The filterscope diagnostic on DIII-D utilizes photomultiplier tubes to measure visible light emission from the plasma. The system has undergone a substantial upgrade since previous attempts to cross-calibrate the filterscope with other spectroscopic diagnostics were unsuccessful. The optics now utilize a dichroic mirror to initially split the light at nearly 99% transmission or reflectance for light below or above 550 nm. This allows the system to measure Dα emission without degrading visible light emission from the plasma for wavelengths below 550 nm (to measure Dβ, Dγ, W–I, C-III, etc.). Additional optimization of the optical components and calibration techniques reduce the error in the signal up to 10% in some channels compared to previous methods. Cross-calibration measurements with two other high resolution spectroscopic diagnostics now show excellent agreement for the first time. This expands the capabilities of the filterscope system allowing measurement of divertor detachment, emission profiles, edge-localized mode behavior, and plasma–wall interactions. SignificanceVisible light optical coherence tomography (OCT) is emerging for spectroscopic and ultrahigh resolution imaging, but challenges remain. Depth-dependent dispersion limits retinal image quality and current correction approaches are cumbersome. Inconsistent group refractive indices during image reconstruction also limit reproducibility.AimTo introduce and evaluate water wavenumber calibration (WWC), which corrects depth-dependent dispersion and provides an accurate depth axis in water.ApproachEnabled by a visible light OCT spectrometer configuration with a 3- to 4-dB sensitivity roll-off over 1 mm in air across a 90-nm bandwidth, we determine the spectral phase of a 1-mm water cell, an affine function of water wavenumber. Via WWC, we reconstruct visible light OCT human retinal images with 1.3-μm depth resolution in water.ResultsImages clearly reveal Bruch's membrane, inner plexiform layer lamination, and a thin nerve fiber layer in the temporal parafovea. WWC halves the processing time, while achieving the same image definition as an assumption-free gold standard approach, suggesting that water wavenumber is a suitable proxy for tissue wavenumber. WWC also provides a depth axis in water without explicitly assuming a group refractive index.ConclusionsWWC is a simple method that helps to realize the full potential of visible light OCT.

Solar-induced chlorophyll fluorescence (SIF) is used as a proxy of photosynthetic efficiency. However, interpreting top-of-canopy (TOC) SIF in relation to photosynthesis remains challenging due to the distortion introduced by the canopy's structural effects (i.e., fluorescence re-absorption, sunlit-shaded leaves, etc.) and sun–canopy–sensor geometry (i.e., direct radiation infilling). Therefore, ground-based, high-spatial-resolution data sets are needed to characterize the described effects and to be able to downscale TOC SIF to the leafs where the photosynthetic processes are taking place. We herein introduce HyScreen, a ground-based push-broom hyperspectral imaging system designed to measure red ( F 687 ) and far-red ( F 760 ) SIF and vegetation indices from TOC with single-leaf spatial resolution. This paper presents measurement protocols, the data processing chain and a case study of SIF retrieval. Raw data from two imaging sensors were processed to top-of-canopy radiance by dark-current correction, radiometric calibration, and empirical line correction. In the next step, the improved Fraunhofer line descrimination (iFLD) and spectral-fitting method (SFM) were used for SIF retrieval, and vegetation indices were calculated. With the developed protocol and data processing chain, we estimated a signal-to-noise ratio (SNR) between 50 and 200 from reference panels with reflectance from 5% to 95% and noise equivalent radiance (NER) of 0.04 (5%) to 0.18 (95%) mW m − 2 sr − 1 nm − 1 . The results from the case study showed that non-vegetation targets had SIF values close to 0 mW m − 2 sr − 1 nm − 1 , whereas vegetation targets had a mean F 687 of 1.13 and F 760 of 1.96 mW m − 2 sr − 1 nm − 1 from the SFM method. HyScreen showed good performance for SIF retrievals at both F 687 and F 760 ; nevertheless, we recommend further adaptations to correct for the effects of noise, varying illumination and sensor optics. In conclusion, due to its high spatial resolution, Hyscreen is a promising tool for investigating the relationship between leafs and TOC SIF as well as their relationship with plants' photosynthetic capacity. Accurate assessment of burn severity is critical for wound care and the course of treatment. Delays in classification translate to delays in burn management, increasing the risk of scarring and infection. To this end, numerous imaging techniques have been used to examine tissue properties to infer burn severity. Spatial frequency-domain imaging (SFDI) has also been used to characterize burns based on the relationships between histologic observations and changes in tissue properties. Recently, machine learning has been used to classify burns by combining optical features from multispectral or hyperspectral imaging. Rather than employ models of light propagation to deduce tissue optical properties, we investigated the feasibility of using SFDI reflectance data at multiple spatial frequencies, with a support vector machine (SVM) classifier, to predict severity in a porcine model of graded burns. Calibrated reflectance images were collected using SFDI at eight wavelengths (471 to 851 nm) and five spatial frequencies (0 to 0.2  mm  -  1). Three models were built from subsets of this initial dataset. The first subset included data taken at all wavelengths with the planar (0  mm  -  1) spatial frequency, the second comprised data at all wavelengths and spatial frequencies, and the third used all collected data at values relative to unburned tissue. These data subsets were used to train and test cubic SVM models, and compared against burn status 28 days after injury. Model accuracy was established through leave-one-out cross-validation testing. The model based on images obtained at all wavelengths and spatial frequencies predicted burn severity at 24 h with 92.5% accuracy. The model composed of all values relative to unburned skin was 94.4% accurate. By comparison, the model that employed only planar illumination was 88.8% accurate. This investigation suggests that the combination of SFDI with machine learning has potential for accurately predicting burn severity.

An optimized compact-based method is introduced that verifies the distance measuring precision of surveying instruments. The paper describes the improvement of the compact based method not only in establishing a compact facility by using shorter lengths and folded path but also in the assessment process by implementing the official ISO standard for electronic distance measuring equipment (ISO 17123-4). The advent of fully coherent free-electron laser and diffraction-limited synchrotron radiation storage ring sources of x-rays is catalyzing the development of new ultrahigh accuracy metrology methods. To fully exploit these sources, metrology needs to be capable of determining the figure of an optical element with subnanometer height accuracy. The major limiting factors of the current absolute accuracy of ex situ metrology are drift errors due to temporal instabilities of the lab's environmental conditions and systematic errors inherent to the metrology instruments. Here, we discuss in detail work at the Advanced Light Source X-Ray Optics Laboratory on building of advanced environmental control that is a key component in the development of ultrahigh accuracy ex situ metrology for x-ray optics. By a few examples, we show how the improvement of the environmental conditions in the lab allows us to significantly gain efficiency in performing ex situ metrology with high-quality x-ray mirrors. The developed concepts and approaches, included in the design of the new X-Ray Optics Laboratory, are described in detail. These data are essential for construction and successful operation of a modern metrology facility for x-ray optics, as well as high-precision measurements in many fields of experimental physics.

We report the development and validation of a hybrid intravascular diagnostic system combining multispectral fluorescence lifetime imaging (FLIm) and intravascular ultrasound (IVUS) for cardiovascular imaging applications. A prototype FLIm system based on fluorescence pulse sampling technique providing information on artery biochemical composition was integrated with a commercial IVUS system providing information on artery morphology. A customized 3-Fr bimodal catheter combining a rotational side-view fiberoptic and a 40-MHz IVUS transducer was constructed for sequential helical scanning (rotation and pullback) of tubular structures. Validation of this bimodal approach was conducted in pig heart coronary arteries. Spatial resolution, fluorescence detection efficiency, pulse broadening effect, and lifetime measurement variability of the FLIm system were systematically evaluated. Current results show that this system is capable of temporarily resolving the fluorescence emission simultaneously in multiple spectral channels in a single pullback sequence. Accurate measurements of fluorescence decay characteristics from arterial segments can be obtained rapidly (e.g., 20 mm in 5 s), and accurate co-registration of fluorescence and ultrasound features can be achieved. The current finding demonstrates the compatibility of FLIm instrumentation with in vivo clinical investigations and its potential to complement conventional IVUS during catheterization procedures.

Single-molecule switching based super-resolution microscopy techniques have been extended into three dimensions through various 3D single-molecule localization methods. However, the localization accuracy in z can be severely degraded by the presence of aberrations, particularly the spherical aberration introduced by the refractive index mismatch when imaging into an aqueous sample with an oil immersion objective. This aberration confines the imaging depth in most experiments to regions close to the coverslip. Here we show a method to obtain accurate, depth-dependent z calibrations by measuring the point spread function (PSF) at the coverslip surface, calculating the microscope pupil function through phase retrieval, and then computing the depth-dependent PSF with the addition of spherical aberrations. We demonstrate experimentally that this method can maintain z localization accuracy over a large range of imaging depths. Our super-resolution images of a mammalian cell nucleus acquired between 0 and 2.5 μm past the coverslip show that this method produces accurate z localizations even in the deepest focal plane.

A polarization-maintaining buffered Fourier domain mode-locked (FDML) swept source with a center wavelength of 1300 nm is demonstrated. The scanning rate of the buffered FDML swept source is doubled without sacrificing the output power of the swept source by combining two orthogonally polarized outputs with a polarization beam combiner. The stability of the swept source is improved because the polarization state of the laser beam inside the laser cavity is maintained without the use of any polarization controllers. The swept source is capable of an edge-to-edge tuning range of more than 150 nm and a FWHM range of 95 nm at a 102 kHz sweeping rate and with an average power of 12 mW. A swept source optical coherence tomography (SSOCT) system is developed utilizing this buffered FDML swept source. The axial resolution of the SSOCT system is measured to be 9.4 µm in air. The sensitivity of the SSOCT system is 107.5 dB at a depth of 0.25 mm with a 6 dB roll-off at a depth of 2.25 mm.

We present a calibration protocol to obtain the alignment factors of a custom-made spectrometer and the nonlinear fitting function between the measured CCD pixel domain and the wavelength domain to apply to the spectral-domain optical coherence tomography (SD-OCT) using fiber Bragg gratings. We have used five gratings with different center wavelengths covering the broadband source spectral range. All have a narrow spectral bandwidth (0.05 nm) and the same reflectivity (92%) to calibrate and align the custom-made spectrometer. The implemented SD-OCT system following the proposed protocol showed the alignment factors as 44.37 deg incident angle, 53.11 deg diffraction angle, and 70.0-mm focal length. The spectral resolution of 0.187 nm was recalculated from the alignment factors.

Sinusoidally structured illumination is used in concert with a phantom-based lookup-table (LUT) to map wide-field optical properties in turbid media with reduced albedos as low as 0.44. A key advantage of the lookup-table approach is the ability to measure the absorption (mu(a)) and reduced scattering coefficients (mu(s) (')) over a much broader range of values than permitted by current diffusion theory methods. Through calibration with a single reflectance standard, the LUT can extract mu(s) (') from 0.8 to 2.4 mm(-1) with an average root-mean-square (rms) error of 7% and extract mu(a) from 0 to 1.0 mm(-1) with an average rms error of 6%. The LUT is based solely on measurements of two parameters, reflectance R and modulation M at an illumination period of 10 mm. A single set of three phase-shifted images is sufficient to measure both M and R, which are then used to generate maps of absorption and scattering by referencing the LUT. We establish empirically that each pair (M,R) maps uniquely to only one pair of (micro(s) ('),micro(a)) and report that the phase function (i.e., size) of the scatterers can influence the accuracy of optical property extraction.

A simple Peltier cooled cold trap aperture is presented to minimize the flux of hydrocarbons on optics in vacuum UV systems. The system can be cooled down to −40 °C under vacuum. To test the effect of the cold trap, the aperture is placed in front of a high-intensity D2 lamp used for calibration in the range of 116 nm to 300 nm which is flanged to a VUV spectrometer. The influence of the aperture temperature is monitored by measuring the intensity loss rate of the Lyman-alpha emission line at 121.6 nm due to the formation of carbon contamination on the MgF2 window of the lamp depending on the Peltier temperature. The application of the aperture significantly reduced the intensity loss from approx. 20% h−1 to less than 2% h−1 and enables the reliable use of a D2 lamp for the relative intensity calibration of the spectrometer. Achieving high form accuracy of optical freeform surfaces in machining is extremely difficult due to their geometric complexity. A positioning or repositioning process is necessary for fabricating optical freeform surfaces during the machining and measuring processes. The concept of fiducial-aided calibration and positioning (FACP) has been developed to provide high-precision relative position data among different coordinate frames to minimize the repositioning errors. This paper attempts to establish an uncertainty analysis model to evaluate the uncertainty and reliability of the FACP method. Firstly, two kinds of the most-used configurations of the FACP systems available for carrying out experiments in both machining and measuring machines are designed with consideration of four main factors. Secondly, a linear transforming model is developed to connect different coordinate frames among the machine tool, on-machine measuring system and off-machine measurement instrument with high precision. Hence, all the uncertainties associated with the FACP method are considered, and a modified chi-squared technique is used to identify the relationship between these uncertainties. Experimental work has been conducted on two machine tools with different on-machine probing systems. The results show that the transformation uncertainty is very small in the proposed linear transforming model, while the final accuracy of the FACP system is sensitive to the measurement results obtained from the on-machine measuring system. This work extends modulated imaging, a recently developed technique based on the projection of structured light on a turbid medium that is able to measure optical properties of the high-scattering medium and perform tomography. We observe that structured light obliquely projected on a turbid medium undergoes a spatial shift during propagation. We propose a method to measure the spatial phase shift of a sinusoidal fringe pattern projected in a turbid medium, and we present a model derived from the diffusion approximation to describe the light propagation. Experimental validation by measurements performed on liquid phantoms is presented.

Multiangle, fiber-based, spectral-domain Doppler optical coherence tomography with a phase-resolved algorithm is presented to measure three components of an arbitrary velocity vector. A beam divider that divides a probe beam to have five independent viewpoints and path length delays was designed. The divider was inserted into the sampling arm of a Doppler optical coherence tomography system between the collimator and the first galvo mirror of a two-axis galvo scanner. The divider produced five independent D k's (the average difference between the wave vectors of incoming and outgoing beams) after passing through the focusing lens while keeping two-axis scanning capability. After calibration, an unknown velocity vector field inside a microtube was quantified by solving a three-dimensional minimization problem.

Solar simulators are a fundamental instrument to characterize solar cells parameters, as they can reproduce the operating conditions under which the solar cells are going to work. However, these systems are frequently big, heavy, and expensive, and a small solar simulator could be a good contribution to test small prototyping devices manufactured in research labs, especially if it could manage the irradiation at any wavelength interval in a custom way. We have designed, developed, and calibrated a small solar simulator made entirely with LEDs, no optics inside, and electronically controlled through a PC using an Arduino microcontroller. The whole structure is 3-D printed in black PLA plastic. The electrical current through the LEDs, and thus the spectral irradiance of the simulator, is controlled with a very intuitive LabVIEW interface. As our calibration proves, we have built an easily reproducible and low-cost Class AAA solar simulator in a central illumination area of 1 cm2, according to the IEC60904-9 standard. This means that the homogeneity in that area is under a 2% deviation in spatial terms, below 0.5% in temporal terms, and is a factor of a 3% close to the AM1.5G sun reference spectrum.
