Anxiolytic and anxiogenic-like behavioral outcomes have been reported for methylenedioxymethamphetamine (MDMA or ecstasy) in rodents. In the present experiment, we attempted to identify behavioral, hormonal and neurochemical outcomes of MDMA treatment to clarify its effects on anxiety-related responses in 2-month-old Balb/c male mice (25-35 g; N = 7-10 mice/group). The behavioral tests used were open field, elevated plus maze, hole board, and defensive behavior against predator odor. Moreover, we also determined striatal dopamine and dopamine turnover, and serum corticosterone levels. MDMA was injected ip at 0.2, 1.0, 5.0, 8.0, 10, or 20 mg/kg. MDMA at 10 mg/kg induced the following significant (P < 0.05) effects: a) a dose-dependent increase in the distance traveled and in the time spent moving in the open field; b) decreased exploratory activity in the hole board as measured by number of head dips and time spent in head dipping; c) increased number of open arm entries and increased time spent in open arm exploration in the elevated plus maze; d) increased time spent away from an aversive stimulus and decreased number of risk assessments in an aversive odor chamber; e) increased serum corticosterone levels, and f) increased striatal dopamine level and turnover. Taken together, these data suggest an anxiogenic-like effect of acute MDMA treatment, despite the fact that behavioral anxiety expression was impaired in some of the behavioral tests used as a consequence of the motor stimulating effects of MDMA.

The influence of highintensity (110 dB) sound vibrations on experimental rats was studied on the behavior level. Classical behavioral tests (open field, hole board, light-dark box) were used to examine behavioral changes during 5min acoustic impact of either audible or infrasound spectrum. Statistically sig nificant changes in locomotor activity and grooming reactions were observed, following either aural or infra sound impact. Summary: The benefits of long-term moderate exercise for health are widely accepted in humans, but few animal studies have been undertaken to characterize the effects of such activity on emotionality and responsiveness to stress. The present study describes the effects of long-term moderate forced treadmill training (36 weeks) on exploratory activity, anxiety-like behaviour, and the resting or stress levels of some physiological variables, including pituitary-adrenal (PA) hormones. Five-week-old male Sprague-Dawley rats were trained on the treadmill (TM) for 36 weeks, using a more moderate training (12m/min, 30min/day, 4–5 days/week) than that currently used in the literature. Two groups were used as controls: a non-handled sedentary (SED) group, receiving no manipulation, and a control (CON) group exposed to a stationary treadmill for the same amount of time as the TM group. In accordance with literature data, TM rats showed lower resting levels of glucose, triglycerides and cholesterol than the other two groups. The TM and CON groups both showed higher ambulation than the SED group in some behavioural tests, without evidence for altered anxiety. Resting levels of adrenocorticotropin (ACTH) and corticosterone did not differ among the groups, but a reduced ACTH response to both a novel environment (mild stressor) and an active escape-avoidance task (severe stressor) was observed in TM rats, whereas changes in corticosterone were modest. The results support the view that the physiological consequences of long-term moderate training are beneficial, including reduced PA responsiveness to stress, even though exercise training did not affect anxiety-like behaviour. Objective: To study the anxiolytic activity of methanol extract of Achyranthes aspera Linn (Amaranthaceae). Materials and Methods: Male Swiss albino mice were used. Methanolic extract of Achyranthes aspera (MEAA) was administered in the doses of 100, 300 and 600 mg/kg p.o. Hole board (HB), open field (OF), elevated plus maze (EPM) and light/dark exploration (LDE) tests were used for determination of anxiolytic activity. Results: The methanolic extract of Achyranthes aspera significantly increased the number and duration of head poking in HB test. The extract also significantly increased the time spent and the number of entries in open arm in EPM. In LDE test, the extract produced significant increase in time spent and number of crossings and decreased the duration of immobility in light box. In OFT, the extract showed significant increase in number of rearing, assisted rearing and the squares crossed. Conclusion: In the present study, MEAA exhibited anxiolytic activity which might be attributed to its phyto-constituents viz. alkaloid, steroid and triterpenes. Since Achyranthes aspera is ubiquitous and abundantly grown, it could be a fairly economical therapeutic agent for management of anxiety disorders. [ABSTRACT FROM AUTHOR]

Abstract: Attention deficit hyperactivity disorder (ADHD) can coexist with epilepsy. Spontaneously hypertensive rats (SHRs) are considered to model ADHD with overactivity, impulsiveness, deficient sustained attention, and alterations in circadian autonomic profiles. The present study explored spontaneous recurrent seizures (SRSs) and behavioral diurnal activity rhythms in normotensive Wistar rats and SHRs in the kainate model of epilepsy. Rats were video monitored (24h/3 months) to detect SRSs. SHRs manifested a lower seizure frequency during the light phase in the 8th and 10th weeks and a lower frequency of SRSs during the night phase accompanied by attenuated responses in hyperexcitability tests. Both epileptic strains were hyperactive, with lower anxiety levels, and their diurnal rhythms were abolished. Epileptic Wistar rats and SHRs exhibited less exploration during the dark phase. This study suggests that SHRs may be useful in modeling some aspects (particularly hypertension-related diurnal rhythm disturbance) of behavior associated with epilepsy. [Copyright &y& Elsevier]
