Fucoidan is a fucose-enriched polysaccharide, obtained from brown algae, with demonstrated antioxidant properties. However, traditional extraction methods using water or chemical-based extraction methods have reduced yield and produced hazardous by-products. In this study, we isolated fucoidan at a high yield using enzyme-assisted extraction; the Celluclast enzyme assisted extract of Undaria pinnatifida sporophylls (FCUS). To examine the antioxidant properties of FCUS, oxidative stress was induced with 2,2'-azobis (2-methylpropionamidine) dihydrochloride (AAPH) in Vero cells and zebrafish model. FCUS was composed of 30.4% sulfate and 52.3% fucose. Pre-treatment of Vero cells with FCUS dose dependently inhibited AAPH-induced reactive oxygen species (ROS) production. Moreover, FCUS remarkably reduced cell death, ROS generation, and lipid peroxidation production in zebrafish larvae. Overall, these findings indicate that the sulfate-rich fucoidan of FCUS, obtained with an eco-friendly process, could be implemented as a beneficial antioxidant agent in the functional food industry.

Alcoholic liver disease (ALD) threatens human health, so it is imperative that we find ways to prevent or treat it. In recent years, the study of polysaccharides has shown that they have different kinds of bioactivities. Among them are many biological effects that have been attributed to polysaccharide precursors. D-Isofloridoside (DIF) is one of the polysaccharide precursors from the marine red alga Laurencia undulata . This study evaluated the effect of DIF on alcohol-induced oxidative stress in human hepatoma cells (HepG2). As a result, DIF attenuated alcohol-induced cytotoxicity, reduced the amount of intracellular reactive oxygen species (ROS), and effectively reduced alcohol-induced DNA damage in HepG2 cells. In addition, a western blot showed that, after DIF treatment, the expression levels of glutathione (GSH), superoxide dismutase (SOD), and B-cell lymphoma-2 (bcl-2) increased, while the expression levels of γ-glutamyl transferase (GGT), BCL2-associated X (bax), cleaved caspase-3, and mitogen-activated protein kinase (p38 and c-Jun N-terminal kinase ) signal transduction proteins reduced. This showed that DIF may protect cells by reducing the amount of intracellular ROS and inhibiting intracellular oxidative stress and apoptotic processes. Finally, molecular docking demonstrated that DIF can bind to SOD, GGT, B-cell lymphoma- 2 , and bax proteins. These results indicated that DIF can protect HepG2 cells from alcohol-induced oxidative stress damage, making it an effective potential ingredient in functional foods.

Gelatin was extracted from the skin of Japanese flounder ( Palatichtys olivaceus ) and was subjected to enzymatic hydrolysis. The peptic hydrolysate resulted in a potent antioxidative peptide Gly-Gly-Phe-Asp-Met-Gly (582 Da), which bears +12.61 kcal/mol hydrophobicity. The antioxidative potential of the peptide was characterized by analyzing the protective effect of the peptide on reactive oxygen species (ROS)-mediated intracellular macromolecule damage. It was found that the peptide is a potent scavenger of intracellular ROS, thereby protecting the radical-mediated damage of membrane lipids, proteins, and DNA. Moreover, the peptide is capable of upregulating the expression of inherent antioxidative enzymes, superoxide dismutase-1, glutathione, and catalase. Collectively, it can be concluded that Japanese flounder skin, a processing byproduct of filleting, can be effectively used to produce a bioactive peptide with potent antioxidant capacity.

Two new peptides derived from seaweed pipefish Syngnathus schlegeli, SPP-1(QLGNLGV) and SPP-2 (SVMPVVA) were assessed for their ability to prevent hydrogen peroxide induced oxidative stress in human dermal fibroblasts (HDFs). Both peptides showed a significant hydroxyl radical scavenging activity when tested by ESR technique. And also the peptides effectively suppressed the hydrogen peroxide induced ROS production and DNA damage in HDF cells. Furthermore the two peptides increase the protein expression levels of intracellular antioxidant enzymes SOD1, GSH and catalase in hydrogen peroxide stressed HDF cells. At the cellular signaling level, SPPs block the NF-κB activation which may lead to the reduction of oxidative stress mediated damage of HDF cells. Positive charges of chitooligomer (COS) enable COS to interact with negatively charged anionic groups on the cell surface resulting in an improvement in the biological activity of COS and its derivatives. In this study, 4-hydroxybenzaldehyde-COS (HB-COS) was synthesized and investigated for its abilities against H 2 O 2 -induced oxidative stress in microglia BV-2 cells. Under oxidative stress, HB-COS significantly attenuated reactive oxygen species (ROS) generation and DNA oxidation, and upregulated the protein levels of antioxidative enzymes. Abstract: In the present study, a peptide possessing antioxidant properties was isolated from Nile tilapia (Oreochromis niloticus) scale gelatin. Gelatin protein was hydrolyzed using alcalase, pronase E, trypsin and pepsin. Antioxidant efficacy of respective hydrolysates were evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical and superoxide radical anion scavenging activities. Moreover, protective effect on DNA damage caused by hydroxyl radicals generated was determined. Further, the level of reactive oxygen species (ROS) was determined using a fluorescence probe, 2′,7′-dichlorofluorescin diacetate (DCFH-DA), which could be converted to highly fluorescent dichlorofluorescein (DCF) with the presence of intracellular ROS on mouse macrophages, RAW 264.7 cells. Among hydrolysates, alcalase-derived hydrolysate exhibited the highest antioxidant activity compared to other enzymatic hydrolysates. Therefore, it was further analyzed and the sequence of an active peptide present in it was identified as Asp-Pro-Ala-Leu-Ala-Thr-Glu-Pro-Asp-Pro-Met-Pro-Phe (1382.57Da). This peptide showed no cytotoxic effect on mouse macrophages (RAW 264.7) and human lung fibroblasts (MRC-5). In addition, it scavenged hydroxyl, DPPH and superoxide radicals at the IC50 values of 7.56, 8.82 and 17.83μM, respectively. These results suggest that the peptide derived from Nile tilapia (O. niloticus) scale gelatin acts as a candidate against oxidative stress and could be used as a potential functional food ingredient. Advanced glycation end-products (AGEs) such as methylglyoxal (MGO) play a vital role in the pathogenesis of nephropathy, a diabetic complication. In the present study, we evaluated the anti-glycation and renal protective properties of Ishige okamurae extract (IOE) against AGE-induced oxidative stress. HPLC analysis confirmed that bioactive phlorotannins such as diphlorethohydroxycarmalol and ishophloroglucin A are predominantly present in IOE. IOE showed strong anti-glycation activities via inhibition of AGE formation, inhibition of AGE–protein cross-linking, and breaking of AGE–protein cross-links. In addition, in vitro studies using mesangial cells demonstrated that IOE effectively suppressed intracellular reactive oxygen species production, intracellular MGO accumulation, and apoptotic cell death by MGO-induced oxidative stress, in addition to regulating the expression of proteins involved in the receptor for AGEs and nuclear factor erythroid 2-related factor 2 (Nrf2)/antioxidant response elements (ARE) signaling pathways. It has been reported that enzymatic digestion of algae could improve the yield and enhance the biological activity compared to water and organic extraction. Our previous research indicated that Celluclast-assisted extract of Sargassum fulvellum (SF) possessed higher carbohydrate content and stronger antioxidant activity compared to water and other enzyme-assisted extracts. In the present study, we evaluated the antioxidant activities of polysaccharides from SF (SFPS) in vitro in Vero cells and in vivo in zebrafish. SFPS was obtained by Celluclast-assisted hydrolysis and ethanol precipitation. Results showed that SFPS contained 74.55 ± 1.26% sulfated polysaccharides and effectively scavenged 1, 1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radicals. SFPS significantly and dose-dependently scavenged intracellular reactive oxygen species (ROS) and improved cell viability. Further studies indicated that SFPS reduced apoptotic body formation through downregulation of proapoptotic protein (Bax and cleaved caspase-3) levels and upregulation of antiapoptotic protein (Bcl-xL and PARP) levels in 2,2-azobis(2-amidinopropane) hydrochloride (AAPH)-treated Vero cells. In addition, SFPS showed strong protective effect against AAPH-stimulated oxidative stress in vivo in zebrafish, as demonstrated by the improved survival rate, reduced heart rate, and decrease in ROS, cell death, and lipid peroxidation levels. These results suggest that SFPS possesses strong in vitro and in vivo antioxidant activity and can be a potential ingredient in the pharmaceutical and cosmeceutical industries. Abstract: Seafood processing by-product, Pacific cod (Gadus macrocephalus) skin, was utilised to purify an active peptide with ACE inhibitory and antioxidant activities. Gelatin was extracted from the skin and it was hydrolysed with gastrointestinal endopeptidases (pepsin, trypsin and α-chymotrypsin). Assay-guided purification of the hydrolysate resulted in an active peptide, Leu-Leu-Met-Leu-Asp-Asn-Asp-Leu-Pro-Pro (1301Da). The peptide showed potent non-competitive ACE inhibition (IC 50 =35.7μM) and effectively protects cellular macromolecules from reactive oxygen species (ROS) mediated damage. The peptide significantly reduced the oxidation levels of membrane lipids, proteins and DNA in RAW264.7 cells by effectively scavenging the intracellular ROS. Moreover, it was found that the peptide treatment upregulated the m-RNA expression of cellular antioxidative enzymes (superoxide dismutase, glutathione and catalase) and thereby enhanced the intracellular antioxidant mechanisms. These findings suggest that Pacific cod skin could be effectively bioconverted to produce a bioactive peptide, which could be used as a functional food ingredient to control ACE activity and oxidative stress. Abstract: The aim of this study was to investigate the antioxidant and anti-inflammatory activities of aminoethyl-chitooligosaccharides (AE-COS) in murine microglial cells (BV-2). At the high concentration of 100μg/ml, the inhibition of reactive oxygen species, DNA, protein and lipid oxidation were determined in BV-2 cells, at the rates of 77%, 80%, 85% and 85%, respectively. Furthermore, AE-COS was studied for its capabilities against lipopolysaccharide-induced inflammatory responses in BV-2 cells. It was found that AE-COS reduced the level of nitric oxide (NO) and prostaglandin E2 production by diminishing the expression of inducible NO synthase and cyclooxygenase-2 without significant cytotoxicity. Moreover, the inhibitory activities of AE-COS on generation of tumor necrosis factor-alpha and interleukin-1beta were performed. Collectively, these results indicate that AE-COS possess potential antioxidant and anti-inflammatory activities in brain microglia, and could be a useful therapeutic agent for the treatment of neuroinflammatory diseases.
