Background The comorbidity of depression and anxiety is associated with an increased risk of prolonged adverse mental health status. However, little is currently known about their genetic and environmental influences that help to explain both the comorbidity and distinctiveness. Using longitudinal twin data, the present study investigated both the overlapping and distinct relationships between depression and anxiety viewed from the perspective of Gray's Reinforcement Sensitivity Theory (RST): two personality traits of the Behavioral Inhibition and Activation Systems (BIS and BAS). Methods A total of 422 twin pairs (298 monozygotic and 124 dizygotic pairs) participated by completing a personality questionnaire at wave 1, and mood symptoms questionnaires at wave 2. The waves were on average 2.23 years apart. Results Multivariate Cholesky decomposition indicated that the genetic variance of the personality traits (BIS and BAS) explained all of the genetic variance in depressive and anxiety symptoms. Additionally, genetic factors related to the BIS positively explained depressive and anxiety symptoms, whereas genetic factors related to the BAS negatively explained only depressive symptoms. Limitations Limitations include shorter time interval and the reliance on self-reported data. Conclusions The present study provided evidence explaining the overlap and differentiation of depressive and anxiety symptoms by using data on personality traits in a longitudinal, genetically-informative design. The findings suggested the personality traits from Gray's RST model played an important role in the prediction, and clarified the description, of both depressive and anxiety symptoms.

Background and Objectives: Altruism is a form of prosocial behavior with the goal of increasing the fitness of another individual as a recipient while reducing the fitness of the actor. Although there are many studies on its heterogeneity, only a few behavioral genetic studies have been conducted to examine different recipient types: family members favored by kin selection, the dynamic network of friends and acquaintances as direct reciprocity, and strangers as indirect reciprocity. Materials and Methods: This study investigated the genetic and environmental structure of altruism with reference to recipient types measured by the self-report altruism scale distinguished by the recipient (the SRAS-DR) and examine the relationship to personality dimensions measured by the NEO-FFI with a sample of 461 adult Japanese twin pairs. Results: The present study shows that there is a single common factor of altruism: additive genetic effects explain 51% of altruism without a shared environmental contribution. The genetic contribution of this single common factor is explained by the genetic factors of neuroticism (N), extraversion (E), openness to experience (O), and conscientiousness (C), as well as a common genetic factor specific to altruism. Only altruism toward strangers is affected by shared environmental factors. Conclusions: Different types of altruistic personality are constructed by specific combinational profiles of general personality traits such as the Big Five as well as a genetic factor specific to altruism in each specific way.

Using a genetically informative design (about 2000 twin pairs), we investigated the phenotypic and genetic and environmental architecture of a broad construct of conscientiousness (including conscientiousness per se, effortful control, self-control, and grit). These four different measures were substantially correlated; the coefficients ranged from 0.74 (0.72–0.76) to 0.79 (0.76–0.80). Univariate genetic analyses revealed that individual differences in conscientiousness measures were moderately attributable to additive genetic factors, to an extent ranging from 62 (58–65) to 64% (61–67%); we obtained no evidence that shared environmental influences were observed. Multivariate genetic analyses showed that for the four measures used to assess conscientiousness, genetic correlations were stronger than the corresponding non-shared environmental correlations, and that a latent common factor accounted for over 84% of the genetic variance. Our findings suggest that individual differences in the four measures of conscientiousness are not distinguishable at both the phenotypic and behavioural genetic levels, and that the overlap was substantially attributable to genetic factors.

Background/objectives: Many personality traits correlate with BMI, but the existence and direction of causal links between them are unclear. If personality influences BMI, knowing this causal direction could inform weight management strategies. Knowing that BMI instead influences personality would contribute to a better understanding of the mechanisms of personality development and the possible psychological effects of weight change. We tested the existence and direction of causal links between BMI and personality. Subjects/methods: We employed two genetically informed methods. In Mendelian randomization, allele scores were calculated to summarize genetic propensity for the personality traits neuroticism, worry, and depressive affect and used to predict BMI in an independent sample (N = 3 541). Similarly, an allele score for BMI was used to predict eating-specific and domain-general phenotypic personality scores (PPSs; aggregate scores of personality traits weighted by BMI). In a direction of causation (DoC) analysis, twin data from five countries (N = 5424) were used to assess the fit of four alternative models: PPSs influencing BMI, BMI influencing PPSs, reciprocal causation, and no causation. Results: In Mendelian randomization, the allele score for BMI predicted domain-general (β = 0.05; 95% CI: 0.02, 0.08; P = 0.003) and eating-specific PPS (β = 0.06; 95% CI: 0.03, 0.09; P < 0.001). The allele score for worry also predicted BMI (β = −0.05; 95% CI: −0.08, −0.02; P < 0.001), while those for neuroticism and depressive affect did not (P ≥ 0.459). In DoC, BMI similarly predicted domain-general (β = 0.21; 95% CI:, 0.18, 0.24; P < 0.001) and eating-specific personality traits (β = 0.19; 95% CI:, 0.16, 0.22; P < 0.001), suggesting causality from BMI to personality traits. In exploratory analyses, links between BMI and domain-general personality traits appeared reciprocal for higher-weight individuals (BMI > ~25). Conclusions: Although both genetic analyses suggested an influence of BMI on personality traits, it is not yet known if weight management interventions could influence personality. Personality traits may influence BMI in turn, but effects in this direction appeared weaker.

This study used a behavioral genetic approach to examine the genetic and environmental etiology of stability and changes in self-esteem in relation to personality. Multiple genetic analyses were conducted on a longitudinal dataset of self-esteem and Big Five personality scores among young adult Japanese twins over the course of a decade. There were 1221 individuals for whom data were available on both self-esteem and the Big Five personality test at Time 1 and 365 at Time 2. The mean interval between the two times was 9.95 years. Genetic effects on self-esteem were robust, and the same genes were responsible for the stability of self-esteem in individuals over time. Nearly half of the variance in self-esteem was explained by a new genetic factor arising during the decade, suggesting that genetic innovation of self-esteem occurred in early adult life. The genetic and environmental covariance structure between personality and self-esteem in individuals was constant over a decade, providing evidence that the stability of self-esteem was largely attributable to personality. However, genetics for self-esteem independent of personality still contributed to stability over time, differentiating the concept of self-esteem from personality as a trait in terms of its genetic and environmental etiological levels.

Mõttus and colleagues (2017) reported evidence that the unique variance in specific personality characteristics captured by single descriptive items often displayed trait-like properties of cross-rater agreement, rank-order stability, and heritability. They suggested that the personality hierarchy should be extended below facets to incorporate these specific characteristics, called personality nuances. The present study attempted to replicate these findings, employing data from 6,287 individuals from 6 countries (Australia, Canada, Czech Republic, Denmark, Japan, and United States). The same personality measure-240-item Revised NEO Personality Inventory-and statistical procedures were used. The present findings closely replicated the original results. When the original and current results were meta-analyzed, the unique variance of nearly all items (i.e., items' scores residualized for all broader personality traits) showed statistically significant cross-rater agreement (median = .12) and rank-order stability over an average of 12 years (median = .24), and the unique variance of the majority of items had a significant heritable component (median = .14). These 3 item properties were intercorrelated, suggesting that items systematically differed in the degree of reflecting valid unique variance. Also, associations of items' unique variance with age, gender, and body mass index (BMI) replicated across samples and tracked with the original findings. Moreover, associations between item residuals and BMI obtained from one group of people allowed for a significant incremental prediction of BMI in an independent sample. Overall, these findings reinforce the hypotheses that nuances constitute the building blocks of the personality trait hierarchy, their properties are robust and they can be useful. The second to fourth digit ratio has been argued to reflect prenatal hormonal influences and is reportedly associated with various psychological and behavioral traits, such as sexual orientation, cognitive abilities, and personality. We examined genetic and environmental influences on the second to fourth digit ratio (2D:4D) using a Japanese twin sample (N = 300). The genetic analysis showed substantial additive genetic influences for both right and left hand 2D:4D. The rest of the variance was explained mainly by environmental influences not shared within twin pairs. These findings were, in general, in accordance with preceding studies with primarily Caucasian twin samples. The bivariate genetic analysis revealed that the additive genetic influences were largely shared between the right and left hand, while the non-shared environmental influences were largely unique to each hand. Results from a comparison of opposite-sex and same-sex twins were not significant, although they were in the predicted direction according to the prenatal hormone transfer hypothesis. Female monozygotic twin pairs discordant in sexual orientation showed significant within-pair differences in left hand 2D:4D, where non-heterosexual twins had lower (more masculinized) 2D:4D. In addition, we found that non-heterosexual male MZ twins had larger (more feminized) 2D:4D than their heterosexual co-twins. These results suggest the existence of non-shared environmental influences that affect both 2D:4D and sexual orientation.

The present research examined the relationship between heritability and response time for the Big Five personality facets in three samples of adults and adolescents. We predicted that the larger the heritability of a facet the faster is the response to the items of that facet. In support of our predictions, the results showed that heritability and response time were indeed negatively correlated. The effect size of the relationship was small but systematic and statistically significant across all samples. The findings underline the generality of personality heritability and highlight the link between heritability and behavioral indicators. The potential usefulness of both heritability estimates and response time in research on personality is discussed.

Since natural selection produces genetically homogeneous populations with regard to adaptively important traits, the relatively high genetic variance associated with human personality is an enigma. In this article, we propose that humans adaptively control the activation of domain-specific mental mechanisms in accordance with personality. This process functions to reduce fitness differences among individuals with different genetic backgrounds associated with personality. Such control would facilitate the evolution of heritable personality traits. We conducted a twin study, showing that the level of general trust (trust of strangers) is controlled not only by environmental factors but also by personality factors, thus producing reactive heritability of general trust. This result supports our hypothesis.

Although there is a great deal of interest in the Behavioral Inhibition System (BIS) and Behavioral Activation System (BAS)—two temperaments formulated by Gray’s reinforcement sensitivity theory—few studies have examined the genetic and environmental etiology underlying their continuity and change. Using a two-wave longitudinal data set of twins from late adolescence to early adulthood, this study examined (1) whether genetic influences contribute to change as well as continuity of the two temperaments and (2) whether the magnitude of genetic influences on the BIS and BAS differs across the two measurement points. The questionnaire was administered to 448 twin pairs at two waves, with an interval of approximately 2–3 years. Univariate genetic analyses revealed that genetic factors accounted for around one-third of the phenotypic variance in the BIS and BAS traits at both waves. Longitudinal bivariate analyses revealed that (1) genetic influences contribute only to continuity, whereas environmental influences contribute to both continuity and change in the two traits, and (2) the degree of genetic influences does not differ across time. These results suggest that in this age period, temporal stability of individual differences in the BIS and BAS owes more to genetic than to environmental factors.

We examined whether effortful control (EC), a temperament proposed by Rothbart and Bates (1998), has genetically coherent structure. A self- report measure of EC was administered to 450 Japanese twins (151 males and 299 females, ages 17 to 32 years) including 152 monozygotic and 73 dizygotic pairs. Univariate genetic analysis revealed that AE model fit best for the total EC as well as its subscales. The heritability estimate for total EC was 49%, and the estimates for subscales ranged between 32% and 45%. Multivariate genetic analysis revealed that the subscales of EC were genetically correlated to a high degree and environmentally correlated to a moderate degree. These results suggest that EC has substantial genetic basis and genetically coherent structure, supporting the validity of the construct. The implications to molecular genetic study and study of psychopathology were discussed.

This study investigated the possible effects of genetic and environmental gender differences in effect on individual differences by using the Bem Sex Role Inventory (BSRI) with twins. A sex/gender-limitation analysis, a behavior genetics methodology was used to the following: (a) effects of gender-specific genes, (b) gender differences in quantitative genetic effects, (c) effects of gender-specific shared environment, (d) gender differences of quantitative shared environment, and (e) gender differences of quantitative nonshared environment. Participants were adolescent and adult twins, including 111 identical male pairs, 241 identical female pairs, 36 fraternal male pairs, 65 fraternal female pairs, and 58 opposite-gender pairs. The results indicated that although masculinity and femininity were explained by genetic factors to some extent, there were no significant gender differences in the genetic factors. Moreover, because our data did not support a model which explained gender differences in the effects of specific common environment factors, no evidence was found to support the prenatal hormonal hypothesis or the existence of parenting which encouraged children's gender role personality.

In three studies, a General Factor of Personality (GFP) was found to occupy the apex of the hierarchical structure. In Study 1, a GFP emerged independent of method variance and accounted for 54% of the reliable variance in a multitrait–multimethod assessment of 391 Italian high school students that used self-, teacher-, and parent-ratings on the Big Five Questionnaire — Children. In Study 2, a GFP was found in the seven dimensions of Cloninger's Temperament and Character Inventory as well as the Big Five of the NEO PI-R, with the GFPtci correlatingr= .72 with the GFPneo. These results indicate that the GFP is practically the same in both test batteries, and its existence does not depend on being extracted using the Big Five model. The GFP accounted for 22% of the total variance in these trait measures, which were assessed in 651 pairs of 14- to 30-year-old Japanese twins. In Study 3, a GFP accounted for 32% of the total variance in nine scales derived from the NEO PI-R, the Humor Styles Questionnaire, and the Trait Emotional Intelligence Questionnaire assessed in 386 pairs of 18- to 74-year-old Canadian and U.S. twins. The GFP was found to be 50% heritable with high scores indicating openness, conscientiousness, sociability, agreeableness, emotional stability, good humor and emotional intelligence. The possible evolutionary origins of the GFP are discussed.

J. M. Digman (1997) proposed that the Big Five personality traits showed a higher-order structure with 2 factors he labeled alpha and beta. These factors have been alternatively interpreted as heritable components of personality or as artifacts of evaluative bias. Using structural equation modeling, the authors reanalyzed data from a cross-national twin study and from American cross-observer studies and analyzed new multimethod data from a German twin study. In all analyses, artifact models outperformed substance models by root-mean-square error of approximation criteria, but models combining both artifact and substance were slightly better. These findings suggest that the search for the biological basis of personality traits may be more profitably focused on the 5 factors themselves and their specific facets, especially in monomethod studies.

Since established in 1998, the Keio Twin Project (KTP) has been dedicated to investigating genetic and environmental sources contributing to human psychological traits in adolescence and young adulthood. A population-based twin registry was constructed by the KTP through the use of official residential records in the Tokyo area, and to date requests to participate in our research have generated 1040 pairs of twins and triplets of age 14 to 30, forming one of the largest twin registries in Asia. Our comprehensive datasets, obtained through questionnaires, performance tests, and physical measurements, cover a wide range of human traits: personality, psychiatry, mental health, sociality, cognition, and physical index. Demographic variables and environment of upbringing are also sought by twins and by some parents. This extensive information allows us to clarify the genetic and environmental overlap across multiple traits as well as specificities unique to single traits. Adding an evolutionary psychology perspective to the behavior genetics framework is currently being attempted in order to develop a grand theory of human genetics.

This study examined whether universality of the 5-factor model (FFM) of personality operationalized by the Revised NEO Personality Inventory is due to genetic influences that are invariant across diverse nations. Factor analyses were conducted on matrices of phenotypic, genetic, and environmental correlations estimated in a sample of 1,209 monozygotic and 701 dizygotic twin pairs from Canada, Germany, and Japan. Five genetic and environmental factors were extracted for each sample. High congruence coefficients were observed when phenotypic, genetic, and environmental factors were compared in each sample as well as when each factor was compared across samples. These results suggest that the FFM has a solid biological basis and may represent a common heritage of the human species.

There is empirical evidence that underlying the Big Five personality factors are two higher-order factors which have come to be known as "alpha" (a) and "beta" (beta). The alpha factor is defined by the agreeableness, conscientiousness, and emotional stability domains; whereas beta is delineated by extraversion and intellect. It has been argued that a and beta are important constructs because they bridge the gap between psychometric studies of personality and theories of personality development. However, it is unclear if alpha and beta are constructs that can be reliably reproduced across a diverse range of independent samples. In a sample of 1209 MZ and 701 DZ twin pairs from Canada, Germany, and Japan who completed the NEO-PI-R, factorial analyses of the five NEO-PI-R domains extracted two factors resembling alpha and beta. Subsequent multivariate genetic analyses revealed that this factor structure was a clear reflection of the organizing effects of multiple genetic influences, providing evidence for alpha and beta as stable heuristic devices that can be used to integrate personality measurement and developmental theory. The multivariate genetic and environmental structure of Cloninger's Temperament and Character Inventory (TCI) was investigated in a sample of 617 pairs of adolescent and young adult twins from Japan. Additive genetic factors accounted for 22% to 49% of the variability on all TCI temperament scales. Although the theory predicts lower heritability for the character scales, all character subscales had a substantial genetic contribution, and nonshared environmental influences accounted for the remainder. Multivariate genetic analyses showed that several subscales used to define one dimension shared a common genetic basis with subscales defining others. Using the degree of shared genetic influence as the basis to rearrange the TCI subscales into new dimensions, it was possible to create genetically independent scales. The implications for personality measurement, theory, and molecular genetic research are discussed.

Theoretical assumptions regarding the genetic and environmental structure of personality proposed in Cloninger's seven-factor model of temperament and character were verified in a Japanese sample by using the twin method. The Temperament and Character Inventory (TCI) was administered to 296 twin pairs ranging in age from 14 to 28 years old. Among four temperament dimensions (novelty seeking [NS], harm avoidance [HA], reward dependence [RD], and persistence [PS]), HA and PS showed significant additive genetic contributions and no shared environmental effect, supporting the original theoretical assumption. NS and RD could be explained by either genetic or shared environmental factors with nonshared environment. All three character dimensions (cooperativeness [CO], self-directedness [SD], and self-transcendence [ST]) could be explained exclusively by additive contributions and no shared environmental effect. Multivariate genetic analysis indicated that there were no significant associations between NS, HA, and RD, as the theory predicts, and the genetic components of PS, SD, and CO were derived from those of the temperament dimensions. The fourth genetic component, which had a substantial load specifically on ST and overlapped with PS, was identified. Although most of the nonshared environmental effects were trait-specific, the phenotypic correlation between NS and HA could be explained by nonshared environmental overlap.

Dimensional approaches have been used to describe the fundamental dimensions that underlie the entire domain of normal and pathological personality. We tested the five factor model of personality structure in a sample of Japanese twins, to clarify the contributions of genetic and environment. The revised NEO personality inventory (NEO-PI-R) was administered to 251 twin pairs, ranging in age from 15 to 27 years of age. The NEO-PI-R is a 240-item questionnaire which was developed to assess the dimensions of personality. Univariate genetic analysis showed that the AE model in which phenotypic covariances are explained only by additive genetic (A) and nonshared environment (E) is still a plausible model, and that the relative proportion of genetic influence was comparable to that reported by Loehlin (1992). Multivariate genetic analysis of the Japanese data suggested/revealed that the five factors are genetically dependent on each other and one common genetic factor mediates their interdependence. Previous studies have assumed that they are phenotypically independent and robust. Although there are sampling biases in the present study, it is noteworthy that the results for all five factors depicted by the NEO-PI-R were comparable to those reported by Western researchers, and the genetic structure of the five-factor model is complex.
