Similar to its bacterial homolog GroEL, Hsp60 in oligomeric conformation is known to work as a folding machine, with the assistance of co-chaperonin Hsp10 and ATP. However, recent results have evidenced that Hsp60 can stabilize aggregation-prone molecules in the absence of Hsp10 and ATP by a different, “holding-like” mechanism. Here, we investigated the relationship between the oligomeric conformation of Hsp60 and its ability to inhibit fibrillization of the Ab40 peptide. The monomeric or tetradecameric form of the protein was isolated, and its effect on beta-amyloid aggregation was separately tested. The structural stability of the two forms of Hsp60 was also investigated using differential scanning calorimetry (DSC), light scattering, and circular dichroism. The results showed that the protein in monomeric form is less stable, but more effective against amyloid fibrillization. This greater functionality is attributed to the disordered nature of the domains involved in subunit contacts.

Partially degalactosylated xyloglucan from tamarind seeds (Deg-XG) is a very appealing biopolymer for the production of in situ gelling systems at physiological temperature. In this work, we observe that the morphology of hydrogels evolves towards high degrees of structural organization with time, yielding to dense stacks of thin membranes within 24 h of incubation at 37 °C. We also explore the possibility offered by gamma irradiation of controlling the time scale of this phenomenon, the final morphology and mechanical properties of the system. Structural and molecular modifications of Deg-XG with dose are investigated by FTIR, dynamic light scattering (DLS) and rotational viscosimetry. The impact on gelation ability and gel strength is studied by rheological analysis. The morphology evolution is investigated by SEM analysis, and absence of cytotoxicity verified by MTS assay and optical microscopy of neuroblastoma cells.

Study of macromolecular structure and dynamics of networks formed by pulsed electron-beam irradiation of poly(N-vinyl pyrrolidone) (PVP) aqueous solutions, at relatively low energy per pulse and across different concentration regimes, provides the basis for the understanding of a new generation of functional nanostructures. Networks are the result of the follow–up reactions initiated by a continuous series of electron pulse-generated hydroxyl radicals, which may have a different fate at the variance of polymer concentration. Different spectroscopic techniques, FT-IR, 13 C { 1 H} CP-MAS NMR and Raman, applied to characterize the formed networks, describe a profound modification of the chemical structure when network size is approaching the nanoscale. Static light scattering measurements provide further information on the average weight molecular weight modification of PVP when forming nanogel particles. From the simultaneous control of network size and modification of chemical functionality stems also an intrinsic fluorescence of these nanogels never observed before. Altogether the obtained radiation-sculptured nanogels exhibit interesting multifunctionality that, coupled with the already proven biocompatibility, can be exploited in the biomedical field.

(1) Background: A new family of nanosystems able to discern between normal and tumor cells and to release a therapeutic agent in controlled way were synthetized by e-beam irradiation. This technique permits to obtain biocompatible, sterile, carboxyl-functionalized polyvinylpyrrolidone (PVP-co-acrylic acid) nanogels (NGs); (2) Methods: Here, we performed a targeting strategy based on the recognition of over-expressed proteins on tumor cells, like the folate receptor. The selective targeting was demonstrated by co-culture studies and flow cytometry analysis, using folate conjugated NGs. Moreover, nanoparticles were conjugated to a chemotherapeutic drug or to a pro-apoptotic siRNA through a glutathione sensitive spacer, in order to obtain a controlled release mechanism, specific for cancer cells. The drug efficiency was tested on tumor and healthy cells by flow cytometric analysis, confocal and epifluorescence microscopy and cytotoxicity assay; the siRNA effect was investigated by RNAi experiment; (3) Results: The data obtained showed that the use of NGs permits a faster cargo release in cancer cells, in response to high cytosolic glutathione level, also improving their efficacy; (4) Conclusion: The possibility of releasing biological molecules in a controlled way and to recognize a specific tumor target allows overcoming the typical limits of the classic cancer therapy.

Several biopolymers are widely employed in food, pharmaceutical and biomedical sectors by virtue of their ability to generate supramolecular structures, typically physical hydrogels. In the case of high methoxyl pectins (HMP) the gel formation is promoted by the presence of cosolutes (sugars or polyols) and low pH. The present investigation regards the structuring kinetics of aqueous HMP solutions having different polymer concentration and equal sucrose content at 20°C. A sequence of consecutive frequency sweep was applied to each sample immediately after its preparation. The time evolution of the linear viscoelastic behavior is described by the sigmoidal profiles of both moduli at each applied frequency and more thoroughly defined through the change of the mechanical spectrum, i.e. the variation of the parameters of the generalized Maxwell model or the Friedrich-Braun model which are both suitable to provide a satisfactory data fitting.

Neuronal membrane damage is related to the early impairments appearing in Alzheimer's disease due to the interaction of the amyloid β-peptide (Aβ) with the phospholipid bilayer. In particular, the ganglioside GM1, present with cholesterol in lipid rafts, seems to be able to initiate Aβ aggregation on membrane. We studied the thermodynamic and structural effects of the presence of GM1 on the interaction between Aβ and liposomes, a good membrane model system. Isothermal Titration Calorimetry highlighted the importance of the presence of GM1 in recruiting monomeric Aβ toward the lipid bilayer. Light and Small Angle X-ray Scattering revealed a different pattern for GM1 containing liposomes, both before and after interaction with Aβ. The results suggest that the interaction with GM1 brings to insertion of Aβ in the bilayer, producing a structural perturbation down to the internal layers of the liposome, as demonstrated by the obtained electron density profiles.

The effects of K(+), Na(+) ions and their mixture on the conformational transition and macroscopic gel properties of kappa-Carrageenan system have been studied using different experimental techniques. The macroscopic gelation properties of kappa-Carrageenan were found to be dependent upon cosolute type. Indeed, a more ordered and strong gel was obtained in the presence of K(+) with respect to Na(+) ions. The gel properties obtained using mixtures of two cosolutes are shown to depend on the [K(+)]/[Na(+)] ratio.

This work concerns the aggregation properties of (Gly-Val-Gly-Val-Pro)(251) rec, a polypentapeptide reflecting a highly conserved repetitive unit of the bioelastomer, elastin. On raising the temperature of aqueous solutions above 25 degrees C, this polypeptide was already known to undergo concurrent conformational changes (hydrophobic folding), phase separation, and self-assembly with formation of aggregated three-stranded filaments composed of dynamic polypeptide helices, called beta-spirals. Aggregates obtained from the solution can be shaped into bands that acquire entropic elastic properties upon gamma-irradiation and can perform a variety of energy conversions. Previous studies have shown that aggregation is prompted by the (diverging) critical fluctuations of concentration occurring in the solution, in vicinity of its spinodal line. Here, we present combined circular dicroism (CD) and light scattering experiments, and independent fittings of experimental data to the theoretical spinodal and binodal (coexistence) lines. Results show the following logical and causal sequence of processes: (a) Smooth and progressive conformational changes promoted by concentration fluctuations occurring as temperature is raised "pull down" (in the temperature scale) the instability region of the solution. (b) This further promotes critical fluctuations. (c) The related locally high concentration prompts a further substantial conformational change ending in triple-helix formation and coacervation. (d) This intertwining of processes, covering different length scales (from that of individual peptides to the mesoscopic one of demixed regions), is related to the fact that solvent-induced interactions play a strong role over the entire scale span. These results concur with other recent ones in pointing out that process interactions over many length-scales probably reflect a frequent if not ubiquitous pattern in protein aggregation. This may be highly relevant to the desirable deep understanding of such phenomenon, whose interests cover many fields.

α,β-polyasparthydrazide (PAHy), a synthetic water-soluble biocompatible polymer, was chemically crosslinked with ethyleneglycol diglycidylether (EGDGE), in order to obtain water swellable microparticies. These were characterized by means of FT-IR spectrophotometry and by means of particle size distribution analysis. The mean pore size of the prepared gels as various crosslinking ratios and the fractal dimensions were determined by light scattering measurements. Swelling measurements gave evidence of the high affinity of PAHy-EGDGE microparticles towards aqueous media at different pH values. The physical state of the prepared networks was evaluated by means of X-rays diffractometry and thermal analysis. A peculiar effect of the degree of cross-linking on the glass transition temperature (T g ) value was shown. In vitro chemical and enzymatic stability studies suggested that the prepared samples do not undergo any degradation alter treatment at pH 1. 7.4 and 10 as well as after incubation with enzymes such as peptin and α-chymotrypsin.

Beta-amyloid (1-40) is one of the two most abundant species of amyloid-beta peptides present as fibrils in the extracellular senile plaques in the brain of Alzheimer's patients. Recently, the molecular aggregates constituting the early stage of fibril formation, i.e., oligomers and protofibrils, have been investigated as the main responsible for amyloid-beta cytotoxic effect. The molecular mechanism leading to neurodegeneration is still under debate, and it is common opinion that it may reside in the interaction between amyloid species and the neural membrane. In this investigation Atomic Force Microscopy and spectroscopy have been used to understand how structural (and mechanical) properties of POPC/POPS lipid bilayers, simulating the phospholipid composition and negative net charge of neuritic cell membranes, are influenced by the interaction with A beta(1-40), in different stages of the peptide aggregation. Substantial differences in the damage caused to the lipid bilayers have been observed, confirming the toxic effect exerted especially by A beta(1-40) prefibrillar oligomers. (c) 2013 Elsevier B.V. Gelled microemulsions are the subject of considerable scientific and commercial interest. Many efforts are currently devoted to improving their toxicological profile and functioning as biocompatible diffusion barrier for the controlled delivery of hydrophobic compounds. In the present investigation, a non-ionic polymeric surfactant was chosen to generate an oil-in-water microemulsion of a model fragrance in the presence of poly(N-vinyl-2-pyrrolidone) (PVP). The microemulsion was then subjected to either electron-beam or UV-irradiation to induce free-radical crosslinking of PVP at low temperature and in the absence of crosslinking agents, catalysts and initiators. Irradiation conditions with the two irradiation sources have been purposely selected to generate PVP hydrogels with similar appearances and mechanical spectra. Despite the macroscopic analogies, specific features are imparted to the two families of hydrogels by the two different irradiation methodologies. A description of the microstructure of both pure PVP and microemulsion-laden hydrogels is given starting from the dynamic light scattering (DLS) characterization of the liquid (unirradiated) formulations, followed by the study the hydrogels' dynamic mechanical, solubility and swelling properties, FTIR analysis of the water insoluble fractions, DLS measurements and cytotoxicity studies.

The use of synergistic interactions between polysaccharides can represent an important approach for the preparation of physical networks with a wide range of industrial applications, such as in the food and biomedical fields. Locust Bean Gum and Xanthan, two biocompatible polymers, are able to form gels when mixed together. A detailed study was carried out on three samples, prepared at different weight ratios and two temperatures, by means of light scattering and rheological techniques. The solid-like character of each sample was evaluated by measurements of the viscoelastic spectrum and intensity autocorrelation function. The contribution of fast and slow fluctuations to the static light scattered intensity was also evaluated at different angles. The power law exponents for the sol–gel transitions were estimated together with the fractal dimensions. The scattering results were found in good agreement with the mechanical ones in revealing a strong dependence of the structural properties of the different samples on their preparation conditions. Indeed, results show that a fine tuning of the properties of the mixed gels is possible through the change of the temperature preparation and/or the polymer weight ratio.

We present a variety of new experiments which concern the self-assembly of a polymeric network from homogeneous solutions of Agarose, a representative biostructural polysaccharide used for previous studies at our laboratories. They allow deriving a semi-quantitative phase diagram in the T, C plane. The diagram includes both the spinodal and gelation lines. Below a value of about 2% w/v, concentration is not sufficient for direct gelation; however, quenching of the sol from high temperatures to below the spinodal line initiates the spinodal demixing. The latter generates two sets of regions having respectively, higher- and lower-than-average polymer concentrations. In the higher-concentration regions the functional polymer-polymer interaction (that is, self-assembly) is favoured. In fact, as in the course of demixing the point representative of higher-concentration regions reaches the region below the gelation line, gelation is allowed and indeed observed to occur. This evidences the possibly more general role of spinodal demixing (extended to multi-component systems) as a pathway for specific, local and biofunctional enhancements of concentrations. At very low concentrations (e.g. 10−4 w/v) the kinetics of demixing is still observed to occur, but the set of higher-concentration regions is no longer percolative. In these conditions, gelation occurs only within each individual region, while the specimen remains free-running. This illustrates a novel aspect of the notion of non-gelling concentrations.

The potential utility of κ-carrageenan gels for preparing drug release devices is here shown. Structural properties of κ-carrageenan gels prepared with different salt composition and containing Ketoprofen sodium salt, as model drug, have been evaluated with static light scattering and rheological measurements. These properties have been correlated with release profiles in vitro at pH 5.5. Release properties from gelled matrices have been compared with those obtained by two commercial products containing the same drug. Results show that: i) in this system it is possible to easily control the gel texture by using different cationic concentration; ii) the kinetics of drug release by κ-carrageenan gels are dependent on the structural properties of matrices; iii) in the typical interval time used in classical local applications, all gel samples release the loaded drug almost completely, at difference with the commercial products. All these findings can provide useful suggestions for the realization of classical topical release systems.

Multicomponent essential oils Tagetes Minuta and Poleo as well as pure limonene were encapsulated in Tween doped-high methoxylated pectin gels. Optical microscopy reveals that the obtained gels containing limonene consisted in a highly heterogeneous oil-in-water emulsion stabilised by the gelled medium. The influence of limonene encapsulation in pectin gelation kinetics and the gel structural properties were followed by dynamic rheological measurements. An electronic nose device developed in our laboratory was used to follow the flavour release of the three systems in order to discriminate the samples according to the main components released to the headspace. PCA and Neural Network Analysis allowed us to discriminate Tagetes Minuta from Poleo due to the difference in their limonene content. It is remarkable that the fingerprints of encapsulated complex mixtures differ from those obtained for the non-encapsulated oils, showing a preferential release of some components. In the case of limonene, the effect of the encapsulated concentration on the detected odour was also studied.

We report data on static and dynamic light scattering of semidilute solutions of high methoxyl pectin in the presence of different amounts of sucrose, ranging from few % (w/w) up to a value just below the threshold required for gel formation. The apparent gyration radius of the pectin is found to slightly increase with sucrose concentration while the mean relaxation time displays a diverging behavior, strongly correlated with the increase of the solvent bulk viscosity. The dynamic behavior on approaching the condition required for gelation exhibits features typical of a glass transition, providing new insight into the role of sucrose in promoting the gelation of pectin.

Protein aggregation has been recognized to be a pathological indicator for several fatal diseases, such as Alzheimer's disease, transmissible spongiform encephalopathies, Creutzfeldt–Jacob disease, etc. Aggregation usually involves conformational changes of proteins that have acquired an intermediate β-structure-rich conformation and can occur even at low protein concentration. Recent work in our laboratory has shown that bovine serum albumin (BSA), even at low-concentration, exhibits self-association properties related to conformational changes, so providing a very convenient model system to study this class of problems. Here we report data (obtained by different experimental techniques) on a mixture of BSA in native and intermediate (β-structure-rich) form. Results show that the interaction between the two species is responsible for a decrease in the thermodynamic stability of the solution. This occurs without requiring noticeable conformational changes of the native protein. Results presented here can provide new insight on the ‘protein only’ hypothesis proposed for the formation of plaques involved in several neurodegenerative diseases.

Viscosities of solutions of Met−, Et−, and n‐PropOH were measured by quasielastic light scattering of polystyrene lattice spheres of 800 A diameter, in the low‐concentration, low‐temperature ranges where effects of alcohols on the structure and properties of liquid water are most pronounced. Raw data already indicate that alcohols promote the formation of clathrate‐like structures of H bonds which add to those occurring naturally in the pure solvent. Evaluation of the number of water molecules taking part in longer‐lived structures further indicates that this promotion is most effective in the infinite‐dilution limit. Available thermodynamic data agree with this evaluation. Mismatches among solute‐promoted clathrate‐like cages are evidenced. The known disruptive effects of alcohols on the anomalous properties of cold and supercooled water are thus understood in terms of limitations to the correlation length of density fluctuations, set by mismatches. A contribution of OH groups to cage promotion is also evidenced at least at low temperature. Constraints or hindrances to the motion of water molecules, irrespective of their nature, appear to be effective in favoring structures of stabler, longer‐lived H bonds, corresponding to water molecules of lower mobility. This offers a unified view of hydrophobic and hydrophilic interactions which agrees with earlier work and with more recent computer experiments. The possibility here evidenced for structures of H bonds promoted in solvent water of being conflictual or synergistic, adds to the microscopic understanding of solvent‐mediated interactions, e.g., of biosolutes.

We report on the effects that the presence of ethylene glycol or glycerol has on the oxygen affinity of hemoglobin. We attribute these effects to an altered equilibrium between T and R quaternary conformations of hemoglobin and separate them into bulk-electrostatic and non-bulk-electrostatic contributions to the standard free-energy difference between the R and T states.
