We determine the optical force for an arbitrarily polarized non-paraxial, strongly aberrated, axially symmetric focusing beam and apply this description to the case of an arbitrarily positioned dielectric microsphere, commonly employed in optical tweezers, not taking into account the contribution of evanescent waves at the interface. We derive the analytical expression of the force profile, finding that the incident polarization does not affect the axial optical force. In addition, we derive an approximated expression for the axial force as a function of beam displacement just outside the microsphere and we show how the information provided by the ripple structure of the optical trapping efficiency versus sphere displacement curve, due to the aberration effect, could be exploited to calibrate the bead axial position versus the experimental beam positioning controls Fechado

The biochemical sensing is almost the only way for communication between unicellular organisms. Prokaryote and Eukaryote chemotaxis has been mechanically studied mainly by observing the directionality and timing of the microorganisms movements subjected to a chemical gradient, but not through the directionality and strength of the forces it generates. To observe the vector force of microorganisms under a chemical gradient we developed a system composed of two large chambers connected by a tiny duct capable to keep the chemical gradient constant for more than ten hours. We also used the displacements of a microsphere trapped in an Optical Tweezers as the force transducer to measure the direction and the strength of the propulsion forces of flagellum of the microorganism under several gradient conditions. A 9 mu m diameter microsphere particle was trapped with a Nd:YAG laser and its movement was measured through the light scattered focused on a quadrant detector. We observed the behavior of the protozoa Leishmania amazonensis (eukaryote) under several glucose gradients. This protozoa senses the gradient around it by swimming in circles for three to five times following by tumbling, and not by the typical straight swimming/tumbling of bacteria. Orientador: Carlos Lenz Cesar Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Fisica Gleb Wataghin Resumo: Microorganismos unicelulares, como os demais seres vivos, necessitam interagir com o ambiente e procurar ativamente por alimentos. Seus orgãos sensoriais, entretanto, se limitam a sensores hidrodinâmicos e receptores bioquímicos de membrana. A quimiotaxia estuda a resposta de organismos unicelulares frente a gradientes de concentração de substâncias atratoras ou repulsoras. Essa resposta é parte essencial do processo de infecção por parasitas no direcionamento e reconhecimento das células a serem infectadas. Dessa forma, um estudo quantitativo da quimiotaxia de parasitas requer o monitoramento tanto dos movimentos do parasita quanto das intensidades, direção e sentido das forças que exerce na presença de gradientes de concentração das substâncias atratoras e repulsoras. Forças de microorganismos com dimensões da ordem de 10 µm são da ordem de pico-Newtons. A pinça óptica é a microferramenta ideal para esse tipo de estudo pelas seguintes razões: (1) tem sensibilidade para medir forças desde 20 femto-Newtons até 200 pico-Newtons, da mesma ordem de grandezas das forças geradas pelos parasitas; (2) é uma técnica remota, sem necessidade de contacto e (3) não destrutiva, pois os lasers no infravermelho não geram calor suficiente para causar danos térmicos. Nesse trabalho mostramos como se pode utilizar o deslocamento da posição de equilíbrio de uma microesfera como transdutor de forças vetoriais, determinando quantitativamente intensidade, direção e sentido das forças na microesfera. Um detector de quadrante da luz do próprio laser da pinça óptica fornece o deslocamento da mesma em duas dimensões. Dessa forma foi possível monitorar a força exercida pelo parasita em tempo real em diferentes gradientes de concentração de várias substâncias. Também desenvolvemos uma câmera de gradiente que garantiu um gradiente unidimensional estacionário na qual se pode aprisionar os parasitas e realizar a medida vetorial da força em função do tempo. Demonstramos a capacidade do nosso sistema de realização de medidas de quimiotaxia utilizando o protozoário Leishmania amazonensis, responsável pela doença leishmaniose, na forma promastigota, na presença de gradientes de glucose. Nossos resultados mostram que além das forças serem fortemente direcionadas na direção do gradiente, que a intensidade das mesmas diminui nas direções contrária ou perpendicular ao gradiente. Com esse trabalho mostramos a construção de um sistema de medidas quantitativa capaz de estudar quantitativamente a quimiotaxia de qualquer parasita frente a qualquer gradiente de concentração de diferentes substâncias Abstract: Unicellular microorganisms, like others live beings, need to interact with environment to find nourishment. They have only hydrodynamics sensors and biochemical receptors on membrane to accomplish this task. The response of the microorganism to concentration gradients of attractive or repulsive chemical substances is called chemotaxis. The investigation of chemotaxis is essential to understand infection processes and how parasites recognizes and directs itself to the cells to be infected. This way a quantitative study of parasites chemotaxis requires the observation of not only its movements, but also the strength, direction and sense of forces exerted in the presence of concentration gradient of attractive or repellent chemical substances. Microorganism¿s forces with dimensions of around 10µm are the order of pico-Newton. Optical tweezers are a suitable tool for this kind of investigation, we can justify this affirmation by the following reasons: (1) it has high sensibility to measure forces from 20 femto-Newton until 200 pico-Newton. These forces are in same order of the forces exerted by the parasites. (2) This technique does not need contact, therefore it is a remote technique; and (3) this tool does not destroy the parasites, because lasers on infrared band do not generate enough heat to cause thermal damage on them. In this work we used the displacements of a microsphere trapped in an Optical Tweezers as the force transducer to measure the direction and the strength of the propulsion forces of flagellum of the microorganism under several gradient conditions. A quadrant detector was utilized to sense the displacement of optical tweeter¿s laser in two dimensions. So we monitor the force exerted by the parasites in real time in several concentrations gradients of different substances. We also developed a system enable to create concentration gradient. This system guaranteed a stationary one-dimensional gradient, which the parasites were arrested and the vectorial measurements of force in time function were realized. In this study, we used the protozoa Leishmania amazonensis in its promastigote form. It is responsible by the disease called leishmaniose and it represents a serious problem of public health. We demonstrated that our system is able to perform chemotaxis measurements in gradients of glucose. Our results suggest that the direction and strength of force can be used to identify the movement of the parasite. We also notice differences in strength forces for both direction x and y for different glucose in several concentrations. Grandes mudanças são esperadas, desde como produzimos alimentos até como combatemos as doenças. O controle dos processos biológicos nos permitirá produzir carne sintética para alimentação, produzir biocombustíveis retirando CO2 da atmosfera, produzir órgãos inteiros para transplante e combater de forma eficiente doenças como câncer, por exemplo. Está claro para o nosso grupo que para se obter esses resultados é necessário entender a biologia na sua unidade mais básica: a célula. A partir do entendimento e domínio das reações químicas que acontecem dentro da célula, e mais especificamente do controle do DNA, é que vamos conseguir atingir essas previsões e revolucionar a maneira como vivemos hoje. Com esse pensamento em mente, o objetivo dessa tese foi desenvolver uma plataforma fotônica integrada para estudos de processos celulares. Nós acreditamos que as ferramentas fotônicas são as ferramentas que preenchem todos os requisitos para os estudos de processos celulares, pois possibilitam o acompanhamento dos processos em tempo real sem causar dano as células. As técnicas presentes são: fluorescência excitada por 1 ou 2 fotons, geração de segundo ou terceiro harmônico, pinças ópticas, imagem por tempo de vida da fluorescência e "fluorescence correlation spectroscopy" (FCS). Nesta tese demonstramos como montar essa plataforma integrada e mostramos sua versatilidade com resultados em várias áreas da biologia e também para o estudo de quantum dots. Abstract: The scientific community believes there is a great chance that the next technological revolution is coming from the control of biological processes. Great changes are expected, from the way we produce food up to the way we fight diseases. The control of biological processes will allow us to produce synthetic meat as food, to produce biofuels extracting CO2 directly from the atmosphere, to produce whole synthetic organs for transplant and to fight diseases, like cancer, in more efficient ways. It is clear to our group that in order to obtain these results it is necessary to understand biology from its most basic unity: the cell. Only from understanding and controlling chemical reactions inside a cell, and more specifically from the DNA controlling, it will be possible to achieve these predictions and cause a revolution in the way we live nowadays. Bearing these thoughts in mind, the objective of this thesis was to develop an integrated photonic platform for study of cellular processes. We believe that photonic tools are the only tools that fulfill all the requeriments for studies of cellular processes because they are capable to follow processes in real time without any damage to the cells. The techniques integrated are: 1 or 2 photon excited fluorescence, second or third harmonic generation, optical tweezers, fluorescence lifetime imaging and fluorescence correlation spectroscopy. In this thesis we demonstraded how to assemble this integrated plataform and we showed its versatility with results from different areas of biology and quantum dots. Suas aplicações nas ciências da vida e biofísica cresceram exponencialmente após a demonstração de que ela permitia manter vivos microorganismos capturados por longos tempos. Informações obtidas destes experimentos requerem um transdutor de força, para o qual se utiliza o deslocamento de uma microesfera capturada. Estamos portanto trabalhando no limite dos regimes de óptica geométrica e Rayleigh, que geralmente são utilizados para simplificar a força óptica. Até hoje não existe consenso entre as teorias das forças, para um regime de tamanho arbitrário, nas pinças ópticas nem para sistemas de alta simetria como microesferas, muito menos em geometrias mais complicadas. Uma das maiores dificuldades encontradas nesse aspecto é a ausência de boas medidas experimentais das forças ópticas, independentes de modelos. Por isso grande parte do trabalho dessa tese foi o desenvolvimento de um sistema de medidas de forças ópticas utilizando pinças duplas para obtenção de toda uma curva da força em função do deslocamento tridimensional da partícula capturada, e não apenas os valores da força em posições fixas. A segunda grande contribuição vem da descrição teórica da força óptica. A grande dificuldade nesse aspecto é a descrição de um feixe incidente de grande abertura numérica e sua decomposição em ondas parciais. É nesse contexto que se encaixa esse trabalho de tese. Acreditamos ter dado uma contribuição extremamente valiosa resolvendo de forma analítica e exata o problema da decomposição de um feixe convergente em ondas parciais relativas a qualquer origem do sistema de coordenadas em três dimensões Abstract: The optical tweezers is an instrument capable of manipulating and trapping dielectric particles through the radiation pressure. Their applications in the life sciences and biophysics increased exponentially after it has been demonstrated that it allowed to microorganisms to be maintained alive trapped for long times. Information obtained from these experiments requires a force transducer, for which the displacement of the captured micro sphere is used. We are therefore working in the limit of the geometrical optics and Rayleigh regime, which are usually used to simplify the optical force. Until today consensus fails to exist among the theories of forces for optical tweezers, of an arbitrary size regime, neither for systems of high symmetry as micro spheres, much less in more complicated geometries. One of the greatest difficulties encountered in this aspect is the absence of good experimental measurements of optical forces, independent of models. Therefore great part of this thesis was the development of a system capable of measuring optical forces using a double tweezers setup to obtain an entire curve of the force as a function of the three-dimensional displacement of the trapped particle, and not just the values of the force for fixed positions. The next grand contribution comes from the theoretical description of the optical force. The large difficulty in this aspect is the description of incident beams of great numerical aperture and its decomposition in partial waves. It is in this context that this thesis fits in. We believed to have given an extremely valuable contribution, solving in an analytical and exact way the problem of the decomposition of a convergent beam partial waves relative the any origin of the three dimensional coordinate system Doutorado Física Doutor em Ciências

Algumas glicoproteínas contém ácido siálico, que é o principal responsável pelas cargas negativas na superfície da hemácia que quando em solução cria um potencial elétrico (Ç) repulsivo. A carga elétrica negativa da superfície eritrocitária influencia na distribuição dos íons da solução ao redor da célula formando uma dupla camada de íons. A primeira, conhecida como camada compacta de cargas ou "Stern" é formada por íons rigidamente ligados à hemácia e a segunda camada é composta por íons distribuídos difusamente e conhecida como camada difusa. O objetivo deste estudo foi medir o potencial zeta (Ç), a espessura da dupla camada das cargas iónicas (DLC) ao redor da hemácia, e a força de agregação eritrocitária com diferentes meios potencializadores da aglutinação utilizando pinça óptica. (...continua) Abstract: The red blood cell (RBC) membrane contains proteins and glycoproteins embedded in a fluid lipid bilayer that confers viscoelastic behavior. Sialylated glycoproteins of the RBC membrane are responsible for a negatively charged surface, which creates a repulsive electric zeta potential (Z) between the cells.The compact layer of charge or Stern consists of ions rigidly bonded to the cell and the double layer includes ions diffusely distributed around the cell. The aim of this study was to measure the RBC double layer thickness of the charge (DLC) around the cell, zeta potential (Z) and cell aggregation force in agglutination potentiator solutions, using optical tweezers. A resolução necessária para atingir esse objetivo requer praticamente ferramentas fotônicas. Contudo, uma integração de diferentes ferramentes fotônicas e uma aproximação funcional serão necessárias para acessar os processos biomecânicos e bioquímicos celulares. Deste modo nós podemos observar eventos bioquímicos disparados mecanicamente ou eventos mecânicos disparados bioquimicamente, ou até mesmo observar simultâneamente eventos biomecânicos e bioquímicos disparados por outros meios, entre outros, eletricamente. Uma das grandes vantagens das ferramentas fotônicas é a sua facilidade de integração. Nós desenvolvemos uma ferramenta integrada incorporando pinça óptica simples com Microscopia Confocal "Single-photon" e Multifóton. O sistema consegue realizar microscopias de fluorescência excitada pela absorção de dois fótons e geração de segundo harmônico em conjunto com manipulações ópticas. Medidas de força, elasticidade e viscosidade de membranes esticadas podem ser monitoradas em tempo real pelas microscopias confocais, bem como protozoários capturados opticamente, como, por exemplo, Trypanosoma cruzi. Nós mostraremos vários exemplos do uso de tal ferramenta integrada e seu potencial para observar processos mecânicos e bioquímicos a nível celular Abstract: The research in biomedical photonics is clearly evolving in the direction of the understanding of biological processes at the cell level. The spatial resolution to accomplish this task practically requires photonics tools. However, an integration of different photonic tools and a multimodal and functional approach will be necessary to access the mechanical and biochemical cell processes. This way we can observe mechanicaly triggered biochemical events or biochemicaly triggered mechanical events, or even observe simultaneously mechanical and biochemical events triggered by other means, e.g. electricaly. One great advantage of the photonic tools is its easiness for integration. Therefore, we developed such integrated tool by incorporating single Optical Tweezers with Confocal Single and Multiphoton Microscopies. This system can perform 2-photon excited fluorescence and Second Harmonic Generation microscopies together with optical manipulations. Force, elasticity and viscosity measurements of stretched membranes can be followed by real time confocal microscopies. Also opticaly trapped living protozoas, such as Trypanosoma cruzi. Integration with CARS microscopy is under way. Orientador: Carlos Lenz Cesar Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Física Gleb Wataghin Resumo: O microscópio de força atômica é uma ferramenta que possibilita a medida de forças precisamente localizadas com resoluções no tempo, espaço e força jamais vistas. No coração deste instrumento está um sensor a base de uma viga (cantilever) que é responsável pelas características fundamentais do AFM. O objetivo desta pesquisa foi usar a deflexão deste cantilever para obter uma calibração rápida e precisa da força da armadilha da pinça óptica, assim como testar e comparar com os método tradicionalmente utilizados para este propósito. Para isso, foi necessário analisar e entender o condicionamento de sinais utilizados no AFM. Foram estudados cantilever tradicionais, cujo sistema de detecção é baseado na deflexão de um feixe laser em conjunto com fotodetectores, bem como cantilevers piezoresistivos. Cantilevers piezoresistivos fornecem uma alternativa simples e conveniente aos cantilevers ópticos. A integração de um elemento sensorial dentro do cantilever elimina a necessidade de um laser externo e de um detector utilizados na maioria dos AFMs. Isto elimina a etapa delicada de alinhamento da laser ao cantilever e fotodetector que normalmente precede uma medida com AFM, uma simplificação que expande o potencial do AFM para o uso em meios adversos, como câmaras de ultra alto vácuo ou, como no caso específico das Pinças Ópticas, onde existem esferas em solução líquida e também restrições de dimensão Abstract: The atomic force microscope (AFM) is a tool that enables the measurement of precisely localized forces with unprecedented resolution in time, space and force. At the heart of this instrument is a cantilever probe that sets the fundamental features of the AFM. The objective of this research has been using the deflection of this cantilever to get a fast and accurate calibration of optical tweezers trap force, as well as testing and comparing to the traditionally used methods of calibration for this purpose. For that it was necessary to resolve and understand the sensors signals conditioning used in the AFM. Traditional cantilevers, whose detection system is based on the deflection of a laser beam in addition with a photodetector, as well as piezoresistive cantilevers has been studied. Piezoresistive cantilevers provide a simple and convenient alternative to optically detected cantilevers. Integration of a sensing element into the cantilever eliminates the need for the external laser and detector used in most AFMs. This removes the delicate step of aligning the laser to the cantilever and photodetector which usually precedes an AFM measurement, a simplification which expands the potential of the AFM for use in difficult environments such as ultrahigh vacuum chambers or, as in Optical Tweezers specific case, where there are spheres into a liquid solution as well as dimensional constraints Mestrado Física Mestre em Física
