Identification of individual single wall nanotubes (SWNTs) within a cellular structure can provide vital information towards understanding the potential mechanisms of uptake, their localisation and whether their structure is transformed within a cell. To be able to image an individual SWNT in such an environment a resolution is required that is not usually appropriate for biological sections. Standard transmission electron microscopy (TEM) techniques such as bright field imaging of these cellular structures result in very weak contrast. Traditionally, researchers have stained the cells with heavy metal stains to enhance the cellular structure, however this can lead to confusion when analysing the samples at high resolution. Subsequently, alternative methods have been investigated to allow high resolution imaging and spectroscopy to identify SWNTs within the cell; here we will concentrate on the sample preparation and experimental methods used to achieve such resolution., (Copyright 2009 Elsevier B.V. Multi-walled carbon nanotubes (MWNTs) have been proposed for use in many applications and concerns about their potential effect on human health have led to the interest in understanding the interactions between MWNTs and human cells. One important technique is the visualisation of the intracellular distribution of MWNTs. We exposed human macrophage cells to unpurified MWNTs and found that a decrease in cell viability was correlated with uptake of MWNTs due to mainly necrosis. Cells treated with purified MWNTs and the main contaminant Fe(2)O(3) itself yielded toxicity only from the nanotubes and not from the Fe(2)O(3). We used 3-D dark-field scanning transmission electron microscopy (DF-STEM) tomography of freeze-dried whole cells as well as confocal and scanning electron microscopy (SEM) to image the cellular uptake and distribution of unpurified MWNTs. We observed that unpurified MWNTs entered the cell both actively and passively frequently inserting through the plasma membrane into the cytoplasm and the nucleus. These suggest that MWNTs may cause incomplete phagocytosis or mechanically pierce through the plasma membrane and result in oxidative stress and cell death.

Water-soluble single-walled nanotubes (SWNTs) are being tested as contrast agents for medical imaging and for the delivery of therapeutically active molecules to target cells. However, before they become used commercially, it will be essential to establish their subcellular distribution and whether they are cytotoxic. Here we characterize uptake of unlabeled, acid-treated, water-soluble SWNTs by human monocyte derived macrophage cells using a combination of Raman spectroscopy and analytical electron microscopy and compare our findings to previous work on unpurified SWNTs. Raman spectroscopy demonstrated that acid-treated SWNTs had a greater number of functional groups on the carbon walls than nontreated SWNT. The acid-treated SWNTs were less aggregated within cells than unpurified SWNTs. Bundles, and also individual acid-treated SWNTs, were found frequently inside lysosomes and also the cytoplasm, where they caused no significant changes in cell viability or structure even after 4 days of exposure.

Multi-walled carbon nanotubes were synthesized by chemical vapor deposition from pure toluene and toluene/diazine mixtures using ferrocene as a catalyst precursor at 760 degrees C. As recently announced, characterization of the resulting nanotube films showed that, unlike pure carbon nanotubes, those grown in the presence of nitrogen have an extremely high degree of internal order, both in terms of the uniform chirality in the nanotube walls and of the crystallographic register between them. Here, the structure, defects, and morphology of the nanotubes were analyzed in depth using advanced electron microscopy techniques, and compared with existing models and observations. Nitrogen, which seems to be responsible for the dramatic structural order, was found to segregate preferentially within the core of the nanotubes.
