Background: Cancer cell metastasis is facilitated by matrix-metalloproteinases through degradation of extracellular matrix (ECM) proteins and is a major cause of mortality. One of the most common remedies for cancer is chemotherapy, which has many side effects. Therefore, it seems necessary to find a way to reduce the side effects of these drugs while maintaining their anticancer effects. Quercetin (que) is a natural substance that has been reported to have anticancer activities. Objectives: This study aims at evaluating the effect of que in combination with doxorubicin (dox) on the migration of the MDA-MB-231 breast cancer cell line. Methods: The effects of que and dox on cell viability in 24h and 48 h was assessed by MTT assay. Also, the effects of the same drugs on the cancer cells migration were evaluated, using the wound healing assay. Lastly, the effects of que and dox were assessed on the expression of MMP-2 and MMP-9 genes. Results: The combination of 50 µMof que with 32 nM of dox was selected by CI comparison. The viability and migration of cancer cells and the gelatinases genes expression were decreased after treatment with individual drugs. The migration and the expression of MMP-2 and MMP-9 genes after treatment with the combination of que and dox was significantly reduced compared to the treatment with que and dox alone. Conclusions: Que inhibits the viability and migration of MDA-MB-231 cancer cells and synergistically enhances the effects of dox on the survival and migration of these cells. Background: Cyrtopodion scabrum is a kind of lizard, widely distributed in southwestern, central, and eastern Iran. In our previous studies, we showed the selective anticancer properties of Cyrtopodion scabrum extract (CSE) against certain gastrointestinal cancer cell lines with no significant harmful effects on the normal cells. Method: This in vitro study was conducted to investigate the mechanism of action of CSE in SW742 (colon cancer cell line) using western blotting, real-time polymerase chain reaction (PCR), and PI staining/flow cytometry analysis. Results: Western blot analysis showed that CSE upregulates P53 protein expression significantly in SW742 cancer cell line, while QRT-PCR results revealed that P53 mRNA did not increase accordingly. It is proposed that TP53 protein play its role as a tumor suppressor through the protein stability mechanism and not through the increase in the gene expression. Considering this fact, for TP53 stabilization, two of its target genes, p21 and mdm2, were regulated. We evaluated the mRNA expression of these two genes as well. The obtained data showed a significant increase in both genes by CSE (1000 μg/ml), suggesting that the induction of G2 cell cycle arrest, which we previously reported, may happen through P53 and P21over-expression. No significant decrease was observed in apoptosis when SW742 cells were co-treated with CSE and the inhibitor of P53 transcriptional activity, (PFT)-α, indicating that the induction of apoptosis with CSE did not occur through P53-dependent transcriptional activity. Conclusion: The obtained results herein revealed that the observed anticancer effect of CSE may occur through TP53 upregulation, yet with P53-independent transcriptional activity. Background and Aim: The diagnosis of COVID-19 is an essential step toward controlling the pandemic. For this purpose, a series of laboratory methods have been developed. This study evaluated the efficiency of the ELISA method for COVID-19 IgM and IgG detection. Materials and Methods: In this case-control study, 46 blood samples from PCR positive COVID-19 patients and 49 samples from PCR negative COVID-19 subjects were collected. Subsequently, the presence of IgM and IgG in all blood samples was assessed using the Pishtaz Teb ELISA kit. Results: The data demonstrated that among 49 PCR negative, 40 (81.6%) were IgM negative and 9 (19.1%) were IgM positive. Besides, out of the 49 PCR-negative patients, 42 (85.7%) and 7 (14.3%) were IgG negative and positive, respectively. In 46 PCR-positive individuals, 40 (87%) were IgM negative, while 6 (13 %) were IgM positive. Of the 46 PCR-positive patients, 24 (52.2%) were IgG negative, and 22(47.8%) were IgG positive. Conclusion: Our results showed that detection of SARS-CoV-2 IgG and IgM using Pishtaz Teb ELISA kit is not enough for COVID diagnosis, but it can serve as a diagnostic RNA supplement to confirm infection with SARS-CoV-2 in approved clinics and other scientific communities, owing to its easy, rapid, and inexpensive availability. Background: The endothelial nitric oxide synthase (eNOS) gene encodes the eNOS enzyme in humans; it synthesizes nitric oxide (NO), which prevents or promotes colorectal cancer (CRC) progression. Polymorphisms of eNOS are correlated with the probability of cancer, but the findings are not consistent. Objectives: The present study aimed at investigating the correlation of eNOS rs1799983 polymorphism with CRC compared to the control group. Methods: The eNOS rs1799983 polymorphism (G894T) was assessed in 100 healthy control subjects and 100 patients with CRC. Polymerase chain reaction (PCR) and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) were utilized to analyze the eNOS polymorphism. Results: The risk of CRC in genotypes TT and GT of eNOS G894T polymorphism was significantly greater than genotype GG. The frequencies of genotypes TT and GT were 6% and 37% in the controls and 28% and 44% in the patients, respectively. The frequency of TT and GT genotypes showed a significant difference between patients with CRC (adjusted odds ratio (OR = 3.1, 95% confidence interval (95% CI) = 1.6 - 3.2; P = 0.001) and controls (adjusted OR = 1.8, 95% CI = 0.6 - 2.5; P = 0.005). Conclusions: According to the results of the present study, allele T of eNOS G894T had a significant correlation with CRC even after adjusting for traditional risk factors. [ABSTRACT FROM AUTHOR]
