Photo-sono therapy (PST) is an innovative anti-vascular approach based on cavitation-induced spallation. Currently, passive cavitation detection (PCD) is the prevalent technique for cavitation monitoring during treatment. However, the limitations of PCD are the lack of spatial information of bubbles and the difficulty of integration with the PST system. To address this, we proposed a new, to the best of our knowledge, cavitation mapping method that integrates Doppler optical coherence tomography (OCT) with PST to visualize bubble dynamics in real time. The feasibility of the proposed system has been confirmed through experiments on vascular-mimicking phantoms and in vivo rabbit ear vessels, and the results are compared to high-speed camera observations and PCD data. The findings demonstrate that Doppler OCT effectively maps cavitation in real time and holds promise for guiding PST treatments and other cavitation-related clinical applications.

SignificanceThe human vocal fold (VF) oscillates in multiple vectors and consists of distinct layers with varying viscoelastic properties that contribute to the mucosal wave. Office-based and operative laryngeal endoscopy are limited to diagnostic evaluation of the VF epithelial surface only and are restricted to axial-plane characterization of the horizontal mucosal wave. As such, understanding of the biomechanics of human VF motion remains limited.AimOptical coherence tomography (OCT) is a micrometer-resolution, high-speed endoscopic imaging modality which acquires cross-sectional images of tissue. Our study aimed to leverage OCT technology and develop quantitative methods for analyzing the anatomy and kinematics of in vivo VF motion in the coronal plane.ApproachA custom handheld laryngeal stage was used to capture OCT images with 800 A-lines at 250 Hz. Automated image postprocessing and analytical methods were developed.ResultsNovel kinematic analysis of in vivo, long-range OCT imaging of the vibrating VF in awake human subjects is reported. Cross-sectional, coronal-plane panoramic videos of the larynx during phonation are presented with three-dimensional videokymographic and space-time velocity analysis of VF motion.ConclusionsLong-range OCT with automated computational methods allows for cross-sectional dynamic laryngeal imaging and has the potential to broaden our understanding of human VF biomechanics and sound production.

Multiple light scattering in biomedical tissue limits the penetration depth of optical imaging systems such as optical coherence tomography. To increase the imaging depth in scattering media, a computational method based on coherent reflection matrix measurement has been developed using low coherence interferometry. The complex reflection matrix is obtained via point-by-point scanning followed by a phase-shifting method; then singular value decomposition is used to retrieve the singly back-scattered light. However, the in vivo application of the current reported method is limited due to the slow acquisition speed of the matrix. In this Letter, a wide-field heterodyne-detection method is adopted to speed up the complex matrix measurement at a deep tissue layer. Compared to the phase-shifting method, the heterodyne-detection scheme retrieves depth-resolved complex amplitudes faster and is more stable without mechanical movement of the reference mirror. As a result, the matrix measurement speed is increased by more than one order of magnitude.

Subglottic stenosis (SGS) is a challenging disease to diagnose in neonates. Long-range optical coherence tomography (OCT) is an optical imaging modality that has been described to image the subglottis in intubated neonates. A major challenge associated with OCT imaging is the lack of an automated method for image analysis and micrometry of large volumes of data that are acquired with each airway scan (1 to 2 Gb). We developed a tissue segmentation algorithm that identifies, measures, and conducts image analysis on tissue layers within the mucosa and submucosa and compared these automated tissue measurements with manual tracings. We present a very simple method of constructing a polarization-sensitive optical coherence tomography (PS-OCT) system. An ordinary fiber-based swept-source OCT system was reconfigured for PS-OCT by adding a long section of polarization-maintaining fiber in the sample arm. Two polarization modes of a large group-delay difference formed spatially distinguished polarization channels. The depth-encoded information on the polarization states was retrieved by an amplitude-based analysis. We found that our method provides an economic scheme of PS-OCT. It demonstrates that an ordinary OCT system can be easily reconfigured for PS-OCT imaging if it has sufficient margins in the imaging range.

Endoscopic imaging technologies, such as endoscopic optical coherence tomography (OCT) and near-infrared fluorescence, have been used to investigate vascular and morphological changes as hallmarks of early cancer in the gastrointestinal tract. Here we developed a high-speed multimodality endoscopic OCT and fluorescence imaging system. Using this system, the architectural morphology and vasculature of the rectum wall were obtained simultaneously from a Sprague Dawley rat in vivo. This multimodality imaging strategy in a single imaging system permits the use of a single imaging probe, thereby improving prognosis by early detection and reducing costs.

We present a class of novel system characterization methods for spectral-domain optical coherence tomography (SD-OCT) particularly on getting optimized axial resolution performance. Our schemes uniquely utilize the autocorrelation interference response, also known as the self-interference product, which is generated by the optical fields from the imaging sample in automatic interferences. In our methods, an autocorrelation-inducing calibration sample was prepared which was made by sandwiching glass plates. OCT images of the calibration sample were captured by an SD-OCT system under testing. And the image data were processed to find various system characteristics based on the unique properties of autocorrelation interferograms, free of dispersion- and polarization-involved modulations. First, we could analyze the sampling characteristic of the SD-OCT's spectrometer for spectral calibration that enables accurate linear-k resampling of detected spectral fringes. Second, we could obtain the systematic polarization properties for quantifying their impact on the achieved axial resolutions. We found that our methods based on the autocorrelation response provide an easy way of self-characterization and self-validation that is useful in optimizing and maintaining axial resolution performances. It was found very attractive that a variety of system characteristics can be obtained in a single-shot measurement without any increased system complexity.

We present an automatic segmentation method for the delineation and quantitative thickness measurement of multiple layers in endoscopic airway optical coherence tomography (OCT) images. The boundaries of the mucosa and the sub-mucosa layers are accurately extracted using a graph-theory-based dynamic programming algorithm. The algorithm was tested with sheep airway OCT images. Quantitative thicknesses of the mucosal layers are obtained automatically for smoke inhalation injury experiments.

Biofilm formation has been linked to ventilator-associated pneumonia, which is a prevalent infection in hospital intensive care units. Currently, there is no rapid diagnostic tool to assess the degree of biofilm formation or cellular biofilm composition. Optical coherence tomography (OCT) is a minimally invasive, nonionizing imaging modality that can be used to provide high-resolution cross-sectional images. Biofilm deposited in critical care patients’ endotracheal tubes was analyzed in vitro. This study demonstrates that OCT could potentially be used as a diagnostic tool to analyze and assess the degree of biofilm formation and extent of airway obstruction caused by biofilm in endotracheal tubes.

We have developed laser-diode-based optical-resolution photoacoustic microscopy (LD-OR-PAM) of superficial microvasculature which has the desirable properties of being compact, low-cost, and label-free. A 300-mW visible pulsed laser diode was operated at a 405 ± 5 nm wavelength with a pulse energy as low as 52 nJ. By using a 3.6 MHz ultrasound transducer, the system was tested on carbon fibers with a lateral resolution of 0.95 µm and an SNR of 38 dB. The subcutaneous microvasculature on a mouse back was imaged without an exogenous contrast agent which demonstrates the potential of the proposed prototype for skin chromophores. Our eventual goal is to offer a practical and affordable multi-wavelength functional LD-OR-PAM instrument suitable for clinical applications.

Detection of an early stage of subglottic edema is vital for airway management and prevention of stenosis, a life-threatening condition in critically ill neonates. As an observer for the task of diagnosing edema in vivo, we investigated spatiotemporal correlation (STC) of full-range optical coherence tomography (OCT) images acquired in the rabbit airway with experimentally simulated edema. Operating the STC observer on OCT images generates STC coefficients as test statistics for the statistical decision task. Resulting from this, the receiver operating characteristic (ROC) curves for the diagnosis of airway edema with full-range OCT in-vivo images were extracted and areas under ROC curves were calculated. These statistically quantified results demonstrated the potential clinical feasibility of the STC method as a means to identify early airway edema.

Microvascular network of the retina plays an important role in diagnosis and monitoring of various retinal diseases. We propose a three-dimensional (3-D) segmentation method with intensity-based Doppler variance (IBDV) based on swept-source optical coherence tomography. The automatic 3-D segmentation method is used to obtain seven surfaces of intraretinal layers. The microvascular network of the retina, which is acquired by the IBDV method, can be divided into six layers. The microvascular network of the six individual layers is visualized, and the morphology and contrast images can be improved by using the segmentation method. This method has potential for earlier diagnosis and precise monitoring in retinal vascular diseases.

We report on a novel acoustic radiation force orthogonal excitation optical coherence elastography (ARFOE-OCE) technique for imaging shear wave and quantifying shear modulus under orthogonal acoustic radiation force (ARF) excitation using the optical coherence tomography (OCT) Doppler variance method. The ARF perpendicular to the OCT beam is produced by a remote ultrasonic transducer. A shear wave induced by ARF excitation propagates parallel to the OCT beam. The OCT Doppler variance method, which is sensitive to the transverse vibration, is used to measure the ARF-induced vibration. For analysis of the shear modulus, the Doppler variance method is utilized to visualize shear wave propagation instead of Doppler OCT method, and the propagation velocity of the shear wave is measured at different depths of one location with the M scan. In order to quantify shear modulus beyond the OCT imaging depth, we move ARF to a deeper layer at a known step and measure the time delay of the shear wave propagating to the same OCT imaging depth. We also quantitatively map the shear modulus of a cross-section in a tissue-equivalent phantom after employing the B scan.

In this Letter, we present a trimodality imaging system and an intravascular endoscopic probe for the detection of early-stage atherosclerotic plaques. The integrated system is able to acquire optical coherence tomography (OCT), fluorescence, and ultrasound images and simultaneously display them in real time. A trimodality intravascular endoscopic probe of 1.2 mm in diameter and 7 mm in length was fabricated based on a dual-modality optical probe that integrates OCT and fluorescence imaging functions and a miniature ultrasound transducer. The probe is capable of rotating at up to 600 rpm. Ex vivo images from rabbit aorta and human coronary arteries showed that this combined system is capable of providing high resolution, deep penetration depth and specific molecular fluorescence contrast simultaneously.

Optical coherence tomography (OCT) is an emerging noninvasive imaging technique, which is based on low-coherence interferometry. OCT images suffer from speckle noise, which reduces image contrast. A shrinkage filter based on wave atoms transform is proposed for speckle reduction in OCT images. Wave atoms transform is a new multiscale geometric analysis tool that offers sparser expansion and better representation for images containing oscillatory patterns and textures than other traditional transforms, such as wavelet and curvelet transforms. Cycle spinning-based technology is introduced to avoid visual artifacts, such as Gibbs-like phenomenon, and to develop a translation invariant wave atoms denoising scheme. The speckle suppression degree in the denoised images is controlled by an adjustable parameter that determines the threshold in the wave atoms domain. The experimental results show that the proposed method can effectively remove the speckle noise and improve the OCT image quality. The signal-to-noise ratio, contrast-to-noise ratio, average equivalent number of looks, and cross-correlation (XCOR) values are obtained, and the results are also compared with the wavelet and curvelet thresholding techniques.

We report on the feasibility of using long-range swept-source optical coherence tomography (OCT) to detect airway changes following smoke inhalation in a sheep model. The long-range OCT system (with axial imaging range of 25 mm) and probe are capable of rapidly obtaining a series of high-resolution full cross-sectional images and three-dimensional reconstructions covering 20-cm length of tracheal and bronchial airways with airway diameter up to 25 mm, regardless of the position of the probe within the airway lumen. Measurements of airway thickness were performed at baseline and postinjury to show mucosal thickness changes following smoke inhalation.

We developed an optical frequency domain imaging (OFDI) system based on an integrated thermo-optic swept laser to achieve three-dimensional surface imaging. The wavelength was swept by applying a heating signal to a thermo-optic polymeric waveguide. The sub-micrometer surface profile was converted from the three-dimensional phase information of the OFDI system on various samples used as resolution targets with a step height of 120 nm. © 2014 IOP Publishing Ltd.

We have developed a novel miniature integrated optical coherence tomography (OCT)-intravascular ultrasound (IVUS) probe, with a 1.5-mm-long rigid part and 0.9-mm outer diameter, for real-time intracoronary imaging of atherosclerotic plaques and guiding of interventional procedures. By placing the OCT ball lens and IVUS transducer back-to-back at the same axial position, this probe can provide automatically coregistered, coaxial OCT-IVUS imaging. To demonstrate its real-time capability, three-dimensional OCT-IVUS imaging of a pig's coronary artery displaying in polar coordinates, as well as images of three major types of atherosclerotic plaques in human cadaver coronary segments, were obtained using this probe and our upgraded system. Histology validation is also presented.

Using fingerprints as a method to identify an individual has been accepted in forensics since the nineteenth century, and the fingerprint has become one of the most widely used biometric characteristics. Most of the modern fingerprint recognition systems are based on the print pattern of the finger surface and are not robust against spoof attaching. We demonstrate a novel vital vascular fingerprint system using Doppler optical coherence tomography that provides highly sensitive and reliable personal identification. Because the system is based on blood flow, which only exists in a livng person, the technique is robust against spoof attaching.

We present an ultrathin fiber-optic endoscopy probe for optical coherence tomography (OCT), which is made of a series of fused optical fibers instead of the conventional scheme based on an objective lens. The large-core fiber with a core diameter of 20 μm was utilized for the probe, while a single-mode fiber of core diameter 8.2 μm mainly delivered the OCT light. Those fibers were spliced with a bridge fiber of an intermediate core size. The guided light was stepwise converted to a beam of a large mode-field diameter to be radiated with a larger depth of focus. We obtained a 125 μm thick all-fiber endoscopy probe with a side-viewing capability implemented by an angled fiber end. Successful OCT imaging was demonstrated with a swept-source OCT system and showed the practical applicability of our lens-free endoscopy probe.

A novel linearized interrogation method is presented for a Fourier domain mode-locked (FDML) fiber Bragg grating (FBG) sensor system. In a high speed regime over several tens of kHz modulations, a sinusoidal wave is available to scan the center wavelength of an FDML wavelength-swept laser, instead of a conventional triangular wave. However, sinusoidal wave modulation suffers from an exaggerated non-uniform wavelength-spacing response in demodulating the time-encoded parameter to the absolute wavelength. In this work, the calibration signal from a polarization-maintaining fiber Sagnac interferometer shares the FDML wavelength-swept laser for FBG sensors to convert the time-encoded FBG signal to the wavelength-encoded uniform-spacing signal.

Different types and sizes of micro- and nanoparticles have been synthesized and developed for numerous applications. It is crucial to characterize the particle sizes. Traditional dynamic light scattering, a predominant method used to characterize particle size, is unable to provide depth resolved information or imaging functions. Doppler variance optical coherence tomography (OCT) measures the spectral bandwidth of the Doppler frequency shift due to the Brownian motion of the particles utilizing the phase-resolved approach and can provide quantitative information about particle size. Spectral bandwidths of Doppler frequency shifts for various sized particles were quantified and were demonstrated to be inversely proportional to the diameter of the particles. The study demonstrates the phase-resolved Doppler variance spectral domain OCT technique has the potential to be used to investigate the properties of particles in highly scattering media.

We review the principle and some recent applications of Doppler optical coherence tomography (OCT). The advances of the phase-resolved Doppler OCT method are described. Functional OCT algorithms which are based on an extension of the phase-resolved scheme are also introduced. Recent applications of Doppler OCT for quantification of flow, imaging of microvasculature and vocal fold vibration, and optical coherence elastography are briefly discussed.

We have investigated the use of multimode fiber in optical coherence tomography (OCT) with a mode filter that selectively suppresses the power of the high-order modes (HOMs). A large-core fiber (LCF) that has a moderate number of guiding modes was found to be an attractive alternative to the conventional single-mode fiber for its large mode area and the consequentially wide Rayleigh range of the output beam if the HOMs of the LCF were efficiently filtered out by a mode filter installed in the middle. For this, a simple mode filtering scheme of a fiber-coil mode filter was developed in this study. The LCF was uniformly coiled by an optimal bend radius with a fiber winder, specially devised for making a low-loss mode filter. The feasibility of the mode-filtered LCF in OCT imaging was tested with a common-path OCT system. It has been successfully demonstrated that our mode-filtered LCF can provide a useful imaging or sensing probe without an objective lens that greatly simplifies the structure of the probing optics.

Many diseases involve changes in the biomechanical properties of tissue, and there is a close correlation between tissue elasticity and pathology. We report on the development of a phase-resolved acoustic radiation force optical coherence elastography method (ARF-OCE) to evaluate the elastic properties of tissue. This method utilizes chirped acoustic radiation force to produce excitation along the sample's axial direction, and it uses phase-resolved optical coherence tomography (OCT) to measure the vibration of the sample. Under 500-Hz square wave modulated ARF signal excitation, phase change maps of tissue mimicking phantoms are generated by the ARF-OCE method, and the resulting Young's modulus ratio is correlated with a standard compression test. The results verify that this technique could efficiently measure sample elastic properties accurately and quantitatively. Furthermore, a three-dimensional ARF-OCE image of the human atherosclerotic coronary artery is obtained. The result indicates that our dynamic phase-resolved ARF-OCE method can delineate tissues with different mechanical properties.

A polarization-maintaining buffered Fourier domain mode-locked (FDML) swept source with a center wavelength of 1300 nm is demonstrated. The scanning rate of the buffered FDML swept source is doubled without sacrificing the output power of the swept source by combining two orthogonally polarized outputs with a polarization beam combiner. The stability of the swept source is improved because the polarization state of the laser beam inside the laser cavity is maintained without the use of any polarization controllers. The swept source is capable of an edge-to-edge tuning range of more than 150 nm and a FWHM range of 95 nm at a 102 kHz sweeping rate and with an average power of 12 mW. A swept source optical coherence tomography (SSOCT) system is developed utilizing this buffered FDML swept source. The axial resolution of the SSOCT system is measured to be 9.4 µm in air. The sensitivity of the SSOCT system is 107.5 dB at a depth of 0.25 mm with a 6 dB roll-off at a depth of 2.25 mm.

There have been many advances in the field of diagnostic and therapeutic pulmonary medicine in the past several years, with major progress in the field of imaging. Optical coherence tomography (OCT) is a high-resolution (micron level) imaging modality currently being advanced with the potential to image airway wall structures in real time and at higher resolution than previously possible. OCT has the potential to increase the sensitivity and specificity of biopsies, create 3D images of the airway to guide diagnostics, and may have a future role in diverse areas such as the evaluation and treatment of patients with obstructive sleep apnea, tracheal stenosis, airway remodeling and inhalation injury. OCT has recently been investigated to monitor airway compliance in chronic obstructive pulmonary disease and asthma patients as well as differentiate causes of pulmonary hypertension. In future clinical and research applications, OCT will likely be combined with other endoscopic based modalities such as ultrasound, spectroscopy, confocal, and/or photoacoustic tomography to determine functional and biomolecular properties. This article discusses the current uses of OCT, its potential applications, as it relates to specific pulmonary diseases, and the future directions for OCT.

We report on the synergy of an integrated ultrasound (US) and photoacoustic (PA) probe system for intravascular imaging. The combined dual-modality probe is based on a 39 MHz ring-shaped US transducer which detects both US echoes and laser-generated PA signals. By combining optical fiber, US transducer, and micromirror, we can obtain intravascular cross-sectional B-scan images by internal illumination of the sample. The performance of the probe is evaluated in a phantom study. Moreover, the coaxially designed probe also provides co-registered US and PA images of a normal rabbit aorta, which demonstrates the imaging ability of the dual-functional system, implying future clinical applications.

Optical coherence tomography (OCT) is a nondestructive imaging technique offering high temporal and spatial resolution, which makes it a natural choice for assessing tissue mechanical properties. We have developed methods to mechanically analyze the compliance of the rabbit trachea in vivo using tissue deformations induced by tidal breathing, offering a unique tool to assess the behavior of the airways during their normal function. Four-hundred images were acquired during tidal breathing with a custom-built endoscopic OCT system. The surface of the tissue was extracted from a set of these images via image processing algorithms, filtered with a bandpass filter set at respiration frequency to remove cardiac and probe motion, and compared to ventilatory pressure to calculate wall compliance. These algorithms were tested on elastic phantoms to establish reliability and reproducibility. The mean tracheal wall compliance (in five animals) was 1.3±0.3×10(-5) (mm Pa)(-1). Unlike previous work evaluating airway mechanics, this new method is applicable in vivo, noncontact, and loads the trachea in a physiological manner. The technique may have applications in assessing airway mechanics in diseases such as asthma that are characterized by significant airway remodeling.

We present a large-core fiber (LCF) with a reduced nonlinear property for a single-mode beam delivery of intense ultrashort pulses. A tapered-fiber mode filter was fabricated in an LCF with the core diameter decreased from 20 μm to 6 μm at the tapered waist region surrounded by index-matched liquid. By the tapered geometry, the high-order mode was rejected so that our mode-filtered LCF acted as a single-mode fiber despite the multimode property of the original LCF. Early detection of cancer remains the best way to ensure patient survival and quality of life. Squamous cell carcinoma is usually preceded by dysplasia presenting as white, red, or mixed red and white epithelial lesions on the oral mucosa (leukoplakia, erythroplakia). Dysplastic lesions in the form of erythroplakia can carry a risk for malignant conversion of 90%. A noninvasive diagnostic modality would enable monitoring of these lesions at regular intervals and detection of treatment needs at a very early, relatively harmless stage. The specific aim of this work was to test a multimodality approach [three-dimensional optical coherence tomography (OCT) and polarimetry] to noninvasive diagnosis of oral premalignancy and malignancy using the hamster cheek pouch model (nine hamsters). The results were compared to tissue histopathology. During carcinogenesis, epithelial down grow, eventual loss of basement membrane integrity, and subepithelial invasion were clearly visible with OCT. Polarimetry techniques identified a four to five times increased retardance in sites with squamous cell carcinoma, and two to three times greater retardance in dysplastic sites than in normal tissues. These techniques were particularly useful for mapping areas of field cancerization with multiple lesions, as well as lesion margins.

We present a calibration protocol to obtain the alignment factors of a custom-made spectrometer and the nonlinear fitting function between the measured CCD pixel domain and the wavelength domain to apply to the spectral-domain optical coherence tomography (SD-OCT) using fiber Bragg gratings. We have used five gratings with different center wavelengths covering the broadband source spectral range. All have a narrow spectral bandwidth (0.05 nm) and the same reflectivity (92%) to calibrate and align the custom-made spectrometer. The implemented SD-OCT system following the proposed protocol showed the alignment factors as 44.37 deg incident angle, 53.11 deg diffraction angle, and 70.0-mm focal length. The spectral resolution of 0.187 nm was recalculated from the alignment factors.

A fiber-based multimodal imaging system that combines multiphoton microscopy (MPM) with Fourier domain optical coherence microscopy (OCM) is reported. The system uses a fiber-based femtosecond laser, a fiber coupler, and a double-clad fiber (DCF) device. The fiber laser has a central wavelength of 1.04 μm and bandwidth of 29 nm. Longer excitation wavelength is used to increase penetration depth and increase the excitation efficiency for dyes, such as red fluorescent dyes. A single mode fiber coupler is used to replace the free-space beam splitter and one arm of the coupler is fused with a double-clad fiber device. The MPM and OCM share the same excitation light path in the core of a double-clad fiber, while the OCM and MPM signals were collected by the core and clad of the double-clad fiber, respectively. The performance of the introduced double-clad device is analyzed. The device can confine all the excitation light in the core and has a collection efficiency of 20% for the MPM signal. The efficiency can be further increased by fusing more multimode fibers with the DCF. Simultaneous optical coherence microscopic imaging, second harmonic generation imaging, and two-photon excitation fluorescence imaging are demonstrated in biological samples.

We report the use of spectral Doppler optical coherence tomography imaging (SDOCTI) for quantitative evaluation of dynamic blood circulation before and after a localized ischemic stroke in a mouse model. Rose Bengal photodynamic therapy (PDT) is used as a noninvasive means for inducing localized ischemia in cortical microvasculature of the mouse. Fast, repeated Doppler optical coherence tomography scans across vessels of interest are performed to record flow dynamic information with high temporal resolution. Doppler-angle-independent flow indices are used to quantify vascular conditions before and after the induced ischemia by the photocoagulation of PDT. The higher (or lower) flow resistive indices are associated with higher (or lower) resistance states that are confirmed by laser speckle flow index maps (of laser speckle imaging). Our in vivo experiments shows that SDOCTI can provide complementary quantified flow information that is an alternative to blood volume measurement, and can be used as a means for cortical microvasculature imaging well suited for small animal studies.

We report a photonic crystal fiber (PCF) coupler having an ultrawide spectral bandwidth keeping single mode operation. The use of the PCF coupler in a fiber-based optical coherence tomography (OCT) system enables us to handle the wide spectral bands of various light sources, including superluminescent diodes (SLDs) at 1300 nm and 820 nm, Ti:sapphire lasers, and white-light sources. The multiband imaging performances of the PCF-based OCT system are demonstrated by obtaining dental images at 1300 nm and 820 nm with the same setup. In addition, we show that the PCF coupler could cover the spectrum over a one octave span and guide both the fundamental wave (1030 nm) and the second harmonic wave (515 nm) simultaneously.

We report on a dual-modality optical coherence tomography (OCT) ultrasound (US) system for intravascular imaging. To the best of our knowledge, we have developed the first integrated OCT-US probe that combines OCT optical components with an US transducer. The OCT optical components mainly consist of a single-mode fiber, a gradient index lens for light-beam focusing, and a right-angled prism for reflecting light into biological tissue. A 40-MHz piezoelectric transducer (PZT-5H) side-viewing US transducer was fabricated to obtain the US image. These components were integrated into a single probe, enabling both OCT and US imaging at the same time. In vitro OCT and ultrasound images of a rabbit aorta were obtained using this dual-modality imaging system. This study demonstrates the feasibility of an OCT-US system for intravascular imaging, which is expected to have a prominent impact on early detection and characterization of atherosclerosis.

We demonstrate the use of Doppler variance (standard deviation) imaging for 3-D in vivo angiography in the human eye. In addition to the regular optical Doppler tomography velocity and structural images, we use the variance of blood flow velocity to map the retina and choroid vessels. Variance imaging is subject to bulk motion artifacts as in phase-resolved Doppler imaging, and a histogram-based method is proposed for bulk-motion correction in variance imaging. Experiments were performed to demonstrate the effectiveness of the proposed method for 3-D vasculature imaging of human retina and choroid.

Hamster cheek pouches were exposed to 2-chloroethyl ethyl sulfide [CEES, half-mustard gas (HMG)] at a concentration of 0.4, 2.0, or 5.0 mg/ml for 1 or 5 min. Twenty-four hours post-HMG exposure, tissue damage was assessed by both stereomicrography and optical coherence tomography (OCT). Damage that was not visible on gross visual examination was apparent in the OCT images. Tissue changes were found to be dependent on both HMG concentration and exposure time. The submucosal and muscle layers of the cheek pouch tissue showed the greatest amount of structural alteration. Routine light microscope histology was performed to confirm the OCT observations.

With the increase of the A-line speed of optical coherence tomography (OCT) systems, real-time processing of acquired data has become a bottleneck. The shared-memory parallel computing technique is used to process OCT data in real time. The real-time processing power of a quad-core personal computer (PC) is analyzed. It is shown that the quad-core PC could provide real-time OCT data processing ability of more than 80 K A-lines per second. A real-time, fiber-based, swept source polarization-sensitive OCT system with 20 K A-line speed is demonstrated with this technique. The real-time 2D and 3D polarization-sensitive imaging of chicken muscle and pig tendon is also demonstrated.

Phase microscopy for high-dynamic-range quantitative phase-contrast imaging of a transparent phase object was demonstrated. Using a common path Fourier domain optical coherence tomography system, this technique is capable of displacement measurement with a sensitivity of 34 pm. The limitation of 2pi ambiguity restriction was overcome by the use of a phase retrieval approach performed in spectral domain. Two-dimensional quantitative phase imaging of human neonatal dermal keratinocyte cells was demonstrated to evaluate the performance of the system for cell imaging.

We report on the feasibility of rapid, high-resolution, 3-D swept-source optical coherence tomography (SSOCT) to detect early airway injury changes following smoke inhalation exposure in a rabbit model. The SSOCT system obtains 3-D helical scanning using a microelectromechanical system motor-based endoscope. Real-time 2-D data processing and image display at the speed of 20 frames/s are achieved by adopting the technique of parallel computing. Longitudinal images are reconstructed via an image processing algorithm to remove motion artifacts caused by ventilation and pulse. Quantitative analyses of tracheal airway thickness as well as thickness distribution along tracheal circumference are also performed based on the comprehensive 3-D volumetric data.

An abnormal level of red blood cell (RBC) aggregation is a clinical condition associated with many pathologies. Our purpose is to investigate the feasibility of spectral domain phase-resolved optical Doppler tomography incorporated with Doppler variance imaging to characterize RBC aggregation in flowing blood. Variance/standard deviation (SD) of the Doppler frequency spectrum of porcine blood flowing through a glass tube (diameter 300 microm) at 4.7 mms is measured. For nonaggregating RBCs with hematocrits of 20, 40, and 60%, the mean centerline SD values were 34.2+/-1.2, 47.8+/-1.0, and 47.2+/-0.5, respectively. Optical coherence tomography (OCT) is an evolving noninvasive imaging modality that has been used to image the human larynx during surgical endoscopy. The design of a long gradient index (GRIN) lens-based probe capable of capturing images of the human larynx by use of swept-source OCT during a typical office-based laryngoscopy examination is presented. In vivo OCT imaging of the human larynx is demonstrated with a rate of 40 frames per second. Dynamic vibration of the vocal folds is recorded to provide not only high-resolution cross-sectional tissue structures but also vibration parameters, such as the vibration frequency and magnitude of the vocal cords, which provides important information for clinical diagnosis and treatment, as well as fundamental research of the voice itself. Office-based OCT is a promising imaging modality to study the larynx for physicians in otolaryngology.

The purpose of this study was to develop a dynamic tunable focal distance graded-refractive-index lens rod-based high-speed 3-D swept-source (SS) optical coherence tomography (OCT) endoscopic system and demonstrate real-time in vivo, high-resolution (10-microm) imaging of pleural-based malignancies in an animal model. The GRIN lens-based 3-D SS OCT system, which images at 39 fps with 512 A-lines per frame, was able to capture images of and detect abnormalities during thoracoscopy in the thoracic cavity, including the pleura, chest wall, pericardium, and the lungs. The abnormalities were confirmed by histological evaluation and compared to OCT findings. The dynamic tunable focal distance range and rapid speed of the probe and SS prototype OCT system enabled this first-reported application of in vivo 3-D thoracoscopic imaging of pleural-based malignancies. The imaging probe of the system was found to be easily adaptable to various sites within the thoracic cavity and can be readily adapted to other sites, including rigid airway endoscopic examinations.

We present multiphoton microendoscopy with a rotational probe and a 1 microm fiber-based femtosecond laser. The rotational probe is based on a double-clad photonic crystal fiber, a gradient index lens, a microprism, and a rotational microelectronicmechanical system (MEMS) motor. The MEMS motor has a diameter of 2.2 mm and can provide 360 degrees full-view rotation. The fiber laser provides ultrashort pulses with a central wavelength at 1.034 microm and a repetition rate of 50 MHz. Second-harmonic-generation images of rat-tail tendon and fish scale are demonstrated with the rotational probe-based multiphoton system.

The salivary pellicle plays an important role in oral physiology, yet noninvasive in situ characterization and mapping of this layer remains elusive. The goal of this study is to develop an optical approach for the real-time, noninvasive mapping and characterization of salivary pellicles using optical coherence tomography (OCT) and optical coherence microscopy (OCM). The long-term goals are to improve diagnostic capabilities in the oral cavity, gain a better understanding of physiological and pathological processes related to the oral hard tissues, and monitor treatment responses. A salivary pellicle is incubated on small enamel cubes using human whole saliva. OCT and OCM imaging occurs at 0, 10, 30, 60 min, and 24 h. For some imaging, spherical gold nanoparticles (15 nm) are added to determine whether this would increase the optical signal from the pellicle. Multiphoton microscopy (MPM) provides the baseline information. In the saliva-incubated samples, a surface signal from the developing pellicle is visible in OCT images. Pellicle "islands" form, which increase in complexity over time until they merge to form a continuous layer over the enamel surface. Noninvasive, in situ time-based pellicle formation on the enamel surface is visualized and characterized using optical imaging.

Optical coherence tomography (OCT) is a noninvasive, high-resolution imaging technology capable of delivering real-time, near-histologic images of tissues. Mustard gas is a vesicant-blistering agent that can cause severe and lethal damage to airway and lungs. The ability to detect and assess airway injury in the clinical setting of mustard exposure is currently limited. The purpose of this study is to assess the ability to detect and monitor progression of half-mustard [2-chloroethylethylsulfide (CEES)] airway injuries with OCT techniques. A ventilated rabbit mustard exposure airway injury model is developed. A flexible fiber optic OCT probe is introduced into the distal trachea to image airway epithelium and mucosa in vivo. Progression of airway injury is observed over eight hours with OCT using a prototype time-domain superluminescent diode OCT system. OCT tracheal images from CEES exposed animals are compared to control rabbits for airway mucosal thickening and other changes. OCT detects the early occurrence and progression of dramatic changes in the experimental group after exposure to CEES. Histology and immunofluorescence staining confirms this finding. OCT has the potential to be a high resolution imaging modality capable of detecting, assessing, and monitoring treatment for airway injury following mustard vesicant agent exposures.

We describe an algorithm based on shrinkage in the curvelet domain to attenuate speckles in optical coherence tomography (OCT) images. The algorithm exploits the curvelet transform's sparse representation of edge discontinuities that are common in OCT images and its ability to map signals and noise into different areas in the curvelet domain. The speckle attenuation is controlled by a single parameter that determines the threshold in the curvelet domain. Applying the algorithm to OCT images shows significant improvement of image quality.

Optical coherence tomography (OCT) is a catheter-based imaging technology with powerful resolution capable of identifying vulnerable plaques and guiding coronary intervention. However, a significant limitation of intravascular OCT imaging is its attenuation by blood. We propose that the use of an oxygen-carrying blood substitute could potentially optimize OCT image quality. Surgical isolation of the descending thoracic aorta of six rabbits is performed, followed by intravascular OCT imaging of the abdominal aorta. Perfluorodecalin (PFD) is oxygenated using a bubble-through technique with 100% oxygen. OCT imaging is performed and compared using three different flushing modalities: PFD; saline; and blood. OCT imaging of the rabbit abdominal aorta is successful in all of the subjects. In each of the six studied subjects, flushing with PFD consistently provides dramatically better imaging of the vessel wall tissue structures. OCT image quality is highly dependent on the ability of the flushing modality to remove blood from the imaging field. From this proof-of-concept study, we demonstrate that endovascular flushing with an oxygen-carrying blood substitute (PFD) is optically superior to saline flushing for intravascular imaging.

Contrast in optical coherence tomography (OCT) images can be enhanced by utilizing surface plasmon resonant gold nanoparticles. To improve the poor in vivo transport of gold nanoparticles through biological barriers, an efficient delivery strategy is needed. In this study, the improved penetration and distribution of gold nanoparticles were achieved by microneedle and ultrasound, respectively, and it was demonstrated that this multimodal delivery of antibody-conjugated PEGylated gold nanoparticles enhanced the contrast in in vivo OCT images of oral dysplasia in a hamster model.
