Intrauterine embryonic development in the microphallid trematode Maritrema feliui is examined by means of transmission electron microscopy. Both fertilization and eggshell formation take place in the ootype. The eggshell is formed from a shell globule material derived from the vitelline cells combined with secretions of Mehlis' gland. The proximal uterus is packed with unembryonated eggs of the oligolecithal type, each composed of a fertilized oocyte and several vitelline cells, all surrounded by the shell. Intrauterine embryonic development of the egg is followed to the early stage of outer embryonic envelope formation, resulting in an embryo of ~20 blastomeres of three different types: macromeres, mesomeres and micromeres. The first equal cleavage division of the zygote produces two macromeres. The outer envelope is of cellular origin and formed by the cytoplasmic fusion of two macromeres, which become situated at opposite poles in the peripheral layer of the embryo just beneath the eggshell. Simultaneously, other blastomeres multiply and differentiate, whereas several micromeres exhibit clear signs of degeneration or apoptosis. These results show that the embryonic development of M. feliui starts in utero and represents an example of early stage ovoviviparity. A reduction in the number of blastomeres results from a continued degeneration of micromeres, which after autolysis and re-absorption, appear to represent an important source of nutritive reserves for the embryo. The embryonic development of this digenean is discussed in relation to its life cycle.

Ultrastructural aspects of the early embryonic development of the aspidogastrean Aspidogaster limacoides are described and their phylogenetic implications discussed. Whereas the proximal regions of the uterine lumen usually contain unembryonated eggs or eggs with early embryos, the posterior or distal regions of the uterus are filled with eggs containing a fully-developed cotylocidium. The eggs of A. limacoides can be classified as polylecithal due to the presence of numerous vitellocytes which accompany each fertilized oocyte or ovum during egg formation. The results of the study are described in details under six headings: (1) general characteristics of the intrauterine eggs; (2) eggshell and operculum formation; (3) unembryonated eggs; (4) zygote formation and early cleavage divisions; (5) embryonic envelope formation; and (6) early degeneration or apoptosis of some blastomeres. The late differentiation of the operculum, possible functions of GER-bodies, and the early degeneration of vitellocytes and some blastomeres in this species are compared, drawn and discussed with corresponding observations reported for other parasitic Platyhelminthes. The most important differences are apparent in the number of egg envelopes and their mode of formation in A. limacoides compared with previous reports for both digeneans and cestodes. The results of the present TEM study indicate that the three macromeres, resulting from two cleavage divisions, take part in the formation of a single embryonic outer envelope in A. limacoides, and that this takes place at a very early stage of embryogenesis. Their fusion results in the formation of a single continuous cytoplasmic layer surrounding the early embryo, which is composed of only a small number of undifferentiated blastomeres. The early separation of the macromeres may indicate an equal cleavage pattern. These results suggest that the systematic position of the Aspidogastrea among the Platyhelminthes still remains somewhat equivocal, and indicate the need for more studies on the embryonic development, larval morphogenesis and molecular phylogeny for the elucidation of the relationships between this enigmatic group and related taxa., (Copyright © 2011 Elsevier Ireland Ltd. In this study the ultrastructural aspects of egg envelope formation in the anoplocephalid cestode Mosgovoyia ctenoides are described. In the early stage of oncospheral morphogenesis, formation of three following primary embryonic envelopes takes place: (1) the capsule, (2) the outer envelope, and (3) the inner envelope. The capsule is formed from the vitellocyte material. Two macromeres contribute to the formation of the outer envelope and three mesomeres take part in the formation of the inner envelope. The three primary envelopes undergo further differentiation and transformation into the secondary envelopes, the so-called oncospheral or egg envelopes. In the advanced preoncospheral phase, the inner envelope undergoes differentiation into three sublayers: (1) a thick extra-embryophoral cytoplasmic layer; (2) an electron-dense embryophore, as a stiff pyriform apparatus; and (3) a thin intra-embryophoral cytoplasmic layer containing mesomere nuclei. The oncosphere is located in the extended cupule-like part of the pyriform apparatus. The two embryophoral horns elongate and fuse, thus forming a rigid cone. Four egg envelopes surround the mature infective oncosphere of M. ctenoides: (1) a thick capsule; (2) the outer envelope; (3) the inner envelope with a characteristic embryophore, in the form of the pyriform apparatus; and (4) the oncospheral membrane. The differentiation and ultrastructure of the egg envelopes of M. ctenoides are compared, in particular to those described in other anoplocephalids, and in general to the oncospheres of other cestode species.

The oncospheral envelopes of infective eggs in Nematotaenia dispar include the outer envelope with 2 sublayers, the inner envelope with a fibrillar embryophore and 2 cytoplasmic sublayers, and the oncospheral membrane. They differentiate from 3 primary embryonic envelopes, capsule, outer and inner envelope. The uterine envelopes are formed around the early embryos by processes of uterine epithelial cells, which surround the capsules. They degenerate rapidly in later stages; however, some structural components of the uterine envelopes were still visible in gravid proglottids as flattened perikarya with pyknotic, lobate nuclei, residual membranous structures and cellular debris situated usually between eggs. The following ultrastructural features of oncospheral envelopes differentiation appear to be characteristic for N. dispar: (1) lack of the outer capsule or shell in the fully mature eggs; (2) bi-layered structure of the outer envelope and tri-layered structure of the inner envelope; (3) absence of hook region membrane resulting probably from its early disintegration; (4) presence of small vesicles or "pits" incorporated into the inner envelope plasma membrane; (5) presence of densely packed microtubules in the external layer of the inner envelope; (6) changes in number of mitochondria and free ribosomes in the external and internal layers of inner envelope during egg maturation; and (7) probable "passage" of mitochondria and free ribosomes through the embryophoral pores in the developing eggs.

The origin, differentiation and functional ultrastructure of egg envelopes surrounding developing and mature miracidia of Schistosoma mansoni and that of developing and mature coracidia of Bothriocephalus clavibothrium have been examined by means of electron microscopy and cytochemistry. Results obtained on these two species were compared with data from our previous studies on the ultrastructure of egg envelopes in different cyclophyllidean (Hymenolepididae, Taeniidae, Anoplocephalidae, Davaineidae) and proteocephalidean cestodes. In the mature infective egg, the three main egg envelopes (the egg-shell, and the outer and inner envelope) that surround the larvae (miracidia of Digenea, coracidia of Pseudophyllidea, and hexacanths of Cyclophyllidea and Proteocephalidea), show evident similarities in their origin, functional ultrastructure and chemical composition. Conclusions of this comparison, concerning analogy and homology in egg envelopes of trematodes and cestodes, are drawn and discussed.

The vitellogenesis in Echeneibothrium beauchampi is studied by means of electron microscopy. In the follicles, the vitelline cells undergo three progressive changes characterized by cellular size, membrane system development and nature of stored inclusions. 1. Immature cells have a gonial morphology. 2. Maturing cells display a very well developed membrane system. Their cytoplasm is gradually filled with abundant lipid droplets. At the same time, a small number of homogeneous vesicles occur which contain protein material moderately electron dense. Glycogen (alpha and beta) appears at the end of the vitellogenesis. 3. Mature cells show a conspicuous regression of the membrane system. Cellular organelles, nucleus and protein vesicles are found in the peripheral cytoplasmic layer which still contains numerous free ribosomes. The amorphous remaining cytoplasm is filled with large amounts of aggregated lipid droplets which often surround glycogen areas. 4. The presence of numerous lipid droplets and glycogen in the vitelline cells of E. beauchampi shows evidence that this Tetraphyllidea is close to Pseudophyllidea, but the aspect of protein vesicles (shell-protein material) brings it near the Cyclophyllidea.
