1. From an experiment with 504 laying hens (ISA Brown strain, 18-40 weeks of age), 90 40-week old hens were used for determining digesta mean retention time (MRT) and gut weight development. This experiment comprised 6 dietary treatments according to a 2 × 3 factorial design. Factors were dietary apparent metabolisable energy (AME) concentration (11·8 vs 10·6 MJ/kg), insoluble nonstarch polysaccharides (NSP) concentration (65 vs 134 g/kg), and fine vs coarse particle sizes of added NSP. Titanium recovery in different gut segments was used as an indicator of MRT. 2. Increasing NSP concentration prolonged MRT in the crop (68 vs 34 min) and total foregut (91 vs 57 min) compared with control NSP. Reducing energy concentration prolonged MRT in the colon (26 vs 7 min), and total hind gut (30 vs 9 min), compared with control energy. Overall MRT was not affected by dietary treatments. 3. Increasing NSP concentration increased relative weights of the empty proventriculus-gizzard and its contents by 30% (25·2 vs 19·4 g/kg) and 18% (15·4 vs 13·0 g/kg), respectively, compared with control NSP diets. 4. MRT in the foregut was prolonged as daily insoluble NSP intake increased, and this was more pronounced in hens given coarsely ground NSP, compared with finely ground. A prolonged MRT in the foregut seemed to indicate a higher level of satiety, which may contribute to a lower feather pecking pressure in laying hens.

An experiment was conducted with 768 non-cage-housed ISA Brown pullets, of which 576 hens were followed during the laying period, to investigate the separate effects of dietary energy dilution and non-starch polysaccharides (NSP) concentration (oat hulls as NSP source) on eating behavior and feather damage. Day-old pullets were allotted to 1 of 6 dietary treatments according to a 3 x 2 factorial arrangement (3 dilution and 2 NSP levels), with 8 replicates (pens) per treatment. At 17 wk of age, pens with hens were allotted to 1 of 8 dietary treatments according to a 4 x 2 factorial arrangement (4 dilution and 2 NSP levels), with 6 replicates per treatment. Compared with 0% dilution level, feed intake of laying hens of 10, 15, and 20% dilution levels increased by 8.4% (9.5 g/hen per d), 16.5% (18.1 g/hen per d), and 20.9% (23.6 g/hen per d), respectively. The ME(n) intake was similar for all dilution levels. Hens fed standard-NSP laying diets had similar insoluble NSP intake for all dilution levels (9.3 g/hen per d). Insoluble NSP intake of hens fed high-NSP laying diets increased from 15.6 g/hen per day (0% dilution) to 18.9 g/hen per day (20% dilution). Providing high- vs. standard-NSP layer diet decreased relative proventriculus contents (1.1 vs. 0.3 g/kg of BW) and increased empty gizzard weight (14.3 vs. 24.4 g/kg of BW). Hens that were fed standard-NSP diets had more feather damage compared with hens fed high-NSP diets (0.58 vs. 0.30 arbitrary units). Increasing the insoluble NSP intake resulted in decreased proventricular weight and increased gizzard weight and its contents, which are indicators of improved functioning of the gut, thereby linearly reducing feather damage. Providing diluted rearing diets increased feed intake from the first weeks of life onwards. It was hypothesized that pullets were increasingly "imprinted" on feed as pecking substrate if dilution level increased. This may decrease feather pecking and could explain the improved feather condition at 49 wk of age when 15% diluted rearing diet was fed.

An experiment was conducted with 504 non-cage-housed ISA Brown laying hens from 18 to 40 wk of age to investigate the separate effects of dietary energy concentration, nonstarch polysaccharide (NSP) concentration, and particle sizes of added NSP source on the eating behavior, feather-pecking behavior, and hen performance of laying hens. Hens were allotted to 1 of 6 dietary treatments according to a 2 x 3 factorial arrangement, with 7 replicates per treatment. The factors were control and low energy concentration (2,825 vs. 2,540 kcal/kg), control and high NSP concentration (133 vs. 195 g/kg), and fine vs. coarse particle size of the added NSP source in the high-NSP diets. We hypothesized that eating time would be increased by feeding low-energy diets or coarsely ground, high-NSP diets, or both, resulting in reduced feather-pecking behavior, without negatively affecting hen performance. Energy reduction, NSP addition, and coarse grinding of NSP increased eating time by 14.2% (P = 0.001), 17.2% (P < 0.001), and 7.9% (P = 0.075), respectively, compared with the control level of these factors. Addition of NSP decreased eating rate (g/min) by 21.0% (P = 0.010). Layers already performed gentle feather-pecking behavior during the fifth week of the rearing period. Dietary treatments did not affect the maximal level of feather condition scores, but arise of feather damage was delayed by 10 wk in hens fed low-energy, coarsely ground, NSP-rich diets compared with hens fed control diets. Hens fed the control NSP diets showed reduced culling rates, because of less cannibalistic pecking, when energy concentration was decreased (44.1 vs. 13.1%), whereas in the high-NSP diets, culling rate decreased slightly when hens were fed the low-energy diets (31.6 vs. 28.6%; P = 0.071). Hens that were fed the low-energy diets compensated for the 10% reduction in energy concentration by a 9.3% higher maximal feed intake (143.0 vs. 130.8 g/d). Hen performance and BW gain of the hens were not affected by dietary treatments. We concluded that hens that were fed low-energy or high (coarsely ground)-NSP diets spend more time on feed intake, compared with hens that were fed the control diets. As a result, hens in some treatments showed less feather-pecking behavior.

An experiment was conducted to investigate the effects of polysaccharide extracts (E) of two mushrooms, Lentinus edodes (LenE) and Tremella fuciformis (TreE), and an herb, Astragalus membranaceus (AstE), on the immune responses of chickens infected with Eimeria tenella. A total of 180 broiler chickens were assigned to nine groups: three groups were fed with each of the extracts (LenE, TreE, and AstE), three groups were fed with the extracts and immunized with live oocyst vaccine (LenE+V, TreE+V, and AstE+V), a group was immunized with the vaccine only, and there were two controls (E. tenella-infected and noninfected groups). The oocyst vaccine was given at 4 days of age, and the extracts (1 g/kg of the diet) were supplemented from 8 to 14 days of age. At 18 days of age, all birds except those in the noninfected group were infected with 9 x 10(4) sporulated oocysts. The results showed that at 7 days postinfection (p.i.), birds fed the extracts without vaccination had lower body weight (BW) gain than those given the vaccine only. However, the extracts in conjunction with the vaccine significantly enhanced BW gain of the infected chickens compared with the vaccine group. Of the three extracts, LenE and TreE showed a better growth-promoting effect. The extracts largely increased oocyst excretion of droppings during the primary response postvaccination. The cecal peak oocyst output and lesion scores measured at 7 days p.i. were higher in the groups fed the extracts than in the group immunized with the vaccine only, whereas those of the groups fed with the extracts and immunized with the vaccine were not significantly different from the vaccine group. Of the three extracts, both LenE- and AstE-fed groups showed lower cecal oocyst output. Thus, as compared with the extracts, the live, attenuated vaccine showed better results with significantly increased immune response in coccidial infected birds. The polysaccharide extracts may prove useful against avian coccidiosis, and, particularly when they are used in conjunction with vaccine, they have shown preliminary promise against the experimental coccidial infection.

The effects of feeding additional starch or fat from d 85 of gestation until parturition on litter performance and on glucose tolerance in sows that were fed a diet with a high level of fermentable nonstarch polysaccharides (NSP) were studied. The day after breeding, 141 multiparous sows were assigned to the experiment. At d 85 of gestation, sows were assigned to the treatments. Sows were fed 3.4 kg/d (as-fed basis) of a high-NSP diet or the same quantity of the high-NSP diet and an additional 360 g of starch (from wheat starch) daily, or the same quantity of the high-NSP diet and an additional 164 g of fat (from soybean oil) daily. During lactation, all sows were given free access to the same lactation diet. Approximately 1 wk before the expected time of parturition, an oral glucose tolerance test was performed in 38 randomly chosen sows by feeding pelleted glucose (3 g/kg BW0.75). Blood samples for glucose analyses were taken at -10, 10, 20, 30, 40, 50, 60, 70, 80, 90, 105, and 120 min after glucose was fed. The supply of additional dietary starch or fat did not increase piglet birth weight or total litter weight at birth. Sows that were fed the high-NSP diet had more (P = 0.097) live-born piglets and fewer (P = 0.084) stillborn piglets than did sows that were fed additional fat, whereas sows that were fed additional starch were intermediate for these variables. Piglet mortality after birth was not affected by dietary treatment. Body weight and backfat gains in the last month of gestation were higher for sows fed additional starch or fat than for sows fed the high-NSP diet (P < 0.001 and P = 0.017, respectively). Feed intake in lactation was greatest by sows fed the high-NSP diet, least by sows fed additional starch at the end of gestation, and intermediate by sows fed additional fat (P = 0.099). The differences in lactation feed intake did not result in differences in BW and backfat losses during lactation. Sows that were fed additional fat had the greatest glucose area under the curve (P = 0.044), indicating that these sows were less tolerant to glucose. In conclusion, feeding additional energy (starch or fat) in late-gestating sows that are fed a high-NSP diet did not increase litter weight at birth or piglet survival, but did increase maternal gain. Feeding sows additional energy from fat might induce glucose intolerance, whereas feeding sows additional energy from starch did not induce glucose intolerance.

(1) This in vivo trial was conducted to study the effects of polysaccharide extracts of two mushrooms, Lentinus edodes (LenE) and Tremella fuciformis (TreE), and a herb, Astragalus membranaceus (AstE) on growth performance, and the weights of organs and the gastrointestinal tract (GIT) of broiler chickens. (2) Three extracts (LenE, TreE and AstE) were supplemented at inclusion rates of 0.5, 1, 2, 3 and 4 g/kg from 7 to 14 d of age and compared with an antibiotic treatment group (20 mg/kg, virginiamycin (VRG) as well as a group of non-supplemented birds. (3) Body weight (BW) gain, feed intake and feed conversion ratio (FCR) of the extract-supplemented groups were not significantly different from those of the antibiotic group. Significant effects of type of extract and concentration on growth performance were found from 7 to 28 d of age. Generally, birds fed with LenE showed higher BW gain and lower FCR from 7 to 28 d of age than those fed with TreE and AstE and 2 g/kg LenE was considered the optimal inclusion rate for enhanced broiler growth. However, the extracts had no significant effect on the relative weights of organs and GIT compared with the antibiotic group. (4) The birds fed the extracts showed better growth performance than the non-supplemented birds, but were not significantly different from those fed VRG. Of the three extracts, LenE appeared to be a potential growth promoter. Future studies are needed to investigate whether the extracts can be used as alternatives for antibiotic growth promoters in challenged birds, and to elucidate the mechanisms for potentially enhanced growth performance in poultry.

We investigated the effects of polysaccharide extracts from 2 mushrooms, Lentinus edodes (LenE) and Tremella fuciformis (TreE), and an herb, Astragalus membranaceus (AstE), on cellular and humoral immune responses of Eimeria tenella-infected chickens. A total of 150 broiler chicks were assigned to 5 treatment groups: 3 groups were infected with E. tenella and fed with extracts (LenE, TreE, and AstE), and 2 control groups were infected with or without E. tenella. The 3 extracts were given at the level of 1 g/kg of the diet from 8 to 14 d of age. Specific systemic and cecum mucosal antibody production, proliferation of splenocytes, and peripheral T and B lymphocytes were measured during the 3 wk following Eimeria infection. A significantly higher production of specific IgA, IgM (at d 14 and 21 postinfection), and IgG (at d 21 postinfection) were detected in the Eimeria-infected groups fed with the extracts than in the infected group not fed with the extracts. Of the 3 extracts, TreE stimulated a slightly higher production of specific IgM (P = 0.052), and a significantly higher IgG production at 21 d postinfection. The cecal antibody production showed a similar trend to that of serum antibodies. The overall mean levels of cecal-specific IgA and IgG of the groups fed with extracts were significantly higher at 14 and 21 d postinfection compared with the group not fed with extracts. Of the 3 extracts, the AstE-fed group showed the highest IgG production at d 7 postinfection. Both TreE- and LenE-fed groups had significantly higher IgM and IgG levels compared with the AstE group at d 21 postinfection. The extract-fed groups also showed a significantly higher antigen-specific proliferation of splenocytes at 14 and 21 d postinfection compared with the group not fed with the extracts. The overall mean of erythrocyte rosette-forming cells (ERFC %) (at d 14 and 21) and erythrocyte-antibody-complement cells (EAC %) (at d 14) of the groups fed with the extracts was significantly higher compared with the group not fed the extracts. It is concluded from this study that supplementation with mushroom and herb extracts resulted in enhancement of both cellular and humoral immune responses in E. tenella-infected chickens.

The objective of this experiment was to study the effects of feeding group-housed gestating sows a diet with a high level of fermentable nonstarch polysaccharides (NSP; approximately 45% sugar beet pulp as fed) ad libitum on the development in individual feed intake characteristics and reproductive performance during three successive reproduction cycles. Performance of the ad libitum-fed sows was compared to the performance of sows that were fed a conventional diet restrictedly. Feed intake characteristics during gestation were only measured in the ad libitum-fed sows. One hundred and nineteen sows were assigned to one of two gestation feeding regimens. Gestating sows were fed a conventional Dutch diet restrictedly or a diet with a high level of fermentable NSP ad libitum. During lactation, sows were given free access to a commercial lactation diet from d 6 after parturition onward. The ad libitum-fed sows ate 1.3 kg/d more during gestation than the restrictedly fed sows (P < 0.001), resulting in higher body weight and backfat gains during gestation (P < 0.05). Sows that were fed ad libitum during gestation lost more body weight and backfat during lactation (P < 0.001) than sows that were fed restrictedly during gestation. Feed intake during lactation, however, did not differ between sows that were fed restrictedly or ad libitum during gestation. The numbers of total piglets born, live-born and stillborn piglets, piglet birth weight, weaning-to-estrus interval, and percentage of sows that returned to estrus after first insemination were not affected by gestation feeding regimen. Mean daily voluntary feed intake (as-fed basis) over the three reproduction cycles in the ad libitum-fed gestating sows was 4.2 kg/d. Depending on the number of preceding reproduction cycles during which a sow was fed ad libitum, the maximum voluntary feed intake was reached in Parity 3, 4, or 5 and then remained stable in subsequent parities. Mean daily feed intake of the ad libitum-fed sows increased from wk 2 to 6 of gestation and then decreased to wk 15 of gestation. The mean number of daily visits with feed intake over the three reproduction cycles was 13.8. On average, ad libitum-fed sows spent 90 min/d on eating. This study shows that it is possible to feed gestating sows a diet with a high level of fermentable NSP ad libitum during three successive reproduction cycles without negative effects on reproductive performance.

An in vivo experiment was conducted to study the potential prebiotic effects of mushroom and herb polysaccharide extracts, Lentinus edodes extract (LenE), Tremella fuciformis extract, and Astragalus membranaceus Radix extract, on chicken growth and the cecal microbial ecosystem, as compared with the antibiotic Apramycin (APR). This investigation was carried out in terms of a dose-response study. The chickens were naturally infected with avian Mycoplasma gallisepticum prior to the experiment. The BW gain, cecal pH, viscosity, and predominant microbial populations were measured 1 wk after the extract and APR treatments. The extracts and APR significantly stimulated growth of the chickens infected with avian Mycoplasma gallisepticum. The average BW gain of the groups fed with the extracts was significantly lower than that of the antibiotic group. The extracts had no significant effect on cecal pH. However, cecal viscosity and microbial populations were significantly affected by feeding extracts and antibiotic. In contrast to APR, the extracts stimulated the number of the potentially beneficial bacteria (bifidobacteria and lactobacilli), while reducing the number of the potentially harmful bacteria (Bacteroides spp. and Escherichia coli). Of the 3 extracts, LenE was associated with the most cecal bifidobacteria and lactobacilli. With each increase in the LenE dose, birds tended to have higher BW gain and total aerobe and anaerobe counts. Numbers of predominant cecal bacteria, in particular, E. coli, bifidobacteria, and lactobacilli, were significantly increased with increases in the LenE dose. It would seem that these specific mushroom and herb polysaccharide extracts hold some promise as potential modifiers of intestinal microbiota in diseased chickens.

In vitro fermentabilities of two mushrooms (Lentinus edodes--LenS; Tremella fuciformis--TreS), an herb (Astragalus membranaceus--AstS), and their polysaccharide fractions (LenE, TreE, and AstE) were investigated using microflora from chicken ceca. Polysaccharides were extracted using the hot water method. The mushrooms had lower polysaccharide yields (8 to 10%) than the herb (31%). Fermentation kinetics were determined using the in vitro cumulative gas production technique. End-products, such as gas, volatile fatty acids (VFA), and ammonia, were also determined. The gas profiles of intact materials were similar for AstS and LenS. The TreS had a diphasic digestion pattern. The extracts had similar profiles to the intact materials though gas production rates were faster. Intact materials tended to produce less VFA than the extracts though LenS and AstE had the highest total VFA production overall. Intact materials contained more protein than the extracts, and therefore resulted in more branched-chain fatty acids and ammonia. Fermentation kinetics and end-point products demonstrated differences in availability of substrates between the mushrooms and herb. These medicinal mushroom and herb materials, particularly their polysaccharide extracts, show promise in altering microbial activities and composition in chicken ceca. In vivo experiments are necessary for confirmation of this hypothesis.

The effect of feeding sows a starch diet or a diet with a high level of nonstarch polysaccharides (NSP) during gestation, lactation, or both gestation and lactation during the first three parities on reproductive performance, body weight, and backfat was studied. Four-hundred and forty-four postpuberal gilts were allotted to a 2 x 2 x 2 factorial experiment. Treatments were diet composition during gestation (including the weaning-to-estrus interval; G-Starch: 274 g/kg of starch and 123 g/kg of fermentable NSP or G-NSP: 86 g/kg of starch and 300 g/kg of fermentable NSP), diet composition during lactation (L-Starch: 293 g/kg of starch and 113 g/kg of fermentable NSP or L-NSP: 189 g/kg of starch and 216 g/kg of fermentable NSP) and group-housing system during gestation (free access stalls or electronic feeding). Both gestation diets were formulated to be isoenergetic. During lactation, sows were given free access to the lactation diets from d 6 after parturition onwards. Body weight and backfat gains during gestation were lower in sows fed the G-NSP diet than in those fed the G-starch diet (P < 0.001). The effects were more pronounced in the electronic feeding system than in the free access stalls. These results indicate an overestimation of the energy value of fermentable NSP. Body weight and backfat losses during lactation were less in sows fed the G-NSP diet during gestation than in those fed the G-starch diet (P < 0.05),which can be explained by a 0.4 kg/d higher (P < 0.001) feed intake during lactation of the sows fed the G-NSP diet. Sows fed the L-NSP diet lost more backfat during lactation than sows fed the L-starch diet (P < 0.05). The number of total piglets born and live-born piglets was 0.5 piglet higher in sows fed the G-NSP diet than in those fed the G-starch diet (P < 0.05). Lactation diet did not affect the number of total piglets born or live-born piglets. This study shows that, although high NSP diets negatively influence body weight and backfat thickness of the sows, it is possible to feed sows a diet with a high level of fermentable NSP diet during both gestation and lactation without negative effects on reproductive performance. Under the conditions of this study, feeding sows a diet with a high level of fermentable NSP during gestation and a high level of starch during lactation seems the most favorable feeding strategy.

A trial was conducted with twenty group-housed pregnant sows to study the adaptation in nutrient digestibility to a starch-rich diet or a diet with a high level of fermentable non-starch polysaccharides (NSP) during a time period of 6 weeks. The starch-rich diet was primarily composed of wheat, peas and tapioca, whereas soya bean hulls and sugar beet pulp, which both are highly fermentable NSP sources, were used to formulate the NSP-rich diet. The starch-rich diet and the NSP-rich diet were formulated to contain different levels of starch (298 vs. 78 g/kg) and fermentable NSP (100 vs. 300 g/kg), but a similar level of net energy (NE) (8.36 MJ/kg). The trial consisted of a 1-week adaptation period followed by a 5-week collection period. Weekly apparent faecal digestibilities of dry matter (DM), crude protein (CP), crude fat, ash and NSP were measured by using the acid-insoluble ash marker method. Apparent faecal digestibilities of DM and organic matter (OM) of both diets were similar. Faecal digestibility of CP and crude fat was lower (p < 0.001) whereas that of NSP was higher (p < 0.001) for sows that received the NSP-rich diet. Calculated NE values of both diets were similar. Sows fed the NSP-rich diet produced faeces that contained a lower (p < 0.001) DM content compared with sows that were fed with the starch-rich diet. The quantity of dry faeces was the same on both diets, therefore total faeces production (as-is basis) was higher (p < 0.01) for the sows fed the NSP-rich diet. During the 5-week collection period, no changes were observed in the digestibility of DM, OM and NSP in the NE value of the diets. Digestibilities of CP and fat, however, were lower in week 1 (p < 0.05) compared with weeks 2-5 for both diets. The DM content of the faeces and the quantity of dry faeces did not change from weeks 1-5. Diet by time interaction was not observed for any of the response variables indicating that sows adapt as quickly to a diet with a high level of fermentable NSP as to a starch-rich diet. The present trial shows that, with regard to digestibility of nutrients, pregnant sows completely adapt to a NSP-rich diet (i.e., NSP from sugar beet pulp) in 2 weeks and that the time period necessary to adapt to a starch-rich diet or a diet with a high level of fermentable NSP is similar.

The dose response effect of dietary nonstarch polysaccharide (NSP) content on physical activity in relation to metabolic rate in pigs was studied. Twelve clusters of 14 pigs (50-kg castrated males) were fed one of four diets, similar in composition, except for the starch and NSP content. The difference in starch and NSP ratio was created by exchanging sugar beet pulp silage (SBPS) for tapioca. On a DM basis, diets contained 0, 5, 10, or 15% SBPS. Pigs were housed in groups and fed at 2.5 times the maintenance energy requirements. Nitrogen and energy balances were measured per cluster during a 7-d experimental period, which was preceded by a 2-wk adaptation period. Dietary composition did not affect ADG. Metabolizability decreased with increasing dietary SBPS content (P < .01). Heat production as well as energy retention were unaffected by the exchange of starch for NSP (P > .1). However, dietary composition affected energy expenditure on physical activity (P < .10). Pigs were more quiet when dietary NSP content increased. Based on heat production data and on apparent digestibility of crude protein, crude fat, and NSP, the estimated net energy value of fermented NSP was 14.8 kJ/g. This relatively high energy value of fermented NSP was mainly related to the lowered energy expenditure for physical activity, 3.9 kJ/g of fermented NSP. The present study demonstrated that the lower energetic utilization of fermented NSP compared with that of starch can be fully compensated in pigs by reducing their physical activity. Thus, energy evaluation systems should account for systematic dietary influences on physical activity.

The current study aimed to evaluate the variation in fermentation activity along the distal canine gastrointestinal tract (GIT, Exp. 1). It also aimed to assess fermentation kinetics and end product profiles of 16 dietary fibers for dog foods using canine fecal inoculum (Exp. 2). For Exp. 1, digesta were collected from the distal ileum, proximal colon, transverse colon, and rectum of 3 adult dogs. Digesta per part of the GIT were pooled for 3 dogs, diluted (1:25, wt/vol), mixed, and filtered for the preparation of inoculum. A fructan, ground soy hulls, and native potato starch were used as substrates and incubated for cumulative gas production measurement as an indicator of the kinetics of fermentation. In addition, fermentation bottles with similar contents were incubated but were allowed to release their gas throughout incubation. Fermentation fluid was sampled at 4, 8, 12, 24, 48, and 72 h after initiation of incubation, and short-chain fatty acids and ammonia were measured. Results showed comparable maximal fermentation rates for rectal and proximal colonic inocula (P > 0.05). Production of short-chain fatty acids was least for the ileal and greatest for the rectal inoculum (P < 0.05). Therefore, for in vitro studies, fecal microbiota can be used as an inoculum source but may slightly overestimate in vivo fermentation. Experiment 2 evaluated the gas production, fermentation kinetics, and end product profiles at 8 and 72 h of incubation for citrus pectin, 3 fructans, gum arabic, 3 guar gums, pea fiber, peanut hulls, soy fiber, sugar beet fiber, sugar beet pectin, sugar beet pulp, wheat fiber, and wheat middlings. Feces of 4 adult dogs were used as an inoculum source. Similar techniques were used as in Exp. 1 except for the dilution factor used (1:10, wt/vol). Among substrates, large variations in fermentation kinetics and end product profiles were noted. Sugar beet pectin, the fructans, and the gums were rapidly fermentable, indicated by a greater maximal rate of gas production (Rmax) compared with all other substrates (P < 0.05), whereas peanut hulls and wheat fiber were poorly fermentable, indicated by the least amount of gas produced (P < 0.05). Sugar beet fiber, sugar beet pulp, soy fiber, and wheat middlings were moderately fermentable with a low Rmax. Citrus pectin and pea fiber showed a similar low Rmax, but time at which this occurred was later compared with sugar beet fiber, sugar beet pulp, soy fiber, and wheat middlings (P < 0.05). Results of this study can be used to formulate canine diets that stimulate dietary fiber fermentation along the distal GIT that may optimize GIT health and stimulate the level of satiety in dogs. The objectives of the present experiment were 1) to study the effects of transport conditions and lairage duration on stress level, muscle glycolytic potential, and pork quality; and 2) to investigate whether the negative effects of high stress immediately pre slaughter are affected by preceding handling factors (transport and lairage). In a 2 × 2 × 2 factorial design, halothane-free pigs (n = 384) were assigned to either short (50 mm) and smooth or long (3 h) and rough transport; long (3 h) or short (<45 mm) lairage; and minimal or high preslaughter stress. Pigs were processed in eight groups (48 pigs per group) on various days at a commercial plant. Blood samples were taken at exsanguination to measure plasma cortisol and lactate concentrations. Muscle pH and temperature were measured at 30 and 40 mm, respectively, and both were measured at 3 h, postmortem. A LM sample was taken 135 mm postmortem to estimate glycogen content and rate of glycolysis. Pork quality attributes were assessed 23 h postmortem. Short transport increased cortisol when followed by short lairage (transport × lairage; P < 0.01). Long transport, but not lairage (P > 0.30), tended to increase (P = 0.06) muscle glycolytic potential. Long transport tended to increase (P = 0.08) electrical conductivity, and decreased a* (P < 0.01) and b* (P < 0.02) values. Decreasing lairage from 3 h to <45 mm decreased (P < 0.05) the L* value, but it did not (P > 0.10) affect other pork quality traits. High stress decreased (P < 0.001) muscle glycolytic potential, and increased (P < 0.001) plasma lactate, cortisol, muscle temperature, rate of pH decline, and ultimate pH. Except for decreased (P < 0.001) b* values, pork color was not (P > 0.40) affected by high stress, but water-holding properties (measured by electrical conductivity, filter paper moisture, and drip loss) were impaired (P < 0.001) by high stress. Fiber optic-measured light scattering and Warner-Bratzler shear force were not (P > 0.12) affected by any treatment. Comparisons with the "optimal" handling (short transport, long lairage, and minimal stress) revealed that, with regard to water-holding properties, the negative effects of high stress were aggravated by suboptimal transport and lairage conditions. High stress alone increased electrical conductivity by 56%, whereas high stress in combination with short lairage led to an 88% increase. Litter moisture contents vary greatly between and within practical poultry barns. The current experiment was designed to measure the effects of 8 different dietary characteristics on litter and excreta moisture content. Additionally, free water content and water activity of the excreta and litter were evaluated as additional quality measures. The dietary treatments consisted of nonstarch polysaccharide content (NSP; corn vs. wheat), particle size of insoluble fiber (coarse vs. finely ground oat hulls), viscosity of a nonfermentable fiber (low- and high-viscosity carboxymethyl cellulose), inclusion of a clay mineral (sepiolite), and inclusion of a laxative electrolyte (MgS04). The 8 treatments were randomly assigned to cages within blocks, resulting in 12 replicates per treatment with 6 birds per replicate. Limited effects of the dietary treatments were noted on excreta and litter water activity, and indications were observed that this measurement is limited in highmoisture samples. Increasing dietary NSP content by feeding a corn-based diet (low NSP) compared with a wheat-based diet (high NSP) increased water intake, excreta moisture and free water, and litter moisture content. Adding insoluble fibers to the wheat-based diet reduced excreta and litter moisture content, as well as litter water activity. Fine grinding of the oat hulls diminished the effect on litter moisture and water activity. However, excreta moisture and free water content were similar when fed finely or coarsely ground oat hulls. The effects of changing viscosity and adding a clay mineral or laxative deviated from results observed in previous studies. Findings of the current experiment indicate a potential for excreta free water measurement as an additional parameter to assess excreta quality besides total moisture. Microbiota plays a role in the release and absorption of nutrients from feed components, thereby affecting digesta composition and moisture content of the excreta. The objective of the current study was to determine the effects of 5 different diets varying in ingredients (medium-chain fatty acids, nonstarch polysaccharides, and starch) on the microbiota composition of ileal digesta of broiler chickens and excreta DM content. Each treatment was repeated 6 times in cages each containing 18 Ross 308 broilers, with growth performance measured from 0 to 34 d of age and excreta DM and ileal microbiota composition analyzed at 34 d of age. Microbiota composition was evaluated using a novel ribosomal RNA microarray technology containing 370 different probes covering various genera, groups of microbial species, and individual species of the chicken gut microbiota, of which 321 had a signal above the background threshold. Replacing part of the animal fat and soybean oil in the wheat-based diet with medium-chain fatty acids (MCFA; 0.3% ClO and 2.7% C12) improved feed efficiency compared with the other dietary treatments. This coincided with a suppression of gram-positive bacteria belonging to the phylum of the Firmicutes, including Lacto bacillus species, and species belonging to the family of the Enterococcaceae and Micrococcaceae, whereas the gram-negative bacteria belonging to the family of the Enterobacteriaceae were promoted. None of the other diets used in the present study notably changed the ileal digesta bacteria composition. Excreta DM content was not affected by dietary treatment. The variation between individual birds per dietary treatment was more pronounced than variation caused by feed composition, with the exception of the digesta microbiota of the birds fed the MCFA diet. It is concluded that a diet with MCFA significantly changes the ileal microbiota composition, whereas the effect of the other diets on the composition of the microbiota and excreta DM content is small in broiler chickens.
