead, many fuse to form what appear to be large syncytia that contain numerous lipid droplets, tightly packed mitochondria and multiple nuclei with condensed chromatin. Fusion of parenchymal cells to form syncytial profiles begins 2-3 days after parturition and the syncytia persist for at least 22 days post partum. By day 35 post partum transformation of the corpus luteum into a corpus albicans is essentially complete.

Large luteal cells of the bovine pregnant luteum can be characterized as steroid-producing cells. Mitochondria of these cells contained intramitochondrial bodies (IMB). The three-dimensional structure of the IMB and the degenerative process of luteal cells were as follows: 1) The IMBs were globular or oval, 0.1 to 1.5 microns in diameter. Their outer surfaces were coarse but their cut surfaces were uniform. Their outer surfaces changed from rough to smooth during pregnancy. 2) At the late stage of pregnancy, the large luteal cells lost their cytoplasmic processes, showing the first sign of degeneration at the cell level in such a way that their mitochondrial cristae transformed from tubular to laminar, and that they contained accumulated lipid droplets. 3) After parturition, the IMBs came out from degenerating large luteal cells, and were conveyed by lymphatic capillaries.

Isolated porcine luteal cells from d 10 and 15 of the estrous cycle (estrus = d 0) were incubated with or without combinations of FSH (0, 10, 10(2), 10(3) ng), LH (0, 10, 10(3) ng), oxytocin, or prostaglandin F2 alpha (PGF2 alpha) (each at 0, 10, 10(3), and 10(5) pg). Progesterone (P4) content was determined after overnight incubation (0 h) then at 2 and 24 h of incubation. The basal (0 h) P4 production of large cells (LC) from d 10 corpora lutea (CL) was 31-fold higher than that by small cells (SC) at 0 h. The LC and SC from d 10 but not those from d 15, were stimulated to a small extent by LH (P < .05). The FSH inhibited P4 production (P < .05) by SC at 24 h on d 10 and by LC after 2 or 24 h of incubation on d 15. There was no interaction between LH and FSH on P4 production. Oxytocin and PGF2 alpha decreased P4 production by d 15 LC at 2 h of incubation (P < .05) and by d 15 SC after 2 or 24 h of incubation (P < .05 and P < .01). The morphology of cells from CL of the cycle or early or mid pregnancy were examined using scanning and transmission electron microscopy (EM). Freshly isolated LC (using scanning EM) from d 10 contained many microvilli arranged in apparent networks on their membranes, but SC had smooth surfaces and contained only a few microvilli. Internally, LC had more small mitochondria than did SC and a different organization of smooth endoplasmic reticulum (SER). The SC from CL of pregnant (d 30 to 60) gilts contained more mitochondria than SC from CL of cyclic gilts. The results indicate that FSH, oxytocin, and PGF2 alpha can have a direct cellular luteolytic effect in the late luteal phase in pigs. The FSH influenced LC, whereas oxytocin and PGF2 alpha effected a more pronounced decrease in P4 from SC. The lower amount of P4 produced overall by SC may be associated with fewer microvilli, mitochondria, and SER in SC.

Uterine tissue was collected from intact pregnant rats on days 4, 10-15, 18, and 22 of pregnancy and day 2 postpartum and from rats on day 22 of pregnancy after ovariectomy (day 9) and progesterone-estrogen treatment. Placental, cervical, mammary, and pituitary tissues were collected from intact pregnant rats on day 22. Ovarian tissue was collected from intact pregnant rats on days 15, 20, and 22. These tissues were evaluated for relaxin immunostaining at the light microscope level using an antiserum to purified rat relaxin and the avidin biotin immunostaining technique. Since the corpus luteum is the major source of relaxin in the rat, this tissue was used as a positive control. Relaxin immunostaining in the corpus luteum was observed in a discrete region of the cytoplasm. This pattern of staining corresponded with the discrete clustering of relaxin-containing secretory granules found in the rat luteal cells. Relaxin was not observed in the day 22 placenta, cervix, mammary gland, pituitary gland, or day 4-13 pregnant uterus. Relaxin immunostaining was detected in endometrial epithelial cells on days 14-22 of pregnancy and day 2 postpartum. Relaxin immunostaining was more evident at implantation sites than between implantation sites. Antimesometrial epithelial cells at implantation sites contained relaxin immunostaining. The mesometrial surface, which consists of the placenta and decidualized endometrium, did not contain relaxin immunostaining. Relaxin immunostaining was also present in the endometrial epithelial cells of the day 22 pregnant rats that had been ovariectomized on day 9 and given subsequent replacement therapy with ovarian steroids. The presence of relaxin immunostaining in the ovariectomized rat indicates the endometrium is not sequestering relaxin secreted from the corpus luteum. These data provide evidence that the pregnant rat endometrium is a source of relaxin in addition to the corpus luteum. The appearance of relaxin in endometrial epithelial cells after implantation and its prominence at implantation sites may indicate that locally secreted conceptus factors stimulate endometrial relaxin synthesis.

Morphometric studies have confirmed that the corpus luteum (CL) of the pregnant rat contains luteal cells with numerous microvilli which directly face an extensive network of sinusoidal capillaries. From this it has been suggested that extensive development of transport structures is necessary to support progesterone synthesis and secretion. The present study was carried out to determine whether these transport structures could be related quantitatively to different rates of total progestin (progesterone plus 20 alpha-hydroxypregn-4-en-3-one) secretion reported to be 32, 10, and 23 micrograms/hr per ovary on day 16 and the morning (AM) and afternoon (PM) of day 22, respectively. Histological analysis was carried out on two CL, fixed by immersion, from each of five rats, at each stage of gestation. The important findings to emerge were that when the progestin secretion rate was greater, there was a significant increase in surface specializations on the luteal cell and a thickening of the capillary walls. There was also a greater volume of interstitial space between luteal cells and capillaries. However, due to the development of microvilli and unevenness in the capillary wall, the physiological diffusion distance (harmonic distance) between luteal cell cytoplasm and blood was not increased. Collectively, these results show that changes in the rate of progestin secretion are accompanied by significant, although disproportionate, changes in transport structures and suggest that the latter are important in supporting luteal function.

The present study was carried out to determine whether the major steroidogenic organelles of luteal cells quantitatively reflect variations in ovarian steroid secretion rates during pregnancy in the rat. Assessments were made on day 16 and in the morning (AM) and afternoon (PM) of day 22 (term is day 23). Ovarian steroidogenesis differs both quantitatively and qualitatively between these stages of pregnancy, so together they provide an ideal physiological model to study structural-functional relationships in the ovary. Corpora lutea of five rats were examined at each stage after progesterone and 20 alpha-hydroxypregn-4-en-3-one (20 alpha-OHP) secretion rates had been determined by a venous outflow technique. Total progestin secretion (progesterone plus 20 alpha-OHP) fell from 32.5 +/- 5.2 to 9.8 +/- 1.2 micrograms/hr per ovary (mean +/- SEM) between day 16 and day 22 AM but then increased to 22.6 +/- 1.4 micrograms/hr per ovary by day 22 PM. The total volume of luteal cell cytoplasm was slightly greater at day 22 AM and PM than at day 16. Similarly, the volumes of smooth endoplasmic reticulum (SER), lipid droplets, and membrane-bound granules all increased, but the volume of mitochondria decreased slightly. In contrast, the surface areas of SER and the inner and outer mitochondrial membranes did not change between day 16 and day 22 AM but then increased substantially by day 22 PM. Therefore, steroid secretion rates per unit area of steroidogenic membrane showed no consistent pattern over the stages examined.(ABSTRACT TRUNCATED AT 250 WORDS)

Aged stages (63) were available for establishment of a timetable of embryonic development of the stripe-faced dunnart. On Day 0 oocytes reaching maturity were found in the ovary. Within +/- 24 h of time 0 (time of minimum morning weight) polymorphonuclear leucocytes appeared and spermatozoa were last detected in the urine of 70% of females. Embryos were collected at intervals during pregnancy by hemihysterectomy and the embryos in the contralateral uterus either were examined at a later stage of pregnancy or allowed to develop to term. Cleavage to the unilaminar blastocyst stage with around 32 cells took 3 days with a cleavage arrest of 24 h at the 4-cell stage. Expansion of the unilaminar blastocyst occurred over the next 3 days. Primitive endoderm cells appeared on Day 6, fully bilaminar blastocysts by the end of Day 7 and trilaminar blastocysts on Day 8. Shell loss and implantation of 13-15-somite stage embryos occurred on Day 8 and organogenesis over the next 2-3 days. The gestation period was 9.5-12.0 days with most births occurring between 10.5 and 11.0 days. Major steps in embryonic development were correlated with stages in the development of the corpora lutea, which were maximal in size, and possibly in secretory activity, when the embryos were at the bilaminar blastocyst stage. Regression commenced when the embryos were at the primitive streak stage. At the time the corpora lutea were maximal the uterine epithelium reached its greatest height and the endometrium was thick and folded. Later in pregnancy villous-like projections of the epithelium formed, and the luminal epithelial cells became rounded. Two cell populations, a tier of 8 smaller cells above the yolk mass and a tier of 8 larger cells around the sides of the yolk mass appeared at the 16-cell stage. From the 16-cell stage to the blastocyst stage, with 150-200 cells, two cell populations distinguished by size, cell cycle time, cytoplasmic appearance and position relative to the yolk mass were present. The two populations were indistinguishable in blastocysts with greater than 200 and less than 2000 cells. They reappeared in blastocysts with greater than 2000 cells, as the darker cells of the embryoblast, and as the paler cells of the trophoblast. The darker cells lay in the yolky hemisphere and the paler cells in the non-yolky hemisphere.

Although a prominent role of conceptuses in the hormonal control of normal parturition in the pig is well founded, mechanisms are entrained that cause a timed relaxin release and an abrupt decrease in progesterone secretion in the complete absence of the uterus. This study focused on the steroidogenic and peptide hormone secretory capacities of aging porcine luteal cells in culture on specific days during a narrow window (days 110-116) when abrupt shifts occur in their function. Dispersed luteal cells (3 x 10(5) cells/well) from hysterectomized and pregnant gilts on days 110, 113, and 116 after estrus were cultured for 24 h with porcine (p) LH, pPRL, prostaglandin E1 (PGE1), PGE2, and (Bu)2cAMP. Progesterone and relaxin concentrations in cells preceding culture and in the spent medium were quantified by RIA. Progesterone in cells before culture was less (P less than 0.05) than that released into the medium, while relaxin levels were greater (P less than 0.05) than those in the medium. Relaxin stored in luteal cells of hysterectomized gilts was consistently greater (P less than 0.01) than that during the same days of late pregnancy; however, progesterone was greater (P less than 0.01) in hysterectomized compared with pregnant gilts only on day 116. Relaxin release was greater (P less than 0.01) in hysterectomized compared with pregnant gilts on days 113 and 116, whereas progesterone was greater (P less than 0.05) on days 110 and 116 in hysterectomized compared with pregnant animals. pLH, pPRL, and (Bu)2cAMP increased (P less than 0.05) relaxin secretion in hysterectomized gilts on days 110, 113, and 116, but only on day 110 in pregnant animals. These hormones consistently increased (P less than 0.05) progesterone release only on day 110 in both groups of gilts. PGE1 increased (P less than 0.01) relaxin release in hysterectomized, but not pregnant, gilts; it was not effective in altering progesterone in either group. PGE2 increased the release of both hormones on day 113 in both groups. The percentage of large cells (i.e. greater than 20 microns in diameter) was consistently greater (P less than 0.01) on days 110-116 in hysterectomized compared with pregnant gilts. Both luteal cell relaxin concentrations and the population of electron-dense granules decreased from days 110-116 in pregnant, but not in hysterectomized, gilts; luteal cells from postpartum animals were nearly depleted of granules, and empty vesicles were prominent. The secretory response of the cells to tropic hormone stimulation is preprogrammed from the previous estrus that occurred more than 110 days previously.(ABSTRACT TRUNCATED AT 400 WORDS)

The size distribution of marmoset luteal cells was determined on Days 6, 14 and 20 after ovulation in non-pregnant cycles and in early pregnancy. Image analysis was used to estimate the cell diameter of dispersed cells prepared from the marmoset corpus luteum (CL). Steroidogenic cells showed a size distribution consistent with one population of cells. There was a significant increase in mean cell diameter (P less than 0.05) from Day 6 to Day 14 in pregnant and non-pregnant animals with no further increase on Day 20. Micrographs of marmoset luteal tissue showed cells of greater than 10 micron containing the organelles typical of steroid-producing cells, and smaller non-steroidogenic cells surrounding the steroid-producing cells. On the basis of microscopy, there were no areas within the CL where cell composition was noticeably different. In contrast, micrographs of human luteal tissue showed two types of steroidogenic cell; most cells were similar to those in the marmoset CL but a smaller population of smaller cells could be distinguished around the periphery and along vascular septa. It is likely that these smaller and larger types of steroidogenic cells are of theca and granulosa cell origin respectively, the two cell populations differing in the degree of electron density and amount of rough endoplasmic reticulum. A distinguishing feature between marmoset and human luteal cells was the shape of the mitochondrian which were considerably rounder in marmoset luteal cells. The origin of steroidogenic cells in the marmoset CL is unclear, although in marmosets and man the luteal cell types display morphological characteristics distinct from the large and small luteal cells described for CL of the domestic ungulates.

The cellular distribution of prostacyclin (PGI2) synthase and specific PGI2 binding sites was investigated by light microscope immunocytochemistry and quantitative autoradiography. The immunostaining for the enzyme was found in small (15-18 microns) and large (18-45 microns) cells as well as in non-luteal cells, arteriole smooth muscle and endothelium. There was no consistent difference between small and large luteal cells and luteal vs. non-luteal cells in the immunostaining. Vascular smooth muscle and endothelial cells, on the other hand, contained less immunostaining than the other cells. Contrary to wide-spread distribution of PGI2 synthase, specific PGI2 binding sites were only found in the luteal cells. On a per cell basis, large luteal cells contained more sites than small luteal cells and vice versa when expressed per unit cell area. The [3H]PGI2 binding to the luteal cells was time and temperature dependent and was inhibited by excess unlabeled PGI2 but not by its metabolite, 6-keto-PGF1 alpha or other eicosanoids which bind to their own receptors. In conclusion, while PGI2 synthase is widely distributed among different cell types in bovine corpora lutea, specific binding sites which may mediate luteotropic actions of PGI2 are only present in small and large luteal cells.

Steroidogenic cells in the corpus luteum of the ferret (Mustela putorius) during early (Days 6 and 13) to midpregnancy (Day 24) were characterized using electron microscopy, immunocytochemical localization of neurophysin, and smears of dispersed cells obtained by dissociating luteal cells with collagenase. The latter were stained for 3 beta-hydroxysteroid dehydrogenase (3 beta-HSD) activity, and the diameters of the cells were determined with an ocular micrometer. Very small cells (less than 12 microns) stained negative for 3 beta-HSD, occurred in clumps of 5-50 cells, and were presumed to be primarily endothelial cells. 3 beta-HSD-positive cells covered a wide spectrum of sizes ranging from 14 to 56 microns and did not exist as two discrete populations. The ratio of small (less than 25 microns) to large (greater than 25 microns) cells was 1.86:1.0 on Day 6, with the 17- to 20-microns cell size class predominating. On the day of implantation (Day 13), about 75% of the cells ranged from 26 to 50 microns, with the 29-microns size predominating. By Day 24, the ratio of small-to-large cells had declined to 0.15. Nearly 90% of the cells were in the 26- to 56-microns range, the predominant size being 35 microns. All size classes of luteal cells stained negative for neurophysin on all 3 days of pregnancy studied. Luteal cells obtained on Days 6, 13, and 24 of pregnancy failed to reveal any evidence of mitosis after in vivo or in vitro colchicine treatment. We interpret these results as indicating that the 3 beta-HSD-positive luteal cells of ferrets progressively increase in size as small luteal cells complete their differentiation from granulosa cells and ultimately form larger luteal cells with somewhat different ultrastructural characteristics.

Gravid female Japanese long-fingered bats were kept in captivity without hibernation at elevated temperatures (23-25 degrees C) in winter. The embryonic growth rate was accelerated and consequently parturition was advanced by a period equivalent to that of exclusion from hibernation as compared with that in the wild population. The corpus luteum became active, as indicated by an increase in the 'light' lutein cells in an experimental bat pregnant with a 15-mm embryo, but was less active (more 'dark' cells) in 2 hibernating control bats with an implanting blastocyst.

Changes in luteal weight from about Day 20 to near term, and in quantitative histology as assessed by ultrastructural morphometry and light microscopic counts of mitosis and cell death on Days 30, 60, 100 and 142, were studied in 168 pregnant ewes. Luteal weight (mean +/- s.d.) remained constant at 0.56 +/- 0.11 g until Day 120, and fell thereafter to reach 0.31 +/- 0.11 g after Day 140 (P less than 0.01). Up to Day 100, quantitative aspects of the composition of the luteal tissue showed no significant change, and values for volume density, cytoplasmic:nuclear ratio, cell number/mm3 and cell volume were comparable to values previously obtained for corpora lutea (CL) of the cycle. By Day 142 structural evidence of luteal regression was present, but regressive changes were much more marked in some CL than others. Mitosis was seen in a few cells (0.02-0.04%) on all of the days studied, but never in large luteal cells. Cell death was rarely seen up to Day 100, but had increased in incidence by Day 142 (P less than 0.01). Luteal progesterone content, 55.2 +/- 15.9 nmol/g on Day 30, was not significantly changed on Days 60, 100 or 142. It is concluded that (1) structural regression of the CL of pregnancy does not begin until much later than the time (about Day 50) when pregnancy ceases to depend on the CL; (2) structural luteal regression begins before parturition, but its time of onset and/or rate of progression vary widely between animals; and (3) large and small luteal cells remain as distinctive populations throughout pregnancy, and their numbers at all stages can be accounted for by survival of the cells which differentiate during the genesis of the CL.

Corpora lutea from sows at 30, 60 and 90 days of gestation were dissociated enzymically, and the components of the resulting cell suspension were separated by sedimentation at unit gravity. Two luteal cell populations of 30-50 micronm diameter and 15-20 micronm diameter were obtained and superfused for up to 18 h with Dulbecco's modified Eagle medium, the cells being supported in a column in a matrix of Biogel. Fractions were collected every 30 min and assayed for progesterone and oestradiol-17beta. At 30 and 60 days of gestation the large luteal cells produced progesterone at an initial rate of approximately 100 ng/h/10(5) cells, which decreased to half this rate at 90 days. The smaller cells also released progesterone into the medium at approximately 15-20 ng/h/10(5) cells at all stages of gestation. At 30 days of gestation, neither cell type released significant amounts of oestradiol-17beta, but from 60 days onwards ,significant and increasing quantities were measured in the superfusates from the larger cells. Both cell types were perfused with porcine LH at the three stages of gestation, and both showed an immediate response in terms of progesterone release which decreased in magnitude with increasing age of gestation. The response of the smaller cells was greater than that of the larger cells.

Electron microscopy evaluation indicated that the large luteal cells from control (saline-injected) cows were morphologically similar: 42-72% of the large luteal cells, in which the nucleus was present in the tissue section, contained secretory granules (100-300 nm diameter). From cell to cell, clustering of granules was observed in various regions of the cell cytoplasm, ranging from locations next to the nucleus to those next to the cell membrane where exocytosis was noted. The large mitochondria containing dense inclusions (500-1800 nm diameter) were uniformly distributed throughout the cytoplasm of 98% of the large luteal cells. The morphology of large luteal cells at 2.5 and 5 min after PGF-2 alpha treatment (25 mg) was similar to that of saline controls. However, at 15 and 30 min after PGF-2 alpha the secretory granules were observed primarily at the cell periphery and at 15 min only 22% of the large luteal cells contained secretory granules. Exocytosis involving fusion of the granule and cell membrane was more evident at this time. At 60 min after PGF-2 alpha few large luteal cells (0.5%) were observed with small granules. We suggest that the secretory granules represent those organelles responsible for packaging secretory proteins for transport out of the cell, and PGF-2 alpha is an effector that will initiate cell depletion of these granules and their contents.

Relaxin was localized in luteal cells of ovaries from nonpregnant, pseudopregnant, and pregnant pigs using porcine relaxin antiserum and peroxidase-antiperoxidase light microscopy immunohistochemistry. The number of immunoreactive cells seemed to increase from Days 17 to 106 of gestation. Luteal cells from pseudopregnant (Day 110) and nonpregnant (Day 14 of the estrous cycle) pigs were also positive for relaxin. However, less than 3% of the luteal cells in the nonpregnant animals were immunoreactive. Electron microscopy immunocytochemistry using porcine relaxin antiserum and goat antirabbit immunoglobulin G-colloidal gold demonstrated that relaxin was packaged in the small membrane-bound granules in luteal cells of pregnant as well as pseudopregnant and nonpregnant pigs. The intensity of labeling (number of gold particles) of the granules increased with pregnancy. There was a 10-fold increase in labeling of granules with the 10-nm versus 25-nm diameter gold. The goat antirabbit labeled with the smaller 10-nm gold particles was necessary to demonstrate the apparent low levels of relaxin in the luteal cells of the nonpregnant pigs. These data further indicate that pregnancy is not required for relaxin synthesis. However, physiologic significance of relaxin in corpora lutea of nonpregnant pigs has not been determined.

Corpora lutea from gerbils on days 1, 4, 8, 12, 16, 20, and 24 of pregnancy were studied electron microscopically. Similarly, luteal tissue from animals on the day of parturition and one day postpartum was studied (gestation: 24 days +/- 8-24h). Agranular endoplasmic reticulum increases in quantity through day 16 and thereafter is somewhat reduced. Granular endoplasmic reticulum and a population of small granules (type I) become abundant during late pregnancy and their possible role in the production and storage of relaxin is discussed. Luteal tissue undergoes a relatively rapid regression which begins on the day of parturition. Conspicuous in the regressing luteal tissue are large (type II) granules (possibly lysosomes), lipid droplets, leucocytic elements and macrophages. Functional correlates of these morphological findings are discussed.

The ultrastructure of lutein cells during pregnancy and the post partum period was examined by transmission electron microscopy in the house musk shrew, Suncus murinus. Lutein cells on day 13 of pregnancy contained an extensive system of anastomosing tubules or cisternae of smooth ER and many enlarged mitochondria with numerous tubulovesicular cristae. From day 13 on, the number of small granules, possibly lysosomes, increased gradually. Between day 20 and 25, the loss of smooth ER began and elongated, or flattened mitochondria increased. Regressing lutein cells observed after parturition were characterized by abundant large dense bodies, bizarre mitochondria and a decrease in the amount of smooth ER. Unusual forms of mitochondria were always observed after day 5 of pregnancy. Two types could be distinguished; one, found frequently in the second third of pregnancy, was ring-, disc-, cup- or dumb-bell-shaped with tubulovesicular cristae, and the other, found exclusively in the last third of pregnancy and after parturition, was elongated, flattened and sometimes twisted. The paucity of lipid droplets was a characteristic feature of the lutein cells of this species. The significance of these ultrastructural changes of cellular organelles is discussed in relation to the ovarian and plasma levels of progesterone.

Secretory granules, which are released by exocytosis and are speculated to contain progesterone, have been described in luteal cells of sheep and other large domestic animals. These granules are small and densely staining. Gemmell and Stacy ('79) suggested that luteal cells of guinea pigs also contain secretory granules, although they could not document exocytosis of granule content at the fine structural level. For the present study, quantitative methods were used to reexamine the possibility that luteal cells of guinea pigs possess secretory granules. Ovaries of guinea pigs were fixed in situ by vascular perfusion at the time of maximum progesterone secretion, when such granules would be most abundant, as well as other stages. Two types of granules that might be confused with secretory granules are microperoxisomes and lysosomes. Therefore, slices of perfusion-fixed corpora lutea were incubated for the fine structural localization of a peroxisomal enzyme, catalase, or for the lysosomal enzymes, acid phosphatase (ACPase) and arylsulfatase. Other tissue was prepared for conventional electron microscopy. Granule types were classified on the basis of size, morphology, and enzyme content. Quantitation of granule types was carried out on both cytochemically reacted and conventionally prepared luteal tissue. More than 5500 microperoxisomes, 2800 lysosomes, and 1100 multivesicular bodies (MVBs) were tabulated. The results indicate that luteal cells of guinea pigs have three main types of granules: 1) Microperoxisomes, about 0.2 micrometer in diameter and containing catalase; 2) lysosomes, about 0.5 micrometer in diameter and positive for ACPase and arylsulfatase; and 3) MVBs, about 0.4 micrometer in diameter and containing small vesicles. At the time of peak steroid secretion during pregnancy and the estrous cycle, the granule population in luteal cells of guinea pigs consists of 73-80% microperoxisomes, 13-17% lysosomes, and 7-9% MVBs. These proportions are similar in tissue reacted for cytochemistry and tissue prepared by conventional means. Greater than 99% of the small 0.2-0.3 micrometer diameter granules in guinea pig luteal cells are catalase reactive. This finding eliminates from further consideration most of the prime candidates for secretory granules in these cells. Finally, neither a sequential appearance of granules nor exocytosis of secretory product was detected. Our data thus argue against the suggestion that luteal cells of guinea pig have secretory granules of the type observed in corpora lutea of large domestic animals.

Relaxin was localized in cells of corpora lutea of pregnant rats at the ultrastructural level using a highly specific antirat relaxin serum and the unlabeled peroxidase-antiperoxidase technique. Electron-dense, membrane-bound granules (maximum diameter, 270 nm), which are present in luteal cells during the last third of gestation, were the only inclusions that were immunochemically stained. The number of granules observed in the luteal cell cytoplasm varied from cell to cell within a particular section. Furthermore, in the granule-rich luteal cells, the granules appeared in clusters. This study establishes that these electron-dense granules represent the subcellular sites of relaxin localization within luteal cells of pregnant rats.

By use of lanthanum tracer and freeze-fracture procedures it was found that granulosa-lutein cells of the pregnant mouse and rat ovaries are connected by gap junctions and septate-like zones of contact. Lutein cell gap junctions enlarge and become partially internalized by the end of the first week of gestation. Expansion of the gap junction domain appears to be due initially to intercalation of particles along borders of small gap junctions devoid of smaller non-junctional particles. The number of gap junction lined processes appearing at the cell border increases concomitantly with hypertrophy of the lutein cell during the second week of pregnancy. Strands of particulate or grooved membrane emanate from the margin of larger gap junctions undergoing interiorization. Most large gap junctions are intimately associated with elements of the smooth endoplasmic reticulum. Spherical gap junctional profiles assume a deeper location in the lutein cell and may form concentric arrays by term while true surface gap junctions appear to fragment in the post-partum corpus luteum. The modifications observed are interpreted with respect to biogenesis of the gap junction and the hormonal control of lutein cell function.

The ultrastructure of large and small cells from corpora lutea of pregnant cows (days 45-280) were evaluated by electron microscopy. The distinguishing features of small cells (10-15 micron in diameter) included stacks of rough endoplasmic reticulum, whorls of smooth endoplasmic reticulum, elongated mitochondria containing crystalline-like inclusions, and cytoplasmic lipid droplets. The large cells (20-50 micron in diameter) contained numerous mitochondria packed tightly together (no elongated structures), an abundance of smooth endoplasmic reticulum (no whorls), and a large number of membrane-bound secretory granules (150-300 nm in diameter). These granules appeared to be packaged in the Golgi, accumulated at a paranuclear region, and migrated as a group to the cell membrane where they were exocytosed. These granules were first observed on day 45 and increased in number to reach a peak around day 200. Lipid droplets became a common cytoplasmic inclusion in the large cells during the third trimester of pregnancy. In addition, during this stage, an electron-dense substance began to accumulate in the mitochondria to such an extent as to occlude the cristae. These mitochondria looked like large (500-1800 nm) membrane-bound granules; however, they did not undergo exocytosis. Their appearance in large cells during the last 3 months of pregnancy may reflect a change in steroid metabolism. Thus, there are two morphologically distinct cell types throughout pregnancy in the cow. The large cell containing the secretory granules underwent what appeared to be a progressive state of apparent deterioration with advancing pregnancy. The morphology of the small cell did not undergo such a dynamic change. No morphological evidence was observed that would support a transition state between the two cell types.

Developmental delay is correlated with torpor in the temperate zone bent-winged bat, Miniopterus schreibersii (latitude 37 degrees S) as a period of pre-implantation delay (delayed implantation) followed by a short post-implantation delay (delayed development). During this time, the number of steroidogenic organelles in luteal cytoplasm is greatly reduced compared with normal embryogenesis, and granular endoplasmic reticulum is prominent. Nidation, which occurs while the animals are hibernating, is not accompanied by marked changes in luteal ultrastructure, although the number of lipid droplets decreases somewhat. Progesterone rises slightly but not significantly; however, a pre-nidation decrease in high 17 beta-estradiol levels may play a role in implantation. Following implantation, the conceptus remains delayed at the blastocyst stage for several weeks. During this time the bats remain torpid and the only change in luteal cell ultrastructure is an increase in smooth endoplasmic reticulum as differentiation begins toward the trilaminar stage. At the end of developmental delay hypertrophy of the luteal cell begins and mitochondria and lipid droplets increase, markedly. By this time arousal from hibernation has occurred, placentation takes place and normal development is underway. At placentation, smooth endoplasmic reticulum reaches its maximum in luteal cytoplasm; estrogen and progesterone levels peak about 6 weeks later. For the remaining 2 months of gestation, signs of luteolysis appear. These observations suggest that the corpus luteum of developmental delay, though sub-optimally functional, is prolonged in its luteinization by the arrival of winter when the bats enter torpor. The capacity for maximal steroidogenesis is acquired at the end of winter, some weeks after implantation, when arousal occurs and normal development ensues.

The fine structure of luteal tissue from sheep and goats at the mid-stage of the estrous cycle and during pregnancy was examined. At the mid-stage of the estrous cycle, when the corpus luteum of both species is secreting progesterone, the ultrastructure of the luteal cells is similar in that granule secretion is prevalent. In late pregnancy, the corpus luteum of the sheep produces very little progesterone and there is an absence of granule secretion. In the pregnant goat, the production of progesterone from the corpus luteum is high and there is plentiful evidence of granule formation and secretion.

The structural changes in the granulosa lutein cells in the pregnant cows between 60 and 245 days, were observed. The polyhedral or pump cells has an acidophilic cytoplasm around the spherical nucleus. The glycogen granules increased in number up to 230 days of pregnancy. The lipid globules increased in size and amount during advanced pregnancy. The mitochondria showed a wide range of variations in shape and size. The matrix of many mitochondria contained dense inclusions which replaced the entire matrix in late pregnancy. The endoplasmic reticulum was chiefly of smooth type but a few stacks of the rough variety could be seen in the early days of pregnancy. Sometimes, whorls of smooth-surfaced endoplasmic reticulum enclosing mitochondria were seen. The Golgi complex was fully developed in the granulosa cells of all cases studied. The dense granules and lipid globules increased in size and number in the older corpora lutea. The cell surface showed numerous infolding of the plasma membrane.

Little information is available on the ultrastructure of macrophages in the corpus luteum or their importance in the regression of luteal tissue. In the present study, the fine structure of activated luteal macrophages during pregnancy and the postpartum period was examined by electron microscopy of guinea pig ovaries fixed by vascular perfusion. In these corpora lutea, macrophages can readily be distinguished from luteal cells. Activated macrophages typically display three prominent inclusions in their cytoplasm: (1) heterophagic vacuoles, (2) distinctive large dense inclusions, and (3) large and small electron-lucent vacuoles. In addition, they contain numerous smaller lysosome-like dense bodies. Activated macrophages in corpora lutea also characteristically show many surface protrusions, such as processes, folds or pseudopodia, which often occur in close contact with nearby luteal cells. Generally, nuclei of macrophages are irregular in shape and display a dense border of heterochromatin, thus differing from those of luteal cells. Macrophages seem to be most abundant in regressing corpora lutea, where they commonly display heterophagic vacuoles containing recognizable luteal cell fragments, evidence that these phagocytes ingest senescent luteal cells. The digestion of luteal cell components in heterophagic vacuoles presumably gives rise to the distinctive large dense inclusions typically seen in macrophages. The findings of this study indicate that macrophages play a central role in luteolysis by phagocytizing luteal cells or their remnants. They therefore appear to bring about the reduction in volume of the corpus luteum that occurs as this tissue regresses. These results taken together with those previously published (Paavola, '78) further indicate that breakdown of the corpus luteum during postpartum luteolysis in guinea pigs involves both autophagy and heterophagy.

Pairs of heifers were inoculated IV with infectious bovine rhinotracheitis virus on postbreeding days (PBD) 7, 14, 21, or 28, and were euthanatized 13 to 15 days after inoculation. Reproductive tracts were examined for cytopathologic changes (light microscopy), virus (cell culture), and viral antigen (immunohistochemical evaluation). Heifers inoculated on PBD 7 or 14 had mild oophoritis characterized by foci of necrosis and mononuclear cell accumulations in the corpus luteum. Most of these heifers also had a few necrotic follicles in at least one ovary. Heifers inoculated on PBD 21 or 28 did not have corpus luteum lesions, but necrotic follicles were numerous in both ovaries. Viral antigen was observed in all ovarian lesions, and infectious virus was isolated from a few of the affected tissues. The uteri of all heifers inoculated on PBD 21 or 28 and 1 heifer inoculated on PBD 7 contained normal-appearing concepti. The uterus of the other PBD 7 heifer contained a degenerating conceptus that was infected with infectious bovine rhinotracheitis virus, as determined by viral isolation, immunohistochemical evaluation, and electron microscopy. Heifers inoculated on PBD 14 were not pregnant at necropsy, but histologic evidence was found that the postbreeding estrous cycle had been longer than normal, indicating that early embryonic death had occurred.
