Bovine tuberculosis (BTB) is a dangerous zoonosis which presents a serious problem for endangered species such as European bison ( Bison bonasus). Little is known about the influence of parasitic co-infections on the course and diagnosis of tuberculosis in animals. The best known co-infection in cattle is Fasciola hepatica and Mycobacterium bovis. The aim of this study was to review the most recent literature regarding tuberculosis and parasite co-infection in ungulates and relate the results to European bison. Our findings indicate that any comprehensive diagnosis of BTB should include parasitological monitoring, and the possible impact of such invasions on cellular response-based tuberculosis tests should be taken into account. The diagnosis of BTB is complex, as is its pathogenesis, and parasitic infestations can have a significant impact on both. Background: The productivity of livestock production has been greatly impacted by parasitic diseases. The infection prevalence of parasites in native and imported slaughtered animals has not received much attention, despite the fact that Saudi Arabia has data on animal parasites. This study aimed to determine the infection prevalence of parasites in slaughtered animals in relation to host origin and host age. Methods: This investigation was conducted at Al Makhwah area in southwestern Saudi Arabia for six months from September 2021 to February 2022. The examined animals were 1208 in total, including goats (504), sheep (440) and cattle (264). All the animals examined were slaughtered at official abattoirs. The collected parasites were preserved, processed and identified. Result: Out of 1208 examined, 72 were infected (5.96%). The parasite community consisted of six species, Dicrocoelium dendriticum, Fasciola hepatica, F. giagantica, Monezia expansa, Echinococcus granulosus (Hydatiid cyst) and Haemonchus contortus. Hydatidosis was a more prevalent parasitic disease. The infection prevalence of hydatidosis in the examined animals was high in cattle (4.85%) while were 3.76 and 4.77% in goats and sheep respectively. Significant differences in prevalence per host age in goats, sheep and cattle were found. The prevalence of parasites in imported animals was higher (16.67, 17.5 and 11.36% in cattle, sheep and goats respectively) as compared to that in native ones (5.42, 6.5 and 3.69% in cattle, sheep and goats respectively). Based on our findings, Hydatidosis was identified as the most prevalent parasitic disease. Imported animals had a higher parasitic infection compared to local animals. It is essential to take necessary measures to prevent the transmission of Hydatidosis and improper organ condemnation from abattoirs. Fasciola hepatica is a parasitic liver trematode that causes fasciolosis in humans and dairy animals. Traditional detection of infection is based on a microscopic examination with low sensitivity. Therefore, this study was carried out to develop an accurate and rapid method to detect F. hepatica in Alborz province. In this cross-sectional study, 386 samples were collected from livers of cattle and sheep in three slaughterhouses of Alborz. DNA was extracted, and nested-PCR was carried out based on the F. hepatica ITS-2 sequence. DNA amplification showed first and second PCR products with expected size of 336 and 208 bp respectively, as unique for F. hepatica. The results showed no cross-reaction with the negative control. Thirty-five liver samples were positive for this parasitic infection. Totally, the prevalence of F. hepatica in the slaughterhouses of Alborz, Iran was 9.07%. This is the first study of the molecular detection of F. hepatica using nested-PCR in Alborz. It is a sensitive and rapid method that will help evaluate the prevalence rate of F. hepatica infection. In this review article, we aim to provide an overview of fasciolosis in ruminants. Diagnosis through new coprological methods (such as Flukefinder®, FLOTAC®, and Mini-FLOTAC®) remains the most suitable approach for farms. Regarding treatment, there is a scarcity of available drugs, and resistance to them has prompted new approaches (including drug combinations, enhanced metabolism, or the use of natural compounds) to address this issue. Additionally, several researchers have developed vaccines to control the disease, but their efficacy varies, and none are currently sufficient for commercial use. Further studies are needed to better understand all aspects discussed in this manuscript, with the goal of improving diagnosis, treatment, and disease control. It is important to note that this manuscript does not delve into in-depth knowledge of the discussed aspects; rather, it provides an overview of the different methodologies related to these three aspects of parasitic disease. This article discusses a rare case of Capillaria hepatica infection in a 2-year-old boy who presented with fever, rash, hepatomegaly, and peripheral eosinophilia. The diagnosis was confirmed through fine needle aspiration cytology (FNAC) of a hepatic lesion, which revealed parasitic eggs. Fasciola hepatica is a parasite of the Trematoda class, found worldwide but primarily seen in cattle-raising countries. It usually settles in the biliary tract and causes nonspecific gastrointestinal symptoms such as abdominal pain, right upper quadrant tenderness, and fever in humans. Parasitological examination of stool or bile samples is required for definitive diagnosis. In our case, demonstrative histopathological features of Fasciola hepatica adult parasite seen in a 38-year-old female patient suffering from abdominal pain were evaluated. Effective management and control of parasitic infections on farms depends on their early detection. Traditional serological diagnostic methods for Fasciola hepatica infection in livestock are specific and sensitive, but currently the earliest detection of the parasite only occurs at approximately three weeks post-infection. At this timepoint, parasites have already entered the liver and caused the tissue damage and immunopathology that results in reduced body weight and loss in productivity. Here, we investigated whether the differential abundance of micro(mi)miRNAs in sera of F. hepatica-infected sheep has potential as a tool for the early diagnosis of infection. Using miRNA sequencing analysis, we discovered specific profiles of sheep miRNAs at both the pre-hepatic and hepatic infection phases in comparison to non-infected sheep. In addition, six F. hepatica-derived miRNAs were specifically identified in sera from infected sheep. Thus, a panel of differentially expressed miRNAs comprising four sheep (miR-3231-3p; miR133-5p; 3957-5p; 1197-3p) and two parasite miRNAs (miR-124-3p; miR-Novel-11-5p) were selected as potential biomarkers. The expression of these candidates in sera samples from longitudinal sheep infection studies collected between 7 days and 23 weeks was quantified using RT-qPCR and compared to samples from age-matched non-infected sheep. We identified oar-miR-133-5p and oar-miR-3957-5p as promising biomarkers of fasciolosis, detecting infection as early as 7 days. The differential expression of the other selected miRNAs was not sufficient to diagnose infection; however, our analysis found that the most abundant forms of fhe-miR-124-3p in sera were sequence variants (IsomiRs) of the canonical miRNA, highlighting the critical importance of primer design for accurate diagnostic RT-qPCR. Accordingly, this investigative study suggests that certain miRNAs are biomarkers of F. hepatica infection and validates miRNA-based diagnostics for the detection of fasciolosis in sheep. Purpose: Fascioliasis is a common parasitic disease in humans and herbivores which is caused by Fasciola hepatica and Fasciola gigantica and has a worldwide distribution. Serological tests such as the enzyme-linked immunosorbent assay (ELISA) technique play a prominent role in the fast diagnosis of the disease. However, there are diagnostic limitations, including cross-reactivity with other worms, which decline the specificity of the results. This study aimed to evaluate the structure of a recombinant multi-epitope antigen produced from linear and conformational B-cell epitopes of three parasitic proteins with sera of individuals with fasciolosis, healthy controls, and those with other diseases to gain accurate sensitivity and specificity. Methods: After designing the multi-epitope structure of cathepsin L1, FhTP16.5, and SAP-2 antigens and then synthesizing, cloning, and expressing, the extracted purified protein was evaluated by indirect ELISA to detect IgG antibodies against Fasciola hepatica parasite among the sera of 39 serum samples of Fasciola hepatica, 35 healthy individual samples, and 20 samples of other types of parasitic diseases. The synthesized multi-epitope produced from cathepsin L1, FhTP16.5, and SAP-2 antigens was evaluated using the indirect ELISA. Results: The analysis of the samples mentioned for IgG antibody diagnosis against Fasciola hepatica showed 97.43% (95% confidence interval, 94.23–100%) sensitivity and 100% (95% confidence interval, 97–100%) specificity. Conclusion: The recombinant B-cell multi-epitope with high antigenic potency may increase the specificity of epitopic peptides and ultimately help improve and develop indirect ELISA commercial kits for the diagnosis of fascioliasis in humans. Fascioliasis is a zoonotic parasitic infection caused by Fasciola species in humans and animals. Despite significant advances in vaccination and new therapeutic agents, little attention has been paid to validating methods for the diagnosis of fascioliasis in humans. Serological techniques are convenient assays that significantly improves the diagnosis of Fasciola infection. However, a more sensitive method is required. The aim of this study was to compare the Real-Time PCR technique with the indirect-ELISA for the detection of Fasciola hepatica in human. Using a panel of sera from patients infected with Fasciola hepatica (n = 51), other parasitic infections (n = 7), and uninfected controls (n = 12), we optimized an ELISA which employs an excretory–secretory antigens from F. hepatica for the detection of human fascioliasis. After DNA extraction from the samples, molecular analysis was done using Real-Time PCR technique based on the Fasciola ribosomal ITS1 sequence. Of 70 patient serum samples, 44 (62.86%) samples were identified as positive F. hepatica infection using ELISA and Real-Time PCR assays. There was no cross-reaction with other parasitic diseases such as toxoplasmosis, leishmaniasis, taeniasis, hydatidosis, trichinosis, toxocariasis, and strongyloidiasis. The significant difference between the agreement and similarity of the results of patients with indirect ELISA and Real-Time PCR was 94.4% and 99.2%, respectively (Cohen's kappa ≥ 0.7; P = 0.02). Based on the Kappa agreement findings, the significant agreement between the results of ELISA and Real-Time PCR indicates the accuracy and reliability of these tests in the diagnosis of F. hepatica in humans. Background: The association of fever, focal hepatic lesions and peripheral hyper-eosinophilia (FHLH) can be observed in both infectious and non-infectious conditions. Fascioliasis, capillariasis, toxocariasis, all causes of visceral larva migrans (VLM), represent most of the former, whilst lymphomas, eosinophilic leukemias and mastocytosis belong in the non-infectious conditions. Methods: We prospectively followed a young patient presenting with FHLH in the Tuscany region of Italy. Results: The patient was subject to serological and parasitological examination in an attempt to clarify the origin of the lesions. Serologies for both Fasciola hepatica and Toxocara spp. were positive, with the latter presenting a higher index. We opted for treatment with a prolonged course of albendazole due to the serological results and being toxocariasis more frequent in our setting. The patient was then subject to radiological follow-up. The patient responded to treatment with albendazole as shown by a decrease in eosinophils, seronegativization for Toxocara spp., clinical and radiological improvement. Toxocariasis was hence considered the most likely diagnosis. Conclusions: Parasitic infections cannot be disregarded in the presence of FHLH. Differential diagnosis between these parasitic infections can be challenging due to the presence of similar clinical presentations and serological cross-reactions, and follow-up of the patient is needed to ensure optimal treatment outcomes. Data from the genetic characterization of Fasciola population shed light on the spread of infections among animals which could help in the development of effective parasite control. The aim of the present work was to genetically characterize Fasciola adult worms isolated from sheep in Saudi Arabia by sequence analysis of ITS-1 region.Methods: A total of 12,653 slaughtered sheep in Jeddah city, Saudi Arabia were examined for the presence of Fasciola spp. adult worms. The ITS-1 region of all parasites was amplified and sequenced.Results: Overall, 12 variants DNA sequences were obtained. The variance of isolates ranged from 0.00771 to 0.34405. BLAST search showed that all obtained sequences were Fasciola hepatica and had > 99.3% similarity with F. hepatica isolates from Spain and USA (from different hosts other than sheep). Phylogenetic analysis showed that Fasciola isolates were closely related to isolates from different countries.Discussion: The current study showed that F. hepatica was the only spp. isolated from sheep in Jeddah. Fasciolosis is an endemic zoonotic parasitic disease with significant impacts on human health and both animal health and production. Early post‐infection impacts on the host remain unclear. The objective of this study was to determine the changes, if any, to levels of endotoxin in cattle plasma in response to early‐stage infection with Fasciola hepatica. Thirty‐six (36) commercial bred cattle were experimentally infected with approximately 400 viable metacercariae. Plasma lipopolysaccharide (endotoxin) levels were examined on 24 occasions from 0 h before infection to 336 h after infection using the Limulus Amoebocyte Lysate chromogenic end point assay and compared with that of six (6) uninfected control animals. Peak lipopolysaccharide levels in infected animals were reached at 52 h after infection and returned to pre‐infection levels at time 144 h after infection. Infected animals had significantly elevated lipopolysaccharide levels between 24 and 120 h after infection when compared to uninfected animals. The mean change in endotoxin units (EU)/mL over time after infection was statistically significant in infected animals. Elevations of lipopolysaccharide occurred in all infected animals suggesting a possible repeatable and titratable endotoxemia conducive to therapeutic agent model development. [ABSTRACT FROM AUTHOR]

Fascioliasis is a worldwide spread parasitic disease with a high impact on the economy and public health in many countries. Therefore, it is important to apply prevention, effective treatment, and education campaigns, to prevent the disease. This work aimed to determine the biochemical markers of liver damage caused by Fasciola hepatica during 21 days (D0 – D21) post albendazole (ABZ) therapy in the blood sera. This study included 24 cows naturally infected with Fasciola hepatica, randomly divided into a control group (n = 12) and a group treated per os with ABZ (7.5 mg per kg body weight) (n = 12). The activity of hepatic enzymes – aspartate aminotransferase (AST), alanine aminotransferase (ALT) and cholestatic enzymes – gamma-glutamyl transferase (GGT), and alkaline phosphatase (ALP), were measured in both groups. The activity of AST has significantly increased in the course of the experiment (D0 – D21) in both groups (P < 0.001). ALT activity during D0 – D21 was significantly decreased in both groups (P < 0.001). Activity of GGT was not significantly changed in the control group in the course of the experiment (P > 0.05). Contrarily, the activity of GGT in the treated group was significantly decreased (P < 0.01) during D0 – D21. The activity of GGT in the blood sera of the treated group was significantly lower in comparison with the control group in D7 and D21 (P < 0.01). This decreased GGT activity in the blood sera of the treated group may be caused by the effect of albendazole resulting in fasciola elimination and bile flow improvement. The results of ALP determination show no significant changes in ALP activity between groups. This can lead to the conclusion that the most useful marker of cholestasis caused by F. hepatica is the GGT activity and may help to evaluate the ABZ therapy effect in cattle. [ABSTRACT FROM AUTHOR]

Objectives. To determine seropositivity to anti-IgG antibodies against Echinococcus granulosus, Fasciola hepatica and Taenia solium cysticercus infection and to describe the characteristics of the infected patients in 13 regions of the Peruvian highlands between 2016 and 2019. Materials and methods. Cross-sectional, observational study, in which we analyzed 7811 epidemiological records of laboratory-based surveillance of parasitic zoonoses from 2016 to 2019. Diagnosis was established by detecting IgG type anti-E. granulosus, F. hepatica and T. solium cysticercus antibodies using native antigens by enzyme-linked immunosorbent assay (ELISA) and Immunoblot. We evaluated the difference in the frequency of the cases according to identified characteristics using Pearson's chi-square test and Fisher's exact test. Results. Seropositivity was 7.9% for fascioliasis, 4.9% for cystic echinococcosis, and 2.3% for T. solium cysticercus. These rates were higher in Cerro de Pasco for cystic echinococcosis (24.5%), in Ayacucho for T. solium cysticercus (4.5%) and in Puno for fascioliasis (40.6%). Regarding the sociodemographic characteristics, we found a statistically significant difference in the frequency of cases for all zoonoses according to age group, occupation, and region of residence. We also found a difference with the consumption of vegetables in emollients, and between clinical-epidemiological characteristics and having a family history of parasitic zoonoses. Conclusions. From the 7811 samples, we found that these parasitic zoonoses are distributed in 13 regions of the Peruvian highlands, and represent a major health problem, with frequencies that change according to different characteristics. Background: Trematode infections are of great importance as they affect the health of many species of mammals as cattle, sheep and goat. Fasciola hepatica represents the main trematode zoonosis and risks to human and cattle and paramphistomosis is one emerging parasitic diseases of ruminants widely distributed in the world. The economic expenses are incurred by the use of ineffective anthelmintics for trematode control. Besides to faecal egg count reduction test (FECRT) to determine the anthelmintic efficacy, can be used in vitro assays, by this the aim of the study was to determine the lethal doses (LD) with hatching egg test (EHT) of the main commercial anthelmintics used for the control of trematodes in cattle. Materials, Methods & Results: Liver and rumen were examined from cattle slaughtered in Tabasco, Chiapas and Campeche states from Mexico. F. hepatica eggs were recovered from gallbladder and rumen fluke eggs collected from adult parasites in saline solution. Subsequently, the hatching egg assays were performed placing 100 trematode eggs in distilled water in each one of 96 wells of polystyrene plates. After making the appropriate dilutions, several concentrations of commercial anthelmintics were evaluated, ranging from 0.04 to 80.63 mM for triclabendazole + 0.046 to 96.87 mM febendazole (TC+FBZ), from 0.04 to 91 mM for rafoxanide (RAFOX), from 0.02 to 43.74 mM for closantel (CLOS), from 0.036 to 76.18 mM for clorsulon + 0.002 to 3.31 mM ivermectin (CLORS+ IVM) and from 0.163 to 334.47 mM for nitroxynil (NITROX). A control group (water) was included in each plate. Lethal doses were obtained using the Probit procedure and analysis of the means with a one-way statistical design. Most drugs used against rumen fluke eggs presented a high LD50 and therefore were ineffective to cause egg mortality, such was the case of RAFOX that presented LD50 from 4,580 to 10,790 µg/mL (7 to 17 mM). CLOS presented the lowest LD50 (80 µg/mL or 0.12 mM) on rumen fluke eggs. TC+FBZ was found to be effective drug against the development of F. hepatica eggs in many samples. In the same way NITROX showed a low LD50 (37 to 63 µg/mL or 0.13 to 0.22 mM), but RAFOX presented a highest LD50 (1,450 µg/mL or 2.32 mM). Discussion: The present study focused on screen the ovicidal activity and determining in vitro lethal doses 50 of main commercial anthelmintics used to control F. hepatica and rumen fluke as rapid tests in a tropical region from Mexico. The FECRT is the main method to detect effectiveness of anthelmintic and other method is the coproantigen reduction test (CRT) by ELISA. Both tests require many infected animals depending the number of treatments and by this the egg hatch assay (EHA) represent a complementary diagnosis of effectiveness of anthelmintic products to compare between regions and even between farms, because few animals are required from the farm to collect trematode eggs, and it is possible to know the effectiveness against various anthelmintics at the same time. Efficacy studies on trematodes using egg hatching tests are scarce, although they have the advantage that they can be applied to both F. hepatica and rumen fluke. TC+FBZ was one of the most effective products in inhibiting the development of F. hepatica eggs. However, RAFOX showed low effectiveness against trematode eggs, with very high lethal doses. These results agree with a study that show low efficacy against the development of Paramphistomum cervi eggs and with the FECRT test reductions of 75% and 80.58% were obtained, in times from 7 to 84 days after treatment with RAFOX. NITROX and CLORS were drugs that had good efficacy on the development of F. hepatica eggs. A differential response between liver and rumen fluke was observed. Simple Summary: Two parasitic diseases affecting humans and ruminants have a particular life cycle, because freshwater molluscs intervene in their transmission by ensuring the development of parasite larval forms. As the climate is changing, researchers have begun to investigate the effects of global warming on these snails and the larval forms of parasites that they harbour. Several authors have already conducted analyses of these diseases, but no field studies have been carried out so far. Therefore, in the present study, snail counts were conducted on 39 farms with acidic soils between 1976 and 1997, in 2013–2014, and in 2020–2021. The results showed that the number of snail populations decreased over time and that many populations have fewer and fewer individuals. This decline has also been faster in recent years. The infection of snails by one of the parasites has decreased over time. Conversely, snails are increasingly infected with the other parasite. These changes are due to the generalized use of a drug used to treat one of the diseases in ruminants and probably also due to the heatwave episodes that occurred for several years. As these larval forms are again infecting animals after their departure from the snails, practitioners must take these changes into account when treating ruminants for these diseases. Field investigations were carried out during three periods (from 1976 to 1997, in 2013–2014, and in 2020–2021) on 39 cattle-raising farms on acidic soils to track changes in the populations of two Lymnaeidae (Galba truncatula and Omphiscola glabra) and their infection with Fasciola hepatica and/or Calicophoron daubneyi. Compared to the survey between 1976 and 1997 on these farms, there was a significant decrease in the number of the two lymnaeid populations and the size of the G. truncatula populations in both 2013–2014 and 2020–2021. This decline was significantly faster in the last nine years than it was before 2013. The area of habitats colonized by G. truncatula showed no significant variation over the years, while that of habitats with O. glabra significantly decreased in the period covered by the three surveys. The prevalence of F. hepatica infection in snails significantly decreased over the years, while C. daubneyi infection increased over time in both lymnaeid species. These changes are due to the use of triclabendazole to treat fasciolosis in ruminants since the 1990s, and are probably a consequence of the successive heatwaves that have occurred since 2018 in the region. Fasciolosis is a food and waterborne disease caused by Fasciola spp., representing a global health burden to various hosts, including humans and other animals. This study investigates the in vitro activity of tellurium- and selenium-containing diaryl dichalcogenides: diacetal ditelluride (LQ07), diacetal diselenide (LQ62), and diacetyl diselenide (LQ68) alone and in combination with ivermectin (IVM) against eggs of Fasciola hepatica. The eggs were exposed for 12 h with each organochalcogen (OC) (0.1 - 2 mmol l -1) and IVM (0.01 - 2 mmol l -1) following an incubation of 15 days, allowing embryonation. The inhibitory concentration of 50 % (IC 50) of each OC or IVM was tested with the IC 10 , IC 30 , and IC 50 of IVM or each OC, respectively. LQ07, LQ62, and LQ68, as well as IVM, demonstrated a concentration-dependent ovicidal activity. The peak ovicidal activity of 99.74 % was achieved when IVM was tested at 2.0 mmol l -1. LQ62 and LQ68 demonstrated greater ovicidal activity, having an IC 50 < 0.32 mmol l -1 being 6.25-fold more toxic than IVM alone. The percentage of dead eggs was significantly higher in the IVM group (early mortality), as Se-containing OCs led to the (miracidia) embryonation of the eggs with no hatching (late mortality). Blending Se-containing OCs and IVM showed an additive effect of up to 27 % against F. hepatica eggs. The present data contribute to the potential use of blending-based therapeutic strategies to combat F. hepatica infections in eradication programs worldwide. Fascioliasis is a parasitic infection caused by liver flukes. Although several cases have been reported in Korea, phylogenetic analysis of isolates is lacking. In this study, a 66-year-old woman with right upper quadrant (RUQ) abdominal pain was diagnosed as fascioliasis involving abdominal muscle by imaging study. She received praziquantel treatment, but symptoms were not improved. Lateral movement of the abscess lesion was followed. Trematode parasite was surgically removed from the patient's rectus abdominis muscle. The fluke was identified as Fasciola hepatica based on sequence analysis of 18S rDNA. To determine the phylogenetic position of this Fasciola strain (named Korean Fasciola 1; KF1), the cox1 gene (273 bp) was analyzed and compared with the genes of 17 F. hepatica strains isolated from cows, sheep, goats, and humans from various countries. Introduction: Livestock, including sheep, are infected with many types of parasitic diseases, and among these diseases is fascioliasis, or the so-called liver rot, which is caused by the Fasciola hepatica worm. This disease is widespread in many regions of the world, especially those with moderate climatic conditions. This disease harms the state's economy and livestock breeders due to the high rates of infection, which greatly affects animal production. Materials: Wax sections were made from F. hepatica, cut with a thickness of 7 microns, and stained with Hematoxylin and Eosin stain and Von Kossa technique to investigate the calcium sites in different organs of the worm. Results: There was a clear discrepancy in the density of calcium present in several internal organs of F. hepatica. Conclusion:We infer from this research that there is a clear variation in the density of calcium as well as the size and shape of the calcareous corpuscles in their different locations in the worm's organs. Fasciola hepatica is a trematode parasite that infects animals and humans causing fasciolosis, a worldwide-distributed disease responsible for important economic losses and health problems. This disease is of growing public health concern since parasite isolates resistant to the current treatment (triclabendazole) have increasingly been described. F. hepatica infects its vertebrate host after ingestion of the encysted parasite (metacercariae), which are found in the water or attached to plants. Upon ingestion, newly excysted juveniles of F. hepatica (FhNEJ) emerge in the intestinal lumen and cross the intestinal barrier, reach the peritoneum and migrate to the biliary ducts, where adult worms fully develop. Despite the efforts made to develop new therapeutic and preventive tools, to date, protection against F. hepatica obtained in different animal models is far from optimal. Early events of host-FhNEJ interactions are of paramount importance for the infection progress in fasciolosis, especially those occurring at the host-parasite interface. Nevertheless, studies of FhNEJ responses to the changing host environment encountered during migration across host tissues are still scarce. Here, we set-up an ex vivo model coupled with quantitative SWATH-MS proteomics to study early host-parasite interaction events in fasciolosis. After comparing tegument and somatic fractions from control parasites and FhNEJ that managed to cross a mouse intestinal section ex vivo, a set of parasite proteins whose expression was statistically different were found. These included upregulation of cathepsins L3 and L4, proteolytic inhibitor Fh serpin 2, and a number of molecules linked with nutrient uptake and metabolism, including histone H4, H2A and H2B, low density lipoprotein receptor, tetraspanin, fatty acid binding protein a and glutathione-S-transferase. Downregulated proteins in FhNEJ after gut passage were more numerous than the upregulated ones, and included the heath shock proteins HSP90 and alpha crystallin, amongst others. This study brings new insights into early host-parasite interactions in fasciolosis and sheds light on the proteomic changes in FhNEJ triggered upon excystment and intestinal wall crossing, which could serve to define new targets for the prevention and treatment of this widespread parasitic disease. Fasciola hepatica, commonly referred to as liver flukes, is a substantial zoonotic parasitic disease of humans and livestock globally. While infection is readily controlled by anthelmintics, namely triclabendazole, the heavy reliance on triclabendazole has resulted in drug resistance appearing worldwide. Due to drug resistance, it is imperative to adopt an integrated parasite management program to preserve the efficacy of currently available anthelmintics. A integrated liver fluke management plan would benefit from a simple rapid, field-deployable diagnostic for detection of F. hepatica in environment and the host. Therefore, a rapid DNA test using loop-mediated isothermal amplification was developed and optimised for the detection of F. hepatica from faecal and water samples to enable the detection of parasites both within the host and from the environment. The assay presented here is fast, with amplification in ≤20 min, and highly sensitive, with a detection limit of 5 × 10-4 ng/µL. The workflow presented here provides a time to result of ≤60 min without requiring a commercial kit for the extraction of DNA from faecal and water samples, and pending further validation from field-samples, could potentially be used to enable real-time decision making to mitigate parasite prevalence on a farming property and with no requirement for sample transportation. Fasciola hepatica is a neglected parasitic infection with significant human health and livestock industry impact. The Andean Altiplano harbors an estimated 50% of the Fasciola's world infection burden. There is scarce data regarding the spatial associations between different Fasciola hosts. In this project, we aimed to determine the geospatial relationships between Fasciola eggs passed in feces of different livestock species and the risk of infection among each household as a unit. We used data from a cross-sectional study evaluating children and livestock feces for Fasciola infection around households in three districts of Anta province, in the Cusco region of Peru. Each sample was geographically tagged and evaluated for fascioliasis using microscopy methods. A total of 2070 households were included, the median age was 9.1 years (6.7–11.8), 49.5% were female, and 7.2% of the households had at least one infected child. A total of 2420 livestock feces samples were evaluated. The infection rate in livestock samples was 30.9%. The highest infection rate was found in sheep with 40.8%, followed by cattle (33.8%), and swine (26.4%). The median distance between a household with an infected child to a positive animal sample was 44.6 meters (IQR 14.7–112.8) and the distance between a household with no infected children to a positive animal sample was 62.2 meters (IQR 18.3–158.6) (p = 0.025). The multivariable logistic regression adjusted by presence of poor sanitation, unsafe water consumption, altitude, and presence of multiple infected children per household demonstrated an association between household infection and any cattle feces at a 50 meters radius (Uninfected: OR 1.42 (95%CI 1.07–1.89), p = 0.017. Infected: OR 1.89 (95%CI 1.31–2.73), p = 0.001), positive cattle feces at a 100 meters radius (OR 1.35 (95% CI 1.08–1.69), p = 0.008), and negative cattle feces at a 200 meters radius (OR 1.08 (95% CI 1.01–1.15), p = 0.022). We identified potential hot and cold spots for fascioliasis in the Anta province. An association between environmental contamination with feces from different livestock species and infected children in rural households was found in our study. Local health authorities may apply this strategy to estimate the risk of infection in human populations and apply targeted interventions. Background: Hypereosinophilic syndrome is commonly found in various diseases such as allergic diseases, parasitic diseases, malignancies, etc. Fasciolosis may present with different clinical features, and it can make a difficult diagnosis of the disease. Laboratory manifestations of fascioliasis are eosinophilia. The purpose of this report was to introduce a child with hypereosinophilia that her diagnosis was fascioliasis. Case Presentation: The patient was a 3-year-old girl who was referred for prolonged fever (more than two weeks) and abdominal pain from another medical center, and she was hospitalized. In abdominal and pelvic ultrasound, splenomegaly was seen and in laboratory tests, she had hypereosinophilia. In the flow cytometry of bone marrow aspiration, the only finding was increased eosinophil level. Abdominal and thoracic a computerized tomography (CT) scans showed an increased size of para-aortic lymph nodes. On her examination, lymphadenopathy was present in the inguinal region. Therefore, a biopsy of an inguinal lymph node was performed to rule out lymphoma. Lymph node biopsy was negative for lymphoma. Fasciola serology was performed for the patient, and the stool exam was collected three times (for one day in between) to rule out parasitic disease, including Fasciola, etc. Due to weakly positive serology Fasciola hepatica, triclabendazole was started for the patient (it was given in two doses, 12 hours apart), despite the absence of Fasciola parasitic eggs in her stool. During hospitalization, the patient’s fever was stopped and by starting the use of mentioned drug, eosinophilia was reduced. The patient received a complete improvement in the follow-up. Conclusion: In patients with hypereosinophilia, parasitic diseases such as fascioliasis should be considered even if the fecal specimen is negative for Fasciola eggs.

Fascioliasis is an important neglected tropical disease that causes severe injury to the bile ducts and liver. Therefore, a rapid and accurate method for detection of Fasciola hepatica infection plays a vital role in early treatment. Currently, the diagnosis of fascioliasis is mainly conducted via serological tests using the excretory/secretory (E/S) products, which might cross-react with antigens from other helminth parasitic diseases. Hence, the development of serodiagnosis test using recombinant antigens may contribute to differentiate fascioliasis from other helminth infections. In the past 20 years, many attempts have been made to exert different F. hepatica recombinant antigens to obtain a well-established standard assay with high accuracy. In this review, we address recent studies that refer to the development of serodiagnosis tests for diagnosis of human fascioliasis based on the candidate recombinant antigens produced by different approaches. Meanwhile, in the present review, some main factors have been highlighted to improve the accuracy of diagnostic tests such as the effect of refolding methods to recover antigens' tertiary structure as well as applying a mixture of recombinant antigens with the highest sensitivity and specificity to improve the accuracy of diagnostic tests. Background: Fasciola hepatica causes economically important disease in livestock worldwide. The relevance of this parasitic infection extends beyond its direct consequences due to its immunoregulatory properties. Objectives: Given the importance of the T helper 1 (Th1) immune response in controlling infections with Mycobacterium avium subspecies paratuberculosis (MAP) in cattle, we aimed to establish the immunological consequences that co‐infection with F. hepatica might have on the course of Johne's disease (JD). Methods: This study compared the in vitro response of bovine immune cells to infection with MAP or exposure to MAP antigens following F. hepatica infection or stimulation with F. hepatica products. Results: We found a decreased proliferation of peripheral blood mononuclear cells (PBMCs) after infection with F. hepatica. This reduction was inversely correlated with fluke burden. Pre‐stimulation with F. hepatica molecules produced a significant reduction of ileocaecal lymph node leucocyte proliferation in response to MAP antigens. Additionally,F. hepatica products reduced expression of the CD14 receptor by macrophages and increased levels of apoptosis and bacterial (MAP) uptake. Conclusions: Overall, F. hepatica infection had little impact on the in vitro response of immune cells to MAP, whereas in vitro co‐stimulation with F. hepatica molecules had a measurable effect. Objective: Unusual clinical course Background: Fascioliasis is a zoonotic disease caused by Fasciola hepatica (F. hepatica). This infection is associated with a broad spectrum of clinical symptoms such as fever, eosinophilia, and gastrointestinal symptoms. Case Report: We report a case of F. hepatica abdominal mass in the peri-pancreatic region in a 58-year-old man, returned from Venezuela. The patient developed abdominal pain, nausea, anorexia, and weakness. Radiological investigations showed hepatomegaly, as well as mild intra-hepatic and extrahepatic ductal dilatation. The increase in eosinophilia, elevated total IgE titer, and anamnestic data suggested the hypothesis of parasitic infection. The diagnosis was established by high serological titer against F. hepatica. Conclusions: The development of abdominal mass, with jaundice and dilation of the biliary tract, does not always suggest the presence of heteroplasia. Hepatic fasciolosis has been implicated as one of the most important parasitic diseases affecting the sanity of cattle and sheep, reflecting in significant economic losses besides being an important anthropozoonotic disease. The eggs of Fasciola hepatica are eliminated with the feces of these hosts, and under the influence of extrinsic and intrinsic factors may remain in the environment for months until they find favorable conditions for embryogenesis. The objective of the present study was to evaluate the viability of Fasciola hepatica eggs exposed for 60 minutes at different concentrations of the enzymatic extract of Pochonia chlamydosporia (Pc-10). Subsequently, they were sedimented and placed on 24-well plates containing the extract of P. chlamydosporia (Pc-10) at concentrations of 500 µL (10%), 400 µL (8%), 300 µL (5%), 200 µL (2%), 100 µL (1%) and distilled water (control group). The experiment was carried out in triplicate, using a total of 900 eggs. After the experimental exposure to the enzyme extract of the fungus (Pc-10), 98% of the eggs exposed to the enzymatic solution demonstrated significant ultrastructural alterations in their respective teguments, when observed in scanning electron microscopy and transmission. The ultrastructure showed a collapse of the internal walls of the egg, interfering in the opening of the operculum. The use of the enzymatic extract of P. chlamydosporia (Pc- 10) compromised both the external tegument, breaking it and pasting it, as well as vital structures of the embryonic activity of the F. hepatica eggs. Common parasites of the European bison include gastro-intestinal and pulmonary nematodes, liver flukes (Fasciola hepatica), tapeworms, and protozoa of the genus Coccidia. This study compared the extensiveness and intensities of European bison parasitic invasions in three north-eastern Polish forests in different seasons and queried the role of parasitological monitoring in sanitary and hygienic control of feeding places. Faecal samples were collected in the Białowieża, Knyszyńska, and Borecka Forests between 2014 and 2016, as were some from an area neighbouring the Białowieża Forest outside the Natura 2000 protected area. Parasites were detected in individual samples with the flotation, decanting and Baermann methods. The eggs of Trichostrongylidae, Aonchotheca sp., Nematodirus sp., Strongyloides spp., Trichuris sp., Moniezia spp., and Fasciola hepatica; the larvae of Dictyocaulus viviparus; and the oocytes of Eimeria spp. were identified. Significant variation in invasion intensity and diversity was seen by origin and season. The relationships were assessed first by univariable tests and next multivariately, when origin and season emerged as the major risk factors for exposure to most of the parasites. The differences in the level of parasitic infection between the forests did not have implications for its sufficiency to cause clinical symptoms. However, the associations and risk factors found enable the necessary preventive measures to be taken to protect the E. bison from exposure or decrease the risks. Additionally, parasitological monitoring is appropriate as the method of sanitary and hygienic control of European bison winter feeding places. Fasciolosis remains a relatively frequent parasitic infection in cattle, despite the great implantation and efficacy of fasciolicides. In the present work, the prevalence of Fasciola hepatica has been observed for five years, analyzing 251 bullfighting cattle, evaluating the presence of macroscopic lesions in the liver. Moreover, the relationship with the temperature and average rainfall of the year, and also it describes the pathophysiological consequences during bullfighting. Methodology: it was carried out in Madrid (July-September 2013-2017). Results: n=251 animals in five years, 50.20±2.68 evaluated per year, which 14 had lesions, on average 2.80±1.64 per year. Likewise, a total of 4 compatible animals with lesions were reported during the five years, with an average of 0.80±0.84 animals per year. The average annual temperature was 15.78±0.48° C. The average annual rainfall was 570.20±102.21mm. A statistical correlation was observed between the carcasses with lesions and the average annual rainfall (P<0.01) and the annual mean temperature (P<0.05). The experiment revealed lower prevalence (5.61±2.82) than the one described by different authors, having also compatible climatological conditions with the growth of the parasite and all its intermediate hosts. Objectives: It is well known that raw vegetables have an important role in human nutrition. And considered as an agent for transmission of intestinal parasites Therefore, the present study is designed to detect the parasite contamination in Misan city . Method: A total of 45 fresh vegetables were randomly collected. All samples were examined according to standard methods for detection of protozoan cysts, oocysts, helminth eggs as well as larva. Results: Intestinal parasites were detected in 23/45 (51.1%). The highest contaminated vegetable were celery (66.7%) followed by cress (46.7%), and lettuces ( 40%). Giardia lamblia cysts were the most prevalent (43.5%) followed by Fasciola hepatica eggs (34.7%), Entamoeba spp. cysts (26.1%), Hymenolepis nana eggs (17.4%), Cryptosporidium spp. oocysts (13.1%), Toxoplasma gondii oocysts (8.7%), Enterobius vermicularis eggs (8.7%) and Emeria spp.oocysts (3.4%). Fasciolosis caused by Fasciola hepatica is a common parasitic disease of livestock especially sheep and cattle. In this study molecular characterization of β-tubulin isotype 3 gene in Fasciola hepatica isolates from cattle and sheep in Turkey was carried out. For this purpose a total of 80 adult Fasciola hepatica isolates were collected from 20 sheep and 20 cattle in Kayseri and Erzurum provinces. PCR-RFLP was performed on β-tubulin isotype 3 gene and MboII revealed two fragments of approximately 350 bp and 390 bp, whereas HphI enzyme yielded 210, 340 and 540 bp bands, HindII yielded 380 and 450 bp bands in all samples. A total of 80 isolates were tested by SSCP and all of them presented the same band profiles. Six samples (4 sheep and 2 cattle) were randomly selected and DNA sequence of a 935 bp coding fragment of β-tubulin isotype 3 was performed. Sheep samples were more polymorphic than the cattle. This study was carried out on 352 sheep 110 of which is below the age of 2, 171 is between 2-6 and 71 is above the age of 6 to determine the prevalence of fasciolosis caused by Fasciola hepatica in sheep in the centre of Van and its districs Özalp, Saray, Gevaş and Gürpınar between January and December 2009. All feces and blood samples were examined in the point of the eggs of parasite in feces and specific antibodies, respectively by Benedek sedimentation and antibody ELISA techniques. The origin of sheep, date of sample collection, age and sex of the sheep were recorded. In the research areas, the coprological prevalence and seroprevalence of fasciolasis in sheep were determined as 15.63% and 49.43%, respectively. In the infected sheep, Fasciola hepatica were found by benedek sendimentation. The highest prevalence seroprevalence of Fasciolosis was observed among the sheep above age of 6 (83.9%) and followed by 2-6 age group (46.19%) and <2 age group of sheep (36.32%). In the point of statistics, the difference between the age groups was significant (According to Chi-Square testing P<0.001). The seroprevalence of fasciolasis was %50.35 among females and %45.71 among males which was not accepted as a remarkable difference (P>0.001). As a result Fasciola hepatica was commonly detected in sheep in Van province. Epidemiology and control of this parasitic infections in the region for further studies are needed. [ABSTRACT FROM AUTHOR]
