The cortical representation of visual perception requires the integration of several-signal processing distributed across many cortical areas, but the neural substrates of such perception are largely unknown. The type of firing pattern exhibited by single neurons is an important indicator of dynamic circuitry within or across cortical areas. Neurons in area PEc are involved in the spatial mapping of the visual field; thus, we sought to analyze the firing pattern of activity of PEc optic flow neurons to shed some light on the cortical processing of visual signals. We quantified the firing activity of 152 optic flow neurons using a spline interpolation function, which allowed determining onset, end, and latency of each neuronal response. We found that many PEc neurons showed multiphasic activity, which is strictly related to the position of the eye and to the position of the focus of expansion (FOE) of the flow field. PEc neurons showed a multiphasic activity comprised of excitatory phases interspersed with inhibitory pauses. This phasic pattern seems to be a very efficient way to signal the spatial location of visual stimuli, given that the same neuron sends different firing patterns according to a specific combination of FOE/eye position.

Neurons in area PEc, a visual area located in the superior parietal lobule, are activated by optic flow stimuli. An important issue is whether PEc neurons are able to integrate multimodal signals, such as those related to optic flow selectivity with those about eye and head position. The aim of this study was to assess if angle of gaze and/or head rotation modify the spatial representation of the focus of expansion (FOE), varying FOE, fixation point and head position in space. We found that the rotation of head modulated the firing activity of PEc optic flow neurons. The head position also changed the angle of gaze effect on the PEc neuronal activity. All recorded neurons showed a main interaction effect between head and eye position upon the selectivity for optic flow stimuli. These results seem to suggest that PEc optic flow neurons use different reference frames depending on the position of the eye and/or the head in space emphasizing a possible contribution of this area in guiding locomotion by integrating multiple extraretinal inputs., (Copyright © 2014 Elsevier Ireland Ltd. Alzheimer's disease is increased in diabetic patients. A defective insulin activity on the brain has been hypothesized to contribute to the neuronal cell dysregulation leading to AD, but the mechanism is not clear. We analyzed the effect of insulin on several molecular steps of amyloid precursor protein (APP) processing and β-amyloid (Aβ) intracellular accumulation in a panel of human neuronal cells and in human embryonic kidney 293 cells overexpressing APP-695. The data indicate that insulin, via its own receptor and the phosphatidylinositol-3-kinase/AKT pathway, influences APP phosphorylation at different sites. This rapid-onset, dose-dependent effect lasts many hours and mainly concerns dephosphorylation at the APP-T668 site. This effect of insulin was confirmed also in a human cortical neuronal cell line and in rat primary neurons. Cell fractionation and immunofluorescence studies indicated that insulin-induced APP-T668 dephosphorylation prevents the translocation of the APP intracellular domain fragment into the nucleus. As a consequence, insulin increases the transcription of antiamyloidogenic proteins such as the insulin-degrading enzyme, involved in Aβ degradation, and α-secretase. In contrast, the transcripts of pro-amyloidogenic proteins such as APP, β-secretase, and glycogen synthase kinase (Gsk)-3β are decreased. Moreover, cell exposure to insulin favors the nonamyloidogenic, α-secretase-dependent APP-processing pathway and reduces Aβ40 and Aβ42 intracellular accumulation, promoting their release in the extracellular compartment. The latter effects of insulin are independent of both Gsk-3β phosphorylation and APP-T668 dephosphorylation, as indicated by experiments with Gsk-3β inhibitors and with cells transfected with the nonphosphorylatable mutated APP-T668A analog. In human neuronal cells, therefore, insulin may prevent Aβ formation and accumulation by multiple mechanisms, both Gsk-3β dependent and independent.

Neurons in area PEc in the superior parietal cortex encode signals from different modalities, such as visual, extraretinal and somatosensory, probably combining them to encode spatial parameter of extrapersonal space to prepare body movements. This study reports the characterization of the functional properties of PEc non-visual neurons that showed saccade-related activity. We analyzed the pre- and post-saccadic firing activity in 189 neurons recorded in five hemispheres of three behaving monkeys. Spiking activity of PEc single neurons was recorded while the monkeys performed visually-guided saccades in a reaction time task. We found that 84% of neurons recorded from area PEc showed pre-saccadic activity with directional tuning. In 26% of neurons, we found inhibition of activity in the pre-saccadic period. The onset of this "pause" always started before the saccade and, in 51% of neurons, it was invariant among different gaze directions. The post-saccadic activity in these cells was either a phasic response with directional tuning (77%) and/or an eye position tuning (75%). The analysis of the preferred direction did not show hemispheric preference, however, for the majority of neurons, the angular difference in the preferred direction, in the pre- and post-saccadic period, was more than 60 degrees . By confirming, therefore, that PEc neurons carry information about eye position, these novel findings open new horizons on PEc function that, to date, is not well documented. The pre-saccadic activity may reflect an involvement in saccade control, whereas post-saccadic activity may indicate a role in informing on the new eye position. These novel results about saccade and eye position processing may imply a role of area PEc in gaze direction mechanisms and, possibly, in remapping visual space after eye movements.

Area PEc, a high order association area, is located in the dorsocaudal portion of the superior parietal cortex. PEc neurons encode visual motion signals, especially the direction of stimulus motion. The present study tested if PEc neurons also process visual correlates of self-motion. The extracellular activity of single neurons in response to optic flow stimuli was recorded in two monkeys (Macaca fascicularis) trained in a fixation task. The stimuli were produced by random dots simulating planar motion, radial expansion and radial contraction. A substantial number of PEc neurons were specifically activated by radial optic flow and were selective for the position of the focus of expansion with respect to the fovea. Eccentric positions of the focus of expansion were preferred. Almost all neurons showed opponent excitatory-inhibitory activity to expanding-contracting visual fields. Planar motion elicited very weak responses. Optic flow responsiveness is not entirely explained by classical bar sensitivity in PEc neurons, suggesting that optic flow and classical bar responses could serve different mechanisms in the integration of visuo-motor signals to prepare body movements.

Area PE of macaques has traditionally been considered a somatosensory association cortex. Recent studies, however, suggest that neurons of this and neighboring areas are involved in the visual control of movement, especially arm movement. We investigated the neuronal sensitivity to local visual stimuli of this region by recording neuronal activity in two behaving macaque monkeys trained in a simple visual fixation task. Recordings were performed from the dorsal surface of the caudal pole of the superior parietal lobule (SPL). Classical receptive fields (RFs) were mapped by using conventional static or moving luminous figures. We found that many neurons in this area were selectively activated by moving visual stimuli. Cell responses were tuned to the movement direction. RFs were usually large; their mean surface covered some 30 x 30 degrees of the visual field. The fovea was often included into RF, in many cases it was along a RF side. The center of RFs was mainly located in the contralateral hemifield, although RFs having the center ipsilaterally sited were also found. No evident retinotopy was found. Visual neurons were especially concentrated in a region of the SPL likely corresponding to area PEc. These results suggest that the caudal part of area PE contains neuronal populations specifically signaling local visual motion, possibly encoding the direction of moving objects. These signals might well be suited for sensorimotor integration mechanisms aimed at motor acts.

The influences of the visual background on the spontaneous and evoked activity of neurons in the striate cortex (V1) of the awake and behaving macaque were investigated using uniform (dark and bright) and textured (dynamic random-dot) large fields (10 degrees) centered on the receptive field of the cell under study. Rhesus monkeys were trained to fixate a small target while visual patterns were presented on monitor displays and the impulse activity of single cortical neurons recorded extracellularly with metal microelectrodes. The discharge rates of the ongoing, spontaneous activity of the vast majority of V1 neurons, as well as their responses to optimally adjusted bar stimuli, were not significantly influenced by the luminance of a uniform background. On the other hand, the activity of more than 50% of V1 neurons was clearly affected by a textured background. Comparison of the effects of a uniformly dark background and a background of dynamic random dots showed that the neuron's spontaneous discharge rate was typically higher in the presence of the textured background, while the evoked response was often reduced in amplitude or even suppressed. The opposite effects were observed in only a few neurons. These findings indicate that neurons in area V1 are highly sensitive to a textured background of dynamic random dots which exert on them an activating effect, chiefly by stimulation of the neuron's receptive field, with consequent increase in the ongoing discharge and a reduction of the dynamic range of impulse activity, leading to a reduction in the amplitude of the response evoked by a contrast stimulus.

Single unitresponses to photic stimuli of variable duration were studied in the cat visual areas 17 and 18. The closed-chamber technique was used to record extracellularly impulsive electrical activity in locally anaesthetized, Flaxedil-paralyzed and artificially ventilated cats. Stimulus duration ranges between 0.020 and 10 sec, the background luminance and the intensity of stimuli being constant. We found a first group of cells which fire independently of the stimulus duration; a second group which fire in close relation to the stimulus duration in a rather wide range of values, and a third group which fire monotonously as long as the stimulus goes on. The three groups of cells are supposed to be three functionally different types of cortical cells.

Extracellular recordings were carried out in the primary visual cortex of behaving macaque monkeys. Neurons were activated by moving a visual stimulus across their receptive fields during steady fixation and by moving their receptive fields (by visual tracking) at the same velocity across a stationary visual stimulus. Out of a total number of 123 cells studied, 111 were activated by the visual stimulation both during fixation and tracking. The remaining 12 cells showed good response during fixation and very weakened response during tracking. Our results show that a small percentage of striate neurons in macaque monkey could discriminate between the real motion of an object in the visual world and the self-induced displacement of its retinal image.

Stimulus-response curves of simple cells of the visual cortex were obtained by using 500-msec stationary stimuli. Background influence on single unit responses was studied. The contrast sensitivity of simple cells increases as a function of background luminance. The resolution power of these cortical cells for detecting differences in stimulus contrast decreases at background levels above 0.09 cd/m2.

A substantial proportion of both simple and complex neurons in the cortex subserving central vision are differentially sensitive to binocular disparity of isolated line patterns (local stereopsis), a sensitivity based on a positional disparity between the neuron's receptive fields in the two eyes. In addition, a subset of cortical neurons, nearly all complex neurons, responds to dynamic random-dot stereograms containing no depth cues other than disparity. These neurons are capable of signaling the correct binocular matches among a multitude of false matches in the stereograms (global stereopsis). The discovery of cyclopean neurons in striate cortex, at early stages of the processing neural network for stereoscopic vision provides a new insight of the basic neural mechanisms underlying binocular depth perception.
