Pig pepsinogen has been reacted with N-carboxymethylisatoic anhydride to form N-carboxymethyl-anthraniloyl-(CMA-) pepsinogen, derivatized at Lysp18, Lysp23, Lysp27, Lysp30, and Lys320. Conformational change associated with activation was detected by following energy transfer from tryptophan residues of the pepsin moiety, excited at 295 nm, to CMA groups, monitored by emission above 415 nm. Efficiency of this energy transfer is a measure of conformational change. For this zymogen derivative the change in efficiency occurs with a first order rate constant of 0.041 s-1 at pH 2.4, 22 degrees, which equals the rate at which, following acidification, alkali-stable potential activity becomes alkali-labile. For the native zymogen the rate of this conversion had been shown to be identical to the rate of cleavage of the scissile bond of pepsinogen. Therefore, the correspondence in this derivative of the rates of conversion to alkali lability and change in energy transfer demonstrates that a conformational change accompanies the peptide bond cleavage of activation.

Acidification induces a conversion of canine pepsinogens by a sequential mechanism to the active pepsins. Activation in the presence of pepstatin, which strongly inhibits the pepsins but does not prevent the first step of activation, allows the isolation of the peptide released in this first step. This peptide inhibits the milk clotting activity of canine and also porcine pepsin. Canine pepsins obtained in the absence of pepstatin were characterized by amino acid composition, molecular weight, and activity against hemoglobin and milk and compared with those of other mammalian pepsins.

Previous experiments have suggested that a substrate binding site may appear relatively early in the sequence of transformations that pepsinogen undergoes during its unimolecular activation. To test this possibility, tert-Boc-L-valyl-[3(S)-hydroxy-4(S)-amino-5-(2-naphthyl) pentanoyl]-L-alanylisoamylamide, a fluorescent analogue of pepstatin, has been used to measure the rate of appearance of the binding site. This probe is a potent inhibitor of pepsin, and its naphthyl group is an environmentally sensitive fluorophore that permits binding to be detected by fluorescence spectroscopy. In stopped-flow experiments, the fluorescence change following acidification of pepsinogen due to binding of the probe was found to obey a two-term exponential decay law. Analysis of the data obtained at various pH values permits us to rule out a sequential model for the kinetics. Rather, we were able to demonstrate that a concurrent model, with two species undergoing simultaneous transformation at different rates, is consistent with the results. The two species are related to each other by the protonation of a single site with a pKa of approximately 2.2. We obtained essentially identical results in an analysis of the early events observed in the acidification of pepsinogen labeled with 6-(p-toluidinyl)naphthalene-2-sulfonyl chloride (Auer & Glick, 1984). The protonic equilibrium probably occurs early after exposure to acid and is proposed to lead to a conformational change that freezes the distribution between the two species. It may be that these two conformers persist as identifiable entities at later stages of activation as well.(ABSTRACT TRUNCATED AT 250 WORDS)

Chicken pepsinogen has been spin-labeled by the attachment of four nitroxides to epsilon-amino groups near the protein's amino terminus. Acidification results in a bond cleavage, generating a nonlabeled, enzymatically active protein. Electron spin resonance spectra of the spin-labeled zymogen, acidified in the presence or absence of pepstatin, are identical and indicate that the nitroxides are quite mobile, compared to the nonacidified zymogen. This mobilization is interpreted as the freeing of the peptide to which the spin-labels are attached, from the protein, subsequent to the acidification that causes a peptide bond cleavage. The rate at which the peptide leaves the protein is 1 order of magnitude slower than the cleavage of the peptide bond, measured by the rate of appearance of milk-clotting activity (first-order rate constants of 0.3 min-1 vs. 6 min-1 at pH 2, 22 degrees C). The inclusion of pepstatin, at molar ratios above 2 during activation, decreases the rate of peptide leaving. These observations, and those previously reported for activation of spin-labeled pig pepsinogen, are incorporated into a model of pepsinogen activation.

The bond cleavage step of pepsinogen activation has been investigated in a kinetic study in which the denatured products of short-term acidifications were separated on SDS-polyacrylamide gels and the peptide products were quantitated by densitometry. Although several peptide products were observed, under the conditions of the experiments (pH values between 2.0 and 2.8, 22 degrees C), the only one that was a product of an initial bond cleavage was the 44-residue peptide, which upon removal from pepsinogen yields pepsin. The rate constant for this bond cleavage is 0.015 s-1 at pH 2.4, which is the same as that at which the alkali-stable potential activity of pepsinogen had been found to convert to the alkali-labile activity of pepsin. When the conversion of zymogen to enzyme was followed by the change in fluorescence of adsorbed 6-(p-toluidinyl)naphthalene-2-sulfonate (TNS), the rate of change in TNS fluorescence was the same as the conversion to alkali lability. However, pepstatin blocked the bond cleavage of pepsinogen to pepsin, but it permitted the fluorescence change to proceed. In fact, it accelerated the apparent rate of change of TNS fluorescence by shifting the pKa of an essential conjugate acid from 1.7 to 2.6. The conversion to alkali lability, therefore, may be considered to be a composite of a relatively slow conformational change (at the measured rate), followed immediately by a relatively fast bond cleavage.

Stopped-flow measurements both with native pig pepsinogen and with a fluorescent derivative, labeled near the carboxyl terminus with a toluidinylnaphthalenesulfonyl (TNS) group at Lys364, show rapid fluorescence changes following acidification. The rate constants observed by intrinsic fluorescence of the native zymogen are distinctly greater than those exhibited by the TNS derivative in the pH range examined. The rate constants for two early events in the activation of the derivative increase as the pH decreases from pH 3 to pH 2. The fluorescent intensities of these two processes also vary with pH. Because the ratios of these amplitudes fit the Henderson-Hasselbalch equation, it is concluded that the two processes represent concurrent events, rather than sequential ones. It is proposed that a protonation separates two forms of the zymogen. The conjugate acid undergoes the slower event, whereas the conjugate base, which predominates at pH 3, undergoes the faster event. It is proposed that both these pathways result in activation.

gen derivative has been prepared, using 3-[[2,5-dioxo-1-pyrrolidinyl)oxy]carbonyl]-2,5-dihydro-2,2,5,5-tetramethyl-1H-pyrrolyl-1-oxy with 3 spin-labels/protein molecule. Two are located in the peptide that is released first (1-16) and the other is in the secondarily removed sequence (17-44). Activation of the labeled zymogen is a faithful model of native pepsinogen activation because the two processes are closely related in rate, pH dependence of rate, and sites of peptide bond cleavage. The ESR signal associated with the bound label changes during activation due to release of the activation peptide. The rate of release (0.5 min(-1) at pH 2, 22 degrees C) is an order of magnitude slower than is the rate of activation, i.e., cleavage of the peptide bond that holds the peptide to the enzyme. Activation in the presence of pepstatin, however, results in peptide release (2 min(-1) at pH 2, 22 degrees C) nearly as fast as activation occurs. At pH values between 2.5 and 3, there is a lag in the change of the ESR signal following acidification. This indicates accumulation of an intermediate with pH 8.5 labile activity that still has its activation peptide attached. The strong temperature dependence of the rate of activation (26 kcal/mol) is reflected neither in reported characteristics of pepsin catalysis nor in the measured rate of release of the activation peptide from the enzyme (13 kcal/mol).

1. A pepsinogen has been purified from dog stomach in 20% yield. 2. At pH values below 4 it spontaneously converts to a pepsin, the first order rate constant at pH 2, 22 degrees C, being 11 min-1. 3. The pepsinogen is stable to denaturation at pH values up to 9, in contrast to the pepsin which is irreversibly denatured above pH 7. 4. Its proteolytic activity against denatured hemoglobin is equal to that of pig pepsin, but in the milk-clotting assay at pH 5.5 its has only 14% the activity.

1. A pepsinogen, Ia on the basis of its electrophoretic mobility, from rainbow trout stomach, has an optimum pH near 2 for activation. 2. The cognate pepsin is denatured at pH values above 7, in contrast to the zymogen, which is slightly more alkali-stable. It has an optimum pH of 3 for proteolysis of denatured hemoglobin. 3. The intrinsic reactivity of the zymogen and pepsin (rates of activation and of proteolysis, respectively) are quite high, but as they operate at the environmental temperature of the fish, are remarkably similar to rates of activation and proteolysis by mammalian pepsinogens and pepsins.
