Analyses of the population genetic structure of schistosomes under the “Schistosomiasis Consortium for Operational Research and Evaluation” (SCORE) contrasting treatment pressure scenarios in Tanzania, Niger, and Zanzibar were performed to provide supplementary critical information with which to evaluate the impact of these large-scale control activities and guide how activities could be adjusted. We predicted that population genetic analyses would reveal information on a range of important parameters including, but not exclusive to, recruitment and transmission of genotypes, occurrence of hybridization events, differences in reproductive mode, and degrees of inbreeding, and hence, the evolutionary potential, and responses of parasite populations under contrasting treatment pressures. Key findings revealed that naturally high levels of gene flow and mixing of the parasite populations between neighboring sites were likely to dilute any effects imposed by the SCORE treatment arms. Furthermore, significant inherent differences in parasite fecundity were observed, independent of current treatment arm, but potentially of major impact in terms of maintaining high levels of ongoing transmission in persistent “biological hotspot” sites. Within Niger, naturally occurring Schistosoma haematobium/Schistosoma bovis viable hybrids were found to be abundant, often occurring in significantly higher proportions than that of single-species S. haematobium infections. By examining parasite population genetic structures across hosts, treatment regimens, and the spatial landscape, our results to date illustrate key transmission processes over and above that which could be achieved through standard parasitological monitoring of prevalence and intensity alone, as well as adding to our understanding of Schistosoma spp. life history strategies in general.

Coprological analysis is the most widely used diagnostic tool for helminth infection in both domestic and wild mammals. Evaluation of the efficacy of this technique is rare, due to the lack of availability of adult worm burden. Where information is available the majority of studies are in small ruminants and seldom in a wild host. This study of 289 wild badgers is the first to report the relationship between faecal egg/larval counts and adult worm burden in badgers whilst also evaluating the reliability of coprological analysis as a diagnostic tool for hookworm (Uncinaria criniformis) and lungworm (Aelurostrongylus falciformis) infection. The prevalence of hookworm and lungworm infection, as assessed through adult worm burden was 59.2% and 20.8% respectively. For both species of helminth, infection was consistently under-reported by coprological analysis compared to adult worm burden with a reported 41% sensitivity for hookworm and 10% for lungworm. A significant positive relationship was found between faecal counts and adult worm burden for both species of helminths. BackgroundClonorchiasis is endemic in East and Southeast Asian countries. For a preventive strategy against infectious diseases, vaccination is the most effective. Here, we evaluated the molecular characteristics and immune responses of CsAg17 protein fromClonorchis sinensis, and investigated its protective effects againstC. sinensischallenge.MethodsA cDNA clone encoding CsAg17 protein and containing a secretory signal peptide at the N-terminus was retrieved from theC. sinensistranscriptome bank. Recombinant CsAg17 B-cell epitope protein and cDNA vaccines were produced and their immune responses were evaluated in FVB mice. The proportional changes of CD3+/CD4+and CD3+/CD8+T cells were detected by flow cytometry, and immune effectors were measured by ELISA.ResultsTheCsAg17mRNA was transcribed at a higher level inC. sinensisadults than in metacercariae. The CsAg17 protein was distributed in the sperms, oral and ventral suckers, and mesenchymal tissues ofC. sinensisadults. In mice challenged withC. sinensismetacercariae, vaccination with CsAg17 protein and cDNA resulted in a reduction to 64% and 69% in worm burden, respectively. Both CsAg17 protein and cDNA vaccines increased the proportion of CD3+/CD4+and CD3+/CD8+T cells and stimulated the production of Th1 type cytokines such as interleukin (IL)-2, IL-12, and interferon-γ, while maintaining minimum levels of Th2 cytokines. The levels of IgG specific to CsAg17 protein steeply increased in the two vaccinated groups from 2 weeks after immunization. The liver tissue retained good morphology in the mice vaccinated with CsAg17 protein or cDNA, whereas severe inflammation and large serous cysts were observed in the liver of the unvaccinated mice.ConclusionsVaccination with CsAg17 protein and cDNA reduced the pathological changes in the bile duct and liver, and ameliorated the worm burdenviacellular and humoral immune responses. Thus, they may serve as good vaccine candidates againstC. sinensisinfections.

The French heartworm Angiostongylus vasorum is found in European red fox (Vulpes vulpes) and dog populations, where it appears to be spreading geographically. Once introduced into new areas, it establishes in local fox populations, typically to over 50% prevalence in a few years. High susceptibility and constant excretion of first stage larvae (L1) by the definitive hosts are prerequisites for sustaining high parasite biomass in a particular habitat. The present study explores the hypothesis that repeated ingestion of gastropods in nature will result in accumulation of adult worms and elevated excretion of L1 in feces. Experimentally infected foxes were subsequently inoculated via stomach tube once (9 weeks post initial inoculation) or twice (9 and 13 weeks post inoculation (wpi)) with 100 third stage A. vasorum larvae (L3) previously isolated from aquatic snails infected with L1 from a naturally infected dog. Despite large variation in fecal larval excretion for the individual animals within the groups, excretion of L1 was significantly higher in foxes twice inoculated as compared to foxes inoculated only once. With an outlier in the once inoculated group removed, excretion became significantly higher in the three times inoculated group. Establishment of adult worms varied and only a trend to higher worm burdens was found in the group of foxes inoculated three times. However, this became significant with the same single outlier removed. Overall, it appears that protective immunity to A. vasorum does not appear to occur in V. vulpes with animals exhibiting high infection intensities without obvious clinical signs. The increasing larval excretion in foxes being repeatedly exposed to A. vasorum L3 support the hypothesis that foxes under natural conditions may repeatedly ingest infected gastropods and remain a source of environmental contamination for several months, potentially contributing to the establishment of endemic foci through increasing L1 excretion., Graphical abstract Image 1, Highlights • Foxes were experimentally infected with 100 Angiostrongylus vasorum larvae (L3). • Two groups were later challenged once or twice. • Increasing larval counts linked to time post infection and repeated inoculation. • Data suggests role of foxes in rapid establishment of the parasite in new areas.

BackgroundSibship reconstruction is a form of parentage analysis that can be used to identify the number of helminth parental genotypes infecting individual hosts using genetic data on only their offspring. This has the potential to be used for estimating individual worm burdens when adult parasites are otherwise inaccessible, the case for many of the most globally important human helminthiases and neglected tropical diseases. Yet methods of inferring worm burdens from sibship reconstruction data on numbers of unique parental genotypes are lacking, limiting the method’s scope of application.ResultsWe developed a novel statistical method for estimating female worm burdens from data on the number of unique female parental genotypes derived from sibship reconstruction. We illustrate the approach using genotypic data onSchistosoma mansoni(miracidial) offspring collected from schoolchildren in Tanzania. We show how the bias and precision of worm burden estimates critically depends on the number of sampled offspring and we discuss strategies for obtaining sufficient sample sizes and for incorporating judiciously formulated prior information to improve the accuracy of estimates.ConclusionsThis work provides a novel approach for estimating individual-level worm burdens using genetic data on helminth offspring. This represents a step towards a wider scope of application of parentage analysis techniques. We discuss how the method could be used to assist in the interpretation of monitoring and evaluation data collected during mass drug administration programmes targeting human helminthiases and to help resolve outstanding questions on key population biological processes that govern the transmission dynamics of these neglected tropical diseases.

Taeniasis is prevalent in all regions of Thailand, except the South. Infections were more frequently found in males than females of any age from 7-83 years. Taenia saginata is the most common species throughout the country. Taenia asiatica was reported only in the province of Kanchanaburi in the Central region. Co-infections, with Taenia solium and T. asiatica or T. solium and T. saginata, were found. Hybridization between T. asiatica and T. saginata is evidence that co-infection is never found between these 2 species. Finding more than 1 worm in a single patient was not entirely rare. Genetic variation was found without correlation to its geographic distribution in T. saginata, whereas no variation was found in T. asiatica.

Helminths are associated with health- and welfare problems in organic laying hens. The present observational cross-sectional study therefore aimed to estimate the prevalence and worm burdens of intestinal helminths in organic flocks of laying hens in 8 European countries, and to identify management factors that might be associated with helminth infections, with emphasis on Ascaridia galli. Data on flock-level management factors (e.g. nutritional factors, litter quality, housing system, opening- and closing hours of popholes, pasture rotation and provision of occupational materials) were collected during a farm visit when the hens were on average 62 weeks old. Worm counts were performed for 892 hens from 55 flocks and the number of ascarid (presumably primarily A. galli) eggs per g faeces (EPG) for 881 hens from 54 flocks. The association between parasitological parameters (prevalence, worm burden and EPG) and the management factors were analysed by multivariate models. Results showed that A. galli was highly prevalent across Europe with an overall mean prevalence of 69.5% and mean worm burden of 10 worms per hen. The overall mean prevalence and worm burden for Heterakis spp. were 29.0% and 16 worms per hen, respectively, with a large variation between countries. On average, the hens excreted 576 ascarid EPG. The mean prevalence of Raillietina spp. was 13.6%. A positive correlation was found between mean A. galli worm burden and ascarid EPG. Of the analysed management factors, only pasture access time had a significant negative association with A. galli worm burden which was in contrast to the general belief that outdoor access may increase the risk of helminth infections in production animals. In conclusion, the complexity of on-farm transmission dynamics is thus a challenge when evaluating the relative importance of management factors in relation to helminth infections.

In a Letter to the Editor, Eberhard et al. question the validity of our model of skin snip sensitivity and argue against the use of skin snips to evaluate onchocerciasis elimination by mass drug administration. Here we discuss their arguments and compare model predictions with observed data to assess the validity of our model.

The aim of this study was to evaluate the efficacy of formulations of sodium alginate matrix (pellets) of the nematode predatory fungi, Duddingtonia flagrans (AC001 isolate) and Arthrobotrys robusta (I-31 isolate), in the biological control of sheep gastrointestinal nematode infections. Thirty young Bergamacia ewes were allocated into three groups: In group 1 (control), the animals received 2 g/10 kg of live weight (l.w.) of pellets without fungus; in group 2, each animal received 2 g/10 kg of l.w. of pellets of D. flagrans (0.2 g of fungus/10 kg l.w.); and in group 3, each animal received 2 g/10 kg of l.w. of pellets of A. robusta (0.2 g of fungus/10 kg l.w.). The animals of each group were kept separately under rotational grazing. Pellets, with or without fungi, were mixed with 1 kg animal food and administered twice a week for 6 months. There was no significant difference in mean live weight and packed cell volume among groups (P > 0.05). Mean nematode fecal egg counts (FEC) did not significantly differ between the control and the remaining groups, except in one or two collections, when FEC was higher in the control group than in group 2 and group 3, respectively. The group that received A. robusta pellets needed less salvage anthelmintic treatments. Haemonchus contortus was the predominant species recovered from tracer lambs. The nematophagous fungi, D. flagrans and A. robusta, did not provide satisfactory results in the prophylaxis of parasitic gastroenteritis in sheep, under the conditions of the present study.

International audience; This paper investigates the relationship between the worm burden and the male proportion of adult Schistosoma mansoni. This relationship was tested using data from the literature on S. mansoni experimentally infected rodents and primates. The analyses were performed within each animal model (rat, mouse, hamster and monkey) and showed positive linear relationships between worm burden and male proportion. An analysis between the four animal models showed the same positive linear relationship. The role of the genetic diversity of male and female schistosomes in this relationship is discussed.
