The goal of this work was to better understand how molybdate and trivalent cations can be used as additives to pH neutral electrolytes to activate the Hydrogen Evolution Reaction (HER). Special emphasis was laid on the chlorate process and therefore also to some of the other effects that the additives may have in that particular process. Cathode films formed from the molybdate and trivalent cations have been investigated with electrochemical and surface analytical methods such as polarization curves, potential sweep, Electrochemical Impedance Spectroscopy (EIS), current efficiency measurements, Scanning Electron Microscope (SEM), Energy-Dispersive X-ray Spectroscopy (EDS), X-ray Photoelectron Spectroscopy (XPS), X-Ray Fluorescence (XRF) and Inductively Coupled Plasma (ICP) analysis. Trivalent cations and molybdate both activate the HER, although in different ways. Ligand water bound to the trivalent cations replaces water as reactant in the HER. Since the ligand water has a lower pKa than free water, it is more easily electrochemically deprotonated than free water and thus catalyzes the HER. Sodium molybdate, on the other hand, is electrochemically reduced on the cathode and form films which catalyze the HER (on cathode materials with poor activity for HER). Molybdate forms films of molybdenum oxides on the electrode surface, while trivalent cation additions form hydroxide films. There is a risk for both types of films that their ohmic resistance increases and the activity of the HER decreases during their growth. Lab-scale experiments show that for films formed from molybdate, these negative effects become less pronounced when the molybdate concentration is reduced. Both types of films can also increase the selectivity of the HER by hindering unwanted side reactions, but none of them as efficiently as the toxic additive Cr(VI) used today in the chlorate process. Trivalent cations are not soluble in chlorate electrolyte and thus not suitable for the chlorate process, whereas molybdate, over a wide pH range, can activate the HER on catalytically poor cathode materials such as titanium. Målsättningen med detta doktorsarbete har varit att bättre förstå hur trivalenta katjoner och molybdat lösta i elektrolyten kan effektivisera elektrokemisk vätgasproduktion. Tillämpningen av dessa tillsatser i kloratprocessen och eventuella sidoeffekter har undersökts. De filmer som bildas på katoden av tillsatserna har undersökts med både elektrokemiska och fysikaliska ytanalysmetoder: polarisationskurvor, potentialsvep, elektrokemisk impedansspektroskopi (EIS), strömutbytesmätningar, svepelektronmikroskopi (SEM), energidispersiv röntgenspektroskopi (EDS), röntgenfotoelektronspektroskopi (XPS), röntgenfluorensens (XRF) och induktivt kopplat plasma (ICP). Både trivalenta katjoner och molybdat kan aktivera elektrokemisk vätgasutveckling, men på olika sätt. Vatten bundet till trivalenta katjoner ersätter fritt vatten som reaktant vid vätgasutveckling. Eftersom vatten bundet till trivalenta katjoner har lägre pKa-värde, går det lättare att producera vätgas från dessa komplex än från fritt vatten. Natriummolybdat däremot reduceras på katoden och bildar filmer som kan katalysera vätgasutvecklingen på substratmaterial som har låg katalytisk aktivitet för reaktionen. Molybdat bildar molybdenoxider på ytan medan trivalenta katjoner bildar metallhydroxider. Båda typerna av film riskerar att bilda filmer som är resistiva och deaktiverar vätgasutvecklingen. Laboratorieexperiment visar att problemen minskar med minskad molybdathalt. Båda filmerna kan öka selektiviteten för vätgasutveckling genom att hindra sidoreaktioner. Filmerna är dock inte lika effektiva som de filmer som bildas från den ohälsosamma tillsatsen Cr(VI), vilken används i kloratprocessen idag. Trivalenta katjoner är inte lösliga i kloratelektrolyt och är därför inte en lämplig tillsats i kloratprocessen. Molybdat har god löslighet och kan aktivera vätgasutveckling i ett stort pH‑intervall på titan och andra substratmaterial som själva har betydlig sämre aktivitet för vätgasutveckling. Vor kurzem wurde MOT1 als Molybdat-Transportprotein in Arabidopsis thaliana identifiziert. Unter Zuhilfenahme von GFP-Fusionsproteinen konnte als subzelluläre Lokalisierung des Proteins die Membran des Endoplasmatischen Retikulums (ER) in dieser Arbeit identifiziert werden. Auch wurde hier demonstriert, dass das mit Abstand nächste Homolog des Proteins, MOT2, ist in der vakuolären Membran, dem Tonoplasten, lokalisiert ist. Unter Standarbedingungen zeigten mot1-KO-Pflanzen reduzierte Molybdatgehalte im Blatt und Keimlinge wiesen Wachstumsdefizite in Abwesenheit von Molybdat auf. Dies führte zu der Annahme, dass MOT1 nicht, wie bislang angenommen, den Hauptimporter für Molybdat in die Zelle darstellt. Durch die Quantifizierung vakuolärer Molybdatgehalte konnte einerseits die Vakuole als Hauptspeicherort für Molybdat in der pflanzlichen Mesophyllzelle und andererseits MOT1 als wichtigstes Protein zur Beladung der Vakuole mit dem Spurenelement identifiziert werden. Die Blätter von mot2-KO-Pflanzen zeigten erhöhte Molybdatgehalte und die Bedeutung des Proteins für die Remobilisierung des essentiellen Molybdats während der Seneszenz konnte mittels Transkriptanalysen und Molybdatquantifizierung in Samen und seneszenten Blättern gezeigt werden. Mittels Fluoreszenzanalysen konnte zusätzlich eine wichtige Determinante der Zielsteuerung von Proteinen zum Tonoplasten identifiziert werden. Durch Mutagenese zweier Leucinreste am N-Terminus von MOT2 ist das korrekte Targeting in die vakuoläre Membran gestört und das Protein lokalisiert fälschlicherweise in der Plasmamembran. Es konnte ein Modell für den intrazellulären Molybdattransport vorgestellt werden. So transportiert MOT1 das Anion ins ER, von wo aus es über Vesikelfluss zur Vakuole gelangt. MOT2 stellt das tonoplastidäre Protein für den Export des Spurenelements aus der Vakuole, besonders während der Seneszenz, dar. Auch konnte eine Korrelation zwischen dem Molybdatgehalt und der Konzentration von Moco, sowie dessen Vorstufe MPT, identifiziert werden. Dies liefert Hinweise auf eine Regulation der Moco-Biosynthese in Abhängigkeit von der zellulären Molybdatkonzentration. As well, the subcellular localization of the close homologue was determined. This protein, named MOT2, resides in the vacuole surrounding membrane, the tonoplast. Arabidopsis mot1 knock out plants exhibited reduced leaf molybdate levels and seedlings grown in the absence of molybdate were retarded in growth and development. This led to the conclusion that MOT1 does not act as the main molybdate importer in the cell, as proposed before. By quantification of vacuolar molybdate contents the vacuole was identified as the main storage compartment for molybdate and MOT1 was identified as the main transporter involved in vacuolar loading with the trace element. Leaves of mot2 knock out plants exhibit increased molybdate levels implying a function of the protein in vacuolar molybdate export. This function is essential during remobilization of the anion from leaves during senescence. Observations leading to this conclusion were obtained in this work by investigating transcript levels of mot2 mRNA and quantification of molybdate in seeds and senescent leaves from mot2 knock out plants. Using fluorescence analyses an important component of protein targeting to the vacuole was identified. Mutagenesis of an N-terminal Dileucin motif in MOT2 led to mistargeting of the protein to the plasma membrane, revealing the importance of two leucin residues for protein sorting to the vacuolar membrane. Furthermore a correlation between molybdate concentration and the levels of Moco and its precursor MPT was observed. This suggests a possible regulation of Moco biosynthesis according to molybdate concentration. A summarizing hypothetic model of intracellular molybdate transport in the plant could be created. According to the data raised in this work, molybdate is loaded to the ER lumen via MOT1 activity and subsequently released to the vacuole by vesicle flux. The export of the trace element from its storage compartment, the vacuole, is fulfilled by the activity of MOT2. Latter process is of special importance during senescence.
