The Potato Introduction Station in Sturgeon Bay Wisconsin has maintained herbarium specimens as part of its operations. These specimens mostly were grown from seeds in the field plots or greenhouses, but also include original specimens made during germplasm collecting expeditions. The herbarium of the Station (herbarium acronym PTIS) has been transferred from the potato genebank in Sturgeon Bay Wisconsin to the Wisconsin State Herbarium (acronym WIS) in Birge Hall, University of Wisconsin-Madison where it is now fully integrated and curated and funded by WIS; PTIS is discontinued and is no longer maintained as a separate herbarium. Solanum section Petota has been the subject of intensive taxonomic work since the description of the cultivated potato in 1753. In total, there are 494 epithets for wild taxa and 626 epithets for cultivated taxa. Different taxonomists applied various taxonomic philosophies and species concepts to the section. Hypotheses of the number of species and their interrelationships have differed greatly among authors. A taxonomic treatment of section Petota by Jack Hawkes in 1990 recognized 228 wild species and seven cultivated species, divided into 21 taxonomic series. In 2014 Spooner and collaborators more than halved this number to 107 wild species and four cultivated species, partitioned into four clades; not using series. The purpose of this paper is to provide a retrospective of the methods and philosophies that have resulted in this drastic decrease in the number of species and their infrasectional classification. With ca. 200 species, the informally named Potato clade represents one of the larger subgroups of the estimated 1500 species of Solarium. Because its members include the potato (S. tuberosum), tomato (S. lycopersicum), and pepino (S. muricatum), it is the most economically important clade in the genus. These crop species and their close relatives have been the focus of intensive research, but relationships among major lineages of the Potato clade remain poorly understood. In this study, we use sequences from the nuclear ITS and waxy (GBSSI), and plastid trnT-trnF and trnS-trnG to estimate a phylogeny and further explore relationships within the Potato clade. With increased sampling over past studies, the Potato clade emerges as a strongly supported clade and comprises 12-13 subclades which, for the most part, correspond to traditionally recognized sections. Solanum sect. Regmandra is sister to the rest of the lineages of the Potato clade which are, in turn, organized into two major subclades: (1) sections Anarrhichomenum, Articulatum, Basarthrum, Etuberosum, Juglandifolia, Lvcopersicoides, Lycopersicon, and Petota, and (2) sections Herpystichum and Pteroidea. As in all other studies including these groups, sections Etuberosum, Juglandifolia, Lycopersicoides, Lycopersicon, and Petota form a strongly supported clade. Solanum oxycoccoides, a high-elevation species endemic to north-central Peru, was tentatively assigned to several groups within Solanum based on morphological evidence, but instead the species represents an independent lineage within the Potato clade, sister to the first major subclade. Crop wild relatives have a long history of use in potato breeding, particularly for pest and disease resistance, and are expected to be increasingly used in the search for tolerance to biotic and abiotic stresses. Their current and future use in crop improvement depends on their availability in ex situ germplasm collections. As these plants are impacted in the wild by habitat destruction and climate change, actions to ensure their conservation ex situ become ever more urgent. We analyzed the state of ex situ conservation of 73 of the closest wild relatives of potato (Solanum section Petota) with the aim of establishing priorities for further collecting to fill important gaps in germplasm collections. A total of 32 species (43.8%), were assigned high priority for further collecting due to severe gaps in their ex situ collections. Such gaps are most pronounced in the geographic center of diversity of the wild relatives in Peru. A total of 20 and 18 species were assessed as medium and low priority for further collecting, respectively, with only three species determined to be sufficiently represented currently. Priorities for further collecting include: (i) species completely lacking representation in germplasm collections; (ii) other high priority taxa, with geographic emphasis on the center of species diversity; (iii) medium priority species. Such collecting efforts combined with further emphasis on improving ex situ conservation technologies and methods, performing genotypic and phenotypic characterization of wild relative diversity, monitoring wild populations in situ, and making conserved wild relatives and their associated data accessible to the global research community, represent key steps in ensuring the long-term availability of the wild genetic resources of this important crop. Potato cyst nematodes ( PCN) collected in six localities in the Leningrad region of North West Russia were identified as Globodera rostochiensis pathotype Ro1 and were used for subsequent resistance tests. Seventy-nine accessions of cultivated and closely related wild potato species from the VIR collection in Russia were screened on resistance to G. rostochiensis pathotype Ro1 and on the presence of molecular markers for H1 and Gro1-4 resistance genes. No associations were detected between the resistance level of diploid and tetraploid Andean and tetraploid Chilean potato landraces (indigenous cultivars) and their related wild species and their geographical distribution or presence of PCR-based markers that are associated with the H1 and Gro1-4 genes. It is included as a quarantine pathogen in 55 countries, with losses in susceptible cultivars reaching 50-100%. The aim of our study was to characterize the resistance to S. endobioticum pathotype 1 in cultivated potatoes from a well-characterized subset of the Vavilov Institute of Plant Industry collection and to determine whether this resistance is associated with cultivated potato species taxonomy, with ploidy, with geographic distance or with a molecular marker Nl25-1400 proposed for molecular screening for resistance to pathotype 1 of S. endobioticum. Within the diversity of 52 landrace genotypes, our work shows a lack of such predictive associations with wart resistance. High intraspecific variation of wart diseases resistance allows the selection of extremely resistant and susceptible genotypes available for future genetic and breeding studies. Background: Recent genomic studies have drastically altered our knowledge of polyploid evolution. Wild potatoes (Solanum section Petota) are a highly diverse and economically important group of about 100 species widely distributed throughout the Americas. Thirty-six percent of the species in section Petota are polyploid or with diploid and polyploid cytotypes. However, the group is poorly understood at the genomic level and the series is ideal to study polyploid evolution. Two separate studies using the nuclear orthologs GBSSI and nitrate reductase confirmed prior hypotheses of polyploid origins in potato and have shown new origins not proposed before. These studies have been limited, however, by the use of few accessions per polyploid species and by low taxonomic resolution, providing clade-specific, but not species-specific origins within clades. The purpose of the present study is to use six nuclear orthologs, within 54 accessions of 11 polyploid species, 34 accessions of 29 diploid species of section Petota representing their putative progenitors, and two outgroups, to see if phenomena typical of other polyploid groups occur within wild potatoes, to include multiple origins, loss of alleles, or gain of new alleles. Results: Our results increase resolution within clades, giving better ideas of diploid progenitors, and show unexpected complexity of allele sharing within clades. While some species have little diversity among accessions and concur with the GBSSI and nitrate reductase results, such as S. agrimonifolium, S. colombianum, S. hjertingii, and S. moscopanum, the results give much better resolution of species-specific progenitors. Seven other species, however, show variant patterns of allele distributions suggesting multiple origins and allele loss. Complex three-genome origins are supported for S. hougasii, and S. schenckii, and one of the ten accessions of S. stoloniferum. A very unexpected shared presence of alleles occurs within one clade of S. verrucosum from Central America, and S. berthaultii from South America in six polyploid species S. demissum, S. hjertingii, S. hougasii, S. iopetalum, S. schenckii, and S. stoloniferum. Conclusions: Our results document considerable genomic complexity of some wild potato polyploids. These can be explained by multiple hybrid origins and allele losses that provide a clear biological explanation for the taxonomic complexity in wild potato polyploids. These results are of theoretical and practical benefit to potato breeders, and add to a growing body of evidence showing considerable complexity in polyploid plants in general. Solanum tuberosum, the cultivated potato of world commerce, is a primary food crop worldwide. Wild and cultivated potatoes form the germplasm base for international breeding efforts to improve potato in the face of a variety of disease, environmental and agronomic constraints. A series of national and international genebanks collect, characterize and distribute germplasm to stimulate and aid potato improvement. A knowledge of potato taxonomy and evolution guides collecting efforts, genebank operations and breeding. Past taxonomic treatments of wild and cultivated potato have differed tremendously among authors with regard to both the number of species recognized and the hypotheses of their interrelationships. In total, there are 494 epithets for wild and 626 epithets for cultivated taxa, including names not validly published. Recent classifications, however, recognize only about 100 wild species and four cultivated species. This paper compiles, for the first time, the epithets associated with all taxa of cultivated potato (many of which have appeared only in the Russian literature), places them in synonymy and provides lectotype designations for all names validly published where possible. Solanum section Petota is taxonomically difficult, partly because of interspecific hybridization at both the diploid and polyploid levels. The taxonomy of cultivated potatoes is particularly controversial. Using DNA sequence data of the waxy gene, we here infer relationships among the four species of cultivated potatoes accepted in the latest taxonomic treatment ( S. ajanhuiri, S. curtilobum, S. juzepczukii and S. tuberosum, the latter divided into the Andigenum and Chilotanum Cultivar Groups). The data support prior ideas of hybrid origins of S. ajanhuiri from the S. tuberosum Andigenum Group (2 x = S. stenotomum) × S. megistacrolobum; S. juzepczukii from the S. tuberosum Andigenum Group (2 x = S. stenotomum) × S. acaule; and S. curtilobum from the S. tuberosum Andigenum Group (4 x = S. tuberosum subsp. andigenum) × S. juzepczukii. For the tetraploid cultivar-groups of S. tuberosum, hybrid origins are suggested entirely within much more closely related species, except for two of three examined accessions of the S. tuberosum Chilotanum Group that appear to have hybridized with the wild species S. maglia. Hybrid origins of the crop/weed species S. sucrense are more difficult to support and S. vernei is not supported as a wild species progenitor of the S. tuberosum Andigenum Group. A major justification for taxonomic and biogeographic research is its assumed ability to predict the presence of traits in a group for which the trait has been observed in only a representative subset of the group. Such predictors are regularly used by breeders interested in choosing potential sources of disease and pest resistant germplasm for cultivar improvement, by genebank managers to organize the collection, and by germplasm collectors planning to gather maximum diversity. The present study tests taxonomic and biogeographic associations with 10,738 disease and pest evaluations, derived from the literature and genebank records, of 32 pest and diseases in five classes of organisms (bacteria, fungi, insects, nematodes, and virus). The data show that ratings for only Colorado potato beetle [Leptinotarsa decemlineata (Say)] and one pathogen (Potato M Carlavirus) are reliably predicted both by host taxonomy and climatic variables. While it is logical to initially take both taxonomy and geographic origin into account while screening genebank materials for pest and disease resistances, such associations will hold for only for a small subset of resistance traits. The fingerprinting of 742 potato landraces with 51 simple sequence repeat (SSR, or microsatellite) markers resulted in improving a previously constructed potato genetic identity kit. All SSR marker loci were assayed with a collection of highly diverse landraces of all species of cultivated potato with ploidies ranging from diploid to pentaploid. Loci number, amplification reproducibility, and polymorphic information content were recorded. Out of 148 SSR markers of which 30 are new, we identified 58 new SSR marker locations on at least one of three potato genetic linkage maps. These results permitted the selection of a new potato genetic identity kit based on 24 SSR markers with two per chromosome separated by at least 10 cM, single locus, high polymorphic information content, and high quality of amplicons as determined by clarity and reproducibility. The comparison of a similarity matrix of 742 landraces obtained with the 24 SSR markers of the new kit and with the entire dataset of 51 SSR markers showed a high correlation ( r = 0.94) by a Mantel test and even higher correlations ( r = 0.99) regarding topological comparisons of major branches of a neighbor joining tree. This new potato genetic identity kit is able to discriminate 93.5% of the 742 landraces compared to 98.8% with 51 SSR markers. In addition, we made a marker-specific set of allele size standards that conveniently and unambiguously provide accurate sizing of all alleles of the 24 SSR markers across laboratories and platforms. The new potato genetic identity kit will be of particular utility to standardize the choice and allele sizing of microsatellites in potato and aid in collaborative projects by allowing cumulative analysis of independently generated data. Neo-Tuberosum refers to cultivated potato adapted to long-day tuberization and a syndrome of related morphological and physiological traits, developed by intercrossing and selection of short-day adapted potatoes of the Solanum tuberosum Andigenum Group, native from the Andes of western Venezuela to northern Argentina. This re-creation of the modern potato helped support the theory of an Andigenum Group origin of potato in temperate regions and the possibility to access the largely untapped diversity of the Andigenum Group germplasm by base broadening breeding. This Neo-Tuberosum derived theory, the re-creation of the modern potato from Andigenum germplasm, has been universally accepted for almost 40 years, and has had tremendous impact in planning some breeding programs and supporting phylogenetic conclusions in cultivated potato. We show, with microsatellite (simple sequence repeat, SSR) and plastid DNA marker data, that Neo-Tuberosum germplasm is closely related to Chilotanum Group landraces from lowland south-central Chile rather than to Andigenum Group germplasm. We interpret this quite unexpected result to be caused by strong rapid selection against the original Andigenum clones after unintended hybridization with Chilotanum Group germplasm. In addition, we show that Neo-Tuberosum and Andigenum Group germplasm did not serve to broaden the overall genetic diversity of advanced potato varieties, but rather that Neo-Tuberosum lines and lines not using this germplasm are statistically identical with regard to genetic diversity as assessed by SSRs. Thirty-six percent of the wild potato (Solanum L. section Petota Dumort.) species are polyploid, and about half of the polyploids are tetraploid species (2n = 4x = 48). Determination of the type of polyploidy and development of the genome concept for members of section Petota traditionally has been based on the analysis of chromosome pairing in species and their hybrids and, most recently, DNA sequence phylogenetics. Based on these data, the genome designation AABB was proposed for Mexican tetraploid species of series Longipedicellata Buk. We investigated this hypothesis with genomic in situ hybridization (GISH) for both representatives of the series, S. stoloniferum Schltdl. and S. hjertingii Hawkes. GISH analysis supports an AABB genome constitution for these species, with S. verrucosum Schltdl. (or its progenitor) supported as the A genome donor and another North or Central American diploid species (S. cardiophyllum Lindl., S. ehrenbergii (Bitter) Rydb., or S. jamesii Torrey) as the B genome donor. GISH analysis of chromosome pairing of S. stoloniferum also confirms the strict allopolyploid nature of this species. In addition, fluorescence in situ hybridization data suggest that 45S rDNA regions of the two genomes of S. stoloniferum were changed during coevolution of A and B genomes of this allotetraploid species. The article focuses on the revision of the Solanum medians Complex (Solanum section Petota) which are widely distributed along the coastal lomas and up the western slopes of the Andes Mountains from central Peru and northern Chile. Research shows that its fertile diploid and sterile triploid cytotypes are common and believed to be associated with morphological variants. It was found that so much overlap of their taxonomy is impractical to use morphology for reliable identifications.

Solanum sect. Petota (tuber-bearing wild and cultivated potatoes) are a group of approximately 190 wild species distributed throughout the Americas from the southwestern United States south to Argentina, Chile, and Uruguay. Solanum series Conicibaccata are a group of approximately 40 species within sect. Petota, distributed from central Mexico to central Bolivia, composed of diploids (2n = 2x = 24), tetraploids (2n = 4x = 48) and hexaploids (2n = 6x = 64); the polyploids are thought to be polysomic polyploids. This study initially was designed to address species boundaries of the four Mexican and Central American species of series Conicibaccata with AFLP data with the addition of first germplasm collections of one of these four species, Solanum woodsonii, as a follow-up to prior morphological, chloroplast DNA, and RAPD studies; and additional species of series Conicibaccata from South America. AFLP data from 12 primer combinations (1722 polymorphic bands) are unable to distinguish polyploid species long thought to be distinct. The data suggest a complex reticulate history of the tetraploids or the need for a broad downward reevaluation of the number of species in series Conicibaccata, a trend seen in other series of sect. Chromosome pairing relationships within cultivated potato (Solonum tuberosum) and its wild tuber-bearing relatives (Solanum sect. Petota) hove been interpreted by genome formulas, developed in the early 1900s, through techniques of classic meiotic analysis of interspecific hybrids. Here we reexamine potato genome hypotheses with the first phylogenetic analysis of all major genomes of sect. Petota using cloned DNA sequences of the single-copy nuclear gene GBSSI (waxy). Our results provide the first molecular confirmation of allopolyploidy in wild potato. They both support prior hypotheses and identify novel genome origins never before proposed. The modern cultivated potato (Solanum tuberosum L.) was first recorded in Europe in the Canary Islands in 1567, but its origin has long been in dispute. Two competing hypotheses have proposed an ‘Andean’ area (somewhere from the Andean uplands from Venezuela to northern Argentina) or a lowland south-central ‘Chilean’ area, but the Andean origin hypothesis is today generally accepted. The identity of extant Canary Island potato landraces as exclusively of upland Andean origin is one part of a multicomponent argument that the European potato originated from there, rather than from Chile. We reassess these two competing hypotheses with nuclear microsatellite and chloroplast DNA analyses of 19 Canary Island landraces, 14 Andean landraces, 11 Chilean landraces, and two wild potato species as outgroups, and with chloroplast DNA data of 150 landraces from South America. Our molecular results document a wide variation of Andean- and Chilean-type cultivars on the Canary islands and possible hybrids of the two. Our new data, integrated with historical, molecular, agronomic, and crossing data, support a hypothesis that there were multiple introductions of Andean and Chilean germplasm to the Canary Islands and that the early European potato was selected from Chilean introductions long before the late blight epiphytotics of the 1840s. The cultivated potato, Solanum tuberosum, ultimately traces its origin to Andean and Chilean landraces developed by pre-Colombian cultivators. These Andean landraces exhibit tremendous morphological and genetic diversity, and are distributed throughout the Andes, from western Venezuela to northern Argentina, and in southern Chile. The wild species progenitors of these landraces have long been in dispute, but all hypotheses center on a group of ≈20 morphologically very similar tuber-bearing (Solanum section Petota) wild taxa referred to as the S. brevicaule complex, distributed from central Peru to northern Argentina. We present phylogenetic analyses based on the representative cladistic diversity of 362 individual wild (261) and landrace (98) members of potato (all tuber-bearing) and three outgroup non-tuber-bearing members of Solanum section Etuberosum, genotyped with 438 robust amplified fragment length polymorphisms. Our analyses are consistent with a hypothesis of a "northern" (Peru) and "southern" (Bolivia and Argentina) cladistic split for members of the S. brevicaule complex, and with the need for considerable reduction of species in the complex. In contrast to all prior hypotheses, our data support a monophyletic origin of the landrace cultivars from the northern component of this complex in Peru, rather than from multiple independent origins from various northern and southern members. The use of genetic resources could be more effective and efficient if we were able to predict the presence or absence of useful traits in different populations or accessions. We analyzed the extent to which taxonomic, geographic and ecological factors can predict the presence of frost tolerance in wild potatoes. We used screening data for 1646 samples from 87 species that had been collected in 12 countries in the Americas. There was a strong association of frost tolerance with species and to a lesser extent with taxonomic series. There was significant geographic clustering of areas with wild potatoes with similar levels of frost tolerance. Areas with a high level of frost tolerance are the central and southern Peruvian Andes, the lowlands of Argentina and adjacent areas, and a small area in the central Chilean Andes. There is a greater chance of finding wild potatoes with high levels of frost tolerance in areas with a yearly mean minimum temperature below 3 °C than there is in warmer areas. However, temperature is only a weak predictor of frost tolerance. Temperature data alone did not predict observed frost tolerance in eastern Argentina/Uruguay and falsely predicted it in the southwestern United States. Morphological data and single- to low-copy nuclear DNA restriction enzyme site data were used to test subspecies circumscriptions of Solanum bulbocastanum and S. cardiophyllum. A prior chloroplast DNA analysis showed these species to be part of two main clades: 1. S. bulbocastanum subsp. bulbocastanum, S. bulbocastanum subsp. dolichophyllum, S. bulbocastanum subsp. partitum, S. cardiophyllum subsp. cardiophyllum and S. cardiophyllum subsp. lanceolatum, and 2. S. cardiophyllum subsp. ehrenbergii and other Mexican diploid species. The nuclear DNA and morphological data showed S. cardiophyllum subsp. ehrenbergii to be slightly distinguishable from the other subspecies of S. cardiophyllum, and do not support the distinction of subspecies within either species. This study tests the genetic difference between landrace populations of Solanum tuberosum L. subsp, tuberosum and subsp, andigenum (Juz. & Bukasov) Hawkes using nuclear DNA microsatellites. Microsatellitc loci were amplified in subsp, andigenum (35 accessions), subsp, tuberosum (35 accessions), and other cultivated and wild species (22 accessions). A total of 208 alleles were scored from 18 microsatellite loci spread throughout all 12 chromosomes of potato. Using an infinite allele model and a least squares method of analysis, microsatellite loci separated subsp, tuberosum from subsp, andigenum, and cultivated and wild species. The Solanum brevicaule complex is a group of morphologically very similar wild and cultivated potato taxa ( Solanum sect. Petota). This study uses single to low-copy nuclear RFLPs and RAPDs to investigate their species boundaries and relationships. Cladistic analyses of both data sets are largely concordant with each other and with a recently published phenetic analyses of the same accessions using morphology. All three data sets separate members of the complex into populations from Peru and immediately adjacent northwestern Bolivia, including most cultivated species accessions, and populations from northwestern Bolivia to Argentina.
