The successful use of lipid nanoparticles (LNPs) for clinical development of the COVID-19 mRNA vaccines marked a breakthrough in mRNA-LNP therapeutics. As one of the vital components of LNPs, poly(ethylene glycol)-lipid conjugates (PEG-lipids) influence particle biophysical properties and stability, as well as interactions within biological environments. Reports suggesting that anti-PEG antibodies can be detected quite commonly within the human population raise concerns that PEG content in commercial LNP products could further stimulate immune responses to PEG. The presence of anti-PEG antibodies has been linked to accelerated clearance of LNPs, potentially a source of variability in the biological response to mRNA-LNP products. This motivated us to explore potential PEG alternatives. Herein, we report physicochemical and biological properties of mRNA-LNPs assembled using poly(2-oxazoline) (POx)- and poly(2-oxazine) (POz)-based polymer-lipid conjugates. Notably, we investigated monoacyl lipids as alternatives to diacyl lipids. mRNA-LNPs produced using monoacyl POx/POz-lipids displayed comparable biophysical characteristics and cytocompatibility. Delivery of reporter mRNA resulted in similar transfection efficiencies, in both adherent and suspension cells, and in mice, compared to PEG-lipid equivalents. Our results suggest that monoacyl POx/POz-lipid-containing LNPs are promising candidates for the development of PEG-free LNP-based therapeutic products.

Poly(2-oxazoline)s (POx) have received substantial attention as poly(ethylene glycol) (PEG) alternatives in the biomedical field due to their biocompatibility, high functionality, and ease of synthesis. While POx have demonstrated strong potential as biomaterial constituents, the larger family of poly(cyclic imino ether)s (PCIE) to which POx belongs remains widely underexplored. One highly interesting sub-class of PCIE is poly(2,4-disubstituted-2-oxazoline)s (PdOx), which bear an additional substituent on the backbone of the polymers' repeating units. This allows fine-tuning of the hydrophilic/hydrophobic balance and renders the PdOx chiral when enantiopure 2-oxazoline monomers are used. Herein, we synthesize new water-soluble ( R -/ S -/ RS -) poly(oligo(2-ethyl-4-methyl-2-oxazoline) methacrylate) (P(OEtMeOxMA)) bottlebrushes and compare them to well-established PEtOx- and PEG-based bottlebrush controls in terms of their physical properties, hydrophilicity, and biological behavior. We reveal that the P(OEtMeOxMA) bottlebrushes show a lower critical solution temperature behavior at a physiologically relevant temperature (∼44 °C) and that the enantiopure ( R -/ S -) variants display a chiral secondary structure. Importantly, we demonstrate the biocompatibility of the chiral P(OEtMeOxMA) bottlebrushes through cellular association and mouse biodistribution studies and show that these systems display higher immune cell association and organ accumulation than the two control polymers. These novel materials possess properties that hold promise for applications in the field of nanomedicine and may be beneficial carriers for therapeutics that require enhanced cellular association and immune cell interaction.

In light of research reporting abnormal pharmacokinetic behavior for therapeutics and formulations containing poly(ethylene glycol) (PEG), a renewed emphasis has been placed on exploring alternative surrogate materials and tailoring specific materials to distinct nanomedicine applications. Poly(2-oxazolines) (POx) have shown great promise in this regard; however, a comparison of POx and PEG interactions with components of the immune system is needed to inform on their distinct suitability. Herein, the interaction of isolated immune cells following injection of hyperbranched polymers comprised of PEG or hydrophilic POx macromonomers was determined via flow cytometry. All materials showed similar association with all of the splenic immune cells analyzed. Interestingly, splenic CD68 hi and CD11b hi macrophages showed similar levels of polymer association, despite CD11b hi being a smaller population, suggesting CD68 is linked to increased recognition and phagocytosis of these nanomaterials. This is of interest given that CD68 is a scavenger receptor and directly facilitates the clearance of cellular debris and promotion of phagocytosis, as opposed to CD11b, which is associated with the mediating inflammation via the production of cytokines as well as complement-mediated uptake of foreign particles. In the liver, PEG and poly(2-methyl oxazoline) hyperbranched polymers showed no discernible differences in their cellular association, while hyperbranched poly(2-ethyl oxazoline) showed increased association with dendrocytes and CD68 hi macrophages, suggesting that this material exhibited a greater propensity to interact with components of the immune system. This work highlights the importance of how subtle changes in chemical structure can influence the immune response.

An understanding of thermally induced aggregation and consequent 19 F magnetic resonance imaging (MRI) performance is essential for improved design of thermoresponsive 19 F MRI contrast agents. Herein we describe a series of novel thermoresponsive perfluoropolyether (PFPE)-based comb-shaped poly(2-oxazoline)s (POxs) with different side-chain structures (2-methyl- (MeOx), 2-ethyl- (EtOx), and 2-( n-propyl)-2-oxazoline (nPrOx)). The comb polymers were prepared through reversible addition-fragmentation chain transfer (RAFT) polymerization of the respective oligo(2-oxazoline)acrylates using a perfluoropolyether macro-RAFT agent. The fluoropolyether chain end drives aggregation of the polymers, with small aggregates forming at 300 K for both poly(OMeOx 5 A) 9 -PFPE and poly(OEtOx 4 A) 9 -PFPE. The aggregates decrease in size and display increases in 19 F MRI intensity with temperature, and at 350 K the MeOx polymers are in the form of unimers in solution, similar to the oligoethylene glycol (OEG)-based PFPE polymer. Above the T CP of poly(OEtOx 4 A) 9 -PFPE, the polymer forms large aggregates, and the 19 F MR imaging performance is degraded. Likewise, poly(OnPrOx 4 A)-PFPE is above the LCST at all temperatures studied (300-350 K), and so weak imaging intensity is obtained. This report of novel thermoresponsive POx-based PFPE polymers highlights the importance of understanding self-association of polymers in solution and provides important insights for the development of "smart" thermoresponsive 19 F MRI contrast agents.

N-Acylated poly(aminoester) (NPAE) macromonomers were synthesised via spontaneous copolymerisation of 2-oxazolines and acrylic acid. Inherent quantitative introduction of α-vinyl end-groups was observed, which enabled the preparation of a series of well-defined NPAE comb polymers through redox-initiated RAFT polymerisation. Investigations of the thermal properties of the comb polymers revealed similarities to poly(2-oxazoline) based systems.

To date, cationic polymers with high transfection efficiencies (TE) often have a high cytotoxicity. By screening an 18-membered library of cationic 2-oxazoline-based polymers, a polymer with similar TE as linear poly(ethylene imine) but no detectable cytotoxicity at the investigated concentrations could be identified. The influence of the polymer side chain hydrophobicity and the type and content of amino groups on the pDNA condensation, the TE, the cytotoxicity, the cellular membrane interaction as well as the size, charge, and stability of the polyplexes was studied. Primary amines and an amine content of at least 40% were required for an efficient TE. We describe the synthesis of matrix supported hydrogel structures based on amine containing poly(2-oxazoline)s and their use to bind and release genetic material for potential applications in diagnostics or pathogen detection. Amine containing poly(2-oxazoline)s were synthesized by copolymerization of 2-ethyl-2-oxazoline with a monomer bearing a tert-butyl oxycarbonyl (Boc) protected amine group in the 2-position and subsequent deprotection. The statistical copolymers were used to generate hydrogels and matrix supported hydrogels by cross-linking of a certain fraction of the amine groups with epichlorhydrin. Supported structures were prepared by soaking porous polyethylene (PE) or polypropylene (PP) filter materials in a copolymer/epichlorhydrin solution, which was cross-linked upon heating. Scanning electron microscopy (SEM) of the composites revealed a bead like structure of the gel phase, which could be attributed to a lower critical solution temperature (LCST) behavior of the initial polymer prior to gelation. The dependency of the LCST behavior on the content of amine groups was investigated. Swelling values and the ratio of hydrogel per composite was determined using water sorption analysis. Subsequently, the ability of the systems to absorb and release labeled DNA was tested. Uptake and stimulated release, triggered by changes in pH, temperature, and heparin concentration, were investigated using fluorescence microscopy. Polymerase chain reaction (PCR) proved the successful recovery of the DNA, demonstrating the potential of the presented system for a broad range of molecular biological applications.

Poly(2-oxazoline)s (POxs) are a versatile class of biocompatible polymers, which have been investigated as poly(ethylene glycol) (PEG) alternatives. In recent years, POxs have drawn significant attention as coatings for antifouling applications. In this tutorial review different approaches to immobilize POxs on surfaces as well as properties and applications of POx coated surfaces will be presented.

Currently there is a lack of choice when selecting synthetic materials with the cell-instructive properties demanded by modern biomaterials. The purpose of this study was to investigate the attachment of cells onto hydrogels prepared from poly(2-oxazoline)s selectively functionalized with cell adhesion motifs. A water-soluble macromer based on the microwave-assisted cationic ring-opening polymerization of 2-methyl-2-oxazoline and 2-(dec-9-enyl)-2-oxazoline was functionalized with the peptide CRGDSG or controls using thiol-ene photochemistry followed by facile cross-linking in the presence of a dithiol cross-linker. The growth of human fibroblasts on the hydrogel surfaces was dictated by the structure and amount of incorporated peptide. Controls without any peptide showed resistance to cellular attachment. The benignity of the cross-linking conditions was demonstrated by the incorporation of fibroblasts within the hydrogels to produce three-dimensional cell-polymer constructs.

Detailed characterization of synthetic polymers often required multiple advanced separation technologies since the various molecular distributions present, e.g. polymer molar mass, chemical composition, functionality distributions, etc. are generally mutually dependent. The complexity of polymeric materials necessitates the use of a variety of analytical methods, either in conjunction or in integrated ("hyphenated") systems. Poly(2-oxazoline) homo- and copolymers with two different side groups rendering the systems hydrophobic, i.e. phenyl and dec-9-enyl substituents, synthesized by living cationic ring-opening polymerization, were investigated. The average chemical composition obtained by pyrolysis-gas chromatography-mass spectrometry (Py-GC-MS) corresponded well with the theoretical composition. The chemical-composition distribution was studied with gradient elution liquid chromatography (GELC) using water and tetrahydrofuran as mobile-phase components. Statistical copolymer samples - in contrast to their block copolymer analogues - revealed two well-separated peaks in GELC. By combining GELC with size-exclusion chromatography (SEC) it was confirmed that the GELC separation was not based on differences in the molar mass. A more likely explanation of the GELC results is the presence of an ionic fraction in the samples of statistical copolymers resulting from either chain-transfer reactions or termination by addition of water. Copoly(2-oxazoline)s, prepared by the cationic ring-opening polymerization of 2-(dec-9-enyl)-2-oxazoline with either 2-methyl-2-oxazoline or 2-ethyl-2-oxazoline, are crosslinked with small dithiol molecules under UV irradiation to form homogeneous networks. In situ monitoring of the crosslinking reaction by photo-rheology reveals the formation of soft gels within minutes. The degree of swelling in water is tunable based on the hydrophilicity of the starting macromers and the proportion of alkene side arms present. Furthermore, degradable hydrogels are prepared based on incorporation of a hydrolytically cleavable dithiol crosslinker. The bulk polymerization of 2-(dec-9-enyl)-2-oxazoline (DecEnOx), a fatty acid-based monomer for the cationic ring-opening polymerization, is reported. Furthermore, under optimal conditions, namely microwave heating at 100 °C, the bulk copolymerization with 2-ethyl-2-oxazoline yielded well-defined copolymers. Due to its pendant alkene groups DecEnOx-based polymers possess the potential to be modified in efficient thiol-ene reactions. The functionalization with thiols, e.g., dodecanethiol and 2,3,4,6-tetra-O-acetyl-1-thio-β-D-glycopyranose in "green" solvents is demonstrated., (Copyright © 2011 WILEY-VCH Verlag GmbH & Co. A new sugar-substituted 2-oxazoline monomer was prepared using the copper-catalyzed alkyne-azide cycloaddition (CuAAC) reaction. Its copolymerization with 2-ethyl-2-oxazoline as well as 2-(dec-9-enyl)-2-oxazoline, yielding well-defined copolymers with the possibility to tune the properties by thiol-ene "click" reactions, is described. Extensive solubility studies on the corresponding glycocopolymers demonstrated that the lower critical solution temperature behavior and pH-responsiveness of these copolymers can be adjusted in water and phosphate-buffered saline (PBS) depending on the choice of the thiol. By conjugation of 2,3,4,6-tetra-O-acetyl-1-thio-β-d-glucopyranose and subsequent deprotection of the sugar moieties, the hydrophilicity of the copolymer could be increased significantly, allowing a cloud-point tuning in the physiological range. Furthermore, the binding capability of the glycosylated copoly(2-oxazoline) to concanavalin A was investigated.

2-Oxazolines are well-known organic compounds which are included in a variety of complex biologically active structures and play a role as catalyst ligands and intermediates for functional compounds. In addition, 2-oxazolines serve as monomers for the synthesis of substituted poly(imine)s by cationic ring-opening polymerization. For the latter application, the feasibility of preparing new 2-substituted-2-oxazolines was investigated using an automated synthesizer. The reaction of various nitriles with 2-aminoethanol under Lewis acid catalysis was utilized for this purpose. Twenty-nine different substituted nitriles were selected out of more than 2000 commercial available nitriles to form the corresponding 2-oxazolines. At first, the reaction conditions were optimized for seven nitriles with regard to solvent and catalyst, including reproducibility tests in an automated parallel robot system. In the next step, the synthesis of all 29 2-oxazolines was screened in an automated parallel manner, whereby the reactions were monitored by GC-MS measurements providing novel insights in the scope of this synthesis route. These insights resulting from the high-throughput screening were validated by performing representative larger-scale syntheses of selected 2-oxazolines.
