Workshops are an important part of the IFPA annual meeting as they allow for discussion of specialized topics. At IFPA meeting 2018 there were nine themed workshops, four of which are summarized in this report. These workshops discussed new knowledge and technological innovations in the following areas of research: 1) viviparity in ocean-living species; 2) placental imaging; 3) epigenetics; and 4) extracellular vesicles in pregnancy., (Copyright © 2019 Elsevier Ltd. Background: Menstruation occurs only in higher primates, some bats, the spiny mouse and the elephant shrew. Our knowledge of the latter species is due to work by C. J. van der Horst., Findings: Changes in the uterine stroma are initially similar in fertile and infertile cycles and are confined to a small area. In pregnant animals, the presence of the conceptus causes further development to an implantation chamber. In infertile cycles an outgrowth of highly glandular stroma (a polyp) appears. With decline of the corpora lutea it is shed in a process equivalent to menstruation. Van der Horst described the further development of the placenta and a decidua pseudocapsularis in pregnant animals. In addition he built a unique collection that has thrown light on embryonic development and placentation in other South African mammals., Conclusions: The changes in endometrial stromal cells during the menstrual cycle appear similar between primates and the elephant shrew and deserve to be studied at the molecular level., (Copyright © 2018 Elsevier Ltd. Introduction: In this study we evaluated the full extent of placental bed changes (centre to periphery) in a pregnant chimpanzee uterus, kept at the Museum for Central Africa in Tervuren, Belgium. According to placental size the specimen was equivalent to an 8 weeks pregnant human uterus., Methods: Histological sections from central to peripheral tissue blocks of the placental bed were stained to reveal the presence of trophoblast, endothelium, vascular smooth muscle and elastic laminae. As an indicator for early arterial remodelling, we evaluated endothelial nuclear rounding and subendothelial vascular changes within the maternal vasculature in decidua and adjacent inner myometrium., Results: While interstitially invading trophoblasts were present, endovascular trophoblast invasion seemed about to start into one spiral artery outlet at the centre of the placental bed, confirming our previous impression of a later onset of endovascular trophoblast invasion as compared to the human. An early sign of spiral artery remodelling was rounding of the endothelial nuclei. This phenomenon was not related to the local presence of interstitial trophoblast., Discussion: Endothelial nuclear rounding turned out to be a feature of the placental bed as a whole, being significantly less prominent in the adjacent non-placental bed part of the uterus, indicating an effect of the presence of the placenta. The different time-course of early spiral artery remodelling in the chimpanzee as compared to the human may have had a significant impact upon our evolution., (Copyright © 2017 Elsevier Ltd. Background: Emil Selenka made important contributions to embryology in marsupials, rodents and primates that deserve wider recognition. Here we review his work on early development of the mouse and placentation in the great apes., Findings: Selenka was intrigued by germ layer theory, which led him to study inversion of the germ layers in the mouse and other rodents. He found it was growth of the ectoplacental cone that caused a downward shift in the position of the underlying ectoderm and endoderm, leading to an inside-outside inversion of these layers. In primates he made the important discovery that the embryos of gibbons and orangutans develop under a decidua capsularis. Thus all great apes, including humans, exhibit interstitial implantation; this is in contrast to other primates where implantation is superficial., Conclusions: Selenka's work was thorough and brilliantly illustrated. It was an important influence on his contemporaries and was well known to scientists of the following generation. Embryologists continue to advance our knowledge of fetal membranes and placentation in the mouse, but Selenka's work on gibbons is unique and our knowledge of orangutan placentation is restricted to his specimens., (Copyright © 2015 Elsevier Ltd. Background: Sigmodontinae, known as "New World rats and mice," is a large subfamily of Cricetidae for which we herein provide the first comprehensive investigation of the placenta., Methods: Placentas of various gestational ages ranging from early pregnancy to near term were obtained for five genera, i.e. Necromys, Euryoryzomys, Cerradomys, Hylaeamys, and Oligoryzomys. They were investigated by means of histology, immunohistochemistry, a proliferation marker, DBA-lectin staining and transmission electron microscopy., Results: The chorioallantoic placenta was organized in a labyrinthine zone, spongy zone and decidua and an inverted yolk sac persisted until term. The chorioallantoic placenta was hemotrichorial. The interhemal barrier comprised fetal capillary endothelium and three layers of trophoblast, an outermost, cellular layer and two syncytial ones, with interspersed trophoblast giant cells (TGC). In addition, accumulations of TGC occurred below Reichert's membrane. The junctional zone contained syncytial trophoblast, proliferative cellular trophoblast, glycogen cells and TGC that were situated near to the maternal blood channels. In three of the genera, TGC were also accumulated in distinct areas at the placental periphery. PAS-positive glycogen cells derived from the junctional zone invaded the decidua. Abundant maternal uNK cells with positive response to PAS, vimentin and DBA-lectin were found in the decidua. The visceral yolk sac was completely inverted and villous., Conclusion: The general aspect of the fetal membranes in Sigmodontinae resembled that found in other cricetid rodents. Compared to murid rodents there were larger numbers of giant cells and in some genera these were seen to congregate at the periphery of the placental disk. Glycogen cells were found to invade the decidua but we did not identify trophoblast in the walls of the deeper decidual arteries. In contrast these vessels were surrounded by large numbers of uNK cells. This survey of wild-trapped specimens from five genera is a useful starting point for the study of placentation in an important subfamily of South American rodents. We note, however, that some of these rodents can be captive bred and recommend that future studies focus on the study of time dated pregnancies.

Background: The guinea pig is an attractive model for human pregnancy and placentation, mainly because of its haemomonochorial placental type, but is rather small in size. Therefore, to better understand the impact of body mass, we studied placental development in the capybara which has a body mass around 50 kg and a gestation period of around 150 days. We paid attention to the development of the lobulated arrangement of the placenta, the growth of the labyrinth in the course of gestation, the differentiation of the subplacenta, and the pattern of invasion by extraplacental trophoblast., Methods: Material was collected from six animals at pregnancy stages ranging from the late limb bud stage to mid gestation. Methods included latex casts, standard histology, immunohistochemistry for cytokeratin, vimentin, alpha-smooth muscle actin, and proliferating cell nuclear antigen as well as transmission electron microscopy., Results: At the limb bud stage, the placenta was a pad of trophoblast covered by a layer of mesoderm from which fetal vessels were beginning to penetrate at folds in the surface. By 70 days, the placenta comprised areas of labyrinth (lobes) separated by interlobular areas. Placental growth resulted predominantly from proliferation of cellular trophoblast situated in nests at the fetal side of the placenta and along internally directed projections on fetal mesenchyme. Additional proliferation was demonstrated for cellular trophoblast within the labyrinth.Already at the limb bud stage, there was a prominent subplacenta comprising cellular and syncytial trophoblast with mesenchyme and associated blood vessels. At 90 days, differentiation was complete and similar to that seen in other hystricognath rodents. Overlap of fetal vessels and maternal blood lacunae was confirmed by latex injection of the vessels. At all stages extraplacental trophoblast was associated with the maternal arterial supply and consisted of cellular trophoblast and syncytial streamers derived from the subplacenta., Conclusion: All important characteristics of placental development and organization in the capybara resembled those found in smaller hystricognath rodents including the guinea pig. These features apparently do not dependent on body size. Clearly, placentation in hystricognaths adheres to an extraordinarily stable pattern suggesting they can be used interchangeably as models of human placenta.

The reproductive tract was examined in four non-pregnant and two gravid specimens of Myzopoda. The ovaries had little interstitial tissue. The uterus was bicornuate and the lenticular placental disk was situated mesometrially in one horn. The interhaemal barrier of the placental labyrinth was of the endotheliomonochorial type. There was a single layer of trophoblast cells. The cells of the maternal endothelium were large and basophilic, contained abundant rough endoplasmic reticulum, and rested on an irregular basement membrane. Blunt projections of endometrium extended into the placental disk and clusters of large cells occurred between the endometrial stroma and labyrinth. At the margins of the disk folds of trophoblast occurred and at the cranial end they formed an haemophagous region. The folds lateral to the disk included some peculiar tubular-appearing structures. There was a persistent yolk sac containing large endodermal cells around a largely collapsed lumen. Several features of placentation, such as the interhaemal barrier and the haemophagous region, are consistent with an association of Myzopodidae with Emballonuridae. No support was found for alternative hypotheses that include Myzopodidae in the noctilionoid or vespertilionoid lineages.

Evidence from several sources supports a close phylogenetic relationship between elephants and sirenians. To explore whether this was reflected in similar placentation, we examined eight delivered placentae from the Amazonian manatee using light microscopy and immunohistochemistry. In addition, the fetal placental circulation was described by scanning electron microscopy of vessel casts. The manatee placenta was zonary and endotheliochorial, like that of the elephant. The interhaemal barrier comprised maternal endothelium, cytotrophoblasts and fetal endothelium. We found columnar trophoblast beneath the chorionic plate and lining lacunae in this region, but there was no trace in the term placenta of haemophagous activity. The gross anatomy of the cord and fetal membranes was consistent with previous descriptions and included a four-chambered allantoic sac, as also found in the elephant and other afrotherians. Connective tissue septae descended from the chorionic plate and carried blood vessels to the labyrinth, where they gave rise to a dense capillary network. This appeared to drain into shorter vessels near the chorionic plate. The maternal vasculature could not be examined in the same detail, but maternal capillaries ran rather straight and roughly parallel to the fetal ones. Overall, there is a close resemblance in placentation between the manatee and the elephant.

The four-toed elephant shrew is the only extant member of the genus Petrodromus. Some features of its placentation are found in other elephant shrews and may be important in consolidating the elephant shrews in the order Macroscelidea. These include implantation in an embryo chamber at the caudal end of the uterine horn; a large yolk sac; a large, lobulated allantoic sac; a distinct layer of proliferative trophoblastic cells between the labyrinthine and spongy zones of the placenta; an accessory epitheliochorial placenta; and the formation of a mesoplacenta by the decidua basalis. Of particular note are the large granulated cells that occupy the walls of decidual, myometrial and mesometrial arteries. They appear to be uterine natural killer cells. The persistence of a relatively large yolk sac and a large lobulated allantoic sac is a feature which Petrodromus and other elephant shrews share with other members of the superordinal clade Afrotheria.

Placentae of three hystricimorph rodents--capybara, agouti and paca--were examined by conventional histology, immunohistochemistry for cytokeratin and vimentin, and TUNEL staining. The placentae were divided into lobules of labyrinthine syncytium separated by interlobular and marginal trophoblast. The subplacenta comprised cytotrophoblasts, supported on lamellae of allantoic mesoderm, and syncytiotrophoblast. The central excavation was still apparent in the definitive placenta of capybara. In agouti and paca, the decidua of the junctional zone formed a mesoplacenta comprising a capsule and a pedicle. Towards term the pedicle formed a tenuous attachment between placenta and uterine wall comprising a few maternal vessels surrounded by degraded tissue. In paca placenta, it was shown by TUNEL staining that breakdown of this tissue occurred by apoptosis. The visceral yolk sac was highly villous and, in agouti, the yolk sac villi were extremely long. Lateral to its attachment to the placenta, the fetal surface was covered with non-vascular yolk sac endoderm. A layer of spongiotrophoblast cells was interposed between the endoderm and the marginal trophoblast., (Copyright 2002 Elsevier Science Ltd. All rights reserved.)

Using antisera directed towards the C-terminal region of gastrin releasing peptide (GRP), significant quantities of GRP-like immunoreactivity (GRPLI) were detected in ovine amniotic fluid and in the fetal and maternal circulations. The highest GRPLI levels were found in amniotic fluid (2135 +/- 829 fmol/ml, n = 12; mean +/- SEM), followed by those in ovine fetal (604 +/- 267 fmol/ml, n = 13) and maternal plasma (229 +/- 89 fmol/ml, n = 13). On gel filtration chromatography, the predominant GRPLI form in each fluid eluted in an identical position consistent with the entity being of apparently larger molecular size than porcine GRP1-27. Certain fetal plasma samples contained a second GRPLI peak eluting at the void volume. Hence, during ovine pregnancy a GRPLI entity circulates in fetal and maternal plasma; the entity is of apparently larger molecular size than GRP1-27 but contains a structure immunologically indistinguishable from the bioactive c-terminal region of GRP1-27. Given the recognized bioactivities of GRP, this entity may be an important hormone during ovine fetal life.

Neuroleptanalgesia was induced in late pregnancy in guinea pigs with diazepam and fentanyl-fluanisone. Hypotension occurred, but the arterial acid-base status after 45-60 min was near normal. When repeated injections of fentanyl-fluanisone were given to maintain neuroleptanalgesia, the blood pressure fell further and severe metabolic acidosis developed. Placental blood flow determined with radioactive microspheres was much lower during maintained neuroleptanalgesia than in guinea pigs anaesthetized with diazepam and pentobarbitone.
