Background: functional near-infrared spectroscopy (fNIRS) is an increasingly popular tool to study cortical activity during movement and gait that requires further validation. This study aimed to assess (1) whether fNIRS can detect the difficult-to-measure leg area of the primary motor cortex (M1) and distinguish it from the hand area; and (2) whether fNIRS can differentiate between automatic (i.e., not requiring one's attention) and non-automatic movement processes. Special attention was attributed to systemic artifacts (i.e., changes in blood pressure, heart rate, breathing) which were assessed and corrected by short channels, i.e., fNIRS channels which are mainly sensitive to superficial scalp hemodynamics., Methods: Twenty-three seated, healthy participants tapped four fingers on a keyboard or tapped the right foot on four squares on the floor in a specific order given by a 12-digit sequence (e.g., 434141243212). Two different sequences were executed: a beforehand learned (i.e., automatic) version and a newly learned (i.e., non-automatic) version. A 36-channel fNIRS device including 12 short channels covered multiple motor-related cortical areas including M1. The fNIRS data were analyzed with a general linear model (GLM). Correlation between the expected functional hemodynamic responses (i.e. task regressor) and the short channels (i.e. nuisance regressors), necessitated performing a separate short channel regression instead of integrating them in the GLM., Results: Consistent with the M1 somatotopy, we found significant HbO increases of very large effect size in the lateral M1 channels during finger tapping (Cohen's d = 1.35, p<0.001) and significant HbO increases of moderate effect size in the medial M1 channels during foot tapping (Cohen's d = 0.8, p<0.05). The cortical activity differences between automatic and non-automatic tasks were not significantly different. Importantly, leg movements produced large systemic fluctuations, which were adequately removed by the use of all available short channels., Discussion: Our results indicate that fNIRS is sensitive to leg activity in M1, though the sensitivity is lower than for finger activity and requires rigorous correction for systemic fluctuations. Activity of sensory and motor cortices is essential for sensorimotor integration. In particular, coherence between these areas may indicate binding of critical functions like perception, motor planning, action, or sleep. Evidence is accumulating that cerebellar output modulates cortical activity and coherence, but how, when, and where it does so is unclear. We studied activity in and coherence between S1 and M1 cortices during whisker stimulation in the absence and presence of optogenetic Purkinje cell stimulation in crus 1 and 2 of awake mice, eliciting strong simple spike rate modulation. Without Purkinje cell stimulation, whisker stimulation triggers fast responses in S1 and M1 involving transient coherence in a broad spectrum. Simultaneous stimulation of Purkinje cells and whiskers affects amplitude and kinetics of sensory responses in S1 and M1 and alters the estimated S1-M1 coherence in theta and gamma bands, allowing bidirectional control dependent on behavioral context. These effects are absent when Purkinje cell activation is delayed by 20 ms. Focal stimulation of Purkinje cells revealed site specificity, with cells in medial crus 2 showing the most prominent and selective impact on estimated coherence, i.e., a strong suppression in the gamma but not the theta band. Granger causality analyses and computational modeling of the involved networks suggest that Purkinje cells control S1-M1 phase consistency predominantly via ventrolateral thalamus and M1. Neural entrainment plays a crucial role in perception and action, especially when stimuli possess a certain temporal regularity, and is also suggested to serve as a neural process to select and attend the relevant stream in situations where there are competing stimulus streams. Beneficial effects of entrainment have led to the suggestion that rhythmic stimuli can improve motor function in patients with Parkinson's disease (PD). Behavioural studies support this suggestion, but neurophysiological studies have shown reduced entrainment of motor areas in PD. However, oscillatory entrainment in PD has only been tested in paradigms with a single isochronous stimulus stream, whereas entrainment has an enhanced benefit in situations where one rhythmic stimulus stream has to be segregated from distractor stimuli. Therefore, we here used an intermodal selective attention task with concurrent auditory and visual stimulus streams while recording oscillatory brain activity with Magnetoencephalography (MEG). We aimed to (i) replicate earlier findings of deficient motor entrainment in PD patients in conditions where there is a single stimulus stream, and (ii) to evaluate whether increasing the benefit of entrainment by introducing a distractor stream would lead to entrainment in PD patients not seen otherwise. Contrary to this hypothesis, PD patients showed reduced motor entrainment compared to controls during both conditions, as indexed by beta oscillatory activity. Ethologically inspired models of movement preparation view the sensorimotor system as sampling information from the environment in a parallel fashion in preparation for multiple potential actions. In support, the configuration of the physical workspace, manipulated by the number or spatial separation of potential targets, has been shown to modulate sensorimotor neural activity. It is unclear, however, whether this modulation is driven by the sensory layout of the workspace or through the associated motor plans. Here, we combine a delayed-movement pre-cuing task with visuomotor adaptation to address this question in human subjects while recording MEG. By dissociating visual and motor coordinates of two targets using visuomotor adaptation, the task was designed to evaluate, in a selective fashion, the effects of visual and movement target separation on movement preparatory activity. The results did not allow the intended comparison due to an unanticipated effect of the direction of visuomotor adaptation on baseline oscillatory power in beta and low-gamma bands. Fortuitously, this effect was dependent on whether the adaptation direction decreased or increased the angular separation between alternative movements. That is, there was a sustained reduction of oscillatory power, which was stronger at small compared with large target separation. These results support a direct influence of movement target separation on motor cortex neural activity, mediated by lateral interactions between simultaneously active motor plans. The results further demonstrate a novel effect of visuomotor adaptation on motor cortex oscillatory activity, with properties that support the local nature of learned changes in visuomotor mapping., Significance Statement: There is growing evidence that the motor cortex routinely prepares for different movements simultaneously, each suited to a possible course of events in the immediate environment. The preparatory motor cortex activity for different movements can be seen as a competition between groups of neurons. This competition is influenced by how similar the alternative movements are; for example, in terms of direction, determined by the proximity of alternative movement goals. This study investigates whether the proximity of alternative reach goals has a direct influence on motor cortex activity (in the form of brain oscillations) or if it has an effect only through conscious evaluation of the separation between targets. Choice behavior is influenced by factors such as reward and number of alternatives but also by physical context, for instance, the relative position of alternative movement targets. At small separation, speeded eye or hand movements are more likely to land between targets (spatial averaging) than at larger separation. Neurocomputational models explain such behavior in terms of cortical activity being preshaped by the movement environment. Here, we manipulate target separation, as a determinant of motor cortical activity in choice behavior, to address neural mechanisms of response selection. Specifically, we investigate whether context-induced changes in the balance of cooperative and competitive interactions between competing groups of neurons are expressed in the power spectrum of sensorimotor rhythms. We recorded magnetoencephalography while participants were precued to two possible movement target locations at different angles of separation (30, 60, or 90°). After a delay, one of the locations was cued as the target for a joystick pointing movement. We found that late delay-period movement-preparatory activity increased more strongly for alternative targets at 30 than at 60 or 90° of separation. This nonlinear pattern was evident in slow event-related fields as well as in beta- and low-gamma-band suppression. A comparable pattern was found within an earlier window for theta-band synchronization. We interpret the late delay effects in terms of increased movement-preparatory activity when there is greater overlap and hence less competition between groups of neurons encoding two response alternatives. Local groups of neurons engaged in a cognitive task often exhibit rhythmically synchronized activity in the gamma band, a phenomenon that likely enhances their impact on downstream areas. The efficacy of neuronal interactions may be enhanced further by interareal synchronization of these local rhythms, establishing mutually well timed fluctuations in neuronal excitability. This notion suggests that long-range synchronization is enhanced selectively for connections that are behaviorally relevant. We tested this prediction in the human motor system, assessing activity from bilateral motor cortices with magnetoencephalography and corresponding spinal activity through electromyography of bilateral hand muscles. A bimanual isometric wrist extension task engaged the two motor cortices simultaneously into interactions and coherence with their respective corresponding contralateral hand muscles. One of the hands was cued before each trial as the response hand and had to be extended further to report an unpredictable visual go cue. We found that, during the isometric hold phase, corticomuscular coherence was enhanced, spatially selective for the corticospinal connection that was effectuating the subsequent motor response. This effect was spectrally selective in the low gamma-frequency band (40-47 Hz) and was observed in the absence of changes in motor output or changes in local cortical gamma-band synchronization. These findings indicate that, in the anatomical connections between the cortex and the spinal cord, gamma-band synchronization is a mechanism that may facilitate behaviorally relevant interactions between these distant neuronal groups.

Objective: Investigate the effectiveness and frequency dependence of central drive transmission via the alpha-motoneuron pool to the muscle., Methods: We describe a model for the simulation of alpha-motoneuron firing and the EMG signal as response to central drive input. The transfer in the frequency domain is investigated. Coherence between stochastical central input and EMG is also evaluated., Results: The transmission of central rhythmicities to the EMG signal relates to the spectral content of the latter. Coherence between central input to the alpha-motoneuron pool and the EMG signal is significant whereby the coupling strength hardly depends on the frequency in a range from 1 to 100 Hz. Common central input to pairs of alpha-motoneurons strongly increases the coherence levels. The often-used rectification of the EMG signal introduces a clear frequency dependence., Conclusions: Oscillatory phenomena are strongly transmitted via the alpha-motoneuron pool. The motoneuron firing frequencies do play a role in the transmission gain, but do not influence the coherence levels. Rectification of the EMG signal enhances the transmission gain, but lowers coherence and introduces a strong frequency dependency. Rhythmic synchronization likely subserves interactions among neuronal groups. One of the best studied rhythmic synchronization phenomena in the human nervous system is the beta-band (15-30 Hz) synchronization in the motor system. In this study, we imaged structures across the human brain that are synchronized to the motor system's beta rhythm. We recorded whole-head magnetoencephalograms (MEG) and electromyograms (EMG) of left/right extensor carpi radialis muscle during left/right wrist extension. We analyzed coherence, on the one hand between the EMG and neuronal sources in the brain, and on the other hand between different brain sources, using a spatial filtering approach. Cortico-muscular coherence analysis revealed a spatial maximum of coherence to the muscle in motor cortex contralateral to the muscle in accordance with earlier findings. Moreover, by applying a two-dipole source model, we unveiled significantly coherent clusters of voxels in the ipsilateral cerebellar hemisphere and ipsilateral cerebral motor regions. The spatial pattern of coherence to the right and left arm EMG was roughly mirror reversed across the midline, in agreement with known physiology. Subsequently, we analyzed the brain-wide pattern of beta-band coherence to the motor cortex contralateral to the contracting muscle. This analysis did not reveal any convincing pattern. Because the prior cortico-muscular analysis had demonstrated the expected pattern in our data, this negative finding demonstrates a current limitation of the applied method for cortico-cortical coherence analysis. We conclude that during an isometric muscle contraction, several distributed brain regions form a brain-wide beta-band network for motor control.

Recent research has demonstrated that cortical motor areas are engaged when observing motor actions of others. However, little is known about the possible contribution of the motor system for evaluating the correctness of others' actions. To address this question we designed an MEG experiment in which subjects were executing and observing motor actions with and without errors. In the execution task subjects were asked to make speeded button presses according to instruction cues. During the observation task, they viewed pictures of an actor's hand making button presses which were correct or incorrect according to the cues. Time-frequency representations of the MEG data demonstrated a depression in oscillatory activity in the beta band activity (15-35 Hz) during execution followed by a beta rebound that was stronger for incorrect compared to correct executions. During the observation task, a similar time-course of the beta activity was identified and importantly the modulations were stronger for the observation of incorrect than correct actions. Sources accounting for the difference in beta activity between correct and incorrect actions were localized using a beamforming technique. Both for the execution and observation conditions sources were identified to the dorsal motor areas comprising both primary and pre-motor cortex. Our findings demonstrate that not only is cortical motor activity modulated by action observation, but the modulation increases when the observed action is erroneous. This suggests that the motor system is engaged in evaluating the correctness of the actions of others.

Lateralized magnetic fields were recorded from 12 subjects using a 151 channel magnetoencephalography (MEG) system to investigate temporal and functional properties of motor activation to the observation of goal-directed hand movements by a virtual actor. Observation of left and right hand movements generated a neuromagnetic lateralized readiness field (LRF) over contralateral motor cortex. The early onset of the LRF and the fact that the evoked component was insensitive to the correctness of the observed action suggest the operation of a fast and automatic form of motor resonance that may precede higher levels of action understanding.

Neuronal groups can interact with each other even if they are widely separated. One group might modulate its firing rate or its internal oscillatory synchronization to influence another group. We propose that coherence between two neuronal groups is a mechanism of efficient interaction, because it renders mutual input optimally timed and thereby maximally effective. Modulations of subjects' readiness to respond in a simple reaction-time task were closely correlated with the strength of gamma-band (40 to 70 hertz) coherence between motor cortex and spinal cord neurons. This coherence may contribute to an effective corticospinal interaction and shortened reaction times.

Speaking requires the temporally coordinated planning of core linguistic information, from conceptual meaning to articulation. Recent neurophysiological results suggested that these operations involve a cascade of neural events with subsequent onset times, whilst competing evidence suggests early parallel neural activation. To test these hypotheses, we examined the sources of neuromagnetic activity recorded from 34 participants overtly naming 134 images from 4 object categories (animals, tools, foods and clothes). Within each category, word length and phonological neighbourhood density were co-varied to target phonological/phonetic processes. Multivariate pattern analyses (MVPA) searchlights in source space decoded object categories in occipitotemporal and middle temporal cortex, and phonological/phonetic variables in left inferior frontal (BA 44) and motor cortex early on. The findings suggest early activation of multiple variables due to intercorrelated properties and interactivity of processing, thus raising important questions about the representational properties of target words during the preparatory time enabling overt speaking. Several electrophysiological studies suggest that Parkinson's disease (PD) patients have a reduced tendency to entrain to regular environmental patterns. Here we investigate whether this reduced entrainment concerns a generalized deficit or is confined to movement-related activity, leaving sensory entrainment intact. Magnetoencephalography was recorded during a rhythmic auditory target detection task in 14 PD patients and 14 control subjects. Participants were instructed to press a button when hearing a target tone amid an isochronous sequence of standard tones. The variable pitch of standard tones indicated the probability of the next tone to be a target. In addition, targets were occasionally omitted to evaluate entrainment uncontaminated by stimulus effects. Response times were not significantly different between groups and both groups benefited equally from the predictive value of standard tones. Analyses of oscillatory beta power over auditory cortices showed equal entrainment to the tones in both groups. By contrast, oscillatory beta power and event-related fields demonstrated a reduced engagement of motor cortical areas in PD patients, expressed in the modulation depth of beta power, in the response to omitted stimuli, and in an absent motor area P300 effect. Together, these results show equally strong entrainment of neural activity over sensory areas in controls and patients, but, in patients, a deficient translation of the adjustment to the task rhythm to motor circuits. We suggest that the reduced activation reflects not merely altered resonance to rhythmic external events, but a compromised recruitment of an endogenous response reflecting internal rhythm generation. Abstract: Neurophysiological studies in non-human primates have provided evidence for simultaneous activation of competing responses in the (pre)motor cortex. Human evidence, however, is limited, partly because experimental approaches have often mapped competing responses to paired effectors represented in opposite hemispheres, which restricts the analysis to between-hemisphere comparisons and allows simultaneous execution. A demonstration of competition between different movement plans in the motor cortex is more compelling when simultaneous execution of the alternative responses is ruled out and they are represented in one motor cortex. Therefore, in the current MEG study we have used a unimanual Eriksen flanker paradigm with alternative responses assigned to flexion and extension of the right index finger, activating different direction-sensitive neurons within the finger representation area of the same motor cortex. Results showed that for stimuli eliciting response competition the pre-response motor cortex beta-band (17–29Hz) power decreased stronger than for stimuli that did not induce response competition. Furthermore, response competition elicited an additional pre-response mid-frontal high-gamma band (60–90Hz) power increase. Finally, larger gamma-band effect sizes correlated with greater behavioral response delay induced by response competition. Taken together, our results provide evidence for co-activation of competing responses in the human brain, consistent with evidence from non-human primates. [Copyright &y& Elsevier]
