Crônica (DOC) é uma imunodeficiência caracterizada por infecções recorrentes graves. Os defeitos moleculares que lavam a DGC são geralmente devidos a mau funcionamento, ausência o baixa expressão de um dos componentes do sistema NADPH oxidase. Este trabalho analisou o uso de RT-PCR para a triagem de defeitos moleculares responsáveis por DGC ligada ao X em 8 pacientes. RNA total foi preparado de linfócitos B transformados com vírus de Epstein-Barr e transcrição reversa com hexâmeros randomicos. O cDNA resultante foi amplificado por PCR com oligonucleotídeos específicos para 3 regiões exônicas abrangendo toda a extensão do gene. Com esta estratégia foi possível a detecção da expressão defeituosa de gp91-phox em 7 pacientes. Concluímos que a análise por meio de RT-PCR, um método alternativo menos complexo, rápido e econômico foi apropriado para detecção inicial de defeitos moleculares em 7 de 8 pacientes com DOC ligada ao X. Posteriormente investigamos em detalhe os defeitos genetico-moleculares de 7 crianças não relacionadas com DGC ligada ao X. Todos os pacientes foram procedentes do Chile e Brasil. Encontramos uma inserção c.1267_1268insA no paciente JY no exon 10 levando a uma mutação tipo "&ameshill:". Esta mutação é um novo registro na literatura. Detectamos duas substituições "nonsense", uma no paciente PT, c.95 G>A no exon 2 que leva a um códon de parada W28X e outra no paciente MF, c.229 C>T no exon 3 que leva a um códon de parada R73X. Em 4 casos, nos pacientes IC, Vin, RS, GO, diferentes erros de "splicing" foram encontrados. Dois pacientes apresentaram uma subtitução c.264 G> A ao final do exon 3. Os dois restantes apresentaram uma subtitução c.1326 + 1 G>A no intron 10 e outra subtitução c.1164 - 2 A>O no intron 9. Esta última mutação também é um novo registro na lit_ratura. As mutações identificadas na proteína gp91-phox confirmam um alto grau de heterogeneidade molecular como é relatado em outros grupos étnicos e a importância de investigar os defeitos moleculares em diferentes populações. Contrastando com esta heterogeneidade, a DGC associada com defeitos na proteína p47-phox, apresentam pouca variabilidade. Neste estudo, os pacientes analisados, dois irmãos, mostram uma deleção homozigota OT (L}.GT) no começo do exon 2. L Também é analisado o caso de um paciente com infecções recorrentes que inicaJmente recebeu o diagnóstico de deficiência de G6PD. Estudos moleculares mostraram que a deficiência de G6PD foi devida a uma mutação 202 G_A, variante Afi:icana. . O paciente também mostrou uma reduzida atividade da explosão respiratória como observado em DGC ligada ao X. A análise do gDNA mostrou uma subtitução 264 G_A na região do splicing do exon 3 da proteína gp91-phox. A seqüência do cDNA detectou uma deleção do exon 3, levando a uma mutante inestável ou não funcional da proteina gp91-phox e resultando no fenótipo de DGC ligada ao X. Propomos que ftente a um paciente com deficiência de G6PD com episódios de infecções graves considerem a possibilidade de um defeito na atividade fagocítica e uma eventual associação com DGC Abstract: Chronic granulomatous disease (CGD) is a rare primary immunodeficiency characterized by ear1y onset recurrent severe infections. The molecular defects causing CGD are generally due to the absence, low expression or malfunctioning of one of the NADPH oxidase components. This work analyzed the potential use of reverse transcription (RT)PCR for screening molecular defects responsible for X-linked CGD in 8 Brazilian patients. Total RNA was prepared from EBV-transformed B-lymphocytes and reverse transcribed using random hexamers. The resultant cDNA was PCR-amplified by specific and overlapping pairs of primers regarding 3 exonic regions of gp91-phox gene. This strategy made possible the detection of defective gp91-phox expression in 7 patients. We conclude that RT -PCR analysis, a less complex, more economic and faster alternative method, was appropriate for screening molecular defects in 7 out 8 X-linked CGD patients. We further investigated the molecular genetic defects in 7 unrelated patients with X-linked CGD, from Chile and Brazil. We found an insertion c.1267_1268insA in exon 10 leading to a frameshift mutation. This mutation is a novel reporto we detected two single base-pair substitutions that lead to nonsense mutations. The first was a c.95 G>A substitution in the exon 2 which predicts a stop codon W28X and the second was a c.229 C> T substitution in the exon 3 which predicts a stop codon R73X. We also identified different splice site mutations in 4 cases. Two patients presented a c.264 G> A substitution at the end of exon 3. The remaining two patients presented either a c.1326 + 1 G>A substitution in intron 10 or a c.1164 - 2 A>G substitution in intron 9. This Iast mutation is also novel. The gp91-phox mutations identified in these patients show a high degree of molecular heterogeneity as reported in other ethnic groups and the importance to investigate molecular genetic defects in diferent populations. Contrasting with the heterogeneity of mutations observed in X-linked CGD, the disease associated with defect in p47-phox shows less variability. In this report, the patients with CGD, two sib1ings, show a homozygotous dinucleotide GT deletion (.6.GT) at the beginning of exon 2. We also reported a child with recurrent infections who initially received the diagnosis of G6PD deficiency. Molecular studies showed that the G6PD deficiency was due a 202 G-+A mutation, the A- variant common in African ethnic groups. The proband also exln'bited severely impaired respiratory burst activity, as observed in X-linked CGD. Sequence analysis of genomic DNA showed a 264 G-+A substitution at the 3' splice junction of gp91-phox exon 3. The cDNA sequence showed a deletion of gp91-phox exon 3, giving rise to an unstable or nonfunctional mutant gp91-phox and to the phenotype of Xlinked CGD. We propose that clinicians in face of a patient with G6PD deficiency under a severe infection episode consider the possibility of temporary or permanent impairment of the phagocytes microbicidal activity, and the eventual association of G6PD deficiencyand chronic granulomatous disease Doutorado Saúde da Criança e do Adolescente Doutor em Saúde da Criança e do Adolescente

Orientador: Marco Aurélio Ramirez Vinolo Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: Os ácidos graxos de cadeia curta (AGCCs), acetato, propionato e butirato, são produtos finais oriundos do metabolismo bacteriano. Além de serem um importante substrato energético às células epiteliais intestinais, estes metabólitos são capazes de modular a ativação do sistema imune e, com isso, parecem exercer um papel importante na manutenção da homeostase do hospedeiro. Um dos principais mecanismos associados aos efeitos dos AGCCs é a ativação de receptores acoplados à proteína G, incluindo o FFAR2, receptor de membrana altamente expresso em neutrófilos. Embora se saiba que as bactérias anaeróbias são capazes de produzir altas quantidades de AGCCs em locais de infecção, os efeitos ocasionados por esta produção local e a ativação de FFAR2 ainda são incertos. Dessa maneira, este trabalho se propôs a investigar os efeitos dos AGCCs, produzidos localmente nos sítios de infecção, na resposta imunológica contra a bactéria patogênica Aggregatibacter actinomycetemcomitans (Aa). Para isso, utilizamos modelo de câmara subcutânea em camundongos C57BL/6 para iniciar e manter uma monoinfecção isolada de Aa no interior do organismo dos animais. Os AGCCs inoculados juntamente com a bactéria na câmara não alteraram o padrão de migração leucocitária ao local da infecção, mas reduziram a capacidade das células em fagocitar e destruir as bactérias ali presentes, além de terem modulado negativamente a produção de citocinas e quimiocinas (TNF-?, IL-10 e Cxcl2) essenciais para orquestrar toda a resposta imune subsequente. Tais efeitos não decorreram de ações diretas dos AGCCs sobre o crescimento bacteriano ou de efeito citotóxico dos AGCCs sobre os leucócitos. In vitro, observamos que a incubação com AGCCs reduziu a capacidade de fagocitose e produção de TNF-? por neutrófilos humanos. Por fim, demonstramos ainda que o receptor FFAR2 é importante na resposta à infecção por Aa, uma vez que na sua ausência, houve uma redução na capacidade de eliminação dessa bactéria. Em suma, estes resultados indicam que a produção de AGCCs no local da infecção interfere com a ação dos neutrófilos e com isso pode contribuir para o início ou progressão do quadro infeccioso Abstract: Short chain fatty acid (SCFAs), acetate, propionate and butyrate, are end products derived from bacterial metabolism. They are not only an important energetic substrate to the intestinal epithelial cells, but they are also capable of modulating the activation of the immune system, thereby helping to maintain the host homeostasis. One of the main mechanisms associated with the effects of the SCFAs is the activation of G protein-coupled receptors, including the FFAR2, a surface receptor highly expressed on neutrophils. Although it is already known that anaerobic bacteria are capable of producing high amounts of SCFAs at their infectious sites, the effects of this local production are still unclear. Thus, this study aimed to investigate the effects of these SCFAs, locally produced at the infectious sites, in the immune response against the pathogenic bacteria Aggregatibacter actinomycetemcomitans (Aa). For that, we have used a subcutaneous chamber model in C57BL/6 mice to initiate and maintain a monoinfection of Aa inside the organism of the animals. The SCFAs, inoculated together with the bacteria inside of the chamber, did not alter the pattern of leukocytes migration to the infectious site, but they reduced the capacity of the migrating cells to phagocytose and to destroy the bacteria, decreasing, as well, the production of certain cytokynes and chemokynes (TNF-?, IL-10 and Cxcl2) that are essential to orchestrate the subsequent immune response. These effects were not caused by direct actions of the SCFAs in the bacterial growth or their cytotoxic effects on leukocytes. In vitro, we have seen that incubation with SCFAs reduced the phagocytic capacity and the production of TNF-? by human neuthophils. Lastly, we have also demonstrated that the FFAR2 receptor is important for the response to Aa infection, since that in its absence, there was a reduction in the capacity to eliminate this bacterium. Painful procedures can affect the function of innate immune cells, such as neutrophils and macrophages, increasing the risk of infectious diseases. The present work aimed to verify if the analgesics flunixin meglumine or ketoprofen can attenuate the pain/discomfort of newborn lambs submitted by elastration tail docking and thereby avoid the impairment of blood granulocytes function. Twenty-one neonate lambs were divided into three treatments: the control group (n=7), not subjected to caudectomy; the flunixin group (n=7), subjected to caudectomy under local anesthesia and analgesia with two doses of flunixin meglumine; and the ketoprofen group (n=7), subjected to caudectomy under local anesthesia and two doses of ketoprofen. Pain indicators were observed by pain posture score (PS), the number of vocalizations (V), frequency of the movement of the ears (EF), and respiratory rates (RR), observed by a 10 minutes videos for each time points: -15min, 6h, 48h, and 144h. At the same time points, the reactive oxygen species (ROS) production and phagocytosis of blood granulocytes were measured by flow cytometry. At 6h after caudectomy, there was a pain indicator increase (RR, V, and PS), a blood granulocyte percentage increase, and a granulocytes phagocytosis reduction for both groups. At 48h, the ketoprofen group spend more time in pain posture and, at 144h, they exhibited a ROS production granulocyte reduction without signs of pain. We conclude the flunixin meglumine and ketoprofen did not prevent the acute pain/discomfort caused by caudectomy, because the groups showed a pain behavior and impaired of the innate immune response however, the flunixin meglumine was effective in controlling the chronic pain and their effects on blood granulocytes function in compare ketoprofen. RESUMO: Procedimentos dolorosos podem afetar a função das células imunes inatas como neutrófilos e macrófagos, aumentando o risco de ocorrer doenças infeciosas. Desta maneira, o presente trabalho pretendeu verificar se os analgésicos flunixin meglumine ou cetoprofeno conseguem atenuar a dor/desconforto de cordeiros neonatos submetidos a caudectomia por elastração, evitando assim o comprometimento da função dos granulócitos sanguíneos. Para tanto 21 cordeiras foram aleatoriamente divididas em três tratamentos: grupo controle (n=7) não submetido a caudectomia, grupo flunixin (n=7) submetido a caudectomia precedida por anestesia local e duas doses de flunixin meglumine, e cetoprofeno (n=7) submetido a caudectomia precedida por anestesia local e duas doses de cetoprofeno. Mensurou-se os indicadores de dor/desconforto após caudectomia por observações de escore de postura de dor (EP), número de vocalizações (V), frequência de movimentar as orelhas (FO) e frequência respiratória (FR), observados em vídeos de 10 minutos nos momentos -15 min e 6, 48 e 144h. Nos mesmos momentos, avaliou-se a as funções de produção de espécies reativas de oxigênio (ERO) e de fagocitose por granulócitos sanguineos em citometria de fluxo. Notou-se aumento dos indicadores de dor (FR, de V e de EP), da porcentagem dos granulócitos sanguíneos e redução da eficiência de fagocitose em ambos os grupos as 6h. As 48h, os animais do grupo cetoprofeno ainda apresentava mais tempo em postura de dor que os demais grupos e as 144h, apresentou redução da produção ERO por granulócitos. Tais achados permitem concluir que tanto o flunexin meglumine como o cetoprofeno não preveniram dor/desconforto agudo promovido pela elastração, pois os dois grupos manifestaram comportamento de dor e redução da resposta imune inata. Ainda, o analgésico flunixin meglumine foi efetivo em controlar a dor mais tardia e seus efeitos na função de granulócitos sanguíneos em comparação ao cetoprofeno.

Objective: To characterize the inflammatory profiles of patients with systemic lupus erythematosus receiving standard treatment compared to healthy controls. Patients and methods: Peripheral venous blood was collected from systemic lupus erythematosus patients (n = 14) and controls (n = 18) at enrollment. Blood samples were used for quantification, by flow cytometry, of CD11b (integrin) and Chemokine receptor CXCR2 expression surface antigen in neutrophils and lymphocytes, while cytokines were assayed in serum samples. Purified neutrophils were assayed by their ability to phagocytize human plasma-opsonized zymosan. Results: Patients had a median (interquartile range) disease activity index of 1.0 (0-2.0) characteristic of patients in remission. Interleukin-6 and interleukin-10 serum concentrations were significantly higher in the patient group compared to controls and the phagocytic index of circulating neutrophils was significantly reduced in patients compared to controls. The levels of interleukin-2, interleukin-5, interleukin-8 and tumor necrosis factor alpha did not significantly differ between patients and controls. Flow cytometric analysis revealed that the integrin expression levels were reduced in lymphocytes (but not in neutrophils) obtained from systemic lupus erythematosus patients, while surface expression of the chemokine receptor 2 was similar in both neutrophils and lymphocytes. Conclusion: Systemic lupus erythematosus patients receiving standard treatment presented with elevated systemic levels of interleukin-6 and interleukin-10, reduced neutrophil phagocytic capacity, and reduced lymphocyte expression of integrin even when symptoms were in remission. These alterations to innate immune components may put these individuals at a greater risk for acquiring infections. RESUMO Objetivo: Caracterizar os perfis inflamatórios de pacientes com lúpus eritematoso sistêmico (LES) que recebiam o tratamento padrão em comparação com controles saudáveis. Pacientes e métodos: Coletou-se o sangue venoso periférico de pacientes com LES (n = 14) e controles (n = 18) no momento da entrada no estudo. As amostras de sangue foram usadas para quantificação, por citometria de fluxo, da expressão dos antígenos de superfície CD11b (integrina) e CXCR2 em neutrófilos e linfócitos, enquanto as citocinas foram avaliadas em amostras de soro. Avaliou-se a capacidade dos neutrófilos purificados de fagocitar zimosan opsonizado com plasma humano. Resultados: Os pacientes apresentavam uma pontuação mediana (intervalo interquartil) no Sledai de 1 (0-2), característica de pacientes em remissão. As concentrações séricas de IL-6 e IL-10 foram significativamente maiores no grupo de pacientes em comparação com os controles; o índice de fagocitose de neutrófilos circulantes estava significativamente reduzido nos pacientes em comparação com os controles. Os níveis de IL-2, IL-5, IL-8 e TNF-α não diferiram significativamente entre pacientes e controles. A análise da citometria de fluxo revelou que os níveis de expressão de CD11b estavam reduzidos nos linfócitos (mas não nos neutrófilos) obtidos de pacientes com LES, enquanto a expressão do receptor de superfície CXCR2 foi semelhante em neutrófilos e linfócitos. Conclusão: Os pacientes com LES que recebiam tratamento padrão apresentaram níveis sistêmicos elevados de IL-6 e IL-10, redução na capacidade fagocítica dos neutrófilos e redução da expressão de CD11b em linfócitos, mesmo quando os sintomas estavam em remissão. Essas alterações nos componentes da imunidade inata podem colocar esses indivíduos em maior risco de adquirir infecções.

Phagocytosis by neutrophils and monocytes constitutes the main defense mechanism against bacterial challenges in periodontitis. Phagocytosis by neutrophils has already been evaluated, whereas phagocytic function of monocytes has hardly been addressed so far. Objectives: The aim of this study was to assess phagocytosis by neutrophils and monocytes in periodontitis. Material and Methods: The sample included 30 subjects with severe periodontitis and 27 control subjects without periodontal disease. The phagocytic index (PhI) was calculated as the mean number of adhered/ingested Saccharomyces cerevisiae per phagocytozing monocyte or neutrophil multiplied by the percentage of phagocytes involved in phagocytosis. Results: A significant reduction in phagocyte functions was observed in individuals with periodontitis. The median of PhI of neutrophils using nonsensitized S. cerevisiae was 3 for the control group, and 1.5 for the periodontitis group (p=0.01, Mann-Whitney test). The median of PhI of monocytes with non-sensitized S. cerevisiae was 26.13 for the control group, and 13.23 for the periodontitis group (p=0.03, Mann Whitney test). The median of PhI of monocytes assessed with sensitized S. cerevisiae was 97.92 for the control group and 60.1 for the periodontitis group (p=0.005, t-test). Conclusion: The data demonstrated a reduction in the function of phagocytes, suggesting a decrease in immune defenses in periodontitis.

Introduction This study aims to optimize some experimental conditions of a flow cytometric assay to examine the human neutrophil ability to phagocytose immune complexes (ICs) via Fcγ and complement receptors (FcγR and CR, respectively). The parameters assessed were: number of cells, concentration of ICs, reaction time, pH and concentration of the Trypan Blue quenching solution. Methods Neutrophils were isolated from peripheral blood of healthy volunteers. Precipitated ICs composed of IgG and fluorescein isothiocyanate (FITC)-labeled ovalbumin, opsonized or not with serum complement, were used to trigger the neutrophil phagocytosis via FcγR, CR, and FcγR + CR. Fluorescence of the internalized ICs was measured by flow cytometry, after quenching the extracellular fluorescence with Trypan Blue. Results The optimal experimental conditions established for the phagocytosis assay were: 1 × 10 6 cells mL − 1 and 40 μg mL − 1 FITC-labeled ICs, incubated for 30 min, at 37 °C, in 0.5 mL of reaction volume. Trypan Blue solution at 1.25 mg mL − 1 pH 4.4 was the best fluorescence quencher of FITC-labeled ICs attached to the outer surface of neutrophils. Discussion The selected experimental conditions were viable to evaluate IC phagocytosis by neutrophils; they are also suitable to compare the efficiency of IC phagocytosis mediated by FcγR and CR classes of membrane receptors, alone or in combination. This method finds application in studies of (i) the receptor-specific phagocytic function of normal and pathogenic neutrophils as well as (ii) the impact of drugs and therapies on this essential effector function of neutrophils.

Insulin is a pivotal regulator of glucose metabolism and exerts an important anabolic function throughout the body. Insulin commands the glucose uptake by the cells and might control the processes in which there is need for energy such as mitogenesis and gene transcription. In certain conditions, diabetes mellitus for example, when insulin is diminished, the homeostasis of many tissues and organs are broken what can lead to a higher mortality due to an enhanced susceptibility to infections. This vulnerability to infections can partially be explained by a change in response to inflammation. In fact, diabetic animals and patients show a deficient inflammatory response. Many animal models have shown that neutrophils chemotaxis and recruitment are dampened and macrophages from diabetic patients have low phagocytic and microbicidal activities. In most cases, once insulin therapy is introduced, clinical symptoms and signs can be reverted. In addition, there are a number of studies trying to demystify pathways under insulin command. Researchers are also trying to understand how insulin is able to keep inflammatory response under control, restores innate immune cells ability to fight against pathogens and harmlessly activates adaptive immunity response. This review provides an overview on how inflammatory response is driven in the absence of insulin in diabetes and discusses recent findings on the influence of insulin on innate imune response. At the end, some signaling pathways are also highlighted and important enzymes and proteins that control DNA transcription are presented.

OBJETIVO: Investigar o efeito de uma sessão de exercício abaixo do limiar anaeróbio sobre as funções de neutrófilos e monócitos circulantes em ratos Wistar. MATERIAL E MÉTODO: Trata-se de um delineamento experimental com um pós-teste em que os sujeitos do grupo experimental exercitaram-se na água por 30 minutos enquanto controles não foram submetidos a qualquer intervenção. A atividade funcional dos fagócitos circulantes foi avaliada por ensaio de fagocitose de Saccharomyces cerevisiae e pelo teste de redução de nitroazul de tetrazólio (NBT). Para comparação entre as médias dos grupos, utilizaram-se testes t para amostras independentes com p < 0,05. RESULTADO: Não foram observadas diferenças significativas no número total e diferencial de leucócitos entre os grupos. Entretanto, neutrófilos do grupo experimental não apenas fagocitaram mais S. cerevisiae (p = 0,005) como também foram mais eficientes em reduzir NBT (p = 0,018) em relação aos controles. Não existiram diferenças significativas na atividade funcional dos monócitos entre os grupos. CONCLUSÃO: O exercício realizado com intensidade abaixo do limiar anaeróbio foi suficiente para incrementar a atividade fagocítica e microbicida de neutrófilos em modelo animal, o mesmo não sendo observado para monócitos circulantes. _________________________________________________________________________________ ABSTRACT OBJECTIVE: To assess the effect of a single session of exercise conducted below the anaerobic threshold on the functions of neutrophils and circulating monocytes in Wistar rats. MATERIAL AND METHOD: This study had an experimental design with a single post-test in which subjects of the experimental group exercised in water for 30 minutes, whereas the control group was not submitted to any intervention. Functional capacity of circulating phagocytes was assessed by means of a phagocytosis assay of Saccharomyces cerevisiae and a nitro blue tetrazolium (NBT) reduction test. To compare means between groups, t tests for independent samples were used with p set at < 0.05. RESULTS: There were no significant differences as to the total and differential number of leucocytes between groups. Nonetheless, not only did the neutrophils of the experimental group phagocyte more S. cerevisiae (p = 0.005), but they also reduced NBT more efficiently (p = 0.018) in comparison with control subjects. No significant differences in these functional activities were observed in circulating monocytes. CONCLUSION: The exercise performed below the anaerobic threshold was sufficient to increase both phagocytic and microbicide activities of neutrophils in animal model, but the same protocol did not yield similar results for circulating monocytes.

A função fagocitária e a produção de radicais microbicidas de oxigênio pelos monócitos e neutrófilos foram avaliadas em 9 pacientes com cardiopatia chagásica com insuficiência cardíaca congestiva e 9 sem a síndrome em comparação com 11 indivíduos controles normais pelo teste de fagocitose de Saccharomyces cerevisiae e redução do nitroblue tetrazolium pelos fagócitos do sangue periférico. O índice fagocitário dos monócitos dos chagásicos sem insuficiência cardíaca foi significantemente 6,7 e 10,6 vezes menor do que o dos controles e chagásicos com a síndrome congestiva, respectivamente, devido ao menor envolvimento na fagocitose e a menor capacidade destas células de ingerirem partículas. Nos chagásicos sem insuficiência cardíaca os neutrófilos também apresentaram o IF 11,2 e 19,8 vezes menor que os controles e chagásicos com insuficiência cardíaca, respectivamente. A porcentagem de redução do NBT foi normal e similar para os três grupos. Efeitos opostos equilibrados das disfunções devidas às alterações cardiovasculares e imunes podem ter atuado nos pacientes chagásicos com insuficiência cardíaca paradoxalmente recuperando a função fagocitária alterada. _______________________________________________________________________________ ABSTRACT We evaluated the in vitro phagocytic function and the production of microbicidal oxygen radicals by monocytes and neutrophils of 9 Chagas’ heart disease subjects with heart failure and 9 without the syndrome in comparison with 11 healthy subjects, by assessing phagocytosis of Saccharomyces cerevisiae and NBT reduction by peripheral blood phagocytes. Phagocytic index of monocytes of chagasics without heart failure was significantly 6.7 and 10.6 times lower than those of controls and chagasics with the congestive syndrome, respectively, due to a lesser engagement in phagocytosis and to an inability of these cells to ingest particles. Neutrophils also show in chagasics without heart failure PI 11.2 and 19.8 times lower than that of controls and chagasics with heart failure, respectively. The percent of NBT reduction was normal and similar for the three groups. Balanced opposite effects of cardiovascular and immune disturbances may be acting in Chagas’ disease subjects with heart failure paradoxically recovering the altered phagocytic function.

Todavia, o tratamento semelhante dos animais com o meio de cultivo infusão de cérebro e coração (brain heart infusion-BHI) estéril exerceu proteção de apenas 23%. O número de neutrófilos que migraram para a cavidade peritoneal representou apenas 10% do verificado nos outros tratamentos. A atividade fagocitária não estava relacionada ao índice de sobrevivência dos animais, uma vez que tal atividade foi duas vezes maior no grupo KS do que nos grupos Tg e Cg, e a taxa de sobrevivência nesse grupo foi semelhante à observada no grupo Cg. Esses resultados sugerem que o aumento da migração de neutrófilos para o sítio infeccioso induzida pelas administrações prévias das diferentes substâncias é um importante fator associado com a proteção dos animais contra os efeitos letais da infecção bacteriana. The intraperitoneal inoculation of the animals with thioglycollate (Tg, 90 mg), carrageenin (Cg, 500 mu g), or killed S. typhimurium (KS, 1.9 x 10(9) CFU) 24 hours before challenge with alive S. typhimurium caused a significant neutrophil migration to the peritoneal cavity (determined 6 hours after challenge) and protected the animals against the lethal effect of the bacteria at a rate of 50%, 83% and 80%, respectively. However, similar treatment of the animals with brain heart infusion broth (BHI, S. typhimurium grout medium) protected the animals by only 23%. The migrating neutrophils in these animals represented less than 10% of those observed with other treatments. The phagocytosis activity was not associated with the survival rate as this activity was 2 times higher in the KS group than in the Tg and Cg groups, and the survival rate in this group was similar to that observed in Cg. These results suggest that the increase of neutrophil migration to the infectious site is an important factor associated with protection of the animals against the lethal effect of the bacterial infection.
