This study presents the initial attempt at introducing a magnetic molecularly imprinted polymer (MIP) designed specifically for lamotrigine with the purpose of functioning as a drug carrier. First, the composition of the magnetic polymer underwent optimization based on bulk polymer adsorption studies and theoretical analyses. The magnetic MIP was synthesized from itaconic acid and ethylene glycol dimethacrylate exhibiting a drug loading capacity of 3.4 ± 0.9 μg g -1 . Structural characterization was performed using powder X-ray diffraction analysis, vibrating sample magnetometry, and Fourier transform infrared spectroscopy. The resulting MIP demonstrated controlled drug released characteristics without a burst effect in the phospahe buffer saline at pH 5 and 8. These findings hold promise for the potential nasal administration of lamotrigine in future applications.

The aim of this study was to develop magnetic molecularly imprinted nano-conjugate sorbent for effective dispersive solid phase extraction of antazoline (ANT) and its metabolite, hydroxyantazoline (ANT-OH) in analytical method employing liquid chromatography coupled with mass spectrometry method. The core-shell material was characterized in terms of adsorption properties, morphology and structure. The heterogeneous population of adsorption sites towards ANT-OH was characterized by two K d and two B max values: K d (1) = 0.319 µg L -1 and B max (1) = 0.240 μg g -1 , and K d (2) = 34.6 µg L -1 and B max (2) = 5.82 μg g -1 . The elemental composition of magnetic sorbent was as follows: 17.55, 37.33, 9.14, 34.94 wt% for Si, C, Fe and O, respectively. The extraction protocol was optimized, and the obtained results were explained using theoretical analysis. Finally, the analytical method was validated prior to application to pharmacokinetic study in which the ANT was administrated intravenously to three healthy volunteers. The results prove that the novel sorbent could be useful in extraction of ANT and ANT-OH from human plasma and that the analytical strategy could be a versatile tool to explain a potential and pharmacological activity of ANT and ANT-OH.

The paper describes the formation of six aromatic N-(2-arylethyl)-2-methylprop-2-enamides with various substituents in benzene ring, viz., 4-F, 4-Cl, 2,4-Cl2, 4-Br, 4-OMe, and 3,4-(OMe)2 from 2-arylethylamines and methacryloyl chloride in ethylene dichloride with high yields (46–94%). The structure of the compounds was confirmed by 1H NMR, 13C NMR, IR, and HR-MS. Those compounds were obtained to serve as functionalized templates for the fabrication of molecularly imprinted polymers followed by the hydrolysis of an amide linkage. In an exemplary experiment, the imprinted polymer was produced from N-(2-(4-bromophenyl)ethyl)-2-methylprop-2-enamide and divinylbenzene, acting as cross-linker. The hydrolysis of 2-(4-bromophenyl)ethyl residue proceeded and the characterization of material including SEM, EDS, 13C CP MAS NMR, and BET on various steps of preparation was carried out. The adsorption studies proved that there was a high affinity towards the target biomolecules tyramine and L-norepinephrine, with imprinting factors equal to 2.47 and 2.50, respectively, when compared to non-imprinted polymer synthesized from methacrylic acid and divinylbenzene only. The objective of this article was to design the selective molecularly imprinted sorbent dedicated to the solid‐phase extraction of S‐pramipexole from the complex matrix such as human urine. For that purpose, S‐2,6‐diamino‐4,5,6,7‐tetrahydrobenzothiazole was used as the template acting as the structural analog of S‐pramipexole and five various monomers were employed in the presence of ethylene glycol dimethacrylate to produce molecularly imprinted polymers. The binding capabilities of resulted polymers revealed that the highest imprinting effect was noted for polymer prepared from the itaconic acid. The comprehensive analysis of morphology and the characterization of binding sites showed not only negligible differences in the extension of surfaces of imprinted and nonimprinted polymers but also higher heterogeneity of binding sites in the imprinted material. Comprehensive optimization of the molecularly imprinted solid‐phase extraction allowed to select the most appropriate solvents for loading, washing, and elution steps. Subsequent optimization of mass of sorbent and volumes of solvents allowed to achieve satisfactory total recoveries of S‐pramipexole from the model multicomponent real sample of human urine that equals to 91.8 ± 3.2% for imprinted sorbent with comparison to only 37.1 ± 1.1% for Oasis MCX. This review presents the current status of molecularly imprinted polymers (MIPs) for drug delivery, in particular the studies that focus on biocompatibility, cytotoxicity, and in vitro or in vivo behavior of MIPs. It also shows the limitations that hamper the introduction of MIPs to pharmacotherapy and prevent this class of polymers from commercialization. MIPs are promising materials in the construction of drug delivery devices because they can provide improved delivery profiles or longer release times and deliver the drugs in the feedback regulated way, which is extremely important in modern pharmacotherapy. Here, a brief overview of the imprinting process and a concise description of drug release mechanisms from the imprinted materials will be presented followed by the discussion of potential MIP drug delivery devices for ocular, dermal, intravenous and oral routes of administration. This study presents a validated strategy for the determination of tryptamine in the presence of its competitors, which involves the molecularly imprinted solid-phase extraction combined with high-performance liquid chromatography coupled with fluorimetric detection. Tryptamine-imprinted microscale sorbent was produced from 4-vinylbenzoic acid and ethylene glycol dimethacrylate in methanol by precipitation polymerization, and its imprinting factor was equal to 15.4 in static experiments or 18.6 in dynamic binding experiments. The method for tryptamine determination in the presence of serotonin and l-tryptophan was validated using a complex matrix of bovine serum albumin yielding the recoveries of tryptamine that ranged between 98.7 and 107.0%. Very low limits of detection and limits of quantification for tryptamine (19.9 and 60.3 nmol/L, respectively) allow the quantification of tryptamine in human cerebrospinal fluid in the presence of tryptophan and serotonin. In this paper, the optimized synthesis and detailed characterization of novel imprinted material for selective extraction of 2-aminothiazoline-4-carboxylic acid (ATCA) were described. The prepolymeric system contained 1-allyl-2-thiourea and ethylene glycol dimethacrylate in methanol, tetrahydrofuran and dimethyl sulfoxide porogenic mixture and 2-aminothiazole-4-carboxylic acid which was used as the template for ATCA. This structural analog of the target analyte was found to provide the imprinted polymer with sufficient binding capacity (60.7 ± 0.9 μg g −1 ) and high selectivity (imprinting factor equal to 18.4) toward ATCA. The adsorption of ATCA was analyzed by the Langmuir model. The heterogeneous population of binding sites on the imprinted polymer was characterized by dissociation constants equal to 3.72 μg L −1 and 435 μg L −1 for high and low affinity binding sites, respectively. The morphology of the polymer was studied employing SEM and BET analyses and the composition was confirmed by EDS and 13 C CP/MAS NMR analyses. Adsorption of amino acids on the imprinted material was tested to analyze the impact of the sample components. The superiority of the imprinted sorbent was proved in a novel dispersive solid phase extraction procedure of ATCA from post-mortem whole blood with respect to the extraction efficacy on the commercial ion-exchange sorbents. The limit of quantification and limit of detection of ATCA in the new analytical method were 12 μg L −1 and 3.5 μg L −1 , respectively. Here, seven cross-linkers (six polar diacrylates or dimethacrylates of different lengths between double bonds, and one aromatic-divinylbenzene) were used to examine the impact of the cross-linker on binding capacity and selectivity of 3,3′-diindolylmethane (DIM) imprinted material. DIM participates in the suppression of viability of human ovarian and human breast cancer cell lines, but has low bioavailability. The investigations of novel imprinted polymer matrices for improvement of DIM release could allow to utilize not only a potency of DIM but also similar alkaloids, which are the important compounds with pharmacological activity. The bulk, thermal radical copolymerization of the cross-linkers in the presence of 3,3′-diindolylmethane (the template) and allylamine (the functional monomer) in dimethyl sulfoxide or in carbon tetrachloride (porogens) was carried out. The binding capacities of imprinted and non-imprinted polymers were compared, and two polymers (these were prepared using ethylene glycol dimethacrylate and polyethylene glycol dimethacrylate as the cross-linkers) with the highest selectivity and binding capacity were selected to desorption test. The desorption profile of polymer prepared using polyethylene glycol dimethacrylate as the cross-linker revealed sustained release of 3,3′-diindolylmethane, and this system was selected for further optimization of the cross-linker amounts. The morphology and structure of the selected particles were analyzed using SEM micrographs, 13 C CP/MAS NMR spectroscopy, and BET measurements. The desorption of 3,3′-diindolylmethane from poly(allylamine- co -polyethylene glycol dimethacrylate) particles was in accordance with pseudo-second-order kinetics and the simplified Higuchi model indicated the diffusion controlled release of 3,3′-diindolylmethane. A 2-(4-methoxyphenyl)ethylamine imprinted polymer (MIP) was successfully applied for the selective separation of tyramine. A computational analysis was used to predict the affinity of the polymer matrix towards tyramine and a preliminary experimental evaluation was made for the target analyte. Then the experimental analysis of polymer towards tyramine was continued. The binding sites were characterized with employment of the Langmuir and Freudlich models. After the optimization of solid phase extraction towards tyramine, the most appropriate systems for the extraction steps were chosen: methanol-water 85:15 v/v for the loading and the washing as well as 0.04 M aq. ammonium acetate-methanol 30:70 v/v for the elution steps. The biogenic compounds as tryptamine, serotonin, octopamine, synephrine, and l-tyrosine were used for the selectivity study on the basis of binding capacities of the analytes on the imprinted and the non-imprinted polymers. The theoretical approach to obtained results allowed to explain the adsorption selectivity of the tested polymer. Finally, the complex matrix of bovine serum albumin was used to show the usefulness of imprinted material for bioanalysis. The obtained recoveries showed the superiority of MIP over the commercial sorbent C18. Total recoveries of tyramine from spiked bovine serum albumin sample were determined as: 95±2%, 14±3%, and 1.9±0.4% for the imprinted, non-imprinted, and commercial C18 sorbents, respectively. A novel sorbent for the separation of 3,3′-diindolylmethane ( DIM) has been developed. DIM was used as a template molecule to prepare twelve imprinted polymers from seven different functional monomers in six various porogens in the presence of ethylene glycol dimethacrylate as a crosslinker. The binding capacity showed that the highest binding specificity was achieved when allylamine as the monomer and carbon tetrachloride as the porogen were used to form bulk polymers. Binding capacities were 130.4 ± 2.8 µg g−1 for imprinted MIP7a polymer versus 49.03 ± 0.44 µg g−1 for non-imprinted NIP7a polymer. An imprinting factor of 3.12 reflected the high specificity of MIP7a. A solid-phase extraction protocol was fully optimized. Two methanol-water systems were selected as the most effective for loading and washing steps, while acetonitrile was chosen as the most efficient eluent suitable for recovery of 91.7 ± 3.8% of DIM. Optimal concentration of DIM and volume of loaded sample were also examined. A loaded volume of 2.5 mL was the most appropriate for 0.625 and 1.25 mg L−1. The use of MIP7a for the separations of DIM from spiked bovine serum albumin was evaluated, showing a total recovery of 87.7 ± 3.5%. Abstract: The aim of the study was to develop an efficient sorbent for the separation of dopamine. 4-(2-Aminoethyl)aniline was chosen as a pseudo-template to produce the imprinted polymers from seven different functional monomers in the presence of ethylene glycol dimethacrylate as a cross-linker. The binding capacity showed that the highest binding specificity towards dopamine was achieved when methacrylic acid was used as the monomer in methanol solution to form a polymer matrix. The imprinting factor value was equal to 22.96. Other biogenic amines were bound much more weakly. A simple theoretical model was used to give an insight into the imprinting process and the selectivity of polymer matrix. Abstract: 2-(4-Methoxyphenyl)ethylamine imprinted polymers were obtained from seven functional monomers in four porogens, and their properties were tested. Binding experiments revealed the highest selectivity towards a template for the polymer prepared from methacrylic acid in toluene (MIP1). The binding capacities and the imprinting factors were different for the stationary and the dynamic evaluation procedures. For MIP1, the binding capacities were 6.991±0.081 or 18.247±0.005μmolg−1, and the imprinting factors were 1.97 or 3.84, for stationary and dynamic procedures, respectively. The Scatchard analysis of MIP1 showed two classes of binding sites with values of the dissociation constants K d equal to 16.2 and 192μmolL−1. Composition of polymers was supported by 13C CP/MAS NMR, FTIR and SEM-EDS analyses. The binding abilities of MIP1 towards the structurally related compounds showed that the ethylamine group together with steric effects governed the recognition mechanism. A 2-(3,4-dimethoxyphenyl)ethylamine imprinted polymer ( MIPpt) was prepared via the precipitation polymerization together with a nonimprinted polymer (NIP). The morphology of particles was investigated by scanning electron microscopy and the specific surface areas were estimated by methylene blue adsorption (60.5 ± 3.5 and 36.9 ± 1.2 m2/g for MIPpt and NIP, respectively). The binding experiments were performed to determine the binding capacity of MIPpt/ NIP particles toward dopamine. Next, the effects of solvents on loading, washing, and eluting steps were examined on solid-phase extraction ( SPE). Methanol-water 85:15 v/v (loading step), methanol (washing step), and 0.04 M aqueous ammonium acetate-methanol 30:70 v/v (eluting step) were selected as the most effective systems. Described SPE protocol was successfully applied for separation of dopamine on 2-(3,4-dimethoxyphenyl)ethylamine imprinted particles. Finally, the molecularly imprinted polymer was used for determination of dopamine in spiked banana extract. The total recovery of dopamine from MIPpt was equal to 88.5 ± 4.6%, but from NIP was only 12.8 ± 2.3%. The developed material and method were demonstrated to be applicable for the separation of dopamine from bananas. The commercial sorbent C18 was not suitable to such application. Abstract: A computational model was proposed to evaluate the affinity and selectivity of 2-(3,4-dimethoxyphenyl)ethylamine (homoveratrylamine) imprinted polymers. Four functional monomers: methacrylic acid, 1-vinylimidazole, 4-vinylpyridine, and allylamine were taken into account. Two dielectric constants were used for solvent simulations: a value of ɛ =2.38r ij for toluene was used in the analysis of prepolymerization complexes, and a value of ɛ =36r ij for methanol–water was used in the investigations of adsorption. Theoretical analysis predicted the highest affinity for the polymer synthesized from methacrylic acid. Experimental results confirmed the finding. The prepolymerization complex formed by homoveratrylamine and four methacrylic acid molecules was used to design the polymer cavity. The selectivity of the polymer was analyzed as a simulation of adsorption of six compounds in the cavity by docking procedure. Selected compounds are structurally related to the template or can be present in biological samples. The designed polymer has high selectivity towards homoveratrylamine. A bulk polymerization method was used to easily and efficiently prepare homo-veratric acid (3,4-dimethoxyphenylacetic acid)-imprinted polymers from eight basic monomers: 2-vinylpyridine, 4-vinylpyridine, 1-vinylimidazole, N-allylaniline, N-allylpiperazine, allylurea, allylthiourea, and allylamine, in the presence of homoveratric acid as a template in N,N-dimethylformamide as a porogen. The imprinted polymer prepared from allylamine had the highest affinity to the template, showing an imprinting factor of 3.43, and allylamine polymers MIP8/NIP8 were selected for further studies. Their binding properties were analyzed using the Scatchard method. The results showed that the imprinted polymers have two classes of heterogeneous binding sites characterized by two pairs of Kd, Bmax values: Kd(1) = 0.060 μmol/mL, Bmax(1) = 0.093 μmol/mg for the higher affinity binding sites, and Kd(2) = 0.455 μmol/mL, Bmax(2) = 0.248 μmol/mg for the lower affinity binding sites. Non-imprinted polymer has only one class of binding site, with Kd = 0.417 μmol/mL and Bmax = 0.184 μmol/mg. A computational analysis of the energies of the prepolymerization complexes was in agreement with the experimental results. It showed that the selective binding interactions arose from cooperative three point interactions between the carboxylic acid and the two methoxy groups in the template and amino groups in the polymer cavities. Those results were confirmed by the recognition studies performed with the set of structurally related compounds. Allylamine polymer MIP8 had no affinity towards biogenic amines. Abstract: The main objective of this paper was to examined the impact of synthetic reagents on morphology and recognition properties of 2-(3,4-dimethoxyphenyl)ethylamine imprinted polymers. The effect of nine different functional monomers, five porogens and four cross-linkers on the binding capacity of particles was analyzed. The results revealed that the highest imprinting factor (1.81) showed the polymer obtained from methacrylic acid and ethylene glycol dimethacrylate in toluene. The binding capacities of imprinted (MIP1) and non-imprinted (NIP1) materials were 135.3±9.8 and 74.8±7.8μmol g−1, respectively. The specific surface areas were 55.05±3.89 for MIP1 and 38.72±2.40m2 g−1 for NIP1. The SEM analysis confirmed that the surface of MIP1 is rougher and denser than NIP1. Structural analysis supported by 13C CP/MAS NMR spectra was also performed. The binding abilities of homoveratrylamine and eight structurally related compounds to MIP1 showed that strong interactions between carboxylic group in the polymer and amine group in the analyte together with its molecular volume govern the recognition mechanism. Eight different functional monomers were used with ethylene glycol dimethacrylate as a cross-linker and molsidomine as a template to obtain molecularly imprinted polymers (MIPs). Non-covalent interactions between molsidomine and each functional monomer in DMSO prior to thermal bulk polymerization were utilized. On the basis of calculated imprinting factors, MIP prepared with N,N'-diallyltartaramide was chosen for further investigations. Examination of interactions in the prepolymerization complex between molsidomine and N,N'-diallyltartaramide was performed using the Job method. The absorbance of isomolar solutions reaching a maximum for the molar ratio of template to monomer equal to 1:4. Scatchard analysis was used for estimation of the dissociation constants and the maximum amounts of binding sites. The polymer based on N,N'-diallyltartaramide has two classes of heterogeneous binding sites characterized by two values of Kd and two Bmax: Kd(1) = 1.17 mM-1 and Bmax(1) = 0.8 μmol/mg for the higher affinity binding sites, and Kd(2) = 200 μM-1 and Bmax(2) = 2.05 μmol/mg for the lower affinity binding sites. In the last 10 years, we have witnessed an extensive development of instrumental techniques in analytical methods for determination of various molecules and ions at very low concentrations. Nevertheless, the presence of interfering components of complex samples hampered the applicability of new analytical strategies. Thus, additional sample pre-treatment steps were proposed to overcome the problem. Solid sorbents were used for clean-up samples but insufficient selectivity of commercial materials limited their utility. Here, the application of molecularly imprinted polymers (MIPs) or ion-imprinted polymers (IIPs) in the separation processes have recently attracted attention due to their many advantages, such as high selectivity, robustness, and low costs of the fabrication process. Bulk or monoliths, microspheres and core-shell materials, magnetically susceptible and stir-bar imprinted materials are applicable to different modes of solid-phase extraction to determine target analytes and ions in a very complex environment such as blood, urine, soil, or food. The capability to perform a specific separation of enantiomers is a substantial advantage in clinical analysis. The ion-imprinted sorbents gained interest in trace analysis of pollutants in environmental samples. In this review, the current synthetic approaches for the preparation of MIPs and IIPs are comprehensively discussed together with a detailed characterization of respective materials. Furthermore, the use of sorbents in environmental, food, and biomedical analyses will be emphasized to point out current limits and highlight the future prospects for further development in the field. Ten molecularly imprinted polymers coded as MIP 1 –MIP 10 were prepared by the radical bulk polymerization using ( R , S )-(±)-2-amino-1-phenylethanol as the structural analog of the target analyte ( R , S )-octopamine. The functional monomers, 4-vinylbenzoic acid ( 1 ), methacrylic acid ( 2 ), acrylic acid ( 3 ), trifluoromethacrylic acid ( 4 ), itaconic acid ( 5 ), acrylamide ( 6 ), isopropenylbenzene ( 7 ), 2-hydroxyethyl methacrylate ( 8 ), 2-(diethylamino)ethyl methacrylate ( 9 ), allylamine ( 10 ) were polymerized consecutively with the ethylene glycol dimethacrylate cross-linker in methanol as the porogen. On the basis of the binding capacity of ( R , S )-octopamine MIP 1 with affinity factor equal to 6.37 was selected for further analysis. The affinity of polymer matrix MIP 1 was tested by the non-competitive binding experiments of eight structurally related analytes. Finally, molecularly imprinted solid phase extraction (MISPE) of ( R , S )-octopamine from spiked human serum albumin was carried out in order to verify the applicability of novel sorbent. In this paper the exploration of specific imprinted sorbent for 4-hydroxy-3-nitrophenylacetic acid (NHPA), an indicator of 3-nitrotyrosine, a biomarker of pathophysiological states, was described. The optimization of synthetic protocol, supported by theoretical studies, allowed to select NHPA imprinted poly(acrylic acid- co -ethylene glycol dimethacrylate) as the most specific polymer with imprinting factor equal to 2.27. The acrylic copolymers were characterized, revealing differences between the imprinted and non-imprinted material in the adsorption binding capacities (0.54 ± 0.12 and 0.335 ± 0.073 μmol g−1, respectively), heterogeneity indices (0.47 and 0.72), pseudo-second order kinetics constants (0.251 and 0.077 g μmol −1 min−1, respectively), and total specific surface areas (51.8 and 292.3 m2 g−1, respectively). The objective of this paper was to extend comprehensive theoretical and experimental investigations at the molecular level to identify factors responsible for the high selectivity of imprinted sorbents. This knowledge was utilized in a new analytical strategy devoted to the analysis of hordenine in human urine after beer consumption. Among the various polymeric compositions tested, the most effective material was built up from methacrylic acid and ethylene glycol dimethacrylate (MIP 1), showing a satisfactory binding capacity (4.44 ± 0.15 µmol g−1) and high specificity towards hordenine (AF = 5.90). The comprehensive analyses of porosity data and surface measurements revealed differences between imprinted polymers. The characterization of binding sites of MIP 1 revealed a heterogeneous population with two values of K d (2.75 and 370 μmol L−1) and two values of B max (1.82 and 99 μmol g−1) for higher and lower affinity respectively. The extensive theoretical analyses of interactions between various analytes and the MIP model cavity showed the highest binding energy for hordenine (ΔE B1 = −175.17 kcal mol−1). The method was validated for selectivity, lowest limit of quantification, calibration curve performance, precision, accuracy, matrix effect, carry-over and stability in urine. The validation criteria were fulfilled, and the method was satisfactorily applied to urine samples collected prior to, and 2 h after, consumption of 2 L of beer, revealing the presence of hordenine at the mean level of 129 ± 27 ng mL−1. Additionally, ability of the sorbent to purify the urine sample was assessed using flow injection analysis tandem mass spectrometry, for comparison with other extraction techniques.
