1. Taxonomic novelties in Magnolia-associated pleosporalean fungi in the Kunming Botanical Gardens (Yunnan, China).

Abstract

This paper represents the first article in a series on Yunnanese microfungi. We herein provide insights into Magnolia species associated with microfungi. All presented data are reported from the Kunming Botanical Gardens. Final conclusions were derived from the morphological examination of specimens coupled with phylogenetic sequence data to better integrate taxa into appropriate taxonomic ranks and infer their relationships. Shearia formosa, the type species of Shearia, lacks type material, and its phylogenetic position accordingly remains unresolved. A fresh collection of Shearia formosa, obtained from Magnolia denudata and M. soulangeana in China, therefore, designated a neotype for stabilizing the application of the species and/or genus name. Phylogenetic analyses of a combined DNA data matrix containing SSU, LSU, RPB2 and TEF loci of representative Pleosporales revealed that the genera Crassiperidium, Longiostiolum and Shearia are a well-defined monophylum. It is recognized as the family Longiostiolaceae and strongly supported by Bayesian and Maximum Likelihood methods. Its members are characterized by immersed to semi-immersed, globose to subglobose ascomata with a central, periphysate ostiole, a peridium composed of rectangular to polygonal cells, cylindrical to clavate asci, broadly fusiform, hyaline to pale brown ascospores, a coelomycetous asexual morph with pycnidial conidiomata, enteroblastic, annellidic, ampulliform, doliiform or cylindrical conidiogenous cells and cylindrical to fusiform, transverse and sometimes laterally distoseptate conidia without a sheath or with a basal lateral sheath. Nigrograna magnoliae sp. nov. is introduced from Magnolia denudata with both asexual and sexual morphs. We observed the asexual morph of Brunneofusispora sinensis from the culture and therefore amended the generic and species descriptions of Brunneofusispora., Competing Interests: No authors have competing interests.

Abstract

Laburnicola gen. nov., with four new species, Paramassariosphaeria gen. nov., with a new species and Montagnula saikhuensis sp. nov. are introduced in this paper. The phylogeny of these taxa and genera of Didymosphaeriaceae are resolved based on analysis of combined LSU, SSU, and ITS sequence data. Laburnicola is characterized by obpyriform, immersed ascomata, with a peridium fused to the host tissues, long pedicellate asci and ellipsoidal to fusoid ascospores, whose central cells have longitudinal septa and a rugged surface. Paramassariosphaeria is characterized by globose to subglobose ascomata, papillate ostioles, a peridium comprising pseudoparenchymatous cells of textura angularis, cylindric-clavate to clavate, pedicellate asci and brown, curved-fusoid ascospores, with multiple transverse septa and a mucilaginous sheath. Montagnula saikhuensis has immersed ascomata, with a peridium fused to the host tissues, long pedicellate asci and blackish brown, ellipsoidal ascospores with a single septum. The relationship of these taxa with other Didymosphaeriaceae species is discussed based on comparative morphology and phylogenetic analysis. Abstract

Nitrogen is fundamental to all life forms and is also one of the most limiting of nutrients for plant growth. Several clades of angiosperms have developed symbiotic relationships with actinorhizal bacteria that fix atmospheric nitrogen and increase access to this nutrient. However, the evolutionary patterns of actinorhizal nitrogen-fixing symbioses remain unclear to date. Furthermore the underlying environmental pressures that led to the gain of symbiotic actinorhizal nitrogen fixation have never been investigated. Here, we present the most comprehensive genus-level phylogenetic analysis of the nitrogen-fixing angiosperms based on three plastid loci. We found that actinorhizal nitrogen-fixing species are distributed in nine distinct lineages. By dating the branching events, we determined that seven actinorhizal nitrogen-fixing lineages originated during the Late Cretaceous, and two more emerged during the Eocene. We put forward a hypothesis that multiple gains of actinorhizal nitrogen-fixing symbioses in angiosperms may have been associated with increased global temperatures and high levels of atmospheric carbon dioxide during these two time periods, as well as the availability of open habitats with high light conditions. Our nearly complete genus-level time-tree for the nitrogen-fixing clade is a significant advance in understanding the evolutionary and ecological background of this important symbiosis between plants and bacteria.

Abstract

This article presents the results of an ongoing inventory of Ascomycota in Yunnan, China, carried out as part of the research project series "Exploring ascomycete diversity in Yunnan". From over 100 samples collected from diverse host substrates, microfungi have been isolated, identified and are currently being documented. The primary objective of this research is to promote the discovery of novel taxa and explore the ascomycete diversity in the region, utilising a morphology-phylogeny approach. This article represents the second series of species descriptions for the project and introduces three undocumented species found in the families Bambusicolaceae, Dictyosporiaceae and Periconiaceae, belonging to the suborder Massarineae (Pleosporales, Dothideomycetes). These novel taxa exhibit typical morphological characteristics of Bambusicola, Periconia and Trichobotrys, leading to their designation as Bambusicola hongheensis, Periconia kunmingensis and Trichobotrys sinensis. Comprehensive multigene phylogenetic analyses were conducted to validate the novelty of these species. The results revealed well-defined clades that are clearly distinct from other related species, providing robust support for their placement within their respective families. Notably, this study unveils the phylogenetic affinity of Trichobotrys within Dictyosporiaceae for the first time. Additionally, the synanamorphism for the genus Trichobotrys is also reported for the first time. Detailed descriptions, illustrations and updated phylogenies of the novel species are provided, and thus presenting a valuable resource for researchers and mycologists interested in the diversity of ascomycetes in Yunnan. By enhancing our understanding of the Ascomycota diversity in this region, this research contributes to the broader field of fungal taxonomy and their phylogenetic understanding. Abstract

Taxonomic studies of bambusicolous fungi in China and Thailand have resulted in the collection of three fascinating saprobic coelomycetes strains. Morphology coupled with combined gene analysis of ITS, LSU, TUB2, and TEF1-α DNA sequence data showed that they belong to the genus Apiospora, family Apiosporaceae. A new species from Thailand, Apiospora mukdahanensis, and new records of A. locuta-pollinis from China are herein described. In addition, based on both morphological data coupled with phylogenetics and nomenclatural analyses, A. mori is proposed as a new combination. Maximum likelihood, maximum parsimony and Bayesian analyses were performed to clarify the phylogenetic affinities of the species obtained in this study. Newly obtained strains are compared with morphologically- and phylogenetically-related taxa. The comprehensive descriptions, illustrations, and updated phylogeny are provided and discussed for intra-and intergeneric relationships within Apiospora species. [ABSTRACT FROM AUTHOR]

Abstract

During our survey into the diversity of woody litter fungi across the Greater Mekong Subregion, three rhytidhysteron-like taxa were collected from dead woody twigs in China and Thailand. These were further investigated based on morphological observations and multi-gene phylogenetic analyses of a combined DNA data matrix containing SSU, LSU, ITS, and tef1-α sequence data. A new species of Rhytidhysteron, R. xiaokongense sp. nov. is introduced with its asexual morph, and it is characterized by semi-immersed, subglobose to ampulliform conidiomata, dark brown, oblong to ellipsoidal, 1-septate, conidia, which are granular in appearance when mature. In addition to the new species, two new records from Thailand are reported viz. Rhytidhysteron tectonae on woody litter of Betula sp. (Betulaceae) and Fabaceae sp. and Rhytidhysteron neorufulum on woody litter of Tectona grandis (Lamiaceae). Abstract

During our survey of the diversity of woody litter fungi in China and Thailand, three Hermatomyces species were collected from dead woody twigs of Dipterocarpus sp. (Dipterocarpaceae) and Ehretia acuminata (Boraginaceae). Both morphology and multigene analyses revealed two taxa as new species (Hermatomyces turbinatus and H. jinghaensis) and the remaining collections as new records of H. sphaericus. Hermatomyces turbinatus is characterized by 1) dimorphic conidia, having circular to oval lenticular conidia and 2) turbinate conidia consisting of two columns with two septa composed of 2-3 cells in each column. Hermatomyces jinghaensis is characterized by dimorphic conidia, having circular to oval lenticular conidia and clavate or subcylindrical to cylindrical conidia and consisting of one or two columns with 6-8 cells in each column. Phylogenetic analyses of combined LSU, ITS, tub2, tef1-a and rpb2 sequence data supports the placement of these new taxa within Hermatomycetaceae with high statistical support. Abstract

Members of Dodonaea are broadly distributed across subtropical and tropical areas of southwest and southern China. This host provides multiple substrates that can be richly colonized by numerous undescribed fungal species. There is a severe lack of microfungal studies on Dodonaea in China, and consequently, the diversity, phylogeny and taxonomy of these microorganisms are all largely unknown. This paper presents two new genera and four new species in three orders of Dothideomycetes gathered from dead twigs of Dodonaea viscosa in Honghe, China. All new collections were made within a selected area in Honghe from a single Dodonaea sp. This suggests high fungal diversity in the region and the existence of numerous species awaiting discovery. Multiple gene sequences (non-translated loci and protein-coding regions) were analysed with maximum likelihood and Bayesian analyses. Results from the phylogenetic analyses supported placing Haniomyces dodonaeae gen. et sp. in the Teratosphaeriaceae family. Analysis of Rhytidhysteron sequences resulted in Rhytidhysteron hongheense sp. nov., while analysed Lophiostomataceae sequences revealed Lophiomurispora hongheensis gen. et sp. nov. Finally, phylogeny based on a combined dataset of pyrenochaeta-like sequences demonstrates strong statistical support for placing Quixadomyceshongheensis sp. nov. in Parapyrenochaetaceae. Abstract

Phosphate-solubilizing fungi (PSF) play an important role in increasing the bioavailability of phosphorus in soils for plants. Thirteen fungal strains, one collected from air and 12 from soil, were screened and described here in detail. These fungal strains were tested for their ability to solubilize tricalcium phosphate (TCP) on both solid and liquid Pikovskaya (PVK) media in vitro. The airborne fungal strain KUMCC 18-0196 (Aspergillus hydei sp. nov.) showed the most significant phosphate solubilizing activity on a solid PVK medium with the solubilization index (SI) (2.58 ± 0.04 cm) and the highest solubilized phosphates (1523.33 ± 47.87 μg/mL) on a liquid PVK medium. To the best of our knowledge, A. hydei sp. nov. is the first phosphate-solubilizing fungus reported from air. We also provide the identification especially for Aspergillus , Penicillium and Talaromyces , generally reported as PSF. It is important to not only screen for PSF but also identify species properly so that researchers have a clearer taxonomic picture for identifying potential taxa for future plant growth-promoting applications. Herein, A. hydei (section Nigri), Gongronella hydei , Penicillium soli (section Lanata-Divaricata) and Talaromyces yunnanensis (section Talaromyces) are fully described and introduced as new to science. These four new species are identified based on both morphological characteristics and multigene phylogenetic analyses, including the genealogical concordance phylogenetic species recognition method where necessary. Abstract

During our studies on asexual fungi colonizing herbaceous litter in northern Thailand, we discovered two new fungal species, viz. Dendryphion hydei and Torula hydei spp. nov. The latter are examined, and their morphological characters are described as well as their DNA sequences from ribosomal and protein coding genes are analysed to infer their phylogenetic relationships with extant fungi. Torula hydei is different from other similar Torula species in having tiny and catenate conidia. Dendryphion hydei can be distinguished from other similar Dendryphion species in having large conidiophores and subhyaline to pale olivaceous brown, 2–4(–5)-septate conidia. Multigene phylogenetic analyses of a combined LSU, SSU, TEF1-α, RPB2 and ITS DNA sequence dataset generated from maximum likelihood and Bayesian inference analyses indicate that T. hydei forms a distinct lineage and basal to T. fici. Abstract

Simplicillium species have a wide host range and an extensive distribution. Some species are associated with rusts, as well as other plant pathogenic fungi and play an important role in biological control. In this study, two specimens of Simplicillium were collected from Chiang Mai Province, Thailand. Simplicillium formicae sp. nov. was isolated from an infected ant and S. lanosoniveum from Ophiocordyceps unilateralis which is a new host record. Species were initially identified using ITS gene sequences and confirmed using morphology coupled with phylogenetic analyses of a combined nrLSU, nrSSU, TEF and RPB1 dataset. Simplicillium formicae differs from other species in the genus by the presence of flask-shaped synnemata and phialides with intercalary nodes. Simplicillium lanosoniveum resembles other collections of the species by its completely solitary, tapering phialides and globose to ellipsoidal conidia which adhere in a slimly head. Abstract

In this study a new record of Ganoderma tropicum is described as from Chiang Rai Province, Thailand. The fruiting body was collected on the base of a living Dipterocarpus tree. The sample is described on the basis of morphological characteristics and phylogenetic analyses, and compared with closely related taxa. Multigene phylogenetic analyses of LSU, ITS, and RPB2 highly support the placement of the G. tropicum group with isolates from China and Taiwan (Maximum likelihood 100%, Maximum parsimony 100%, and Bayesian posterior probabilities 1.00). The optimal media, pH, and temperature for mycelial growth of the G. tropicum strain KUMCC18-0046 was also investigated and is reported as: PDA, MEA, and YPD, at pH 7-8 and 25-28 °C, respectively. Abstract

There has been increasing interest in integrating a regional tree of life with community assembly rules in the ecological research. This raises questions regarding the impacts of taxon sampling strategies at the regional versus global scales on the topology. To address this concern, we constructed two trees for the nitrogen-fixing clade: (i) a genus-level global tree including 1023 genera; and (ii) a regional tree comprising 303 genera, with taxon sampling limited to China. We used the supermatrix approach and performed maximum likelihood analyses on combined matK, rbcL, and trnL-F plastid sequences. We found that the topology of the global and the regional tree of the N-fixing clade were generally congruent. However, whereas relationships among the four orders obtained with the global tree agreed with the accepted topology obtained in focused analyses with more genes, the regional topology obtained different relationships, albeit weakly supported. At a finer scale, the phylogenetic position of the family Myricaceae was found to be sensitive to sampling density. We expect that internal support throughout the phylogeny could be improved with denser taxon sampling. The taxon sampling approach (global vs. regional) did not have a major impact on fine-level branching patterns of the N-fixing clade. Thus, a well-resolved phylogeny with relatively dense taxon sampling strategy at the regional scale appears, in this case, to be a good representation of the overall phylogenetic pattern and could be used in ecological research. Abstract

More than 1,000 species of bambusicolous fungi have been reported worldwide, although most lack modern morphological description and phylogenetic investigation. Two species of bambusicolous fungi were collected in northern Thailand. Based on morphological characters, and on a comparison with type specimens, they were determined to be Pteridiospora javanica and Roussoellopsis macrospora. In a multigene phylogenetic analysis Pteridiospora javanica formed a sister clade with Astrosphaeriella stellata, while Roussoellopsis macrospora clustered with R. tosaensis in Roussoellaceae. Because the current placement of Astrosphaeriella in Dothideomycetes cannot presently be resolved, the family placement of Pteridiospora javanica is also unresolved. Based on morphology and phylogeny we epitypify our strains under the names Pteridiospora javanica and Roussoellopsis macrospora. Asexual states are also described for both species. Abstract

Dematiopleospora mariae, isolated from Ononis spinosa collected in Forlí-Cesena Province in Italy, is introduced as a new ascomycete genus and species based on morphology and combined LSU and SSU sequence analyses. Phylogenetic analysis based on maximum parsimony (MP), maximum likelihood (ML) and Mr Bayes all support Dematiopleospora as being a distinct genus within the Phaeosphaeriaceae. Dematiopleospora is distinguished from other genera in this family in having ascospores whose central cells have longitudinal septa with light end cells, ascomata with a thick peridium and necks comprising short, light brown setae. Phylogenetic analysis also separates Dematiopleospora from other genera in the Phaeosphaeriaceae. Dematiopleospora forms a sister group with Entodesmium and Chaetosphaeronema, but these three genera are clearly separated in the molecular analysis with relatively high bootstrap support (89% and 99% respectively). The new genus is compared with similar genera of Phaeosphaeriaceae and a comprehensive description, and micrographs are provided. [ABSTRACT FROM AUTHOR]

Abstract

An undetermined saprobic fungal taxon from Yunnan (China) is revealed as a new genus in Scortechiniaceae (Coronophorales). The novel taxon, Yuxiensis, is characterized by immersed to erumpent, semi-globose ascomata, which are not surrounded by any tomentum or conspicuous subiculum, a subcylindrical quellkörper in the centrum, clavate asci with long pedicels and allantoid hyaline ascospores with granular contents. Maximum likelihood and Bayesian posterior probability analyses based on LSU, ITS, tef1 and rpb2 sequence data depict a close phylogenetic relationship of the new genus to Pseudocatenomycopsis, hence, confirming its placement in Scortechiniaceae. Parasympodiellaceae, thus far belonging to Parasympodiellales, is transferred to Coronophorales based on multi-gene phylogenetic evidence. Additionally, the incertae sedis monotypic genus Arthrocristula is treated as a synonym of Parasympodiella, with Arthrocristula hyphenata recombined as Parasympodiella hyphenata comb. nov., as the type strain of Arthrocristula hyphenata clusters inside the Parasympodiellaceae clade along with other Parasympodiella taxa.
