In the present study, the adsorption of glucose oxidase (GOD) to a mesoporous aluminum oxide (MAO) film was examined with in-situ neutron reflectometry (NR) measurements. The MAO film was deposited on a cover glass slip and a Si disc, and its pore structure was characterized by X-ray reflectometry (XRR) and NR. The Si disc with MAO film was applied for an in-situ NR experiment, and its NR profiles before/after adsorption of GOD were continuously measured with a flow cell. The results indicated that the negatively-charged GOD molecules hardly penetrate into the narrow pore channel (pore diameter = ca. 10 nm) with opposite surface charge.

Protein arrays are used in a wide range of applications. The array described here binds IgG antibodies, produced in rabbit, to gold surfaces via a scaffold protein. The scaffold protein is a fusion of the monomeric E. coli porin outer membrane protein A (OmpA) and the Z domain of Staphylococcus aureus protein A. The OmpA binds to gold surfaces via a cysteine residue in a periplasmic turn and the Z domain binds immunoglobulins via their constant region. Polarised Neutron Reflection is used to probe the structure perpendicular to the gold surface at each stage of the assembly of the arrays. Polarised neutrons are used as this provides a means of achieving extra contrast in samples having a magnetic metal layer under the gold surface. This contrast is attained without resorting to hydrogen/deuterium exchange in the biological layer. Polarised Neutron Reflection allows for the modelling of many and complex layers with good fits. The total thickness of the biological layer immobilised on the gold surface is found to be 187 A and the layer can thus far be separated into its lipid, protein and solvent parts.

The crystallization of DL-aspartic acid beneath nylon 6 spread films has been studied for 150% supersaturated systems using neutron reflectivity and external reflection FTIR. The neutron reflectivity data showed the gradual incorporation of DL-aspartic acid within a nylon 6 spread film layer over a period of 6-8 h, culminating in over 50 vol % of the "film" layer comprising DL-aspartic acid. Accumulation of further DL-aspartic acid material to produce microscopic/macroscopic surface crystals occurred, but on a more limited scale, resulting in approximately 1-5% surface coverage of crystals over the same period. External reflection FTIR studies revealed very weak bands attributable to DL-aspartic acid in surface regions devoid of visible crystals, in agreement with the neutron reflectivity studies. In regions with visible crystals, much larger and sharper DL-aspartic acid bands were seen. Changes in the intensity of the nylon 6 NH stretch band were often observed during the visible crystallization and dissolution of DL-aspartic acid and were consistent with the reversible accumulation of nylon 6 around the growing crystals.
