The medial preoptic nucleus (MPN) and the bed nucleus of the stria terminalis (BNST) of mice contain sexually dimorphic nuclei (SDNs) that are larger and have more neurons expressing calbindin D-28K (CB), a calcium-binding protein, in males than females. However, it is largely unknown whether such SDNs exist in species other than rodents. In this study, we performed an immunohistochemical study of CB in the MPN and BNST of musk shrews and Japanese quails to examine the existence of homologs of SDNs in mice. Like mice, musk shrews had a SDN exhibiting male-biased sex differences in volume and CB-immunoreactive (ir) cell number in the MPN. The BNST of musk shrews also contained a male-biased SDN, but consisted of non-CB neurons. The paratenial thalamic nucleus of musk shrews, but not mice, had more CB-ir cells in males than females. In Japanese quails of both sexes, CB-ir cells in the MPN and BNST were extremely small in number and did not cluster. These results suggest that the distribution of CB neurons differs among these species. We subcutaneously injected 0.5 mg/kg veratrine into the musk shrew (Suncus murinus), observed the presence or absence, latency, and the incidence of vomiting in each animal for 90 min, and selected animals that frequently vomited (FV group) and those that did not vomit (NV group). Subsequently, animal brains were removed, and the induction of c-fos protein (Fos) was immunohistochemically examined to evaluate neuronal activity in the medulla oblongata. The distribution of Fos-positive neurons in the medulla oblongata was similar between FV and NV groups, with numerous neurons along the entire length of the nucleus of the solitary tract and in the ventrolateral reticular formation. Both veratrine-injected groups showed higher numbers of positive neurons than the saline administered group. However, while the FV group showed a high concentration of positive neurons in the dorsal-dorsomedial reticular formation of the nucleus ambiguus in the rostral medulla, the NV group showed few positive neurons in this area. Fos activity in neurons in this area appeared to be higher in animals with a higher incidence of vomiting.

Nitric oxide (NO) is a novel neuronal messenger that likely influences retinal function by activating retinal guanylyl cyclase to increase levels of cGMP. In the present study, the localization of neuronal nitric oxide synthase (nNOS, Type I NOS) in the cone-dominant tree shrew retina was studied using NADPH-d histochemistry and nNOS immunocytochemistry. Both NADPH-d and nNOS-immunoreactivity (IR) labeled the inner segments of rods and the myoids of a regular subpopulation of cones, with their corresponding nuclei outlined. The labeled cone myoids were co-localized with a marker for short-wave-sensitive (SWS) cones (S-antigen) and also displayed the regular triangular packing and density (7%) characteristic of SWS cones in tree shrew and other mammalian retinas. These measures confirmed the identity of the labeled cones as SWS cones. Photoreceptor ellipsoids of all cones were strongly labeled by NADPH-d reactivity, but lacked nNOS-IR. Another novel finding in tree shrew retina was that both NADPH-d and nNOS-IR labeled Muller cells, which have not been labeled by nNOS-IR in other mammalian retinas. Consistent with findings in rod-dominant retinas, two types of amacrine cells at the vitreal edge of the inner nuclear layer and a subpopulation of displaced amacrine cells at the scleral edge of the ganglion cell layer were labeled by both NADPH-d and nNOS-IR. Processes of these labeled cells were seen to extend into the inner plexiform layer, where dense punctate label was seen, especially in the central sublamina. These results show that localization of NOS in the cone-dominant tree shrew retina shares some common properties with rod-dominant mammalian retinas, but also shows some species-specific characteristics. The new finding of nNOS localization in tree shrew SWS cones and rods, but not in other cones, raises interesting questions about the roles of NO in the earliest level of visual processing.

The organization of the main olfactory bulb of an insectivore, the laboratory shrew (Suncus murinus), was studied morphologically. We found particular small spherical regions, nidi, at the border between the glomerular and external plexiform layer (EPL), which were intensely GAD positive, 30-60 microm in diameter, and where no olfactory nerves were seen. Around the nidus small calbindin D28k-positive GABAergic neurons, perinidal cells, were clustered. Furthermore, a distinctive type of newly discovered neurons, which we named tasseled cells, located at the middle of the EPL extended dendrites to the nidus, where their small tuft-like complicated branches made synapses with perinidal cells. The present study showed that the basic components of the olfactory bulb are not necessarily constant in all mammals.

In many mammalian species the neuroendocrine regulation of male and female reproductive behavior is sexually dimorphic. By contrast, many features of female sexual behavior in the musk shrew (Suncus murinus) more closely resemble those of males than of females of other species. Female musk shrews require testosterone (T), which is neurally aromatized to estrogen, to induce sexual behavior. Aromatization occurs in the medial preoptic area (MPOA), and this region is critical for the expression of female receptivity. To compare neural responses to sexual behavior in females and males, we compared the number of Fos-like immunoreactive (Fos-ir) neurons after mating in musk shrews. In both males and females the number of Fos-ir neurons was increased by mating activity in the granule layer of the accessory olfactory bulb (gr-AOB), the bed nucleus of the stria terminalis (BNST), MPOA, the medial amygdala (MeA), and the region corresponding to the midbrain central tegmental field (CTF). Although Fos was induced by mating in several regions, this response was only dimorphic in the ventral medial nucleus of the hypothalamus (VMN), where mating significantly increased Fos-ir in females, but not in males. In both sexes, only the gr-AOB displayed an increase in Fos-ir after exposure to chemosensory cues alone. Thus, the pattern of Fos expression in the brain after mating is only sexually dimorphic in one region, the VMN. Further, in spite of past behavioral studies done in this species, which show a role for pheromones in induction of receptivity, these data show that exposure to pheromones does not induce Fos in structures caudal to the olfactory bulbs., (Copyright 1998 Published by Elsevier Science B. The nervous system responds to both internal and external cues, integrating these signals to coordinate behavior and physiology. Mating interactions can promote dramatic changes in neuroendocrine cells which trigger successful copulation, ovulation, fertilization, and pregnancy. The neurons that transduce behavioral cues into neuroendocrine signals are distributed in a loose continuum along the medial ventral forebrain where they produce and secrete gonadotropin-releasing hormone (GnRH). In the past we have reported changes in GnRH-immunoreactive (GnRH-ir) cell numbers in brains of female musk shrews sacrificed during, and after, brief mating interactions. The purpose of the current study was twofold: first to determine which aspect of intracellular GnRH production is stimulated by behavioral interactions; second, to characterize the specific aspects of the social exchange that trigger GnRH production. We report that 1 h after copulation the production of proGnRH protein is stimulated. Non-copulatory behavioral interactions resulted in a rapid decrease in the numbers of neurons containing GnRH-ir peptide. This change was accompanied by an increase in the GnRH-ir fibers in the median eminence, but no surge in luteinizing hormone. These data suggest that behavioral interactions stimulate release of mature GnRH peptide from cell bodies followed by accumulation of available GnRH in cell terminals. Copulation triggers increased production of proGnRH in cell bodies. The data highlight the usefulness of behavioral paradigms for the examination of the dynamics of neuropeptide production.

5-Hydroxy-L-tryptophan (5-HTP) and L-3,4-dihydroxyphenylalanine (L-DOPA) were injected intraperitoneally (i.p.) into the laboratory shrew (Suncus murinus). Immunocytochemical and immunofluorescence studies were carried out on serial or same sections of the brain, which were reacted with specific antisera to dopamine (DA) or serotonin (5-HT) produced in our laboratory. We observed that cell bodies and nerve terminals of many catecholaminergic (CAnergic) neurons exhibited 5-HTP uptake and conversion of the precursor into 5-HT. However, the locus ceruleus showed scarcely any 5-HT immunoreactivity. This suggests that the precursor uptake mechanism may be different among various CAnergic groups. In contrast to these findings on CAnergic neurons, all serotoninergic (5-HTnergic) neurons after L-DOPA administration showed DA immunopositive reaction in their cell bodies and nerve terminals, suggesting that 5-HTnergic neurons may have the same capacity for precursor uptake. On the other hand, we observed that all aromatic L-amino acid decarboxylase (AADC)-only-positive neuron groups showed both DA and 5-HT immunoreactions after L-DOPA and 5-HTP administration, respectively, in the double-staining immunofluorescence method. From these results, AADC-only-positive neurons may be considered to belong to the amine precursor uptake and decarboxylation (APUD) system.

After 5-hydroxy-L-tryptophan (5-HTP) and L-3,4-dihydroxyphenylalanine (L-DOPA) were injected i.p. in the laboratory shrew Suncus murinus, immunocytochemical and immunofluorescence studies were conducted on continuous or same sections of the brain, using specific anti-tyrosine hydroxylase (TH), anti-aromatic L-amino acid decarboxylase (AADC), anti-dopamine (DA) and anti-serotonin (5-HT) antisera which were produced in our laboratory. The results of double-staining by the immunofluorescence method as well as immunoelectron microscopy strongly indicate that the cells of the premammillary nucleus of the laboratory shrew brain (AADC-only-positive neurons) are capable of synthesizing DA and 5-HT simultaneously upon simultaneous administration of L-dopa and 5-HTP.
