Both toxicology and epidemiology are used to inform hazard and risk assessment in regulatory settings, particularly for pesticides. While toxicology studies involve controlled, quantifiable exposures that are often administered according to standardized protocols, estimating exposure in observational epidemiology studies is challenging, and there is no established guidance for doing so. However, there are several frameworks for evaluating the quality of published epidemiology studies. We previously developed a preliminary list of methodology and reporting standards for epidemiology studies, called Good Epidemiology Practice (GEP) guidelines, based on a critical review of standardized toxicology protocols and available frameworks for evaluating epidemiology study quality. We determined that exposure characterization is one of the most critical areas for which standards are needed. Here, we propose GEP guidelines for pesticide exposure assessment based on the source of exposure data (i.e., biomonitoring and environmental samples, questionnaire/interview/expert record review, and dietary exposures based on measurements of residues in food and food consumption). It is expected that these GEP guidelines will facilitate the conduct of higher-quality epidemiology studies that can be used as a basis for more scientifically sound regulatory risk assessment and policy making.

The status of seven currently used pesticides were assessed under the influence of soil parameters in surface soils of cash crop growing areas of Pakistan. Chlorpyrifos occurred in highest mean concentration (1.18 mg kg -1 ). Selected pesticides exhibited higher affinity towards both organic carbon and black carbon fractions. The δ 13 C stable carbon isotopic fraction of inorganic carbon was also used as a tracer and disclosed high retention of total organic carbon in Swat and Swabi sites. Statistical analysis revealed that carbon storage was primarily influenced by altitude and temperature. Soil clay mineral oxides of aluminum and iron positively correlated with organic carbon and selected pesticides (chlorpyrifos and cyprodinil). Soil to plant bio-concentration ratios predicted heightened uptake of azinfos and diazinon in major cash crop bio mass. There are few studies documenting the dust loaded with pesticides as a potential non-dietary exposure source for occupational worker and populations living near agricultural farms and pesticides formulation plants. In present study we have evaluated the pesticide concentration in dust from potential sites and relevant health risk from dust ingestion. Furthermore, the effect of currently used pesticides was investigated on blood and urine parameters of subjects: farmer, factory worker, urban resident and rural resident and controlled subjects with presumably different levels of exposure. The urinary metabolites (TCPY and IMPY) were quantified as biomarkers of exposure to chlorpyrifos and diazinon in relation with biomarkers of effect including BuChE, LH, FSH, testosterone and oxidative stress. Results showed that chlorpyrifos and diazinon were present in higher concentration in dust and posed a high health risk to exposed subjects. The mean SOD value was high among the farmer (3048U/g Hb) followed by factory worker (1677.6U/g Hb). The urinary biomarkers - TCPY and IMPY- were found higher in exposed subjects as compared to control. Furthermore, testosterone was found in higher concentration in factory worker than control (12.63ng/ml vs 4.61ng/ml respectively). A decreased BuChE activity was noticed in occupational group and significant differences were observed in control verses exposed subjects. The PCA analysis evidenced the impact of pesticides on exposure biomarkers and male reproductive hormones. The study suggests that dust contaminated with pesticides engenders significant health risk particularly related to the nervous and endocrine system, not only for occupational workers exposed to direct ingestion but also for nearby residential community. Worldwide amphibian populations are declining due to habitat loss, disease and pollution. Vulnerability to environmental contaminants such as pesticides will be dependent on the species, the sensitivity of the ontogenic life stage and hence the timing of exposure and the exposure pathway. Herein we investigated the biochemical tissue 'fingerprint' in spawn and early-stage tadpoles of the Common frog, Rana temporaria, using attenuated total reflection-Fourier-transform infrared (ATR-FTIR) spectroscopy with the objective of observing differences in the biochemical constituents of the respective amphibian tissues due to varying water quality in urban and agricultural ponds. Our results demonstrate that levels of stress (marked by biochemical constituents such as glycogen that are involved in compensatory metabolic mechanisms) can be observed in tadpoles present in the pond most impacted by pollution (nutrients and pesticides), but large annual variability masked any inter-site differences in the frog spawn. ATR-FTIR spectroscopy is capable of detecting differences in tadpoles that are present in selected ponds with different levels of environmental perturbation and thus serves as a rapid and cost effective tool in assessing stress-related effects of pollution in a vulnerable class of organism.

Selected organochlorine pesticides (OCs) and halogenated flame retardants (HFRs) were analyzed in surficial fjord sediments collected down the length of Kongsfjorden, Svalbard in the Norwegian high Arctic. Hexachlorocyclohexane (α-HCHs) was found to be the most abundant OC in the sediment, followed by BDE-209>chlordane>α-endosulfan>Dechlorane Plus (anti-DP)>trifluralin concentration ranges were high over the relatively small study area of the fjord (e.g. ∑HCH: 7.2-100 pg g(-1) dry weight (dw)), with concentrations broadly similar to, or lower than, measurements conducted in other parts of the Arctic. Concentrations of legacy OCs, including both HCH isomers and chlordane showed a decreasing trend from the outer, seaward end of the fjord to the inner, glacier end of the fjord. Conversely, sediment concentrations of α- and β-endosulfan (0.1-12.5 pg g(-1) dw) increased from the outer fjord to the inner fjord. This contrasting pattern may be attributed to the influence of historical vs. contemporary sources of these chemicals to the fjord area, whereby the North Atlantic/West Spitzbergen oceanic current dominates the transport and input of the legacy OCs, whereas atmospheric deposition and meltwater runoff from the glaciers influence the inner fjord sediments for endosulfan. Interestingly, BDE-209 and Dechlorane Plus did not reveal any clear spatial trend. It is plausible that both glacial runoff and oceanic current end members are playing a role in introducing these chemicals to the fjord sediments. The relatively low fractional abundance of the syn-DP isomer (fsyn), however, indicates the long-range transport of this chemical to this Arctic site., (Copyright © 2014 Elsevier Ltd. Concentrations of selected organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) are reported in air and surface soil in an extensive spatial survey across Azerbaijan, a country bordering the Caspian Sea with a history of OCP production and extensive use. Polyurethane foam disc passive air samplers (PAS) were deployed during October-November 2008 with soil samples collected in July 2009. Levels of Σ(7)PCB in ambient air were generally low (mean of 0.046 ng m(-3), n = 13) and comparable to concentrations reported in countries within Eastern Europe and similar to or lower than concentrations reported in urban air in the UK and other western countries. Surprisingly, PCB concentrations in rural/background soil fell below the method detection limits at most sites, although concentrations were 0.209 and 0.071 ng Σ(7)PCB g(-1) dry weight (dw) for two urban sites, again comparable to PCB levels measured at background sites in Europe. Levels of α-HCH, β-HCH, γ-HCH and p,p'-DDT/E were elevated in ambient air across Azerbaijan in comparison to PAS-derived concentrations reported elsewhere, with concentrations of α-HCH in air ranging from 0.085 to 2.699 ng m(-3) and p,p'-DDE, 0.037-2.290 ng m(-3). High concentrations of OCPs occurred at several of the urban sites and at sites in proximity to old pesticide storage facilities with concentrations in soil >0.1 μg g(-1) dw for p,p'-DDE and p,p'-DDT at several sites. The ratio of p,p'-DDT/p,p'-DDE was close to unity in the soil at these sites, but elsewhere, the ratio was <1, indicating a weathered DDT pattern, which was also reflected in the air at all sites. A fugacity approach revealed the strong likelihood of net soil-to-air transfer at the majority of sites for all OCPs. The calculated annual fluxes or loading to the atmosphere from a rural/agricultural area (representing land as vineyards and cotton cultivation) were estimated to be on the order of ≈10-100 kg year(-1) for the HCH isomers (including the β -isomer) and p,p'-DDE. The high levels of OCPs in soils, particularly in the vicinity of obsolete-pesticide storage facilities and at select urban sites are of concern and warrant regular monitoring activities and the development of containment or mitigation strategies.

Here we investigate the photodegradation of structurally similar organophosphorus pesticides; methyl-parathion and fenitrothion in water (20 degrees C) and ice (-15 degrees C) under environmentally-relevant conditions with the aim of comparing these laboratory findings to limited field observations. Both compounds were found to be photolyzed more efficiently in ice than in aqueous solutions, with quantum yields of degradation being higher in ice than in water (fenitrothion > methyl-parathion). This rather surprising observation was attributed to the concentration effect caused by freezing the aqueous solutions. The major phototransformation products included the corresponding oxons (methyl-paraoxon and fenitroxon) and the nitrophenols (3-methyl-nitrophenol and nitrophenol) in both irradiated water and ice samples. The presence of oxons in ice following irradiation, demonstrates an additional formation mechanism of these toxicologically relevant compounds in cold environments, although further photodegradation of oxons in ice indicates that photochemistry of OPs might be an environmentally important sink in cold environments.

Aqueous solutions of fenitrothion and methyl parathion were photochemically degraded in an Atlas Suntest solar simulator (500 W m(-2)) as well as under ambient sunlight at Lancaster University (June & August 2007, 54 degrees N) and the degradation kinetics and disappearance quantum yields are reported. Fenitrothion degradation confirmed to first order kinetics (r(2) = 0.90-0.99) with a half-life range of 4.9 h-5.3 h, shorter than previously reported studies. In contrast, methyl parathion did not show significant degradation over the duration of these experiments. Light irradiances were monitored with a spectroradiometer during the Suntest simulator and outdoor experiments. The filtered-xenon arc lamp of the Suntest yielded spectral irradiances comparable to natural sunlight in the UVB and UVA wavelength range (280-400 nm), but with higher irradiances in the visible region (400-750 nm). Nonetheless, as both compounds have light absorption spectra at wavelengths < 400 nm, then the half-lives and disappearance quantum yields were similar between the Suntest and natural sunlight, and demonstrated that the Suntest is suitable for environmentally-relevant photochemical degradation experiments.

Arctic seawater concentrations of two currently used pesticides, endosulfan and gama-HCH, were collated from a variety of cruises undertaken throughout the 1990s up to 2000 for different regions of the Arctic Ocean. Surface seawater concentrations for alpha- and beta-endosulfan ranged from <0.1-8.8 (mean 2.3) pg/L and 0.1-7.8 (mean 1.5) pg/L, while gamma-HCH concentrations were approximately 100 fold higher than alpha-endosulfan, ranging between <0.70 and 894 (mean 250) pg/L. Geographical distributions for alpha-endosulfan revealed the highest concentrations in the western Arctic, specifically in the Bering and Chukchi Seas with lowest levels toward the central Arctic Ocean. In contrast, gamma-HCH revealed higher concentrations toward the central Arctic Ocean, with additional high concentrations in the coastal regions near Barrow, Alaska and the White Sea in northwest Russia, respectively. A fugacity approach was employed to assess the net direction of air-water transfer of these two pesticides, using coupled seawater and air concentrations. For alpha-endosulfan, water-air fugacity ratios (FR) were all <1 indicating net deposition to all regions of the Arctic Ocean, with the lowest values (0.1-0.2) evident in the Canadian Archipelago. Given the uncertainty in the temperature-adjusted Henry's Law constant (factor approximately10), it is plausible that equilibrium may have been reached for this compound in the western fringes of the Arctic Ocean where the highest water concentrations were observed. Similarly, FR values for gamma-HCH were generally <1 and in agreement

Plastic cladding is increasingly used in agriculture to create micro-environments to improve crop yield/growth. Many of these plastics can alter the solar light spectrum by inhibiting or reducing the transmittance of certain parts of the solar spectrum. In this study, we investigated the aqueous photolysis of fenitrothion, under a selection of different plastic films commonly used in agriculture. Three different grades of polyethylene film were used: ¿standard¿, ¿transparent¿ and ¿opaque¿. The transmittance of light wavelengths in the UV region (290¿400nm), measured with a spectroradiometer, was found to decrease in the order of transparent>standard>opaque. Fenitrothion, an organophosphorothioate insecticide possesses molar absorptivity in the solar wavelength range of 290¿400nm. Aqueous first order degradation rate constants for fenitrothion determined over a 12h period were found to be considerably less for those experiments conducted under the standard and opaque plastic films, compared to the transparent film and no-film control. The experiments were conducted in an Atlas Suntest solar simulator using a UV-filtered Xenon arc lamp to simulate sunlight. The first order half-life for fenitrothion was 100 and 250h under the standard and opaque films, respectively, compared to ¿10h for the transparent film and no-film experiments. Our results suggest that pesticide longevity could be greatly extended within these plastic micro-environments, especially for those chemicals which may degrade/transform via photolytic or photochemical pathways.
