Chemical contaminants are becoming an increasingly greater concern for water quality and it is well known that interactions with geochemical interfaces impact the fate and transport of these contaminants in the environment. In this study, we investigated the interactions of one such chemical contaminant, monoethanolamine (MEA), with oxide surfaces, particularly titanium dioxide (TiO 2 ) and iron oxide (α-Fe 2 O 3 ). Attenuated Total Reflectance Fourier Transform Infrared (ATR-FTIR) spectroscopy was used to probe the adsorption behavior of MEA on titanium dioxide (TiO 2 ) and iron oxide (α-Fe 2 O 3 ) nanoparticles as a function of pH and other environmental conditions including concentration and ionic strength. Both the extent and initial rates of adsorption of MEA on these oxide surfaces increases with increasing pH. Adsorption on these oxide surfaces increases with solution concentration until saturation occurs and MEA adsorbs more readily at higher pH. Furthermore, adsorption decreases with increasing ionic strength, demonstrating the importance of electrostatic interactions to this process. Based on these results, a mechanistic picture emerges for the interaction of MEA with titanium dioxide and iron oxide across a range of pH values. Stem cell therapy in heart disease is challenged by mis-injection, poor survival, and low cell retention. Here, we describe a biocompatible multifunctional silica-iron oxide nanoparticle to help solve these issues. The nanoparticles were made via an in situ growth of Fe 3 O 4 nanoparticles on both the external surfaces and pore walls of mesocellular foam silica nanoparticles. In contrast to previous work, this approach builds a magnetic moiety inside the pores of a porous silica structure. These materials serve three roles: drug delivery, magnetic manipulation, and imaging. The addition of Fe 3 O 4 to the silica nanoparticles increased their colloidal stability, T 2 -based magnetic resonance imaging contrast, and superparamagnetism. We then used the hybrid materials as a sustained release vehicle of insulin-like growth factor-a pro-survival agent that can increase cell viability. In vivo rodent studies show that labeling stem cells with this nanoparticle increased the efficacy of stem cell therapy in a ligation/reperfusion model. The nanoparticle-labeled cells increase the mean left ventricular ejection fraction by 11 and 21% and the global longitudinal strain by 24 and 34% on days 30 and 60, respectively. In summary, this multifunctional nanomedicine improves stem cell survival via the sustained release of pro-survival agents.

Bovine serum albumin (BSA) adsorbed on amorphous silicon dioxide (SiO 2 ) nanoparticles was studied as a function of pH across the range of 2 to 8. Aggregation, surface charge, surface coverage, and protein structure were investigated over this entire pH range. SiO 2 nanoparticle aggregation is found to depend upon pH and differs in the presence of adsorbed BSA. For SiO 2 nanoparticles truncated with hydroxyl groups, the largest aggregates were observed at pH 3, close to the isoelectric point of SiO 2 nanoparticles, whereas for SiO 2 nanoparticles with adsorbed BSA, the aggregate size was the greatest at pH 3.7, close to the isoelectric point of the BSA-SiO 2 complex. Surface coverage of BSA was also the greatest at the isoelectric point of the BSA-SiO 2 complex with a value of ca. 3 ±   1 × 10 11 molecules cm -2 . Furthermore, the secondary protein structure was modified when compared to the solution phase at all pH values, but the most significant differences were seen at pH 7.4 and below. It is concluded that protein-nanoparticle interactions vary with solution pH, which may have implications for nanoparticles in different biological fluids (e.g., blood, stomach, and lungs).

The interaction of a model protein, bovine serum albumin (BSA) with two different metal oxide nanoparticles, TiO 2 (∼22nm) and SiO 2 (∼14nm), was studied at both physiological and acidic pH. The pH- and nanoparticle-dependent differences in protein structure and protein adsorption were determined using attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) and thermogravimetric analysis (TGA). The results indicated that the surface coverage of BSA decreases with decreasing pH on both TiO 2 and SiO 2 surfaces, and BSA coverage is higher by a factor of ca. 3-10times more on TiO 2 compared to SiO 2 . The secondary structure of BSA changes upon adsorption to either nanoparticle surface at both pH 7.4 and 2. At acidic pH, BSA appears to completely unfold on TiO 2 nanoparticles whereas it assumes an extended conformation on SiO 2 . These differences highlight for the first time the extent to which the protein corona structure is significantly impacted by protein-nanoparticle interactions which depend on the interplay between pH and specific nanoparticle surface chemistry., (Copyright © 2017 Elsevier Inc. Understanding complex chemical changes that take place at nano-bio interfaces is of great concern for being able to sustainably implement nanomaterials in key applications such as drug delivery, imaging, and environmental remediation. Typical in vitro assays use cell viability as a proxy to understanding nanotoxicity but often neglect how the nanomaterial surface can be altered by adsorption of solution-phase components in the medium. Protein coronas form on the nanomaterial surface when incubated in proteinaceous solutions. Herein, we apply a broad array of techniques to characterize and quantify protein corona formation on silica nanoparticle surfaces. The porosity and surface chemistry of the silica nanoparticles have been systematically varied. Using spectroscopic tools such as FTIR and circular dichroism, structural changes and kinetic processes involved in protein adsorption were evaluated. Additionally, by implementing thermogravimetric analysis, quantitative protein adsorption measurements allowed for the direct comparison between samples. Taken together, these measurements enabled the extraction of useful chemical information on protein binding onto nanoparticles in solution. Overall, we demonstrate that small alkylamines can increase protein adsorption and that even large polymeric molecules such as poly(ethylene glycol) (PEG) cannot prevent protein adsorption in these systems. The implications of these results as they relate to further understanding nano-bio interactions are discussed.

Nanoparticles in biological media form dynamic entities as a result of competitive adsorption of proteins on nanoparticle surfaces called protein coronas. The protein affinity toward nanoparticle surfaces potentially depends on the constituent amino acid side chains which are on the protein exterior and thus exposed to the solution and available for interaction. Therefore, studying the adsorption of individual amino acids on nanoparticle surfaces can provide valuable insights into the overall evolution of nanoparticles in solution and the protein corona that forms. In the current study, the surface adsorption of l-histidine on TiO2 nanoparticles with a diameter of 5 nm at pH 7.4 (physiological pH) is studied from both macroscopic and molecular perspectives. Quantitative adsorption measurements of l-histidine on 5 nm TiO2 particles yield maximum adsorption coverage of 6.2 ± 0.3 × 10(13) molecules cm(-2) at 293 K and pH 7.4. These quantitative adsorption measurements also yield values for the equilibrium constant and free energy of adsorption of K = 4.3 ± 0.5 × 10(2) L mol(-1) and ΔG = -14.8 ± 0.3 kJ mol(-1), respectively. Detailed analysis of the adsorption between histidine and 5 nm TiO2 nanoparticle surfaces with attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy indicates both the imidazole side chain and the amine group interacting with the nanoparticle surface and the adsorption to be reversible. The adsorption results in no change in surface charge and therefore does not change nanoparticle-nanoparticle interactions and thus aggregation behavior of these 5 nm TiO2 nanoparticles in aqueous solution.

Background: Although ZnO nanoparticles (NPs) are used in many commercial products and the potential for human exposure is increasing, few in vivo studies have addressed their possible toxic effects after inhalation. We sought to determine whether ZnO NPs induce pulmonary toxicity in mice following sub-acute or sub-chronic inhalation exposure to realistic exposure doses., Methods: Mice (C57Bl/6) were exposed to well-characterized ZnO NPs (3.5 mg/m3, 4 hr/day) for 2 (sub-acute) or 13 (sub-chronic) weeks and necropsied immediately (0 wk) or 3 weeks (3 wks) post exposure. Toxicity was assessed by enumeration of total and differential cells, determination of total protein, lactate dehydrogenase activity and inflammatory cytokines in bronchoalveolar lavage (BAL) fluid as well as measurements of pulmonary mechanics. Generation of reactive oxygen species was assessed in the lungs. Lungs were evaluated for histopathologic changes and Zn content. Zn concentration in blood, liver, kidney, spleen, heart, brain and BAL fluid was measured., Results: An elevated concentration of Zn2+ was detected in BAL fluid immediately after exposures, but returned to baseline levels 3 wks post exposure. Dissolution studies showed that ZnO NPs readily dissolved in artificial lysosomal fluid (pH 4.5), but formed aggregates and precipitates in artificial interstitial fluid (pH 7.4). Sub-acute exposure to ZnO NPs caused an increase of macrophages in BAL fluid and a moderate increase in IL-12(p40) and MIP-1α, but no other inflammatory or toxic responses were observed. Following both sub-acute and sub-chronic exposures, pulmonary mechanics were no different than sham-exposed animals., Conclusions: Our ZnO NP inhalation studies showed minimal pulmonary inflammation, cytotoxicity or lung histopathologic changes. An elevated concentration of Zn in the lung and BAL fluid indicates dissolution of ZnO NPs in the respiratory system after inhalation. Exposure concentration, exposure mode and time post exposure played an important role in the toxicity of ZnO NPs. Exposure for 13 wks with a cumulative dose of 10.9 mg/kg yielded increased lung cellularity, but other markers of toxicity did not differ from sham-exposed animals, leading to the conclusion that ZnO NPs have low sub-chronic toxicity by the inhalation route.

Given the importance of nanoparticle surface composition in nanotoxicology, analytical tools that can probe nanoparticle surfaces in aqueous media are crucial but remain limited. Attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy is a technique capable of in situ characterization of the liquid-solid interface to probe surface adsorption on nanoparticle surfaces in environmentally and biologically relevant media. Furthermore, given that the interfacial region in these media is dynamic, ATR-FTIR spectroscopy facilitates monitoring these dynamics by interrogating a layer of immobilized nanoparticles coated on the ATR element while changing the overlying aqueous phase. The molecular information acquired from this technique allows for the determination of the adsorption mode, including conformational and structural changes of the coordinating ligand, and can directly measure ligand displacement reactions. Furthermore, in some cases, ATR-FTIR spectroscopy can be used as a quantitative surface analytical tool. In this article, we briefly review the fundamentals of the technique and then provide several examples of using ATR-FTIR spectroscopy to probe nanoparticle surfaces in general with respect to: (i) the adsorption of different environmentally and biologically relevant coordinating ligands; (ii) competitive ligand adsorption and; (iii) the determination of kinetic and thermodynamic parameters. We have also investigated surface adsorption of TiO2 nanoparticles in different biological media typically used for toxicity studies and show that the surface composition of TiO2 nanoparticles depends to a large extent on the composition of the medium due to surface adsorption. This result has important implications for the interpretation of toxicity data as well as inter-comparisons between toxicity studies.

Copper nanomaterials are being used in a large number of commercial products because these materials exhibit unique optical, magnetic, and electronic properties. Metallic copper nanoparticles, which often have a thin surface oxide layer, can age in the ambient environment and become even more oxidized over time. These aged nanoparticles will then have different properties compared to the original nanoparticles. In this study, we have characterized three different types of copper-based nanoparticle (NP) samples designated as Cu(new) NPs, Cu(aged) NPs, and CuO NPs that differ in the level of oxidation. The solution phase behavior of these three copper-based nanoparticle samples is investigated as a function of pH and in the presence and absence of two common, complexing organic acids, citric and oxalic acid. The behavior of these three copper-based NP types shows interesting differences. In particular, Cu(aged) NPs exhibit unique chemistry including oxide phases that form and surface adsorption properties. Overall, the current study provides some insights into the impacts of nanoparticle aging and how the physicochemical characteristics and reactivity of nanomaterials can change upon aging.

Background: There is increasing interest in the environmental and health consequences of silver nanoparticles as the use of this material becomes widespread. Although human exposure to nanosilver is increasing, only a few studies address possible toxic effect of inhaled nanosilver. The objective of this study was to determine whether very small commercially available nanosilver induces pulmonary toxicity in mice following inhalation exposure., Results: In this study, mice were exposed sub-acutely by inhalation to well-characterized nanosilver (3.3 mg/m³, 4 hours/day, 10 days, 5 ± 2 nm primary size). Toxicity was assessed by enumeration of total and differential cells, determination of total protein, lactate dehydrogenase activity and inflammatory cytokines in bronchoalveolar lavage fluid. Lungs were evaluated for histopathologic changes and the presence of silver. In contrast to published in vitro studies, minimal inflammatory response or toxicity was found following exposure to nanosilver in our in vivo study. The median retained dose of nanosilver in the lungs measured by inductively coupled plasma-optical emission spectroscopy (ICP-OES) was 31 μg/g lung (dry weight) immediately after the final exposure, 10 μg/g following exposure and a 3-wk rest period and zero in sham-exposed controls. Dissolution studies showed that nanosilver did not dissolve in solutions mimicking the intracellular or extracellular milieu., Conclusions: Mice exposed to nanosilver showed minimal pulmonary inflammation or cytotoxicity following sub-acute exposures. However, longer term exposures with higher lung burdens of nanosilver are needed to ensure that there are no chronic effects and to evaluate possible translocation to other organs.

Background: Nanotechnology offers great promise in many industrial applications. However, little is known about the health effects of manufactured nanoparticles, the building blocks of nanomaterials., Objectives: Titanium dioxide (TiO(2)) nanoparticles with a primary size of 2-5 nm have not been studied previously in inhalation exposure models and represent some of the smallest manufactured nanoparticles. The purpose of this study was to assess the toxicity of these nanoparticles using a murine model of lung inflammation and injury., Materials and Methods: The properties of TiO(2) nanoparticles as well as the characteristics of aerosols of these particles were evaluated. Mice were exposed to TiO(2) nanoparticles in a whole-body exposure chamber acutely (4 hr) or subacutely (4 hr/day for 10 days). Toxicity in exposed mice was assessed by enumeration of total and differential cells, determination of total protein, lactate dehydrogenase (LDH) activity and inflammatory cytokines in bronchoalveolar lavage (BAL) fluid. Lungs were also evaluated for histopathologic changes, Results: Mice exposed acutely to 0.77 or 7.22 mg/m(3) nanoparticles demonstrated minimal lung toxicity or inflammation. Mice exposed subacutely (8.88 mg/m(3)) and necropsied immediately and at week 1 or 2 postexposure had higher counts of total cells and alveolar macrophages in the BAL fluid compared with sentinels. However, mice recovered by week 3 postexposure. Other indicators were negative., Conclusions: Mice subacutely exposed to 2-5 nm TiO(2) nanoparticles showed a significant but moderate inflammatory response among animals at week 0, 1, or 2 after exposure that resolved by week 3 postexposure.

Abstract Background Human exposure to nanoparticles (NPs) and environmental bacteria can occur simultaneously. NPs induce inflammatory responses and oxidative stress but may also have immune-suppressive effects, impairing macrophage function and altering epithelial barrier functions. The purpose of this study was to assess the potential pulmonary effects of inhalation and instillation exposure to copper (Cu) NPs using a model of lung inflammation and host defense. Methods We used Klebsiella pneumoniae (K.p.) in a murine lung infection model to determine if pulmonary bacterial clearance is enhanced or impaired by Cu NP exposure. Two different exposure modes were tested: sub-acute inhalation (4 hr/day, 5 d/week for 2 weeks, 3.5 mg/m3) and intratracheal instillation (24 hr post-exposure, 3, 35, and 100 μg/mouse). Pulmonary responses were evaluated by lung histopathology plus measurement of differential cell counts, total protein, lactate dehydrogenase (LDH) activity, and inflammatory cytokines in bronchoalveolar lavage (BAL) fluid. Results Cu NP exposure induced inflammatory responses with increased recruitment of total cells and neutrophils to the lungs as well as increased total protein and LDH activity in BAL fluid. Both inhalation and instillation exposure to Cu NPs significantly decreased the pulmonary clearance of K.p.-exposed mice measured 24 hr after bacterial infection following Cu NP exposure versus sham-exposed mice also challenged with K.p (1.4 × 105 bacteria/mouse). Conclusions Cu NP exposure impaired host defense against bacterial lung infections and induced a dose-dependent decrease in bacterial clearance in which even our lowest dose demonstrated significantly lower clearance than observed in sham-exposed mice. Thus, exposure to Cu NPs may increase the risk of pulmonary infection.

There is great concern regarding the adverse health implications of engineered nanoparticles. However, there are many circumstances where the production of incidental nanoparticles, i.e., nanoparticles unintentionally generated as a side product of some anthropogenic process, is of even greater concern. In this study, metal-based incidental nanoparticles were measured in two occupational settings: a machining center and a foundry. On-site characterization of substrate-deposited incidental nanoparticles using a field-portable X-ray fluorescence provided some insights into the chemical characteristics of these metal-containing particles. The same substrates were then used to carry out further off-site analysis including single-particle analysis using scanning electron microscopy and energy-dispersive X-ray spectroscopy. Between the two sites, there were similarities in the size and composition of the incidental nanoparticles as well as in the agglomeration and coagulation behavior of nanoparticles. In particular, incidental nanoparticles were identified in two forms: submicrometer fractal-like agglomerates from activities such as welding and supermicrometer particles with incidental nanoparticles coagulated to their surface, herein referenced as nanoparticle collectors. These agglomerates will affect deposition and transport inside the respiratory system of the respirable incidental nanoparticles and the corresponding health implications. There is an increasing need to evaluate concentrations of nanoparticles in occupational settings due to their potential negative health effects. The Nanoparticle Respiratory Deposition (NRD) personal sampler was developed to collect nanoparticles separately from larger particles in the breathing zone of workers, while simultaneously providing a measure of respirable mass concentration. This study compared concentrations measured with the NRD sampler to those measured with a nano Micro Orifice Uniform-Deposit Impactor (nanoMOUDI) and respirable samplers in three workplaces. The NRD sampler performed well at two out of three locations, where over 90% of metal particles by mass were submicrometer particle size (a heavy vehicle machining and assembly facility and a shooting range). At the heavy vehicle facility, the mean metal mass concentration of particles collected on the diffusion stage of the NRD was 42.5 ± 10.0 µg/m3, within 5% of the nanoMOUDI concentration of 44.4 ± 7.4 µg/m3. At the shooting range, the mass concentration for the diffusion stage of the NRD was 5.9 µg/m3, 28% above the nanoMOUDI concentration of 4.6 µg/m3. In contrast, less favorable results were obtained at an iron foundry, where 95% of metal particles by mass were larger than 1 µm. The accuracy of nanoparticle collection by NRD diffusion stage may have been compromised by high concentrations of coarse particles at the iron foundry, where the NRD collected almost 5-fold more nanoparticle mass compared to the nanoMOUDI on one sampling day and was more than 40% different on other sampling days. The respirable concentrations measured by NRD samplers agreed well with concentrations measured by respirable samplers at all sampling locations. Overall, the NRD sampler accurately measured concentrations of nanoparticles in industrial environments when concentrations of large, coarse mode, particles were low. [ABSTRACT FROM AUTHOR]

Titanium dioxide (TiO2) particles, including nanoparticles with diameters smaller than 100 nm, are used extensively in consumer products. In a 2011 current intelligence bulletin, the National Institute of Occupational Safety and Health (NIOSH) recommended methods to assess worker exposures to fine and ultrafine TiO2 particles and associated occupational exposure limits for these particles. However, there are several challenges and problems encountered with these recommended exposure assessment methods involving the accurate quantitation of titanium dioxide collected on air filters using acid digestion followed by inductively coupled plasma optical emission spectroscopy (ICP-OES). Specifically, recommended digestion methods include the use of chemicals, such as perchloric acid, which are typically unavailable in most accredited industrial hygiene laboratories due to highly corrosive and oxidizing properties. Other alternative methods that are used typically involve the use of nitric acid or combination of nitric acid and sulfuric acid, which yield very poor recoveries for titanium dioxide. Therefore, given the current state of the science, it is clear that a new method is needed for exposure assessment. In this current study, a microwave-assisted acid digestion method has been specifically designed to improve the recovery of titanium in TiO2 nanoparticles for quantitative analysis using ICP-OES. The optimum digestion conditions were determined by changing several variables including the acids used, digestion time, and temperature. Consequently, the optimized digestion temperature of 210°C with concentrated sulfuric and nitric acid (2:1 v/v) resulted in a recovery of >90% for TiO2. A granular bed was designed to collect nanoparticles as an alternative to nylon mesh screens for use in a nanoparticle respiratory deposition (NRD) sampler. The granular bed consisted of five layers in series: a coarse mesh, a large-bead layer, a small-bead layer, a second large-bead layer, and a second coarse mesh. The bed was designed to primarily collect particles in the small-bead layer, with the coarse mesh and large-bead layers designed to hold the collection layer in position. The collection efficiency of the granular bed was measured for varying depths of the small-bead layer and for test particles with different shape (cuboid, salt particles; and fractal, and stainless steel and welding particles). Experimental measurements of collection efficiency were compared to estimates of efficiency from theory and to the nanoparticulate matter (NPM) criterion, which was established to reflect the total deposition in the human respiratory system for particles smaller than 300 nm. The shape of the collection efficiency curve for the granular bed was similar to the NPM criterion in these experiments. The collection efficiency increased with increasing depth of the small-bead layer: the particle size associated with 50% collection efficiency,d50, for salt particles was 25 nm for a depth of 2.2 mm, 35 nm for 3.2 mm, and 45 nm for 4.3 mm. The best-fit to the NPM criterion was found for the bed with a small-bead layer of 3.2 mm. Compared to cubic salt particles, the collection efficiency was higher for fractal-shaped particles larger than 50 nm, presumably due to increased interception. Despite their potential for a variety of applications, copper nanoparticles induce very strong inflammatory responses and cellular toxicity following aerosolized delivery. Coating metallic nanoparticles with polysaccharides, such as biocompatible and antimicrobial chitosan, has the potential to reduce this toxicity. In this study, copper nanoparticles were coated with chitosan using a newly developed and facile method. The presence of coating was confirmed using x-ray photoelectron spectroscopy, rhodamine tagging of chitosan followed by confocal fluorescence imaging of coated particles and observed increases in particle size and zeta potential. Further physical and chemical characteristics were evaluated using dissolution and x-ray diffraction studies. The chitosan coating was shown to significantly reduce the toxicity of copper nanoparticles after 24 and 52 h and the generation of reactive oxygen species as assayed by DHE oxidation after 24 h in vitro. Conversely, inflammatory response, measured using the number of white blood cells, total protein, and cytokines/chemokines in the bronchoalveolar fluid of mice exposed to chitosan coated versus uncoated copper nanoparticles, was shown to increase, as was the concentration of copper ions. These results suggest that coating metal nanoparticles with mucoadhesive polysaccharides (e.g. chitosan) could increase their potential for use in controlled release of copper ions to cells, but will result in a higher inflammatory response if administered via the lung. Citric acid plays an important role as a stabilizer in several nanomaterial syntheses and is a common organic acid found in nature. Here, the adsorption of citric acid onto TiO2 anatase nanoparticles with a particle diameter of ca. 4 nm is investigated at circumneutral and acidic pHs. This study focuses on both the details of the surface chemistry of citric acid on TiO2, including measurements of surface coverage and speciation, and its impact on nanoparticle behavior. Using macroscopic and molecular-based probes, citric acid adsorption and nanoparticle interactions are measured with quantitative solution phase adsorption measurements, attenuated total reflection-FTI R spectroscopy, dynamic light scattering techniques, and zeta-potential measurements as a function of solution pH. The results show that surface coverage is a function of pH and decreases with increasing pH. Surface speciation differs from the bulk solution and is time dependent. After equilibration, the fully deprotonated citrate ion is present on the surface regardless of the highly acidic solution pH indicating pKa values of surface adsorbed species are lower than those in solution. Nanoparticle interactions are also probed through measurements of aggregation and the data show that these interactions are complex and depend on the detailed interplay between bulk solution pH and surface chemistry. The increasing use of manufactured nanoparticles ensures these materials will make their way into the environment. Silver nanoparticles in particular, due to use in a wide range of applications, have the potential to get into water systems, e.g., drinking water systems, ground water systems, estuaries, and/or lakes. One important question is what is the chemical and physical state of these nanoparticles in water? Are they present as isolated particles, agglomerates or dissolved ions, as this will dictate their fate and transport. Furthermore, does the chemical and physical state of the nanoparticles change as a function of size or differ from micron-sized particles of similar composition? In this study, an electrospray atomizer coupled to a scanning mobility particle sizer (ES-SMPS) is used to investigate the state of silver nanoparticles in water and aqueous nitric acid environments. Over the range of pH values investigated, 0.5–6.5, silver nanoparticles with a bimodal primary particle size distribution with the most intense peak at 5.0 ± 7.4 nm, as determined from transmission electron microscopy (TEM), show distinct size distributions indicating agglomeration between pH 6.5 and 3 and isolated nanoparticles at pH values from 2.5 to 1. At the lowest pH investigated, pH 0.5, there are no peaks detected by the SMPS, indicating complete nanoparticle dissolution. Further analysis of the solution shows dissolved Ag ions at a pH of 0.5. Interestingly, silver nanoparticle dissolution shows size dependent behavior as larger, micron-sized silver particles show no dissolution at this pH. Environmental and health studies on nanomaterials are appearing in the literature at a rapid pace. These studies will address important issues related to the environmental health and safety (EHS) of nanomaterials. As noted in many recent workshop and agency reports, studies devoted toward the environmental fate and transport, nanomaterial–biological interactions, toxicity, and overall risk assessment of nanomaterials should have nanomaterial characterization as a central component of the study design. This aspect of the study design is necessary so that risks associated with nanomaterials can be fully understood and related to specific material properties. For studies that use commercially manufactured nanomaterials, the company often provides characterization data (e.g., chemical composition, phase, and size) of the purchased materials. One question is, how good are these data? Another is, what methods of analysis are used to characterize the properties of commercial nanomaterials? In the present study, some examples are presented that show marked differences between independent characterization of commercially manufactured nanomaterials and that provided by the company. Furthermore, information provided by the manufacturer may be incomplete and nonrepresentative of the entire sample and, in some cases, the information can, in fact, be wrong. Inhalation toxicity studies of nanoparticles require production of test aerosols with a number of acceptable characteristics. Five different generation methods for producing a nanoparticle aerosol from the bulk powder were evaluated with the goal of producing an acceptable aerosol that is homogenous, has a consistent concentration over time in which the magnitude may be intentionally varied, and has a size distribution that is both unimodal and has a small geometric mean diameter relative to the primary particle size of the nanoparticle bulk powder. Four types of bulk powders were evaluated including titanium dioxide, silicon dioxide, polymer-coated silver, and single walled carbon nanotubes. A nebulizer was the only tested method able to produce a consistent aerosol of sufficient magnitude. However, the resulting aerosol contains particles derived from the carrier water. Prior sonication of the nanoparticle suspension alone was found to be not useful for deagglomerating nanoparticles. [ABSTRACT FROM AUTHOR]

We examined pulmonary inflammatory responses of mice following whole-body inhalation exposure to copper and iron nanoparticles in acute and sub-acute studies. Concentrations for sub-acute copper and iron exposures were 3.6 mg m-3. No significant pathology was found following acute exposure. Immediately following sub-acute exposure, both iron- and copper-exposed mice showed increased inflammation compared to sentinels. Copper nanoparticle-exposed mice had significantly higher lavage cytokines as well as perivasculitis and alveolitis. Three weeks post-exposure, all inflammatory markers decreased for iron nanoparticle-exposed mice, however, some remained elevated for copper-exposed mice. At biologically relevant pHs, in vitro studies showed that copper nanoparticles displayed a greater propensity for dissolution compared to iron. We conclude that the presence of dissolved ions, the concomitant formation of smaller nanoparticles and the absence of particles in stained lung sections immediately postexposure (inferring either translocation or more dispersed aerosol distribution) contributed to the increased inflammation observed in copper nanoparticle-exposed mice. TiO2 is a widely used manufactured nanomaterial and the opportunity for human exposure makes it necessary to study its health implications. Using murine models for inflammation, size effects of inflammatory response in instillation and acute inhalation exposures of TiO2 nanoparticles with manufacturers' average particles sizes of 5 and 21 nm were investigated. The properties of the primary nanoparticles, nanoparticle agglomerates aerosol and instillation solution for both sized nanoparticles were evaluated. Mice were acutely exposed in a whole-body exposure chamber or through nasal instillation and toxicity was assessed by enumeration of total and differential cells, determination of total protein, LDH activity and inflammatory cytokines in BAL fluid. Lungs were also evaluated for histopathological changes. Results show the larger TiO2 nanoparticles were found to be moderately, but significantly, more toxic. The nanoparticles had different agglomeration states which may be a factor as important as the surface and physical characteristics of the primary nanoparticles in determining toxicity. Environmental molecular surface science is an expanding area of current research. This review focuses on advances in the molecular level understanding of oxide surfaces as they play an important role in several environmental chemical processes. Oxide surfaces are often used as catalysts and adsorbents in environmental remediation. The surface of oxide particles in the troposphere can adsorb and catalyze reactions of trace gases and thus change the chemical balance of the atmosphere. Mineral oxide surfaces in contact with ground water can adsorb and catalyze reactions of pollutant molecules. Surface science studies of these environmental chemical processes provide the basis for delineating molecular-level information about surface structure under environmentally relevant conditions, adsorbate–surface interactions, surface reaction mechanisms, structure–reactivity relationships and an overall understanding of these processes on the molecular level. In order to glean insights into environmental processes, these studies need to be done under environmentally relevant conditions of temperature, pressure and relative humidity. Thus the need for the further development and use of techniques that can be employed under ambient conditions are discussed. [Copyright &y& Elsevier]

Abstract: In this study, in situ transmission FT-IR spectroscopy was employed to investigate the thermal and selective catalytic reduction of NO2 with propylene in nanocrystalline NaY zeolite. NO was the major product in the thermal reduction of NO2. In absence of oxygen and water, selective catalytic reduction of NO2 with propylene at low temperatures (T ≤473K) resulted in the complete reduction of NO2 to N2 and O2. Isocyanate and nitrile species formed on extra framework alumina sites are found to be important intermediates. Based on kinetic studies, a pathway involving radical reactions appears to be the most plausible mechanism for the complete reduction of NO2. [Copyright &y& Elsevier]

Inhalation of nanoparticles has been implicated in respiratory morbidity and mortality. In particular, carbon black nanoparticles are found in many different environmental exposures. Macrophages take up inhaled nanoparticles and respond via release of inflammatory mediators and in some cases cell death. Based on new data, we propose that exposure of macrophages (both a macrophage cell line and primary human alveolar macrophages) to carbon black nanoparticles induces pyroptosis, an inflammasome-dependent form of cell death. Exposure of macrophages to carbon black nanoparticles resulted in inflammasome activation as defined by cleavage of caspase 1 to its active form and downstream IL-1β release. The cell death that occurred with carbon black nanoparticle exposure was identified as pyroptosis by the protective effect of a caspase 1 inhibitor and a pyroptosis inhibitor. These data demonstrate that carbon black nanoparticle exposure activates caspase 1, increases IL-1β release after LPS priming, and induces the proinflammatory cell death, pyroptosis. The identification of pyroptosis as a cellular response to carbon nanoparticle exposure is novel and relates to environmental and health impacts of carbon-based particulates. [ABSTRACT FROM AUTHOR]

Abstract: In this paper, focused on environmental nanotechnology, we review some recent results for carbon dioxide (CO2) adsorption on hydroxylated Fe2O3, γ-Al2O3, and TiO2 nanoparticle surfaces at 296K as followed by transmission FTIR spectroscopy. In the absence of water vapor (<1% relative humidity, RH), following exposure to CO2 different species formed on the oxide surface due to the presence of adsorption sites with different basicities. While the majority surface species on Fe2O3, γ-Al2O3 is determined to be adsorbed bicarbonate, on TiO2 nanoparticles bidentate carbonate was more prevalent. A carboxylate species was observed on TiO2 nanoparticles under dry conditions as well. When water is present at 40% RH, the nature of the adsorbed CO2 species changed to that of solvated carbonate formation in the adsorbed water layer. Observed initial adsorption rates were calculated from time–course experiments under dry conditions and in the presence of 40% RH. When initial adsorption rates were compared between dry and wet experiments, a larger value was found for dry experiments suggesting that CO2 molecules have to compete for adsorption sites with water on these nanoparticle surfaces. As discussed here, quantum chemical calculations provide some additional insights into CO2 adsorption on hydroxylated metal oxide surfaces in the presence and absence of molecularly adsorbed water.
