European badgers, Meles meles, are group-living in the UK, and demarcate their ranges with shared latrines. As carnivores, badgers possess paired anal glands, but olfactory information on the content of badger anal gland secretion (AGS) is largely uninvestigated. Here, we examined the volatile organic compounds (VOCs) of AGS samples from 57 free-living badgers using solid-phase microextraction (SPME) and gas chromatography-mass spectrometry. AGS was rich in alkanes (C7-C15, 14.3% of identified compounds), aldehydes (C5-C14, 9.7%), phenols (C6-C15, 9.5%), alcohols (C5-C10, 7.3%), aromatic hydrocarbons (C6-C13, 6.8%), ketones (C6-C13, 6.3%) and carboxylic acids (C3-C12, 5.6%) and contained a variety of esters, sulfurous and nitrogenous compounds, and ethers. The number of VOCs per profile ranged from 20 to 111 (mean = 65.4; ± 22.7 SD), but no compound was unique for any of the biological categories. After normalization of the raw data using Probabilistic Quotient Normalization, we produced a resemblance matrix by calculating the Euclidian distances between all sample pairs. PERMANOVA revealed that AGS composition differs between social groups, and concentration and complexity in terms of number of measurable VOCs varies between seasons and years. AGS VOC profiles encode individual identity, sex and vary with female reproductive state, indicating an important function in intraspecific communication. Because AGS is excreted together with fecal deposits, we conclude that chemical complexity of AGS enables particularly latrine-using species, such as badgers, to advertise more complex individual-specific information than in feces alone.

Female specimen of the ragworm, Nereis succinea, employs a tetra-peptide, cysteinyl-glutathione (CSSG) as mate recognition and gamete release pheromone during reproduction. In the present study we review the role of peptide-based pheromones in Nereid worms focusing on pheromone production in females. New results demonstrate that the female ragworms produce the pheromone in the course of the oocyte maturation directly correlated to the oocyte diameter. Nevertheless, CSSG production is significantly increasing during the reproductive process, the nuptial dance, itself. The concentrations of the pheromone, nereithione (CSSG) and those of the possible precursors, reduced glutathione (GSH) and oxidized glutathione (GSSG), in the coelomic fluid were determined by HPLC showing that during spawning the female worms excrete 68.5 +/- 12.5 microg/h of the pheromone. GSH is mobilized, or synthesized and continuously converted into the pheromone enabling female N. succinea to maintain a constant level of pheromone release until the GSH reserves are depleted. Upon release CSSG induces not only gamete release in males but in doses as low as 10(-8) to 10(-9) M also significantly increases male swimming activity and as such may guide males towards the slower swimming females.

Nereis succinea and Platynereis dumerilii (Annelida, Polychaeta) are broadcast spawners and reproduce semelparously. The final events in reproduction, swarming and spawning are co-ordinated by sex pheromones. A water-soluble fraction of crude oil, the volatile fraction (C9--C16) of EKO FISK crude oil was found to induce release of gametes in male nereids at levels <0.3 ppm. Using vacuum distillation, column chromatography, preparative GC and GC-MS analysis we showed that C5-alkylated benzenes were most potent in inducing sperm release, of those n-butyl-4-methylbenzene and 1,4-diethyl-2-methylbenzene were found to induce release of gametes at concentrations ≥4 nM. This threshold is lower than those reported for natural pheromones (nereithione: 60 nM, uric acid: 600 nM) but higher than background levels of aromatic compounds of 0.05 nM and below. Other oil fractions showed additional effects, blocking pheromone reception or narcotising and intoxicating animals. Part of these effects could be assigned to naphthalenes at levels down to approx. 320 nM. In the original mixtures, their action was modified or compensated by the presence of gamete release inducing alkylated benzenes. Other highly paralysing substances remained elusive.
