Understanding which structural and functional traits are linked to species’ vulnerability to embolism formation (P50) may provide fundamental knowledge on plant strategies to maintain an efficient water transport. We measured P50, wood density (WD), mean conduit area, conduit density, percentage areas occupied by vessels, parenchyma cells (PATOT) and fibers (FA) on branches of angiosperm and gymnosperm species. Moreover, we compiled a dataset of published hydraulic and anatomical data to be compared with our results. Species more vulnerable to embolism had lower WD. In angiosperms, the variability in WD was better explained by PATOT and FA, which were highly correlated. Angiosperms with a higher P50 (less negative) had a higher amount of PATOT and total amount of nonstructural carbohydrates. Instead, in gymnosperms, P50 vs PATOT was not significant. The correlation between PATOT and P50 might have a biological meaning and also suggests that the causality of the commonly observed relationship of WD vs P50 is indirect and dependent on the parenchyma fraction. Our study suggests that angiosperms have a potential active embolism reversal capacity in which parenchyma has an important role, while in gymnosperms this might not be the case.

Embolism repair ability has been documented in numerous species. Although the actual mechanism driving this phenomenon is still debated, experimental findings suggest that non-structural carbohydrates (NSC) stored in wood parenchyma would provide the osmotic forces to drive the refilling of embolized conduits. We selected 12 broadleaved species differing in vulnerability to xylem embolism (P-50) and amount of wood parenchyma in order to check direct evidence about the possible link(s) between parenchyma cells abundance, NSC availability and species-specific capacity to reverse xylem embolism. Branches were dehydrated until similar to 50% loss of hydraulic conductivity was recorded (PLC similar to 50%). Hydraulic recovery (Delta PLC) and NSC content was, then, assessed after 111 of rehydration. Species showed a different ability to recover their hydraulic conductivity from PLC similar to 50%. Removing the bark in the species showing hydraulic recovery inhibited the embolism reversal. Strong correlations between the Delta PLC and: a) the amount of parenchyma cells (mainly driven by the pith area), b) the consumption of soluble NSC have been recorded. Our results support the hypothesis that refilling of embolized vessels is mediated by the mobilization of soluble NSC and it is mainly recorded in species with a higher percentage of parenchyma cells that may be important in the hydraulic recovery mechanism as a source of carbohydrates and/or as a source of water.

The maintenance of hydraulic function during and after a drought event is crucial for tree survival, but the importance of non-structural carbohydrates (NSCs) in the recovery phase is still debated. We tested whether higher NSC availability facilitates post-drought hydraulic recovery, by applying a short-term drought (Sdr) and a long-term drought combined with shading (Ldr+sh) in Fraxinus ornus and Ostrya carpinifolia. Plants were then re-irrigated and recovery was checked 24 h later, by measuring water potential, stem percentage loss of hydraulic conductance (PLC) and NSC content. The relative magnitude of hydraulic and carbon constraints was also assessed in desiccated plants. During drought, PLC increased only in F. ornus, while it was maintained almost constant in O. carpinifolia due to tighter stomatal control of xylem pressure (i.e. more isohydric). In F. ornus, only Sdr plants maintained high NSC contents at the end of drought and, when re-irrigated, recovered PLC to control values. Whereas hydraulic failure was ubiquitous, only F. ornus depleted NSC reserves at mortality. Our results suggest that preserving higher NSC content at the end of a drought can be important for the hydraulic resilience of trees.

• The hydraulic conductance of angiosperm xylem has been suggested to vary with changes in sap solute concentrations because of intervessel pit properties. • The magnitude of the ‘ionic effect’ was linked with vessel and pit dimensions in 20 angiosperm species covering 13 families including six Lauraceae species. • A positive correlation was found between ionic effect and vessel grouping parameters, especially the portion of vessel walls in contact with neighbouring vessels. Species with intervessel contact fraction (FC) values < 0.1 showed an ionic effect between 2% and 17%, while species with FC values > 0.1 exhibited a response between 10% and 32%. The ionic effect increased linearly with the mean fraction of the total vessel wall area occupied by intervessel pits as well as with the intervessel contact length. However, no significant correlation occurred between the ionic effect and total intervessel pit membrane area per vessel, vessel diameter, vessel length, vessel wall area, and intervessel pit membrane thickness. • Quantitative vessel and pit characters are suggested to contribute to interspecific variation of the ionic effect, whereas chemical properties of intervessel pit membranes are likely to play an additional role.

Recent studies have shown that, in some species, xylem hydraulic conductivity (K(h)) increases with increasing cation concentration of xylem sap. Evidence indicates that K(h) increases as a result of the de-swelling of pit membrane pectins caused by cation neutralization of polygalacturonanes. We tested whether this ionic effect partly compensates for the embolism-induced loss of stem hydraulic conductivity (PLC) by increasing K(h) of functioning conduits. We report changes in PLC, leaf water status and potassium concentration ([K(+)]) of xylem sap measured in April and July in two evergreens (Ceratonia siliqua L. and Phytolacca dioica L.) and one deciduous tree (Platanus orientalis L.) growing in the field in Sicily. In summer, Ceratonia siliqua and Phytolacca dioica showed similar native embolism (PLC = 30-40%) and [K(+)] of xylem sap (14 to 17 mM), and K(h) of stems perfused with 10 to 25 mM KCl increased by 15 to 18% compared with K(h) of stems perfused with a low concentration of a multi-ionic solution. In contrast, native [K(+)] of sap of Platanus orientalis was 50% of that in the two evergreens in summer, with a parallel lack of detectable changes in PLC that was below 10% in both spring and summer. The ionic effect was PLC-dependent: the enhancement of K(h) induced by 10 to 25 mM KCl changed from 15% for fully hydrated stems to 50-75% for stems with PLC = 50%. In Ceratonia siliqua, PLC was less than 10% in spring and about 40% in summer; concurrently, xylem sap [K(+)] increased from 3 to about 15 mM. This [K(+)] at the recorded PLC would cause an increase in residual K(h) of about 30%. Hence, the actual reduction in water transport capacity of Ceratonia siliqua stems in summer is about 20%. Similar calculations for Phytolacca dioica suggest that the actual loss of hydraulic conductivity in stems of this species in summer would be only about 10%, and not 30% as suggested by hydraulic measurements performed in the laboratory. We conclude that an increase in [K(+)] of xylem sap might be involved in the up-regulation of residual K(h), thus substantially alleviating the embolism-induced reduction in leaf water supply.

The physiological significance of ion-mediated enhancement of xylem hydraulic conductivity (K h ) in planta has recently been questioned. The phenomenon has been suggested to be an artefact caused by the use of deionized water as a reference fluid during measurements of the impact of different ions on K h . In the present study, ion-mediated changes in K h were measured in twigs of five woody species during perfusion with 25 mM KCl compared with different reference fluids like deionized water, a commercial mineral water containing different ions (including 0.5 mM Ca 2+ ), and a 1 mM CaCl 2 solution. Both fully hydrated twigs and twigs with about 50% loss of hydraulic conductivity due to cavitation-induced embolism were tested. Adding 25 mM KCl to the three reference fluids caused K h to increase by about 20%. The KCl-mediated increase of K h was even larger (up to 100%) in embolized twigs. The presence of Ca 2+ in the reference solution decreased, but not suppressed, the KCl-mediated enhancement of K h in fully hydrated twigs of three species, but not in the other two species tested. Ca 2+ did not affect the K h response to KCl in embolized twigs. These data suggest that the recently reported suppression of the ‘ionic effect’ by the presence of calcium in the xylem sap is not a general phenomenon and that ion-mediated changes of K h may play a role in planta partially to compensate for cavitation-induced loss of xylem hydraulic conductivity.

Two clones of Olea europaea L. were studied for their potential impact on hydraulic architecture and vulnerability to xylem cavitation, when used as rootstocks. The clones used were “Leccino Minerva” (LM), showing vigorous growth and “Leccino Dwarf” (LD) with strongly reduced growth. Self-rooted LM and LD plants as well as their grafting combinations were compared, namely, LM/LD (Leccino Minerva grafted onto Leccino Dwarf rootstock) and LD/LM (Leccino Dwarf grafted onto Leccino Minerva rootstocks). Plants with LD roots (LD and LM/LD) showed significantly reduced leaf surface area compared with plants with LM roots. Xylem conduits of LD shoots were 25% more numerous than in LM shoots. When grafted onto LM rootstocks, however, LD shoots produced consistently wider and longer vessels than measured in LD self-rooted plants. This caused LD/LM plants to increase stem vulnerability to cavitation with threshold pressures for cavitation (P c) of less than 0.5 MPa compared with LD self-rooted plants that had P c of over 2.0 MPa. By contrast, although LD rootstocks caused some reduction of vessel diameter and length of LM scions, their influence on LM hydraulic architecture was too small to reduce vulnerability to cavitation of LM scions with respect to that measured for LM self-rooted plants. Our conclusion is that although dwarfing rootstocks effectively reduce grafted plant size, they do not necessarily confer higher resistance to xylem cavitation to scions which would improve plant resistance to drought.

We studied changes in the hydraulic conductance of leaves (K(leaf)) between dawn and dusk during the growth period (July) and at midday at the beginning of autumn in four tree species. The main objectives of the study were to check the extent of diurnal and seasonal changes in K(leaf) and the relationships between K(leaf), irradiance and leaf gas exchange. Two evergreen (Aleurites moluccana and Persea americana) and two deciduous trees (Platanus orientalis and Quercus rubra) were studied. Leaf hydraulic conductance was measured every 2 h between 0700 and 1900 h in July and compared with values measured between 0900 and 1300 h in October. Other variables measured were photosynthetically active radiation (PAR), leaf conductance to water vapor (gL) and water potential (psiL). In July, K(leaf) varied by up to 75% in Pe. americana on a diurnal basis and by at least 44% in Q. rubra. The diurnal time course of K(leaf) showed a distinct increase between dawn and late morning (1100 h) and a subsequent decrease in the evening in A. moluccana and Pl. orientalis, whereas in the other two species, K(leaf) was highest just after dawn and lowest in the evening. In October, K(leaf) of all the species studied was lower than in July, with differences of 20 to 28% for A. moluccana and Pl. orientalis and of 66 to over 70% in Pe. americana and Q. rubra, respectively. Significant correlations were found between PAR and K(leaf) (in all species) as well as between gL and K(leaf) (in three out of four species). Leaf habit (evergreen or deciduous) did not influence absolute values of K(leaf) or its diurnal variation.

The kinetics of leaf vein recovery from cavitation-induced embolism was studied in plants of sunflower cv. Margot, together with the impact of vein embolism on the overall leaf hydraulic conductance (Kleaf). During the air-dehydration of leaves to leaf water potentials (Psi L) of -1.25 MPa, Kleaf was found to decrease by about 46% with respect to values recorded in well-hydrated leaves. When leaves, previously dehydrated to Psi L= -1.1 MPa (corresponding to the turgor loss point), were put in contact with water, Kleaf recovered completely in 10 min and so did leaf water potential. Functional vein density was estimated in both dehydrating and rehydrating leaves in terms of total length of red-stained veins infiltrated with a Phloxine B solution per unit leaf surface area. Veins were found to embolize (unstained) with kinetics showing a linear relationship with Kleaf so that about a 70% loss of functional veins corresponded with a Kleaf loss of 46%. Cavitated veins recovered from embolism within 10 min from the beginning of leaf rehydration. These data indicate that: (a) leaves of sunflower underwent substantial vein embolism during dehydration; (b) vein embolism and leaf hydraulic efficiency apparently recovered from dehydration completely and rapidly upon rehydration; (c) vein refilling occurred while conduits were still at more negative xylem pressures than those required for spontaneous bubble dissolution on the basis of Henry's law. The possible consistent contribution of vital mechanisms for vein refilling is discussed.
